Abstract:The reproductive barriers that prevent gene flow between closely related species are a major topic in evolutionary research. Insect clades with parasitoid lifestyle are among the most species‐rich insects and new species are constantly described, indicating that speciation occurs frequently in this group. However, there are only very few studies on speciation in parasitoids. We studied reproductive barriers in two lineages of Lariophagus distinguendus (Chalcidoidea: Hymenoptera), a parasitoid wasp of pest beetle larvae that occur in human environments. One of the two lineages occurs in households preferably attacking larvae of the drugstore beetle Stegobium paniceum ("DB‐lineage"), the other in grain stores with larvae of the granary weevil Sitophilus granarius as main host ("GW‐lineage"). Between two populations of the DB‐lineage, we identified slight sexual isolation as intraspecific barrier. Between populations from both lineages, we found almost complete sexual isolation caused by female mate choice, and postzygotic isolation, which is partially caused by cytoplasmic incompatibility induced by so far undescribed endosymbionts which are not Wolbachia or Cardinium. Because separation between the two lineages is almost complete, they should be considered as separate species according to the biological species concept. This demonstrates that cryptic species within parasitoid Hymenoptera also occur in Central Europe in close contact to humans.

Abstract:Abstract Ontsira mellipes Ashmead is a gregarious larval ectoparasitoid of woodboring cerambycids that is native to North America but can readily attack the exotic Asian longhorned beetle, Anoplophora glabripennis (Motschulsky). To evaluate the potential of the parasitoid as a novel association control agent for the pest beetle, this study investigated some key reproductive traits of the parasitoid, including egg maturation dynamics, and host size preference and suitability in association with the beetle. Results showed that female wasps emerged with a substantial portion (38%) of their lifetime complement of mature eggs and matured eggs rapidly, reaching a peak 4–6 d post-eclosion. The number of mature eggs was positively related to the female wasp’s body size. Oviposition prompted production of more mature eggs by young female wasps. The parasitoid did not show a significant preference for large over small hosts in a choice test. Host size did not affect the parasitoid’s offspring survival, developmental time, or sex ratio. However, clutch size increased with increasing host size. Female wasps that developed from large hosts had larger body size and consequently a higher mature egg load than those reared from small hosts. Neither longevity nor the total number of parasitized hosts over a female’s lifetime was affected by the female’s size, but the total number of offspring produced per female increased with the female’s size. These results have important implications for improving rearing and field-release strategies as well as understanding the ecological mechanisms underlying host size selection in gregarious parasitoids.

Abstract:The solitary endoparasitoid Aenasius vexans Kerrich (Hymenoptera: Encyrtidae) is used for augmentative releases against the cassava mealybug, Phenacoccus herreni Cox & Williams (Sternorrhyncha: Pseudococcidae), an important pest on cassava in South America. In light of the need for large numbers of high quality females, experiments were conducted on host stage suitability and sex allocation. In choice and no‐choice experiments, individual female wasps were offered second and third instar, as well as adult, hosts. During the first five days after emergence, the wasps showed a steady increase in the number of hosts they successfully parasitised per day, but the respective secondary sex ratio for each instar remained constant. Parasitism was highest for third instar hosts in no‐choice tests, while in choice tests parasitism was highest in both third instars and adults. The later the developmental stage of the host at oviposition, the faster the parasitoids developed and emerged, and for each host stage, the development time of males was shorter than for females. The sex ratio of the wasps emerging from hosts that were parasitised as second instars was strongly male‐biased, while the apparently preferred later stages yielded significantly more females than males. Female and male A. vexans emerging from hosts parasitised at the third instar were significantly larger than for the other stages. This may explain the preference for the third instar as well as the female‐biased sex ratio, as size is usually positively correlated with higher fitness, especially in females. The results suggest that third instar hosts are the most suitable for rearing high numbers of large females.

Abstract:Sex pheromones are crucial for mate finding in many animals. Long-range attraction, mate recognition, and the elicitation of sexual receptiveness during courtship are typically mediated by different compounds. It is widely unknown, however, how the different components of a species’ pheromone system influence each other. Here, we demonstrated in the parasitoid wasp Nasonia vitripennis that females quickly cease to respond to the male sex attractant after they contact a male’s oral secretion during courtship. We used this behavioral switch to monitor the fractionation of head extracts from male wasps for identification of the bioactive compounds as a blend of ethyl oleate, ethyl linoleate, and ethyl α-linolenate. This is the first identification of a cephalic courtship pheromone in parasitic Hymenoptera. Plasticity in pheromone-mediated sexual behavior of female insects has hitherto been attributed to the transfer of bioactive proteinaceous molecules with the male ejaculate. The pheromone interaction reported here sheds new light on the sexual communication of insects by showing that the sex pheromone response of females can be terminated by males independent of sperm transfer.

