Abstract:Amino acids derivative are well established molecular probes for diagnosis of a variety of cancer using positron emission tomography (PET). Recently, boramino acid (BAAs) was found as a prospective molecular platform for developing PET tracer. The objective of this study was to develop a 18F-labeled alanine derivative through displacing its carboxylate by trifluoroborate as a selective ASCT2 marker for cancer imaging. 18F-Ala-BF3 was first evaluated in healthy FVB/N mice in vivo, exhibiting rapid renal clearance with almost negligible uptake in stomach (1.53 ± 0.31%ID/g). Notable uptake was observed in thyroid (3.71 ± 0.49%ID/g, 40 min post injection), of which the uptake was significantly inhibited by co-injection with natural L-alanine. In addition, we further established 18F-Ala-BF3 on a human gastric cancer cell (BGC-823) xenografts bearing mouse model. Dynamic PET-CT scan revealed the optimal time window for tumor imaging, it was between 40 and 60 min post injection, when the BGC-823 xenografts uptake was 5.49 ± 1.47%ID/g ( n = 4), and the tumor-to-stomach, tumor-to-blood, tumor-to-muscle, and tumor-to-brain ratios were 3.27 ± 1.53, 3.80 ± 1.48, 3.47 ± 1.48, and 6.20 ± 1.47, respectively.
A Comparative Study of Radiolabeled Bombesin Analogs for the PET Imaging of Prostate Cancer.

Abstract:Radiolabeled bombesin (BBN) analogs that bind to the gastrin-releasing peptide receptor (GRPR) represent a topic of active investigation for the development of molecular probes for PET or SPECT of prostate cancer (PCa). RM1 and AMBA have been identified as the 2 most promising BBN peptides for GRPR-targeted cancer imaging and therapy. In this study, to develop a clinically translatable BBN-based PET probe, we synthesized and evaluated 18F-AlF- (aluminum-fluoride) and 64Cu-radiolabeled RM1 and AMBA analogs for their potential application in PET imaging of PCa. Methods: 1,4,7-triazacyclononane, 1-glutaric acid-4,7 acetic acid (NODAGA)–conjugated RM1 and AMBA were synthesized and tested for their GRPR-binding affinities. The NODAGA-RM1 and NODAGA-AMBA probes were further radiolabeled with 64Cu or 18F-AlF and then evaluated in a subcutaneous PCa xenograft model (PC3) by small-animal PET imaging and biodistribution studies. Results: NODAGA-RM1 and NODAGA-AMBA can be successfully synthesized and radiolabeled with 64Cu and 18F-AlF. 64Cu- and 18F-AlF-labeled NODAGA-RM1 demonstrated excellent serum stability and tumor-imaging properties in the in vitro stability assays and in vivo imaging studies. 64Cu-NODAGA-RM1 exhibited tumor uptake values of 3.3 ± 0.38, 3.0 ± 0.76, and 3.5 ± 1.0 percentage injected dose per gram of tissue (%ID/g) at 0.5, 1.5, and 4 h after injection, respectively. 18F-AlF-NODAGA-RM1 exhibited tumor uptake values of 4.6 ± 1.5, 4.0 ± 0.87, and 3.9 ± 0.48 %ID/g at 0.5, 1, and 2 h, respectively. Conclusion: The high-stability, efficient tumor uptake and optimal pharmacokinetic properties highlight 18F-AlF-NODAGA-RM1 as a probe with great potential and clinical application for the PET imaging of prostate cancer.
A bis-boron boramino acid PET tracer for brain tumor diagnosis

