Abstract:In this manuscript, we have studied the microstructure of the axonal cytoskeleton and adopted a bottom-up approach to evaluate the mechanical responses of axons. The cytoskeleton of the axon includes the microtubules (MT), Tau proteins (Tau), neurofilaments (NF), and microfilaments (MF). Although most of the rigidity of the axons is due to the MT, the viscoelastic response of axons comes from the Tau. Early studies have shown that NF and MF do not provide significant elasticity to the overall response of axons. Therefore, the most critical aspect of the mechanical response of axons is the microstructural topology of how MT and Tau are connected and construct the cross-linked network. Using a scanning electron microscope (SEM), the cross-sectional view of the axons revealed that the MTs are organized in a hexagonal array and cross-linked by Tau. Therefore, we have developed a hexagonal Representative Volume Element (RVE) of the axonal microstructure with MT and Tau as fibers. The matrix of the RVE is modeled by considering a combined effect of NF and MF. A parametric study is done by varying fiber geometric and mechanical properties. The Young's modulus and spacing of MT are varied between 1.5 and 1.9 GPa and 20-38 nm, respectively. Tau is modeled as a 3-parameter General Maxwell viscoelastic material. The failure strains for MT and Tau are taken to be 50 and 40%, respectively. A total of 4 RVEs are prepared for finite element analysis, and six loading cases are inspected to quantify the three-dimensional (3D) viscoelastic relaxation response. The volume-averaged stress and strain are then used to fit the relaxation Prony series. Next, we imposed varying strain rates (between 10/sec to 50/sec) on the RVE and analyzed the axonal failure process. We have observed that the 40% failure strain of Tau is achieved in all strain rates before the MT reaches its failure strain of 50%. The corresponding axonal failure strain and stress vary between 6 and 11% and 5-19.8 MPa, respectively. This study can be used to model macroscale axonal aggregate typical of the white matter region of the brain tissue.
A Fractional Viscoelastic Model Of The Axon In Brain White Matter

Abstract:Traumatic axonal injury occurs when loads experienced on the tissue-scale are transferred to the individual axons. Mechanical characterization of axon deformation especially under dynamic loads however is extremely difficult owing to their viscoelastic properties. The viscoelastic characterization of axon properties that are based on interpretation of results from in-vivo brain Magnetic Resonance Elastography (MRE) are dependent on the specific frequencies used to generate shear waves with which measurements are made. In this study, we aim to develop a fractional viscoelastic model to characterize the time dependent behavior of the properties of the axons in a composite white matter (WM) model. The viscoelastic powerlaw behavior observed at the tissue level is assumed to exist across scales, from the continuum macroscopic level to that of the microstructural realm of the axons. The material parameters of the axons and glia are fitted to a springpot model. The 3D fractional viscoelastic springpot model is implemented within a finite element framework. The constitutive equations defining the fractional model are coded using a vectorized user defined material (VUMAT) subroutine in ABAQUS finite element software. Using this material characterization, representative volume elements (RVE) of axons embedded in glia with periodic boundary conditions are developed and subjected to a relaxation displacement boundary condition. The homogenized orthotropic fractional material properties of the axon-matrix system as a function of the volume fraction of axons in the ECM are extracted by solving the inverse problem.

Hyper-viscoelastic 3D response of axons subjected to repeated tensile loads in brain white matter