Abstract:The suicide prevention gatekeeper training program QPR (Question, Persuade, and Refer) was evaluated among school personnel using a nonequivalent control group design. Substantial gains were demonstrated from pre- to post-test for attitudes, knowledge, and beliefs regarding suicide and suicide prevention. Exploratory analyses revealed the possible moderating effects of age, professional role, prior training, and recent contact with suicidal youth on QPR participants' general knowledge, questioning, attitudes toward suicide and suicide prevention, QPR quiz scores, and self-efficacy. The need for replication using a more rigorous experimental design in the context of strong community collaboration is discussed.
The Evolution of Queen Pheromone Production and Detection in the Reproductive Division of Labor in Social Insect Colonies

Abstract:Structurally diverse queen pheromones and fertility signals regulate the reproductive division of labor of social insects, such as ants, termites, some bees, and some wasps. The independent evolution of sociality in these taxa allows for the exploration of how natural history differences in sender and receiver properties led to the evolution of these complex communication systems. While describing the different effects and the structural diversity of queen pheromones, we identify two major syndromes that mostly separate ants and wasps from bees and termites in their use of different pheromone classes. We compare olfactory receptor evolution among these groups and review physiological and hormonal links to fecundity and pheromone production. We explore the cases in which queen pheromone evolution is conserved, convergent, or parallel and those in which queen pheromone responses are more likely to be learned or innate. More mechanistic information about the pathways linking fecundity to queen pheromone production and perception could help close major knowledge gaps.

Abstract:Attracting a mate at the right time is critical for many species that reproduce sexually. In insects, short-range communication between potential mates is often mediated by cuticular hydrocarbons (CHCs), which cover most of the insect cuticle. Although the CHC profiles of many insects have been studied, we know little about what aspects of a CHC profile cause changes in mate attractiveness over the lifetime of an individual. We addressed this question by studying the mason wasp Odynerus spinipes, whose females exhibit age-related quantitative changes in their CHC profile composition. First, we created an ethogram of the male mating behavior. We knew from preliminary investigations that males do not attempt to mate with recently eclosed adult females. By coating wasp dummies with different CHC extracts, we were able to show that the CHC profiles of 3-day-old females are indeed more attractive to males than those of 0-day-old females. The increased attractiveness of 3-day-old females compared to younger females was negatively correlated with the relative abundance of methyl-branched alkanes in the CHC profile of the females. These results, and the fact that the CHC profile of males is characterized by a high relative abundance of methyl-branched alkanes throughout the adult wasp lifetime, suggest that in O. spinipes, methyl-branched alkanes may act as anti-aphrodisiacs that reduce the harassment of females by males before the females are ready to mate.

Abstract:In honeybees, worker reproduction is mainly regulated by pheromones produced by the brood and the queen. The source of one of the queen pheromones influencing worker reproduction has been located in the mandibular glands. In nonlaying workers, this gland's profile is dominated by fatty acids that are incorporated into the food given to the brood and to nest mates. After queen loss and onset of reproductive activity, workers are able to synthesize different fatty acids, which are normally only produced by queens and that contribute to their reproductive success. Apis mellifera capensis workers have the ability to rapidly produce queen-like mandibular profiles that could represent an important factor in their ability to behave as facultative intraspecific social parasites. Indeed, A. m. capensis workers can take over reproduction from the host queens in colonies of other subspecies. Here, we show that in the presence of their own queen, the mandibular gland profile of A. m. capensis workers is dominated by the precursor of the major compound of the queen pheromone. This is a unique trait among honeybee workers and suggests that A. m. capensis workers are primed for reproduction and that this phenomenon represents a pheromonal predisposition to social parasitism. We identified geographical variation in the ratio of queen-to worker-specific compounds in themandibular gland profile of A. m. capensis workers, which corresponds with the introgression with the neighboring subspecies A. m. scutellata.

