Abstract:Cyanobacteria obtain their energy through photosynthesis and live embedded in a matrix of extracellular polymeric substances (EPS) containing valuable products, e.g., polysaccharides, lipids, proteins, and antimicrobials. Besides chlorophyll a and carotenoids, they have light-absorbing compounds in the form of light-harvesting complexes, the so-called phycobilisomes, consisting of different phycobiliproteins. Together they close the "green gap" whereby cyanobacteria can use light more effective than higher plants. Cultivation of cyanobacteria on a lab-scale results in small amounts of biomass for their characterization or a comprehensive screening. EPS are, for example, produced as a protection against suboptimal culture conditions. Carotenoids are essential light-harvesting pigments for photosynthesis, play a key role in photoprotective reactions, and are produced for cell wall stabilization. Essentially, the pigment composition of cyanobacteria depends on the available light spectrum, nitrogen content, and temperature. Especially the production of EPS and pigments are indicators for the cell-condition. Therefore, different EPS extraction methods were tested including the determination of inhibitory effects of extracts against Escherichia coli. Based on the best EPS extraction method, a new strategy for downstream processing (DSP) was developed to determine EPS, the pigments chlorophyll a and carotenoids, and phycobiliproteins from only one sample. As cyanobacterial model organisms Trichocoleus sociatus and Nostoc flagelliforme were used, and DSP strategy was successfully transferred to four additional cyanobacteria. The final DSP includes the following steps: (i) EPS extraction, (ii) lyophilization of biomass, (iii) extraction of phycobiliproteins, and a final (iv) chlorophyll a and carotenoid extraction. The new strategy allows a comprehensive characterization of cyanobacterial cells.

Abstract:Microalgae and cyanobacteria are photosynthetic organisms that can produce/accumulate biomolecules with industrial interest. Among these molecules, EPSs are macromolecular polysaccharidic compounds that present biological activities and physico-chemical properties, allowing to consider their valorization in diverse commercial markets, such as cosmetic, therapeutic, nutraceutic, or hydrocolloids areas. The number of microalgae and cyanobacteria strains described to produce such EPSs has increased in recent years as, among the 256 producing strains gathered in this review, 86 were published in the last 10 years (~33%). Moreover, with the rise of research on microalgae EPSs, a variety of monosaccharides compositions have been discovered, highlighting the versatility of these organisms. If some production strategies can be applied to increase EPS production yields, it appears that case by case studies are needed to promote EPS synthesis by a strain, as many responses exist. This paper proposes an up-to-date state of the art of the diversity of microalgae and cyanobacteria EPS-producing strains, associated to the variability of compositions. The strategies for the production and extraction of the polymers are also discussed. Finally, an overview of the biological activities and physico-chemical properties allow one to consider their use on several commercial markets.

Abstract:Exopolysaccharides (EPS) are high molecular weight biomaterials of industrial interest due to their variety of applications in the pharmaceutical, cosmetic, environmental, and food industries. EPS produced by Rhodotorula mucilaginosa UANL-001 L has sparked interest due to its bio-adsorbent and wide spectrum antimicrobial properties. However, full exploitation and commercial application of EPS has been restrained due to low yields and high production costs. In the present work, the production and separation of EPS from Rhodotorula mucilaginosa UANL-001L was attempted through extractive fermentation in order to increase EPS production while simplifying the recovery process. Extractive fermentation was implemented with a thermoseparating polymer for phase formation (EOPO 970 and EOPO 12,000); culture viability, biomass generation, EPS production, rheological system properties, and phase formation time and temperature were monitored throughout the process. Extractive fermentation of Rhodotorula mucilaginosa UANL-001L with EOPO 970 resulted in a 42% EPS and 7% biomass recovery on the top phase after 5 to 13-min phase formation time and temperatures between 30 and 40 °C. This is the first report of extractive fermentation application for EPS production by yeast of the genera Rhodotorula, resulting in an interesting strategy for EPS production and recovery, although further optimization is needed.

