Abstract:BACKGROUND: The present study was conducted to investigate the effects of dietary novel alkaline protease from Bacillus licheniformis on the growth performance, meat quality, antioxidant status and intestinal morphology of broilers. In total, 4000 broilers were randomly assigned into five groups and treated with normal control, normal control + 100 mg kg(-1) protease, normal control + 200 mg kg(-1) protease, normal control + 300 mg kg(-1) protease and normal control + 400 mg kg(-1) protease. RESULTS: Supplementing protease impacted final body weight (linear, P = 0.003; quadratic, P = 0.006) and decreased feed conversion rate (linear, P = 0.036) in broilers. Moreover, dietary protease significantly increased breast muscle rate (linear, P = 0.005; quadratic, P = 0.021) and decreased drip loss (linear, P < 0.001; quadratic, P < 0.001). In addition, dietary protease notably increased protein digestibility (linear, P = 0.001; quadratic, P = 0.006) and trypsin activity (linear, P = 0.002; quadratic, P = 0.009) in jejunum. Light microscopy revealed that the jejunum villi in the 300 mg kg(-1) and 400 mg kg(-1) groups exhibited greater height and a denser arrangement compared to those in the control group. The addition of protease decreased malondialdehyde content (linear, P < 0.001; quadratic, P < 0.001) and increased total antioxidant capacity (linear, P = 0.001; quadratic, P < 0.001) in pectoral muscles. CONCLUSION: The results of the present study suggest that dietary novel alkaline protease from B. licheniformis improved growth performance by affecting trypsin activity, protein digestibility, antioxidant capacity and intestinal health. Abstract:Background Different types of exogenous protease supplements have a positive impact on animal performance, but their effects on the nutritional value of meat and the gut microbial community of broilers have not been extensively studied. The objective of this investigation was to determine the impact of supplementation with a novel alkaline protease derived from Bacillus licheniformis (at doses of 0, 100, 200, 300, and 400 g/t) on the fatty acid and amino acid profiles, inosine monophosphate (IMP) levels, total volatile basic nitrogen (TVB-N) content found within the breast muscle, as well as the impact on the cecal microbiota and metabolites. Results Supplementation with 200–400 g/t of the novel protease resulted in a significant elevation in the concentration of essential amino acids ( P < 0.001), flavor amino acids ( P < 0.001), and total protein ( P = 0.013) within the breast muscle. Results derived from the 16S rRNA sequencing and untargeted metabolomics analysis of the cecal content revealed that the novel protease reshaped the cecal microbial and metabolite profiles. In particular, it led to increased relative abundances of Bacteroides , Lactobacillus , Alistipes , and Eubacterium , while simultaneously causing a reduction in the metabolites of D-lactic acid and malonic acid. Moreover, correlation analyses unveiled significant relationships between distinct microbes and metabolites with the contents of IMP, fatty acids, and amino acids in the broiler's breast muscle. Conclusion In summary, the novel protease regulated the intestinal microbial community and metabolism, thereby inducing changes in the compositions of fatty acids and amino acids profiles, as well as IMP levels in broiler meat. These alterations significantly contributed to the enhancement of the nutritional value and flavor of the meat. Graphical abstract

Abstract:The tenderization effect of a new elastase from Bacillus sp. EL31410 was investigated on beef meat. Meat tenderization was done by dipping the meat cut in a solution containing proteolytic enzymes after freeze-dehydration. It was found that a marination time of 4h was enough for enzyme adsorption. The samples were treated for 4h in different enzyme solutions and then was stored at 4°C for 24, 48, 72h, and subjected to texture measurement, sensory evaluations, biochemical analysis and histological observations. A marked decrease in hardness, by texture measurements, was observed in the meats with papain and elastase and higher sensory scores for tenderness were observed in the meats treated with enzymes than in the control. The papain-treated beef meat received the highest score for tenderness, but the scores given for juiciness and taste were lower than that of the control. Rapid increases of fragmentation of myofibrils from the enzyme-treated meat were observed in the first 24h of storage, especially for papain-treated meat. Meantime, elastin of myofibrilar structure was selectively degraded by elastase compared with the control when stored at 4°C for 48h as shown by electron microscopy. These findings suggest that Bacillus elastase (EL31410) is a promising substitute for papain as a favourable meat tenderizer.

