Abstract:Methods toward the dimerization and homocoupling of terminal alkynes have been established that employ Co complexes along with tBuOK under different atmosphere. A variety of Z-1,3-enynes were furnished with a high Z:E (up to 93:7) ratio in moderate to very good yields under inert atmosphere. Alternatively, the Glaser homocoupling reaction of terminal alkynes was also accomplished with a Co catalyst, successfully affording various 1,3-diynes in excellent yields (up to 93%) under oxygen.

Abstract:Transition metal-catalyzed alkyne dimerization represents a powerful method for the construction of enynes. However, the ambiguous hydrogen transfer mechanism during the dimerization has resulted in controlling the regio-, stereo-, and, where applicable, chemoselectivity remaining a long-standing challenge. Herein, a combination of DFT calculations and quasi-classical MD simulations was used to interrogate the dynamic motion of hydrogen in cobalt-catalyzed alkyne dimerization. The collective results inspired us to propose, for the first time, a substrate-dependent differential hydride transfer model involving either concerted oxidative hydrogen transfer or stepwise oxidative addition, followed by alkyne insertion. The practicability and universality of this oxidative hydride transfer mechanism were further validated by the theoretical studies of experimentally observed selective cross- and homo-dimerization. Charge distribution analyses depicted that the differentiation between those two hydride transfer mechanisms originates from the alpha-silicon effect, which can stabilize the neighboring negative charge of the alkyne. Furthermore, a comprehensive DFT study of the substituent effects of alkynes reveals that the electron-withdrawing group will accelerate the oxidative hydride transfer process, which can open up avenues for mechanistic-oriented selective dimerization.

Abstract:C-H activation and functionalization are on the forefront of modern synthetic chemistry. Imagine if any C-H bond of a molecule could be converted to a C-X bond, where X is the target functionality-this would alter the synthetic blueprints for complex target molecules since it would provide novel disconnections in retrosynthetic analysis. Collaborations between many experimental and computational groups have led to rapid developments of new C-H functionalization methods. Our groups represent an example of this; we were brought together as part of the NSF-supported Center for Selective C-H Functionalization. Many examples of experimental-computational synergy for selective Pd(II)-catalyzed C-H activation of aryl and alkyl groups are described in this Account. We describe computations by the Houk group made in response to experimental stimuli by the Yu group. The first section discusses the experimental and computational investigations of oxazoline-directed stereoselective Pd(II)-catalyzed C(sp3)-H bond activation that occurs through the concerted metalation-deprotonation (CMD) pathway involving a monomeric Pd(II) complex. The second section involves two types of bidentate ligands, mono-N-protected amino acid (MPAA) and acetyl-protected aminoethyl quinoline (APAQ) ligands that promote the C-H activation reactions with the ligand as the internal base. In the MPAA-assisted remote C-H bond activation, the basic dianionic amidate ligand participates in the deprotonation of a specific C-H bond. This mechanism accounts for the improved reactivity and selectivity in C-H activation reactions with MPAA ligands. The chiral APAQ ligands enable asymmetric palladium insertion into prochiral C-H bonds on a single methylene carbon center. The dianionic amidate of the APAQ ligand acts as an intramolecular base to deprotonate the methylene C-H asymmetrically and facilitate chiral Pd-C bond formation. The origins of the dramatic differences of five-membered (relatively inactive) and six-membered (highly active) chelation in β-methylene C(sp3)-H activation reactions by a Pd(II) catalyst were explained with density functional theory (DFT) calculations. This is mainly due to the steric repulsions between the ArF group of the substrate and the quinoline group of the ligand. The steric repulsion between the ArF group of the substrate and the quinoline group of the APAQ ligand destabilizes the five-membered chelate transition structure, increasing the energy of the transition state. The third section discusses a mechanism involving a Pd-Ag heterodimeric complex intermediate in the template-directed, Pd(II)-catalyzed remote meta functionalization of toluene derivatives and benzoic acid derivatives. The nitrile directing group of the template coordinates with Ag while the Pd is placed adjacent to the meta-C-H bond in the transition state, leading to the observed high meta selectivity. The selective activation of remote meta-C-H bonds at various distances can be achieved by tuning the template. The dual role of AgOAc as both an oxidant and part of the heteronuclear active species in the mechanism involving PdAg(OAc)3 was determined by DFT calculations and is in accord with literature information about complexes. For the systems discussed in these three sections, the similarity is that they all proceed via the CMD mechanism. The differences lie in the proton acceptors and the active Pd species. Common CMD involves a monomeric Pd mechanism with acetate as the proton acceptor. Both MPAA and APAQ ligands react via monomeric Pd mechanisms with a ligand moiety (the amidate oxygen) as the proton acceptor. Nitrile-containing template-mediated meta-C-H activations proceed via a Pd-Ag heterodimeric mechanism, still with acetate as the proton acceptor. The interaction between our two groups, experts in experiment and computation, and the discoveries made possible by that interplay are highlighted in this Account.
Mechanism, Reactivity, and Selectivity of Nickel-Catalyzed [4 + 4 + 2] Cycloadditions of Dienes and Alkynes

