Abstract:Developmental scientists have examined the independent effects of peer presence, social cues, and rewards on adolescent decision-making and cognitive control. Yet, these contextual factors often co-occur in real world social situations. The current study examined the combined effects of all three factors on cognitive control, and its underlying neural circuitry, using a task to better capture adolescents' real world social interactions. A sample of 176 participants ages 13-25, was scanned while performing an adapted go/no-go task alone or in the presence of a virtual peer. The task included brief positive social cues and sustained periods of positive arousal. Adolescents showed diminished cognitive control to positive social cues when anticipating a reward in the presence of peers relative to when alone, a pattern not observed in older participants. This behavioral pattern was paralleled by enhanced orbitofrontal activation. The results demonstrate the synergistic impact of social and reward influences on cognitive control in adolescents.

Joint Effects of Peer Presence and Fatigue on Risk and Reward Processing in Late Adolescence

Abstract:Peers are thought to increase adolescents’ risk-taking behavior, at least in part, by heightening their sensitivity to rewards. In this study, we investigate whether the effect of peers on late adolescent males is exacerbated when youth are cognitively fatigued, a state characterized by weakened cognitive control and heightened orientation toward rewards, and well established as a factor that compromises decision making. We hypothesized that fatigued adolescents’ top-down regulation of reward-related impulses may be compromised, thereby potentially amplifying the effect of peers on reward- and risk-seeking behavior. Late adolescent males between 18 and 22 years old (mean age = 19.64, SD = 1.22; 61% Caucasian) completed a decision-making battery either alone or in the presence of 3 same-sex peers, and were either cognitively fatigued or non-fatigued. We compared behavior between four experimental groups—fatigued adolescents in a peer group, non-fatigued adolescents in a peer group, fatigued adolescents by themselves, and non-fatigued adolescents by themselves. The findings showed that cognitive fatigue and peer presence evinced independent effects on risk taking and sensitivity to rewards, but that these factors do not influence adolescent decision-making in an additive or synergistic fashion. To our surprise, being fatigued reduces (but does not eliminate) the effect of peers of risk taking. Moreover, the impact of peers on adolescent males’ ability to learn from negative consequences is not compromised when adolescents are in a state of mental fatigue. Our results suggest that mental fatigue increases late adolescent males’ reward sensitivity to the same extent as peer presence, but does not amplify the peer effect on risk-taking behavior. In this regard, grouping adolescents when they are fatigued may be less dangerous than when they are rested. Similarly, the added presence of peers does not further exacerbate the effect of fatigue on adolescent’s reward- and risk-seeking inclinations. In fact, given peers’ unique effect on adolescents’ ability to learn from costly decisions, our findings suggest that seeking the presence of peers—which is often a rewarding experience in and of itself—may be an adaptive response to mitigate the impact of fatigue on decision making.

Abstract:Functional magnetic resonance imaging (fMRI) has illuminated the development of human brain function. Some of this work in typically-developing youth has ostensibly captured neural underpinnings of adolescent behavior which is characterized by risk-seeking propensity, according to psychometric questionnaires and a wealth of anecdote. Notably, cross-sectional comparisons have revealed age-dependent differences between adolescents and other age groups in regional brain responsiveness to prospective or experienced rewards (usually greater in adolescents) or penalties (usually diminished in adolescents). These differences have been interpreted as reflecting an imbalance between motivational drive and behavioral control mechanisms, especially in mid-adolescence, thus promoting greater risk-taking. While intriguing, we caution here that researchers should be more circumspect in attributing clinically significant adolescent risky behavior to age-group differences in task-elicited fMRI responses from neurotypical subjects. This is because actual mortality and morbidity from behavioral causes (e.g. substance abuse, violence) by mid-adolescence is heavily concentrated in individuals who are not neurotypical, who rather have shown a lifelong history of behavioral disinhibition that frequently meets criteria for a disruptive behavior disorder, such as conduct disorder, oppositional-defiant disorder, or attention-deficit hyperactivity disorder. These young people are at extreme risk of poor psychosocial outcomes, and should be a focus of future neurodevelopmental research.

Abstract:Theories of adolescent neurodevelopment have largely focused on group-level descriptions of neural changes that help explain increases in risk behavior that are stereotypical of the teen years. However, because these models are concerned with describing the “average” individual, they can fail to account for important individual or within-group variability. New methodological developments now offer the possibility of accounting for both group trends and individual differences within the same modeling framework. Here we apply GIMME, a model-based approach which uses both group and individual-level information to construct functional connectivity maps, to investigate risky behavior and neural changes across development. Adolescents (N = 30, M age  = 13.22), young adults (N = 23, M age  = 19.19), and adults (N = 31, M age  = 43.93) completed a risky decision-making task during an fMRI scan, and functional networks were constructed for each individual. We took two subgrouping approaches: 1) a confirmatory approach where we searched for functional connections that distinguished between our a priori age categories, and 2) an exploratory approach where we allowed an unsupervised algorithm to sort individuals freely. Contrary to expectations, we show that age is not the most influence contributing to network configurations. The implications for developmental theories and methodologies are discussed.

