A quantitative model to predict the effect of magnesium stearate on punch sticking

Abstract:A new model has been developed to predict the tablet sticking propensity of lubricated direct compression blends based on the active pharmaceutical ingredient (API), lubricant, blend particle properties (size and surface area), and compact tensile strength. A variety of blends containing different APIs, common diluents, and magnesium stearate were prepared for this study to examine the impact of their physical properties on sticking propensity. From this dataset, a Modified Sticking Index model was built to further incorporate the lubricant qualities to the previously reported Sticking Index (SI) [1]. Results showed that incorporating the mass fraction of the lubricant into the SI resulted in a highly predictive correlation with the measured adhered mass of each blend in a standard sticking test. In practice, formulators can use this new model in a material sparing paradigm to aid in formulation development.
