The halogen effect of bis-truxene substituted BODIPY photosensitizers for potential photodynamic therapy

Abstract:Photodynamic therapy (PDT) is important for cancer treatment and has attracted great attention in recent years. Herein, three bis-truxene substituted BODIPY dyes, BDP-2I4H , BDP-2I4Br , and BDP-2I4I , have been designed and synthesized with two same iodo atoms and four different hydrogen, bromo, and iodo tails to explore the halogen effect for potential PDT. UV–Vis spectra show three dye-decorated nanoparticles exhibit normalized absorption bands at approximately 700 nm and the near-edge absorption bands reach 800 nm. TEM images and DLS profiles reveal the sphere-like morphology and the average hydrodynamic diameters of 143, 136, and 205 nm, respectively, for BDP-2I4H NPs , BDP-2I4Br NPs , and BDP-2I4I NPs . Furthermore, the photothermal effect of BDP-2I4H NPs , BDP-2I4Br NPs , and BDP-2I4I NPs in solution is increasingly apparent with the enhancement of radiation time from 0 to 600 s and power from 0.1 to 0.7 W/cm 2 . MTT experiment demonstrates a clear concentration-dependent 4T1 cell death of BDP-2I4H NPs , BDP-2I4Br NPs , and BDP-2I4I NPs under laser irradiation. These research results indicate that the photothermal synergistic sustainable PDT approach has significant potential for biomaterial applications.