Abstract:Purpose: Boramino acids are a class of amino acid biomimics that replace the carboxylate group with trifluoroborate and can achieve the 18F-labeled positron emission tomography (PET) and boron neutron capture therapy (BNCT) with identical chemical structure. Methods: This study reports a trifluoroborate-derived boronophenylalanine (BBPA), a derived boronophenylalanine (BPA) for BNCT, as a promising PET tracer for tumor imaging. Results: Competition inhibition assays in cancer cells suggested the cell accumulation of [18F]BBPA is through large neutral amino acid transporter type-1 (LAT-1). Of note, [18F]BBPA is a pan-cancer probe that shows notable tumor uptake in B16-F10 tumor-bearing mice. In the patients with gliomas and metastatic brain tumors, [18F]BBPA-PET shows good tumor uptake and notable tumor-to-normal brain ratio (T/N ratio, 18.7 ± 5.5, n = 11), higher than common amino acid PET tracers. The [18F]BBPA-PET quantitative parameters exhibited no difference in diverse contrast-enhanced status (P = 0.115-0.687) suggesting the [18F]BBPA uptake was independent from MRI contrast-enhancement. Conclusion: This study outlines a clinical trial with [18F]BBPA to achieve higher tumor-specific accumulation for PET, provides a potential technique for brain tumor diagnosis, and might facilitate the BNCT of brain tumors.

Abstract:[18F]Gln-OSO2F, [18F]Arg-OSO2F, and [18F]FSY-OSO2F were designed by introducing sulfonyl 18F-fluoride onto glutamine, arginine, and tyrosine, respectively. [18F]FSY-OSO2F can be prepared directly by sulfur 18F-fluoride exchange, while [18F]Gln-OSO2F and [18F]Arg-OSO2F require a two-step labeling method. Those tracers retain their typical transport characteristics for unmodified amino acids. Both PET imaging and biodistribution confirmed that [18F]FSY-OSO2F visualized MCF-7 and 22Rv1 subcutaneous tumors with high contrast, and its tumor-to-muscle ratio was better than that of [18F]FET. However, [18F]Gln-OSO2F and [18F]Arg-OSO2F poorly image MCF-7 subcutaneous tumors, possibly due to differences in the types and amounts of transporters expressed in tumors. All three tracers can visualize the U87MG glioma. According to our biological evaluation, none of the tracers evaluated in this study exhibited obvious defluorination, and subtle structural changes led to different imaging characteristics, indicating that the application of sulfur 18F-fluoride exchange click chemistry in the design of radioactive sulfonyl fluoride amino acids is feasible and offers significant advantages.

Abstract:Three tertiary benzenesulfonamide inhibitors 4a–c were radiolabeled with 18F and evaluated for imaging carbonic anhydrase IX (CA IX) expression with positron emission tomography. All three inhibitors exhibit <10nM affinity for CA IX with no measurable affinity for CA II. Despite good affinity/selectivity to CA IX and excellent stability in plasma, uptake of [18F]4a–c in CA IX-expressing HT-29 tumours was low without significant contrast. [18F]4a,b were excreted rapidly, while [18F]4c exhibited significant in vivo defluorination leading to high bone uptake. Due to minimal uptake in HT-29 tumours compared to normal organs/tissues, 18F-labeled benzenesulfonamides [18F]4a–c are not suitable as CA IX imaging agents.
Preclinical Evaluation of a High-Affinity 18F-Trifluoroborate Octreotate Derivative for Somatostatin Receptor Imaging

Abstract:Recent studies have highlighted the high sensitivity of PET imaging with 68Ga-labeled octreotide derivatives for the detection and staging of neuroendocrine tumors. A somatostatin receptor ligand that is easily radiolabeled with 18F-fluoride could improve the availability of PET imaging of neuroendocrine tumors. We report an alkyltrifluoroborate–octreotate conjugate that is radiolabeled in a 1-step 18F exchange reaction in high yield and with high specific activity. Methods: We conjugated a new alkyltrifluoroborate to octreotate to obtain AMBF3-TATE, which was stored in 50-nmol aliquots for radiolabeling. 18F labeling was performed by 18F-19F isotope exchange with 18F-fluoride, and the tracer was purified by C18 cartridge separation. The radiochemical yield was 20%–25%. PET imaging and biodistribution were performed on mice bearing AR42J tumor xenografts. Results: AMBF3-TATE bound the somatostatin receptor subtype 2 with high affinity (inhibition constant, 0.13 ± 0.03 nM). Starting with 29.6–37 GBq (0.8–1 Ci) of 18F-fluoride, more than 7.4 GBq (>200 mCi) of 18F-AMBF3-TATE were obtained in 25 min (n = 5) with greater than 99% radiochemical purity at high specific activity (>111 GBq [3 Ci]/μmol). 18F-AMBF3-TATE was stable in plasma. PET imaging and biodistribution showed rapid renal excretion with low liver activity. High tumor uptake (10.11% ± 1.67% injected dose/g, n = 5) was detected at 60 min after injection. Bone uptake was negligible. Tumor-to-liver, tumor-to-blood, tumor-to-muscle, and tumor-to-bone ratios (at 60 min) were 26.2 ± 0.8, 25.1 ± 1.0, 89.0 ± 3.1, and 21.3 ± 3.6, respectively. Conclusion: 18F-AMBF3-TATE was radiolabeled in high yield and at high specific activity, did not require high-performance liquid chromatography purification, exhibited unexpectedly high binding affinity to somatostatin receptor subtype 2, and showed excellent pharmacokinetic properties in vivo, with high tumor uptake and high contrast ratios.

