Abstract:The non-chloroplastic α-glucan phosphorylase (EC 2.4.1.1) from spinach leaves has been purified to homogeneity as revealed by dodecylsulfate gel electrophoresis. Both purification and separation from the chloroplastic phosphorylase were achieved by chromatography on Sepharose-bound dextrin. The chloroplastic phosphorylase did not bind to Sepharose-dextrin and was removed from the column by washing with buffer, as verified by polyacrylamide gel electrophoresis of the buffer eluate and by chromatography of a preparation from isolated intact chloroplasts. The non-chloroplastic phosphorylase did bind to a high extent to Sepharose-dextrin and could be eluted by a dextrin gradient. Based on dodecylsulfate gel electrophoresis and pyridoxal phosphate determination, a molecular weight of about 90,000 was found for the monomer. Molecular-weight determination by porosity density gradient electrophoresis and gel filtration on Sephadex G-200 suggested that the native enzyme is a dimer, as are other phosphorylases.
Inactivation Kinetics of the Reduced Spinach Chloroplast Fructose-1,6-bisphosphatase by Subtilisin.

Abstract:The course of inactivation of the reduced spinach chloroplast fructose-1,6-bisphosphatase by digestion with subtilisin has been followed by the progress curve method [Tsou, C. L. (1988) Adv. Enzymol. 61, 381-436] and found to follow first-order kinetics. On the basis of the hydrolysis of the substrate, fructose 1,6-bisphosphate, at different concentrations during proteolysis by subtilisin, the first-order inactivation rate constants for the free enzyme and the enzyme-substrate complex can both be determined. The ratio between the inactivation rate constants for the free enzyme and the enzyme-substrate complex indicates strong protection against subtilisin proteolysis by the substrate. It is proposed that the above ratio can be used as a quantitative measure of substrate protection for enzyme inactivation generally. As it has been found that the site of proteolysis is located in a loop region near the N-terminus and well away from the active site, the substrate protection indicates a conformation change of the enzyme away from the substrate binding site.
Purification and characterisation of soluble invertases from leaves of Arabidopsis thaliana

Abstract:Multiple isoforms of β-fructofuranosidase (invertase, EC 3.2.1.26) were identified in mature green leaves of the cruciferous plant Arabidopsis thaliana (L.) Heynh. There were four major and one minor isoforms of soluble acid invertase and an additional activity which could be released from the cell wall by buffers of high ionic strength. This study reports the separation and characterisation of three soluble isoforms following ammonium sulphate and polyethylene glycol 6000 precipitations, Concanavalin A, MonoQ ion exchange, Superose 12 sizeexclusion chromatography and chromatofocusing. These isoforms, designated INV1, INV2 and INV3, had isoelectric points of 4.75, 4.70 and 4.65 and a Km for sucrose of 5, 12 and 5 mM, respectively. Each had a pH optimum of 5.5, exhibited optimal activity at 45 °C and used sucrose as the preferred substrate. All fractions containing these isoforms contained a 52-kDa polypeptide which was specifically detected by immunoblotting with an antibody raised against deglycosylated wheat invertase. The N-terminal amino-acid sequence of this polypeptide was homologous to acid invertases isolated from other plant species. The possible origin of isoforms of soluble acid invertase is discussed.
Purification and Characterization of Gβγ-responsive Phosphoinositide 3-Kinases from Pig Platelet Cytosol

