Abstract:Rice straw was subjected to fungal pretreatment using Pleurotus ostreatus and Trichoderma reesei to improve its biodegradability and methane production via solid-state anaerobic digestion (SS-AD). Effects of moisture content (65%, 75% and 85%), and incubation time (10, 20 and 30 d) on lignin, cellulose, and hemicellulose degradation during fungal pretreatment and methane yield during anaerobic digestion were assessed via comparison to untreated rice straw. Pretreatment with P. ostreatus was most effective at 75% moisture content and 20 d incubation resulting in 33.4% lignin removal and a lignin/cellulose removal ratio (selectivity) of 4.2. In comparison Trichoderma reesei was most effective at 75% moisture content and 20 d incubation resulting in 23.6% lignin removal and a lignin/cellulose removal ratio (selectivity) of 2.88. The corresponding methane yields were 263 and 214 L/kg volatile solids (VS), which were 120% and 78.3% higher than for the untreated rice straw, respectively. (C) 2016 Elsevier Ltd. Combinations of Fungal and Milling Pretreatments for Enhancing Rice Straw Biogas Production During Solid-State Anaerobic Digestion.

Abstract:Rice straw was pretreated by different combinations of physical (milling) and biological (incubation with Pleurotus ostreatus fungus) treatment to improve its biodegradability and biogas production during solid-state anaerobic digestion (SS-AD). Effects of milling (⩽2mm) and incubation time (10, 20 and 30d), on lignin, cellulose, and hemicellulose degradation during fungal pretreatment and methane yield during digestion were assessed by comparison with untreated rice straw. Both incubation time and milling had significant impacts on both lignin removal during fungal pre-treatment and methane yield during digestion. A combination of fungal pretreatment at 30days followed by milling prior to anaerobic digestion resulted in 30.4% lignin removal, the highest selectivity value (the ratio between relative lignin removal and relative cellulose removal) of 4.22, and the highest methane yield of 258L/kgVS. This was equivalent to a 165% increase in methane yield from SS-AD compared to untreated rice straw.

Abstract:Abstract Biological treatment of cereal straw for ruminant nutrition purposes might present an environmentally friendly option of valorizing a widely available by-product of grain production for farming systems with low external input. Several strains of white-rot fungi have been selected in the past under mostly controlled laboratory conditions for their capacity of lignin degradation. The study adapted to conditions on farm for upscaling purposes. The development of the in vitro straw digestibility with two different moistening pre-treatments and inoculated with three different fungi species, namely Pleurotus ostreatus , Ceriporiopsis subvermispora and Volvariella volvacea , was determined up to 42 days of fermentation with five sampling times. The effect of physical straw pre-treatments on nutritional parameters was evaluated. The neutral detergent fiber digestibility (NDFD 30h ), enzymatically soluble organic substance (ELOS) and the gas production (Hohenheim Feed value Test, HFT) as indicators for in vitro ruminal degradability decreased over time independent of the fungus: HFT, ELOS and NDFD 30h by up to 50, 35 and 30% of the original straw. Remoistening and autoclaving the straw increased the gas production significantly by 2.6 mL/200 g dry matter (DM), and ELOS and NDFD 30h by 45 and 51 g/kg DM compared to the original straw (34.9 mL/200 mg DM, 342 g/kg DM, 313 g/kg NDF).

Evaluation of fungal degradation of wheat straw cell wall using different analytical methods from ruminant nutrition perspective

