Abstract:The enzymatic interesterification of a solvent‐free, equimolar mixture of trilaurin and 1,3‐palmitin‐2‐olein was studied using three immobilized lipase preparations as catalysts. Analysis of triacylglycerol (TAG) content and fatty acid (FA) distribution monitored the lipase‐catalyzed interesterification in sn‐1,3 positions and FA exchange in the sn‐2 position caused by acyl migration. Lipase from Rhizopus oryzae immobilized on polypropylene showed high sn‐1,3 regioselectivity, and minimal exchange in the sn‐2 position. With lipase from Thermomyces lanuginosus on silica (Lipozyme® TL IM), completely randomized FA distribution was obtained in 24 h. T. lanuginosus lipase on polypropylene caused a moderate rate of FA exchange in the sn‐2 position. Thus, the T. lanuginosus lipase and silica promoted randomization of FA distribution, whereas the R. oryzae lipase and polypropylene did not. Higher water activity promoted hydrolysis and thereby increased concentrations of partial acylglycerols, but at the same time a decrease in the acyl migration rate of these intermediates was also observed. The net result was that at a certain degree of interesterification, there was no significant effect of water activity on the degree of exchange in the sn‐2 position. On the other hand, a low water activity had the major advantage of giving a high yield of TAG.Practical applications: Both the type and the position of FA affect the properties of TAG. In TAG interesterification, there are thus different requirements for regiospecificity in FA exchange, depending on which product is desired. According to our results, lipase‐based catalysts can be used for TAG interesterification either to achieve unchanged FA composition in sn‐2 position or fast FA randomization in all positions. This helps to broaden the application of lipases in interesterification. The production of TAG can be tailor‐made for lipid mixtures with particular TAG composition and FA distribution using proper lipases, support materials, water activity, and reaction time.The enzymatic LLL–POP model reaction was used to investigate effects of lipase, support material, and water activity on acyl migration. Abstract:Aspergillus terreus MTCC 11096 isolated from the soils of agricultural fields cultivating sweet sorghum was previously identified to produce feruloyl esterases (FAEs). The enzymes responsible for feruloyl esterase activity were purified to homogeneity and named as AtFAE-1, AtFAE-2, and AtFAE-3. The enzymes were monomeric having molecular masses of 74, 23 and 36 kDa, respectively. Active protein bands were identified by a developed pH-dependent zymogram on native PAGE. The three enzymes exhibited variation in pH tolerance ranging between pH 5-8 and thermostability of up to 55°C. Inhibition studies revealed that the serine residue was essential for feruloyl esterase activity; moreover aspartyl and glutamyl residues are not totally involved at the active site. Metal ions such as Ca(2+), K(+), and Mg(2+) stabilized the enzyme activity for all three FAEs. Kinetic data indicated that all three enzymes showed catalytic efficiencies (k(cat) /K(m)) against different synthesized alkyl and aryl esters indicating their broad substrate specificity. The peptide mass fingerprinting by MALDI/TOF-MS analysis and enzyme affinity toward methoxy and hydroxy substituents on the benzene ring revealed that the AtFAE-1 belonged to type A while AtFAE-2 and AtFAE-3 were type C FAE. The FAEs could release 65 to 90% of ferulic acid from agrowaste substrates in the presence of xylanase.

Abstract:Producing ferulic acid (FA) from the natural substrate with feruloyl esterase is promising in industries, screening and engineering new enzymes with high efficiency to increase the FA yield is of great concern. Here, the feruloyl esterase of Lactobacillus acidophilus (FAELac) was heterologous expressed and the FAELac with different oligomerization states was separated. Interestingly, the activity of dimer was 37-fold higher than high-polymer. To further enhance the efficiency of FAELac, eight mutants were generated based on the simulated structure, of which Q198A, Q134T enhanced the catalytic efficiency by 5.4- and 4.3-fold in comparison with the wild type. Moreover, higher yields of FA (2.21, 6.60, and 1.67 mg/g substrate, respectively) were released by the mutants from de-starched wheat bran, insoluble wheat arabinoxylan, and steam-exploded corn stover. These results indicated that improving the purification process, engineering new FAELac and substrates bias studies hold great potential for increasing FA production yield.

