Abstract:Membrane fusion is essential for many cellular physiological functions, which is modulated by highly precise molecular mechanism involving multiple energy barriers. Nanoparticles (NPs), which exhibit immense potential in the field of biomedical applications, can act as fusogen proteins to initiate and regulate membrane fusion. However, the underlying mechanisms of NP-induced membrane fusion and the molecular details involved remain largely elusive. Here, using coarse-grained molecular dynamics simulations, we systematically investigate the NP-induced membrane fusion behaviors and the influences of NP properties (size, hydrophobicity and hydrophilicity). Our results show that the vesicle-bilayer fusion induced by a hydrophobic NP is an intricately state-wise process, involving the approach and local deformation of the vesicle and bilayer bridging by the NP, the flip-flop of lipids from proximal leaflets and the formation of a fusion stalk, as well as further lipid interactions between distal leaflets and complete fusion. Moreover, we find that NP properties have distinct effects on membrane fusion and thus the optimal NP conditions for facilitating membrane fusion are obtained. Our work provides a mechanistic understanding of NP-induced membrane fusion and offers useful insights for efficient and controlled regulation of membrane fusion.

Abstract:Membrane fusion is a ubiquitous phenomenon linked to many biological processes, and represents a crucial step in liposome-based drug delivery strategies. The ability to control, ever more precisely, membrane fusion pathways would thus be highly valuable for next generation nano-medical solutions and, more generally, the design of advanced biomimetic systems such as synthetic cells. In this article, we present fusogenic nanostructures constructed from synthetic DNA which, different from previous solutions, unlock routes for modulating the rate of fusion and making it conditional to the presence of soluble DNA molecules, thus demonstrating how membrane fusion can be controlled through simple DNA-based molecular circuits. We then systematically explore the relationship between lipid-membrane composition, its biophysical properties, and measured fusion efficiency, linking our observations to the stability of transition states in the fusion pathway. Finally, we observe that specific lipid compositions lead to the emergence of complex bilayer architectures in the fusion products, such as nested morphologies, which are accompanied by alterations in biophysical behaviour. Our findings provide multiple, orthogonal strategies to program lipid-membrane fusion, which leverage the design of either the fusogenic DNA constructs or the physico/chemical properties of the membranes, and could thus be valuable in applications where some design parameters are constrained by other factors such as material cost and biocompatibility, as it is often the case in biotechnological applications.

Abstract:Phospholipid-nanoparticle constructs, formed by allowing nanoparticles to adsorb to the outer leaflet of liposomes, are found to be stabilized against fusion with one another. Here, through single-particle tracking by epifluorescence microscopy, we explore their use as novel colloidal particles-flexible and hollow colloidal particles that contrast strikingly with colloids of the conventional type. At the single-liposome level, the distribution of diffusion coefficients is quantified. Biomolecular function is addressed through experiments in which we explore the access of receptor to liposome-immobilized ligand, finding that receptor binding persists over a range of nanoparticle surface coverage where liposome fusion and large-scale aggregation is prevented. This opens the door to designing newer and more flexible types of tailor-made materials with desirable functionality.

Abstract:The fusion between two lipid membranes is a ubiquitous mechanism in cell traffic and pathogens invasion. Yet it is not well understood how two distinct bilayers overcome the energy barriers towards fusion and reorganize themselves to form a unique continuous bilayer. The magnitudes and numbers of these energy barriers are themselves an open question. To tackle these issues, we developed a new tool that allows to control the forces applied between two supported lipid bilayers (SLBs) deposited on superparamagnetic beads. By applying a magnetic field, the beads self-organize along field lines in chains of beads and compress the two membranes on the contact zone. Using the diffusion of fluorescently labelled lipids from one bilayer to the other allows us to identify fusion of the bilayers in contact. We applied increasing forces on SLBs and increased the occurrence of fusion. This experimental system allows the simultaneous study of tens of facing bilayers in a single experiment and mitigates the stochasticity of the fusion process. It is thus a powerful tool to test the various parameters involved in the membrane fusion process.
Nanopore and nanoparticle formation with lipids, undergoing polymorphic phase transitions

Abstract:We describe several unexpected phenomena, caused by a solid-solid phase transition (gel-to-crystal) typical for all main classes of lipid substances - phospholipids, triglycerides, diglycerides, alkanes, etc. We discovered that this transition leads to spontaneous formation of a network of nanopores, spreading across the entire lipid structure. These nanopores are spontaneously impregnated (flooded) by water when appropriate surfactants are present, thus fracturing the lipid structure at a nano-scale. As a result, spontaneous disintegration of the lipid into nanoparticles or formation of double emulsions is observed, just by cooling and heating of an initial coarse lipid-in-water dispersion around the lipid melting temperature. The process of nanoparticle formation is effective even after incorporation of medical drugs of high load, up to 50 % in the lipid phase. The role of the main governing factors is clarified, the procedure is optimized, and the possibility for its scaling-up to industrially relevant amounts is demonstrated.

