Abstract:Radionuclide therapy (RNT) stands out as a highly effective method for treating solid tumors. However, its therapeutic efficiency faces challenges due to the radioresistance of tumors, the limited penetration depth and intracellular deposition of rays in tumor tissue, which causes residue of living cancer cells. Herein, we report a novel approach by utilizing radionuclide 131 I-labelled polydopamine encapsulated gold nanoparticle co-loaded with the classical anticancer drug gemcitabine within a hydrogel formed from oxidized glucan and chitosan hydrochloride, combining RNT with chemotherapy for cancer treatment. Au, as a high Z element, is able to interact with shortrange /3-rays to emit bremsstrahlung and secondary charged particles which in turn increase the dose deposited in tumor cells. Simultaneously, gemcitabine is able to affect cell cycle redistribution, resulting in an increase in the radiosensitive cellular component of the cycle, and gemcitabine also inhibits the repair of radioactive damage to cellular DNA, which has a radiosensitizing effect. In both in vivo and vitro experiments, the injectable hydrogel demonstrates excellent biosecurity, stability in radionuclide labeling, and capabilities for single-photon emission computed tomography (SPECT) imaging. Compared analysis with single RNT revealed that combination therapy markedly inhibits the growth of triple-negative breast cancer. This integrated therapeutic strategy establishes an efficient tumor synergistic treatment platform, offering new avenues for advancing radionuclide therapy in the clinical treatment of cancer.

Abstract:The integration of theranostics, which combines diagnostics with therapeutics, has markedly improved the early detection of diseases, precise medication management, and assessment of treatment outcomes. In the realm of oncology, organoplatinum-based supramolecular coordination complexes (SCCs) that can coload therapeutic agents and imaging molecules have emerged as promising candidates for multimodal theranostics of tumors. To address the challenges of tumor-targeted delivery and multimodal theranostics for SCCs, this study employs a cell membrane cloaking strategy to fabricate biomimetic metallacage nanoparticles (MCNPs) with multimodal imaging capabilities and homologous targeting capabilities. Specifically, a photosensitizer molecule (BTTP) containing AIE-active groups was assembled into a metallacage of C-BTTP through Pt-N coordination. This process endows the metallacage with strong NIR-II fluorescence in the aggregated state and significantly superior ROS generation compared to that of the precursor ligand. After being encapsulated with F127, the MCNPs were further cloaked with U87 cancer cell membranes, creating biomimetic MCNPs that achieve tumor-targeting capabilities. Verified by in vitro and in vivo experiments, MCNPs enable multimodal imaging and initiate immunotherapy under photothermal and photodynamic stimulation, leading to synergistic antitumor effects. Furthermore, the evaluation of immunogenic cell death and dendritic cell maturation rate in U87 tumor-bearing mice confirmed the mechanism of photothermal and photodynamic synergistic immunotherapy. This study provides an innovative strategy for enhancing the tumor-targeting and therapeutic efficiency of SCCs, offering a versatile strategy for efficient and minimally invasive theranostics of tumors. The development of such biomimetic nanoparticles represents a significant advancement in the field of nanomedicine, potentially transforming cancer treatment through personalized and targeted therapies.
MOF‐Coated Upconversion Nanoparticle Agents Enable Synergistic Photodynamic Therapy and Immunotherapy

Abstract:Although conventional therapeutic approaches demonstrate significant success in treating primary tumors, their efficacy diminishes when addressing distant and metastatic tumors, calling for new technologies to transport and activate the therapeutic agents. Here, a core-shell UCNP@MOF (upconversion nanoparticle - metal organic framework) nanoagents is constructed for Type I and Type II photodynamic therapy under biocompatible near-infrared light irradiance (at 808 nm) and utilize their pore loading capacity to further realize synergistic immunotherapy in solid tumors. The Cu2+ to Cu+ reduction at the nodes in the MOFs shell is demonstrated to promote the generation of reactive oxygen species via depletion of overexpressed glutathione (GSH) compounds, while tumor microenvironment-enriched hydrogen peroxide is found to substantially augment the PDT effects. Importantly, the MOFs pore-loaded indoleamine 2,3-dioxygenase (IDO) inhibitor, along with s-nitroso-n-acetylpenicillamine (SNAP), upregulated tumor-infiltrating lymphocytes and induced prominent immunotherapeutic effects in vivo. This core-shell UCNP@MOF nanoagents have demonstrated effectiveness against both primary and distant tumors in xenografted tumor models, holding promise for treating solid tumors in metastasis. Upconversion Nanoparticles Intercalated in Large Polymer Micelles for Tumor Imaging and Chemo/Photothermal Therapy