Abstract:Trifluoroborate boronophenylalanine (BBPA) is a boron amino acid analog of 4‐boronophenylalanine (BPA) but with a trifluoroborate group (-BF3-) instead of a carboxyl group (-COOH). Clinical studies have shown that 18F-labeled BBPA ([18F]BBPA) can produce high-contrast tumor images in positron emission tomography (PET). Beyond PET imaging, BBPA is a theranostic agent for boron neutron capture therapy (BNCT). Because BBPA possesses an identical chemical structure to BNCT and PET, it can potentially predict the boron concentration for BNCT using [18F]BBPA-PET. The synthesis of BBPA was achieved by selectively fluorinating the α-aminoborate compound, taking advantage of the varying rates of solvolysis of the B-F bond. The study showcased the high-contrast [18F]BBPA-PET imaging in various tumor models, highlighting its broad applicability for both [18F]BBPA-PET and BBPA-BNCT. [18F]BBPA-PET tumor uptake remains consistent across various doses, including those used in BNCT. This enables accurate estimation of the boron concentration in tumors using [18F]BBPA-PET. With its dual boron structure, BBPA increases boron concentration in tumor cells and tumor tissues compared to BPA. Thus, less boron carrier is needed. This study introduces a new theranostic boron carrier that enhances boron accumulation in tumors, predicts boron concentration, and enhances the accuracy and effectiveness of BNCT.

Trimeric Radiofluorinated Sulfonamide Derivatives to Achieve in Vivo Selectivity for Carbonic Anhydrase IX-Targeted PET Imaging.

Abstract:Carbonic anhydrase IX (CA-IX), a transmembrane enzyme, mediates cell survival under hypoxic conditions and is overexpressed in solid malignancies. In this study, we synthesized four F-18 sulfonamide derivatives and evaluated their potential for imaging CA-IX expression with PET. Methods: Azide derivatives of 2 carbonic anhydrase inhibitors, 4-(2-aminoethyl)benzenesulfonamide (AEBS) and 4-aminobenzensulfonamide (ABS), were coupled to radiosynthons with either 1 or 3 alkynes and a pendent ammoniomethyltrifluoroborate (AmBF3) to generate monovalent or trivalent enzyme inhibitors. Binding affinity to CA-IX and other CA isoforms was determined via a stopped-flow, CA-catalyzed CO2 hydration assay. Tracers were radiolabeled via F-18-F-19 isotope exchange reactions. Imaging/biodistribution studies were performed using HT-29 tumor-bearing immunocompromised mice. Results: Monomeric AmBF3-AEBS and AmBF3-ABS were obtained in 41% and 40% yields, whereas trimeric AmBF3-(AEBS)(3) and AmBF3-(ABS)(3) were obtained in 47% and 55% yields, respectively. Derivatives bound CA-I, -II, -IX, and -XII with good affinity (0.49-100.3 nM). F-18-labeled sulfonamides were obtained in 16.3%-36.8% non-decay-corrected radiochemical yields, with 40-207 GBq/mmol specific activity and greater than 95% radiochemical purity. Biodistribution/imaging studies showed that the tracers were excreted through both renal and hepatobiliary pathways. At 1 h after injection, HT-29 tumor xenografts were clearly visualized in PET images with modest contrast for all 4 tracers. Tumor uptake was 2-fold higher for monovalent tracers (similar to 0.60 percentage injected dose per gram [%ID/g]) than for trivalent tracers (similar to 0.30 %ID/g); however, tumor-to-background ratios were significantly better for F-18-AmB(F)3-(ABS)(3). Preblocking with acetazolamide reduced more than 80% uptake of F-18-AmBF3(ABS)(3) in HT-29 tumors. Conclusion: Our data suggest that trimerization of an otherwise nonspecific CA inhibitor greatly enhances the selectivity for CA-IX in vivo and represents a promising strategy for creating multivalent enzyme inhibitors for selectively imaging extracellular enzyme activity by PET.