Abstract:BACKGROUND: White rot fungi have been used to improve the nutritive value of lignocellulose for ruminants. In feed analysis, the Van Soest method is widely used to determine the cell wall contents. To assess the reliability of this method (Method A) for determination of cell wall contents in fungal-treated wheat straw, we compared a combined monosaccharide analysis and pyrolysis coupled to gas chromatography with mass spectrometry (Py-GC/MS) (Method B). Ruminal digestibility, measured as in vitro gas production (IVGP), was subsequently used to examine which method explains best the effect of fungal pretreatment on the digestibility of wheat straw.RESULTS: Both methods differed considerably in the mass recoveries of the individual cell wall components, which changed on how we assess their degradation characteristics. For example, Method B gave a higher degradation of lignin (61.9%), as compared to Method A (33.2%). Method A, however, showed a better correlation of IVGP with the ratio of lignin to total structural carbohydrates, as compared to Method B (Pearson's r of -0.84 versus -0.69). Nevertheless, Method B provides a more accurate quantification of lignin, reflecting its actual modification and degradation. With the information on the lignin structural features, Method B presents a substantial advantage in understanding the underlying mechanisms of lignin breakdown. Both methods, however, could not accurately quantify the cellulose contents - among others, due to interference of fungal biomass.CONCLUSION: Method A only accounts for the recalcitrant residue and therefore is more suitable for evaluating ruminal digestibility. Method B allows a more accurate quantification of cell wall, required to understand and better explains the actual modification of the cell wall. The suitability of both methods, therefore, depends on their intended purposes. © 2019 The Authors. Abstract:BackgroundThe white-rot fungi Ceriporiopsis subvermispora (Cs), Pleurotus eryngii (Pe), and Lentinula edodes (Le) have been shown to be high-potential species for selective delignification of plant biomass. This delignification improves polysaccharide degradability, which currently limits the efficient lignocellulose conversion into biochemicals, biofuels, and animal feed. Since selectivity and time efficiency of fungal delignification still need optimization, detailed understanding of the underlying mechanisms at molecular level is required. The recently developed methodologies for lignin quantification and characterization now allow for the in-depth mapping of fungal modification and degradation of lignin and, thereby, enable resolving underlying mechanisms.ResultsWheat straw treated by two strains of Cs (Cs1 and Cs12), Pe (Pe3 and Pe6) and Le (Le8 and Le10) was characterized using semi-quantitative py-GC–MS during fungal growth (1, 3, and 7 weeks). The remaining lignin after 7 weeks was quantified and characterized using 13C lignin internal standard based py-GC–MS and whole cell wall HSQC NMR. Strains of the same species showed similar patterns of lignin removal and degradation. Cs and Le outperformed Pe in terms of extent and selectivity of delignification (Cs ≥ Le >> Pe). The highest lignin removal [66% (w/w); Cs1] was obtained after 7 weeks, without extensive carbohydrate degradation (factor 3 increased carbohydrate-to-lignin ratio). Furthermore, though after treatment with Cs and Le comparable amounts of lignin remained, the structure of the residual lignin vastly differed. For example, Cα-oxidized substructures accumulated in Cs treated lignin up to 24% of the total aromatic lignin, a factor two higher than in Le-treated lignin. Contrarily, ferulic acid substructures were preferentially targeted by Le (and Pe). Interestingly, Pe-spent lignin was specifically depleted of tricin (40% reduction). The overall subunit composition (H:G:S) was not affected by fungal treatment.ConclusionsCs and Le are both able to effectively and selectively delignify wheat straw, though the underlying mechanisms are fundamentally different. We are the first to identify that Cs degrades the major β-O-4 ether linkage in grass lignin mainly via Cβ–O–aryl cleavage, while Cα–Cβ cleavage of inter-unit linkages predominated for Le. Our research provides a new insight on how fungi degrade lignin, which contributes to further optimizing the biological upgrading of lignocellulose.

Abstract:Five white-rot fungi Pleurotus ostreatus, Lentinus edodes, Hericium erinaceus, Pleurotus eryngii and Flammulina filiformis were studied (solid-state incubation and in vitro gas production) to determine lignin degradation and optimal duration of fermentation of corn straw. All fungi significantly decreased lignin, with optimal reductions after 28 d. Although cellulose also decreased, L. edodes and P. eryngii minimized these losses. In intro dry matter digestibility, total volatile fatty acid concentration and total gas production of fermented corn straw decreased (P < 0.001) as fermentation was prolonged, with improved rumen fermentability for all fungal treatments except F. filiformis. Total gas production in L. edodes did not decrease but peaked on day 28, whereas F. filiformis reduced methane emission. In conclusion, fermentation of corn straw with P. eryngii or L. edodes for 28 d degraded lignin and improved nutritional value as ruminant feed.