Abstract:Frontiers in theranostics are driving the demand for multifunctional nanoagents. Upconversion nanoparticle (UCNP)-based systems activated by near-infrared (NIR) light deeply penetrating biotissue are a powerful tool for the simultaneous diagnosis and therapy of cancer. The intercalation into large polymer micelles of poly(maleic anhydride-alt-1-octadecene) provided the creation of biocompatible UCNPs. The intrinsic properties of UCNPs (core@shell structure NaYF4:Yb3+/Tm3+@NaYF4) embedded in micelles delivered NIR-to-NIR visualization, photothermal therapy, and high drug capacity. Further surface modification of micelles with a thermosensitive polymer (poly-N-vinylcaprolactam) exhibiting a conformation transition provided gradual drug (doxorubicin) release. In addition, the decoration of UCNP micelles with Ag nanoparticles (Ag NPs) synthesized in situ by silver ion reduction enhanced the cytotoxicity of micelles at cell growth temperature. Cell viability assessment on Sk-Br-3, MDA-MB-231, and WI-26 cell lines confirmed this effect. The efficiency of the prepared UCNP complex was evaluated in vivo by Sk-Br-3 xenograft regression in mice for 25 days after peritumoral injection and photoactivation of the lesions with NIR light. The designed polymer micelles hold promise as a photoactivated theranostic agent with quattro-functionalities (NIR absorption, photothermal effect, Ag NP cytotoxicity, and Dox loading) that provides imaging along with chemo- and photothermal therapy enhanced with Ag NPs.

Abstract:Cortical necrosis commonly manifests clinically as a rapid loss of glomerular filtration rate, often with oliguria, granular casts, and low-level proteinuria. Cortical necrosis may occur at any age. Causes include complications of vascular anastomosis in the transplanted kidney. In the native kidney, it occurs after thrombosis of interlobular or larger arteries, massive cholesterol emboli, septic abortion, or other catastrophic obstetrical complications (including eclampsia), and in the neonate, it occurs as complications of sepsis and placental hemorrhage. Causes of native kidney cortical necrosis typically affect both kidneys. Prognosis depends on the underlying cause. Light microscopy: There is coagulative necrosis involving all tubular segments and glomeruli. Nuclei may be pale and pyknotic, or may no longer be apparent. Thrombi may be present in vessels at the edge of the infarct. Immunofluorescence microscopy: No specific staining. Electron microscopy: Widespread necrosis.
Radiolabeled Human Protein-Functionalized Upconversion Nanoparticles for Multimodal Cancer Imaging

Abstract:Multimodal imaging removes the intrinsic limitations of individual modalities, while the multipurpose nanoagents reduce the toxicity associated with the use of multiple contrast agents. However, limited functionality, inefficient or incompatible targeting, and unknown biodistribution are significant drawbacks for such complicated nanodesigns. We report a biofriendly pentamodal imaging agent with the synergistic dual-targeting ability (passive and active) based on fluoromagnetic NaGdF4:Yb,Er upconverting nanoparticles (UCNPs). Besides the nano-enabled passive targeting, the apo-human serum transferrin protein (Tf) conjugation actively targets the transferrin receptors (TfR) over-expressed on different cancer cells. The radiolabel, 99mTc, chelates in the Tf core increase the radiolabeling efficiency (∼95%) and stability (∼100% for 24 h), enabling efficient single-photon emission computed tomography imaging. Deep-penetrating 980 nm excited UCNPs are inherently active luminescent and photothermal agents enabling fluorescence and photothermal imaging, respectively. The Gd in UCNPs produces a T1-weighted MRI signal, and the Yb helps in computed tomography (CT) as contrast agents. A pentamodal imaging agent (UCNP@Tf-99mTc) with synergistic dual-targeting ability is thus designed. In vitro studies demonstrated receptor-mediated endocytosis and minimal cytotoxicity of the UCNP@Tf-99mTc nanoformulation (>75% cell viability at 1000 ppm). The in vivo biodistribution studies in the mice model under SPECT/CT revealed its near-ideal bioavailability and renal clearance behavior. The MRI investigation on 4 T1 tumor-bearing mice brought direct evidence of passive and synergistic passive-active targeted tumor accumulation of the nanoformulation. The in vivo multimodal tumor imaging (MRI/SPECT/thermal-camera/CT) demonstrates the potency of UCNP@Tf-99mTc as a single replacement for multiple imaging agents and a perfect candidate for the ultramodern Advanced Multimodality Image Guided Operating (AMIGO) suite.