Abstract:The expression of prostate-specific membrane antigen (PSMA) in prostate cancer is 100–1000 times higher than that in normal tissues, and it has shown great advantages in the diagnosis and treatment of prostate cancer. The combination of PSMA and PET imaging technology based on the principle of metabolic imaging can achieve high sensitivity and high specificity for diagnosis. Due to its suitable half-life (109 min) and good positron abundance (97 %), as well as its cyclotron accelerated generation, 18 F has the potential to be commercialize, which has attracted much attention. In this article, we synthesized a series of fluorosulfate PET tracers targeting PSMA. All four analogues have shown high affinity to PSMA (IC 50 = 1.85–5.15 nM). After the radioisotope exchange labeling, [ 18 F]L9 and [ 18 F]L10 have PSMA specific cellular uptake (0.65 ± 0.04 % AD and 1.19 ± 0.03 % AD) and effectively accumulated in 22Rv1 xenograft mice model. This study demonstrates that PSMA-1007-based PSMA-targeted aryl [ 18 F]fluorosulfate novel tracers have the potential for PET imaging in tumor tissues.

Synthesis and Evaluation of an 18F-Labeled Bradykinin B1 Receptor-Targeting Small Molecule for PET Imaging

Abstract:1169 Objectives The bradykinin B1 receptor (B1R), a G-protein coupled receptor, is overexpressed in several pathophysiological conditions including inflammation and cancer. B1R mediates nociception in the context of inflammation, and it is believed to contribute to tumourigenesis by promoting cell proliferation, invasion and angiogenesis. Previously, our lab reported the evaluation of several promising B1R-targeting peptides for cancer imaging. These peptides have high affinity to B1R and generate high-contrasted images of B1R-expressing tumour xenografts in mice. A significant number of potent B1R small molecule antagonists have been developed by pharmaceutical companies for the treatment of chronic pain, and could potentially be exploited for the design of B1R-targeting imaging agents. In this study, we investigated the use of an 18F-labeled small molecule (4-methoxy-N,2,6-trimethyl-N-[2-[2-[4-(1-[18F]fluoroethyl-4-piperidinyl)-1-piperazinyl]-2-oxoethoxy]ethyl]benzenesulfonamide, 18F-Z02035) as an alternative to peptide-based B1R-imaging. Methods The non-radioactive Z02035 standard and its des-fluoroethyl precursor (4-methoxy-N,2,6-trimethyl-N-[2-[2-[4-(1-H-4-piperidinyl)-1-piperazinyl]-2-oxoethoxy]ethyl]benzenesulfonamide, Z02153) were synthesized via multi-step synthesis approach. The binding affinity of Z02035 for B1R was determined by in vitro competition binding assay using B1R-expressing cell membrane. 18F-Z02035 was prepared in two steps: (1) synthesis of 2-[18F]fluoroethyl tosylate by aliphatic nucleophilic substitution using 18F-fluoride and 1,2-bis(tosyloxy)ethane; and (2) coupling of 2-[18F]fluoroethyl tosylate with the precursor Z02153. PET imaging and biodistribution studies were performed at 1 h post-injection in mice bearing both B1R-positive HEK293T::hB1R and B1R-negative wild-type HEK293T tumours. Results The overall synthesis yield for the non-radioactive standard Z02035 was 1% (seven steps), and the yield for the precursor Z02153 was 47% (seven steps). Competition assays showed that Z02035 is a potent B1R binder with a Ki value of 0.93 ± 0.44 nM. By reacting Z02153 with 2-[18F]fluoroethyl tosylate, 18F-Z02035 was obtained in 6 ± 1% non-decay corrected yield with > 99% radiochemical purity and 0.9 ± 0.8 Ci/µmol specific activity. From PET images, 18F-Z02035 enabled visualization of the B1R-positive HEK293T::hB1R tumours with better contrast than the B1R-negative HEK293T tumours. In terms of clearance, 18F-Z02035 was excreted primarily through the hepatobiliary pathway. The average uptake in HEK293T::hB1R tumours (3.77 ± 0.79 %ID/g) was higher than in HEK293T tumours (2.11 ± 0.37 %ID/g). The tumour-to-muscle ratio for HEK293T::hB1R was 3.38 ±1.05. 18F-Z02035 exhibited slight defluorination in vivo as bone uptake was 4.63 ± 0.65 %ID/g. Conclusions We took a novel approach for targeting B1R expression by synthesizing an 18F-labeled small molecule. Although absolute uptake and overall contrast are lower than reported B1R tracers derived from peptide sequences, 18F-Z02035 was able to differentiate between B1R expressing and non-expressing tumour xenografts. Optimization to enhance tumour uptake/contrast and reduce in vivo defluorination is currently underway.