Abstract:Solid-state fermentation with white-rot fungi is an interesting alternative to chemical straw treatment for ruminant nutrition. However, for practical implementation on farms, feasible handling and its effect on nutritional characteristics have to be tested beforehand. Chopped wheat straw was either soaked and drained or just remoistened to about 24% dry matter without subsequent sterilization. Moist straw was inoculated with Pleurotus ostreatus, Ceriporiopsis subvermispora, or Volvariella volvacea. Fermentation lasted up to 42 d with weekly or shorter sampling intervals. Fiber fractions, minerals and elements, and non-starch polysaccharides were analyzed, and microflora was plate counted. Lactic acid bacteria of selected samples were identified by MALDI-TOF. All inoculated fungi grew well under the selected conditions expressed by the visible mycelium and specific smell. P. ostreatus developed fruiting bodies in the given time. Initial numbers of lactic acid bacteria were >8.0 log cfu/g. In the beginning, Weissella confusa/cibaria dominated. However, neither decrease in lignin nor cellulose concentration was observed during the period in either of the treatments, thus seeming to be inappropriate for ruminant nutrition purposes. Some elements and minerals peaked, especially towards the second and third week (Na, Cl, Mg, Fe, and Mn). Growth conditions for mushrooms were optimized by remoistening the straw with a defined amount of water.

Structural Motifs of Wheat Straw Lignin Differ in Susceptibility to Degradation by the White-Rot Fungus Ceriporiopsis subvermispora

Abstract:The white-rot fungus Ceriporiopsis subvermispora delignifies plant biomass extensively and selectively and, therefore, has great biotechnological potential. We previously demonstrated that after 7 weeks of fungal growth on wheat straw 70% w/w of lignin was removed and established the underlying degradation mechanisms via selectively extracted diagnostic substructures. In this work, we fractionated the residual (more intact) lignin and comprehensively characterized the obtained isolates to determine the susceptibility of wheat straw lignin's structural motifs to fungal degradation. Using 13C IS pyrolysis gas chromatography–mass spectrometry (py-GC-MS), heteronuclear single quantum coherence (HSQC) and 31P NMR spectroscopy, and size-exclusion chromatography (SEC) analyses, it was shown that β-O-4′ ethers and the more condensed phenylcoumarans and resinols were equally susceptible to fungal breakdown. Interestingly, for β-O-4′ ether substructures, marked cleavage preferences could be observed: β-O-4′-syringyl substructures were degraded more frequently than their β-O-4′-guaiacyl and β-O-4′-tricin analogues. Furthermore, diastereochemistry (threo > erythro) and γ-acylation (γ-OH > γ-acyl) influenced cleavage susceptibility. These results indicate that electron density of the 4′-O-coupled ring and local steric hindrance are important determinants of oxidative β-O-4′ ether degradation. Our findings provide novel insight into the delignification mechanisms of C. subvermispora and contribute to improving the valorization of lignocellulosic biomass.This paper provides insight into the cleavage preference of wheat straw lignin's structural motifs by the industrially promising white-rot fungus Ceriporiopsis subvermispora.

Abstract:Lignin degradation is important for enhancing the digestibility and improving the nutritive quality of ruminant feeds. White rot fungi are well known for their bioconversion of lignocellulosic biomass. The objective of this paper was to evaluate whether Lentinus sajor-caju, Pleurotus ostreatus, Phyllotopsis rhodophylla, Pleurotus djamor, Pleurotus eryngii, and Pleurotus citrinopileatus treatments altered the chemical compositions of highland barley straw constituents and enhanced their nutritional value as a ruminant feed. All white rot fungi significantly increased the relative crude protein (CP), ethyl ether extract (EE), starch, soluble protein (SP), and non-protein nitrogen (NPN) contents but decreased the ash, neutral detergent fiber (NDF), acid detergent fiber (ADF), acid detergent lignin (ADL), and acid detergent insoluble protein (ADFIP) contents. In addition, L. sajor-caju treatment increased (p < 0.001) the levels of PA, PB2, PB3, CA, CB1, CB2, and CNSC, but reduced (p < 0.001) the PC and CC in the solid-state fermentation of highland barley straw. Maximum ligninlysis (50.19%) was optimally produced in the presence of 1.53% glucose and 2.29% urea at 22.72 °C. The in vitro dry matter digestibility and total volatile fatty acid concentrations of fermented highland barley straw, as well as the fermentability, were optimized and improved with L. sajor-caju, which degraded the lignocellulose and improved the nutritional value of highland barley straw as a ruminant feed.