Abstract:State- and flow-driven process modelling paradigms are both widely practised in the business process modelling domain. From the perspective of both from their syntax structure and intentioned application scopes, they are quite different. But one of our findings is that any one of them usually can be replaced by the other in a variety of real cases. It is shown with theoretical approach in this work that these two paradigms are equal in expressiveness power in that they can be mutually transformed into each other without any semantic losses. The motivation of this work is to suggest that researchers argue less about which method is more powerful and pay more attention to the exploration of better ways to combine different style models in addressing real problems. In this paper, (bi) imulation of Pi calculus is applied to prove the correctness of the bitransformation of two paradigms. A hybrid business model composed of a state-driven ATM machine and flow-driven withdraw processes is also illustrated with a concrete banking scenario to show how the result of bitransformation can be applied to facilitate the integration, understanding and verification of hybrid business models into real business cases.
Sustainable Exopolysaccharide Production by Rhizobium viscosum CECT908 Using Corn Steep Liquor and Sugarcane Molasses as Sole Substrates

Abstract:Microbial exopolysaccharides (EPS) are promising alternatives to synthetic polymers in a variety of applications. Their high production costs, however, limit their use despite their outstanding properties. The use of low-cost substrates such as agro-industrial wastes in their production, can help to boost their market competitiveness. In this work, an alternative low-cost culture medium (CSLM) was developed for EPS production by Rhizobium viscosum CECT908, containing sugarcane molasses (60 g/L) and corn steep liquor (10 mL/L) as sole ingredients. This medium allowed the production of 6.1 ± 0.2 g EPS/L, twice the amount produced in the standard medium (Syn), whose main ingredients were glucose and yeast extract. This is the first report of EPS production by R. viscosum using agro-industrial residues as sole substrates. EPSCSLM and EPSSyn exhibited a similar carbohydrate composition, mainly 4-linked galactose, glucose and mannuronic acid. Although both EPS showed a good fit to the Herschel–Bulkley model, EPSCSLM displayed a higher yield stress and flow consistency index when compared with EPSSyn, due to its higher apparent viscosity. EPSCSLM demonstrated its potential use in Microbial Enhanced Oil Recovery by enabling the recovery of nearly 50% of the trapped oil in sand-pack column experiments using a heavy crude oil.

Abstract:Unusual composition of an exopolymer (EP) from an obligate halophilic bacterium Chromohalobacter canadensis 28 has triggered an interest in development of an effective bioreactor process for its production. Its synthesis was investigated in 2-L bioreactor at agitation speeds at interval 600-1000 rpm, at a constant air flow rate of 0.5 vvm; aeration rates of 0.5, 1.0, and 1.5 vvm were tested at constant agitation rate of 900 rpm. EP production was affected by both, agitation and aeration. As a result twofold increase of EP yield was observed and additionally increased up to 3.08 mg/mL in a presence of surfactants. For effective scale-up of bioreactors mass transfer parameters were estimated and lowest values of KLa obtained for the highest productivity fermentation was established. Emulsification activity of EP exceeded that of trade hydrocolloids xanthan, guar gum, and cellulose. A good synergism between EP and commercial cellulose proved its potential exploration as an enhancer of emulsifying properties of trade emulsions. A pronounced lipophilic effect of EP was established toward olive oil and liquid paraffin. Cultivation of human keratinocyte cells (HaCaT) with crude EP and purified γ-polyglutamic acid (PGA) showed higher viability than control group.

Abstract:Virgibacillus salarius BM02 was identified as a highly exopolysaccharide (EPS) producing bacterium. The EPS production and its physico-chemical properties (intrinsic viscosity and total sugars/protein (TS/P) ratio) were optimized using Box-Behnken experimental design. Maximum EPS production of 5.87 g L-1 with TS/P ratio of 12.56 and intrinsic viscosity of 0.13 dL g-1 was obtained at optimal conditions of sucrose (4.0% w/v), peptone (0.75% w/v) and incubation period of 4.69 day. The monosaccharide composition of EPS was mannose, arabinose and glucose at a molar ratio of 1.0:0.26:0.08. The EPS showed high water solubility (38.5%), water holding capacity (514.46%) and foaming capacity (55.55%). The EPS showed moderate antioxidant activity in vitro and good emulsion stabilizing properties against several hydrophobic compounds. The emulsifying activity was stable at different temperatures, pH and ionic strength. Additionally, the acid hydrolysate of the EPS was evaluated as a carbon source for the mixotrophic cultivation of industrially important Spirulina platensis. It induced an enhancement of not only biomass production of S. platensis, but also cellular contents (pigments, proteins and lipids) leading to higher nutritional value.

