Abstract:Goldfish and crucian carp at low temperature exhibit plasticity in gill morphology during exposure to hypoxia to enhance gas exchange. Hypoxia-induced changes in gill morphology and cellular ultrastructure of the high altitude scaleless carp from Lake Qinghai, China, were investigated to determine whether this is a general characteristic of cold water carp species. Fish were exposed to acute hypoxia (0.3 mg O-2 l(-1)) for 24 h followed by 12 h recovery in normoxic water (6 mg O-2 l(-1) at 3200 m altitude), with no mortality. Dramatic alterations in gill structure were initiated within 8 h of hypoxia and almost complete by 24 h, and included a gradual reduction of filament epithelial thickness (> 50%), elongation of respiratory lamellae, expansion of lamellar respiratory surface area (> 60%) and reduction in epithelial water-blood diffusion distance (< 50%). An increase in caspase 3 activity in gills occurred following 24 h exposure to hypoxia, indicating possible involvement of apoptosis in gill remodeling. Extensive gill mucous production during hypoxia may have been part of a general stress response or may have played a role in ion exchange and water balance. The large increase in lamellar surface area and reduction in diffusion distance presumably enhances gas transfer during hypoxia (especially in the presence of increased mucous production) but comes with an ionoregulatory cost, as indicated by a 10 and 15% reduction in plasma [Na+] and [Cl-], respectively, within 12-24 h of hypoxia. Within 12 h of hypoxia exposure, 'wavy-convex '-mitochondria rich cells (MRCs) with large apical crypts and numerous branched microvilli were transformed into small 'shallow-basin' cells with a flattened surface. As the apical membrane of MRCs is the site for active ion uptake from the water, a reduction in apical crypt surface area may have contributed to the progressive reduction in plasma [Na+] and [Cl-] observed during hypoxia. The changes in the macro-and ultra-structure of fish gills, and plasma [Na+] and [Cl-] during hypoxia were reversible, showing partial recovery by 12 h following return to normoxia. Although the large morphological changes in the gill observed in the scaleless carp support the hypothesis that gill remodeling during hypoxia is a general characteristic of cold water carp species, the reduced magnitude of the response in scaleless carp relative to goldfish and crucian carp may be a reflection of their more active lifestyle or because they reside in a moderately hypoxic environment at altitude.

Abstract:Hypoxia is common in aquatic environments, and exposure to hypoxia followed by re-oxygenation is often believed to induce oxidative stress. However, there have been relatively few studies of reactive oxygen species (ROS) homeostasis and oxidative status in fish that experience natural hypoxia-re-oxygenation cycles. We examined how exposure to acute hypoxia (2 kPa O2) and subsequent re-oxygenation (to 20 kPa O2) affects redox status, oxidative damage and anti-oxidant defenses in estuarine killifish (Fundulus heteroclitus), and whether these effects were ameliorated or potentiated by prolonged (28 days) acclimation to either constant hypoxia or intermittent cycles of nocturnal hypoxia (12 h:12 h normoxia:hypoxia). Acute hypoxia and re-oxygenation led to some modest and transient changes in redox status, increases in oxidized glutathione, depletion of scavenging capacity and oxidative damage to lipids in skeletal muscle. The liver had greater scavenging capacity, total glutathione concentrations and activities of anti-oxidant enzymes (catalase, glutathione peroxidase) than muscle, and generally experienced less variation in glutathiones and lipid peroxidation. Unexpectedly, acclimation to constant hypoxia or intermittent hypoxia led to a more oxidizing redox status (muscle and liver) and it increased oxidized glutathione (muscle). However, hypoxia-acclimated fish exhibited little to no oxidative damage (as reflected by lipid peroxidation and aconitase activity), in association with improvements in scavenging capacity and catalase activity in muscle. We conclude that hypoxia acclimation leads to adjustments in ROS homeostasis and oxidative status that do not reflect oxidative stress, but may instead be part of the suite of responses that killifish use to cope with chronic hypoxia.

