Abstract:A multivalent recognition and alkaline phosphatase (ALP)-responsive electrogenerated chemiluminescence (ECL) biosensor for cancer cell detection and in situ evaluation of cell surface glycan expression was developed on a poly(amidoamine) (PAMAM) dendrimer-conjugated, chemically reduced graphene oxide (rGO) electrode interface. In this strategy, the multivalency and high affinity of the cell-targeted aptamers on rGO provided a highly efficient cell recognition platform on the electrode. The ALP and concanavalin A (Con A) coated gold nanoparticles (Au NPs) nanoprobes allowed the ALP enzyme-catalyzed production of phenols that inhibited the ECL reaction of Ru(bpy)(3)(2+) on the rGO electrode interface, affording fast and highly sensitive ECL cytosensing and cell surface glycan evaluation. Combining the multivalent aptamer interface and ALP nanoprobes, the ECL cytosensor showed a detection limit of 38 CCRF-CEM cells per mL in human serum samples, broad dynamic range and excellent selectivity. In addition, the proposed biosensor provided a valuable insight into dynamic profiling of the expression of different glycans on cell surfaces, based on the carbohydrates recognized by lectins applied to the nanoprobes. This biosensor exhibits great promise in clinical diagnosis and drug screening.

Abstract:<p>Biomarkers play an important role in disease diagnosis and prognosis, which demand reliable, sensitive, rapid, and economic detection platform to conduct simultaneous multiple-biomarkers analysis in serum or body liquid. Here, we developed a universal biosensing platform through integrating the advantages of unique nanostructure and biochemistry properties of graphene oxide quantum dots and high throughput and low cost of microfluidic chip for reliable and simultaneous detection of multiple cancer antigen and antibody biomarkers. The performance of the proposed biosensing platform is validated through the representative cancer biomarkers including carcino-embryonic antigen (CEA), carbohydrate antigen 125 (CA125), α-fetoprotein (AFP), carbohydrate antigen 199 (CA199) and carbohydrate antigen 153 (CA153). It has a large linear quantification detection regime of 5-6 orders of magnitude and an ultralow detection limit of 1 pg/mL or 0.01 U/mL. Moreover, the proposed biosensing chip is capable of conducting 5-20 kinds of biomarkers from at least 60 persons simultaneously in 40 minutes with only 2 μL serum of each patient, which essentially reduces the detection cost and time to at least 1/60 of current popular methods. Clinical breast cancer and healthy samples detection results indicated its promising perspective in practical applications including cancer early diagnosis, prognosis, and disease pathogenesis study.</p>

Abstract:With a unique chemical enhancement capability, graphene oxide is exceptionally suitable to serve as an alternative surface-enhanced Raman scattering (SERS) substrate, which is usually defined as GERS. However, such a GERS matrix with both promising uniformity and molecule enrichment ability has not been applied in the quantitative detection of tumor markers. Herein, an ultrasensitive and specific immunoassay of carcinoembryonic antigens mediated by the GERS matrix was demonstrated. With the assistance of Au NRs as immunoprobes, a reliable limit of detection as low as 3.01 pg/mL was finally achieved because of the collective effect of chemical and electromagnetic enhancements. Meanwhile, a calibration curve with a high R2 value of 0.996 was obtained in the range from 1 μg/mL to 10 pg/mL. These results exhibit that the GERS matrix has a great promise for participating in immunoassay, which may pave a potential avenue for the real utilization of the SERS technique in clinical biomedicine.

Abstract:AbstractBy dint of unique physical/chemical properties and bio-compatibility, graphene can work as a building block for a SERS substrate and open up a unique platform for tumor cells detection with high sensitivity. Herein we demonstrate a facile system with highly enhanced surface enhanced Raman spectroscopy of graphene (G-SERS). The system consists of a reduced graphene oxide (rGO) sandwiched by silver and gold nanostructures. Due to the ultrasmall thickness of rGO, the inter-coupling between Ag and Au nanoparticles is precisely controlled and the local field enhancement has been improved to more than 70 times. Associated with the unique chemical mechanism of rGO, the hybrid system has been utilized to identify tumor cells without using any biomarkers. We believe this research will be important for the applications of rGO in cancer screening.
In-Situ Integration of 3D C-MEMS Microelectrodes with Bipolar Exfoliated Graphene for Label-Free Electrochemical Cancer Biomarkers Aptasensor

