Abstract:Membrane electropermeabilization is the observation that the permeability of a cell membrane can be transiently increased when a micro-millisecond external electric field pulse is applied on a cell suspension or on a tissue. Applicative aspects for the transfer of foreign molecules (macromolecules) into the cytoplasm are routinely used. But only a limited knowledge about what is really occurring in the cell and its membranes at the molecular levels is available. This chapter is a critical attempt to report the present state of the art and to point out some of the still open problems. The experimental facts associated to membrane electropermeabilization are firstly reported. They are valid on biological and model systems. Secondly, soft matter approaches give access to the bioelectrochemical description of the thermodynamical constraints supporting the destabilization of simplified models of the biological membrane. It is indeed described as a thin dielectric leaflet, where a molecular transport takes place by electrophoresis and then diffusion. This naïve approach is due to the lack of details on the structural aspects affecting the living systems as shown in a third part. Membranes are part of the cell machinery. The critical property of cells as being an open system from the thermodynamical point of view is almost never present. Computer simulations are now contributing to our knowledge on electropermeabilization. The last part of this chapter is a (very) critical report of all the efforts that have been performed. The final conclusion remains that we still do not know all the details on the reversible structural and dynamical alterations of the cell membrane (and cytoplasm) supporting its electropermeabilization. We have a long way in basic and translational researches to reach a pertinent description.

Abstract:Pulsed electric fields are used to permeabilize cell membranes in biotechnology and the clinic. Although molecular and continuum models provide compelling representations of the mechanisms underlying this phenomenon, a clear structural link between the biomolecular transformations displayed in molecular dynamics (MD) simulations and the micro- and macroscale cellular responses observed in the laboratory has not been established. In this paper, plasma membrane electropermeabilization is characterized by exposing Jurkat T lymphoblasts to pulsed electric fields less than 10ns long (including single pulse exposures), and by monitoring the resulting osmotically driven cell swelling as a function of pulse number and pulse repetition rate. In this way, we reduce the complexity of the experimental system and lay a foundation for gauging the correspondence between measured and simulated values for water and ion transport through electropermeabilized membranes. We find that a single 10MV/m pulse of 5ns duration produces measurable swelling of Jurkat T lymphoblasts in growth medium, and we estimate from the swelling kinetics the ion and water flux that follows the electropermeabilization of the membrane. From these observations we set boundaries on the net conductance of the permeabilized membrane, and we show how this is consistent with model predictions for the conductance and areal density of nanoelectropulse-induced lipid nanopores.

Abstract:Most experimental studies of electroporation focus on permeabilization of the outer cell membrane. Some experiments address delivery of ions and molecules into cells that should survive; others focus on efficient killing of the cells with minimal temperature rise. A basic method for quantifying electroporation effectiveness is measuring the membrane's diffusive permeability. More specifically, comparisons of membrane permeability between electroporation protocols often rely on relative fluorescence measurements, which are not able to be directly connected to theoretical calculations and complicate comparisons between studies. Here we present part I of a 2-part study: a research method for quantitatively determining the membrane diffusive permeability for individual cells using fluorescence microscopy. We determine diffusive permeabilities of cell membranes to propidium for electric field pulses with durations of 1 to 1000 μs and strengths of 170 to 400 kV/m and show that diffusive permeabilities can reach 1.3±0.4×10-8 m/s. This leads to a correlation between increased membrane permeability and eventual propidium uptake. We also identify a subpopulation of cells that exhibit a delayed and significant propidium uptake for relatively small single pulses. Our results provide evidence that cells, especially those that uptake propidium more slowly, can achieve large permeabilities with a single electrical pulse that may be quantitatively measured using standard fluorescence microscopy equipment and techniques.