Abstract:Air-breathing fish risk losing aerially sourced oxygen to ambient hypoxic water since oxygenated blood from the air-breathing organ returns through the heart to the branchial basket before distribution. This loss is thought to help drive the evolutionary reduction in gill size with the advent of air-breathing. In many teleost fish, gill size is known to be highly plastic by modulation of their anatomic diffusion factor (ADF) with inter-lamellar cell mass (ILCM). In the anoxia-tolerant crucian carp, ILCM recedes with hypoxia but regrows in anoxia. The air-breathing teleost Chitala ornata has been shown to increase gill ADF from normoxic to mildly hypoxic water by reducing ILCM. Here, we test the hypothesis that ADF is modulated to minimize oxygen loss in severe aquatic hypoxia by measuring ADF, gas-exchange, and by using computed tomography scans to reveal possible trans-branchial shunt vessels. Contrary to our hypothesis, ADF does not modulate to prevent oxygen loss and despite no evident trans-branchial shunting, C. ornata loses only 3% of its aerially sourced O2 while still excreting 79% of its CO2 production to the severely hypoxic water. We propose this is achieved by ventilatory control and by compensating the minor oxygen loss by extra aerial O2 uptake.

Abstract:Amphibious fishes have evolved multiple adaptive strategies for respiring out of water, but there has been less focus on reversible plasticity. We tested the hypothesis that when amphibious fishes leave water, enhanced respiratory performance on land is the result of rapid functional phenotypic flexibility of respiratory traits. We acclimated four isogenic strains of Kryptolebias marmoratus to air for 0, 1, 3 or 7 days. We compared respiratory performance out of water with traits linked to the O2 cascade. Aerial O2 consumption rate was measured over a step-wise decrease in O2 levels. There were significant differences between strains, but time out of water had the largest impact on measured parameters. K. marmoratus had improved respiratory performance (lower aerial critical oxygen tension (Pcrit), higher regulation index (RI)) after only 1 day of air exposure and these changes were strongly associated with the change in hematocrit and dorsal cutaneous angiogenesis. Additionally, we found that 1-hour of air exposure induced the expression of four angiogenesis-associated genes, vegfa, angpt2, pecam-1 and efna1 in the skin. After 7 days in air, respiratory traits were not significantly linked to the variation in either aerial Pcrit or RI. Overall, our data indicate that there are two phases involved in the enhancement of aerial respiration; an initial rapid response (1 day) and a delayed response (7 days). We found evidence for the hypothesis that respiratory performance on land in amphibious fishes is the result of rapid flexibility in both O2 uptake and O2 carrying capacity.

Hypoxic regulation and oxidative stress in marine invertebrates and fish

Abstract:With the exception of mammalian hibernators, entering and emerging from a state of metabolic rate depression is a physiological capacity, alien to warm blooded animals. Many marine ectotherms, including some benthic fish, however, make use of metabolic rate reduction to survive prolonged periods of environmental hypoxia, common in coastal and oceanic habitats. Hypoxic specialist sediment dwellers can survive for several weeks under conditions close to anoxia, and some of these animals even avoid exposure to fully normoxic conditions. We have studied oxidative stress markers in several infauna worms and mud clams, exposed to environmental hypoxia and found them to maintain strong antioxidant capacity.Entering a hypoxic state causes a regulatory response in mammals, involving activation of hypoxia inducible transcription factor HIF-1 α, which is stabilized under severe tissue hypoxia and, after dimerization with HIF-1?, induces up-regulation of genes that preserve functionality in hypoxic tissues. An oxidized tissue redox state and reactive oxygen species are know to prevent HIF-1 α stabilization and interfere with the hypoxic signalling cascade. We were interested in HIF-1 α functionality in hypoxia tolerant marine ectotherms and studied HIF-1 regulation in response to thermal stress and hypoxic exposure in North Sea and polar confamilial fishes (eelpout). We found constitutive HIF-1 α protein levels in fish liver and an increase of DNA binding activity of the HIF-1 dimer under severe cooling in the North Sea eelpout, presumably related to functional hypoxia in liver tissue. Generally, HIF-1 activation was more pronounced at more reduced tissue redox state, however oxidative stress markers were simultaneously increased. Several HIF-1α sequences were since obtained from temperate and polar fish species and will be discussed with respect to the functional peculiarities of the transcription factor in hypoxia tolerant ectotherms.