Abstract:Density functional theory (DFT) calculations with B3LYP and M06 functionals elucidated the reactivities of alkynes and Z/E selectivity of cyclodecatriene products in the Ni-catalyzed [4 + 4 + 2] cycloadditions of dienes and alkynes. The Ni-mediated oxidative cyclization of butadienes determines the Z/E selectivity. Only the oxidative cyclization of one s-cis to one s-trans butadiene is facile and exergonic, leading to the observed 1Z,4Z,8E-cyclodecatriene product. The same step with two s-cis or s-trans butadienes is either kinetically or thermodynamically unfavorable, and the 1Z,4E,8E- and 1Z,4Z,8Z-cyclodecatriene isomers are not observed in experiments. In addition, the competition between the desired cooligomerization and [2 + 2 + 2] cycloadditions of alkynes depends on the coordination of alkynes. With either electron-deficient alkynes or alkynes with free hydroxyl groups, the coordination of alkynes is stronger than that of dienes, and alkyne trimerization prevails. With alkyl-substituted alkynes, the generation of alkyne-coordinated nickel complex is much less favorable, and the [4 + 4 + 2] cycloaddition occurs.

Abstract:FeCl2 ligated with 2-(benzimidazolyl)-6-(1-(arylimino)ethyl)pyridine in the presence of zinc powder and zinc iodide could effectively catalyze cyclotrimerization of intermolecular alkynes to afford benzene derivatives in high regioselectivity. The synthetic usefulness of this catalytic system consisting of Fe(II) complex C1, Zn and ZnI2 in acetonitrile was tested with various terminal alkynes. In the cases of using aryl substituted alkynes, the 1,2,4-trisubstituted benzenes 2 were found to be the major products.

Abstract:The asymmetric allylic alkylation (AAA), which features employing active allylic substrates, has historical significance in organic synthesis. The allylic C-H alkylation is principally more atom- and step-economic than the classical allylic functionalizations and thus can be considered a transformative variant. However, asymmetric allylic C-H alkylation reactions are still scarce and yet underdeveloped. Herein, we have found that Z/ E- and regioselectivities in the Pd-catalyzed asymmetric allylic C-H alkylation of 1,4-dienes are highly dependent on the type of nucleophiles. A highly stereoselective allylic C-H alkylation of 1,4-dienes with azlactones has been established by palladium-chiral phosphoramidite catalysis. The protocol proceeds under mild conditions and can accommodate a wide scope of substrates, delivering structurally divergent α,α-disubstituted α-amino acid surrogates in high yields and excellent levels of diastereo-, Z/ E-, regio-, and enantioselectivities. Notably, this method provides key chiral intermediates for an efficient synthesis of lepadiformine marine alkaloids. Experimental and computational studies on the reaction mechanism suggest a novel concerted proton and two-electron transfer process for the allylic C-H cleavage and reveal that the Z/ E- and regioselectivities are governed by the geometry and coordination pattern of nucleophiles.

