Abstract:In January 2014, severe haze episodes which sweep across Chinese cities have attracted public concern and interest at home and abroad. In addition to the physicochemical properties of air pollutants, bacteria are thought to be responsible for the spread of respiratory diseases and various allergies. We attempted the bacterial characterization of submicron particles (PM0.18-0.32, PM0.32-0.56, and PM0.56-1) under severe haze episodes using high-throughput sequencing and real-time quantitative PCR detecting system based on 21 samples collected from January to March 2014 at Ji'nan, China. The high bacterial concentration in PM0.32-0.56 (7314cells m-3), PM0.18-0.32 (7212cells m-3), and PM0.56-1 (6982cells m-3) showed significant negative correlations with SO2, NO2, and O3. Under sufficient sequencing depth, 37 phyla, 71 classes, 137 orders, 236 families, and 378 genera were classified, and the bacterial community structure varied significantly in different size fractions. For example, Holophagaceae (Acidobacteria) in PM0.32-0.56 showed 6-fold higher abundance than that in PM0.18-0.32. Moreover, functional categories and bacterial species (Lactococcus piscium, Pseudomonas fragi, Streptococcus agalactiae, and Pseudomonas cichorii) that may potentially be responsible for infections and allergies were also discovered. Source track analysis showed that the ambient bacteria mainly originated from soils, leaf surfaces, and feces. Our results highlighted the importance of airborne microbial communities by understanding the concentration, structure, ecological and health effects, especially those in submicron particles during haze episodes.

Abstract:Bacteria are abundant in atmospheric water phase with the potential to influence atmospheric processes and human health, yet relatively little information is known about the bacterial characteristics at high altitudes. Here we investigated the bacterial community by high throughput sequencing in 24 cloud water samples collected from September 26 to October 31, at the summit of Mt. Tai (36°15' N, 117°06' E, 1534m a.s.l) in China. Diverse bacterial population were identified and the gram-negative bacteria contributed the majority of total bacteria including Proteobacteria (81.6%) and Bacteroidetes (3.9%), followed by gram-positive bacteria Firmicutes (7.1%) and Actinobacteria (2.3%). These gram-negative taxa mainly inhabited in leaf-surface and cold environments. Meanwhile bacteria involved in the cloud condensation nuclei and ice nuclei formation were observed such as Sphingomonas (6.7%), Pseudomonas (4.1%), and Bacillus (1.1%). In addition, Sphingmonas was more active than that in daytime and participated in the cloud chemistry process. Meanwhile O3 and SO2 critically contributed to the variation of bacterial community. It is the first report on the bacterial community structure of cloud water over Asian area. Our results can serve as an important reference for environmental scientists, and biologists.
Bacterial community structure in atmospheric particulate matters of different sizes during the haze days in Xi'an, China

Abstract:Serious air pollution events have frequently occurred in China associated with the acceleration of urbanization and industrialization in recent years. Exposure to atmospheric particulate matter (PM) of high concentration can lead to adverse effects on human health. Airborne bacteria are important constituents of microbial aerosols and contain lots of pathogens. However, variations in bacterial community structure in atmospheric PM of different sizes (PM2.5, PM10 and TSP) have not yet been explored. In this study, PM samples of different sizes were collected during the hazy days from Jul.2016 to Apr.2017 to determine bacterial diversity and community structure. Samples from soils and leaf surfaces were also collected to determine potential sources of bacterial aerosols. High-throughput sequencing technology was used generate bacterial community profiles, where we determined their diversity and abundances in the samples. Results showed that the dominant bacterial community structures in PM2.5, PM10 and TSP were strongly similar. Compared with non-haze days, the relative abundances of most bacterial pathogens on the haze days did not increase. Meanwhile, temperature, O3 and NO2 had more significant effects on bacterial community than the other environmental factors. Source tracking analysis indicated that the airborne bacteria might be not from local environment. It may come from the entire city or other regions by long distance airflow transport. Results of this study improved our understanding of the influence of bioaerosols on human health and the potential sources of airborne microbes.