Abstract:A G-protein beta gamma subunit (G beta gamma)-responsive phosphoinositide 3-kinase (PI 3-kinase) was purified approximately 5000-fold from pig platelet cytosol, The enzyme was purified by polyethylene glycol precipitation of the cytosol followed by column chromatography on Q-Sepharose fast flow, gel filtration, heparin-Sepharose, and hydroxyapatite. The major G beta gamma-responsive PI 3-kinase is distinct from p85 containing PI 3-kinase as the activities can be distinguished chromatographically and immunologically and is related to p110 gamma as it crossreacts with anti-p110 gamma-specific antibodies. The p110 gamma-related PI 3-kinase cannot be activated by G-protein alpha(i/o) subunits, and it has an apparent native molecular mass of 210 kDa, The p110 gamma-related PI 3-kinase phosphorylates phosphatidylinositol (PtdIns), phosphatidylinositol 4-phosphate (PtdIns4P), and phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P-2). The apparent K-m values for ATP were found to be 25 mu M with PtdIns, 44 mu M with PtdIns4P, and 37 mu M with PtdIns(4,5)P-2 as the substrate, G beta gamma subunits did not alter the K-m of the enzyme for ATP; however, V-max increased 2-fold with PtdIns as substrate, 3.5-fold with PtdIns4P, and 10-fold with PtdIns(4,5)P-2. Under basal conditions the apparent K-m values for lipid substrates were 64, 10, and 15 mu M for PtdIns, PtdIns4P, and PtdIns(4,5)P-2, respectively, In the presence of G beta gamma subunits the dependence of PI 3-kinase activity on the concentrations of lipid substrates became complex with the highest level of stimulation occurring at high substrate concentration, suggesting that the binding of G beta gamma and lipid substrate (particularly PtdIns(4,5)P-2) may be mutually cooperative, Wortmannin and LY294002 inhibit the G beta gamma-responsive PI 3-kinase activity with IC50 values of 10 nM and 2 mu M, respectively, Unlike the p85 containing PI 3-kinase in platelets, the p110 gamma-related PI 3-kinase is not associated with a PtdIns(3,4,5)P-3 specific 5-phosphatase.The p85-associated PI 3-kinase was not activated by G beta gamma alone but could be synergistically activated by G beta gamma and phosphotyrosyl platelet-derived growth factor receptor peptides, This may represent a form of coincidence detection through which the effects of tyrosine kinase and G-protein-linked receptors might be coordinated.

Abstract:Spinach (Spinacia oleracea L.) leaf sucrose-phosphate synthase (SPS) can be inactivated by phosphorylation of Ser-158 by calmodulin-like domain protein kinases (CDPKs) or SNF1-related protein kinases (SnRK1) in vitro. While the phosphorylation site sequence is relatively conserved, most of the deduced sequences of SPS from dicot species surrounding the Ser-158 regulatory phosphorylation site contain a Pro residue at P-4 (where P is the phosphorylated Ser); spinach is the exception and contains an Arg at P-4. We show that a Pro at P-4 selectively inhibits phosphorylation of the peptide by a CDPK relative to a SnRK1. The presence of a Pro at P-4, by allowing a tight turn in the peptide substrate, may interfere with proper binding of residues at P-5 and beyond. Both kinases had greater activity with peptides having basic residues at P-6 and P+5 (in addition to the known requirement for an Arg at P-3/P-4), and when the residue at P-6 was a His, the pH optimum for phosphorylation of the peptide was acid shifted. The results are used to predict proteins that may be selectively phosphorylated by SnRK1s (as opposed to CDPKs), such as SPS in dicot species, or may be phosphorylated in a pH-dependent manner.
Partial purification and characterization of a protein inhibitor of phosphoenolpyruvate carboxylase kinase

Abstract:Abstract. The activity of phosphoenolpyruvate carboxylase (PEPCase) kinase in leaf extracts increased markedly on dilution. This was shown to be caused by the presence of a protein that inhibits the kinase. The inhibitor protein was separated from the kinase and purified partially. It inhibited the kinase reversibly, presumably by a direct interaction; it was neither a protease nor a protein phosphatase. The amounts of kinase and inhibitor in leaves were estimated following separation by hydrophobic chromatography. The amount of inhibitor in the crassulacean acid metabolism plant Kalanchoë fedtschenkoi Hamet et Perrier was sufficient to inhibit the basal level of kinase activity present during the light period and the early stages of the dark period. Similarly, the amount of inhibitor in the C4 plant Zea mays L.was sufficient to inhibit the low amount of kinase activity present in the dark and at moderate light intensity. Analogous to the role of the protein inhibitor of mammalian cyclic AMP-dependent protein kinase, the function of the PEPCase kinase inhibitor may be to inhibit the basal level of kinase present in conditions under which rapid flux through PEPCase is not required.