Abstract:The electrochemical label-free aptamer-based biosensors (also known as aptasensors) are highly suitable for point-of-care applications. The well-established C-MEMS (carbon microelectromechanical systems) platforms have distinguishing features which are highly suitable for biosensing applications such as low background noise, high capacitance, high stability when exposed to different physical/chemical treatments, biocompatibility, and good electrical conductivity. This study investigates the integration of bipolar exfoliated (BPE) reduced graphene oxide (rGO) with 3D C-MEMS microelectrodes for developing PDGF-BB (platelet-derived growth factor-BB) label-free aptasensors. A simple setup has been used for exfoliation, reduction, and deposition of rGO on the 3D C-MEMS microelectrodes based on the principle of bipolar electrochemistry of graphite in deionized water. The electrochemical bipolar exfoliation of rGO resolves the drawbacks of commonly applied methods for synthesis and deposition of rGO, such as requiring complicated and costly processes, excessive use of harsh chemicals, and complex subsequent deposition procedures. The PDGF-BB affinity aptamers were covalently immobilized by binding amino-tag terminated aptamers and rGO surfaces. The turn-off sensing strategy was implemented by measuring the areal capacitance from CV plots. The aptasensor showed a wide linear range of 1 pM-10 nM, high sensitivity of 3.09 mF cm-2 Logc-1 (unit of c, pM), and a low detection limit of 0.75 pM. This study demonstrated the successful and novel in-situ deposition of BPE-rGO on 3D C-MEMS microelectrodes. Considering the BPE technique's simplicity and efficiency, along with the high potential of C-MEMS technology, this novel procedure is highly promising for developing high-performance graphene-based viable lab-on-chip and point-of-care cancer diagnosis technologies.

Abstract:Rapid and efficient measurement of cancer cells is a major challenge in early cancer diagnosis. In the present study, a miniature multiplex chip was created for in situ detection of cancer cells by implementing a novel graphene oxide (GO)-based Förster resonance energy transfer (FRET) biosensor strategy, i.e. assaying the cell-induced fluorescence recovery from the dye-labeled aptamer/graphene oxide complex. Fluorescence intensity measurement and image analyses demonstrated that this microfluidic biosensing method exhibited rapid, selective and sensitive fluorescence responses to the quantities of the target cancer cells, CCRF-CEM cells. Seven different cancer cell samples can be measured at the same time in such a microfluidic chip. The linear response for target CCRF-CEM cells in a concentration range from 2.5 × 10(1) to 2.5 × 10(4) cells mL(-1) was obtained, with a detection limit about 25 cells mL(-1), which is about ten times lower than those of normal biosensors. The novel fluorescence biosensing microfluidic chip supplies a rapid, visible and high-throughput approach for early cancer diagnosis with high sensitivity and specificity.

Abstract:Summary Sirolimus is a potent immunosuppressive agent with a novel mechanism of action. It inhibits the transduction of cytokine signals necessary for the proliferation and maturation of T cells. Because sirolimus blocks a broad spectrum of cytokine signals, it seems logical to use it as an adjunct to CsA-based immunosuppression. The high degree of synergy between these two agents, as suggested by the rigorous median-effect analysis, has been confirmed by a reduced rate of rejection episodes among human renal allograft recipients. However, Phase I studies document wide interindividual variation in the pharmacokinetic parameters of 26 stable renal transplant patients, thereby suggesting that optimal therapy may require monitoring of drug concentrations, which is a task that has been somewhat simplified by the good correlation of trough level to AUC. Development of a monoclonal antibody assay system may simplify the monitoring of drug concentrations further. Additional studies of sirolimus will be required to determine the therapeutic concentrations and ratios of sirolimus to CsA that provide optimal immunosuppression, and to assess the possibility of a steroid-free regimen.