Abstract:Urban aerosols harbour diverse bacterial communities in Shanghai. • The functional groups were associated with nitrogen, carbon, and sulfur cycling. • Temperature, SO2, and wind speed were key drivers for the bacterial community. Airborne bacteria play key roles in terrestrial and marine ecosystems and human health, yet our understanding of bacterial communities and their response to the environmental variables lags significantly behind that of other components of PM2.5. Here, atmospheric fine particles obtained from urban and suburb Shanghai were analyzed by using the qPCR and Illumina Miseq sequencing. The bacteria with an average concentration of 2.12 × 103 cells/m3, were dominated by Sphingomonas, Curvibacter, Acinetobacter, Bradyrhizobium, Methylobacterium, Halomonas, Aliihoeflea, and Phyllobacterium, which were related to the nitrogen, carbon, sulfur cycling and human health risk. Our results provide a global survey of bacterial community across urban, suburb, and high-altitude sites. In Shanghai (China), urban PM2.5 harbour more diverse and dynamic bacterial populations than that in the suburb. The structural equation model explained about 27%, 41%, and 20%–78% of the variance found in bacteria diversity, concentration, and discrepant genera among urban and suburb sites. This work furthered the knowledge of diverse bacteria in a coastal Megacity in the Yangtze river delta and emphasized the potential impact of environmental variables on bacterial community structure.
Fungi Diversity in PM2. 5 and PM1 at the Summit of Mt. Tai: Abundance, Size Distribution, and Seasonal Variation

Abstract:Fungi are ubiquitous throughout the near-surface atmosphere, where they represent an important component of primary biological aerosol particles. This study combined internal transcribed spacer region sequencing and quantitative real-time polymerase chain reaction (qPCR) to investigate the ambient fungi in fine (PM2. 5, 50 % cutoff aerodynamic diameter Da50 = 2.5 µm, geometric standard deviation of collection efficiency σg = 1.2) and submicron (PM1, Da50 = 1 µm, σg = 1.2) particles at the summit of Mt. Tai located in the North China Plain, China. Fungal abundance values were 9.4 × 104 and 1.3 × 105 copies m−3 in PM2. 5 and PM1, respectively. Most of the fungal sequences were from Ascomycota and Basidiomycota, which are known to actively discharge spores into the atmosphere. The fungal community showed a significant seasonal shift across different size fractions according to Metastats analysis and the Kruskal–Wallis rank sum test. The abundance of Glomerella and Zasmidium increased in larger particles in autumn, whereas Penicillium, Bullera, and Phaeosphaeria increased in smaller particles in winter. Environmental factors, namely Ca2+, humidity, and temperature, were found to be crucial for the seasonal variation in the fungal community. This study might serve as an important reference for fungal contribution to primary biological aerosol particles.

Abstract:Primary biological aerosol particles are ubiquitous in the global atmosphere and can affect cloud formation, deteriorate air quality, and cause human infections. Mt. Tai (1,534 m a.s.l.) is an elevated site in the North China Plain where atmospheric aerosols reflect both regional advection and long‐range transport. In this study, we deployed a Wideband Integrated Bioaerosol Sensor (WIBS‐4A) and collected total suspended particles and eight‐stage size‐segregated aerosol samples at the summit of Mt. Tai in spring from 21 March to 8 April 2017 to quantify the abundance, size distributions, and diurnal variations of fluorescent bioaerosols and to investigate the effect of different fluorescence thresholds of WIBS for ambient bioaerosol recognition. During the whole sampling period, the number concentration of fluorescent particles (>0.8 μm) was 647 ± 533 L−1, accounting for 26.9% ± 10.0% by number of the total particles in that size range. Three‐dimensional excitation‐emission matrix fluorescence of water‐soluble organic matter in size‐segregated aerosols shows that humic‐like substances (HULIS) are mainly in the fine mode (<2.1 μm) while protein‐like substances are mainly in the coarse mode (>2.1 μm). From the diurnal variation, it is shown that bioaerosols can undergo transformation during long‐range transport and contribute to HULIS. For bioaerosol recognition, we find that 6σ‐threshold can lead to better classification of fluorescent aerosol particles for fungal spores. Overall, our results constrain the abundance of primary bioaerosols in the troposphere over East Asia and elucidate the processes for their evolution via mountain/valley breezes and long‐range transport.
Characteristics of Bacterial Community in Cloud Water at Mt Tai: Similarity and Disparity under Polluted and Non-Polluted Cloud Episodes