Abstract:Recent studies showed that nanosecond pulsed electric fields (nsPEFs) can activate voltage-gated ion channels (VGICs) and trigger action potentials (APs) in excitable cells. Under physiological conditions, VGICs' activation takes place on time scales of the order 10-100 µs. These time scales are considerably longer than the applied pulse duration, thus activation of VGICs by nsPEFs remains puzzling and there is no clear consensus on the mechanisms involved. Here we propose that changes in local electrical properties of the cell membrane due to lipid oxidation might be implicated in AP activation. We first use MD simulations of model lipid bilayers with increasing concentration of primary and secondary lipid oxidation products and demonstrate that oxidation not only increases the bilayer conductance, but also the bilayer capacitance. Equipped with MD-based characterization of electrical properties of oxidized bilayers, we then resort to AP modelling at the cell level with Hodgkin-Huxley-type models. We confirm that a local change in membrane properties, particularly the increase in membrane conductance, due to formation of oxidized membrane lesions can be high enough to trigger an AP, even when no external stimulus is applied. However, excessive accumulation of oxidized lesions (or other conductive defects) can lead to altered cell excitability.

Abstract:Exceeding physiological limits of the cell membrane potential compromises structural integrity, enabling the passage of normally impermeant solutes and disrupting cell function. Electropermeabilization has been studied extensively at the cellular scale, but not at the individual membrane lesion level. We employed fast total internal reflection fluorescence (TIRF) imaging of Ca2+ entry transients to discern individual lesions in a hyperpolarized cell membrane and characterize their focality, thresholds, electrical conductance, and the lifecycle. A diffuse and momentary membrane permeabilization without a distinct pore formation was observed already at a -100 mV threshold. Polarizing down to -200 mV created focal pores with a low 50- to 300-pS conductance, which disappeared instantly once the hyperpolarization was removed. Charging to -240 mV created high-conductance (> 1 nS) pores which persisted for seconds even at zero membrane potential. With incremental hyperpolarization steps, persistent pores often emerged at locations different from those where the short-lived, low-conductance pores or diffuse permeabilization were previously observed. Attempts to polarize membrane beyond the threshold for the formation of persistent pores increased their conductance adaptively, preventing further potential build-up and "clamping" it at a certain limit (-270 ± 6 mV in HEK cells, -284 ± 5 mV in CHO cells, and -243 ± 9 mV in neurons). The data suggest a previously unknown role of electroporative lesions as a protective mechanism against a potentially fatal membrane overcharging and cell disintegration.

Abstract:The molecular mechanism of how insects recognize intruding microorganisms and parasites and distinguish them from own body structures is not well known. We explored evolutionary adaptations in an insect parasitoid host interaction to identify components that interfere with the recognition of foreign objects and cellular encapsulation. Because some parasitoids provide protection for the developing wasp in the absence of an overt suppression of the insect host defense, we analyzed the surface of eggs and symbiotic viruses for protective properties. Here we report on the molecular cloning of a 32-kDa protein (Crp32) that is one of the major protective components. It is produced in the calyx cells of the female wasp ovaries and attached to the surface of the egg and other particles including polydnaviruses. The recombinant protein confers protection to coated objects in a cellular encapsulation assay suggesting that a layer of Crp32 may prevent cellular encapsulation reactions by a local inactivation of the host defense system.

Abstract:The formation of pores over lipid membranes by the application of electric fields, termed membrane electroporation, is widely used in biotechnology and medicine to deliver drugs, vaccines, or genes into living cells. Continuum models for describing the free energy landscape of membrane electroporation were proposed decades ago, but they have never been tested against spatially detailed atomistic models. Using molecular dynamics (MD) simulations with a recently proposed reaction coordinate, we computed potentials of mean force of pore nucleation and pore expansion in lipid membranes at various transmembrane potentials. Whereas the free energies of pore expansion are compatible with previous continuum models, the experimentally important free energy barrier of pore nucleation is at variance with established models. The discrepancy originates from different geometries of the transition state; previous continuum models assumed the presence of a membrane-spanning defect throughout the process, whereas, according to the MD simulations, the transition state of pore nucleation is typically passed before a transmembrane defect has formed. A modified continuum model is presented that qualitatively agrees with the MD simulations. Using kinetics of pore opening together with transition state theory, our free energies of pore nucleation are in excellent agreement with previous experimental data. https://doi.org/10.1103/PhysRevLett.132.148401 © 2024 American Physical Society