Abstract:This study aims to determine the effectiveness of papaya‐fruit latex and yellow‐senesced leaves as a natural and organic tenderizer. The fruit and leaves of the plant were ground to powder, while 0 g, 10 g, 15 g and 20 g variations were used to cover 50 g of meat for 4 h. Subsequently, the Bradford and Kjeldahl methods were used to determine the protein content, while the protein profile was analyzed using SDS‐PAGE and confirmed using a Scanning Electron Microscope (SEM). The results showed that the protein concentration in mutton after fruit latex treatment was 41%, which was higher than the concentration of beef at 29.86%. Furthermore, the beef lost protein bands and its molecular weight fell from 225 kDa to 86 KDa, while the mutton experienced a reduction from 100 kDa to 65 kDa, which was significantly smaller than for raw meat. A single protein band was also observed at 21.6 kDa in the sample, indicating the presence of papain enzyme protein. Meanwhile, the SEM results showed that collagen and myofibril in the muscles were damaged in the treated meats. Based on these results, treatment with papaya fruit latex and yellow papaya leaves increases the tenderness of meat.

Abstract:Sous vide meat is an emerging food category, the consumption of which has increased owing to greater convenience, sensory traits, elderly consumers acceptance, and low-cost cuts use. However, required prolonged thermal treatment to achieve desired tenderness, impact energy-consumption besides triggering lipid oxidation, undesired off-flavors, and cooked meat profiles. Using a response surface methodology (RSM), this study evaluated the effects of the vegetal proteolytic papain (0 to 20 mg/kg) and low-temperature sous vide cooking (SVC) time (1 to 8 h at 65°C) in low-value marinated M. semitendinosus beefsteaks on technological characteristics associated with tenderness, and lipid oxidation. Additionally, the sensory profile traits of the pre-selected treatments were described using check-all-that-apply (CATA) and preference mapping. Shear force (WBsSF) was reduced with greater papain addition, whereas higher cooking losses (CL) were observed with longer SVC cooking times. Both the released total collagen and TBARS values increased with increasing papain concentrations and SVC times. Combining high levels of papain (>10 mg/kg) and SVC time (>6 h) resulted in lower WBsSF values (<20 N) but higher CL (>27%) and the CATA descriptors "aftertaste" and "mushy." The optimized conditions (14 mg/kg papain; 2 h SVC) also reduced WBsSF values (<26 N) with lower CL (<20%) and were most preferred and described as "juicy" and "tender" by consumers. Observed results suggest that combined mild SVC and papain may potentiate tenderness, conjointly favor juiciness and oxidation, further representing a promising tool for reducing SVC time without compromising valued sous vide sensory traits.

Abstract:Objective: Given the social constraints imposed by a gluten-free diet, it can be hypothesized that children with celiac disease (CD) living in the United States have a reduced health-related quality of life (HRQOL); however, there is no validated CD-specific HRQOL instrument for children living in the United States. The goals of this study were to develop and validate a CD-specific HRQOL instrument for children 8 to 18 years of age with CD and to report HRQOL in these children using both generic- and disease-specific instruments. Methods: This was a prospective study using focus group methodology to develop a CD-specific HRQOL instrument that was then administered to children 8 to 18 years of age with CD living throughout the United States. Instrument validation methods included construct, convergent, and divergent validities. Results: Two instruments were developed: CD-specific pediatric HRQOL instrument (CDPQOL) 8 to 12 and CDPQOL 13 to 18. A total of 181 children with CD completed the CDPQOL as well as a comparator generic instrument. Exploratory factor analysis restructured the CDPQOL and reduced the total number of items. The CDPQOL showed a moderate agreement with the Psychosocial dimensions of the generic instrument confirming convergent validity and low-to-moderate agreement with the Physical Health Summary dimension of the generic instrument confirming divergent validity. Conclusions: The CDPQOL, consisting of 13 to 17 questions, is a validated instrument for the measurement of HRQOL in children 8 to 18 years of age with CD living in the United States.