Abstract:A novel finite element method (FEM) is developed to study mechanical response of axons embedded in extra cellular matrix (ECM) when subjected to harmonic uniaxial stretch under purely non-affine kinematic boundary conditions. The proposed modeling approach combines hyper-elastic (such as Ogden model) and time/frequency domain viscoelastic constitutive models to evaluate the effect of parametrically varying oligodendrocyte-axon tethering under harmonic stretch at 50Hz. A hybrid hyper-viscoelastic material (HVE) model enabled the analysis of repeated uniaxial load on stress propagation and damage accumulation in white matter. In the proposed FEM, oligodendrocyte connections to axons are depicted via a spring-dashpot model. This tethering technique facilitates contact definition at various locations, parameterizes connection points and varies stiffness of connection hubs. Results from a home-grown FE submodel configuration of a single oligodendrocyte tethered to axons at various locations are presented. Root mean square deviation (RMSD) are computed between stress-strain plots to depict trends in mechanical response. Steady-state dynamic (SSD) simulations show stress relaxation in axons. Gradual axonal softening under repetitive loads is illustrated employing Prony series - HVE models. Representative von-Mises stress plots indicate that undulated axons experience bending stresses along their tortuous path, suggesting greater susceptibility to damage accumulation and fatigue failure due to repeated strains.

Visco-hyperelastic characterization of human brain white matter micro-level constituents in different strain rates

Abstract:In this study, we propose a computational characterization technique for obtaining the material properties of axons and extracellular matrix (ECM) in human brain white matter. To account for the dynamic behavior of the brain tissue, data from time-dependent relaxation tests of human brain white matter in different strain rates are extracted and formulated by a visco-hyperelastic constitutive model consisting of the Ogden hyperelastic model and the Prony series expansion. Through micromechanical finite element simulation, a derivative-free optimization framework designed to minimize the difference between the numerical and experimental data is used to identify the material properties of the axons and ECM. The Prony series expansion parameters of axons and ECM are found to be highly affected by the Prony series expansion coefficients of the brain white matter. The optimal parameters of axons and ECM are verified through micromechanical simulation by comparing the averaged numerical response with that of the experimental data. Moreover, the initial shear modulus and the reduced shear modulus of the axons are found for different strain rates of 0.0001, 0.01, and 1 s-1. Consequently, first- and second-order regressions are used to find relations for the prediction of the shear modulus at the intermediate strain rates. The macro-level experimental data in different strain rates are used in the context of simulation-based optimization to obtain the properties of axons and extracellular matrix material.
Bidirectional hyperelastic characterization of brain white matter tissue

Abstract:Biomechanical study of brain injuries originated from mechanical damages to white matter tissue requires detailed information on mechanical characteristics of its main components, the axonal fibers and extracellular matrix, which is very limited due to practical difficulties of direct measurement. In this paper, a new theoretical framework was established based on microstructural modeling of brain white matter tissue as a soft composite for bidirectional hyperelastic characterization of its main components. First the tissue was modeled as an Ogden hyperelastic material, and its principal Cauchy stresses were formulated in the axonal and transverse directions under uniaxial and equibiaxial tension using the theory of homogenization. Upon fitting these formulae to the corresponding experimental test data, direction-dependent hyperelastic constants of the tissue were obtained. These directional properties then were used to estimate the strain energy stored in the homogenized model under each loading scenario. A new microstructural composite model of the tissue was also established using principles of composites micromechanics, in which the axonal fibers and surrounding matrix are modeled as different Ogden hyperelastic materials with unknown constants. Upon balancing the strain energies stored in the homogenized and composite models under different loading scenarios, fully coupled nonlinear equations as functions of unknown hyperelastic constants were derived, and their optimum solutions were found in a multi-parametric multi-objective optimization procedure using the response surface methodology. Finally, these solutions were implemented, in a bottom-up approach, into a micromechanical finite element model to reproduce the tissue responses under the same loadings and predict the tissue responses under unseen non-equibiaxial loadings. Results demonstrated a very good agreement between the model predictions and experimental results in both directions under different loadings. Moreover, the axonal fibers with hyperelastic characteristics stiffer than the extracellular matrix were shown to play the dominant role in directional reinforcement of the tissue.