Abstract:The easy recurrence and high metastasis of fatal tumors require the development of a combination therapy, which is able to overcome the drawbacks of monomodal strategies such as surgery, photodynamic therapy (PDT), and radiotherapy (RT). Taking the complementary advantages of PDT and RT, we present herein the integration of lanthanide-doped upconversion nanoparticles (UCNPs) with chlorin e6 (Ce6)-imbedded RBC membrane vesicles as a near-infrared-induced PDT agent for achieving synchronous depth PDT and RT with reduced radiation exposure. In such a nanoagent, gadolinium-doped UCNPs with strong X-ray attenuation ability act not only as a light transductor to activate the loaded photosensitizer Ce6 to allow PDT but also as a radiosensitizer to enhance RT. PDT with enhanced low-dose RT can achieve synergistic inhibition of tumor growth by producing reactive oxygen species to destroy local tumor cells and inducing strong T-cell-dependent immunogenic cell death to arrest systemic cancer metastasis. This combination of PDT and RT might be a potential appealing strategy for tumor eradication.

Abstract:Nuclear-targeted therapy by delivering anticancer drug directly into cancer cell nuclei can elicit synergistic therapeutic effects and kill these cancer cells with much enhanced efficiencies. Besides nuclear targeting, another difficulty in nuclear-targeted therapy is how to achieve real-time monitoring of the therapy process simultaneously. In this article we report on the development of multifunctional upconversion nanoparticles (UCNPs) which were able to target cancer cell nuclei, and thus deliver the anticancer drug directly to the nuclear region and simultaneously image cell nucleus by magnetic resonance (MR)/upconversion fluorescent for real-time guidance of their therapeutic action simultaneously. The Er/Yb-doped NaYF4 core and NaGdF4 shell endow the core/shell structured UCNPs with enhanced upconversion fluorescent imaging and more sensitive T1-MR imaging performances, and the surface conjugation of TAT peptide served as a key role in the nuclear targeting and nuclear transport process. This multifunctional UCNPs-based nano-theranostic was used to improve the efficacy of DOX in Hela humor tumor models, by direct DOX delivery to the nucleus under the synchronous monitoring of the nano-theranostics. Further development of this technology may provide more exciting opportunities in treating cancer disease by nuclear-targeted therapy.
Acidity-Activated Charge Conversion of 177 Lu-Labeled Nanoagent for the Enhanced Photodynamic Radionuclide Therapy of Cancer

Abstract:Nanomaterials in combination with radionuclide therapy (RNT) provide new opportunities for cancer treatment. However, nanomaterials with efficient tumor accumulation have been less exploited for effective radionuclide-based therapy. Here, we report glycol chitosan-based nanoparticles (GCP-NPs) with acidic pH-dependent surface charge conversion for efficient radionuclide-based combination therapy. The nanoplatform can change the surface charge of nanoparticles from slight negative to positive in the acidic tumor microenvironment, which facilitates cellular internalization and penetration and thus improves the tumor accumulation efficiency of nanomaterials. Radiolabeling of GCP-NPs with 99mTc enables in vivo radioactive imaging in the mouse subcutaneous tumor model, showing 8.1-fold enhanced tumor uptake relative to pH-insensitive control nanoparticles (termed as GCOP-NPs). Afterward, therapeutic radioisotope 177Lu-labeled GCP-NPs (177Lu-GCP-NPs) that utilize RNT synergistic with photodynamic therapy (PDT) derived from conjugated pyropheophorbide-a within nanoparticles endow superior antitumor efficacy in living cells and tumor-bearing mouse model. More importantly, the combination of RNT and PDT using 177Lu-GCP-NPs can effectively inhibit lung metastasis and eliminate splenomegaly, which is not possible for individual RNT or PDT. Therefore, this study proposes a facile radionuclide-based combination therapy strategy toward complete cancer remission.

