Abstract:Lipopolysaccharides (LPSs) are major components of the outer membranes of Gram-negative bacteria. In this work, the structure of the O-polysaccharide of Ochrobactrum quorumnocens T1Kr02 was identified by nuclear magnetic resonance (NMR), and the physical–chemical properties and biological activity of LPS were also investigated. The NMR analysis showed that the O-polysaccharide has the following structure: →2)-β-d-Fucf-(1→3)-β-d-Fucp-(1→. The structure of the periplasmic glucan coextracted with LPS was established by NMR spectroscopy and chemical methods: →2)-β-d-Glcp-(1→. Non-stoichiometric modifications were identified in both polysaccharides: 50% of d-fucofuranose residues at position 3 were O-acetylated, and 15% of d-Glcp residues at position 6 were linked with succinate. This is the first report of a polysaccharide containing both d-fucopyranose and d-fucofuranose residues. The fatty acid analysis of the LPS showed the prevalence of 3-hydroxytetradecanoic, hexadecenoic, octadecenoic, lactobacillic, and 27-hydroxyoctacosanoic acids. The dynamic light scattering demonstrated that LPS (in an aqueous solution) formed supramolecular particles with a size of 72.2 nm and a zeta-potential of –21.5 mV. The LPS solution (10 mkg/mL) promoted the growth of potato microplants under in vitro conditions. Thus, LPS of O. quorumnocens T1Kr02 can be recommended as a promoter for plants and as a source of biotechnological production of d-fucose.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to determine the structure, physicochemical properties and biological activities of lipopolysaccharide (LPS) isolated from the potato rhizosphere bacterium *Ochrobactrum quorumnocens* T1Kr02. Specifically, the researchers analyzed the structure of the O - polysaccharide part of the LPS of this strain by nuclear magnetic resonance (NMR), and studied its physicochemical properties and biological activities. In addition, the formation of supramolecular particles of LPS in aqueous solution and its influence on the growth of potato micro - plants were also explored. ### Research Background Lipopolysaccharide (LPS) is a major component of the outer membrane of Gram - negative bacteria and has important biological functions. The LPS structures of different bacteria vary, and these differences can be used for the rapid identification of bacteria, especially in distinguishing pathogenic *Brucella* and saprophytic *Ochrobactrum*. In addition, the structure and properties of LPS are crucial for understanding its interaction with environmental objects and its potential application value. ### Research Objectives 1. **Determine the structure of LPS**: Use NMR technology to analyze the detailed structure of the O - polysaccharide part of the LPS of the *Ochrobactrum quorumnocens* T1Kr02 strain. 2. **Analyze the physicochemical properties of LPS**: Including the formation of supramolecular particles of LPS in aqueous solution, such as particle size and ζ - potential. 3. **Evaluate the biological activities of LPS**: Especially the effect on promoting the growth of potato micro - plants. ### Main Findings 1. **Structure of O - polysaccharide**: - The repeating unit structure of the O - polysaccharide part is: →2)-β - D - fructofuranose-(1→3)-β - D - fructopyranose-(1→. - Some D - fructofuranose residues are O - acetylated at the 3 - position (about 50%), and some D - glucose residues are linked to succinic acid at the 6 - position (about 15%). - This is the first report on the polysaccharide structure containing D - fructofuranose and D - fructopyranose residues. 2. **Physicochemical properties**: - The results of dynamic light scattering show that LPS forms supramolecular particles with a diameter of 72.2 ± 3.6 nm and a ζ - potential of –21.5 ± 0.7 mV in aqueous solution. - Electrophoresis analysis shows that LPS is mainly composed of high - molecular - weight molecules and does not show the typical ladder - like band pattern of Enterobacteriaceae LPS. 3. **Biological activities**: - A 10 μg/mL LPS solution can promote the growth of potato micro - plants. ### Conclusion This study not only provides the detailed structure and physicochemical properties of the LPS of the *Ochrobactrum quorumnocens* T1Kr02 strain, but also shows its potential application in promoting plant growth. These findings provide a theoretical basis for the future development of LPS - based biotechnology products.