Abstract:Sarcopeltis skottsbergii is an endemic species of the southern region of South America, with R-phycoerythrin (R-PE) as an accessory photosynthetic pigment. The production of S. skottsbergii is around twenty thousand tons of dry alga per year. The evaluation of (R-PE) in a biorefinery model is still incipient in the algal biotechnology area and will be used in the food, pharmaceutical, cosmeceutical, and nutraceutical industries. This work evaluated the cell disruption and separation processes by using two green technologies, ultrasound-assisted extraction (UAE) and high-pressure homogenization (HPH), to obtain an R-phycoerythrin enriched extract from S. skottsbergii . Two-levels three-factor central composite design (CCD) and response surface methodology (RSM) were carried out to optimize the extraction conditions, including the factors for UAE (time, amplitude, and solvent) and HPH (Pressure, number of passes, and solvent). Additionally, a second-order polynomial fit was performed to fit the experimental data by the green method. HPH method was the most efficient extraction method under the conditions obtained of 100-400 MPa pressure power, 2-3 number of passes, and distilled water as solvent. Furthermore, the experimental extraction yields ranged from 4.4-5.7 mg of PE g -1 of dry biomass under the optimal extraction conditions (400 MPa; 2 passes), which agreed with the predictive yield of 4.6-5.5 mg g -1 DW. The ultrafiltration membrane used for the separation process for both methods exhibited a rejection of R-phycoerythrin concentrated at 30 KDa. Furthermore, R-phycoerythrin showed a positive correlation between the antioxidant capacity (ORAC) in the best-selected extractions. After the extraction, the same pattern was observed in Chlorophyll a and total carotenoids with DPPH. Thus, it was an attractive non-aggressive extraction alternative with biological activity of interest for formulating biotechnological products for the food industry is suggested.

Abstract:The autotrophic cyanobacterium Phormidium sp. is filamentous, blue-green, sheath-prominent, and trichome-constricted. DNA barcoding of the 16S rRNA sequence was submitted to NCBI-GenBank under the accession number MW436689, with a similarity of 97–99% with varied sequences. The Phormidium sp. barcode region had a high G + C content (52.7%). An investigation is underway to extract and characterize exocellular polymeric substances from Phormidium sp. (EPS- P ) for efficacy analysis. FTIR, GC-MS, and NMR were used to characterize (EPS- P ). FTIR suggests the presence of hydroxy groups (O-H moieties) and amine salts (N-H). GCMS shows D-galactose, L-rhamnose, D-mannose, uronic acid, and D-glucuronic acid. The specific chemical shifts could be identified in the 1 H NMR spectra. EPS has been used in a variety of industrial and environmental applications. The ability to emulsify EPS- P (0.1%) was analyzed with various oils. The emulsifying index (EI) was observed to be gingelly oil > brake oil > neem oil > coconut oil > petrol oil > diesel oil (EI 24 -EI 120 hours). EPS- P performed bio-flocculating activity at various concentrations (10-50 mg mL 1 ). The flocculating activity of EPS- P shows 89.5% at 50 mg ml −1 . The EPS- P could be an excellent candidate for bio-flocculation and bio-emulsification aspects that are also eco-friendly.

Abstract:Understanding the impact of variables involved in soluble-extracellular polymeric substance (S-EPS) production processes is crucial for reducing production costs and enhancing sustainability. Response surface methodology (RSM) provides essential tools that assist in developing predicted interactions among process variables for both industrial and non-industrial applications. The present study offers a simple and systematic demonstration of RSM capabilities, focusing on maximizing efficiency and minimizing production costs of S-EPS produced by Cyanocohniella rudolphia. RSM was employed to (1) design the production setup; (2) fit the collected data into a second-order polynomial model; (3) statistically evaluate the model's validity and the significance of the involved variables; and (4) identify and optimize production variables to enhance output and reduce costs. Focused on four key variables, each at three levels, RSM designed 25 distinct S-EPS production conditions, each with three replicates. Statistical analysis identified the most significant variables affecting S-EPS production as the culture medium/wet biomass ratio, production days, and nitrogen concentration. The model's validation demonstrated a strong correlation between the predicted and experimental values, with S-EPS production ranging from 70.46 to 228.65 mg/L and a maximum variation of 11.6%. This study demonstrates the effectiveness of RSM in optimizing S-EPS production, with the developed model showing a strong correlation between the variables and the response. The RSM model offers a promising approach for the bioprocessing industry, enhancing productivity and efficiency, minimizing costs, and leading to sustainable, cost-effective practices.