Regioselective chemo-enzymatic syntheses of ferulate conjugates as chromogenic substrates for feruloyl esterases

Abstract:Generally, carbohydrate-active enzymes are studied using chromogenic substrates that provide quick and easy color-based detection of enzyme-mediated hydrolysis. In the case of feruloyl esterases, commercially available chromogenic ferulate derivatives are both costly and limited in terms of their experimental application. In this study, we describe solutions for these two issues, using a chemoenzymatic approach to synthesize different ferulate compounds. The overall synthetic routes towards commercially available 5-bromo-4-chloro-3-indolyl and 4-nitrophenyl O -5-feruloyl-α-l-arabinofuranosides 1a and 1b were significantly shortened (7-8 steps reduced to 4-6) and transesterification yields enhanced (from 46 to 73% for 1a and 47 to 86 % for 1b ). This was achieved using enzymatic (immobilized Lipolase 100T from Thermomyces lanuginosus ) transesterification of unprotected vinyl ferulate to the primary hydroxyl group of α‐l‐arabinofuranosides. Moreover, a novel feruloylated-butanetriol 4-nitrocatechol-1-yl analog 12 , containing a cleavable hydroxylated linker was also synthesized in 29% overall yield in 3 steps (convergent synthesis). The latter route combined regioselective functionalization of 4-nitrocatechol and enzymatic transferuloylation. The use of 12 as a substrate to characterize type A feruloyl esterase from Aspergillus niger reveals the advantages of this substrate for the characterizations of feruloyl esterases.

Abstract:Lipase from Candida rugosa (CRL) is an efficient enzyme for phytosterol esterification. However, the poor stability limits its application. A novel cross-linked enzyme aggregates of CRL (CRL-FIL-CLEAs@Fe3O4) was designed by lipase surface modification with functional ionic liquid (FIL) and immobilization with magnetic Fe3O4 nanoparticles as a carrier simultaneously. The loading of CRL was 0.16 mg protein/mg CRL-FIL-CLEAs@Fe3O4. The hydrolysis expressed activity of CRL-FIL-CLEAs@Fe3O4 was 1.36±0.04 U/mg CRL-FIL-CLEAs@Fe3O4. The hydrolysis specific activity of CRL-FIL-CLEAs@Fe3O4 (8.48±0.25 U/mg protein) was significantly higher than conventional CRL-CLEAs (2.86±0.26 U/mg protein). After 15 days of storage at room temperature, CRL-FIL-CLEAs@Fe3O4 retained 2.6 times higher activity (73.31%) than that of free lipase (28.52%). After eleven times of reuse, the CRL-FIL-CLEAs@Fe3O4 still retained high hydrolysis activity (83.42%). CRL-FIL-CLEAs@Fe3O4 was further applied in solvent-free esterification reaction of phytosterol with oleic acid, and the conversion rate of phytosterol was up to 93.24% after 22 hours (reaction temperature was 48 °C, substrate molar ratio of oleic acids to phytosterols was 10:1, the number of biocatalysts and substrate phytosterol was 5 mg and 0.1mmol, respectively). It was expected that a good reference could be provided for the more efficient and green production of food-grade phytosterol esters.

Novel Feruloyl Esterase from Lactobacillus fermentum NRRL B-1932 and Analysis of the Recombinant Enzyme Produced in Escherichia coli

Abstract:A total of 33 Lactobacillus strains were screened for feruloyl esterase (FE) activity using agar plates containing ethyl ferulate as the sole carbon source, and Lactobacillus fermentum NRRL B-1932 demonstrated the strongest FE activity among a dozen species showing a clearing zone on the opaque plate containing ethyl ferulate. FE activities were monitored using high-performance liquid chromatography with an acetonitrile-trifluoroacetic acid gradient. To produce sufficient purified FE from L. fermentum strain NRRL B-1932 (LfFE), the cDNA encoding LfFE (Lffae) was amplified and cloned by using available closely related genome sequences and overexpressed in Escherichia coli A 29.6-kDa LfFE protein was detected from the protein extract of E. coli BL21(pLysS) carrying pET28bLffae upon IPTG (isopropyl-β-d-thiogalactopyranoside) induction. The recombinant LfFE containing a polyhistidine tag was purified by nickel-nitrilotriacetic acid affinity resin. The purified LfFE showed strong activities against several artificial substrates, including p-nitrophenyl acetate and 4-methylumbelliferyl p-trimethylammoniocinnamate chloride. The optimum pH and temperature of the recombinant LfFE were around 6.5 and 37°C, respectively, as determined using either crude or purified recombinant LfFE. This study will be essential for the production of the LfFE in E. coli on a larger scale that could not be readily achieved by L. fermentum fermentation. Importance: The production of feruloyl esterase (FE) from Lactobacillus fermentum NRRL B-1932 reported in this study will have immense potential commercial applications not only in biofuel production but also in pharmaceutical, polymer, oleo chemical, cosmetic additive, and detergent industries, as well as human health-related applications, including food flavoring, functional foods, probiotic agents, preventive medicine, and animal feed. Given the essential role FE plays in the production of hydroxycinnamic acids and ferulic acid, plus the generally regarded as safe status of lactobacilli, which therefore have less regulatory concerns, LfFE from the probiotic L. fermentum reported in this work can be directly used for increased production of high-value hydroxycinnamates and ferulic acid from natural or synthetic carbon sources.
The Combined Cultivation of Feruloyl Esterase-Producing Strains with CMCase and Xylanase-Producing Strains Increases the Release of Ferulic Acid