Abstract:Analogues of androsta-1,4-diene-3,17-dione (3a) in which the D ring is modified were prepared and tested as suicide inactivators and competitive inhibitors of human placental aromatase. As long as the five-membered ring is intact, modifications of the D ring such as reduction or removal of the carbonyl group or conversion to a gamma-butyrolactone cause a less than 6-fold decrease in affinity for and rate of inactivation of aromatase, compared to 3a. Thus, an oxygen atom at C-17 is not required for binding of these inhibitors to aromatase, suggesting that hydrogen bonding to the D-ring oxygen does not play a major role in binding. Opening the D ring converts the cyclopentane ring to an alkyl chain and causes a greater than 300-fold decrease in affinity; this can be partially reversed by shortening the chain length. These results are consistent with a model in which the free chain of the opened D ring adopts conformations that sterically interfere with binding of the inhibitor to the enzyme. These findings may have practical applications in drug design, by allowing the preparation of 17-deoxo analogues that have high affinity for aromatase but that are not subject to reduction of the 17-carbonyl group, which is a major mode of metabolism of 3a.
The Electrostatic Component of the Disjoining Pressure and the Pore Creation Rate in Electroporation Models and Theory

Abstract:Under externally applied electric fields, lipid membranes tend to permeate and change their electrical resistance by the combined processes of pore creation and pore evolution (expansion or contraction). This study is focused on the pore creation process, represented by an empirical expression currently used in the electroporation (EP) models, for which an alternative theoretically based expression was provided. The choice of this expression was motivated by the role the DLVO's (disjoining) pressures may play in the process of EP. The electrostatic energy effects on each sides of a lipid membrane were evaluated in terms of the electrostatic component of the disjoining pressure. Thus the pore creation energy considerations in the current EP models, associated with the necessity of an idealized non conducting circular pre-pore were avoided. As a result, a new expression for the onset of the electroporation was proposed. It was found that this new theoretically determined expression is in good agreement with the one estimated from an experiment that specifically targeted the pore nucleation by eliminating the pore evolution process. Furthermore, the proposed expression is dependent on the electrolyte and the lipid properties and should provide better predictability than the currently used pore creation EP model, which is only temperature dependent.

Abstract:The aim of the present study was to explore whether acupuncture may improve the absorption of baicalin from the extracts of Scutellaria baicalensis Georgi in rats with pelvic inflammation. 24 female Sprague-Dawley (SD) rats were randomly divided into groups A (a combination of oral administration of Scutellaria baicalensis Georgi extracts and acupuncture), B (oral administration of Scutellaria baicalensis Georgi extracts), C (model group) and D (normal control group), with 6 rats in each group. After treatment, the plasma concentration of baicalin was determined using high performance liquid chromatography (HPLC), the serum levels of tumor necrosis factor-alpha (TNF-α) and Interleukin 6 (IL-6) were detected with enzyme-linked immunosorbent assay (ELISA). The results showed that the plasma concentration of baicalin in group A was significantly higher than in group B, and the serum levels of TNF-α and IL-6 in group A were significantly lower than in group B. This indicates that acupuncture at certain acupoints may significantly improve the curative effects of Scutellaria baicalensis Georgi in rats with pelvic inflammation by enhancing the absorption of baicalin from the Scutellaria baicalensis Georgi extracts.