Development of injectable upconversion nanoparticle‐conjugated doxorubicin theranostics electrospun nanostructure for targeted photochemotherapy in breast cancer

Abstract:Nanotheranostic‐based photochemotherapies with targeted drug delivery have considerably surfaced in cancer therapy. In the presented work, polyethyleneimine‐coated upconversion nanoparticles were engineered to conjugate covalently with doxorubicin. Upconversion nanoparticles (UCNP)‐Doxorubicin (DOX)/synthesized epidermal growth factor receptor‐targeting peptide blended with polymer composite was electrospun and formulated as the injectable dosage form. The size of the UCNP and the nanofiber diameter were assessed as 26.75 ± 1.54 and 162 ± 2.82 nm, respectively. The optimized ratio of dopants resulted in UCNP photoluminescence with maximum emission intensity at around 800 nm upon 980 nm excitation wavelength. The paramagnetic nature of UCNPs and amide conjugation with the drug was confirmed analytically. The loading capacity of UCNP for doxorubicin was determined to be 54.56%, while nanofibers exhibited 98.74% capacity to encapsulate UCNP‐DOX. The release profile of UCNP‐DOX from nanofiber formulation ranged from sustained to controlled, with relative enhancement in acidic conditions. The nanofiber demonstrated good mechanical strength, robust swelling, and degradation rate. Biocompatibility tests showed more than 90% cell viability on L929 and NIH/3T3 cell lines with UCNP‐DOX@NF/pep nanoformulation. The IC50 values of 2.15 ± 0.54, 2.87 ± 0.67, and 3.42 ± 0.45 μg/mL on MDA‐MB‐231, 4T1, and MCF‐7 cancer cell line, respectively, with a significant cellular uptake, has been reported. The UCNP protruded a ≈62.7°C temperature rise within 5 min of 980 nm laser irradiation and a power density of 0.5 W cm−2. The nanoformulation induced reactive oxygen species of 65.67% ± 3.21% and apoptosis by arresting the cell cycle sub‐G1 phase. The evaluation conveys the effectiveness of the developed injectable theranostic delivery system in cancer therapy.

Abstract:Nuclear medicine with radioisotopes is extremely useful for clinical cancer diagnosis, prognosis, and treatment. Herein, polyethylene glycol (PEG)-modified nanoscale coordination polymers (NCPs) composed of hafnium (Hf4+) and tetrakis (4-carboxyphenyl) porphyrin (TCPP) are prepared via a one-pot reaction. By chelation with the porphyrin structure of TCPP, such Hf-TCPP-PEG NCPs could be easily labeled with 99mTc4+, an imaging radioisotope widely used for single-photon emission computed tomography (SPECT) in a clinical environment. Interestingly, Hf, as a high- Z element in such 99mTc-Hf-TCPP-PEG NCPs, could endow nontherapeutic 99mTc with the therapeutic function of killing cancer cells, likely owing to the interaction of Hf with γ rays emitted from 99mTc to produce charged particles for radiosensitization. With efficient tumor retention, as revealed by SPECT imaging, our 99mTc-Hf-TCPP-PEG NCPs offer exceptional therapeutic results in eliminating tumors with moderate doses of 99mTc after either local or systemic administration. Importantly, those biodegradable NCPs could be rapidly excreted without much long-term body retention. Our work, showing the success of applying NCPs for radioisotope therapy (RIT), presents a potential concept for the realization of highly effective cancer treatment with 99mTc, a short-half-life (6.0 h) diagnostic radioisotope, which is promising for cancer RIT with enhanced efficacy and reduced side effects.
Y-86/90-based Theranostics of Breast Cancer in a Preclinical Model