Abstract:Feruloyl esterase (FAE)-producing micro-organisms to obtain ferulic acid (FA) from natural substrates have good industrial prospects, and the synergistic effect of multiple bacteria can better improve the yield of FA. In this study, on the premise of the synergistic effect of FAE, hemicellulose, and cellulase, the key strain Klebsiella oxytoca Z28 with FAE was combined with CMCase and Xylanase-producing strains to produce FA. The combination of strains with higher FA production are Klebsiella oxytoca Z28, Klebsiella pneumoniae JZE, Bacillus velezensis G1, and their FA production can reach 109.67 μg/g, which is 15% higher than that of single bacteria. To explore the effects of temperature, Ph, inoculum amount, distillers grains concentration and fermentation time on the FAE activity of the combination of strains in the fermentation process, and determined that temperature, Ph, and fermentation time were the main influencing factors and optimized through orthogonal design. The optimized fermentation conditions are 34 °C, Ph 8.0, and fermentation days for 6 days, the FAE activity can reach 270.78 U/L, and the FA yield of the combined strain is 324.50 μg/g, which is 200% higher than that of single-strain fermentation.

Transesterification of Methyl Oleate for Sustainable Production of Biolubricant: Process Optimization and Kinetic Study

Abstract:Trimethylolpropane fatty acid triester (TFATE) is a promising biolubricant as an environmentally friendly substitute to conventional mineral-based lubricants. However, the issues of extensive energy consumption from vacuum maintenance and feedstock saponification caused by strong alkaline catalysts exist in current TFATEbased biolubricant synthesis. Here, sustainable production of biolubricant was achieved with methyl oleate (MO) from non-edible oil sources as the feedstock and moderate alkaline K2CO3 as the catalyst. Nitrogen gas stripping method instead of vacuum was adopted for methanol removal. The effects of agitation speed, N-2 flow rate, catalyst amount, MO to trimethylolpropane (TMP) molar ratio and temperature on MO conversion and TFATE selectivity were examined. The TFATE selectivity reached 95.6% under the optimized conditions. Leaching tests indicated neglectable K2CO3 dissolution in reaction products. The kinetic parameters for the three consecutive transesterification reactions were calculated. In addition, the synthesized biolubricants exhibit low acid value and volatility, moderate iodine value, superior thermal stability, and good rheological and frictional properties.

Abstract:Feruloyl esterase (FAE; EC 3.1.1.73) cleaves the ester bond between ferulic acid (FA) and sugar, to assist the release of FAs and degradation of plant cell walls. In this study, two FAEs (Fae13961 and Fae16537) from the anaerobic fungus Pecoramyces sp. F1 were heterologously expressed in Pichia pastoris (P. pastoris). Abstract:Short-chain alkyl esters and sugar esters are widely used in the food, pharmaceutical and cosmetic industries due to their flavor and emulsifying characteristics, respectively. Both compounds can be synthesized via biocatalysis using lipases. This work aims to compare the performance of commercial lipases covalently attached to dry acrylic beads functionalized with oxirane groups (lipases from Candida antarctica type B-IMMCALB-T2-350, Pseudomonas fluorescens-IMMAPF-T2-150, and Thermomyces lanuginosus-IMMTLL-T2-150) and a home-made biocatalyst (lipase from Pseudomonas fluorescens adsorbed onto silica coated with octyl groups, named PFL-octyl-silica) in the syntheses of short- and long-chain carboxylic acid esters. Esters with flavor properties were synthetized by esterification of acetic and butyl acids with several alcohols (e.g., ethanol, 1-butanol, 1-hexanol, and isoamyl alcohol), and sugar esters were synthetized by esterification of oleic and lauric acids with fructose and lactose. All biocatalysts showed similar performance in the syntheses of short-chain alkyl esters, with conversions ranging from 88.9 to 98.4%. However, in the syntheses of sugar esters the performance of PFL-octyl-silica was almost always lower than the commercial IMMCALB-T2-350, whose conversion was up to 96% in the synthesis of fructose oleate. Both biocatalysts showed high operational stability in organic media, thus having great potential for biotransformations.

