Abstract:Lethal cardiac arrhythmias contribute to mortality in a number of pathological conditions. Several parameters obtained from a non-invasive, easily obtained electrocardiogram (ECG) are established, well-validated prognostic indicators of cardiac risk in patients suffering from a number of cardiomyopathies. Increased heart rate, decreased heart rate variability (HRV), and increased duration and variability of cardiac ventricular electrical activity (QT interval) are all indicative of enhanced cardiac risk. In animal models, it is valuable to compare these ECG-derived variables and susceptibility to experimentally induced arrhythmias. Intravenous infusion of the arrhythmogenic agent aconitine has been widely used to evaluate susceptibility to arrhythmias in a range of experimental conditions, including animal models of depression and hypertension, following exercise and exposure to air pollutants, as well as determination of the antiarrhythmic efficacy of pharmacological agents. It should be noted that QT dispersion in humans is a measure of QT interval variation across the full set of leads from a standard 12-lead ECG. Consequently, the measure of QT dispersion from the 2-lead ECG in the rat described in this protocol is different than that calculated from human ECG records. This represents a limitation in the translation of the data obtained from rodents to human clinical medicine. Status epilepticus (SE) is a single seizure or series of continuously recurring seizures lasting more than 30 min, and results in mortality in 20% of cases. Many individuals survive the SE, but die within 30 days. The mechanism(s) of this delayed mortality is not fully understood. It has been suggested that lethal ventricular arrhythmias contribute to many of these deaths. In addition to SE, patients experiencing spontaneously recurring seizures, i.e. epilepsy, are at risk of premature sudden and unexpected death associated with epilepsy (SUDEP). As with SE, the precise mechanisms mediating SUDEP are not known. It has been proposed that ventricular abnormalities and resulting arrhythmias make a significant contribution. To investigate the mechanisms of seizure-related cardiac death, and the efficacy of cardioprotective therapies, it is necessary to obtain both ECG-derived indicators of risk and evaluate susceptibility to cardiac arrhythmias in animal models of seizure disorders. Here we describe methods for implanting ECG electrodes in the Sprague-Dawley laboratory rat (Rattus norvegicus), following SE, collection and analysis of ECG recordings, and induction of arrhythmias during iv infusion of aconitine. These procedures can be used to directly determine the relationships between ECG-derived measures of cardiac electrical activity and susceptibility to ventricular arrhythmias in rat models of seizure disorders, or any pathology associated with increased risk of sudden cardiac death.
Absorption of Epinephrine from the Subcutaneous Tissue of the Rat

Abstract:In a previous investigation of the carbohydrate metabolism of rats a dose of 0.2 mg. of epinephrine per kilo was injected subcutaneously. 1 This made it desirable to find out whether such a dose produced a rise in blood pressure. The animals received amytal intraperitoneally, followed by the injection of a small amount of urethane in order to steady the blood pressure. A glass cannula was tied into the carotid artery and connected with a mercury manometer of small dimensions. Heparin was used as anticoagulant in the cannula. Intravenous injections were made into a femoral vein and were timed by the beats of a metronome. In 4 experiments of the type shown in Fig. 1 the minimal pressoric rate of intravenously injected epinephrine was established before the subcutaneous injection was made. It was found that an intravenous injection of 0.001 mg. per kilo per minute was always followed by a rise in blood pressure, while a subcutaneous injection of 0.2 mg. per kilo made shortly afterwards had no effect on blood pressure. This result permits the conclusion that the absorption of epinephrine from the subcutaneous tissue of the rat proceeds at a rate less than 0.001 mg. per kilo per minute. How much less the rate is can of course not be judged from these experiments. Since a subcutaneous injection leads to hyperglycemia and since an intravenous injection of 0.0002 mg. per kilo per minute is the smallest amount which still produces hyperglycemia in rats, the rate of absorption of epinephrine from the subcutaneous tissue must be above this figure. In order to ascertain how long epinephrine remains in the subcutaneous tissue, chemical determinations were carried out. Rats of 250 to 300 gm. of body weight were anesthetized with amytal.