Abstract:The initial, nanometer-sized connection between the plasma membrane and a hormone- or neurotransmitter-filled vesicle -the fusion pore- can flicker open and closed repeatedly before dilating or resealing irreversibly. Pore dynamics determine release and vesicle recycling kinetics, but pore properties are poorly known because biochemically defined single-pore assays are lacking. We isolated single flickering pores connecting v-SNARE-reconstituted nanodiscs to cells ectopically expressing cognate, "flipped" t-SNAREs. Conductance through single, voltage-clamped fusion pores directly reported sub-millisecond pore dynamics. Pore currents fluctuated, transiently returned to baseline multiple times, and disappeared ~6 s after initial opening, as if the fusion pore fluctuated in size, flickered, and resealed. We found that interactions between v- and t-SNARE transmembrane domains (TMDs) promote, but are not essential for pore nucleation. Surprisingly, TMD modifications designed to disrupt v- and t-SNARE TMD zippering prolonged pore lifetimes dramatically. We propose that the post-fusion geometry of the proteins contribute to pore stability.
Nanoparticles Binding to Lipid Membranes: from Vesicle-Based Gels to Vesicle Inversion and Destruction

Abstract:Cells offer numerous inspiring examples where proteins and membranes combine to form complex structures that are key to intracellular compartmentalization, cargo transport, and specialization of cell morphology. Despite this wealth of examples, we still lack the design principles to control membrane morphology in synthetic systems. Here we show that even the relatively simple case of spherical nanoparticles binding to lipid-bilayer membrane vesicles results in a remarkably rich set of morphologies that can be controlled quantitatively via the particle binding energy. We find that when the binding energy is weak relative to a characteristic membrane-bending energy, the vesicles adhere to one another and form a soft solid, which could be used as a useful platform for controlled release. When the binding energy is larger, the vesicles undergo a remarkable destruction process consisting first of invaginated tubules, followed by vesicles turning inside-out, yielding a network of nanoparticle-membrane tubules. We propose that the crossover from one behavior to the other is triggered by the transition from partial to complete wrapping of nanoparticles. This model is confirmed by computer simulations and by quantitative estimates of the binding energy. These findings open the door to a new class of vesicle-based, closed-cell gels that are more than 99% water and can encapsulate and release on demand. Our results also show how to intentionally drive dramatic shape changes in vesicles as a step toward shape-responsive particles. Finally, they help us to unify the wide range of previously observed responses of vesicles and cells to added nanoparticles.

Abstract:Significance The plasma membrane protects cell content from the outer environment. Overcoming this obstacle and delivering drugs efficiently into cells still remain a major hurdle for modern pharmacology and medicine. We developed and demonstrated a simple nanomaterial platform—a dimer of small-diameter carbon nanotube porins (CNTPs)—which functions as a potent membrane fusogen. Molecular simulations revealed a distinct fusion mechanism. CNTP-studded vesicles loaded with a chemotherapeutic agent, doxorubicin, delivered the drug to cancer cells, killing a majority of them. Our work provides new opportunities for understanding membrane fusion mechanisms, designing synthetic fusogens, and developing simple and efficient drug carriers for delivery of therapeutics and vaccines.

Abstract:Abstract Introduction of microfluidic mixing technique opens a new door for preparation of the liposomes and lipid-based nanoparticles by on-chip technologies that are applicable in a laboratory and industrial scale. This study demonstrates the role of phospholipid bilayer fragment as the key intermediate in the mechanism of liposome formation by microfluidic mixing in the channel with “herring-bone” geometry used with the instrument NanoAssemblr. The fluidity of the lipid bilayer expressed as fluorescence anisotropy of the probe N,N,N-Trimethyl-4-(6-phenyl-1,3,5-hexatrien-1-yl) was found to be the basic parameter affecting the final size of formed liposomes prepared by microfluidic mixing of an ethanol solution of lipids and water phase. Both saturated and unsaturated lipids together with various content of cholesterol were used for liposome preparation and it was demonstrated, that an increase in fluidity results in a decrease of liposome size as analyzed by DLS. Gadolinium chelating lipids were used to visualize the fine structure of liposomes and bilayer fragments by CryoTEM. Experimental data and theoretical calculations are in good accordance with the theory of lipid disc micelle vesiculation.