Superior Immunomodulatory Effects of Intravenous Immunoglobulins on Human T-cells and Dendritic Cells: Comparison to Calcineurin Inhibitors

Abstract:Background. Prophylactic administration of anti-HBs intravenous immunoglobulins (IVIg) in hepatitis B infected-liver transplant patients protects against acute rejection. To explore the suitability of intravenous immunoglobulins (IVIg) as prophylaxis of acute rejection and graft-versus-host disease (GVHD) after allograft transplantation, the effects of IVIg and calcineurin inhibitors (CNI) on human blood-derived T-cells and DC were compared. Methods. T-cells were stimulated with phytohemagglutinin (PHA) or allogeneic spleen antigen-presenting cells (APC) and T-cell proliferation and cytokine production were determined in presence or absence of IVIg or CNI. Immature blood dendritic cells (DC) were stimulated in presence or absence of IVIg or CNI, and allogeneic T-cell stimulatory capacity, cell death, and phenotypic maturation were established. Results. IVIg and CNI equally inhibited T-cell proliferation and IFN-&ggr; production after PHA stimulation or allogeneic stimulation. CD8+ T-cells were preferentially affected by both IVIg and CNI after allogeneic stimulation. Like CNI, addition of IVIg at later time points after T-cell activation suppressed mitotic progression of responding T-cells. IVIg-treated DC were suppressed in their capacity to stimulate allogeneic T-cell proliferation by 73±12%, whereas DC-function was not affected by CNI. The decreased allogeneic T-cell stimulatory capacity of IVIg-treated DC correlated to induction of cell death in DC and decreased up-regulation of CD40 and CD80. Conclusions. In vitro IVIg functionally inhibit the two principal immune cell-types involved in rejection and GVHD, i.e. T-cells and DC, whereas CNI only suppress T-cells. By targeting both T-cells and DC, IVIg may be a promising candidate for immunosuppressive treatment after allograft transplantation.

Abstract:Adolescent risky behavior is related to developmental changes in decision-making processes and their neural correlates. Yet, research investigating how the family environment relates to risk processing in the adolescent brain is limited. In this study, longitudinal data were collected from 167 adolescents (13-15 years, 53% male) who self-reported household chaos and their parent's monitoring practices, and completed a decision-making task during functional MRI at Time 1 and Time 2 (1 year apart). Parental knowledge was positively related to insular risk processing only among adolescents in low-chaos environments at both time points. Results highlight environmental correlates of insular risk processing in the developing brain.

Abstract:Adolescents' peer world is highly dynamic with constant dissolution of old friendships and formation of new ones. Though many of adolescents' risky decisions involve their peers, little is known about how adolescents' ever-changing friendships shape their ability to make these peer-involving risky decisions, particularly adaptive ones, and whether this association shifts over time. In a 5-wave longitudinal fMRI study, 173 adolescents (at wave 1: M<sub>age</sub> = 12.8, SD<sub>age</sub> = 0.52; range = 11.9-14.5) made risky choices to win money for their best friend. We assessed whether participants nominated the same or different best friend as their previous participation year (a total of 340 data points of friendship maintenance / change). In early adolescence, adolescents with the same best friend took more adaptive risks for that best friend than those with a different best friend. In late adolescence, however, adolescents with a different best friend took more adaptive risks for the new best friend than those with the same best friend. Further, the amygdala was differentially sensitive to friendship maintenance / change during these peer-involving adaptive risks across time. This study has implications for how stable and flexible peer landscapes differentially modulate social motivation and social decision-making over the course of adolescence.

Abstract:Influenced by individual, family, and social factors, risk-taking often occurs in childhood as a multi-determined outcome. And the ability to govern risky decision-making in a balanced manner is a hallmark of the integrity of cognitive and affective development from childhood to adulthood. The Triadic Neural Systems Model posits that nuanced coordination of motivational approach, avoidance and prefrontal control system is crucial to regulate adaptive risk-taking and related behaviors. Although widely studied in adolescence and adulthood, how these systems develop in childhood remains elusive. Here, we show heterogenous age-related differences in the triadic neural systems involved in risky decision-making in 218 school-age children relative to 80 young adults. Children exhibited gradual increases in risk-taking measures from 6 to 12 years-old, which are associated with age-related differences in brain activity patterns underlying reward and risk processing. In comparison to adults, children exhibited weaker activation in control-related prefrontal systems, but stronger activation in reward-related striatal systems. Network analyses revealed that children showed greater reward-related functional connectivity within and between the triadic systems. Our findings support an immature and unbalanced developmental view of the core neurocognitive systems involved in risky decision-making and related behaviors in middle to late childhood.