Abstract:PurposeThis study evaluated the treatment of naked oat straw with four species of white rot fungi (Phanerochaete chrysosporium, Pleurotus ostreatus, Irpex lacteus, and Phlebia acerina) for solid-state fermentation in terms of the changes in chemical composition, lignocellulosic structure, and in vitro ruminal digestion.MethodsAll the fungi were inoculated in naked oat straw for 28 days to evaluate the changes of above indexes weekly.ResultsThe results showed that all of the fungus species could alter the multi-scale structure of the straw to different extents when compared to the control. P. acerina caused the greatest degradation of acid detergent lignin (46.51%), followed by P. chrysosporium (44.20%), while P. ostreatus and I. lacteus were slightly weaker in terms of acid detergent lignin degradation than the other two species. The dry matter loss for all treatments was below 20% except for P. chrysosporium, and all of the fungus species caused holocellulose degradation during treatment, with P. chrysosporium in particular exhibiting non-selectivity. Compared with the other fungi, I. lacteus treatment resulted in moderate losses of hemicellulose and cellulose (31.92% and 15.44%, respectively). In addition to P. chrysosporium, the in vitro dry matter digestion was improved with treatment by the other three fungi. Treatment with I. lacteus, led to improvement in dry matter, neutral detergent fiber and acid detergent fiber degradability of 16.22%, 37.12%, and 40.05%, respectively.ConclusionIrpex lacteus outperformed the other three fungi in terms of decomposing three-dimensional structures and improving the nutritional value of naked oat straw.Graphic Abstract

Synergistic Effect of Cellulase and Xylanase on in Vitro Rumen Fermentation and Microbial Population with Rice Straw As Substrate

Abstract:The in vitro gas test was conducted to investigate the effects of cellulase (CEL) and xylanase (XYL) on in vitro rumen fermentation and microbial population with rice straw as substrate. Three levels of CEL (0, 7.5 and 15 U/g of DM) and XYL (0, 15 and 30 U/g of DM) were tested in a 3x3 factorial arrangement. Addition of CEL and XYL could affect the gas production parameters. Methane emission was increased linearly by GEL addition (P < 0.01), while the methane production was greater when XYL added at the intermediate dose (quadratic, P < 0.01). Total volatile fatty acids (CEL, quadratic, P < 0.01; XYL, linear and quadratic, P < 0.05) were enhanced by either CEL or XYL addition. Ammonia nitrogen was reduced by CEL (linear and quadratic, P < 0.05), while microbial protein was enhanced by addition of CEL (quadratic, P < 0.01) and XYL (linear and quadratic, P < 0.01). In vitro degradability of DM and NDF were increased by addition of CEL (quadratic, P < 0.05) or XYL (quadratic, P < 0.01). Addition of CEL increased the copy number of total bacteria (linear, P < 0.01) and Fibrobacter succinogens (linear and quadratic, P < 0.05). The outcome of this research indicated that the application of CEL and XYL could improve rumen fermentation, increase rice straw digestion and affect the rumen microbial population. Combination of CEL and XYL at the middle level is more effective than a single CEL or XYL.

Abstract:Abstract Background Identifying new high-performance enzymes or enzyme complexes to enhance biomass degradation is the key for the development of cost-effective processes for ethanol production. Irpex lacteus is an efficient microorganism for wheat straw pretreatment, yielding easily hydrolysable products with high sugar content. Thus, this fungus was selected to investigate the enzymatic system involved in lignocellulose decay, and its secretome was compared to those from Phanerochaete chrysosporium and Pleurotus ostreatus which produced different degradation patterns when growing on wheat straw. Extracellular enzymes were analyzed through 2D-PAGE, nanoLC/MS-MS, and homology searches against public databases. Results In wheat straw, I. lacteus secreted proteases, dye-decolorizing and manganese-oxidizing peroxidases, and H 2 O 2 producing-enzymes but also a battery of cellulases and xylanases, excluding those implicated in cellulose and hemicellulose degradation to their monosaccharides, making these sugars poorly available for fungal consumption. In contrast, a significant increase of β-glucosidase production was observed when I. lacteus grew in liquid cultures. P. chrysosporium secreted more enzymes implicated in the total hydrolysis of the polysaccharides and P. ostreatus produced, in proportion, more oxidoreductases. Conclusion The protein pattern secreted during I. lacteus growth in wheat straw plus the differences observed among the different secretomes, justify the fitness of I. lacteus for biopretreatment processes in 2G-ethanol production. Furthermore, all these data give insight into the biological degradation of lignocellulose and suggest new enzyme mixtures interesting for its efficient hydrolysis.