Abstract:Axon is a long protrusion from the neuronal cell body whose interconnection forms the nervous network and allows biomechanical signals to be transmitted among neurons. From the mechanic’s point of view, the axon cytoskeleton consists of bundles of microtubules (MTs), cross-linked by microtubule-associated protein (MAP) tau, and supported by a periodic array of actin–spectrin rings. Yet, the fundamental question of how these actin–spectrin rings and microtubule bundles behave in synergy to provide the required mechanical strength to the whole axon is still poorly understood. Here, we developed a coarse-grained molecular dynamics model of axon to address this outstanding issue. We show that the dynamic response of spectrin filaments plays a vital role in the strain-softening of axon, which serves as a buffering mechanism for neurons to accommodate externally imposed deformation. Furthermore, the actin–spectrin structure is found to be essential for maintaining the mechanical stability of axon and allowing it to bear larger compressive forces. Our model and predictions not only explain recent experimental observations on axonal mechanics, but also provide insights on how to potentially modify the mechanical response of axonal cytoskeleton and therefore tune its capability in executing different biological duties.
Modeling links softening of myelin and spectrin scaffolds of axons after a concussion to increased vulnerability to repeated injuries

Abstract:Damage to the microtubule lattice, which serves as a rigid cytoskeletal backbone for the axon, is a hallmark mechanical initiator of pathophysiology after concussion. Understanding the mechanical stress transfer from the brain tissue to the axonal cytoskeleton is essential to determine the microtubule lattice's vulnerability to mechanical injury. Here, we develop an ultrastructural model of the axon's cytoskeletal architecture to identify the components involved in the dynamic load transfer during injury. Corroborative in vivo studies were performed using a gyrencephalic swine model of concussion via single and repetitive head rotational acceleration. Computational analysis of the load transfer mechanism demonstrates that the myelin sheath and the actin/spectrin cortex play a significant role in effectively shielding the microtubules from tissue stress. We derive failure maps in the space spanned by tissue stress and stress rate to identify physiological conditions in which the microtubule lattice can rupture. We establish that a softer axonal cortex leads to a higher susceptibility of the microtubules to failure. Immunohistochemical examination of tissue from the swine model of single and repetitive concussion confirms the presence of postinjury spectrin degradation, with more extensive pathology observed following repetitive injury. Because the degradation of myelin and spectrin occurs over weeks following the first injury, we show that softening of the myelin layer and axonal cortex exposes the microtubules to higher stress during repeated incidences of traumatic brain injuries. Our predictions explain how mechanical injury predisposes axons to exacerbated responses to repeated injuries, as observed in vitro and in vivo.
Mechanical characterization of spectrin at the molecular level

Abstract:Spectrin, a large cytoskeletal protein, consists of a heterodimeric structure comprising α and β subunits. Here, we have studied the mechanics of spectrin filament as a major constituent of dendrites and dendritic spines. Given the intricate biological details and compact biological construction of spectrin, we've developed a constitutive model of spectrin that describes its continuous deformation over three distinct stages and it's progressive failure mechanisms. Our model closely predicts both the force at which uncoiling begins and the ultimate force at which spectrin fails, measuring approximately 93 ~ 100 pN. Remarkably, our predicted failure force closely matches the findings from AFM experiments focused on the uncoiling of spectrin repeats, which reported a force of 90 pN. Our theoretical model proposes a plausible pathway for the potential failure of dendrites and the intricate connection between strain and strain rate. These findings deepen our understanding of how spectrin can contribute to traumatic brain injury risk analysis.

Abstract:Axonal injury represents a critical target area for the prevention and treatment of traumatic brain and spinal cord injuries. Finite element (FE) models of the head and/or brain are often used to predict brain injury caused by external mechanical loadings, such as explosive waves and direct impact. The accuracy of these numerical models depends on correctly determining the material properties and on the precise depiction of the tissues' microstructure (microscopic level). Moreover, since the axonal microstructure for specific regions of the brain white matter is locally oriented, the stress, and strain fields are highly anisotropic and axon orientation dependent. Additionally, mechanical strain has been identified as the proximal cause of axonal injury, which further demonstrates the importance of this multi-scale relationship. In this study, our previously developed FE and kinematic axonal models are coupled and applied to a pseudo 3-dimensional representative volume element of central nervous system white matter to investigate the multi-scale mechanical behavior. An inverse FE procedure was developed to identify material parameters of spinal cord white matter by combining the results of uniaxial testing with FE modeling. A satisfactory balance between simulation and experiment was achieved via optimization by minimizing the squared error between the simulated and experimental force-stretch curve. The combination of experimental testing and FE analysis provides a useful analysis tool for soft biological tissues in general, and specifically enables evaluations of the axonal response to tissue-level loading and subsequent predictions of axonal damage.
A transversely isotropic viscoelastic constitutive equation for brainstem undergoing finite deformation