Abstract:Graphene is an emerging nanomaterial increasingly being used in electrochemical biosensing applications owing to its high surface area, excellent conductivity, ease of functionalization, and superior electrocatalytic properties compared to other carbon-based electrodes and nanomaterials, enabling faster electron transfer kinetics and higher sensitivity. Graphene electrochemical biosensors may have the potential to enable the rapid, sensitive, and low-cost detection of cancer biomarkers. This paper reviews early-stage research and proof-of-concept studies on the development of graphene electrochemical biosensors for potential future cancer diagnostic applications. Various graphene synthesis methods are outlined along with common functionalization approaches using polymers, biomolecules, nanomaterials, and synthetic chemistry to facilitate the immobilization of recognition elements and improve performance. Major sensor configurations including graphene field-effect transistors, graphene modified electrodes and nanocomposites, and 3D graphene networks are highlighted along with their principles of operation, advantages, and biosensing capabilities. Strategies for the immobilization of biorecognition elements like antibodies, aptamers, peptides, and DNA/RNA probes onto graphene platforms to impart target specificity are summarized. The use of nanomaterial labels, hybrid nanocomposites with graphene, and chemical modification for signal enhancement are also discussed. Examples are provided to illustrate applications for the sensitive electrochemical detection of a broad range of cancer biomarkers including proteins, circulating tumor cells, DNA mutations, non-coding RNAs like miRNA, metabolites, and glycoproteins. Current challenges and future opportunities are elucidated to guide ongoing efforts towards transitioning graphene biosensors from promising research lab tools into mainstream clinical practice. Continued research addressing issues with reproducibility, stability, selectivity, integration, clinical validation, and regulatory approval could enable wider adoption. Overall, graphene electrochemical biosensors present powerful and versatile platforms for cancer diagnosis at the point of care.
Electroporation-Assisted Surface-Enhanced Raman Detection for Long-Term, Label-Free, and Noninvasive Molecular Profiling of Live Single Cells

Abstract:Molecule characterization of live single cells is greatly important in disease diagnoses and personalized treatments. Conventional molecule detection methods, such as mass spectrography, gene sequencing, or immunofluorescence, are usually destructive or labeled and unable to monitor the dynamic change of live cellular molecules. Herein, we propose an electroporation-assisted surface-enhanced Raman scattering (EP-SERS) method using a microchip to implement label-free, noninvasive, and continuous detections of the molecules of live single cells. The microchip containing microelectrodes with nanostructured EP-SERS probes has a multifunction of cell positioning, electroporation, and SERS detection. The EP-SERS method capably detects both the intracellular and extracellular molecules of live single cells without losing cell viability so as to enable long-term monitoring of the molecular pathological process in situ. We detect the molecules of single cells for two breast cancer cell lines with different malignancies (MCF-7 and MDA-MB-231), one liver cancer cell line (Huh-7), and one normal cell line (293T) using the EP-SERS method and classify these cell types to achieve high accuracies of 91.4-98.3% using their SERS spectra. Furthermore, 24 h continuous monitoring of the heterogeneous molecular responses of different cancer cell lines under doxorubicin treatment is successfully implemented using the EP-SERS method. This work provides a long-term, label-free, and biocompatible approach to simultaneously detect intracellular and extracellular molecules of live single cells on a chip, which would facilitate research and applications of cancer diagnoses and personalized treatments.
A highly sensitive nanobiosensor based on aptamer-conjugated graphene-decorated rhodium nanoparticles for detection of HER2-positive circulating tumor cells

Abstract:Abstract Assessment of human epidermal growth factor receptor-2 (HER2) tumor marker status is an impressive factor in screening, diagnosing and monitoring breast cancer (BC). The electrochemical biosensor is a revolutionary method in cancer diagnosis, which is used in this research to detect HER2 + circulating tumor cells. The electrochemical activity, size, shape, and morphology of the synthesized nanomaterials were analyzed. The hybrid nanocomposite established by the coupling of reduced graphene oxide nanosheets (rGONs) and rhodium nanoparticles (Rh-NPs) on the surface of graphite electrode resulted in improved surface area, electrochemical activity, and biocompatibility. The graphite electrode-based aptasensor (g-aptasensor) demonstrated exceptional performance against HER2-overexpressed SKBR3 cancer cells, with a linear dynamic range of 5.0 to 10.0 × 10 4 cells/mL, an analytical limit of detection (LOD) as low as 1.0 cell/mL, and a limit of quantification (LOQ) of 3.0 cells/mL. The G-rich DNA aptamers can fold into an intermolecular G-quadruplex, which specifically bind to the target molecule. Consequently, the advantages of this highly efficient nanocomposite platform include broad dynamic range, high specificity, selectivity, stability, reproducibility, and low cost. These characteristics indicate that the fabricated nanobiosensor has a high potential for use in detecting and monitoring HER2 level for the care of BC patients and clinical diagnosis.