Abstract:The freshwater green microalgae, Dictyosphaerium chlorelloides (CCALA 330), has the ability to produce extracellular polysaccharides (EPS). Conditions for optimum growth and EPS overproduction were determined in laboratory-scale tubular photobioreactors (PBR) with a working volume of 300 mL. Multiple limitations in nutrient supply were proven to be an effective method for EPS overproduction. Salinity stress was also applied to the culture, but no significant increase in EPS production was observed. The effects of different nitrogen sources were examined and the microalgae exhibited the fastest growth and EPS production in medium containing ammonium nitrate. Under determined optimal conditions, EPS concentration reached 10 g/L (71% of the total biomass) and a total biomass of 14 g/L at the end of 17 days cultivation. Pilot-scale cultivation was also carried out in a column type airlift photobioreactor (PBR) with a working volume of 60 L. A new and efficient methodology was developed for separating cells from the EPS-containing culture broth. Due to the strong attachment between cells and EPS, high-pressure homogenization was carried out before a centrifugation process. The EPS in the supernatant was subsequently purified using ultrafiltration. The green microalgae Dictyosphaerium chlorelloides may therefore be appropriate for the commercial production of EPS.

Abstract:Cyanobacteria can produce extracellular polysaccharides (EPS) and release them to the environment, which is one of the adaptive mechanisms of cyanobacteria evolved to face complicated and variable environment. As a protective barrier between cell and environment, the EPS play a protective role against desiccation, ultraviolet radiation, bio-mineralization, or predation by protozoans. The EPS produced by cyanobacteria are a kind of complex anionic heteropolymer polysaccharides, more than 75% of which are composed of six or more monosaccharaides. Glucose is the most frequent kind of monosaccharaides. The characteristics of the EPS vary with cyanobacterial species. Most of the EPS show an anionic feature due to the charged groups, such as uronic acid and sulfate groups. Sulfate group is the basis of the anti-viral properties of polysaccharides, while hydrophobic groups such as ester-linked acetyl groups, peptides moieties, and deoxy sugars contribute to emulsifying properties. Therefore, the EPS produced by cyanobacteria have a broad application prospect in many industries, such as food additives, cosmetology, pharmacology, and wastewater treatment. The industrial application of the EPS can not only reuse the cyanobacteria bloom, bringing considerable economic benefits, but also provide a solution to the salvaging cyanobacteria distribution problem, avoiding its secondary pollution. Until now, there is no commodity from cyanobacterial EPS, and many technical problems are needed to be solved between the theoretical study and large batch industrial production.

Abstract:Exopolysaccharides (EPS) are produced by various microbial groups including bacteria. EPS produced from Lactobacilli is gaining popularity as a new generation functional food to replace various chemically synthesized food additives. EPS production by strains of L. reuteri were reported previously in sourdough fermentation. The submerged cultivation of L. reuteri strain DSM 20016 T was optimised for crude EPS production. Initially, the factors controlling crude EPS generation were explored using one-at-a-time (OFAT). Among the investigated carbon sources, sucrose supplemented medium increased maximal crude EPS generation. Yeast extract was determined to be the greatest nitrogen source for increasing crude EPS and cell mass. Plackett-Burman experimental design (PBD) was employed to screen the important parameters impacting crude EPS production in the growing medium. Three important factors were identified: sucrose, yeast extract and sodium acetate. The RSM quadratic model for crude EPS generation was significant (R2:0.9373) and correlated with the observed response at roughly 94%. About 3.4 g L−1 crude EPS was produced under optimal fermentation conditions of sucrose (106.0 g L−1), yeast extract (30.0 g L−1) and sodium acetate (7.0 g L−1). This study shows the crude EPS synthesis of L. reuteri from sourdough can be used in submerged fermentation and biotechnological applications.