Abstract:Bacteria are widely distributed in atmospheric aerosols and are indispensable components of clouds, playing an important role in the atmospheric hydrological cycle. However, limited information is available about the bacterial community structure and function, especially for the increasing air pollution in the North China Plain. Here, we present a comprehensive characterization of bacterial community composition, function, variation, and environmental influence for cloud water collected at Mt Tai from 24 July to 23 August 2014. Using Miseq 16S rRNA gene sequencing, the highly diverse bacterial community in cloud water and the predominant phyla of Proteobacteria, Bacteroidetes, Cyanobacteria, and Firmicutes were investigated. Bacteria that survive at low temperature, radiation, and poor nutrient conditions were found in cloud water, suggesting adaption to an extreme environment. The bacterial gene functions predicted from the 16S rRNA gene using the Phylogenetic Investigation of Communities by Reconstruction of Unobserved States (PICRUSt) suggested that the pathways related to metabolism and disease infections were significantly correlated with the predominant genera. The abundant genera Acinetobacter, Stenotrophomonas, Pseudomonas, and Empedobacter originated from a wide range of habitats including cloud condensation nuclei and ice nuclei active species, opportunistic pathogens, and functional species, demonstrating the importance of ecology and health in cloud water. Cluster analysis including hierarchical cluster (Hcluster) and principal coordinate analysis (PCoA) indicated a significant disparity between polluted and non-polluted samples. Linear discriminant analysis effect size (LEfSe) demonstrated that potential pathogens were enriched in the polluted cloud samples, whereas the diverse ecological function groups were significant in the non-polluted samples. Discrepant community structure determined by redundancy analysis (RDA) indicated that the major ions in cloud water and PM2.5 in the atmosphere have a negative impact on bacteria, playing a vital role in shaping microbial community structure. The major ions might provide nutrition to bacteria and directly influence the bacterial community, whereas PM2.5 in air has an indirect impact on bacterial community structure. During wet deposition, soluble particulate matter was dissolved in water droplets resulting in elevated concentration in cloud water. PM2.5 was possibly associated with different origins and pathways of air mass as determined using source tracking by the backward trajectory, mainly related to long-range transport. This work enhanced our understanding of the characteristics of bacterial ecology in the atmospheric aqueous phase, highlighting the potential influence of environmental variables on the bacterial community in cloud processes. It may provide fundamental information of the bacterial community response in cloud water under increasing pollution. However, due to the limited sample size (13 samples) collected at the summit of Mt Tai, these issues need in-depth discussion. Further studies based on an annual series of field observation experiments and laboratory simulations will continue to track these issues.

Abstract:Particulate matter (PM) air pollution poses a formidable public health threat to the city of Beijing. Among the various hazards of PM pollutants, microorganisms in PM2.5 and PM10 are thought to be responsible for various allergies and for the spread of respiratory diseases. While the physical and chemical properties of PM pollutants have been extensively studied, much less is known about the inhalable microorganisms. Most existing data on airborne microbial communities using 16S or 18S rRNA gene sequencing to categorize bacteria or fungi into the family or genus levels do not provide information on their allergenic and pathogenic potentials. Here we employed metagenomic methods to analyze the microbial composition of Beijing's PM pollutants during a severe January smog event. We show that with sufficient sequencing depth, airborne microbes including bacteria, archaea, fungi, and dsDNA viruses can be identified at the species level. Our results suggested that the majority of the inhalable microorganisms were soil-associated and nonpathogenic to human. Nevertheless, the sequences of several respiratory microbial allergens and pathogens were identified and their relative abundance appeared to have increased with increased concentrations of PM pollution. Our findings may serve as an important reference for environmental scientists, health workers, and city planners.
Effects of Haze Pollution on Microbial Community Changes and Correlation with Chemical Components in Atmospheric Particulate Matter.