Subcellular compartmentation of pyrophosphate and alkaline pyrophosphatase in leaves

Abstract:The subcellular localisation of pyrophosphate and alkaline pyrophosphatase in leaves has been studied using non-aqueous density gradient centrifugation of spinach leaves, and membrane filtration of wheat mesophyll protoplasts. The pyrophosphate was measured in extracts prepared in trichloroacetic acid, and could be quantitatively recovered from the leaf material. It was located predominantly in the cytosol, with a concentration of 0.2–0.3 mM. In contrast, the alkaline pyrophosphatase was largely, if not, exclusively, located in the chloroplast. By comparing the pyrophosphate levels in the cytosol with previously published data on the cytosolic levels of phosphate and metabolic intermediates, it is shown that the reactions catalysed by pyrophosphate: fructose-6-phosphate phosphotransferase and UDP-glucose pyrophosphorylase are close to the thermodynamic equilibrium and, thus, freely reversible in vivo. Comparison of the pyrophosphate levels with the reported electrical and pH gradient across the tonoplast membrane shows the free energy released during pyrophosphate hydrolysis is similar to that required to move a proton across the tonoplast membrane. It is suggested that pyrophosphate could operate as a secondary energy donor in the cytosol of plant cells.
Isolation and Expression Analysis of Fructokinase Genes from Citrus

Abstract:Fructokinase (FRK) is of primary importance in phosphorylation of fructose in plants. Two genomic DNA fragments encoding putative fructokinase gene were isolated from Citrus unshiu Marc. using PCR, which were named Cufrk1 and Cufrk2. A cDNA sequence same to the exons' sequences of Cufrk1 was amplified by RT-PCR and the full-length cDNA sequences of this gene was isolated using rapid amplification of cDNA ends (RACE) from mature fruit, named CuFRK1 (GenBank number: AY561840). Sequencing analysis showed that the amino acid sequences were 68% identical between Cufrk1 and Cufrk2. The full-length cDNA of CuFRK1 was 1 459 bp in length and contained a complete ORF from 168 to 1 220 bp, encoding 350 amino acids with a calculated molecular weight of 37.5 kD and a predicted pI of 5.03. The deduced amino acids of CuFRK1 possessed two sugar-binding domains, three ATP-binding domains and were 62% to 78% identical to the previously characterized fructokinase genes of other plants. Northern analysis showed that transcripts of CuFRK1 (Cufrk1) and Cufrk2 were detected at a high level in leaves, fruit at early developmental stages, but undetectable in peels and stems. Their expression patterns were distinct in petals and mature fruit. Enzymatic analysis showed that the activity of citrus fructokinase was decreasing with the fruit development, coincided with the accumulation of fructose in fruit. There was a significantly negative correlation between the decreasing fructokinase activity and the increasing fructose content during fruit development.

Abstract:Fructokinase (FRK) (ATP: D -fructose 6-phosphotransferase; EC 2.7.1.4) is a key enzyme in fructose metabolism in the sink organs of plants. In developing potato ( Solanum tuberosum L.) tubers, fructose released by sucrose synthase (SuSy)-mediated sucrolytic pathway is converted to fructose-6-phosphate by FRKs. In plants, FRK isoforms are encoded by the nuclear multigene families. Here, we report the isolation of a 1110-bp cDNA clone encoding a 319-aa FRK2 isoform (designated StFRK2) from an Indian potato cultivar, Kufri Chipsona-1 by RT-PCR using tuber total RNA. Two more cDNAs encoding a 256-aa FRK2 variant and a 266-aa FRK-like protein were isolated using total RNA from tuber and leaf, respectively. Six more potato FRK sequences designated StFRK2A, StFRK2B, StFRK1, StFRK3, StFRK4A and StFRK4B comprising 319, 329, 347, 389, 385, 375 amino acids, respectively were retrieved from the published reports/databases and examined. All these available potato FRKs could be categorized into 4 potentially active isoforms i.e., FRK1-4 as reported in tomato. Phylogenetic analysis was carried out using mostly the FRK sequences from the Solanaceae family. Sequence alignments with the catalytic motifs, crucial amino acids and secondary structures were presented; 3-D structures were predicted. Gene-specific expression patterns were shown. Significantly higher level of FRK activities were noticed during the early stages of tuber development as compared with the other organs. Aqueous two-phase system purified tuber extract could show FRK activity and allowed to detect a ~ 35 kDa protein band by denaturing SDS-PAGE. This is a comprehensive report on the FRK isoforms in potato and other Solanaceae family members.