Abstract:Radiolabeled urea-based low-molecular weight inhibitors of the prostate-specific membrane antigen (PSMA) are under intense investigation as imaging and therapeutic agents for prostate and other cancers. In an effort to provide agents with less nontarget organ uptake than the ureas, we synthesized four (18)F-labeled inhibitors of PSMA based on carbamate scaffolds. 4-Bromo-2-[(18)F]fluorobenzoyllysineoxypentanedioic acid (OPA) carbamate [(18)F]23 and 4-iodo-2-[(18)F]fluorobenzoyllysine OPA carbamate [(18)F]24 in particular exhibited high target-selective uptake in PSMA+ PC3 PIP tumor xenografts, with tumor-to-kidney ratios of >1 by 4 h postinjection, an important benchmark. Because of its high tumor uptake (90% injected dose per gram of tissue at 2 h postinjection) and high tumor-to-organ ratios, [(18)F]23 is promising for clinical translation. Prolonged tumor-specific uptake demonstrated by [(18)F]24, which did not reach equilibrium during the 4 h study period, suggests carbamates as alternative scaffolds for mitigating dose to nontarget tissues.
(18)F-Ambf3-mj9: a Novel Radiofluorinated Bombesin Derivative for Prostate Cancer Imaging.

Abstract:A novel radiofluorinated derivative of bombesin, (18)F-AmBF3-MJ9, was synthesized and evaluated for its potential to image prostate cancer by targeting the gastrin releasing peptide receptor (GRPR). AmBF3-MJ9 was prepared from an ammoniomethyl-trifluoroborate (AmBF3) conjugated alkyne 2 and azidoacetyl-MJ9 via a copper-catalyzed click reaction, and had good binding affinity for GRPR (Ki=0.5±0.1nM). The (18)F-labeling was performed via a facile one-step (18)F-(19)F isotope exchange reaction, and (18)F-AmBF3-MJ9 was obtained in 23±5% (n=3) radiochemical yield in 25min with >99% radiochemical purity and 100±32GBq/μmol specific activity. (18)F-AmBF3-MJ9 was stable in mouse plasma, and was partially (22-30%) internalized after binding to GRPR. Positron emission tomography (PET) imaging and biodistribution studies in mice showed fast renal excretion and good uptake of (18)F-AmBF3-MJ9 by GRPR-expressing pancreas and PC-3 prostate cancer xenografts. Tumor uptake was 1.37±0.25%ID/g at 1h, and 2.20±0.13%ID/g at 2h post-injection (p.i.) with low background uptake and excellent tumor visualization (tumor-to-muscle ratios of 75.4±5.5). These data suggest that (18)F-AmBF3-MJ9 is a promising PET tracer for imaging GRPR-expressing prostate cancers.