Abstract:An increased propensity for risk taking is a hallmark of adolescent behavior with significant health and social consequences. Here, we elucidated cortical and subcortical regions associated with risky and risk-averse decisions and outcome evaluation using the Balloon Analog Risk Task in a large sample of adolescents (n = 256, 56% female, age 14 ± 0.6), including the level of risk as a parametric modulator. We also identified sex differences in neural activity. Risky decisions engaged regions that are parts of the salience, dorsal attention, and frontoparietal networks, but only the insula was sensitive to increasing risks in parametric analyses. During risk-averse decisions, the same networks covaried with parametric levels of risk. The dorsal striatum was engaged by both risky and risk-averse decisions, but was not sensitive to escalating risk. Negative-outcome processing showed greater activations than positive-outcome processing. Insula, lateral orbitofrontal cortex, middle, rostral, and superior frontal areas, rostral and caudal anterior cingulate cortex were activated only by negative outcomes, with a subset of regions associated with negative outcomes showing greater activation in females. Taken together, these results suggest that safe decisions are predicted by more accurate neural representation of increasing risk levels, whereas reward-related processes play a relatively minor role.

Peer-mediated Social Signals Alter Risk Tolerance in Teenage Boys Depending on their Peers

Abstract:Introduction: During early adolescence, peer influences play a crucial role in shaping learning and decision preferences. When teens observe what their peers are doing, they can learn and change their behavior, especially when they are taking risks. Our study incorporated an economical behavioral task and computational modeling framework to examine whether and how early male adolescents' risk attitudes change when they see information about their peers' choices. Methods: We recruited 38 middle school male students aged 12-15 years. The experiment consisted of three sessions: The first and third sessions were designed to evaluate the risk attitude of the participants. In the second session, participants were asked to guess the choices made by their peers, and then, the computer gave them feedback on the correctness of their predictions. Each participant was randomly assigned to risk-taking or risk-averse peers. Results: Our results revealed that teenagers who predicted risk-averse peers exhibited significant declines in their risk attitudes during the last session. On the other hand, participants with risk-seeking peers exhibited a significantly higher level of risk attitudes after predicting their peers. The data showed that these peer-biased changes in risk attitudes are proportional to the gap between teens and their peers' risk perspectives. Results showed that their perspectives aligned closer after receiving the information, and approximately a third of the gap was eliminated. Conclusion: Here, by combining choice data and computational modeling, we demonstrate that risky behavior is contagious among male adolescents. According to our data, peer-biased risk contagion, a socially motivated and deliberate process, is associated with social distance in teens. There's no causal directionality here, but we could speculate that peer influence goes hand-in-hand with social integration as an adaptive process.

Abstract:Previous neuroscience studies have investigated neural correlates of risky decision-making in a single-brain frame during pseudo social (predominantly non face-to-face) contexts. To fully understand the risky decision-making behavior in more natural social interactions, the present study employed a functional near-infrared spectroscopy (fNIRS) hyperscanning technique to simultaneously measure pairs of participants' fronto-temporal activations in a face-to-face gambling card-game. The intra-brain results revealed that both those who identified as males and as females showed higher activations in their mPFC and in the inferior parts of the frontopolar area, as well as in the tempo-parietal junction (TPJ) in cases involving higher versus lower risk. This is consistent with previous findings suggesting importance of the mentalizing network in decision tasks. The fNIRS results of inter-brain neural synchronization (INS) also revealed that males and females showed increased inter-brain coherence in the mPFC and dlPFC. Females, however, uniquely showed increased inter-brain coherence in the left TPJ. This INS result suggests that males may primarily depend on non-social cognitive ability to make a risky decision in a social interaction, while females may use both social and non-social cognitive abilities. The implications are also discussed for general topics of human interaction and two-person neuroscience.
HIV infection in a defined population of Swedish haemophiliacs.