Abstract:Evaluation of extracellular domain of human epidermal growth factor receptor-2 (HER2-ECD) oncomarker status is an impressive factor in screening, diagnosing and monitoring early-stage breast cancer (BC). Electrochemical aptamer-based nanobiosensor with high sensitivity and selectivity for quantitative and qualitative measurement of HER2-ECD oncomarker was developed. In this study, the nanocomposite made by distinct materials included reduced graphene oxide nano-sheets (rGONs) and rhodium nanoparticles (Rh-NPs) on the graphite electrode (GE) surface. This structure resulted in amplified electrochemical activity, high surface area, stability, and bio-compatibility. Each of the steps of preparing nanomaterials and setting up biosensor were carefully examined by analytical and electrochemical techniques. Various modified electrodes were constructed and analyzed in terms of electrochemical performance, morphology, size, and shape of nanomaterials. The GE-based aptasensor had a noteworthy and conducive results against HER2-ECD with a wide dynamic range of 10.0-500.0 ng/mL, a low limit of detection (LOD) of 0.667 ng/mL (significantly less than the clinical cut-off), and a low limit of quantification (LOQ) of 2.01 ng/mL. The benefits provided by this aptasensor such as broad dynamic range, high sensitivity, selectivity, stability, reproducibility, and low cost suggest tremendous potential for non-invasive detection and monitoring of the HER2-ECD levels of BC care and clinical diagnosis.

Abstract:Starting from an earlier benchmark analytical calculation of the luminosity process ${e}^{+}{e}^{\ensuremath{-}}\ensuremath{\rightarrow}{e}^{+}{e}^{\ensuremath{-}}+(\ensuremath{\gamma})$ at the SLAC Linear Collider (SLC) and the CERN ${e}^{+}{e}^{\ensuremath{-}}$ collider LEP, we use the methods of Yennie, Frautschi, and Suura to develop an analytical improved naive exponentiated formula for this process. The formula is compared to our multiple-photon Monte Carlo event generator bhlumi (1.13) for the same process. We find agreement on the overall cross-section normalization between the exponentiated formula and bhlumi below the 0.2% level. In this way, we obtain an important cross-check on the normalization of our higher-order results in bhlumi and we arrive at formulas which represent the LEP/SLC luminosity process in the below 1% ${Z}^{0}$ physics tests of the $\mathrm{SU}{(2)}_{L}\ifmmode\times\else\texttimes\fi{}\mathrm{U}(1)$ theory in complete analogy with the famous high-precision ${Z}^{0}$ line-shape formulas for the ${e}^{+}{e}^{\ensuremath{-}}\ensuremath{\rightarrow}{\ensuremath{\mu}}^{+}{\ensuremath{\mu}}^{\ensuremath{-}}$ process discussed by Berends et al., for example.

Abstract:Abstract Background In this label-free bioassay, an electrochemiluminescence (ECL) immunosensor was developed for the quantification of breast cancer using HER-2 protein as a metastatic biomarker. Method For this purpose, the ECL emitter, [Ru(bpy) 3 ] 2+ , was embedded into biocompatible chitosan (CS) polymer. The prepared bio-composite offered high ECL reading due to the depletion of human epidermal growth factor receptor 2 (HER-2) protein. Reduced graphene oxide (rGO) was used as substrate to increase signal stability and achieve greater sensitivity. For this, rGO was initially placed electrochemically on the glassy carbon electrode (GCE) surface by cyclic voltammetry (CV) technique. Next, the prepared CS/[Ru(bpy) 3 ] 2+ biopolymer solution was coated on a drop of the modified electrode such that the amine groups of CS and the carboxylic groups of rGO could covalently interact. Using EDC/NHS chemistry, monoclonal antibodies (Abs) of HER-2 were linked to CS/[Ru(bpy) 3 ] 2+ /rGO/GCE via amide bonds between the carboxylic groups of Ab molecules and amine groups of CS. The electrochemical behavior of the electrode was studied using different electrochemical techniques such as electrochemical impedance spectroscopy (EIS), differential pulse voltammetry (DPV) and square wave voltammetry (SWV) and also ECL tests. Results After passing all optimization steps, the lower limit of detection (LLOQ) and linear dynamic range (LDR) of HER-2 protein were practically obtained as 1 fM and 1 fM to 1 nM, individually. Importantly, the within and between laboratory precisions were performed and the suitable relative standard deviations (RSDs) were recorded as 3.1 and 3.5%, respectively. Conclusions As a proof of concept, the designed immunosensor was desirably applied for the quantification of HER-2 protein in breast cancer suffering patients. As a result, the designed ECL-based immunosensor has the capability of being used as a conventional test method in biomedical laboratories for early detection of HER-2 protein in biological fluids. Abstract:Two-dimensional (2D) graphene, sp2-hybridized carbon, and its two major derivatives, graphene oxide (GO) and reduced graphene oxide (rGO) have played an important role in immunoassays (IAs) and immunosensing (IMS) platforms for the detection of carcinoembryonic antigen (CEA), an implicated tumor biomarker found in several types of cancer. The graphene family with high surface area is functionalized to form stable nanocomposites with gold nanoparticles (AuNPs) and electron mediators. The capture anti-CEA antibody (Ab) with high density can be anchored on AuNPs of such composites to provide remarkable detection sensitivity, significantly below the level found in normal subjects and cancer patients. Electrochemical and fluorescence/chemiluminescence-quenching properties of graphene-based nanocomposites are exploited in various detection schemes. Future endeavors are envisioned for the development of an array platform with high-throughput for CEA together with other tumor biomarkers and C-reactive protein, a universal biomarker for infection and inflammation. The ongoing efforts dedicated to the replacement of a lab-based detector by a cellphone with smart applications will further enable cost-effective and frequent monitoring of CEA in order to establish its clinical relevance and provide tools for real-time monitoring of patients during chemotherapy.
Laser-scribing graphene-based electrochemical biosensing devices for simultaneous detection of multiple cancer biomarkers