Abstract:The expression of prostate-specific membrane antigen (PSMA) in prostate cancer is 100-1000 times higher than that in normal tissues, and it has shown great advantages in the diagnosis and treatment of prostate cancer. The combination of PSMA and PET imaging technology based on the principle of metabolic imaging can achieve high sensitivity and high specificity for diagnosis. Due to its suitable half-life (109 min) and good positron abundance (97%), as well as its cyclotron accelerated generation, 18F has the potential to be commercialize, which has attracted much attention. In this article, we synthesized a series of fluorosulfate PET tracers targeting PSMA. All four analogues have shown high affinity to PSMA (IC50 = 1.85-5.15 nM). After the radioisotope exchange labeling, [18F]L9 and [18F]L10 have PSMA specific cellular uptake (0.65 +/- 0.04% AD and 1.19 +/- 0.03% AD) and effectively accumulated in 22Rv1 xenograft mice model. This study demonstrates that PSMA-1007-based PSMAtargeted aryl [18F]fluorosulfate novel tracers have the potential for PET imaging in tumor tissues.
Synthesis and Evaluation of F-18-Ambf3-Psma Derivatives As Pet Imaging Agents for Prostate Cancer

Abstract:1155 Objectives Prostate specific membrane antigen (PSMA) is a membrane bound glycoprotein that is overexpressed on the cell surface of prostate cancer cells. A few PSMA-targeting radiotracers have been successfully introduced in clinical studies for imaging prostate cancer by PET. 18F-DCFPyL is currently the most sensitive tracer for the identification of PSMA-positive prostate cancer. However, the procedure for producing 18F-DCFPyL involves multiple synthesis steps. In this study, two 18F-labeled PSMA-targeting tracers (HTK-01069 and HTK-01070) that could be radiolabeled via 18F-19F isotope exchange on the ammoniomethyl-trifluoroborate (AmBF3) motif were synthesized and evaluated for PSMA imaging. Methods HTK-01069 was synthesized by coupling succinimido 4-(N-trifluoroborylmethyl-N,N-dimethylammonio)methylbenzoate to Lys-C(O)-Glu urea. HTK-01070 was prepared by coupling tetrafluorophenyl 4-(azidemethyl)benzoate to t-butyl protected Lys-C(O)-Glu urea, followed by deprotection and click reaction with N-propargyl-N,N-dimethyl-ammoniomethyltrifluoroborate. 18F-19F isotope exchange reactions were conducted for radiolabeling and both tracers were purified by HPLC. The stability of the tracers was assessed in mouse plasma for 2 h and LogD7.4 values were determined by the shake-flask method. PET imaging and biodistribution studies were performed in mice bearing PSMA-expressing LNCap prostate cancer xenografts. For blocking studies, mice were co-injected with 0.5 mg of DCFPyL. Results 18F-HTK-01069 and 18F-HTK-01070 were obtained in 9% and 3% decay-corrected radiochemical yields with > 99% radiochemical purity. The specific activities for 18F-HTK-01069 and 18F-HTK-01070 were 2.0 and 1.3 Ci/μmol, respectively. Both tracers were stable after 2 h incubation in plasma and were highly hydrophilic with LogD7.4 values between -3.4 and -4.3. From the PET and biodistribution data, 18F-HTK-01069 and 18F-HTK-01070 demonstrated clear PSMA-dependent uptake in LNCap prostate cancer xenografts at 1 h post-injection (p.i.). The clearance of both 18F-HTK-01069 and 18F-HTK-01070 was predominantly renal with minor involvement of the hepatobiliary pathway. At 1 h p.i., 18F-HTK-01070 exhibited higher tumour uptake (8.28 ± 1.25 %ID/g) than 18F-HTK-01069 (5.82 ± 1.09 %ID/g). 18F-HTK-01070 was also superior to 18F-HTK-01069 in terms of tumour-to-muscle ratio (37.7 ± 9.53 vs. 18.9 ± 6.05) and tumour-to-blood ratio (16.0 ± 1.37 vs. 11.9 ± 4.60). With PSMA blockade, uptake in tumours decreased significantly to 0.33 ± 0.07 and 0.27 ± 0.06 %ID/g for 18F-HTK-01069 and 18F-HTK-01070, respectively. Conclusions Two novel imaging agents, 18F-HTK-01069 and 18F-HTK-01070, were synthesized and evaluated for targeting PSMA expression with PET. Administration of 18F-HTK-01069 or 18F-HTK-01070 allowed for clear visualization of LNCap tumours in mice, with 18F-HTK-01070 yielding higher uptake and contrast. The data suggests that 18F-AmBF3-PSMA derivatives are promising agents for detecting PSMA-expressing prostate cancers.