Abstract:The results of a clinical follow-up of 124 Swedish patients with haemophilia A and B or severe form of von Willebrand's disease are reported, especially with regard to HIV infection and complications thereof. 44 of the patients were anti-HIV positive. In this group there was an increasing prevalence of lymphadenopathy (18 cases), diarrhoea and fungal infections. 10 patients had generalized persistent lymphadenopathy. Platelet counts were significantly lower and IgG, IgA and IgM levels significantly higher than in the anti-HIV negative group. Oligoclonal protein bands in the gammaglobulin region were found in 8 of the anti-HIV positive patients. The vast majority of these changes have appeared during the past year, and 1-5 years after seroconversion. Presently 22/44 (50%) anti-HIV positive haemophiliacs have at least one symptom related to this infection. Most of the clinical complications have been possible to treat or to mitigate so far.
Susceptibility to peer influence in adolescents: Associations between psychophysiology and behavior

Abstract:The current study investigated in-the-moment links between adolescents' autonomic nervous system activity and susceptibility to three types of peer influence (indirect, direct, continuing) on two types of behavior (antisocial, prosocial). The sample included 144 racially ethnically diverse adolescents (46% male, 53% female, 1% other; M age = 16.02 years). We assessed susceptibility to peer influence behaviorally using the Public Goods Game (PGG) while measuring adolescents' mean heart rate (MHR) and pre-ejection period (PEP). Three key findings emerged from bivariate dual latent change score modeling: (1) adolescents whose MHR increased more as they transitioned from playing the PGG alone (pre-influence) to playing while simply observed by peers (indirect influence) displayed more prosocial behavior; (2) adolescents whose PEP activity increased more (greater PEP activity = shorter PEP latency) as they transitioned from indirect influence to being encouraged by peers to engage in antisocial behavior (direct influence) engaged in more antisocial behavior; and (3) adolescents whose PEP activity decreased less as they transitioned from direct influence on prosocial behavior to playing the PGG alone again (continuing influence) displayed more continuing prosocial behavior (marginal effect). The discussion focuses on the role of psychophysiology in understanding adolescents' susceptibility to peer influence.
Anxiety may alter the role of fronto-striatal circuitry in adolescent risky decision-making

Abstract:Background Anxiety disorders often emerge in adolescence and are associated with risk aversion. Risk aversion conflicts with the typical adolescent approach-motivated phenotype and can interfere with learning and contribute to symptom maintenance. Methods We investigated the neural and behavioral correlates of risk avoidance in a diverse sample of adolescents ( N = 137; M Age = 11.3; 34.3 % white, 22.1 % Latino, 20 % Asian, 14.3 % Black, 9.3 % Mixed Race) as they completed a task involving risky decision-making and response inhibition during fMRI. Voluntary cautious choice was compared to successful response inhibition to isolate the neural systems underlying the decision to avoid a risk and identify their relation to risk-taking and anxiety in adolescents. Results Anxious adolescents self-reported more avoidance but demonstrated normative risk-taking on the laboratory task. Interestingly, they responded quickly during response inhibition but took longer to decide in the face of risk. All youth showed widespread recruitment of decision-making and salience network regions when deciding to avoid risk. The neural mechanisms driving avoidance differed based on anxiety such that left inferior frontal gyrus (IFG) activation was linked to risk avoidance in adolescents with low anxiety and risk-taking in anxious adolescents, while striatal connectivity was linked to risk avoidance in anxious adolescents and risk-taking in those with low anxiety. Limitations This work is cross-sectional and therefore cannot speak to causality or directionality of effects. Conclusions These results suggest that the neural mechanisms contributing to adolescent risk-taking may function to promote avoidance in anxious youth, increasing vulnerability to maladaptive avoidance and further anxiety development.

Abstract:Abstract The influence of peers is particularly strong during adolescence, however, its impacts on the functional and structural organization of the developing brain, cognition and psychopathological symptoms still remain unknown. Here, we capitalized on data from 6,064 pre-adolescents (mean age = 11.93 years) from the Adolescent Brain Cognitive Development (ABCD) database, to determine associations between two distinct peer environments (proportion of prosocial or delinquent friends) and the structural and functional architecture of the brain, cognition, as well as behavioral and emotional dysregulation. A higher proportion of prosocial friends was associated with fewer behavioral problems and larger fronto-cingulo-insular regions characterized by a high density of serotonin receptors. In contrast, a higher proportion of delinquent friends was associated with increased behavioral and mental problems but lower neurocognitive performance and smaller posterior cingulate/precuneus, as well as decreased functional connectivity in default-mode and fronto-striato-limbic circuits characterized by high expression of dopamine receptors. Prosocial friends mediated the association between the volume of certain brain regions (e.g., rACC) and prodromal psychosis / behavioral inhibition, while delinquent friends mediated the relationship between fronto-striato-limbic connectivity and behavioral problems. Prosocial friends also attenuated the development of internalizing problems whereas delinquent friends promoted externalizing symptoms. These results underscore the critical importance of a positive adolescent peer environment to promote mental health and resilience, indicate a direct link between peer environment and adolescent neuro-maturational processes, and suggest a holistic approach to healthy brain development and early interventions during adolescence.