Abstract:Membrane fusion, a fundamental biological process of the fusion of the membrane composition between cells, is vital for cell-cell communication and cargo transport between living cells. This fusion interaction achieves the transportation of the inner content to the cellular cytosol as well as the simultaneous blending of foreign substances with the cell membrane. Inspired by this biological process, emerging membrane-fusogenic particles have been developed, opening a new area for bioengineering and biomedical applications. Especially, membrane-fusion-mediated transfer of inner cargoes can bypass endosomal entrapment to maximize the transportation efficiency, emerging as a unique cytoplasmic delivery platform distinct from those depending on conventional endocytosis-based pathways. In addition, the membrane fusion enables cell surface modification through lipid diffusion and mixing, providing a tool for direct cell membrane engineering. In this review, we focus on the development of membrane-fusogenic particles and their up-to-date progress. We briefly introduce the concept of membrane fusion, elaborate inspiring strategies of membrane-fusogenic particles, and highlight the recent advances and the promising applications of membrane-fusogenic particles as a next-generation bioengineering tool. In the end, we conclude with the present challenges and opportunities, providing insights in the future research of membrane-fusogenic particles.
Spontaneous Fusion of Core‐Shell Nanocapsules with Oil Cores and Oppositely Charged Polysaccharide Shells

Abstract:Oppositely charged nanocapsules composed of chitosan‐based and hyaluronate‐based shells and oil cores are shown to spontaneously merge at optimized conditions serving as a model for fused nanoreactors. Efficient energy transfer processes between the encapsulated dyes as well as a model fluorogenic reaction are shown to take place in such confined hydrophobic environments of water‐dispersable nanocapsules only after their fusion. Polymer nanocapsules with hydrophobic cores are promising candidates for nanoreactors to carry out (bio)chemical reactions mimicking the performance of natural cellular systems. Their architecture allows reagents to be encapsulated in the cores enabling reactions to proceed in confined environments in a controlled, and efficient manner. Polysaccharide‐shell oil‐core nanocapsules are proposed here as facile mergeable nanoreactors. Spontaneous fusion of oppositely charged polysaccharide capsules is demonstrated for the first time. Such capsules are formed and easily loaded with reagents by nanoemulsification of an aqueous solution of hydrophobically modified polysaccharides (chitosan, hyaluronate) and oleic acid with dissolved desired hydrophobic compounds. Efficient fusion of the formed nanocapsules dispersed in an aqueous medium at optimized conditions (pH, ionic strength) is followed using fluorescence microscopy by labeling both their cores and shells with fluorescent dyes. As a proof of concept, a model fluorogenic synthesis is also realized by fusing the capsules containing separated reagents and the catalyst. The nanocapsules and fusion process developed here establish a platform for realization of versatile reactions in a confined environment including model studies on biologically relevant processes taking place in natural systems.

High Efficiency Liposome Fusion Induced by Reducing Undesired Membrane Peptides Interaction

Abstract:A full membrane fusion model which attains both complete lipid mixing and content mixing liposomal membranes mediated by coiled-coil forming lipopeptides LPK [L-PEG(12)-(KIAALKE)(3)] and LPE [L-PEG(12)-(EIAALEK)(3)] is presented. The electrostatic effects of lipid anchored peptides on fusion efficiency was investigated. For this, the original amino acid sequence of the membrane bound LPK was varied at its 'f'-position of the helical structure, i.e. via mutating the anionic glutamate residues by either neutral serines or cationic lysines. Both CD and fluorescence measurements showed that replacing the negatively charged glutamate did not significantly alter the peptide ability to form a coiled coil, but lipid mixing and content mixing assays showed more efficient liposome-liposome fusion resulting in almost quantitative content mixing for the lysine mutated analogue (LPKK) in conjunction with LPE. A mechanism is proposed for a fusion model triggered by membrane destabilizing effects mediated by the membrane destabilizing activety of LPK in cooperation with the electrostatic activity of LPE. This new insight may enlightens the further development of a promising nano carrier tool for biomedical applications.

Abstract:Liposomes are synthetic phospholipid vesicles containing an aqueous lumen confined by a lipid bilayer. These vesicles have been used as nanoreactors because they offer confined environments that can result in significant changes to chemistry. However, a major limitation of using liposomes as nanocontainers is the impermeability of their lipid membranes. To overcome this, scientists have tested the use of porous liposomes generated by membrane phase changes or by pore formation proteins. In this study, the selective permeability of porous liposomes to molecules smaller than the threshold pore size is demonstrated for triggering internal reactions. Furthermore, in order to deliver macromolecules larger than the threshold pore size into lipid-based nanoreactors, fusogenic soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) proteins were employed to mediate membrane fusion between two liposomes to initiate reactions on demand without losing previously encapsulated macromolecules. This novel approach circumvents limitations associated with inserting molecules larger than the threshold pore size of porous lipid membranes.
On fusogenicity of positively charged phased-separated lipid vesicles: experiments and computational simulations

Abstract:This paper studies the fusogenicity of cationic liposomes in relation to their surface distribution of cationic lipids and utilizes membrane phase separation to control this surface distribution. It is found that concentrating the cationic lipids into small surface patches on liposomes, through phase-separation, can enhance liposome's fusogenicity. Further concentrating these lipids into smaller patches on the surface of liposomes led to an increased level of fusogenicity. These experimental findings are supported by numerical simulations using a mathematical model for phase-separated charged liposomes. Findings of this study may be used for design and development of highly fusogenic liposomes with minimal level of toxicity.