Novel method for induction of myocardial infarction in rats by ultrasound guided electrocoagulation of the left anterior descending artery

Main funding source(s): Minerva Imaging Background Murine models of acute myocardial infarction (MI) are commonly established through ligation of the left anterior descending artery (LAD). Despite inherent procedural challenges, including high mortality rates, limited improvements of the induction procedure have been made. Ultrasound-guided electrocoagulation of the LAD could be advantageous by improving animal welfare and decreasing procedural mortality. Purpose To investigate the feasibility of MI induction by ultrasound guided electrocoagulation in rats in comparison to traditional LAD ligation. Methods Thirty-eight female Sprague Dawley rats were randomized to MI by either of the two induction methods leading to 19 rats in each group. Infarct size and left ventricular ejection fraction (LVEF) were evaluated using positron emission tomography (PET) with [18F]fluorodeoxyglucose ([18F]FDG) 9 days post-MI. Myocardial infarct size was categorized as non-infarcted, mild, moderate, or severe based on [18F]FDG uptake. Animal welfare was assessed twice daily for the initial 48h post-surgery using a behavioural scoring system. Results There was no significant difference in the mortality rate of rats with MI induced by electrocoagulation compared to LAD ligation (16% vs 37%, P=0.27). Among the surviving animals both methods produced an equal number of myocardial infarcts (n=5). Electrocoagulation resulted in 1 mild, 2 moderate, and 2 severe MIs, while LAD ligation produced 1 mild, 0 moderate, and 4 severe MIs. Infarct size was similar in the electrocoagulation group compared to the LAD ligation group (mean±SD electrocoagulation: 15±5% vs ligation: 28±13%, P=0.13). The number of non-infarcted surviving rats in the electrocoagulation group was 11 compared to 7 in the LAD ligation group (P=0.33). The LVEF of infarcted rats showed no significant difference between groups (electrocoagulation: 47±8% vs ligation: 38±10%, P=0.14). The LVEF for non-infarcted rats was 53±5%. Post-surgical animal welfare appeared to be improved in the electrocoagulation group, however, the accumulated behavioural score did not reveal a significant difference (median [interquartile range] electrocoagulation: 0 [0-1] vs ligation 2 [0-2], P=0.19). Conclusion Induction of myocardial infarction by ultrasound-guided electrocoagulation is feasible in rats and should be considered as an alternative to LAD ligation. Further development is needed to improve the rate of success and consistency in infarct size.

Determination of Epimedin B in Rat Plasma and Tissue by LC-MS/MS: Application in Pharmacokinetic and Tissue Distribution Studies

Abstract:A simple, sensitive, and specific liquid chromatography tandem mass-spectrometric method was developed and validated for the determination of epimedin B in rat plasma and tissue samples. After being processed with a protein precipitation method, these samples were separated on an Agilent Eclipse XDB-C 18 column with an isocratic mobile phase consisting of acetonitrile and 0.1% formic acid aqueous solution (32 : 68, v/v). The calibration curve of epimedin B was linear over the concentration range from 1 to 500 ng/mL in plasma and tissue homogenate. The method was then applied to pharmacokinetic and tissue distribution studies after a single oral administration of Herba Epimedii extract to SD rats. Results showed that epimedin B reached the plasma peak concentration at 0.4 h and the terminal elimination half-life was 1.6 h in rat plasma, and the plasma area under the curve from time zero to infinity ( A U C 0 – ∞ ) was 14.35 μ g/L·h. The concentration distribution of epimedin B in rat tissue was in the following order: liver > ovary > womb > lung > kidney > spleen > heart > brain, indicating that the compound could be widely distributed in rat, and the reproductive system may be the principal target of epimedin B for female rat.