Abstract:In this study, particulate matter (PM) with aerodynamic diameters of <= 2.5 and <= 10 mu m (PM2.5 and PM10, respectively), which was found at different concentrations in spring, was collected in Beijing. The chemical composition and bacterial community diversity of PM were determined, and the relationship between them was studied by 16S rRNA sequencing and mathematical statistics. Chemical composition analysis revealed greater relative percentages of total organic compounds (TOC) and secondary ions (NO3-, SO42-, and NH4+). The concentrations of Ca2+, Na+, Mg2+, K+ and SO42+ increased in high-concentration PM, which was associated with the contribution of soil, dust and soot. Microbiological analysis revealed 1191 operational taxonomic units. Microbial community structure was stable at the phylum level. The most abundant phyla were Proteobacteria, Actinobacteria, Firmicutes, Bacteroidetes and Cyanobacteria. Community clustering analysis at the genus level showed that the difference in bacterial community structure between different PM concentrations (clean air vs. smog) was greater than that between different particle sizes. The dominant genera varied in different concentrations of PM. An unclassified genus of Cyanobacteria and Comamonadaceae were most abundant in low-and high-concentration PM, respectively. The microbial community structure was dynamic at the genus level due to different environmental factors. The dominant bacteria in high-concentration PM were widely distributed in soils, indicating that the soil contributed more to the increase in the PM. The individual microbes that were detected did not increase significantly as the PM concentration increased. The bacterial community structure was strongly correlated with K+, Ca2+, Na+, Mg2+, SO42- and TOC in high-concentration PM and had a good correlation with NO3-, Cl-, NH4+ and TIC in low-concentration PM. Soil and dust contributed to the increase in the concentration of the particles, and the relevant chemical components also produced differences in the bacterial community structure in different concentrations of PM. [Characterizing Beijing's Airborne Bacterial Communities in PM2.5 and PM1 Samples During Haze Pollution Episodes Using 16S Rrna Gene Analysis Method].

Abstract:During 8th-14th Jan., 2013, severe particulate matter (PM) pollution episodes happened in Beijing. These air pollution events lead to high risks for public health. In addition to various PM chemical compositions, biological components in the air may also impose threaten. Little is known about airborne microbial community in such severe air pollution conditions. PM2.5 and PM10 samples were collected during that 7-day pollution period. The 16S rRNA gene V3 amplification and the MiSeq sequencing were performed for analyzing these samples. It is found that there is no significant difference at phylum level for PM2.5 bacterial communities during that 7-day pollution period both at phylum and at genus level. At genus level, Arthrobacter and Frankia are the major airborne microbes presented in Beijing winter.samples. At genus level, there are 39 common genera (combined by first 50 genera bacterial of the two analysis) between the 16S rRNA gene analysis and those are found by Metagenomic analysis on the same PM samples. Frankia and Paracoccus are relatively more abundant in 16S rRNA gene data, while Kocuria and Geodermatophilus are relatively more abundant in Meta-data. PM10 bacterial communities are similar to those of PM2.5 with some noticeable differences, i.e., at phylum level, more Firmicutes and less Actinobacteria present in PM10 samples than in PM2.5 samples, while at genus level, more Clostridium presents in PM10 samples. The findings in Beijing were compared with three 16S rRNA gene studies in other countries. Although the sampling locations and times are different from each other, compositions of bacterial community are similar for those sampled at the ground atmosphere. Airborne microbial communities near the ground surface are different from those sampled in the upper troposphere.
PM2.5 Drives Bacterial Functions for Carbon, Nitrogen, and Sulfur Cycles in the Atmosphere.

Abstract:Airborne bacteria may absorb the substance from the atmospheric particles and play a role in biogeochemical cycling. However, these studies focused on a few culturable bacteria and the samples were usually collected from one site. The metabolic potential of a majority of airborne bacteria on a regional scale and their driving factors remain unknown. In this study, we collected particulates with aerodynamic diameter ≤2.5 μm (PM2.5) from 8 cities that represent different regions across China and analyzed the samples via high-throughput sequencing of 16S rRNA genes, quantitative polymerase chain reaction (qPCR) analysis, and functional database prediction. Based on the FAPROTAX database, 326 (80.69%), 191 (47.28%) and 45 (11.14%) bacterial genera are possible to conduct the pathways of carbon, nitrogen, and sulfur cycles, respectively. The pathway analysis indicated that airborne bacteria may lead to the decrease in organic carbon while the increase in ammonium and sulfate in PM2.5 samples, all of which are the important components of PM2.5. Among the 19 environmental factors studied including air pollutants, meteorological factors, and geographical conditions, PM2.5 concentration manifested the strongest correlations with the functional genes for the transformation of ammonium and sulfate. Moreover, the PM2.5 concentration rather than the sampling site will drive the distribution of functional genera. Thus, a bi-directional relationship between PM2.5 and bacterial metabolism is suggested. Our findings shed light on the potential bacterial pathway for the biogeochemical cycling in the atmosphere and the important role of PM2.5, offering a new perspective for atmospheric ecology and pollution control.