Abstract:Radiolabeled acetates have potential use in tumor imaging. Moreover, aryl fatty acid displays antitumor effect with phenylacetate currently in clinical trial. A series of 2-R 1 -2-(4-(2-fluoroethoxy)benzamido)acetate were synthesized in the present investigation. 2-(4-(2-[ 18 F]fluoroethoxy)benzamido)acetate ([ 18 F]FEBA) was evaluated in tumor-bearing mice. It was compared with 2-[ 18 F]fluoro-2-deoxy- d -glucose ([ 18 F]FDG), showing satisfied stability. Excitingly, the radiochemical purity of [ 18 F]FEBA was more than 99% and the radiochemical yield was up to 25%. Although 2-R 1 -2-(4-(2-fluoroethoxy)benzamido)acetate deserved further evaluations, the results might provide many experimental foundations and principles for the future study of the novel fluorine-18-labeled pharmaceuticals.
Synthesis and Biological Evaluation of a Radiolabeled PET (Positron Emission Tomography) Probe for Visualization of In Vivo α-Fucosidase Expression

Abstract:The acidic hydrolase α-fucosidase (AF) is a biomarker for maladies such as cancer and inflammation. The most advanced probes for α-fucosidase are unfortunately constrained to ex vivo or in vitro applications. The in vivo detection and quantification of AF using positron emission tomography would allow for better discovery and diagnosis of disease as well as provide better understanding of disease progression. We synthesized, characterized, and evaluated a radiolabeled small molecule inhibitor of AF based on a known molecule. The radiosynthesis involved the 11C methylation of a phenoxide, which was generated in situ by ultrasonification of the precursor with sodium hydride. The tracer was produced with a decay corrected yield of 41.7 ± 16.5% and had a molar activity of 65.4 ± 30.3 GBq/μmol. The tracer was shown to be stable in mouse serum at 60 min. To test the new tracer, HCT116 colorectal carcinoma cells were engineered to overexpress human AF. In vitro evaluation revealed 3.5-fold higher uptake in HCT116AF cells compared to HCT116 controls (26.4 ± 7.8 vs. 7.5 ± 1.0 kBq/106 cells). Static PET scans 50 min post injection revealed 2.5-fold higher tracer uptake in the HCT116AF tumors (3.0 ± 0.8%ID/cc (n = 6)) compared with the controls (1.2 ± 0.8 (n = 5)). Dynamic scans showed higher uptake in the HCT116AF tumors at all time-points (n = 2). Ex vivo analysis of the tumors, utilizing fluorescent DDK2 antibodies, confirmed the expression of human AF in the HCT116AF xenografts. We have developed a novel PET tracer to image AF in vivo and will now apply this to relevant disease models.

Abstract:A novel synthesis of 4-[(18)F]fluorobenzylamine ([(18)F]FBA) by means of transition metal-assisted sodium borohydride reduction of 4-[(18)F]fluorobenzonitrile ([(18)F]FBN) is described. This approach could successfully be extended to borohydride exchange resin (BER) enabling a viable option for use in automated syntheses. [(18)F]FBA was used for the synthesis of 4-[(18)F]fluorobenzylamine-based thiol group-reactive prosthetic groups 4-[(18)F]fluorobenzyl-2-bromoacetamide ([(18)F]FBBA) and 4-[(18)F]fluorobenzylamidopropionyl maleimide ([(18)F]FBAPM). [(18)F]FBBA and [(18)F]FBAPM were obtained in radiochemical yields of 75% and 55%, respectively. Feasibility of using [(18)F]FBAPM as novel prosthetic group for peptide and protein labelling was demonstrated with cysteine-containing tripeptide glutathione (GSH). [(18)F]FBBA was used for labelling of a fully phosphorothioated 20mer oligodesoxynucleotide (ODN).