Abstract:The use of chemical cues and signals is essential for communication in insects. Wasps of the genus Nasonia (Hymenoptera, Pteromalidae) are gregarious parasitoids that lay their eggs into puparia of cyclorrhaphous flies. During their life cycle, various kinds of semiochemicals are used: (1) a male abdominal sex pheromone that attracts females and induces site fidelity in males, (2) a female-derived contact sex pheromone eliciting courtship behavior in males, (3) an oral male aphrodisiac eliciting receptivity signaling in females and causing a switch in the females' olfactory preferences, (4) chemicals derived from host habitat and host puparia used in olfactory host finding by female wasps, and (5) chemicals used by females to assess the quality and parasitization status of potential hosts. We review the literature on the chemical ecology of Nasonia spp. following the wasps' life cycle from emergence to oviposition. We depict biosynthetic pathways where available, discuss ecological implications, highlight differences among Nasonia species, summarize insights into their olfactory perception and associative learning abilities, and point out gaps in our understanding of the chemical ecology of these parasitoids to be addressed in future studies.

Sex pheromone communication in an insect parasitoid, Campoletis chlorideae Uchida

Abstract:Sex pheromones are pivotal for insect reproduction. However, the mechanism of sex pheromone communication remains enigmatic in hymenopteran parasitoids. Here we have identified the sex pheromone and elucidated the olfactory basis of sex pheromone communication in Campoletis chlorideae (Ichneumonidae), a solitary larval endoparasitoid of over 30 lepidopteran pests. Using coupled gas chromatography-electroantennogram detection, we identified two female-derived pheromone components, tetradecanal (14:Ald) and 2-heptadecanone (2-Hep) (1:4.6), eliciting strong antennal responses from males but weak responses from females. We observed that males but not females were attracted to both single components and the blend. The hexane-washed female cadavers failed to arouse males, and replenishing 14:Ald and 2-Hep could partially restore the sexual attraction of males. We further expressed six C. chlorideae male-biased odorant receptors in Drosophila T1 neurons and found that CchlOR18 and CchlOR47 were selectively tuned to 14:Ald and 2-Hep, respectively. To verify the biological significance of this data, we knocked down CchlOR18 and CchlOR47 individually or together in vivo and show that the attraction of C. chlorideae to their respective ligands was abolished. Moreover, the parasitoids defective in either of the receptors were less likely to court and copulate. Finally, we show that the sex pheromone and ( Z )-jasmone, a potent female attractant, can synergistically affect behaviors of virgin males and virgin females and ultimately increase the parasitic efficiency of C. chlorideae . Our study provides new insights into the molecular mechanism of sex pheromone communication in C. chlorideae that may permit manipulation of parasitoid behavior for pest control.

Abstract:The cuticle of virtually any insect is covered by a thin layer of cuticular hydrocarbons (CHCs) typically consisting of a complex mixture of n-alkanes, methyl-branched alkanes, and unsaturated hydrocarbons. Apart from their putative primary function as a desiccation barrier, CHCs are used by insects for communication. In many species of parasitoid wasps, males use CHCs to recognize females, and thus the composition of CHC profiles is typically species- and sex-specific. Sometimes, the biological activity of CHCs is synergized by more polar lipids. In most species, however, the contribution of polar lipids and the role of individual CHCs or CHC classes is unclear. Here we report a CHC-based contact sex pheromone in the pteromalid wasp Urolepis rufipes. Males of U. rufipes were arrested on and showed courtship behavior (wing fanning) and copulation attempts toward cadavers of females, whereas male cadavers and solvent-washed cadavers of either sex elicited no responses. Whole body extracts from females and the non-polar CHC fraction thereof elicited arrestment, courtship, and copulation attempts by males, whereas the fractions containing polar lipids were behaviorally inactive. Analyses of male- and female-derived CHC profiles revealed that they consisted exclusively of n-alkanes and methyl-branched alkanes. Removal of n-alkanes from female-derived CHCs with molecular sieves rendered the remaining methyl-branched fraction inactive. Synthetic n-alkanes in female-specific ratios also did not elicit any responses by males. Two 7-methylalkanes (7-MeC31 and 7-MeC33) were the only two components with a higher relative abundance in females compared to males. However, supplementation of male-derived CHCs with synthetic racemic 7-MeC31 and 7-MeC33 to levels found in females was not sufficient to render male-derived CHCs attractive to males. Enantiopure synthetic 7-MeC31 and 7-MeC33 might have caused different effects but were not available. We conclude that female-derived CHCs function as a contact sex pheromone in U. rufipes. Our data suggest that methyl-branched and n-alkanes act synergistically and that the sex-specific relative composition of the whole profile, rather than the abundances of single components, underlies the elicitation of male courtship behavior.