Abstract:The objective of this study was to define the constitutive response of brainstem undergoing finite shear deformation. Brainstem was characterized as a transversely isotropic viscoelastic material and the material model was formulated for numerical implementation. Model parameters were fit to shear data obtained in porcine brainstem specimens undergoing finite shear deformation in three directions: parallel, perpendicular, and cross sectional to axonal fiber orientation and determined using a combined approach of finite element analysis (FEA) and a genetic algorithm (GA) optimizing method. The average initial shear modulus of brainstem matrix of 4-week old pigs was 12.7 Pa, and therefore the brainstem offers little resistance to large shear deformations in the parallel or perpendicular directions, due to the dominant contribution of the matrix in these directions. The fiber reinforcement stiffness was 121.2 Pa, indicating that brainstem is anisotropic and that axonal fibers have an important role in the cross-sectional direction. The first two leading relative shear relaxation moduli were 0.8973 and 0.0741, respectively, with corresponding characteristic times of 0.0047 and 1.4538 s, respectively, implying rapid relaxation of shear stresses. The developed material model and parameter estimation technique are likely to find broad applications in neural and orthopaedic tissues.

Abstract:Constitutive behaviors and material properties of brain tissue play an essential role in accurately modeling its mechanical responses. However, the measured mechanical behaviors of brain tissue exhibit a large variability, and the reported elastic modulus can differ by orders of magnitude. Here we develop the micromechanical models based on the actual microstructure of the longitudinally anisotropic plane of brain tissue to investigate the microstructural origins of the large variability. Specifically, axonal fiber bundles with the specified configurations are distributed in an equivalent matrix. All micromechanical models are subjected to multiple loading modes, such as tensile, compressive, and shear loading, under periodic boundary conditions. The predicted results agree well with the experimental results. Furthermore, we investigate how brain tissue elasticity varies with its microstructural features. It is revealed that the large variability in brain tissue elasticity stems from the volume fraction of axonal fiber, the aspect ratio of axonal fiber, and the distribution of axonal fiber orientation. The volume fraction has the greatest impact on the mechanical behaviors of brain tissue, followed by the distribution of axonal fiber orientation, then the aspect ratio. This study provides critical insights for understanding the microstructural origins of the large variability in brain tissue elasticity.

Viscoelastic modelling and analysis of two-dimensional woven CNT-based multiscale fibre reinforced composite material system