Abstract:Polyhydroxyalkanoates (PHAs) are bioplastics produced by microorganisms under nutrient limiting conditions and in the presence of excess carbon source. PHAs have gained popularity as a sustainable alternative to traditional plastics. However, large scale production of PHAs is economically challenging due to the relatively high costs of organic carbon. Alternative options include using organisms capable of phototrophic or mixotrophic growth. This study aimed at the production of poly(3-hydroxybutyrate) P(3HB), a type of PHA, at pilot scale using the freshwater cyanobacterium Synechocystis sp. PCC6803. First, to identify optimal conditions for P(3HB) production from Synechocystis sp. PCC6803, different supplemental carbon source concentrations and salinity levels were tested at laboratory scale. The addition of 4 g/L acetate with no added NaCl led to P(3HB) accumulation of 10.7% dry cell weight on the 28th day of cultivation. Although acetate additions were replicated in an outdoor 400 L serpentine photobioreactor, P(3HB) content was lower, implying uncontrolled conditions impact on biopolymer production efficiency. An optimized P(3HB) extraction methodology was developed to remove pigments, and the biopolymer was characterized and subjected to 3D printing (fused deposition modelling) to confirm its processability. This study thus successfully led to the large-scale production of P(3HB) using sustainable and environmentally friendly cyanobacterial fermentation.

Abstract:Microbial exopolysaccharides (EPS) are long-chain polysaccharides that are synthesized and secreted by microorganisms into the external matrix. In bacteria, EPS can either be associated with the cell surface in the form of capsules or be secreted into the environment. The yield of EPS is influenced by various factors, including the growth conditions and composition of the growth media. EPSs are high molecular-weight carbohydrates that consist of a backbone of repeated subunits of monosaccharides in varying ratios. Recently, there has been an increasing interest in EPS-producing microbes with Generally Recognized as Safe (GRAS) status. These food-grade organisms have the potential to produce polymers that can control the rheological and functional properties of food systems. EPSs have also been reported to have prebiotic and immune-modulating functions like anticancer, antidiabetic, antiviral, etc. The increasing recognition of the association between EPS and health benefits suggests the potential of EPS-producing starters with functional characteristics in the production of value-added functional products. Such products align with consumer demand for natural and healthy alternatives with fewer additives. The exploration of functional means of EPS in Pharmacology will provide an opportunity to identify novel and robust microbial resources producing unique EPSs.
Production of exopolysaccharide from Klebsiella oxytoca: Rheological, emulsifying, biotechnological properties, and bioremediation applications

Abstract:Bacteria can synthesize a broad spectrum of multifunctional polysaccharides including extracellular polysaccharides (EPS). Bacterial EPS can be utilized in the food, pharmaceutical, and biomedical areas owing to their physical and rheological properties in addition to generally presenting low toxicity. From an ecological viewpoint, EPS are biodegradable and environment compatible, offering several advantages over synthetic compounds. This study investigated the EPS produced by Klebsiella oxytoca (KO-EPS) by chemically characterizing and evaluating its properties. The monosaccharide components of the KO-EPS were determined by HPLC coupled with a refractive index detector and GC-MS. The KO-EPS was then analyzed by methylation analysis, FT-IR and NMR spectroscopy to give a potential primary structure. KO-EPS demonstrated the ability to stabilize hydrophilic emulsions with various hydrophobic compounds, including hydrocarbons and vegetable and mineral oils. In terms of iron chelation capacity, the KO-EPS could sequester 41.9 % and 34.1 % of the most common iron states, Fe2+ and Fe3+, respectively. Moreover, KO-EPS exhibited an improvement in the viscosity of aqueous dispersion, being proportional to the increase in its concentration and presenting a non-Newtonian pseudoplastic flow behavior. KO-EPS also did not present a cytotoxic effect indicating that the KO-EPS could have potential applications as a natural thickener, bioemulsifier, and bioremediation agent.
Characterization of soluble and bound EPS obtained from 2 submerged membrane bioreactors by 3D-EEM and HPSEC