Abstract:As an important part of atmospheric aerosol, airborne bacteria have major impacts on human health. However, variations of airborne community structure due to human-induced activities and their possible impact on human health have not been well understood. In this study, we sampled atmospheric microbial aerosols in three Chinese cities (Shanghai, Xiamen, and Zhangzhou) with contrasting social-economic levels and analyzed the bacterial composition using high-throughput sequencing methods. A high similarity of the predominant phyla was observed in three cities but the relative abundances were quite different. At the genus level, the most dominant genus in Shanghai and Xiamen was Deinococcus while the most dominant genus in Zhangzhou was Clostridium. The different characteristics of airborne bacterial in the three cities above may be ascribed to the environmental variables affected by human over-activities such as the vehicle exhausts and coal-burning emissions in Shanghai, the tourist aggregation and construction works in Xiamen, the extensive uses of chemical fertilizers, and agricultural activities in Zhangzhou. The variation of the bacterial community and the pathogenic bacteria detected in three cities would have a potential threat to human health

Abstract:Microorganisms existing in airborne fine particulate matter (PM2.5) have key implications in biogeochemical cycling and human health. In this study, PM2.5 samples, collected in the typical basin cities of Xi’an and Linfen, China, were analyzed through high-throughput sequencing to understand microbial seasonal variation characteristics and ecological functions. For bacteria, the highest richness and diversity were identified in autumn. The bacterial phyla were dominated by Proteobacteria, Actinobacteria, Firmicutes, and Bacteroidetes. Metabolism was the most abundant pathway, with the highest relative abundance found in autumn. Pathogenic bacteria (Pseudomonas, Acinetobacter, Serratia, and Delftia) were positively correlated with most disease-related pathways. Besides, C cycling dominated in spring and summer, while N cycling dominated in autumn and winter. The relative abundance of S cycling was highest during winter in Linfen. For fungi, the highest richness was found in summer. Basidiomycota and Ascomycota mainly constituted the fungal phyla. Moreover, temperature (T) and sulfur dioxide (SO2) in Xi’an, and T, SO2, and nitrogen dioxide (NO2) in Linfen were the key factors affecting microbial community structures, which were associated with different pollution characteristics in Xi’an and Linfen. Overall, these results provide an important reference for the research into airborne microbial seasonal variations, along with their ecological functions and health impacts.

Microbial Aerosol Characteristics in Highly Polluted and Near-Pristine Environments Featuring Different Climatic Conditions

Abstract:There is an increasing interest in understanding ambient bioaerosols due to their roles both in health and in climate. Here, we deployed an Ultraviolet Aerodynamic Particle Sizer to monitor viable (fluorescent) bioaerosol concentration levels at city centers (highly polluted) and their corresponding suburbs (near pristine) (total 40 locations) in 11 provinces featuring different climate zones in China between July 16 and 28, 2013. The concentration levels of viable bioaerosol particles (BioPM) of >0.5 μm were measured, and corresponding percentages of BioPM% (biological fraction of total PM) and BioPM2.5% (biological fraction of PM2.5) in particulate matter (PM) and BioPM, respectively, were determined. For some key cities, indoor viable bioaerosol levels were also obtained. In addition, bacterial structures of the air samples collected across these monitoring locations were studied using pyrosequencing. BioPM concentration levels ranged from 2.1 × 104 to 2.4 × 105/m3 for city centers [BioPM% = 6.4% (±6.3%)] and 0.5 × 104 to 4.7 × 105/m3 for suburbs [BioPM% = 10% (±8.7%)]. Distinctive bioaerosol size distribution patterns were observed for different climate zones, e.g., some had fluorescence peaks at 3 μm, while the majority had peaks at 1 μm. Ambient bacterial aerosol community structures were also found different for different geophysical locations. Results suggest that there was a poor overall relationship between PM and BioPM across 40 monitoring locations (R2 = 0.081, two-tailed P value = 0.07435). Generally, city centers had higher PM concentrations than suburbs, but not BioPM and BioPM%. Indoor bioaerosol levels were found at least tenfold higher than those corresponding outdoors. Bacillus was observed to dominate the bacterial aerosol community in the air sample.
Unique Atmospheric Microbiota Patterns for 31 Major Chinese Cities