Abstract:INTRODUCTION:The emerging evidence that demonstrated the extra-demand of glutamine for cancer surviving strongly called for the development of PET tracer for imaging glutamine uptake in cancer. In this work, [18F]Gln-BF3 as a natural glutamine derivative was synthesized to explore its potential application of imaging glutamine uptake for cancer diagnosis. METHODS:[18F]Gln-BF3 was prepared by deprotection of purified precursor trityl-Gln-BF3 (amide bond protected with triphenylmethyl group) using TFA and radiolabeled using 18F-19F isotope exchange protocol. PET imaging and biodistribution studies were conducted in Balb/c mice bearing 4T1 xenograft. RESULTS:Gln-BF3 was identified with HRMS ([M-H]- = 169.0765). [18F]Gln-BF3 was radiolabeled in high radiochemical yield (RCY > 25%) and characterized with Radio-HPLC-MS. Preliminary PET imaging showed the radioactivity was fast cleared from muscle tissue and excreted mainly via the renal pathway. PET study demonstrated that uptake of [18F]Gln-BF3 in 4T1 xenografts was significant. The biodistribution results of [18F]Gln-BF3 in mice bearing 4T1 xenograft indicate a tumor-to-muscle ratio of 2.58 ± 0.64 (n = 4) and a 6.29 ± 0.42%ID/g (n = 4) uptake in tumor at 45 min post injection. CONCLUSION:[18F]Gln-BF3 was radiolabeled in a "kit-like" manner and showed notable and tumor-selective uptake in tumor-bearing animal models, suggesting that this new strategy of radiolabeling amino acid provided a promising solution for the future development of glutamine-derived PET tracers.

Abstract:Four trifluoroborate derivatives of phosphonium cations 2a–d were radiolabeled with fluorine-18 (18F) and evaluated for imaging myocardial perfusion with positron emission tomography (PET). Tracers were radiolabeled simply via 18F–19F isotope exchange reaction in acidic (pH 2) aqueous solution. On average, [18F]2a–d were obtained in 10–17% non-decay-corrected radiochemical yield with 25.9–48.1GBq/μmol specific activity, and >96% radiochemical purity. In vitro stability study showed no decomposition of [18F]2a–d after being incubated in mouse plasma for up to 2h. Myocardial uptake in mice was visualized in PET images by using [18F]2b–d but not [18F]2a. [18F]2a–d were stable against in vivo defluorination as no significant bone uptake was observed. Despite sub-optimal heart uptake of [18F]2b–d, we successfully demonstrated that 18F–19F isotope exchange reaction on trifluoroborates could be a promising strategy for the design of potential 18F-labeled tracers even for intracellular targets.

Abstract:Positron Emission Tomography (PET) is a rapidly expanding, cutting edge technology for preclinical evaluation, cancer diagnosis and staging, and patient management. A one-step aqueous (18)F-labeling method, which can be applied to peptides to provide functional in vivo images, has been a long-standing challenge in PET imaging. Over the past few years, we have sought a rapid and mild radiolabeling method based on the aqueous radiosynthesis of in vivo stable aryltrifluoroborate (ArBF(3)(-)) conjugates. Recent access to production levels of (18)F-Fluoride led to a fluorescent-(18)F-ArBF(3)(-) at unprecedentedly high specific activities of 15Ci/μmol. However, extending this method to labeling peptides as imaging agents has not been explored.In order to extend these results to a peptide of clinical interest in the context of production-level radiosynthesis, we applied this new technology for labeling RGD, measured its specific activity by standard curve analysis, and carried out a preliminary evaluation of its imaging properties.RGD was labeled in excellent radiochemical yields at exceptionally high specific activity (~14Ci/μmol) (n = 3). Preliminary tumor-specific images corroborated by ex vivo biodistribution data with blocking controls show statistically significant albeit relatively low tumor uptake along with reasonably high tumor:blood ratios (n = 3).Isotope exchange on a clinically useful (18)F-ArBF(3)(-) radiotracer leads to excellent radiochemical yields and exceptionally high specific activities while the anionic nature of the aryltrifluoroborate prosthetic results in very rapid clearance. Since rapid clearance of the radioactive tracer is generally desirable for tracer development, these results suggest new directions for varying linker arm composition to slightly retard clearance rather than enhancing it.This work is the first to use production levels of (18)F-activity to directly label RGD at specific activities that are an order of magnitude higher than most reports and thereby increases the distribution window for radiotracer production and delivery.