Reactive oxygen species (ROS) homeostasis is essential for the maintenance of normal physiological conditions in an organism. Direct evidence that environmental hypoxia leads to ROS overproduction is scarce in marine fish. Furthermore, the sources of ROS overproduction in marine fish under hypoxic stress are poorly known. In this study, we investigated the effects of hypoxia on redox homeostasis in L. crocea and the impact of impaired redox homeostasis on fish. We first confirmed that hypoxia drove ROS production mainly via the mitochondrial electron transport chain and NADPH oxidase complex pathways in L. crocea and its cell line (large yellow croaker fry (LYCF) cells). We subsequently detected a marked increase in the antioxidant systems of the fish. However, imbalance between the pro-oxidation and antioxidation systems ultimately led to excessive ROS and oxidative stress. Cell viability showed a remarkable decrease while oxidative indicators, such as malondialdehyde, protein carbonylation, and 8-hydroxy-2 deoxyguanosine, showed a significant increase after hypoxia, accompanied by tissue damage. N-acetylcysteine (NAC) reduced ROS levels, alleviated oxidative damage, and improved cell viability in vitro. Appropriate uptake of ROS scavengers (e.g., NAC and elamipretide Szeto-Schiller-31) and inhibitors (e.g., apocynin, diphenylene iodonium, and 5-hydroxydecanoate) may be effective at overcoming hypoxic toxicity. Oxygen consumption rate during recovery from loss of equilibrium induced by warming, hypoxia, or exhaustive exercise in rainbow darter (Etheostoma caeruleum)

Abstract:Animals routinely encounter environmental (e.g., high temperatures and hypoxia) as well as physiological perturbations (e.g., exercise and digestion) that may threaten homeostasis. However, comparing the relative threat or "disruptiveness" imposed by different stressors is difficult, as stressors vary in their mechanisms, effects, and timescales. We exploited the fact that several acute stressors can induce the loss of equilibrium (LOE) in fish to (i) compare the metabolic recovery profiles of three environmentally relevant stressors and (ii) test the concept that LOE could be used as a physiological calibration for the intensity of different stressors. We focused on Etheostoma caeruleum, a species that routinely copes with environmental fluctuations in temperature and oxygen and that relies on burst swimming to relocate and avoid predators, as our model. Using stop-flow (intermittent) respirometry, we tracked the oxygen consumption rate (MO2) as E. caeruleum recovered from LOE induced by hypoxia (PO2 at LOE), warming (critical thermal maximum, CTmax), or exhaustive exercise. Regardless of the stressor used, E. caeruleum recovered rapidly, returning to routine MO2 within ~3 h. Fish recovering from hypoxia and warming had similar maximum MO2, aerobic scopes, recovery time, and total excess post-hypoxia or post-warming oxygen consumption. Though exhaustive exercise induced a greater maximum MO2 and corresponding higher aerobic scope than warming or hypoxia, its recovery profile was otherwise similar to the other stressors, suggesting that "calibration" to a physiological state such as LOE may be a viable conceptual approach for investigators interested in questions related to multiple stressors, cross tolerance, and how animals cope with challenges to homeostasis.
Air Pocket for a Non-Air-Breathing Fish: Developing Respirometry Methodology for the critically endangered Delta Smelt