Abstract:In this study, a graphene electrochemical sensor based on laser graphene polymer material was proposed to induce graphene formation on polyimide substrates via fiber laser. The laser produces stable power and results to achieve the benefits of consistency, conductivity, and flexibility. The electrochemical three-electrodes were manufactured on polyimide to replace the traditional three-electrodes by achieving small size and portability. An electrode activation is the modification of laser-scribed graphene electrodes (LSG) to facilitate the binding of liver cancer sites. The evaluation is performed by differential pulsed Voltammetry (DPV) to detect cancer proteins in the phosphate buffer saline (PBS) buffers and serum. In a serum environment, the concentrations of alpha-fetoprotein (AFP) and Carcinoembryonic antigen (CEA) were detected from 0.75 ng ml −1 to 100 ng ml −1 , AFP and CEA electrodes have a good linear range (R 2 = 0.96 and R 2 = 0.98), indicating the sensor's sensitivity and specificity for cancer detection. In addition, two types of carcinogenic proteins were monitored in the PBS buffer and successfully detected in this experiment. Based on the results, the appropriate LSG sensor may be used for monitoring with limited resources. Multiplex detection of pancreatic cancer biomarkers using a SERS-based immunoassay

Abstract:Early diagnosis of pancreatic cancer (PC) is critical to reduce the mortality rate of this disease. Current biological analysis approaches cannot robustly detect several low abundance PC biomarkers in sera, limiting the clinical application of these biomarkers. Enzyme linked immunosorbent assay and radioimmunoassay are two common platforms for detection of biomarkers; however, they suffer from some limitation. This study demonstrates a novel system for multiplex detection of pancreatic biomarkers CA19-9, MMP7 and MUC4 in sera samples with high sensitivity using surface enhanced Raman spectroscopy. Measuring the levels of these biomarkers in PC patients, pancreatitis patients, and healthy individuals reveals the unique expression pattern of these markers in PC patients, suggesting the great potential of using this approach for early diagnostics of PCs.
Aptamer-Functionalized and Gold Nanoparticle Array-Decorated Magnetic Graphene Nanosheets Enable Multiplexed and Sensitive Electrochemical Detection of Rare Circulating Tumor Cells in Whole Blood

Abstract:The identification and monitoring of circulating tumor cells (CTCs) in human blood plays a pivotal role in the convenient diagnosis of different cancers. However, it remains a major challenge to monitor these CTCs because of their extremely low abundance in human blood. Here, we describe the synthesis of a new aptamer-functionalized and gold nanoparticle (AuNP) array-decorated magnetic graphene nanosheet recognition probe to capture and isolate rare CTCs from human whole blood. In addition, by employing the aptamer/electroactive species-loaded AuNP signal amplification probes, multiplexed electrochemical detection of these low levels of CTCs can be realized. The incubation of the probes with the sample solutions containing the target CTCs can lead to the efficient separation of the CTCs and result in the generation of two distinct voltammetric peaks on a screen-printed carbon electrode, whose potentials and current intensities, respectively, reflect the identity and number of CTCs for the multiplexed detection of the Ramos and CCRF-CEM cells with detection limits down to 4 and 3 cells mL-1. With the successful demonstration of the concept, further extension of the developed sensing strategy for the determination of various CTCs in human whole blood for the screening of different cancers can be envisioned in the near future.