Abstract:Carbon nanotube (CNT) has fostered research as a promising nanomaterial for a variety of applications due to its exceptional mechanical, optical, and electrical characteristics. The present article proposes a novel and comprehensive micromechanical framework to assess the viscoelastic properties of a multiscale CNT-reinforced two-dimensional (2D) woven hybrid composite. It also focuses on demonstrating the utilisation of the proposed micromechanics in the dynamic analysis of shell structure. First, the detailed constructional attributes of the proposed trans-scale composite material system are described in detail. Then, according to the nature of the constructional feature, mathematical modelling of each constituent phase or building block's material properties is established to evaluate the homogenised viscoelastic properties of the proposed composite material system. To highlight the novelty of this study, the viscoelastic characteristics of the modified matrix are developed using the micromechanics method of Mori-Tanaka (MT) in combination with the weak viscoelastic interphase (WI) theory. In the entire micromechanical framework, the CNTs are considered to be randomly oriented. The strength of the material (SOM) approach is used to establish mathematical frameworks for the viscoelastic characteristics of yarns, whereas the unit cell method (UCM) is used to determine the viscoelastic properties of the representative unit cell (RUC). Different numerical results have been obtained by varying the CNT composition, interface conditions, agglomeration, carbon fibre volume percentage, excitation frequency, and temperature. The influences of geometrical parameters like yarn thickness, width, and the gap length to yarn width ratio on the viscoelasticity of such composite material systems are also explored. The current study also addresses the issue of resultant anisotropic viscoelastic properties due to the use of dissimilar yarn thickness. The results of this micromechanical analysis provide valuable insights into the viscoelastic properties of the proposed composite material system and suggest its potential applications in vibration damping. To demonstrate the application of developed novel micromechanics in vibration analysis, as one of the main contributions, comprehensive numerical experiments are conducted on a shell panel. The results show a significant reduction in vibration amplitudes compared to traditional composite materials in the frequency response and transient response analyses. To focus on the aspect of micromechanical behaviour on dynamic response and for the purpose of brevity, only linear strain displacement relationships are considered for dynamic analysis. These insights could inform future research and development in the field of composite materials.
Effect of Strain Rate on Single Tau, Dimerized Tau and Tau-Microtubule Interface: A Molecular Dynamics Simulation Study

Abstract:Microtubule-associated protein (MAP) tau is a cross-linking molecule that provides structural stability to axonal microtubules (MT). It is considered a potential biomarker for Alzheimer’s disease (AD), dementia, and other neurological disorders. It is also a signature protein for Traumatic Brain Injury (TBI) assessment. In the case of TBI, extreme dynamic mechanical energies can be felt by the axonal cytoskeletal members. As such, fundamental understandings of the responses of single tau protein, polymerized tau protein, and tau-microtubule interfaces under high-rate mechanical forces are important. This study attempts to determine the high-strain rate mechanical behavior of single tau, dimerized tau, and tau-MT interface using molecular dynamics (MD) simulation. The results show that a single tau protein is a highly stretchable soft polymer. During deformation, first, it significantly unfolds against van der Waals and electrostatic bonds. Then it stretches against strong covalent bonds. We found that tau acts as a viscoelastic material, and its stiffness increases with the strain rate. The unfolding stiffness can be ~50–500 MPa, while pure stretching stiffness can be >2 GPa. The dimerized tau model exhibits similar behavior under similar strain rates, and tau sliding from another tau is not observed until it is stretched to >7 times of original length, depending on the strain rate. The tau-MT interface simulations show that very high strain and strain rates are required to separate tau from MT suggesting Tau-MT bonding is stronger than MT subunit bonding between themselves. The dimerized tau-MT interface simulations suggest that tau-tau bonding is stronger than tau-MT bonding. In summary, this study focuses on the structural response of individual cytoskeletal components, namely microtubule (MT) and tau protein. Furthermore, we consider not only the individual response of a component, but also their interaction with each other (such as tau with tau or tau with MT). This study will eventually pave the way to build a bottom-up multiscale brain model and analyze TBI more comprehensively.

Embedded finite element modeling of the mechanics of brain axonal fiber tracts under head impact conditions

Abstract:Investigating and understanding the biomechanical kinematics and kinetics of human brain axonal fibers during head impact process is crucial to study the mechanisms of Traumatic Axonal Injury (TAI). Such a study may require the explicit incorporation of brain fiber tracts into the host brain in order to distinguish the mechanical states of axonal fibers and brain tissue. Herein we extend our previously developed human head model by using an embedded element method to include fiber tracts reconstructed from diffusion tensor images in a host brain with the purpose of numerically tracking the deformation state of axonal fiber tracts during a head impact simulation. The updated model is validated by comparing its prediction of intracranial pressures with experimental data, followed by a thorough study of the effects of element types used for fiber tracts and the stiffness ratios of fiber to host brain. The validated model is also used to predict and visualize the damaged region of fiber tracts during the head impact process based on different injury criteria. The model is promising in tracking the state of fiber tracts and can add more objective functions such as axonal fiber deformation if used in the future design optimization of head protective equipment such as a football helmet.
Revealing the nonlinear mechanical behavior of white matter brain tissue by analyzing the asynchronous deformation and damage of matrix and axonal fibers