Abstract:Respirometry quantifies O2 consumption to estimate metabolic activity across stressor(s), and the resulting data is central for devising conservation strategies for fishes. Due to its rapid adoption and widespread utility, protocols have been established to foster quality control and increase consistency. However, the application of established respirometry protocols on the Delta Smelt (Hypomesus transpacificus) have been largely unsuccessful, with reports over the past 12 years documenting high mortality rates that can only be circumvented with very short measurement duration (~4 h). This prevents the measurement of standard metabolic rate (SMR) and critical O2 tension (Pcrit), and ultimately obstructed physiological research, management actions and conservation tools development. In this study, we detailed the anecdotal, behavioral and empirical evidence that culminated in successful measurements of Delta Smelt SMR and Pcrit at 10, 12, 15, 17 and 19 degrees C. We discovered the Delta Smelt is physotomous and requires frequent refilling of its air bladder to maintain buoyancy. Inspired by air-breathing fish respirometry, the inclusion of an air pocket within their chamber led to considerably more robust respirometry measurements compared to past attempts. The influence of fasting and time of day on O2 consumption were also examined. Altogether, this methodology paves the way for acquiring critical knowledge needed to inform Delta Smelt research and conservation (e.g. the development of metabolic index). The lessons learned may also have implications for other aspects of Delta Smelt conservation such as decreasing conservation aquaculture operating costs and improving survival during fish transport and supplementation.

Abstract:Naturalistic data are reported concerning the first year of hospital use of fluphenazine decanoate. Observations revealed that staff psychiatrists were cautious in their initial use of this product, consistently reserving it for those patients who were very uncooperative about taking oral medications. Outcome of treatment was regarded as satisfactory in only 38% of the patients and a number of unusually severe and prolonged side effects were encountered. A conservative position about the use of depot medications in the treatment of schizophrenia is advocated until further studies of specific target areas for such treatment can be accomplished.

Abstract:Background Water deprivation-induced hypoxia stress (WDIHS) has been extensively investigated in numerous fish species due to their adaptation with accessory respiratory organs to respire air but this has not been studied in Indian stinging fish Heteropneustes fossilis . Data regarding WDIHS-induced metabolism in accessory respiratory organ (ARO) and gills and its relationship with oxidative stress (OS) in respiratory organs of air-breathing fish H. fossilis , are limited. So, this study aimed to investigate the effects of WDIHS (0, 3, 6, 12, and 18 h) on hydrogen peroxide (H 2 O 2 ) as reactive oxygen species (ROS), OS, redox regulatory enzymes, and electron transport enzymes (ETC) in ARO and gills of H. fossilis . Methods Fish were exposed to air for different hours (up to 18 h) against an appropriate control, and ARO and gills were sampled. The levels of oxygen saturation in the body of the fish were assessed at various intervals during exposure to air. Protein carbonylation (PC) and thiobarbituric acid reactive substances (TBARS) were used as OS markers, H 2 O 2 as ROS marker, and various enzymatic activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR), along with the assessment of complex enzymes (I, II, III, and V) as well as the levels of ascorbic acid (AA) and the reduced glutathione (GSH) were quantified in both the tissues. Results Discriminant function analyses indicate a clear separation of the variables as a function of the studied parameters. The gills exhibited higher levels of GSH and H 2 O 2 compared to ARO, while ARO showed elevated levels of PC, TBARS, AA, SOD, CAT, and GPx activities compared to the gills. The activities of GR and ETC enzymes exhibited similar levels in both the respiratory organs, namely the gills, and ARO. These organs experienced OS due to increased H 2 O 2 , TBARS, and PC levels, as observed during WDIHS. Under WDIHS conditions, the activity/level of CAT, GPx, GR, and GSH decreased in ARO, while SOD activity, along with GR, GSH, and AA levels decreased in gills. However, the activity/level of SOD and AA in ARO and CAT in gills was elevated under WDIHS. Complex II exhibited a positive correlation with WDIHS, while the other ETC enzymes (complex I, III, and V) activities had negative correlations with the WDIHS. Discussion The finding suggests that ARO is more susceptible to OS than gills under WDIHS. Despite both organs employ distinct redox regulatory systems to counteract this stress, their effectiveness is hampered by the inadequacy of small redox regulatory molecules and the compromised activity of the ETC, impeding their ability to effectively alleviate the stress induced by the water-deprivation condition.