Abstract:Biomarker testing plays a crucial role in the early detection of liver cancer. Herein, we developed a dual-signal amplification approach utilizing magnetic aggregation and a recognition competition strategy for the simultaneous detection of alpha-fetoprotein (AFP) and manganese superoxide dismutase (MnSOD) in serum. 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 were synthesized by functionalizing Raman signaling molecules and aptamer complementary chains onto the surface of gold nanoparticles (AuNPs). The detection complex Raman signal molecule@AuNPs@H-Fe3O4@cDNA was assembled by conjugating 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 with two nucleic acid aptamers (cDNA1 and cDNA2) modified with Fe3O4 through partial base complementary pairing. The target protein exhibited specific binding with the aptamer, leading to the competitive displacement of 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 from the Fe3O4 array surface, consequently resulting in a reduction of the Surface-Enhanced Raman Spectroscopy (SERS) signal. Through this approach, the limit of detection (LOD) for AFP and MnSOD in serum was achieved at remarkably low levels of 5.89 pg/mL and 6.23 pg/mL, respectively, with a reaction incubation period of only 5 min. Finally, the platform was utilized for the quantification of AFP and MnSOD in the serum of a nude mouse hepatocellular carcinoma model. The results obtained through SERS were consistent with those from enzyme-linked immunosorbent assay (ELISA), validating its accuracy. This methodology presents a novel approach for the swift and concurrent detection of proteins, holding significant clinical promise for the early diagnosis of hepatocellular carcinoma.

Abstract:In this report, a label-free reflectometric interference spectroscopy (RIfS) based microchip biosensor for the detection of circulating tumour cells (CTCs) is demonstrated. Highly ordered nanoporous anodic aluminium oxide (AAO) fabricated by electrochemical anodization of aluminium foil was used as the RIfS sensing platform. Biotinylated anti-EpCAM antibody that specifically binds to human cancer cells of epithelial origin such as pancreatic cancer cells (PANC-1) was covalently attached to the AAO surface through multiple surface functionalization steps. Whole blood or phosphate buffer saline spiked with low numbers of pancreatic cancer cells were successfully detected by specially designed microfluidic device incorporating an AAO RIfS sensor, without labour intensive fluorescence labelling and/or pre-enhancement process. Our results show that the developed device is capable of selectively detecting of cancer cells, within a concentrations range of 1000-100,000 cells/mL, with a detection limit of <1000 cells/mL, a response time of <5 min and sample volume of 50 μL of. The presented RIfS method shows considerable promise for translation to a rapid and cost-effective point-of-care diagnostic device for the detection of CTCs in patients with metastatic cancer.

Abstract:A novel reusable and dual-potential responsive electrogenerated chemiluminescence (ECL) biosensor was fabricated for synchronous detection of cancer cells and their surface N-glycan. In this strategy, a cancer cell recognized aptamer hybridized with a capture DNA was immobilized on electrochemically reduced MoS2 nanosheets, and Ru(phen)3(2+) as ECL probes was intercalated into the grooves of the double-strand DNA. In the presence of target cells, the capture DNA and Ru(phen)3(2+) were released from the electrode interface owing to the specific interaction between cancer cells and the aptamer. Meanwhile, concanavalin A (Con A), a mannose binding protein, and a conjugated gold nanoparticle modified graphite-C3N4 (Con A@Au-C3N4) was used as a negative ECL nanoprobe and applied for the cell surface N-glycan evaluation owing to the excellent ECL properties of g-C3N4 at negative potential. The cytosensing and cell surface N-glycan evaluation could be simultaneously realized with high sensitivity and excellent selectivity based on the ratio of ECL intensity between the negative potential and positive potential (ΔECLn/ΔECLp), avoiding the traditional routing cell counting procedures. Moreover, the aptamer modified electrode can be regenerated in the presence of capture DNA solutions for cyclic utilization. As a proof-of-concept, the ECL cytosensor showed excellent performances for the analysis of the MCF-7 cancer cell and its surface N-glycan evaluation in human serum samples. The reusable and dual potential response ECL biosensor endows a feasibility tool for clinical diagnosis and drug screening especially in complex biological systems.