Abstract:Abstract The iconic European stag beetle ( Lucanus cervus ) (Coleoptera: Lucanidae) is one of the largest terrestrial beetles in Europe. Due to decreasing population numbers, thought to be a consequence of habitat loss, this beetle has become a near-threatened species across much of Europe, and a reliable monitoring system is required to measure its future population trends. As part of a programme aimed at conserving UK populations, we have investigated the chemical ecology of the beetle, with a view to developing an efficient semiochemical-based monitoring system. Such a scheme will be beneficial not only in the UK but across the European range of the species, where the beetle is of conservation concern. Here, we report on a surprising discovery of a male-produced pheromone, which provokes initial sexual receptivity in females, and which has not been previously identified in the animal kingdom. Furthermore, we assign sex pheromone function to a previously described female-specific compound.

Abstract:Abstract Parasitoid wasps are among the most speciose animals, yet have relatively few available genomic resources. We report a draft genome assembly of the wasp Diachasma alloeum (Hymenoptera: Braconidae), a host-specific parasitoid of the apple maggot fly Rhagoletis pomonella (Diptera: Tephritidae), and a developing model for understanding how ecological speciation can “cascade” across trophic levels. Identification of gene content confirmed the overall quality of the draft genome, and we manually annotated ∼400 genes as part of this study, including those involved in oxidative phosphorylation, chemosensation, and reproduction. Through comparisons to model hymenopterans such as the European honeybee Apis mellifera and parasitoid wasp Nasonia vitripennis, as well as a more closely related braconid parasitoid Microplitis demolitor, we identified a proliferation of transposable elements in the genome, an expansion of chemosensory genes in parasitoid wasps, and the maintenance of several key genes with known roles in sexual reproduction and sex determination. The D. alloeum genome will provide a valuable resource for comparative genomics studies in Hymenoptera as well as specific investigations into the genomic changes associated with ecological speciation and transitions to asexuality.

Plasticity in life features, parasitism and super-parasitism behavior of Bracon hebetor, an important natural enemy of Galleria mellonella and other lepidopteran host species

Abstract:The wasp, Bracon hebetor Say, is an important potential biocontrol agent of a wide range of lepidopteran insect species. The current study was subjected to compare these ectoparasitoid fitness traits on different host species belonging to the order Lepidoptera. Moreover, we determine the population dynamic with different host densities, sex ratio patterns, super-parasitism, longevity, paralysis success, and reproductive potential under laboratory conditions. Our results revealed that oviposition increased with an increase in host density, while the adult emergence and egg hatching were decreased due to the super-parasitism on host larvae. A higher male and female-biased population were observed when virgin and mated females offered fresh hosts. Adults' longevity was recorded more in females than males when kept only with bee honey + royal jelly + host larvae. The mean duration of egg-adult development was recorded higher on Galleria mellonella and lowest on Busseola fusca. The parasitization rate, super-parasitism, and cumulative fecundity of mated and virgin female wasps with different host species were observed higher on specific host Galleria mellonella while it was lower on Phthorimaea operculella. Furthermore, the parasitoids having mating experience preferred fresh, while the parasitoids' lack of mating experiences preferred paralyzed host under olfactometer test. Besides, this research has produced novel facts on the biology of parasitic wasp, B. hebetor that may guide the advancement of sustainable biological control programs to control lepidopteran pests.
A scent shield to survive: identification of the repellent compounds secreted by the male offspring of the cuckoo bumblebee Bombus vestalis

Abstract:Abstract In social insects, the high cost of parental investment has promoted the evolution of traits that reduce or avoid care for unrelated offspring. They have evolved a sophisticated recognition system which enables them to reject foreign individuals, and therefore prevent their nest's exploitation by parasites and predators. Nonetheless, many social parasites are able to overcome social insect recognition and parasitize their parental care behaviour by laying their eggs in host nests. Although several studies have already been conducted on the strategies used by social parasites to successfully invade host nests, the strategy used by its offspring to evade host rejection is still poorly understood. In bumblebees, previous investigations indicated that male offspring of B ombus vestalis ( G eoffroy) (Hymeno‐ptera: A pidae), the specific obligate social parasite of B ombus terrestris (L.), may use allomones to evade host worker attacks. Following these studies, we performed electrophysiological studies and behavioural experiments to identify the potential repellents. Six compounds elicited electroantennographic responses in host worker antennae. The corresponding mixture of synthetic volatiles as well as pure tetradecyl acetate, and to a lesser extent pure ( Z )‐11‐hexadecenyl acetate, showed a repellent effect on starved host workers in a feeding bioassay. We hypothesize that tetradecyl acetate has a key function in the repellent effect of the secretions of B . vestalis male offspring towards B . terrestris host workers.

