Abstract:Background: Metastasis-associated protein 1 (MTA1) plays an important role in various cancers by promoting epithelial-mesenchymal transition (EMT), which also contributes to the development of endometriosis. However, the association between MTA1 and endometriosis is little known and needs to be revealed. Methods: The study population consisted of 33 women with histologically proven endometriosis and 20 endometriosis-free controls. Immunohistochemistry(IHC) and Western blotting(WB) were performed to evaluate MTA1 expression in different tissues. Primary cells isolation and culture, gene transfection, Scratch wound assay and Transwell assay were used to analyse the effect of MTA1 on migration and invasion in endometrial cells. We analyzed Gene Expression Omnibus (GEO) and RNA Sequencing (RNA-seq) datesets to find a promising target—ZEB2. Immunoprecipitation (IP) was performed to show a interaction between MTA1 and ZEB2. Establishing a mouse model of endometriosis with 18 nude mice. Besides, treatment with resveratrol in both cells and mice were used to validate the findings. Findings: MTA1 expression was significantly higher in patients with endometriosis compared with controls and was related to revised American Fertility Society (r-AFS) classification. Up-regulation or down-regulation of MTA1 in ESCs resulted in an enhancement or inhibition on cells migration and invasion abilities respectively, as well as the change of E-cadherin and Vimentin expression. Via analyzing the dateset of GEO and RNA-seq after MTA1 knockdown, we found ZEB2 were high expressed in endometriosis tissues and contributed to endometrial cells migration and invasion abilities, which could be down-regulated by MTA1. Further, we found a physical combination between MTA1 and ZEB2. In addition, we found resveratrol repressed endometrial cells metastasis via decreasing MTA1 and ZEB2 expression. To valid these findings, we established a mice endometriosis model. The results coincided with the in vitro study that MTA1 and ZEB2 were overexpressed in ectopic tissues and resveratrol treatment led to an inhibition of ectopic tissue growth and decreased expression of MTA1 and ZEB2, as well as decreased Vimentin and increased E-cadherin expression. Overall, MTA1-ZEB2 acts as a regulator of EMT and enhances endometrial cells migration and invasion abilities, subsequently appears to promote the pathogenesis of endometriosis. Interpretation: This study provides new insight that MTA1 is upregulated and promotes EMT via interacting with ZEB2 in endometriosis.