Abstract:Existing methods of heart rate recording in animals have shortcomings, which significantly obscure the influence of experimental factors on heart rate. We have developed a method of electrocardiographic recording of heart rate in rats without these drawbacks. To contact the animal limbs used liquid electrodes which are two small baths filled with conductive fluid (saline solution). To provide the relative immobility (and quiet) of the animal the two baths was covered with a dark chamber without a bottom and with an entrance for the rat. During the experiment, a rat placed near the chamber comes into it (for the innate preference for darkness) and locates itself inside with its head for the exit. At that moment ECG recording starts. This method allows to record heart rate in the intact rodents (without anesthesia and stress) and does not require substantial preparation. It is not suitable for standard ECG analysis of the heart condition and function, but this is a good way for recording heart rate for the further analysis of its variability.
Determination of catecholamine secreted from rat adrenal medulla by in vivo voltammetry

Abstract:In vivo voltammetry using carbon fiber electrodes was employed to determine the catecholamine secreted from the adrenal medulla. The basal level of catecholamine in the medulla was between 0.1 and 0.5 micromol/L, but it increased markedly up to 5 30 micromol/L when the gland was made ischemic. The spontaneous release of catecholamine from the medulla was observed in both normal and ischemia conditions, but the frequency and amplitude of spontaneous release were increased significantly by ischemia. This implied that the animal was under serious stress condition. The above results also show that acetylcholine can induce the secretion of catecholamine in a dose-dependent manner.

Abstract:OBJECTIVE:To understand the effect of pinacidil on rat myocardial Ca(2+)regulation.METHODS:After baseline measurement and a period of equilibrium, myocytes were randomly allocated to one of 4 treatment groups: Control group (8 myocytes): incubation in Lactate Ringer's solution at 24 degrees C for 2 hours; K group (8 myocytes): incubation in Lactate Ringer's solution containing 16 mmol/L potassium at 24 degrees C for 2 hours; K+P group (8 myocytes): incubation in Lactate Ringer's solution containing potassium 16 mmol/L and pinacidil 50 micromol/L at 24 degrees C for 2 hours; K+P+G group (8 myocytes): incubation in Lactate Ringer's solution containing potassium 16 mmol/L, pinacidil 50 micromol/L and glibenclamide 10 micromol/L at 24 degrees C for 2 hours. After each incubation, myocytes were resuspended in cell culture media at the same temperature and intracellular [Ca(2+)](i) and SR Ca(2+) release were measured.RESULTS:The amplitude percent of [Ca(2+)](i) transient evoked by electrical stimulation in the K group was significantly decreased to 67.05% - 80.11% compared to 90.27% - 95.57% in the K+P group during reperfusion after ischemia (P<0.01). The percent amplitude of the [Ca(2+)](i) transient evoked by the rapid application of 10 mmol caffeine in the K group myocyte was approximately 112.00%+/-16.93% compared with that of the [Ca(2+)](i) transient evoked by electrical stimulation. However, in the K+P group myocyte the peak amplitude of the caffeine induced Ca(2+) release was 173.15%+/-26.01% compared with electrical stimulation (P<0.01). The duration of transient evoked by caffeine in K+P group (3.20+/-0.71 ms was significantly shorter than that in K group (3.93+/-0.46) ms (P<0.05).CONCLUSION:Cardioplegic arrest with simultaneous activation of KATP channels preserves rat myocardial Ca2+ by inducing sarcoplasmic reticulum Ca(2+) release and by alteration of Na(+)-Ca(2+) exchanger to better maintain [Ca(2+)](i) homeostasis.