Purification and properties of inosine-guanosine phosphorylase from Escherichia coli K-12

Abstract:A xanthosine-inducible enzyme, inosine-guanosine phosphorylase, has been partially purified from a strain of Escherichia coli K-12 lacking the deo-encoded purine nucleoside phosphorylase. Inosine-guanosine phosphorylase had a particle weight of 180 kilodaltons and was rapidly inactivated by p-chloromercuriphenylsulfonic acid (p-CMB). The enzyme was not protected from inactivation by inosine (Ino), 2'-deoxyinosine (dIno), hypoxanthine (Hyp), Pi, or alpha-D-ribose-1-phosphate (Rib-1-P). Incubating the inactive enzyme with dithiothreitol restored the catalytic activity. Reaction with p-CMB did not affect the particle weight. Inosine-guanosine phosphorylase was more sensitive to thermal inactivation than purine nucleoside phosphorylase. The half-life determined at 45 degrees C between pH 5 and 8 was 5 to 9 min. Phosphate (20 mM) stabilized the enzyme to thermal inactivation, while Ino (1 mM), dIno (1 mM), xanthosine (Xao) (1 mM), Rib-1-P (2 mM), or Hyp (0.05 mM) had no effect. However, Hyp at 1 mM did stabilize the enzyme. In addition, the combination of Pi (20 mM) and Hyp (0.05 mM) stabilized this enzyme to a greater extent than did Pi alone. Apparent activation energies of 11.5 kcal/mol and 7.9 kcal/mol were determined in the phosphorolytic and synthetic direction, respectively. The pH dependence of Ino cleavage or synthesis did not vary between 6 and 8. The substrate specificity, listed in decreasing order of efficiency (V/Km), was: 2'-deoxyguanosine, dIno, guanosine, Xao, Ino, 5'-dIno, and 2',3'-dideoxyinosine. Inosine-guanosine phosphorylase differed from the deo operon-encoded purine nucleoside phosphorylase in that neither adenosine, 2'-deoxyadenosine, nor hypoxanthine arabinoside were substrates or potent inhibitors. Moreover, the E. coli inosine-guanosine phosphorylase was antigenically distinct from the purine nucleoside phosphorylase since it did not react with any of 14 monoclonal antisera or a polyvalent antiserum raised against deo-encoded purine nucleoside phosphorylase.

Relaxin activity in foaling mares.

Abstract:Plasma relaxin concentrations were measured hourly by radio immunoassay in 4 pregnant mares from 11 days before until 4 days after natural foaling. Pre-partum levels ranged from 4 to 7 ng/ml without any surge until the second stage of labour when they increased rapidly to about 11 ng/ml. In 3 of these mares, relaxin activity declined immediately after the expulsion of the placenta and was below detectable levels within 36 h. In the other mare relaxin activity did not fall until after the mechanical removal of the placenta 7 h after foaling. Eight mares were induced to foal by the administration of oxytocin. In 5 out of 6 mares induced with a single i.v. injection of 40 i.u. plasma relaxin increased following injection and peaked at 17 ng/ml shortly after foaling and expulsion of placenta. Activity then rapidly declined. In the 6th mare, which retained the placenta, relaxin activity did not decrease after foaling. Administration of more oxytocin to this mare after foaling resulted in a further increase in plasma relaxin activity, but oxytocin treatment of other mares after placental delivery failed to elicit a similar increase. In the 7th mare 2 i.v. injections of 40 i.u. oxytocin induced a relaxin concentration of 34 ng/ml whereas in the remaining mare one i.m. injection of 40 i.u. oxytocin led to a relaxin profile similar to that in naturally foaling mares. Oxytocin may therefore be responsible for the increased secretion rate of relaxin at foaling. These data support the concept that the placenta is the sole significant source of relaxin in the pregnant mare.
Purification and characterization of human erythrocyte pyridoxine kinase

Abstract:A 6000-fold purification of pyridoxine kinase from human erythrocytes has been achieved by a combination of DEAE-cellulose chromatography, ammonium sulfate fractionation, gel filtration and preparative disc polyacrylamide gel electrophoresis. Analytic disc polyacrylamide gel electrophoresis at pH 8.7 reveals two protein bands with similar mobility in the purified enzyme, but only one of these has catalytic activity. The enzyme is found to have a pI of 5.5 and a molecular weight of 65000 by gel filtration, and a broad pH optimum of 8.5. The enzyme is labile at acidic pH. The order of activation of divalent metal ions on the enzyme is Co-2+ greater than Mn-2+ greater than Mg-2+ greater than Zn-2+ greater than Cu-2+ greater than Ni-2+ greater than Fe-2+. Of the monovalent cations studied, K+ is the most effective activator, NH+4 is slightly less effective while Na+ is inhibitory. ATP is the specific phosphate donor for the enzyme. Sulfhydryl reagents did not significantly inhibit the enzyme. The Km for pyridoxine is 5.7 with 10-minus 6 M. Pyridoxal, pyridoxamine and 4-deoxypyridoxine inhibit pyridoxine kinase. Inhibition by pyridoxal is competitive and pyridoxal is observed to be phosphorylated effectively by the enzyme. Young human red cells have a higher activity of pyridoxine kinase than old red cells.
A role for PFK-2/FBPase-2, as distinct from fructose 2,6-bisphosphate, in regulation of insulin secretion in pancreatic beta-cells