Abstract:While much research on adolescent risk behaviour has focused on the development of prefrontal self-regulatory mechanisms, prior studies have elicited mixed evidence of a relationship between individual differences in the capacity for self-regulation and individual differences in risk taking. To explain these inconsistent findings, it has been suggested that the capacity for self-regulation may be, for most adolescents, adequately mature to produce adaptive behaviour in non-affective, "cold" circumstances, but that adolescents have a more difficult time exerting control in affective, "hot" contexts. To further explore this claim, the present study examined individual differences in self-control in the face of affective and non-affective response conflict, and examined whether differences in the functioning of cognitive control processes under these different conditions was related to risk taking. Participants completed a cognitive Stroop task, an emotional Stroop task, and a risky driving task known as the Stoplight game. Regression analyses showed that performance on the emotional Stroop task predicted laboratory risk-taking in the driving task, whereas performance on the cognitive Stroop task did not exhibit the same trend. This pattern of results is consistent with theories of adolescent risk-taking that emphasise the impacts of affective contextual influences on the ability to enact effective cognitive control.

Abstract:Previous research has clearly documented that risky decision making is different in young and older adults. Yet, there has been a relative dearth of research that seeks to understand such age-related changes in the neural activities associated with risk taking. To address this research issue, 21 men (12 young men, mean age 29.9 +/- 6.2 years and 9 older men, mean age 65.2 +/- 4.2 years) performed a risky-gains task while their brain activities were monitored by an fMRI scanner. The older adults, relative to their younger peers, presented with contralateral prefrontal activity, particularly at the orbitofrontal cortex. Furthermore, stronger activation of the right insula was observed for the older-aged participants compared to the younger-aged adults. The findings of this study are consistent with the a priori speculations established in accordance with the HAROLD model as well as previous findings. Findings of this study suggest that when making risky decisions, there may be possible neuropsychological mechanisms underlying the change in impulsive and risk-taking behaviors during the course of natural ageing.

Abstract:Humans are sophisticated social beings. Social cues from others are exceptionally salient, particularly during adolescence. Understanding how adolescents interpret and learn from variable social signals can provide insight into the observed shift in social sensitivity during this period. The present study tested 120 participants between the ages of 8 and 25 years on a social reinforcement learning task where the probability of receiving positive social feedback was parametrically manipulated. Seventy-eight of these participants completed the task during fMRI scanning. Modeling trial-by-trial learning, children and adults showed higher positive learning rates than did adolescents, suggesting that adolescents demonstrated less differentiation in their reaction times for peers who provided more positive feedback. Forming expectations about receiving positive social reinforcement correlated with neural activity within the medial prefrontal cortex and ventral striatum across age. Adolescents, unlike children and adults, showed greater insular activity during positive prediction error learning and increased activity in the supplementary motor cortex and the putamen when receiving positive social feedback regardless of the expected outcome, suggesting that peer approval may motivate adolescents toward action. While different amounts of positive social reinforcement enhanced learning in children and adults, all positive social reinforcement equally motivated adolescents. Together, these findings indicate that sensitivity to peer approval during adolescence goes beyond simple reinforcement theory accounts and suggest possible explanations for how peers may motivate adolescent behavior.

Abstract:Abstract Social learning theory posits that adolescents learn to adopt social norms by observing the behaviors of others and internalizing the associated outcomes. However, the underlying neural processes by which social learning occurs is less well-understood, despite extensive neurobiological reorganization and a peak in social influence sensitivity during adolescence. Forty-four adolescents (Mage = 12.2 years) completed an fMRI scan while observing their older sibling within four years of age (Mage = 14.3 years) of age complete a risky decision-making task. Group iterative multiple model estimation (GIMME) was used to examine patterns of directional brain region connectivity supporting social learning. We identified group-level neural pathways underlying social observation including the anterior insula to the anterior cingulate cortex and mentalizing regions to social cognition regions. We also found neural states based on adolescent sensitivity to social learning via age, gender, modeling, differentiation, and behavior. Adolescents who were more likely to be influenced elicited neurological up-regulation whereas adolescents who were less likely to be socially influenced elicited neurological down-regulation during risk-taking. These findings highlight patterns of how adolescents process information while a salient influencer takes risks, as well as salient neural pathways that are dependent on similarity factors associated with social learning theory.