Abstract:This research study deals with the quantification and characterization of the EPS obtained from two 25 L bench scale membrane bioreactors (MBRs) with micro-(MF-MBR) and ultrafiltration (UF-MBR) submerged membranes. Both reactors were fed with synthetic water and operated for 168 days without sludge extraction, increasing their mixed liquor suspended solid (MLSS) concentration during the experimentation time. The characterization of soluble EPS (EPSs) was achieved by the centrifugation of mixed liquor and bound EPS (EPSb) by extraction using a cationic resin exchange (CER). EPS characterization was carried out by applying the 3-dimensional excitation-emission matrix fluorescence spectroscopy (3D-EEM) and high-performance size exclusion chromatography (HPSEC) with the aim of obtaining structural and functional information thereof. With regard to the 3D-EEM analysis, fluorescence spectra of EPSb and EPSs showed 2 peaks in both MBRs at all the MLSS concentrations studied. The peaks obtained for EPSb were associated to soluble microbial by-product-like (predominantly protein-derived compounds) and to aromatic protein. For EPSs, the peaks were associated with humic and fulvic acids. In both MBRs, the fluorescence intensity (FI) of the peaks increased as MLSS and protein concentrations increased. The FI of the EPSs peaks was much lower than for EPSb. It was verified that the evolution of the FI clearly depends on the concentration of protein and humic acids for EPSb and EPSs, respectively. Chromatographic analysis showed that the intensity of the EPSb peak increased while the concentrations of MLSS did. Additionally, the mean MW calculated was always higher the higher the MLSS concentrations in the reactors. MW was higher for the MF-MBR than for the UF-MBR for the same MLSS concentrations demonstrating that the filtration carried out with a UF membrane lead to retentions of lower MW particles.
Exopolysaccharide from Leuconostoc mesenteroides XR1: Yield optimization, partial characterization and properties

Abstract:The production conditions of exopolysaccharide (EPS) from Leuconostoc mesenteroides XR1 were optimized by response surface methodology (RSM). Maximum EPS yield was 56.59 ± 0.51 g/L under fermentation conditions with 2.6 g/L ammonium citrate, initial pH 6.5 and temperature 23 °C, which was 6.21-fold greater than the EPS yield before optimization. Characterization of the chain conformation using Congo red test and circular dichroism (CD) showed that EPS exhibited a random coil structure in aqueous solution. The CD results revealed that the EPS concentration altered its hydrogen-bond interactions and chirality, but did not change its chain conformation. The average polydispersity index (PDI) of the EPS solution was only 27.16 %, indicating that it was uniformly distributed in the aqueous solution with high stability. The degradation temperature of EPS was 253.11 °C, indicating high thermal stability. EPS possessed the ability to scavenge activities of free radicals and was protective against oxidative stress-induced plasmid DNA damage. In addition, stable hydrogels could be formed at EPS concentrations above 5 % (w/v). These results collectively showed that EPS can be used commercially as an antioxidant and drug delivery carrier.

Abstract:Given the necessity for bioprocesses scaling-up, the present study aims to explore the potential of three marine cyanobacteria and a consortium, cultivated in semi-continuous mode, as a green approach for i ) continuous exopolysaccharide-rich biomass production and ii ) removal of positively charged metals (Cu, Ni, Zn) from mono and multi-metallic solutions. To ensure the effectiveness of both cellular and released exopolysaccharides, weekly harvested whole cultures were confined in dialysis tubings. The results revealed that all the tested cyanobacteria have a stronger affinity towards Cu in mono and three-metal systems. Despite the amount of metals removed per gram of biomass decreased with higher biosorbent dosage, the more soluble carbohydrates were produced, the greater was the metal uptake, underscoring the pivotal role of released exopolysaccharides in metal biosorption. According to this, Dactylococcopsis salina 16Som2 showed the highest carbohydrate productivity (142 mg L -1 d -1 ) and metal uptake (84 mg Cu g -1 biomass) representing a promising candidate for further studies. The semi-continuous cultivation of marine cyanobacteria here reported assures a schedulable production of exopolysaccharide-rich biosorbents with high metal removal and recovery potential, even from multi-metallic solutions, as a step forward in the industrial application of cyanobacteria.