Abstract:To study membrane fusion mediated by synaptic proteins, proteoliposomes have been widely used for in vitro ensemble measurements with limited insights into the fusion mechanism. Single-particle techniques have proven to be powerful in overcoming the limitations of traditional ensemble methods. Here, we summarize current single-particle methods in biophysical and biochemical studies of fusion mediated by soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) and other synaptic proteins, together with their advantages and limitations.
Examination Of Membrane Fusion By Dissipative Particle Dynamics Simulation And Comparison With Continuum Elastic Models

Abstract:A dissipative particle dynamics model is applied to probe the lipidic membrane fusion. This model is verified by reproducing the lipid phase behavior. The classical stalk model has been visited and modified. The tilt deformation of the lipids and the noncircular shape of the stalk are supported. The stalk is shown to undergo asymmetric expansion to form the trans-monolayers contact (TMC). Unlike previous models, an energy barrier between the stalk and the TMC has been identified, implying that the TMC should be a metastable formation. This shows good agreement with the fusion experiments. Two typical elastic continuum models are compared with our result and possible modifications to the two elastic models are suggested. The effect of spontaneous curvature of lipid on selection of fusion pathway is also examined. It is observed that a bent stalk with pore or an inverted micellar intermediate will have more chance to occur than traditional stalk when the spontaneous curvature of the lipid becomes more negative. An antifouling membrane-fusogenic liposome for effective intracellular delivery in vivo

Abstract:The membrane-fusion-based internalization without lysosomal entrapment is advantageous for intracellular delivery over endocytosis. However, protein corona formed on the membrane-fusogenic liposome surface converts its membrane-fusion performance to lysosome-dependent endocytosis, causing poorer delivery efficiency in biological conditions. Herein, we develop an antifouling membrane-fusogenic liposome for effective intracellular delivery in vivo. Leveraging specific lipid composition at an optimized ratio, such antifouling membrane-fusogenic liposome facilitates fusion capacity even in protein-rich conditions, attributed to the copious zwitterionic phosphorylcholine groups for protein-adsorption resistance. Consequently, the antifouling membrane-fusogenic liposome demonstrates robust membrane-fusion-mediated delivery in the medium with up to 38% fetal bovine serum, outclassing two traditional membrane-fusogenic liposomes effective at 4% and 6% concentrations. When injected into mice, antifouling membrane-fusogenic liposomes can keep their membrane-fusion-transportation behaviors, thereby achieving efficient luciferase transfection and enhancing gene-editing-mediated viral inhibition. This study provides a promising tool for effective intracellular delivery under complex physiological environments, enlightening future nanomedicine design.

Abstract:Soft-matter nanoparticles are of great interest for their applications in biotechnology, therapeutic delivery, and in vivo imaging. Underpinning this is their biocompatibility, potential for selective targeting, attractive pharmacokinetic properties, and amenability to downstream functionalisation. Morphological diversity inherent to soft-matter particles can give rise to enhanced functionality. However, this diversity remains untapped in clinical and industrial settings, and only the simplest of particle architectures [spherical lipid vesicles and lipid/polymer nanoparticles (LNPs)] have been routinely exploited. This is partially due to a lack of appropriate methods for their synthesis. To address this, we have designed a scalable microfluidic hydrodynamic focusing (MHF) technology for the controllable, rapid, and continuous production of lyotropic liquid crystalline (LLC) nanoparticles (both cubosomes and hexosomes), colloidal dispersions of higher-order lipid assemblies with intricate internal structures of 3-D and 2-D symmetry. These particles have been proposed as the next generation of soft-matter nano-carriers, with unique fusogenic and physical properties. Crucially, unlike alternative approaches, our microfluidic method gives control over LLC size, a feature we go on to exploit in a fusogenic study with model cell membranes, where a dependency of fusion on particle diameter is evident. We believe our platform has the potential to serve as a tool for future studies involving non-lamellar soft nanoparticles, and anticipate it allowing for the rapid prototyping of LLC particles of diverse functionality, paving the way toward their eventual wide uptake at an industrial level.