Abstract:539 Objectives: Angiogenesis is one of the hallmarks of cancer, enabling increased tumor growth and metastasis. Therefore, imaging of angiogenesis may enable sensitive detection of tumors, and therapy targeted to angiogenic vessels can greatly reduce tumor burden of both primary and disseminated disease. CD105 is highly expressed on angiogenic vessels in a number of cancers, making it a widely-applicable target for both imaging and therapy with a chimeric CD105-targeted monoclonal antibody, TRC105. The use of the theranostic pair 86Y/90Y enables accurate tracking and therapy with the same chemical entity, Y-DTPA-TRC105. Methods: The chimeric anti-CD105 antibody, TRC105 (TRACON Pharmaceuticals) was conjugated with diethylenetriaminepentaacetic acid (DTPA) for radiolabeling with two isotopes of yttrium: 86Y (t1/2: 14.7 h, a positron emitter) and 90Y (t1/2: 64 h, a higher energy beta-emitter). Serial PET imaging with the positron-emitting 86Y-DTPA-TRC105 was conducted to map the biodistribution and investigate the pharmacokinetics of the tracer in 4T1 tumor-bearing mice. For therapeutic studies, four groups were employed: 90Y-DTPA-TRC105 (120 µCi, 45 µg), 90Y-DTPA-IgG (an isotype control, 120 µCi, 45 µg), TRC105 only (45 µg), and PBS only. Tumor growth and mouse body weight were monitored for 14 days. Blood analyses were performed in the treatment group at 0, 7, and 14 days post-injection, and histological analysis also compared tumors and main organs pre- and post-therapy. Cherenkov luminescence imaging was also performed for the 90Y-injected mice throughout the course of the study to qualitatively map the biodistribution of 90Y-DTPA-TRC105. Estimation of radiation doses from 90Y-DTPA-TRC105 was also enabled through the analysis of 86Y-DTPA-TRC105 PET data and OLINDA. Results: High uptake of 86Y-DTPA-TRC105 was found in 4T1 tumors, increasing over time (from 2.3 ± 0.6 %ID/g at 30 min p.i. to 9.1 ± 1.1 %ID/g at 48 h, n=4), with the liver showing the highest off-target accumulation, at 7.1 ± 0.9 %ID/g at 48 h p.i. Ex vivo biodistribution studies further verified this trend, with tumor uptake of 7.2 ± 0.41 %ID/g. Radiation extrapolation to an adult human female revealed a whole-body effective dose of 0.57 ± 0.02 mSv/MBq from 90Y-DTPA-TRC105, via analysis of the 86Y-DTPA-TRC105 PET data. Cherenkov imaging demonstrated persistent uptake of 90Y-DTPA-TRC105 in the 4T1 tumors throughout the entire study. In therapeutic studies, 90Y-DTPA-TRC105 was found to slow tumor growth relative to the control groups, with significantly smaller (p<0.05) tumor volumes at 10 days p.i. However, high doses to the liver (the clearance organ for antibody-based agents, 0.079 ± 0.008 mSv/MBq) limited the amount of radioactivity that could be administered. Histological analysis of ex vivo tissues revealed significant damage to the treated tumors; at the same time, noticeable damage was evident in the liver and clearance organs as well. Conclusions: The use of the theranostic pair of 86/90Y allows PET imaging and treatment of CD105-expressing angiogenic tumors. While promising therapeutic outcomes were achieved, the treatment with 90Y-DTPA-TRC105 requires further optimization to minimize off-target doses.
Plasmon-Triggered Upconversion Emissions and Hot Carrier Injection for Combinatorial Photothermal and Photodynamic Cancer Therapy

Abstract:Despite the unique ability of lanthanide-doped upconversion nanoparticles (UCNPs) to convert near-infrared (NIR) light to high-energy UV-vis radiation, low quantum efficiency has rendered their application unpractical in biomedical fields. Here, we report anatase titania-coated plasmonic gold nanorods decorated with UCNPs (Au NR@aTiO2@UCNPs) for combinational photothermal and photodynamic therapy to treat cancer. Our novel architecture employs the incorporation of an anatase titanium dioxide (aTiO2) photosensitizer as a spacer and exploits the localized surface plasmon resonance (LSPR) properties of the Au core. The LSPR-derived near-field enhancement induces a threefold boost of upconversion emissions, which are re-absorbed by neighboring aTiO2 and Au nanocomponents. Photocatalytic experiments strongly infer that LSPR-induced hot electrons are injected into the conduction band of aTiO2, generating reactive oxygen species. As phototherapeutic agents, our hybrid nanostructures show remarkable in vitro anticancer effect under NIR light [28.0% cancer cell viability against Au NR@aTiO2 (77.3%) and UCNP@aTiO2 (98.8%)] ascribed to the efficient radical formation and LSPR-induced heat generation, with cancer cell death primarily following an apoptotic pathway. In vivo animal studies further confirm the tumor suppression ability of Au NR@aTiO2@UCNPs through combinatorial photothermal and photodynamic effect. Our hybrid nanomaterials emerge as excellent multifunctional phototherapy agents, providing a valuable addition to light-triggered cancer treatments in deep tissue.