Abstract:Microbial aerosol deposition and exposure represent a major health threat toward humans and ecology. Here, particulate matter (PM) samples from a total of 465 automobile air conditioning (AC) filters from 31 major Chinese cities were analyzed for the airborne patterns of bacterial, fungal and RNA viral communities and their influencing factors. Atmospheric microbiota were shown to vary greatly among 31 major cities. The relative abundances of some bacterial genera were extremely high, e.g., in Changsha PM about 20 times than other some cities for Brevibacterium. Significant correlations were found between latitude and bacteria diversity indexes (Shannon and Simpson index: negative correlation) as well as the relative abundance of some bacterial, fungal and viral genera (p-value <0.05). The relative abundances of genus Aspergillus were found much higher in PM samples from Changsha (51.22%) and Shanghai (23.25%) than other cities. Varying RNA virus structures were detected in 16 cities' samples with family Virgaviridae abundant from over half of the 16 cities. Using redundancy (RDA), variance partition (VPA) and aggregated boosted tree (ABT) analysis, the work showed that environ -mental factors (including geography factors), especially relative humidity, have played a leading role on the fungal structures, followed by air pollutants and social factors. In contrast, social factors, especially the sec-ondary industry, had the greatest impact on the bacteria community structures, followed by environmental factors and air pollutants. However, only temperature was shown to be correlated with the relative abundances of top 20 bacteria and fungi genera (random forest regression R-2 = 0.774). Here, distinctive microbiota patterns were detected being formed by environmental, social-economic and ecological factors, and further influenced by interactions among the microbial agents across 31 geophysical locations.
Morphology, Mixing State, and Hygroscopicity of Primary Biological Aerosol Particles from a Chinese Boreal Forest

Abstract:Abstract. Biological aerosols play an important role in atmospheric chemistry, clouds, climate, and public health. Here, we studied the morphology and composition of primary biological aerosol particles (PBAPs) collected in the Lesser Khingan Mountain boreal forest of China in summertime using transmission electron microscopy and scanning electron microscopy. Of all detected particles > 100 nm in diameter, 13 % by number were identified as PBAPs. In addition, 57 % of the PBAPs were identified as bacteria, followed by brochosomes (24 %) and fungi (19 %). The dominant size of bacteria was 1–4 μm, fungi was 2–4 μm, and brochosomes was 300–500 nm. The number size distribution of PBAPs coupled with the mass concentrations of PM2.5 and PM10 were used to estimate the total mass concentration of PBAPs, which is approximately 1.9 μg m−3 and accounts for 47 % of the in situ PM2.5–10 mass. C, N, O, P, K, and Si are detected in all PBAP particles, and P represented a major marker to identify PBAPs. Moreover, there is a higher frequency and concentration of PBAPs at night compared with day. Bacterial and fungal particles displayed weak hygroscopicity with a growth factor of ~ 1.09 at RH = 94 %. Electron microscopy shows that approximately 20 % of the bacterial particles were internally mixed with metal, mineral dust, and inorganic salts in the boreal forest air. This work provides a database for both further understanding physicochemical state of individual PBAP particles from natural sources and expanding the scope of atmospheric implications.
Effect of air pollution on the total bacteria and pathogenic bacteria in different sizes of particulate matter.

Abstract:In recent years, air pollution events have occurred frequently in China during the winter. Most studies have focused on the physical and chemical composition of polluted air. Some studies have examined the bacterial bioaerosols both indoors and outdoors. But few studies have focused on the relationship between air pollution and bacteria, especially pathogenic bacteria. Airborne PM samples with different diameters and different air quality index values were collected in Hangzhou, China from December 2014 to January 2015. High-throughput sequencing of 16S rRNA was used to categorize the airborne bacteria. Based on the NCBI database, the "Human Pathogen Database" was established, which is related to human health. Among all the PM samples, the diversity and concentration of total bacteria were lowest in the moderately or heavily polluted air. However, in the PM2.5 and PM10 samples, the relative abundances of pathogenic bacteria were highest in the heavily and moderately polluted air respectively. Considering the PM samples with different particle sizes, the diversities of total bacteria and the proportion of pathogenic bacteria in the PM10 samples were different from those in the PM2.5 and TSP samples. The composition of PM samples with different sizes range may be responsible for the variances. The relative humidity, carbon monoxide and ozone concentrations were the main factors, which affected the diversity of total bacteria and the proportion of pathogenic bacteria. Among the different environmental samples, the compositions of the total bacteria were very similar in all the airborne PM samples, but different from those in the water, surface soil, and ground dust samples. Which may be attributed to that the long-distance transport of the airflow may influence the composition of the airborne bacteria. This study of the pathogenic bacteria in airborne PM samples can provide a reference for environmental and public health researchers.