Abstract:Various pretreatments are employed to increase the utilization of rice straw as a ruminant feed ingredient to minimize its negative environmental impact. However, an efficient alternative is still needed. The purpose of this study was to evaluate the ability of ammonia and/or white-rot fungi (Pleurotus ostreatus) to degrade lignin, increase the nutritional value, and enhance the rumen fermentability of rice straw. Rice straw was treated with ammonia and/or basidiomycete white-rot fungi (P. ostreatus) with untreated straw as control under solid-state fermentation employing a completely randomized design. The crude protein increased from 2.05% in the control to 3.47% in ammoniated rice straw, 5.24% in basidiomycete white-rot fungi (P. ostreatus), and 6.58% in ammoniated-basidiomycete white-rot fungi-treated (P. ostreatus) rice straw. The ammoniated-basidiomycete white-rot fungi-treated (P. ostreatus) rice straw had the least lignin content (3.76%). Ammoniated-basidiomycete white-rot fungi-treated (P. ostreatus) rice straw had improved in vitro dry matter digestibility (65.52%), total volatile fatty acid (76.56 mM), and total gas production (56.78 mL/g) compared to ammoniated rice straw (56.16%, 67.71 mM, 44.30 mL/g) or basidiomycete white-rot fungi-treated (P. ostreatus) rice straw (61.12%, 75.36 mM, 49.31 mL/g), respectively. The ammoniated-basidiomycete white-rot fungi (P. ostreatus) treatment improved rice straw’s nutritional value, in vitro dry matter digestibility, volatile fatty acids, and gas production.

Abstract:This study was conducted to investigate the effects of treatment of rice straw (RS) with sodium hydroxide (SH) and ammonium bicarbonate (AB) on fermentation characteristics, fibrolytic enzyme activities and populations of liquid- and solid-associated ruminal microbes in vitro. In vitro gas test was performed to evaluate the nutritive value of the treated straw. Specific activities of fibre-degrading enzymes, namely carboxymethyl cellulase (CMCase), avicelase that catalyzes the breakdown of crystalline cellulose (Avicel) to cellobiose and xylanase were estimated from the amount of reducing sugars released from the solid-bound microbes. Total deoxyribonucleic acid (DNA) was extracted from the liquid- and solid-associated ruminal microbes, respectively, and populations of rumen microbes were determined by real-time quantitative polymerase chain reaction (PCR). Populations of total rumen fungi, Ruminococcus flavefaciens and Fibrobacter succinogenes were expressed as a proportion of total rumen bacterial 16S ribosomal deoxyribonucleic acid (rDNA). The cumulative gas production (GP) at all incubation times, potential GP and rate constant of GP were highest for SH-RS, followed by AB-RS and lowest for the untreated RS (P<0.05). Microbial crud protein increased with incubation time, and was higher for the treated straws than for the untreated (P<0.05). Both treated and untreated straws maintained a typical roughage type of fermentation with a high proportion of acetate. The SH treatment increased CMCase and avicelase activities at all incubation times. Both SH and AB treatments significantly increased xylanase activity. Rumen fungi were significantly increased with incubation time in both liquid and solid phases for SH-RS, but not affected by AB treatment. Both solid- and liquid-associated R. flavefaciens were higher in treated straws than in the untreated, with higher solid-associated R. flavefaciens in SH-RS than in AB-RS at early incubation periods. Solid-associated F. succinogenes was lower, and liquid-associated F. succinogenes was higher in proportion of total ruminal bacterial 16S rDNA in SH-RS than in other two straws, with little difference in solid-associated F. succinogenes between untreated and AB straws. Liquid-associated F. succinogenes was lower at early incubation in AB-RS. It is inferred that chemical treatments enhance the nutritive value of RS through improving rumen fermentation and fibrolytic enzyme activities that are mainly resultant from more available substrate, and has great influences on rumen microbial distribution and populations, but their fluctuating pattern with incubation time is slightly different between two treated straws.
Combination of Pretreatment with White Rot Fungi and Modification of Primary and Secondary Cell Walls Improves Saccharification