Abstract:In this work, we develop a multifunctional nano-platform by coating upconversion nanoparticles (UCNPs) with bovine serum albumin (BSA), obtaining UCNP@BSA nanoparticles with great solubility and stability in physiological environments. Two types of dye molecules, including a photosensitizer, Rose Bengal (RB), and an NIR-absorbing dye, IR825, can be simultaneously loaded into the BSA layer of the UCNP@BSA nanoparticles. In this carefully designed UCNP@BSA-RB&; IR825 system, RB absorbs green light emitted from UCNPs under 980-nm excitation to induce photodynamic cancer cell killing, while IR825 whose absorbance shows no overlap with upconversion excitation and emission wavelengths, offers nanoparticles a strong photothermal perform under 808-nm laser irradiation. Without showing noticeable dark toxicity, the obtained dual-dye loaded nanoparticles are able to kill cancer via combined photothermal and photodynamic therapies, both of which are induced by NIR light with high tissue penetration, by a synergetic manner both in vitro and in vivo. In addition, the intrinsic paramagnetic and optical properties of Gd(3+)-doped UCNPs can further be utilized for in vivo dual modal imaging. Our study suggests that UCNPs with well-designed surface engineering could serve as a multifunctional nano-platform promising in cancer theranostics.

Abstract:The design and synthesis of a novel generation of a nanoscaled platform with imaging-guided therapy remain a real challenge. It can not only improve the imaging sensitivity of tumor tissues for guiding all kinds of treatments but also reduce the harm for healthy tissues. Here, polydopamine (PDA), polyethylene glycol (PEG), and c(RGDyK) peptide (RGD)-modified and cisplatin-loaded Gd2Hf2O7 nanoparticles (Gd2Hf2O7@PDA@PEG-Pt-RGD NPs) are designed for magnetic resonance imaging (MRI)-guided combined chemo-/photothermal-/radiotherapy of resistant tumors. The as-prepared NPs display high relaxivity (r1 = 38.28 mM-1 s-1) as an MRI contrast agent because of their ultrasmall size and surface modification with polyacrylic acid and PDA. Gd2Hf2O7@PDA@PEG-Pt-RGD NPs exhibit pH and NIR dual-stimuli responsiveness for cisplatin release. Based on competent NIR absorption and high X-ray attenuation efficiency, Gd2Hf2O7@PDA@PEG-Pt-RGD NPs show potential photothermal effect by exposing to an 808 nm NIR laser and significantly improve the generation of reactive oxygen species after X-ray radiation. Combined chemo-/photothermal-/radiotherapy can effectively treat the resistant A549R cells, providing the enhanced therapeutic efficiency to cancer tissues and the reduced side effect to healthy tissues. Furthermore, Gd2Hf2O7@PDA@PEG-Pt-RGD NPs present no obvious toxicity during the treatment, which demonstrates the potential as an efficient MRI-guided combined chemo-/photothermal-/radiotherapy nanoplatform for drug-resistant tumors.

Abstract:Theranostics, the integration of diagnostics and therapies, has become a new concept in the battles with various major diseases such as cancer. Here, we report a multifunctional nanoplatform, which is developed by covalently grafting core-shell structured upconversion nanoparticles (UCNPs) with nanographene oxide (NGO) via bifunctional polyethylene glycol (PEG), and then loading phthalocyanine (ZnPc) on the surface of NGO. The obtained UCNPs-NGO/ZnPc nanocomposites are not only be used as upconversion luminescence (UCL) imaging probes of cells and whole-body animals with high contrast for diagnosis, but also can generate cytotoxic singlet oxygen under light excitation for photodynamic therapy (PDT), as well as rapidly and efficiently convert the 808 nm laser energy into thermal energy for photothermal therapy (PTT). A remarkably improved and synergistic therapeutic effect compared to PTT or PDT alone is obtained, providing high therapeutic efficiency for cancer treatment. Therefore, benefiting from the unique multifunctional hybrid nanostructure, UCNPs-NGO/ZnPc nanocomposites developed herein are promising as an integrated theranostic probe for potential UCL image-guided combinatorial PDT/PTT of cancer.