Abstract:The mechanical behavior of the brain plays a vital role in regulating brain morphology and brain function. The experimental results show that the brain tissue behaves with strong nonlinearity, heterogeneity, loading rate and direction dependence, and complicated damage behavior. However, the underlying mechanism attributed to the interlacement of extremely soft matrix and complex axonal fibers network in white matter brain tissue is still not well understood. In this work, the nonlinear mechanical behavior of white matter brain tissue is analyzed by developing a mesoscale visco-hyperelastic constitutive model that considers isotropic matrix and anisotropic axonal fibers separately. Based on the diffusion tensor imaging data, the generalized structure tensor is introduced to reflect axonal fiber's orientation and distribution characteristics in different parts of white matter. Furthermore, the damage models of matrix and axonal fibers are established to characterize their damage evolution differences. In particular, a damage initiation criterion based on the equivalent strain of axonal fibers stretch is proposed by analogy with the Tsai-Hill criterion, according to the distribution characteristics of axonal fibers. Then the mesoscale constitutive model is applied to reveal the effects of matrix and axonal fibers on the deformation and damage of white matter. On the one hand, it is found that the "S-shaped" nonlinear stress curve of white matter in uniaxial tension is the superposition of the convex stress curve of matrix and the "J-shaped" stress curve of axonal fibers, and the nonlinearity is stronger with the increase of strain rate. On the other hand, with the change of the distribution of axonal fibers from uniaxially oriented to dispersed, the anisotropy of white matter caused by the stress reinforcement of axonal fibers gradually evolves from transverse isotropy to isotropy. With the increase of strain rate, the reinforcement effect of axonal fibers tends to be more isotropic, and the proportion of isotropic matrix stress is increased, resulting in the isotropic tendency of white matter. Moreover, the asynchrony of matrix and axonal fibers damage also results in the complex transition of white matter deformation behavior. Along with the increase of deformation, the anisotropic mechanical behavior of white matter rapidly degenerates to isotropy with the damage of axonal fibers. Overall, the strong coupling of nonlinear, anisotropic, and strain rate-dependent mechanical behaviors of white matter is well explained by combining the asynchronous deformation and damage of matrix and axonal fibers in this study. It will help understand brain injury, brain growth and development, physiological brain diseases, and other relevant problems.

Abstract:BackgroundMechanical properties of the brain tissue are crucial to understand the mechanisms of traumatic brain injury (TBI). Injured brain tissue could induce changes of mechanical properties and anatomical structures. However, limited data is available for the injured tissue.New MethodWe developed a custom-built device to introduce controlled cortical impact (CCI) to brain with controlled impact velocity and direction. A study protocol for measuring the viscoelastic properties of injured brain tissue was also developed. Micro-scale morphological changes of the vasculature were quantified by analyzing confocal images of the brain tissue using CLARITY method.ResultsResults showed significant differences of the instantaneous shear modulus of the impact region from different impact angles. However, no significant differences were found for long-term shear modulus by varying the impact angles and velocities. Analysis of the vasculature showed an increased radius of the vessels in the injured tissue compared with that in the control group.Comparison with existing methodsA combination of three different impact velocities and three different impact angles were adopted for producing injury to the brain. In addition, viscoelastic properties were compared between the injured and non-injured regions. The corresponding morphological changes of the vasculature system were also investigated.ConclusionsThe instantaneous shear modulus at the impact region was significantly different for the three impact angles. Compared to that of the control group, increased radius of the vasculature was also observed in the injured brain tissue. Results indicated that the biomechanical and structural changes of the injured tissue were closely related to the impact angles and velocities. Viscoelastic measurements could also help validation of computational models.