Abstract:Particulate matter (PM) has been a threat to the environment and public health in the metropolises of developing industrial countries such as Beijing. The microorganisms associated with PM have an impact on human health if they are exposed to the respiratory tract persistently. There are few reports on the microbial resources collected from PM and their antimicrobial activities. In this study, we greatly expanded the diversity of available commensal organisms by collecting 1,258 bacterial and 456 fungal isolates from 63 PM samples. A total of 77 bacterial genera and 35 fungal genera were included in our pure cultures, with Bacillus as the most prevalent cultured bacterial genus, Aspergillus , and Penicillium as the most prevalent fungal ones. During heavy-haze days, the numbers of colony-forming units (CFUs) and isolates of bacteria and fungi were decreased. Bacillus, Paenibacillus , and Chaetomium were found to be enriched during haze days, while Kocuria , Microbacterium , and Penicillium were found to be enriched during non-haze days. Antimicrobial activity against common pathogens have been found in 40 bacterial representatives and 1 fungal representative. The collection of airborne strains will provide a basis to greatly increase our understanding of the relationship between bacteria and fungi associated with PM and human health.

Abstract:Samples of atmospheric microbial aerosols were collected before, during, and after a dust invasion in Shanghai and analyzed using 16S rRNA high-throughput sequencing. The bacterial community structures in the mixed pollutive aerosols and dust were characterized, and the key environmental factors were identified. The dominant phyla were Proteobacteria, Actinomycetes, and Firmicutes, and the relative abundance of Acidobacteria increased significantly during the episode. Additionally, marked differences in the relative abundances of the 22 detected genera were observed between the three sampling stages: The dominant genera were Rubellimicrobium and Paracoccus prior to the arrival of the dust but became Deinococcus and Chroococcidiopsis during the invasion and then Clostridium and Deinococcus afterward. Notably, the relative abundance of Cyanobacteria, which is known to cause hepatotoxicity and promote tumor growth in humans, grew substantially during the event. Finally, statistical analysis revealed the largest environmental factors affecting the bacterial communities to be wind speed and the SO2, SO42-, NO3-, PM10, NH4+, and Ca-2(+) concentrations.
Characterization of Atmospheric Bioaerosols along the Transport Pathway of Asian Dust During the Dust-Bioaerosol 2016 Campaign

Abstract:Previous studies have shown that bioaerosols are injected into the atmosphere during dust events. These bioaerosols may affect leeward ecosystems, human health, and agricultural productivity and may even induce climate change. However, bioaerosol dynamics have rarely been investigated along the transport pathway of Asian dust, especially in China where dust events affect huge areas and massive numbers of people. Given this situation, the Dust-Bioaerosol (DuBi) Campaign was carried out over northern China, and the effects of dust events on the amount and diversity of bioaerosols were investigated. The results indicate that the number of bacteria showed remarkable increases during the dust events, and the diversity of the bacterial communities also increased significantly, as determined by means of microscopic observations with 4,6-diamidino-2-phenylindole (DAPI) staining and MiSeq sequencing analysis. These results indicate that dust clouds can carry many bacteria of various types into downwind regions and may have potentially important impacts on ecological environments and climate change. The abundances of DAPI-stained bacteria in the dust samples were 1 to 2 orders of magnitude greater than those in the non-dust samples and reached 105–106 particles m−3. Moreover, the concentration ratios of DAPI-stained bacteria to yellow fluorescent particles increased from 5.1 % ± 6.3 % (non-dust samples) to 9.8 % ± 6.3 % (dust samples). A beta diversity analysis of the bacterial communities demonstrated the distinct clustering of separate prokaryotic communities in the dust and non-dust samples. Actinobacteria, Bacteroidetes, and Proteobacteria remained the dominant phyla in all samples. As for Erenhot, the relative abundances of Acidobacteria and Chloroflexi had a remarkable rise in dust events. In contrast, the relative abundances of Acidobacteria and Chloroflexi in non-dust samples of R-DzToUb were greater than those in dust samples. Alphaproteobacteria made the major contribution to the increasing relative abundance of the phylum Proteobacteria in all dust samples. The relative abundance of Firmicutes did not exceed 5 % in all the air samples, even though it is the predominant phylum in the surface sand samples from the Gobi Desert. These results illustrate that the bacterial community contained in dust aerosol samples has a different pattern compared with non-dust aerosol samples, and the relative abundances of airborne bacteria are different from those in the surface sand or soil and differ by location and transmitting vector.