Abstract:Objective A small animal model would be an effective tool for research on the pathophysiology of cardiopulmonary bypass (CPB). However, numerous CPB models do not involve myocardial arrest and resuscitation. The aim of this research is to establish an easily achievable myocardial arrest and resuscitation CPB model through hyperkalemia and landiolol, simulating clinical cardiac surgery. Materials and methods Ten Sprague-Dawley rats were chosen for CPB. Rats underwent sevoflurane inhalation induction anesthesia and were sustained in an anesthesia state by intubation and intraperitoneal injection’s of esketamine and propofol. The entire CPB circuit include a reservoir, a membrane oxygenator and a roller pump, which were connected into a complete loop via silicon tubes and infusion tube. After CPB was established through the tail artery and internal jugular vein, cardioplegic arrest was induced and maintained for 5 min at a rectum temperature of 28.5 ± 0.5°C with hyperkalemia and landiolol. Calcium chloride, epinephrine and insulin were then used for resuscitation. Result All rats successfully finished cardioplegic arrest, resuscitation procedure and survived 2 h postoperatively. Mean hematocrit during CPB was significantly lower than physiologic values of the baseline. The mean time of arrest-resuscitation and CPB was 5.4 ± 0.8 min and 98.5 ± 5.0 min. The blood gas at each detection point were in range with the normal standard requirement of CPB. Conclusion The establishment of cardioplegic arrest and resuscitation procedure via hyperkalemia and landiolol during CPB of WD rat could be achieved successfully. This animal model could be an alternative organ injury research on organ injury of patients undergoing cardiac surgery.
In situ detection of cyclic AMP-phosphodiesterase activity in the heart of Lewis and Sprague-Dawley rats: the effect of restraint stress or amphetamine application

Abstract:Cyclic AMP plays an important role in heart functions under normal as well as pathological conditions. Since phosphodiesterase (PDE), responsible for the hydrolysis of cAMP, is equally important as synthesizing adenylyl cyclase, we decided to determine its activity by cytochemical procedure after exposure of rats to restraint stress or an acute dose of amphetamine. Sprague-Dawley (S-D) and Lewis (LE) rats, the latter known to have a deficient hypothalamo-pituitary-adrenal axis activity, were used in order to disclose the possible significance of rat strain on PDE activity. Animals were divided into 3 groups: controls, rats treated with an acute dose of amphetamine (8 mg/kg, i.p., for 60 min) and rats under restraint stress for 60 min. Control hearts of both strains revealed PDE activity on sarcolemma of cardiomyocytes and plasmalemma of endothelial cells of microvessels. In LE rats we observed an additional enzyme reaction in junctional sarcoplasmic reticulum. In addition, cardiomyocytes of LE rats revealed a higher PDE activity when compared to S-D rats. Restraint stress decreased PDE activity in cardiomyocytes of LE rats while amphetamine markedly inhibited enzyme activity in cardiomyocytes of S-D rats. Endothelial PDE was more resistant to stress. Our results indicate differences in PDE localization and variations in sensitivity of myocardial cAMP-PDE of LE and S-D rat strains to restraint stress and amphetamine application.
[Determination of adenosine content in heart tissue by ultra performance liquid chromatography-triple quadrupole mass spectrometry]