Abstract:This study investigates the impact of bromelain, a plant enzyme, on the physicochemical and sensory properties of horse meat, as well as the effects of different bacterial cultures (Lactococcus lactis, Lactococcus lactis subsp. lactis biovar diacetylactis, Lactobacillus acidophilus, and Bifidobacterium longum) on the inhibition of lipid oxidation and control of pH during chilled storage. Horse meat (longissimus dorsi) samples (n = 14) were treated with bromelain in two forms (powder and aqueous solution) and with three methods: immersion in enzyme solution, spreading enzyme powder on meat, and syringing enzyme solution into the meat. After fermentation, a part of the meat samples (n = 6) was treated with different bacteria compositions at a 5% weight ratio and stored at 0–2 °C for 6 days. Injecting 3–5% bromelain solutions was most effective at tenderizing the meat, reducing shear force by up to 56% after 8 h. This injection also maximized the water-holding capacity (78–81%) and minimized cooking losses (21–26%), compared to 38% for the control meat sample without treatment. Syringing with 3% bromelain yielded the highest sensory scores across the tenderness, flavor, and overall palatability parameters. The combination of L. acidophilus, Lc. lactis, and B. longum at a ratio of 1.5:1.5:2 was highly effective in reducing oxidative spoilage and optimizing pH levels, thereby ensuring extended meat storability. This study demonstrates that bromelain treatment is an effective method for improving the tenderness, WHC, and sensory properties of horse meat. The LAB combination showed efficient acid formation, crucial for enhancing meat preservation.

Abstract:In 1994, NIOSH issued a report to increase the recognition of Organic Dust Toxic Syndrome (ODTS), a flu-like illness following respiratory exposure to organic dusts (1). It was noted that ODTS was a poorly recognized problem possibly affecting up to 40% of heavily exposed US workers (2). A review of the medical literature failed to locate any papers regarding poison center recognition of this syndrome. We undertook a study to determine whether poison control centers are properly diagnosing ODTS and other, related pulmonary exposures.

Abstract:The aim of this study was to investigate the effects of bromelain and papain enzymes for tenderizing of squid (Loligo vulgaris) muscle. Squid samples were immersed into the enzyme solutions and then they were kept in a water bath at 60 °C for 20 min to activate enzymes. After enzyme treatment, pH values, hardness and shear force values of squid muscle decreased. Total protein, free amino acid contents and sensory scores increased. Use of bromelain and papain enzymes was found as effective method to tenderize of squid muscle. Better results were obtained with papain compared to bromelain.

Abstract:Alxa Bactrian camel meat is an organic diet that provides balanced nutrition and is easy to digest and absorb. Despite its potential, it is currently underutilized. To develop a new type of camel jerky, this study utilized a single-factor design method to optimize the formula and fermentation process parameters of Alxa Bactrian camel jerky. Additionally, the physical and chemical composition, nutritional components, protein degradation, and microbial changes of the camel jerky were analyzed to evaluate the nutritional benefits of the new camel jerky created using modern fermentation technology. The results show that the optimal addition amounts of camel fermented jerky ingredients are 2.2% black pepper, 3% salt, 1.8% white sugar, and 9% cooking wine, whereas the optimal fermentation conditions by compound starter culture are 0.07% starter, 23 h of fermentation time, and a 30°C fermentation temperature. Compared to the single starter, the compound starter significantly increased the protein content, total amino acids, unsaturated fatty acids, trace elements, and a*, L*, and e-values of jerky; however, it also decreased the water activity (a w), thiobarbituric acid, and pH values of jerky during storage. Compound starter strains can compensate for the shortage of single starters and improve the overall quality of meat products. These findings could provide new insights and serve as a reference for the development of camel meat products and hold significant importance for the growth of the camel industry.
Effects of Muscle Type and Aging on Glycolysis and Physicochemical Quality Properties of Bactrian camel (Camelus bactrianus) Meat

Abstract:Poor tenderness of camel meat has seriously hampered the development of the camel meat industry. This study investigated the effects of muscle fiber composition and ageing time on meat quality, glycolytic potential, and glycolysis-related enzyme activities. Muscle samples of the longissimus thoracis (LT), psoas major (PM), and semitendinosus (ST) were collected from eight 8-10 year old Sonid Bactrian camels (females). Muscle fiber composition was examined by ATPase staining and immunohistochemistry. Meat quality indexes, glycolytic potential, and activities of major glycolytic enzymes were examined at 4 °C aging for 1, 6, 24, 72, and 120 h. The results showed that LT was mainly composed of type IIb muscle fibers, whereas PM and ST were mainly composed of type I muscle fibers. The PCR results of the myosin heavy chain (MyHC) were consistent with the ATPase staining results. During aging, the shear force of LT muscle was always greater than that of PM and ST, and its glycolysis was the strongest; type IIa, IIb, and IIx muscle fibers were positively correlated with muscle shear force and glycolysis rate, and type I muscle fibers were significantly and negatively correlated with the activities of the key enzymes of glycolysis within 6 h. The results showed that the muscle fibers of LT muscle had the greatest glycolysis capacity. These results suggest that an excessive type IIb muscle fiber number percentage and area in camel meat accelerated the glycolysis process, but seriously affected the sensory profile of the camel meat. The results of this study provide directions for the camel industry when addressing the poor tenderness of camel meat.
Enhanced Oligopeptide and Free Tryptophan Release from Chickpea and Lentil Proteins: A Comparative Study of Enzymatic Modification with Bromelain, Ficin, and Papain