A skilled examiner may perform DO to identify clinical signs in the eye that may signify a sightthreatening disorder such as diabetic retinopathy, glaucoma and neovascular agerelated macular degeneration, or a lifethreatening disorder such as raised intracranial pressure or retinoblastoma/choroidal melanoma (Figure 1 ). These disorders are treatable, so it is important that all clinicians are competent at DO.

Abstract:ABSTRACT Oxygen (O2) is required for aerobic energy metabolism but can produce reactive oxygen species (ROS), which are a wide variety of oxidant molecules with a range of biological functions from causing cell damage (oxidative distress) to cell signalling (oxidative eustress). The balance between the rate and amount of ROS generated and the capacity for scavenging systems to remove them is affected by several biological and environmental factors, including oxygen availability. Ectotherms, and in particular hypoxia-tolerant ectotherms, are hypothesized to avoid oxidative damage caused by hypoxia, although it is unclear whether this translates to an increase in ecological fitness. In this Review, we highlight the differences between oxidative distress and eustress, the current mechanistic understanding of the two and how they may affect ectothermic physiology. We discuss the evidence of occurrence of oxidative damage with hypoxia in ectotherms, and that ectotherms may avoid oxidative damage through (1) high levels of antioxidant and scavenging systems and/or (2) low(ering) levels of ROS generation. We argue that the disagreements in the literature as to how hypoxia affects antioxidant enzyme activity and the variable metabolism of ectotherms makes the latter strategy more amenable to ectotherm physiology. Finally, we argue that observed changes in ROS production and oxidative status with hypoxia may be a signalling mechanism and an adaptive strategy for ectotherms encountering hypoxia.

Abstract:Surviving hypoxia is one of the most critical challenges faced by vertebrates. Most species have adapted to changing levels of oxygen in their environment with specialized organs that sense hypoxia, while only few have been uniquely adapted to survive prolonged periods of anoxia. The goal of this review is to present the most recent research on oxygen sensing, adaptation to hypoxia, and mechanisms of anoxia tolerance in nonmammalian vertebrates. We discuss the respiratory structures in fish, including the skin, gills, and air-breathing organs, and recent evidence for chemosensory neuroepithelial cells (NECs) in these tissues that initiate reflex responses to hypoxia. The use of the zebrafish as a genetic and developmental model has allowed observation of the ontogenesis of respiratory and chemosensory systems, demonstration of a putative intracellular O2 sensor in chemoreceptors that may initiate transduction of the hypoxia signal, and investigation into the effects of extreme hypoxia on cardiorespiratory development. Other organisms, such as goldfish and freshwater turtles, display a high degree of anoxia tolerance, and these models are revealing important adaptations at the cellular level, such as the regulation of glutamatergic and GABAergic neurotransmission in defense of homeostasis in central neurons.

Abstract:Fish are among the best-studied aquatic animals due to their economic and ecological values. Fish meat is the most affordable protein source for the economically weaker section of people. The environment of almost all aquatic ecosystems has a specific influential role on or by fishes. Therefore, studying their stress biology, especially oxidative stress, is vital because it can influence their growth, production, reproduction, etc. To review the above topic, peer-reviewed electronic databases, including Web of Science, science direct, PubMed, Google Scholar, Scopus, and AGRICOLA, were searched with specific keywords associated with fish, oxidative stress, diseases, etc. The influence of abiotic stress, such as the effects of water dissolved oxygen, temperature, salinity, water hardness, alkalinity, pH, pollutants, heavy metals, and anthropogenic activities, was reviewed in the current article to draw a conclusion on the updated relation that exists between fish physiology, disease, and abiotic stressors. Oxidative stress and redox regulatory levels under the above parameters were reviewed as the stress or anti-stress responses differ in various fish models. Undoubtedly, the reviewed abiotic factors modulate fish oxidative health status to a greater extent, and therefore, these factors must be considered on a priority basis to improve the general health and immunity status of fish. The statement above remains valid in both saline and freshwater habitats.