Efficient Ferulic Acid and Xylo-Oligosaccharides Production by a Novel Multi-Modular Bifunctional Xylanase/feruloyl Esterase Using Agricultural Residues As Substrates

Abstract:Liberating high value-added compounds ferulic acid (FA) and xylo-oligosaccharides (XOSs) from agricultural residues is a promising strategy for the utilization of lignocellulose. In this study, a bifunctional xylanase/feruloyl esterase from bacterial consortium EMSD5 was heterogeneously expressed in Escherichia coli. Depending on the inter-domain synergism of the recombinant enzyme rXyn10A/Fae1A, high yields of FA (2.78, 1.82, 1.15 and 7.31 mg/g substrate, respectively) were obtained from 20 mg in-soluble wheat arabinoxylan, de-starched wheat bran, ultrafine-grinding corn stover and steam-exploded corncob. Meanwhile, 3.210, 1.235, 1.215 and 0.823 mg xylose/XOSs were also released. For cost-saving enzyme production, we firstly constructed a recombinant E. coli, which could secrete the bifunctional xylanase/feruloyl esterase out of cells. When the recombinant E. coli was cultured in medium containing 200 mg de-starched wheat bran, 474 mu g FA and 18.2 mg xylose/XOSs were also detected. Hence, rXyn10A/Fae1A and the recombinant strain showed great applied potential for FA and XOSs production.
[Rifampicin in the treatment of Staphylococcus arthritis].

Abstract:Rifampicin is used for treating 20 Staphylococcus septic arthritis, in combination with another antibiotic. The former is chosen for its striking bone diffusion. The average duration of the whole antibiotic treatment is 3 months. Rifampicin is usually prescribed, in daily doses of 1 200 mg for 60 days, after one month of parenteral antibiotherapy. The bactericidal activity is controlled regularly by measuring serum inhibitory titre. For 2 patients, the administration of rifampicin is suspended after 12 and 25 days respectively, due to digestive disorders. Results are evaluated at the end of a period averaging 18 months. Articular infection has been cured in all patients. Taking these results into account, we are able to emphasize that the inclusion of rifampicin in the antibiotic scheme is of great value in treating bone and articular infections.

Abstract:This work aimed to evaluate the lipases from Yarrowia lipolytica as a biocatalyst for synthesizing fatty acid ethyl esters (FAEE). The Yarrowia lipolytica lipases were evaluated in three distinct forms: firstly, as the crude enzymatic liquid extract derived from the submerged fermentation process of Y. lipolytica (YLL); secondly, as YLL immobilized on silica-alumina (YLL-S40); and thirdly, as the fermented solid material obtained from solid-state fermentation of Y. lipolytica (SEP-YLL). The highest FAEE content (54 wt.%) was obtained after 240 h in transesterification reactions using SEP-YLL. The influence of temperature, biocatalyst concentration, and reaction time was studied in the ethyl oleate synthesis. The SEP-YLL also showed better results compared to YLL and YLL-S40. The reaction catalyzed by SEP-YLL achieved 81% conversion at 30 °C. The SEP-YLL was evaluated in a batch-stirred reactor (BSTR) and a continuous packed-bed reactor (PBR), obtaining an oleic acid conversion of 80% and 70%, respectively. The low-cost biocatalyst (SEP-YLL) demonstrated potential for application in biodiesel production by reducing free fatty acids from an acid oil source through PBR and BSTR.

Abstract:Brewer's spent grain (BSG), the most abundant by-product of the beer industry, can be considered a sustainable source of ferulic acid (4-hydroxy-3-methoxycinnamic acid) (FA). FA is esterified to the arabinoxylan in the BSG. FA exhibits a variety of biological activities and is a target for valorization of BSG. We present a novel enzyme-assisted approach that utilizes the synergistic action of specific endo-1,4-β-xylanase and ferulic acid esterase activity to selectively release high yields of FA from BSG. The study compares the efficacy of three different fungal feruloyl esterases (E.C. 3.1.1.73) of carbohydrate esterase family 1 (CE1), Hi Fae, Ao Fae1, and An Fae1, acting separately or in combination with two different endo-1,4-β-xylanases (EC 3.2.1.8), a bacterial GH30 enzyme from Dickeya chrysanthemi , and Shearzyme®, a commercial GH10 enzyme from Aspergillus aculeatus , respectively. On an equimolar dosage basis, the An Fae1-GH30 was the most efficient combination in releasing FA from BSG. This combination showed significant synergy in a dose-dependent manner, enabling up to 85% FA recovery from BSG within 30 min. of treatment. The results demonstrate a novel enzymatic approach for valorization of BSG as FA is considered a high-value natural compound for use in e.g. anti-aging skin care products.