Abstract:A method based on ultra performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-MS/MS) was developed and validated for the rapid and accurate determination of adenosine (Ado) in cardiac tissues with high sensitivity and specificity. The samples were dissolved in 1 mL of ultrapure water containing 10 μmol/L 2-hydroxy-3-nonyladenine hydrochloride (EHNA) as a stabilizer, ground at low temperature for 2 min, and then ultrasonically extracted at 60 Hz in an ice-water bath for 40 min. Methanol and 5 mmol/L ammonium acetate solution were used as the mobile phases under a flow rate of 0.4 mL/min, a column temperature of 40 ℃ and an injection volume of 3 μL. The Ado in cardiac tissue was qualitatively and quantitatively analyzed by electrospray ionization (ESI) positive-ion-switching in multiple reaction monitoring (MRM) mode. A solvent standard curve and the external standard method were used for the accurate quantification of Ado. The results showed that the matrix effect of Ado in cardiac tissue was very low. A good linear relationship was obtained in the range of 0.1-160 ng/mL, and the correlation coefficient (r2) was 0.9930. The limits of detection (LOD) and quantification (LOQ) were 0.03 and 0.1 ng/mL, respectively. The spiked recoveries of Ado in murine cardiac tissue were 113.6%, 96.3%, and 102.9% at three spiked levels of low, medium, and high, respectively. The intra-day repeatability (RSDs) were 1.7%-8.4%, and the inter-day reproducibility (RSDs) were 2.6%-7.4%. Based on the correlation and consistency results, a positive bias was observed between the proposed UPLC-MS/MS method and the double-antibody sandwich method. Moreover, the Ado contents detected by these two methods were significantly positively correlated (P<0.0001). Cardiac tissue samples were collected from 17 mice and 17 rats and detected in our laboratory. The content ranges of Ado in the cardiac tissues of mice and rats determined by the developed UPLC-MS/MS method were 3.25-8.78 mg/kg and 10.24-15.19 mg/kg, respectively (average adenosine contents: 5.37 and 12.60 mg/kg, respectively). The developed method is simple, accurate, sensitive, and it is suitable for the determination of Ado in cardiac tissues. It also provides important technical support for cardiac clinical research and disease diagnosis.
Comparative Study of Functional Changes in Heart Mitochondria in Two Modes of Epinephrine Exposure Modeling Myocardial Injury in Rats

Abstract:The parameters of coupled respiration and transport of calcium ions in mitochondria isolated from the heart of rats were studied in two modes of exposure to epinephrine for modelling myocardial damage. In 24 h after injection of 1.5 mg/kg epinephrine to rats, we observed a decrease in the efficiency of oxidative phosphorylation in heart mitochondria in the presence of both NADH- and FADH-dependent respiratory substrates. Increasing the epinephrine dose and exposure (2 mg/kg, 72 h) led to a more pronounced decrease in the ADP/O coefficient when succinate was used as a substrate, which indicated a predominant decrease in the activity of complex II of the respiratory chain. The injection of epinephrine in the two modes resulted in a decrease in the rate of calcium entry in rat heart mitochondria, but had no effect on mitochondrial calcium retention capacity, which reflects the resistance of the organelles to the induction of the Са2+-dependent pore. These findings suggest that both cardiomyopathy models in rats can be used to study the effectiveness of pharmacological therapy using mitochondria-targeted agents.
Real-time ventricular volumes and cardiac function in the working rat heart

Abstract:The measurement of instantaneous left ventricular volumes throughout the cardiac cycle in a rat heart beating at rates of 300 to 600 min-1 with a chamber volume of only a few tenths of a milliliter is difficult. Combining an x-ray scatter detection system with a small x-ray source collimated to scatter low energy x-rays off a working rat heart, measurements of real-time changes in ventricular volumes can be determined. The x-ray scatter signal is proportional to the volume and can be calibrated to measure the actual volume. Using this system, changes in ventricular function can readily be detected. An example of data obtained from increasing preload with a fixed afterload is shown. This technique not only distinguishes an increasing stroke volume, but also more rapid early diastolic filling and systolic ejection rates with increasing preload. This new x-ray scatter technique appears to be a promising way to measure rapidly changing left ventricular volumes and function in the working rat heart. This could significantly enhance the scientific use of the rat model.
The rat telemetry assay and venous catheter access buttons for use in cardiovascular safety pharmacology assessments - Surgical methods, refinements and colony maintenance