Alternative mating strategies in the Wellington tree weta expressed via genetic polymorphism for precocial male maturation

Abstract:The expression of alternative mating strategies is generally hypothesized to be a plastic response of the genome to the environment or a genetic polymorphism. The Wellington tree weta ( Hemideina crassidens ) is an orthopteran insect in which males express three alternative mating morphotypes related to the size of mandibular weaponry. My common garden experiment in the laboratory shows that male weapon size at maturity in this species is nether condition- nor environment-dependent; therefore, supporting the hypothesis that it is genetically polymorphic. Males that matured at the 8th instar had the shortest development time and the smallest weaponry, males that matured at the 10th instar had the longest development time and the largest weaponry, and males that matured at the 9th instar were intermediate to the other two morphs. These morph-related differences in development time suggest that male alternative mating strategies in this species are expressed via precocial male maturation, a rare mode of strategy expression in insects. Additionally, adult lifespans differed significantly among morphs with 8th instar males having the shortest, and 9th instar males having the longest lifespans. I discuss how these differences in lifespan align with morph-specific investment in pre- and post-copulatory processes to equalize fitnesses among morphs.

Polyandry, genetic diversity and fecundity of emigrating beetles: understanding new foci of infestation and selection

Abstract:The frequency of polyandry and its ecological (and therefore selective) advantages remain unclear, especially in the field, where such tests are seldom conducted. We quantified the fecundity and level of polyandry in field populations of two stored grain beetles, Tribolium castaneum (Herbst) and Rhyzopertha dominica (F.), by comparing resident with dispersing individuals at common sites. Almost all females caught in flight had mated, with paternity analysis from 110 parent–offspring arrays revealing that most females had mated with more than one male (T. castaneum: 90.6, R. dominica: 70.2%). No difference in the extent of polyandry between beetles collected from grain in storage and beetles caught in flight was detected for either species, and the degree of polyandry did not impact overall fecundity. Levels of polyandry were similar to those in recently established (first generation) laboratory cultures of both species. Our results provide the first direct evidence of polyandry in wild populations of T. castaneum and R. dominica, and that females that have mated several times have the potential to produce offspring of multiple genotypes in the field. Females thus invade resource patches (whether previously colonised or not) with considerable genetic diversity to distribute across their offspring, an advantage in a newly colonised site because it increases the chances of her offspring establishing there. This advantageous consequence of polyandry to dispersing individuals, and the fact that most of the individual beetles sampled in the field had mated multiple times, provides justification as to why polyandry should not be viewed as an evolutionary paradox.

Abstract:Abstract A major hypothesis for the evolution of chemical signals is that pheromones arise from non-communicative precursor compounds. However, data supporting this hypothesis are rare, primarily because the original functions of the antecedent compounds often have been lost. A notable exception, however, is the parasitoid wasp species Leptopilina heterotoma , whose compound (−)-iridomyrmecin is used as a defensive secretion, a cue for females to avoid competition with con- and hetero-specific females, and as the primary component of the females’ sex pheromone. To better understand the evolution of sex pheromones from defensive compounds, we examined the chemical ecology of L. pacifica , the sister species of L. heterotoma . Here, we show that L. pacifica also produces a defensive secretion containing a species-specific mixture of mostly iridoid compounds. However, the composition of the secretion is more complex than in L. heterotoma , and iridomyrmecin is only a minor component. Moreover, in contrast to L. heterotoma , conspecific female competitors were not avoided by female subjects, and a role of the iridoids in the female sex pheromone of L. pacifica can be excluded, as only the females’ cuticular hydrocarbons (CHCs) resulted in the elicitation of courtship by males. Although closely related, the two sister species show substantial differences in the use of the defensive secretion for communicative purposes. Variation in pheromone usage in this genus still presents a conundrum, highlighting the need for additional studies to understand the selective forces shaping the evolution of pheromone composition.