Abstract:PFK-2/FBPase-2 (6-phosphofructo-2-kinase/fructose 2,6-bisphosphatase) catalyses the formation and degradation of fructose 2,6-P(2) (fructose 2,6-bisphosphate) and is also a glucokinase-binding protein. The role of fructose 2,6-P(2) in regulating glucose metabolism and insulin secretion in pancreatic beta-cells is unresolved. We down-regulated the endogenous isoforms of PFK-2/FBPase-2 with siRNA (small interfering RNA) and expressed KA (kinase active) and KD (kinase deficient) variants to distinguish between the role of PFK-2/FBPase-2 protein and the role of its product, fructose 2,6-P(2), in regulating beta-cell function. Human islets expressed the PFKFB2 (the gene encoding isoform 2 of the PFK2/FBPase2 protein) and PFKFB3 (the gene encoding isoform 3 of the PFK2/FBPase2 protein) isoforms and mouse islets expressed PFKFB2 at the mRNA level [RT-PCR (reverse transcription-PCR)]. Rat islets expressed PFKFB2 lacking the C-terminal phosphorylation sites. The glucose-responsive MIN6 and INS1E cell lines expressed PFKFB2 and PFKFB3. PFK-2 activity and the cell content of fructose 2,6-P(2) were increased by elevated glucose concentration and during pharmacological activation of AMPK (AMP-activated protein kinase), which also increased insulin secretion. Partial down-regulation of endogenous PFKFB2 and PFKFB3 in INS1E by siRNA decreased PFK-2/FBPase-2 protein, fructose 2,6-P(2) content, glucokinase activity and glucoseinduced insulin secretion. Selective down-regulation of glucose-induced fructose 2,6-P(2) in the absence of down-regulation of PFK-2/FBPase-2 protein, using a KD PFK-2/FBPase-2 variant, resulted in sustained glycolysis and elevated glucose-induced insulin secretion, indicating an over-riding role of PFK-2/FBPase-2 protein, as distinct from its product fructose 2,6-P(2), in potentiating glucose-induced insulin secretion. Whereas down-regulation of PFK-2/FBPase-2 decreased glucokinase activity, overexpression of PFK-2/FBPase-2 only affected glucokinase distribution. It is concluded that PFK-2/FBPase-2 protein rather than its product fructose 2,6-P(2) is the over-riding determinant of glucose-induced insulin secretion through regulation of glucokinase activity or subcellular targeting.
Studies on the entry of fructose-2,6-bisphosphate into chloroplasts

Abstract:The regulatory metabolite fructose-2,6-bisphosphate (Fru-2,6-P(2)) has an important function in controlling the intermediary carbon metabolism of leaves. Fru-2,6-P(2) controls two cytosolic enzymes involved in the interconversion of fructose-6-phosphate and fructose-1,6-bisphosphate (fructose-1,6-bisphosphatase and pyrophosphate, fructose-6-phosphate 1-phosphotransferase) and thereby controls the partitioning of photosynthate between sucrose and starch. It has been demonstrated that Fru-2,6-P(2) is present mainly in the cytosol. Here we present evidence that Fru-2,6-P(2) can be taken up by isolated intact chloroplasts but at a very slow rate (about 0.01 micromoles per milligram of chlorophyll per hour). This uptake is time and concentration dependent and is inhibited by PPi. When provided a physiological concentration of Fru-2,6-P(2) (10 micromolar), chloroplasts accumulated up to 0.6 micromolar Fru-2,6-P(2) in the stroma. Elevated plastid Fru-2,6-P(2) levels had no effect on overall photosynthetic rates of isolated chloroplasts. The results indicate that, while Fru-2,6-P(2) enters isolated chloroplasts at a sluggish rate, caution should be exercised in ascribing physiological importance to effects of Fru-2,6-P(2) on chloroplast enzymes.