Abstract:Introduction: Rat telemetry is the assay of choice to assess the potential effects of novel drug candidates on cardiovascular parameters during early drug discovery. Telemetry device implantation can be combined with venous catheter and access button implantation when intravenous administration of the drug substance is required. Methods: Rats (Sprague Dawley or Han Wistar) were implanted with telemetry devices for arterial blood pressure measurement using either direct aortic catheterisation (n = 131) or aortic catheterisation via the femoral artery (n = 17). Bipolar leads for ECG recording were also implanted in some of the animals (n = 102). Femoral vein catheters and access buttons were implanted as a separate surgery after the initial telemetry implantation (n = 43). Results: 128 animals (86%) were implanted successfully with telemetry devices without any notable surgical or post-surgical problems. When considering the 2 different catheterisation methods separately, the success rate of the direct aortic approach was 88% compared to 76% with the aortic placement via the femoral artery. Lameness was the most common post-surgical problem. Blood loss during surgery and ischaemic patches on the tail were also observed at a low incidence with the direct aortic approach. Catheter pull-out occurred in some rats before the first signal check reducing the overall success rate for blood pressure measurement using the direct aortic approach to 85%. A 95% success rate was observed for catheter and access button implantation. Discussion: A high success rate is possible when implanting telemetry devices in rats with and without venous catheters and access buttons. We have attempted to provide solutions to problems and describe refinements to the procedure which may further improve surgical outcomes.
Effects of Epinephrine and Phenylephrine on Evoked Potentials in a Rat Model of Acute Hemorrhagic Shock

Abstract:Objective To evaluate the effects of epinephrine and phenylephrine on evoked potentials in a rat model of acute hemorrhagic shock. Methods Eighteen SPF healthy male Sprague-Dawley rats, aged 11 weeks, weighing 250-350 g, were divided into 3 groups ( n=6 each) using a random number table meth-od: acute hemorrhagic shock group ( HS group) , epinephrine group ( E group) and phenylephrine group ( P group). The animals were anesthetized with intraperitoneal 10％ chloral hydrate 300 mg∕kg and tracheos-tomized, and spontaneous breathing was kept. Acute hemorrhagic shock was induced by withdrawing 40％ of the blood volume from the right internal jugular vein. Epinephrine 2 μg·kg-1 ·min-1 ( 0. 01 mg∕ml, 0. 3 ml) and phenylephrine 20μg·kg-1·min-1 (0. 1 mg∕ml, 0. 3 ml) were intravenously infused after the end of blood letting in E and P groups, respectively. Mean arterial pressure ( MAP ) , somatosensory evoked po-tential (SSEP) and motor evoked potentials (MEP) were recorded before blood letting, at the time of with-drawing 20％of the blood volume, at the end of blood letting, and at 3 min after administration ( at the corre-sponding time point in group HS) . Results The MAP and amplitudes of MEP and SSEP were significantly decreased at the end of blood letting than at the time of withdrawing 20％ of the blood volume in three groups (P<0. 05). Compared with HS group, the MAP and amplitudes of MEP and SSEP were significantly in-creased at 3 min after administration in E group, and the MAP was increased at 3 min after administration ( P<0. 05 or 0. 01) , and no significant change was found in amplitudes of MEP and SSEP in P group ( P>0. 05) . Conclusion Epinephrine exerts no effect on evoked potential monitoring, however, phenylephrine affects the accuracy of evoked potential monitoring in a rat model of acute hemorrhagic shock.
Effect of epinephrine on the absorption of lidocaine following application to the oral mucosa in rats

Abstract:Abstract Background We investigated the role of epinephrine in prolonging the localization of lidocaine on the oral mucosa and inhibiting its absorption in the blood of rats. Methods We used 7–8-week-old pathogen-free Wistar male rats (n = 128) for our study. We divided them into the control group administered with 14 C-labeled lidocaine hydrochloride gel only and the study group administered with 14 C-labeled lidocaine hydrochloride gel with epinephrine. The medications were administered in the palatal mucosa of the rats. The amount of mucosa, palatine bone, and serum lidocaine was measured by radioactivity using a liquid scintillation counter and was observed using autoradiograms. Results Initially, there was no significant difference in the lidocaine levels between the lidocaine and lidocaine with epinephrine groups in the palatal mucosa (751.9 ± 133.8 vs. 669.8 ± 101.6 ng/mg [2 min]). After 4 min, the values were significantly lower in the lidocaine with epinephrine group (1040.0 ± 142.8 vs. 701.2 ± 109.0 ng/mg [20 min]). After 40 min, the lidocaine level became significantly higher in the lidocaine with epinephrine group (586.8 ± 112.4 vs. 1131.3 ± 155.2 ng/mg [40 min]). Similar results were observed in the palatine bone and serum. Conclusion Epinephrine prolonged the localization of lidocaine applied to the mucosa and inhibited its absorption into the bloodstream of rats. Clinical studies are required to evaluate the use of epinephrine-containing topical anesthetics on the oral mucosa.