Abstract:Plant cell walls have protective and structural functions conferring resistance to degradation. The lignin and hemicellulose network surrounding the cellulose microfibrils is insoluble unless subjected to harsh treatments. As lignin, pectin and xylan are effective barriers to cellulose extraction and hydrolysis, reducing their presence in cell walls improves saccharification. Microorganisms that can depolymerise lignin are of extreme interest to the biofuel industry. White rot fungi can be effective in pretreatment of lignocellulosic biomass prior to saccharification. Here, we show the cumulative effects of pretreating biomass with two white rot fungi, Phanerochaete chrysosporium and Trametes cingulata, on tobacco lines with reduced lignin or xylan, caused by suppression of the CINNAMOYL-CoA REDUCTASE, CINNAMATE-4-HYDROXYLASE, TOBACCO PEROXIDASE 60 or UDP-GLUCURONATE DECARBOXYLASE and on Arabidopsis thaliana with reduced de-esterified homogalacturonan content, obtained by overexpressing a pectin methyl esterase inhibitor or constitutively expressing the Aspergillus nigerPOLYGALACTURONASE II gene. Tests were extended to fresh material from an Arabidopsis mutant for a cell wall peroxidase. We demonstrate that fungal pretreatment is a reliable method of improving cellulose accessibility in biofuel feedstocks, fresh material and cell wall residues from different plants. These results contribute to the understanding of the consequences of primary and secondary cell wall perturbations on lignocellulosic biomass accessibility to white rot fungi and on saccharification yield. A comparison of the effects of P. chrysosporium and T. cingulata on tobacco saccharification also highlights the limitation of current knowledge in this research field and the necessity to systematically test culture conditions to avoid generalisations.

Abstract:Lignin is main composition of agricultural biomass which can be decomposed through enzymatic hydrolysis by fungi. However, there are still needs to identify more efficient and effective fungal stain for biomass valorization. In this study, lignin degrading fungi from birch forest were screened for sustainable degradation of waste agricultural straws. The most effective strain was identified as Cerrena unicolor GC.u01 using 18 S rDNA gene-sequencing technology. Three different crop straws (corn stalk, rice and wheat straws) were used for the biotreatment studies. The activities of lignin degrading enzymes, laccase (Lac), cellulase and xylanase, secreted by C. unicolor were also determined. Scanning electron microscopy (SEM), fourier transform infrared spectroscopy (FTIR) and thermal gravimetric analyzer (TGA) were further used to monitor the effects of the biotreatment process. The results showed that C. unicolor degraded 34.3% rice straw lignin, a percentage which was higher than other isolated strains after 15 d straw liquid fermentation. The highest Lac activity (8.396 U•mL − 1 ) was observed with corn stalk on the 7 d. Cellulase and xylanase activities, in the same biomass, were higher than those of wheat and rice straws after 15 d. Furthermore, SEM, FTIR and TGA analyses showed that C. unicolor pretreatment process had significant effects on corn stalk, rice and wheat straws' structures. The newly isolated stain of C. unicolor demonstrated high lignin degradation potential that can provide effective, ecofriendly means of valorizing biomass to industrial useable raw-material.