Abstract:Here, we reported for the first time a mechanistically distinctive cobalt-catalyzed Markovnikov-type sequential semihydrogenation/hydrohydrazidation of aliphatic terminal alkynes in one pot. A cobalt hydride species was employed as two roles for both a unique metal-catalyzed Markovnikov-type insertion of the aliphatic terminal alkynes and then metal-catalyzed hydrogen atom transfer of alkenes. This operationally simple protocol exhibits excellent functional group tolerance and step economy. The hydrazone products could be easily transferred to various valuable amine derivatives.
Z-selective alkene isomerization by high-spin cobalt(II) complexes

Abstract:The isomerization of simple terminal alkenes to internal isomers with Z-stereochemistry is rare, because the more stable E-isomers are typically formed. We show here that cobalt(II) catalysts supported by bulky β-diketiminate ligands have the appropriate kinetic selectivity to catalyze the isomerization of some simple 1-alkenes specifically to the 2-alkene as the less stable Z-isomer. The catalysis proceeds via an "alkyl" mechanism, with a three-coordinate cobalt(II) alkyl complex as the resting state. β-Hydride elimination and [1,2]-insertion steps are both rapid, as shown by isotopic labeling experiments. A steric model explains the selectivity through a square-planar geometry at cobalt(II) in the transition state for β-hydride elimination. The catalyst works not only with simple alkenes, but also with homoallyl silanes, ketals, and silyl ethers. Isolation of cobalt(I) or cobalt(II) products from reactions with poor substrates suggests that the key catalyst decomposition pathways are bimolecular, and lowering the catalyst concentration often improves the selectivity. In addition to a potentially useful, selective transformation, these studies provide a mechanistic understanding for catalytic alkene isomerization by high-spin cobalt complexes, and demonstrate the effectiveness of steric bulk in controlling the stereoselectivity of alkene formation.

Abstract:Positional and stereochemical selectivity in the isomerization of terminal alkenes to internal alkenes is observed using the cis-Mo(CO)4(PPh3)2 precatalyst. A p-toluenesulfonic acid (TsOH) cocatalyst is essential for catalyst activity. Various functionalized terminal alkenes have been converted to the corresponding 2-alkenes, generally favoring the Z isomer with selectivity as high as 8:1 Z:E at high conversion. Interrogation of the catalyst initiation mechanism by 31P NMR reveals that cis-Mo(CO)4(PPh3)2 reacts with TsOH at elevated temperatures to yield a phosphine-ligated Mo hydride (MoH) species. Catalysis may proceed via 2,1-insertion of a terminal alkene into a MoH group and stereoselective β-hydride elimination to yield the (Z)-2-alkene.

Abstract:The frustrated Lewis pair (FLP) comprised of B(C6F5)(3) and 1,2,2,6,6-pentamethylpiperidine (PMP) can efficiently catalyze Z-selective hydrophosphonylation of terminal ynones with a Z/E selectivity of up to 20 :1. Mechanistic studies suggest that the trans arrangement of the phosphite nucleophilic attack and hydrogen bond formation on the alkyne moiety is responsible for the observed Z-selectivity.

Abstract:Y[N(TMS)(2)](3)/FeCl3 has been found to be an efficient bimetallic catalyst system for the cyclotrimerization of terminal alkynes, which cannot be achieved by either trivalent iron or trivalent lanthanide catalysts. Furthermore, this reaction also occurs efficiently in the presence of Fe[N(TMS)(2)](3) and YCl3. Both aromatic and aliphatic alkynes are compatible with this catalytic system. It is postulated that the catalytic cyclotrimerization proceeds through a tandem intermolecular diinsertion of alkynes into the yttrium-alkynyl bond and intramolecular electrophilic addition of a pi-coordinated alkyne moiety, and the pi-coordination of Fe3+ to alkyne may play an important role in controlling the insertion degree of advancement and selectivity. The observed catalytic reaction is sharply in contrast with the cyclotrimerization of alkynes, known to proceed through a typical metallacyclopentadiene intermediate.