Abstract:Plant-based foods offer a sustainable alternative to meet the growing protein demand. Legumes are the most promising of these, as they contain relatively high concentrations of protein, low digestible starch, and dietary fiber, as well as them possibly featuring low levels of fat. Enzymatically modified legume proteins provide us with tempting perspectives in terms of enhancing foods' biological values. However, their bioavailability and digestibility are generally less sufficient than that of proteins of animal origin, which may be improved by well-tailored enzyme modification. In this study, the efficacy of three plant-based proteases (bromelain, ficin, and papain) were evaluated at two distinct concentrations (2.5% and 10%) and three hydrolysis durations (1, 2, and 12 h) when transforming chickpea and lentil proteins. The degree of hydrolysis (DH), peptide profiles, and free amino acid content were analyzed to determine the efficiency of each enzyme. Results showed significant variations in DH, which was influenced by enzyme type, concentration, and hydrolysis duration. Papain exhibited the highest DH, particularly at a 10% concentration, reaching 27.8% efficiency in chickpea and 34.8% in lentils after 12 h. Bromelain and ficin were proven to be less effective, with ficin showing the least hydrolytic activity. SDS-PAGE analysis revealed substantial protein degradation, especially subsequent to papain treatment, pointing out that most proteins were cleaved into smaller peptides. SEC-HPLC indicated a predominant release of peptides within the 200–1000 Da range, suggesting enhanced bioavailability. Papain and bromelain treatments resulted in a significant release of oligopeptides and dipeptides. UHPLC analysis highlighted a marked post-hydrolysis increase in total free amino acids, with arginine, leucine, and lysine being the most abundant ones. Notably, tryptophan, being undetectable in untreated samples, was released in measurable amounts post-hydrolysis. These findings demonstrate papain's superior performance in protein hydrolysis and its potential in producing bioactive peptides, highlighting its applicability in food processing and the development of both nutraceuticals and functional foods.

Culinary Recipes Development from Camel Meat Treated with the Enzyme Preparation Protosubtilin

Abstract:Most recently, non-traditional sources of animal protein, including camel meat, are of increasing interest. Due to the coarse-grained fibrous structure of the camel muscle tissue, ready-made dishes from this meat raw material have a hard consistency. In this regard, the study aim is to develop a rational regimen for softening Kalmyk Bactrian camel breed meat using the enzyme preparation protosubtilin G20x, to develop culinary dishes from meat raw materials pre-treated with this preparation, and also to compare with the study results of the protosubtilin G3x impact on softening meat raw materials. In terms of organoleptic quality indicators, camel meat complies with the GOST 7269-2015 and has the following content of the main nutrients: proteins – 21.6 %; fats – 7.2 %; minerals – 0.97 %. The moisture mass fraction is 76.7 %; energy value accounts for 151 kcal/100 g; moisture-binding capacity ranges from 73.3 to 80.7 %. By the microstructure, the camel muscle tissue is the same as in most butcher beast species, but the muscle fibers diameter ranges from 10 to 250 microns, indicating a coarse-grained fibrous structure. Moreover, there is a large amount of connective tissue in different cuts. Therefore, meat raw materials will remain tough after standard heat treatment. According to the listed indicators, a man can identify camel meat as a dietary full-fledged meat raw material. Still due to the meat hardness, pre-treatment is necessary to soften it. The optimal processing mode of meat raw materials treatment with an enzyme preparation protosubtilin G20x is the following: the aqueous solution concentration – 0.05 %; medium temperature – 40 °С; duration – 1 hour. The finished culinary dish from pre-processed raw materials received the highest score – 5 points. A comparative analysis of the protosubtilin G3x and G20x impact on the meat raw materials structure demonstrated that the preparation G20x was optimal for processing Kalmyk Bactrian camel breed meat, since at a lower concentration it enabled to increase the moisture-binding capacity of raw materials and finished dish yield.
Combined treatment of lactic acid-ultrasound-papain on yak meat and its tenderization mechanism