Abstract:The novel lutecium‐based rare earth nanoparticles RENPs offer the potential for imaging‐guided tumor delineation with CXCR4 target, radio‐sensitization with ROS production and tumor microenvironment immune activation with immunogenic cell death and immune cell recruitment. This multifunctional nanoprobe hold promises for enhancing the effectiveness of radioimmunotherapy in triple negative breast cancer synergistically. Radiotherapy demonstrates a synergistic effect with immunotherapy by inducing a transformation of "immune cold" tumors into "immune hot" tumors in triple negative breast cancer (TNBC). Nevertheless, the effectiveness of immunotherapy is constrained by low expression of tumor‐exposed antigens, inadequate inflammation, and insufficient tumor infiltrating lymphocyte (TILs). To address this predicament, novel lutecium‐based rare earth nanoparticles (RENPs) are synthesized with the aim of amplifying radiation effect and tumor immune response. The nanoprobe is characterized by neodymium‐based down‐conversion fluorescence, demonstrating robust photostability, biocompatibility, and targetability. The conjugation of RENPs with a CXCR4 targeted drug enables precise delineation of breast tumors using a near‐infrared imaging system and improves radiation efficacy via lutetium‐based radio‐sensitizer in vivo. Furthermore, the study shows a notable enhancement of immune response through the induction of immunogenic cell death and recruitment of TILs, resulting in the inhibition of tumor progression both in vitro and in vivo models following the administration of nanoparticles. Hence, the novel multifunctional nanoprobes incorporating various lanthanide elements offer the potential for imaging‐guided tumor delineation, radio‐sensitization, and immune activation post‐radiation, thus presenting an efficient radio‐immunotherapeutic approach for TNBC.

Theranostic nanocomposite from upconversion luminescent nanoparticles and black phosphorus nanosheets

Abstract:An anti-cancer campaign might not be easily achieved through a single therapeutic modality. Collaboration of multimodal therapies and diagnosis could be vital to win the battle against cancer. In this context, we synthesized a multifunctional theranostic nanocomposite (UCNP-BPNS) from upconversion nanoparticles (UCNP) and black phosphorus nanosheets (BPNS) for synergistic photothermal/photodynamic therapies in vitro and dual modal imaging. Core-shell UCNP (NaYF4:Yb,Er@NaGdF4) and BPNS were synthesized using solvo-thermal and liquid exfoliation methods, respectively, and then covalently conjugated after UCNP was modified with polyacrylic acid and BPNS with methoxypolyethylene glycol amine. The experimental results validate that the nanocomposite exhibited a good photothermal therapy (PTT) effect under 808 nm laser irradiation, endorsing the apparent heat conversion effect of BPNS. Besides, a very good photodynamic therapy (PDT) effect was achieved under 980 nm laser irradiation of the nanocomposite due to Förster resonance energy transfer from UCNP to BPNS that generated singlet oxygen (1O2). The synergistic PTT/PDT therapeutic effect provided by UCNP-BPNS under simultaneous 808 and 980 nm laser irradiation was significantly higher than either PTT or PDT alone. Furthermore, due to the merit of the outer shell coated on the surface of the core of UCNP, the nanocomposite exhibited good potential for magnetic resonance and upconversion luminescence imaging. These results demonstrated that our multifunctional nanocomposite has promising theranostic efficacy under near infrared laser irradiation.

Abstract:Radionuclide therapy (RNT) is an effective method for the clinical precise treatment of cancer. However, the uneven dose distribution and rapid metabolism of nuclides limit the effective killing of tumors. To overcome the limitations of radionuclide therapeutic approaches, combining different therapeutic strategies to treat cancer has manifested great promise in basic and clinical research. Here, a new combination therapy strategy was developed to combine radionuclide therapy, sonodynamic therapy and photothermal therapy (RNT-SDT-PTT) under radionuclide imaging guided achieve highly effective combination therapy. We prepared a polydopamine-modified Au nanostar (AN), then loaded with the acoustic sensitizer protoporphyrin (IX) and labeled with diagnostic ( 99m Tc) or therapeutic ( 131 I) radionuclides ( 131 I/ 99m /IX) for the precise diagnosis and treatment of pancreatic cancer. After intratumor administration, single photon emission computed tomography imaging showed that the nanocarriers were mostly retained in the tumor compared to free radionuclide. As well as using near-infrared light to trigger PTT and ultrasound with high penetration depth to activate IX to generate reactive oxygen species achieved SDT of tumor. The ultimate significantly improved the inhibitory effects by the RNT-SDT-PTT combined therapy for pancreatic cancer. Therefore, this study proposes an effective radionuclide combination therapy regimen consisting of three widely used treatments, offering promising prospects for the future of oncology.