Abstract:Abstract The study sought to investigate the potentials of axenic cultures of Pleurotus ostreatus , Phanerochaete chrysosporium and their coculture ( P. chrysosporium and P. ostreatus ) to break down lignin and to enhance the rumen fermentability of rice straw. Rice straw was fermented by two lignin-degrading fungi, namely, P. ostreatus , P . chrysosporium and its coculture ( P . ostreatus and P . chrysosporium ) with uninoculated straw as control under solid-state fermentation employing a completely randomized research design. The coculture exhibited a mutual intermingling plus inhibition interaction. The fungi treatment increased the crude protein from (5.1%) in the control to (6.5%, 6.6%, and 6.7%) in the P . ostreatus , P . chrysosporium and coculture respectively. The coculture treated straw had a lower lignin content (5.3%) compared to the P . chrysosporium (6.2%) with the P . ostreatus recording the least (3.3%) lignin fraction. Treatment of rice straw with coculture improved the in vitro dry matter digestibility (68.1%), total volatile fatty acids (35.3 mM), and total gas (57.4 ml/200 mg) compared to P . chrysosporium (45.1%, 32.2 mM, 44.4 ml/200 mg) but was second to P . ostreatus (75.3%, 38.3 mM, 65.6 ml/200 mg). Instead of an anticipated synergistic effect from the coculture, a competitive antagonistic effect was rather observed at the end of the study, a condition that can be attributed to the coculture behavior.

Abstract:Abstract Background Wheat straw is one of the most abundant crop residues produced in the world, making it highly interesting as a substrate for biogas production. However, due to the complex structure, its degradability and gas yield are low. The degradability can be improved by pre-treatment, making the material more accessible to microbial degradation. Methods To investigate the microbial response to straw as a feed stock for biogas production, this study examined the community structure of cellulose-degrading bacteria in lab-scale biogas digesters operating with manure, alone or in co-digestion with straw, with and without pre-treatment (steam-explosion) at different temperatures. The community was studied by targeting the functional gene encoding glycoside hydrolases of families 5 and 48 using T-RFLP, clone libraries and qPCR. Results In general, bacteria belonging to the phyla Firmicutes and Bacteroidetes dominated the cellulose-degrading bacteria community in all digesters. The degree of similarity to the characterised bacteria was often low, and some clones were more closely related to the uncultured bacteria. The addition of straw, pre-treatment of straw and increasing operating temperature all affected the cellulose-degrading community structure, with differing responses in the cel48 and cel5 communities. Both communities changed in response to temperature, while only the cel5 community was affected by the addition of straw and cel48 community by straw pre-treatment. Conclusions The addition of straw, pre-treatment of straw and operating temperature all affected the cellulose-degrading community in biogas digesters, but there were no major differences in the digester performance and gas yield.

Abstract:A great body of evidence is available on the in vitro efficacy of white-rot fungi (WRF) to degrade lignin in fibre-rich biomass (e.g. wheat straw, wood chips and rice straw (RS)) and improve the biomass' nutritive value for ruminants. Determining the impact of incubation temperature of three WRF to improve the nutritional value of rice straw. Growth of Ceriporiopsis subvermispora , Lentinula edodes and Pleurotus eryngii on RS for 26 days at the following six temperature regimes: continuous at 24°C, 30°C, 35°C and 40°C, and 3 days at 35°C and 40°C, with subsequent days at 24°C. In a follow-up experiment, improvement in fermentability in buffered rumen fluid of RS treated by the three WRF at 24°C and 30°C for up to 8 weeks was investigated. All three fungi grew at temperatures up to 35°C, with no growth observed at 40°C, with C. subvermispora being more temperature sensitive. There were significant differences in cellulose, hemicellulose and lignin degradation of RS at 24°C and 30°C, with C. subvermispora degrading 69% and 90% of the hemicellulose and lignin respectively at 30°C, greater than at 24°C (55% and 80% respectively). For L. edodes , there were significant differences in cellulose degradation between 24°C and 30°C, with 12% more degradation at 30°C, but not for hemicellulose and lignin. In vitro gas production showed no significant differences between the two incubation temperatures for either of the two fungi. Pleurotus eryngii treatment did not show any improvement in terms of in vitro gas production. Treatment of RS with L. edodes and C. subvermispora , but not P. eryngii , is robust and temperature changes will not have a major impact on their efficacy as long as the temperature remains below 30°C. Temperature during the incubation of WRF with rice straw needs to be below 30°C for this biotechnology to be applied in practice.