Purification of polysaccharides from Phellinus linteus by using an aqueous two-phase system and evaluation of the physicochemical and antioxidant properties of polysaccharides in vitro

Abstract:In this study, Phellinus linteus polysaccharides (PLPS) and proteins were simultaneously separated from P. linteus mycelia by using an aqueous two-phase system (ATPS) based on choline chloride ([Chol]Cl)/K2HPO4, and the physicochemical and antioxidant properties of PLPS after ATPS extraction were evaluated. Results demonstrated that the maximal extraction efficiencies of 68.53% ± 0.29% PLPS and 82.37% ± 0.41% proteins were obtained when the cholinium-based ATPS contained 68.9% K2HPO4, 20% [Chol]Cl, 10.0 mg mL−1 crude water extract (1.0 mL), and distilled water (4.0 mL) at shaking time and temperature of 30 min and 21.2 °C, respectively. Compared with C-PLPS obtained using traditional ethanol precipitation and isolation protocols, PLPS had higher carbohydrate content (63.58% ± 1.12%), lower molecular weight (15.2 kDa, 80%), different monosaccharide compositions, and showed similar preliminary structural characterizations. Moreover, PLPS exhibited more evident scavenging effects on free radicals and in vitro antioxidant activities than C-PLPS. Therefore, the method of [Chol]Cl/K2HPO4 ATPS can be developed as an effective strategy for the separation/purification of highly bioactive polysaccharides.

Path Planning for Altruistically Negotiating Systems: The Near-Sighted Tarzan Algorithm

Abstract:This paper introduces a variant of the Rapidlyexploring Random Leafy Tree, called the Near-sighted Tarzan Algorithm, that performs path planning (for message routing) in a discrete graph simulating the communication network of an Altruistically Negotiating System. The Near-sighted Tarzan Algorithm is designed to operate in environments with similar characterstics as an ad hoc network. These characteristics include an unordered topology, large quantities of agents, network connectivity, and limited knowledge of the network by any given agent. Comparison tests with Dijkstra’s algorithm as a benchmark show the Tarzan algorithm to be superior when searching for paths between large distances. The Tarzan algorithm can be easily run in parallel making it suitable for use in an Altruistically Negotiating System. Abstract:Inulin is widely used as a prebiotic and emerging as a priming compound to counteract plant diseases. We isolated inulin-degrading strains from the lettuce phyllosphere, identified as Bacillus subtilis and Priestia megaterium, species hosting well-known biocontrol organisms. To better understand their varying inulin degradation strategies, three intracellular β-fructofuranosidases from P. megaterium NBRC15308 were characterized after expression in Escherichia coli: a predicted sucrose-6-phosphate (Suc6P) hydrolase (SacAP1, supported by molecular docking), an exofructanase (SacAP2), and an invertase (SacAP3). Based on protein multiple sequence and structure alignments of bacterial glycoside hydrolase family 32 enzymes, we identified conserved residues predicted to be involved in binding phosphorylated (Suc6P hydrolases) or nonphosphorylated substrates (invertases and fructanases). Suc6P hydrolases feature positively charged residues near the structural catalytic pocket (histidine, arginine, or lysine), whereas other β-fructofuranosidases contain tryptophans. This correlates with our phylogenetic tree, grouping all predicted Suc6P hydrolases in a clan associated with genomic regions coding for transporters involved in substrate phosphorylation. These results will help to discriminate between Suc6P hydrolases and other β-fructofuranosidases in future studies and to better understand the interaction of B. subtilis and P. megaterium endophytes with sucrose and/or fructans, sugars naturally present in plants or exogenously applied in the context of defense priming.

Abstract:To measure the kinetic properties of photorespiratory enzymes, it is necessary to work with purified proteins. Protocols to purify photorespiratory enzymes from leaves of various plant species require several time-consuming steps. It is now possible to produce large quantities of recombinant proteins in bacterial cells. They can be rapidly purified as histidine-tagged recombinant proteins by immobilized metal affinity chromatography using Ni2+-NTA-agarose. This chapter describes protocols to purify several Arabidopsis thaliana His-tagged recombinant photorespiratory enzymes (phosphoglycolate phosphatase, glycolate oxidase, and hydroxypyruvate reductase) from Escherichia coli cell cultures using two bacterial strain-plasmid systems: BL21(DE3)-pET and LMG194-pBAD.