Abstract:We used the AAMR's Adaptive Behavior Scale to ascertain current menstrual status in a population-based sample of 157 women with Down's syndrome (DS) and 187 women with other intellectual disability, all 40 years of age or older. The age-adjusted likelihood of menopause was twice as high in women with DS syndrome as in women with other intellectual disability (OR = 2.3; 95% CI = 1.1-4.9). Treated thyroid conditions did not influence menstrual status and did not modify the relationship between DS and menstrual status. These findings support the hypothesis that women with DS experience menopause at an earlier age and that this may be associated with accelerated aging.
Osmo-respiratory compromise in the mosshead sculpin (Clinocottus globiceps): effects of temperature, hypoxia, and re-oxygenation on rates of diffusive water flux and oxygen uptake

Abstract:In nature, mosshead sculpins (Clinocottus globiceps) are challenged by fluctuations in temperature and oxygen levels in their environment. However, it is unclear how mosshead sculpins modulate the permeability of their branchial epithelia to water and O2 in response to temperature or hypoxia stress. Acute decrease in temperature from 13 to 6 oC reduced diffusive water flux rate by 22% and ṀO2 by 51%, whereas acute increase in temperature from 13 to 25 oC increased diffusive water flux rate by 217% and ṀO2 by 140%, yielding overall Q10 values of 2.08 and 2.47 respectively. Acute reductions in oxygen tension from >95% to 20% or 10% air saturation did not impact diffusive water flux rates, however, ṀO2 was reduced significantly by 36% and 65% respectively. During 1-h or 3-h recovery periods diffusive water flux rates were depressed while ṀO2 exhibited overshoots beyond the normoxic control level. Many responses differed from those seen in our parallel earlier study on the tidepool sculpin, a cottid with similar hypoxia tolerance but much smaller gill area that occupies a similar environment. Overall, our data suggest that during temperature stress, diffusive water flux rates and ṀO2 follow the traditional osmo-respiratory compromise pattern, but during hypoxia and re-oxygenation stress, diffusive water flux rates are decoupled from ṀO2.
Gill ionocyte remodeling mediates blood pH regulation in rockfish (Sebastes diploproa) exposed to environmentally relevant hypercapnia

Abstract:Marine fishes excrete excess H + using basolateral Na + /K + -ATPase (NKA) and apical Na + /H + -exchanger 3 (NHE3) in gill ionocytes. However, the mechanisms that regulate H + excretion during exposure to environmentally relevant hypercapnia (ERH) remain poorly understood. Here, we explored transcriptomic, proteomic, and cellular responses in gills of juvenile splitnose rockfish ( Sebastes diploproa) exposed to three days of ERH conditions (pH ~7.5; ~1,600 μatm pCO 2 ). Blood pH was fully regulated at ~7.75 despite a lack of significant changes in gill (1) mRNAs coding for proteins involved in blood acid-base regulation, (2) total NKA and NHE3 protein abundance, and (3) ionocyte density. However, ERH-exposed rockfish demonstrated increased NKA and NHE3 abundance on the ionocyte plasma membrane coupled with wider apical membranes and greater extension of apical microvilli. The observed gill ionocyte remodeling is consistent with enhanced H + excretion that maintains blood pH homeostasis during exposure to ERH and does not necessitate changes at the expression or translation levels. These mechanisms of phenotypic plasticity may allow fishes to regulate blood pH during environmentally relevant acid-base challenges, and thus have important implications for both understanding how organisms respond to climate change and for selecting appropriate metrics to evaluate its impact on marine ecosystems.