Mechanical Responses of a Single Myelin Layer: A Molecular Simulation Study

Abstract:The myelin sheath provides insulation to the brain's neuron cells, which aids in signal transmission and communication with the body. Degenerated myelin hampers the connection between the glial cells, which are the front row responders during traumatic brain injury mitigation. Thus, the structural integrity of the myelin layer is critical for protecting the brain tissue from traumatic injury. At the molecular level, myelin consists of a lipid bilayer, myelin basic proteins (MBP), proteolipid proteins (PLP), water and ions. Structurally, the myelin sheath is formed by repeatedly wrapping forty or more myelin layers around an axon. Here, we have used molecular dynamic simulations to model and capture the tensile response of a single myelin layer. An openly available molecular dynamic solver, LAMMPS, was used to conduct the simulations. The interatomic potentials for the interacting atoms and molecules were defined using CHARMM force fields. Following a standard equilibration process, the molecular model was stretched uniaxially at a deformation rate of 5 Å/ps. We observed that, at around 10% applied strain, the myelin started to cohesively fail via flaw formation inside the bilayers. Further stretching led to a continued expansion of the defect inside the bilayer, both radially and transversely. This study provides the cellular-level mechanisms of myelin damage due to mechanical load.
Osmotically driven beading instability in axons: Continuum theory, perturbation analysis and finite element implementation

Abstract:Axonal beading or formation of multiple beads along an axon is characteristic of many brain pathological states like Alzheimer's, Parkinson's and traumatic injuries. Despite the many existing experimental studies, the underlying mechanisms of this shape instability remain still poorly understood. In this paper, we establish a combined theoretical and numerical framework to study the governing key factors of this morphological transformation. We develop a three-dimensional (3D) non-equilibrium large deformation thermodynamic model with two main parts: the central axoplasm which is considered as a polyelectrolyte hydrogel and the encapsulating cortical membrane which is modeled as an incompressible hyperelastic layer with surface energy and growing surface. The model constitutive and evolution equations are then extracted employing thermodynamic balance principles for both bulk and surface material points. It is shown that the second law of thermodynamics indicates that the axolemma growth rate is proportional to the membrane tension which is in perfect agreement with the available experimental findings. While the developed model is general and can be extended to cover other types of axonal beadings, for the sake of simplicity, here, we focus on osmotically driven axisymmetric beadings which are compressible viscoelastic periodic modulations. We solve the corresponding governing equations using the linear perturbation method. This perturbation analysis proves that: 1) the beading instability is a rate dependent phenomenon that is controlled by the axolemma growth, 2) the initially dominant beading waves (the fastest waves) might be replaced only by longer waves which are more stable and 3) the wavelength of the fastest beads should vary roughly linearly with the axonal radius. These main findings are all in good agreement with the existing experimental results. Finally, the finite element implementation of the model is also presented to verify the results of the linear stability analysis for slow waves. The obtained axisymmetric finite element results are in good agreement with the corresponding theoretical findings.

Abstract:In this study, we report a computational investigation on how the mechanochemical characteristics of crosslinking molecules influence the viscoelasticity of three dimensional F-actin networks, an issue of key interest in analyzing the behavior of living cells and biological gels. In particular, it was found that the continuous breakage and rebinding of cross-linkers result in a locally peaked loss modulus in the rheology spectrum of the network, reflecting the fact that maximum energy dissipation is achieved when the driving frequency of the applied oscillating shear becomes comparable to the dissociation/association rate of crosslinking molecules. In addition, we showed that when subjected to constant rate of shear, an actin network can exhibit either strain hardening or softening depending on the ratio between the loading rate and unbinding speed of cross-linkers. A criterion for predicting the transition from softening to hardening was also obtained, in agreement with recent experiments. Finally, significant structural evolution was found to occur in random networks undergoing mechanical "training" (i.e. under a constant applied shear stress over a period of time), eventually leading to a pronounced anisotropic response of the network afterward which again is consistent with experimental observations.