Abstract:Objective Hemodynamic effect of epinephrine is associated with its dose,infusion rate and route of administration.The present study was to observe the hemodynamic effect of low-dose epinephrine intravenously injected before cardiopulmonary bypass(CPB) and added in blood reservoir during CPB.Methods We established a model of CPB in 36 SD male rats and randomized them into 3 groups of equal number: an ES group,intravenously injected with epinephrine 0.5 μg/kg before CPB and saline 0.4 mL added in blood reservoir during CPB;an SE group,injected with saline 0.4 mL before CPB and epinephrine 0.5 μg/kg added in blood reservoir during CPB;and an SS group: injected with saline 0.4 mL before CPB and added in blood reservoir during CPB.We recorded the mean arterial pressure(MAP) and heart rate(HR) at 0 s(baseline),5s,10s,15s,30s,1 min,2 min,3 min,4 min and 5 min after the injection of epinephrine or saline before and during CPB.Results Before CPB,MAP showed no significant changes in the SS and SE groups but increased at 5s(P0.05),decreased at 15s and 30 s(P0.05),and returned to the baseline level at 1 min in the ES group as compared with the baseline.During CPB,MAP exhibited no significant changes in the SS and ES groups but increased from 15s to 1 min in the SE group in comparison with the baseline(P0.05).Conclusion Intravenous injection of epinephrine 0.5 μg/kg elicited biphasic changes in MAP,first increased and then decrease before CPB.Epinephrine 0.5 μg/kg added in blood reservoir induced an obvious increase in MAP during CPB.
Preparation of C57/BL6 mice myocardial infarction model and method of improving survival rate

Abstract:Objective To investigate how to develop a credible,steady and repeatable rat model of acute myocardial infarction with a low mortality,improve the method and decreased those factors that influence death rate of creating the model.Methods 40 C57/BL6 mice were anesthetized by intraperitoneal injection of pentobarbital sodium.After tracheotomy intubations,respiration machine was linked.Left anterior thoracotomy through 3,4 intercostal region was performed,and then the left anterior descending coronary arteries were ligated to produce acute myocardial infarction.During the operation,some details to affect rat mortality were analyzed and improved,which involving anesthesia,artificial respiration,thoracotomy and area of ligating the left anterior descending coronary artery,etc.Results The model of acute myocardial infarction was established successfully.Following the details were highlighted,survival rate was 95% after operation and 92.5% beyond three weeks.Conclusion C57/BL6 mice acute myocardial infarction model can be made quickly by this way and the model's survival rate was improved effectively.

Abstract:Cardiovascular disease (CVD) is the main cause of death worldwide, with myocardial infarction (MI) being the most prevalent pathology involved in CVD. MI is characterized by a deficiency in oxygen supply to the myocardium, thereby promoting ventricular remodeling of the ischemic and remote zone of the heart. Cardiac remodeling associated with MI could promote the development of heart failure and finally death. For these reasons, it is important to develop animal models that mimic human cardiac disease which could help to identify new mechanisms involved in the pathology and, consequently, develop new therapeutic strategies. We herein describe in detail a protocol for MI induction with low mortality rate (<15%) in rats by ligation of the left anterior descending artery. In addition, we also describe two imaging techniques which allow to evaluate cardiac structure and function—including deformation parameters in rats such as transthoracic echocardiography and cardiac magnetic resonance. This animal model could be useful for acute and chronic studies and for evaluating the potential usefulness of different treatments.