Abstract:The combined effects of lactic acid-ultrasound-papain on yak meat tenderization were investigated, and its tenderization mechanism elucidated. The optimal condition for combined tenderization was obtained with 0.03% lactic acid, ultrasound for 30 mins, and 200 U/g of papain. When compared with the untreated yak meat, the cutting force and the cooking loss of the tenderized yak meat were decreased by 62.16% and 31.25%, respectively, while the myofibrillar fragmentation index increasing 4.3 times. After the combined tenderization, it was observed a loose arrangement of muscle with larger gaps and cavities. The random-coil content of myofibrillar protein increased to 48.3%, while α-helix decreased to 14.4%, indicating that the myofibrillar protein underwent some unfolding and stretching. These results demonstrated that this combined treatment improved the tenderization of yak meat by disrupting muscle integrity, increasing its water-holding capacity, denaturing myofibrillar protein, and changing its spatial structure. The combined process using lactic acid-ultrasound-papain showed some potential to improve meat texture.

Abstract:This study investigates vacuum impregnation technology, a non-thermal approach to treat local beef cubes (2 cm × 2 cm × 2 cm) with three distinct marinades: distilled water (CONTROL), 10% (v/v) pineapple juice (PINE), and 10% (v/v) papaya juice (PAPAYA), both containing plant proteases (bromelain and papain). The primary objective was to evaluate vacuum impregnation (VI) and traditional marination (TM) techniques and assess their impact on beef’s physicochemical attributes, shelf-life stability, and its plant protease activity. Shelf-life tests over 5 days revealed that all beef samples’ pH reached its lowest on day 4, subsequently increasing on day 5. Throughout storage, colour parameters (L*, a*, b*, and ∆E) of treated samples consistently decreased. VI-PINE exhibited the lowest water holding capacity (WHC) over 5 days. Texture profile analysis (TPA) indicated that VI samples' hardness significantly reduced in VI-PINE and VI-PAPAYA compared to TM samples, attributed to coupled plant proteases. VI-PINE showed the highest marinade uptake improvement (21.6%); while having the lowest WHC (15.52%), and all samples' pH ranged in 5.15–6.12. The colour parameters of treated samples increased compared to raw untreated samples. Compared to TM, VI-PINE (1115 g) and VI-PAPAYA (1333 g) significantly reduced beef hardness, with VI-marinated beef also exhibiting higher tyrosine release compared to TM techniques. The application of VI on beef cubes with plant protease-containing marinades provides practical insights for the food industry, offering potential avenues to enhance beef quality, improve shelf life, and optimize flavour incorporation.

Abstract:Spiral is an easily expandable, high fault-tolerant self-routing computer network topology. The architecture is built around modules of four fully connected computer nodes each, connected by top and bottom spirals. Six theorems (without proof) and analytical results based on those theorems are reported. The theorems concern disjoint paths, maximum path length, traffic between source-destination pairs, expected average link traffic, total one-way link traffic, and mean path length.<<ETX>>
Spray-drying and ultrasonication processing of camel whey protein concentrate: Characterization and impact on bioactive properties

Abstract:The production of whey protein concentrates (WPC) from camel milk whey represents an effective approach to valorize this processing byproduct. These concentrates harbor active ingredients with significant bioactive properties. Camel WPC were spray-dried at inlet temperature of 170, 185 and 200°C, or ultrasonicated (US) for 5, 10, and 15 min, then freeze-dried to obtain fine powder. The effect of both treatments on protein degradation was studied by sodium dodecyl sulfate-PAGE and reverse-phase ultraperformance liquid chromatography techniques. Significantly Substantially enhanced protein degradation was observed after US treatment when compared with spray-drying (SPD). Both SPD and US treatments slightly enhanced the WPC samples' antioxidant activities. The US exposure for 15 min exhibited the highest 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) scavenging activity (12.12 mmol Trolox equivalent per gram). Moreover, US treatment for 10 min exhibited the highest in vitro antidiabetic properties (α-amylase and α-glucosidase inhibition), and dipeptidyl peptidase-IV inhibitory activity among all samples. In addition, the US for 10 min and SPD at 170°C showed the lowest median inhibitory concentration (IC50) values for in vitro antihypercholesterolemic activities in terms of pancreatic lipase and cholesteryl esterase inhibition. Conclusively, these green techniques can be adapted in the preservation and processing of camel milk whey into active ingredients with high bioactive properties.
