Abstract:Background Patients with resected oral squamous cell carcinoma (OSCC) harboring metastatic lymph nodes without extranodal extension (pN^+ENE^-) exhibit substantial variations in treatment and outcomes. Information from next-generation sequencing (NGS) might improve risk stratifications and therapeutic strategies. Methods The hotspot-mutation regions of 45 cancer-related genes were investigated by NGS using an ultra-deep (> 1,000×) sequencing approach on formalin-fixed paraffin-embedded samples obtained from 144 patients with resected OSCC with pN^+ENE^-. Results Pathologic T4 (pT4) was the most important traditional prognosticator for disease-specific survival (DSS) (hazard ratio [HR]: 3.33; 95% confidence interval [CI]: 1.81-6.12; P < 0.001).