Abstract:In transition metal catalysis, the nature of ligand coordination plays a vital role in defining the reactivity of the catalytic system. Achieving multi-substrate coordination and coupling by leveraging dynamic ligand dissociation is rare. Here, we report the discovery of such a phenomenon in nickel/copper-co-catalyzed divergent allylic alkylation of vinyl epoxides and aziridines using alkynes. Under otherwise identical conditions, the use of either strong bisphosphine ligands or hemilabile P,N-ligands leads to bimolecular allylic alkynylation or trimolecular dienyne formation, respectively. DFT calculations provide key insights for these ligand-induced divergent reactivities, particularly ligand-dissociation-enabled coordination of three substrates on nickel for trimolecular coupling. This catalytic system couples a remarkably broad range of readily available terminal alkynes with vinyl epoxides, carbonates, and aziridines in high regio- and stereo-selectivity. The utility of the versatile enyne and dienyne products, coupled with the use of abundant nickel/copper catalysts, makes this a practical method in synthetic and medicinal chemistry.

Abstract:A counterion-controlled reactivity tuning in Pd-catalyzed highly chemoselective and regioselective dimerization and hydration of terminal alkynes is reported. The use of acetate as counterion favors the formation of an alkenyl alkynyl palladium intermediate which forms hitherto less reported 1,3-diaryl-substituted conjugated enynes after reductive elimination. Using chloride, which is a better leaving. group, leads to anion exchange on the alkenylpalladium intermediate with hydroxide which after reductive elimination and tautomerization delivered the hydration products.

Abstract:Because internal alkenes are more challenging synthetic targets than terminal alkenes, metal-catalyzed olefin mono-transposition (i.e., positional isomerization) approaches have emerged to afford valuable E- or Z- internal alkenes from their complementary terminal alkene feedstocks. However, the applicability of these methods has been hampered by lack of generality, commercial availability of precatalysts, and scalability. Here, we report a nickel-catalyzed platform for the stereodivergent E/Z-selective synthesis of internal alkenes at room temperature. Commercial reagents enable this one-carbon transposition of terminal alkenes to valuable E- or Z-internal alkenes via a Ni–H-mediated insertion/elimination mechanism. Though the mechanistic regime is the same in both systems, the underlying pathways that lead each of the active catalysts are distinct, with the Z-selective catalyst forming from comproportionation of an oxidative addition complex followed by oxidative addition with substrate and the E-selective catalyst forming from protonation of the metal by the trialkylphosphonium salt additive. In each case, ligand sterics and denticity control stereochemistry and prevent over-isomerization.

Abstract:Alkenyl metal Z/E isomerization involving use of the alkyne-tethered electrophile is an important topic in transition-metal catalyzed alkyne functionalization. In this report, we present computational evidence that supports a mechanism in which alkyne coordination to the nickel center as a η2-type ligand in assist of Z/E isomerization. This η2-vinyl-nickel type Z/E isomerization is found to be lower in energy compared with carbonyl-assisted Z/E isomerization from a syn-alkenylnickel intermediate, providing new insights into the mechanism of Ni-catalyzed anti-carbometallative cyclizations of alkyne. The newman projections analysis were conducted to characterize the key transition states of carbonyl insertion process and elucidate the origin of diastereoselectivity in this type reaction. Theoretical calculations further revealed that the Z/E isomerization mode of alkenyl nickel is more inclined to rotate through the carbon-carbon double bond, which is also confirmed by NBO charge and the Wiberg bond indices (WBI). We anticipate that the novel η2-vinyl-nickel type Z/E isomerization mode may extend to other alkyne functionalization processes and explain the unique effectiveness of this reaction mode in many instances.