Abstract:Plant biomass is a kind of renewable, abundant, eco-friendly, and clean natural resource. Recent increase in industrial demand, coupled with development and utilization of biocatalysts for utilization and production of high value-added products from plant biomass have attracted numerous research interest. The ester bond, between plant cell wall polysaccharides and ferulic acid (FA), can be hydrolysed by feruloyl esterases (FAE), thereby destroying compactness of lignocellulosic materials, which is beneficial during utilization of biomass resources. This is this research focus. In this article, we reviewed classification, characteristics, and application of FAE from different species, then highlighted the synergistic effect of FAE action with other enzymes. The findings are considered to provide insights into role of FAE in functional research, biochemical properties, and industrial application, especially with focus on processing of lignocellulosic biomass, harnessing hydroxycinnamate from agricultural by-products, and production of biofuels among other industrial uses.
Magnetic cross-linked lipase aggregates coupled with ultrasonic pretreatment for efficient synthesis of phytosterol oleate

Abstract:In this study, coupling with substrate ultrasonic pretreatment technology, magnetic cross-linked enzyme aggregates (CRL-FIL-CLEAs@Fe3O4) was used as novel biocatalyst for efficient preparation of phytosterol oleate in solvent-free reaction system. The CRL loading was 0.16 mg protein/mg CRL-FIL- CLEAs@Fe3O4. The effects of ultrasonic parameters, catalyst dosage, substrate molar ratio, and reaction temperature on the synthesis of phytosterol esters were studied. The results showed that the catalytical properties of CRL-FIL-CLEAs@Fe3O4 (7.09±0.11 μmol phytosterol/mg protein. hour) was significantly improved compared with natural lipase from Candida rugosa (CRL, 5.13±0.21 μmol phytosterol/mg protein. hour). Simultaneously, ultrasonic pretreatment greatly enhanced substrate mass transfer, further reducing the esterification time. The max conversion rate 94.72% of phytosterol was obtained after 9 h reaction under optimum condition (the molar ratio of oleic acid to phytosterol was 6:1, the concentration of CRL-FIL-CLEAs@Fe3O4 was 10 wt% of the phytosterol mass). After five recycles, the activity of CRL-FIL-CLEAs@Fe3O4 (80.02%) was still 2.68 times higher than that of natural lipase system (29.91%). Meanwhile, the novel immobilized lipase (CRL-FIL-CLEAs@Fe3O4) before and after reaction was characterized by SEM, AFM, VSM, TEM, and TGA. The results showed that CRL-FIL-CLEAs@Fe3O4 was a robust and recyclable biocatalyst for phytosterol esterification.

Abstract:Using lipases to catalyze the synthesis of the most differentiated type of compounds remains one of the major challenges among scientists. Seeking more economic and advantageous catalysts is a current goal of green chemistry. In this work, we demonstrate the potential of a chemically modified form of lipase from Thermomyces lanuginosus (cmLTL) for the synthesis of both hydrophobic (heptyl heptanoate, heptyl octanoate, heptyl decanoate, decyl heptanoate, decyl octanoate and decyl decanoate) and amphiphilic (2‐(2‐ethoxyethoxy)ethyl oleate and 2‐(2‐ethoxyethoxy)ethyl linoleate) esters, in bulk. The results were compared with its native (LTL) and immobilized (imLTL) forms. The data revealed that LTL showed poor activity for all reactions performed with n‐heptane (h 60%), with exception of the short ester, heptyl heptanoate. cmLTL was the only form of LTL capable of producing hydrophobic and amphiphilic esters, without compromising the yield when the reactions were performed under solvent‐free conditions (>50%). Molecular modeling showed that the active pocket of cmLTL is able to deeply internalize transcutol, with stronger interactions, justifying the outstanding results obtained. Furthermore, owing to the possibility of cmLTL filtration, the reusability of the catalyst is ensured for at least 6 cycles, without compromising the reaction yields.