Abstract:Hyperventilation is a common response in fish exposed to elevated water CO2. It is believed to lessen the respiratory acidosis associated with hypercapnia by lowering arterial PCO2, but the contribution of hyperventilation to blood acid–base compensation has yet to be quantified. Hyperventilation may also increase the flux of irons across the gill epithelium and the cost of osmoregulation, owing to the osmo-respiratory compromise. Therefore, hypercapnia exposed fish may increase standard metabolic rate (SMR) leaving less energy for physiological functions such as foraging, migration, growth and reproduction. Here we show that gill ventilation, blood PCO2 and total blood [CO2] increased in red drum (Sciaenops ocellatus) exposed to 1000 and 5000 µatm water CO2, and that blood PCO2 and total blood [CO2] decrease in fish during hypoxia induced hyperventilation. Based on these results we estimate the ventilatory contributions to total acid–base compensation in 1000 and 5000 µatm water CO2. We find that S. ocellatus only utilize a portion of its ventilatory capacity to reduce the acid–base disturbance in 1000 µatm water CO2. SMR was unaffected by both salinity and hypercapnia exposure indicating that the cost of osmoregulation is small relative to SMR, and that the lack of increased ventilation in 1000 µatm water CO2 despite the capacity to do so is not due to an energetic tradeoff between acid–base balance and osmoregulation. Therefore, while ocean acidification may impact ventilatory parameters, there will be little impact on the overall energy budget of S. ocellatus.

Abstract:Due to different sizes of humans and rodents, the performance of clinical imaging devices is not enough for a scientifically reliable evaluation in mice and rats; therefore dedicated small-animal systems with a much higher sensitivity and spatial resolution, compared to the ones used in humans, are required. Smallanimal imaging represents a cutting-edge research method able to approach an enormous variety of pathologies in which animal models of disease may be used to elucidate the mechanisms underlying the human condition and/or to allow a translational pharmacological (or other) evaluation of therapeutic tools. Molecular imaging, avoiding animal sacrifice, permits repetitive (i.e. longitudinal) studies on the same animal which becomes its own control. In this way also the over time evaluation of disease progression or of the treatment response is enabled. Many different rodent models have been applied to study almost all kind of human pathologies or to experiment a wide series of drugs and/or other therapeutic instruments. In particular, relevant information has been achieved in oncology by in vivo neoplastic phenotypes, obtained through procedures such as subcutaneous tumor grafts, surgical transplantation of solid tumor, orthotopic injection of tumor cells into specific organs/sites of interest, genetic modification of animals to promote tumor-genesis; in this way traditional or innovative treatments, also including gene therapy, of animals with a cancer induced by a known carcinogen may be experimented. Each model has its own disadvantage but, comparing different studies, it is possible to achieve a panoramic and therefore substantially reliable view on the specific subject. Small-animal molecular imaging has become an invaluable component of modern biomedical research that will gain probably an increasingly important role in the next few years.
Mechanisms of sustaining oxygen extraction efficiency in dragonfly nymphs during aquatic hypoxia

Abstract:Despite breathing water using their tidally ventilated rectal gills, dragonfly nymphs show a surprising ability to maintain oxygen (O ) extraction from the water during hypoxia. However, an increase in convective O transfer is insufficient to sustain aerobic demands by itself, which suggests that diffusive mechanisms must also be involved. This study examines the contributions of changing the O partial pressure gradient (PO ) and/or O conductance across the rectal gill in maintaining O extraction efficiency (OEE) of dragonfly nymphs during hypoxia. Data were collected using the same custom-designed respiro-spirometer described in a previous study with the addition of an implanted O sensor to measure hemolymph PO . Results show that the implantation of the O sensor does not affect the respiratory and ventilatory response of nymphs to hypoxia. Hemolymph PO fell from 6.3 ± 1.6 kPa at normoxia to 2.5 ± 0.6 kPa at 16.0 kPa, which resulted in the PO diffusion gradient remaining statistically constant at these two water PO s (17.5 ± 1.7 and 15.4 ± 0.7 kPa during normoxia and 16.0 kPa respectively). Beyond 16.0 kPa, a progressive reduction in hemolymph PO was unable to sustain the diffusion gradient. Mathematical modeling revealed that while the addition of hemolymph PO in tandem with ventilation frequency was able to elevate OEE during 16.0 kPa to that of normoxia, both were still insufficient during severe hypoxia and required an increase in O conductance. Estimating the change in whole-gill conductance showed that nymphs are indeed increasing their conductance as the water becomes hypoxic, demonstrating a reliance on both diffusion gradient and O conductance to enhance diffusive O transfer in conjunction with convective mechanisms to maintain O extraction during hypoxia.
