Abstract:Acupuncture has a long history and definite effectiveness in traditional Chinese medicine. Manual acupuncture and electroacupuncture stimulate specific acupoints on meridians to obtain local and/or systemic biological effects, but the mechanism behind these effects remains unclear. In the present study, we used three-dimensional tissue-clearing technology to study four specific acupoints on the Ren meridian of mice, which is located within the anatomic landmark of the linea alba. The distribution, density, branching, and relationships between blood vessels and nerves were quantitated. The acupoints and meridian before and after electroacupuncture were visualized and analyzed together with the surrounding structures using topological Mapper methods. We found that capillary branching and sympathetic neurovascular interactions (SNIs) were denser in the CV 4 acupoint (Guanyuan) compared with surrounding non-acupoints. Furthermore, SNIs in CV 4 indicated capabilities in topological amplification, which was confirmed by high resolution in vivo dynamic vascular imaging. After electro or manual acupuncture, CV 4 showed different degrees of increased blood flow detected with NIR-II probe of LZ-1105, compared with little increased in non-acupoints.