Abstract:Electroporation causes a temporal increase in cell membrane permeability and leads to prolonged changes in transmembrane voltage (TMV) in both excitable and non-excitable cells. However, the mechanisms of these TMV changes remain to be fully elucidated. To this end, we monitored TMV over 30 min after exposing two different cell lines to a single 100 μs electroporation pulse using the FLIPR Membrane Potential dye. In CHO-K1 cells, which express very low levels of endogenous ion channels, membrane depolarization following pulse exposure could be explained by nonselective leak current, which persists until the membrane reseals, enabling the cells to recover their resting TMV. In U-87 MG cells, which express many different ion channels, we unexpectedly observed membrane hyperpolarization following the initial depolarization phase, but only at 33 °C and not at 25 °C. We developed a theoretical model, supported by experiments with ion channel inhibitors, which indicated that hyperpolarization could largely be attributed to the activation of calcium-activated potassium channels. Ion channel activation, coupled with changes in TMV and intracellular calcium, participates in various physiological processes, including cell proliferation, differentiation, migration, and apoptosis. Therefore, our study suggests that ion channels could present a potential target for influencing the biological response after electroporation.

Abstract:Sneddon syndrome is characterized by livedo reticularis and multiple cerebral infarctions. Skin and central nervous system symptoms usually have a synchronous onset and at times initial symptoms affect one of them, the other lagging several years behind. We here report a patient with Sneddon syndrome who developed multiple cerebral infarctions more than 10 years after the onset of livedo reticularis. While the neurological symptoms were apparent, the patient did not display active skin manifestations. Laboratory findings excluded collagen diseases, antiphospholipid antibody syndrome, and inherited quantitative deficiency of protein C, protein S and antithrombin III. Abnormal findings included extremely elevated levels of β‐thromboglobulin and platelet factor‐4 in the blood, although these acute phase markers of thrombosis were examined several years after the onset of cerebral infarctions. Platelet activation may have caused Sneddon syndrome in the present case.
Lipid oxidation alters membrane mechanics favoring permeabilization

Abstract:Ferroptosis is a form of regulated necrosis characterized by the iron-dependent accumulation of lipid peroxides in cell membranes. However, how lipid oxidation affects biophysical properties of cellular membranes and how these changes contribute to the opening of membrane pores are major questions in the field. Therefore, understanding the interplay between oxidized phospholipids and alterations in membrane biophysical properties is essential to unravel the underlying mechanisms of ferroptosis. Here, we characterized membrane alterations upon lipid oxidation in in vitro model membrane systems using lipid vesicles and supported lipid bilayers (SLBs) in which lipid oxidation was induced by Fenton reactions. We find that vesicle permeabilization kinetically correlates with the appearance of malondialdehyde (MDA), a product of lipid oxidation. Lipid oxidation also alters the lateral organization of SLBs exhibiting liquid-ordered (Lo)/liquid-disordered (Ld) phase coexistence, reducing the lipid mismatch between domains and decreasing the circularity of Lo domains, indicating a decrease in line tension at the phase boundaries. Further characterization of SLBs by force spectroscopy reveals a significant reduction in the average membrane breakthrough force upon oxidation, indicating changes in lipid bilayer organization that make it more susceptible to permeabilization. Our findings suggest that lipid oxidation induces strong changes in membrane lipid interactions and in the mechanical properties of membranes leading to reduced tension in the permeabilized state of the bilayer, which promotes membrane pore formation.

Abstract:In the study of electroporation, the conductivity of the cell membrane is a crucial factor in assessing the permeability of cell membrane. In previous researches, the oversimplified equivalent resistance models were utilized to describe changes in cell membrane permeability, resulting in inaccurate results and a significant gap between experimental and simulation outcomes. In this paper, an electroporation model that incorporates a dynamic conductivity equation based on ion transmembrane transport is proposed, enabling more accurate assessments of changes in cell membrane permeability. A two-dimensional (2-D) finite-element model with coupled multi-physical field is developed to validate the proposed model. The changes in the cell conductivity is used to reveal evolutionary trends in membrane permeability. The simulation results align well with literature data and provide a mechanistic explanation for previous experimental results. Meanwhile, the effect of transmembrane transport of ions on cell membrane perforation is discussed, with results indicating that the migration of ions significantly alter the conductive properties around the cell membrane, further impacting the evolution of membrane permeability. The proposed electroporation model provides a significant improvement over previous models, allowing for more accurate predictions of cell membrane permeability. This model is of great value in the study of electroporation-mediated small molecule delivery and in developing new strategies for intracellular drug delivery.
