Abstract:NADH and NADPH are ubiquitous coenzymes in all living cells that play vital roles in numerous redox reactions in cellular energy metabolism. To accurately detect the distribution and dynamic changes of NAD(P)H under physiological conditions is essential for understanding their biological functions and pathological roles. In this work, we developed a near-infrared (NIR)-emission fluorescent small-molecule probe (DCI-MQ) composed of a dicyanoisophorone chromophore conjugated to a quinolinium moiety for in vivo NAD(P)H detection. DCI-MQ has the advantages of high water solubility, a rapid response, extraordinary selectivity, great sensitivity (a detection limit of 12 nM), low cytotoxicity, and NIR emission (660 nm) in response to NAD(P)H. Moreover, the probe DCI-MQ was successfully applied for the detection and imaging of endogenous NAD(P)H in both living cells and tumor-bearing mice, which provides an effective tool for the study of NAD(P)H-related physiological and pathological processes.

Abstract:Nicotinamide adenine dinucleotide (phosphate) (NAD(P)H/NAD(P)+) is crucial for oxidation-reduction equi-librium in all living cells, and disturbance of NAD(P)H/NAD(P)+ indicates many diseases. To accurately detect and track NAD(P)H in vitro and in vivo, we have developed a small organic dye (Rh-QL) based on intramolecular charge transfer (ICT) mechanism for multimodally detecting NAD(P)H by visible and near-infrared (Vis-NIR) dual-channel lighting-up fluorescence and by near-infrared spectrophotometry. This quinoline-based Indicator showed high selectivity and sensitivity toward NAD(P)H with a large redshift of absorption and dual-channel emissions at 580 nm and 750 nm, spatiotemporally and synchronously. For liquid samples, Rh-QL succeeded to screen NAD(P)H-related G6PD enzymatic activity by responsive absorption at 710 nm. In living cells, Rh-QL exhibited a good bio-compatibility and remarkable mitochondrial-targeted capacity to visualize intracellular endogenous and exogenous NAD(P)H variation utilizing Vis-NIR dual-channel fluorescence. In vivo, the near -infrared fluorescence was applied to differentiate tumor-bearing mice by NAD(P)H imaging. Moreover, "glycolysis" in both cancer cells and tumors of mice were traced to produce extra NAD(P)H using Rh-QL. As the first Vis-NIR dual-channel lighting-up probe to multimodally detect and track NAD(P)H in vitro and in vivo, it is appealing for high efficiency in understanding NAD(P)H-related physiological and pathological processes of disease diagnosis and therapy.

Abstract:The development of fluorescent probes for nitroreductase (NTR) has received intense attention because of its biological significance and wide application. In this work, a novel fluorescent probe for the detection of NTR in aqueous solution was designed and synthesized on a 1,8-naphthalimide scaffold. In the presence of NTR and nicotinamide adenine dinucleotide (NADH) under physiological conditions, the probe was converted into a 4-hydroxy-1,8-naphthalimide derivative and exhibited a sharp fluorescence enhancement at 550 nm, with a high selectivity for NTR over various analytes. The detection limit for NTR was determined to be 9.8 ng/ml by this probe. Due to its low signal background, this probe showed > 70-fold fluorescence enhancement. Theoretical calculations revealed that the reason for the fluorescence quenching of this probe is the photoinduced electron transfer (PET) from both the nitrobenzene and morpholine groups to the naphthalimide fluorophore.

Abstract:The reduced coenzymes (NADH and NADPH) are an important product in energy metabolism and closely related to the occurrence and development of cancer. So it is necessary to use a powerful detection tool to visualize NAD(P)H in energy metabolism of tumor cells and find a new strategy to improve cancer treatment based on NAD(P)H. Herein, a novel multifunctional probe (Cy-N) is synthesized with good near-infrared fluorescence (NIRF) response to NAD(P)H and the photoacoustic (PA) and photothermal properties are successfully activated by NAD(P)H. The probe is successfully applied in visualizing NAD(P)H in energy metabolism of tumor cells and imaging NAD(P)H in bacteria. Moreover, the probe can be used to image NAD(P)H in energy metabolism of tumor-bearing mice by dual-modal imaging (NIRF and PA). More importantly, in terms of the role of NAD(P)H in energy metabolism, the photothermal therapy (PTT) is activated by NAD(P)H and a novel strategy of enhanced PTT is proposed by injecting glucose. As far as we know, this is the first probe to detect NAD(P)H in energy metabolism through dual-modal imaging, and also the first probe to activate PTT based on NAD(P)H, which will provide important information of the diagnosis and treatment of cancer.

Abstract:A new ratiometric fluorescent probe for hydrazine based on a 4-hydroxynaphthalimide fluorophore was developed. The selective deprotection of the acetate group of the probe by hydrazine led to a prominent fluorescence ratiometric change, as well as a remarkable color change which could be distinguished by naked-eye. The reaction between the probe and hydrazine completed in 5min under the analytical system, and displayed a high selectivity and sensitivity for hydrazine against other primary amines and environmental and/or biological abundant ions. Moreover, the probe could be applied for hydrazine detection both in water samples and in vivo.

Abstract:A new "turn-on" fluorescent probe, composed of a protected phenol group with a p-nitrobenzyl moiety that functions as a latent donor and conjugated with two benzo[f] indolinium acceptors, was developed and applied for imaging nitroreductase (NTR) in hypoxic tumor cells. Nitrobenzyl moieties on the substrate can be conveniently converted into aminobenzyl groups with a nitroreductase-catalyzed reaction in the presence of reduced nicotinamide adenine dinucleotide (NADH). This is followed by a cleavage reaction and the release of the free phenol moiety, which is manifested in enhanced fluorescence intensity during the detection process. Our experimental results show that the NTR detection ability is over 100 equivalents of other biological reductants (1 mM), whereas the confocal fluorescence imaging of tumor cells indicates the possibility of its application in biomedical research fields for tumor hypoxia diagnosis.

Abstract:Nitroxyl (HNO) is one of the important derivatives of nitric oxide (NO). It was intertwined with various biological and pharmacological events as a well-defined active molecule. Developing fluorescent probes for high specific and in situ trapping of HNO in living samples is still challenging. In this project, we constructed a near-infrared (NIR) metal-free fluorescent probe (DCM-P) for monitoring HNO in vitro and in vivo. The novel probe, DCM-P, contains a dicyanomethylene-4H-pyran (DCM) fluorophore as the reporter and a triarylphosphine as the recognition moiety. Upon exposure to HNO, the probe emits fluorescence at the wavelength of 688 nm, which belongs to near-infrared region and endows great beneficial for imaging in vivo. Moreover, DCM-P shows high chemoselective detection and fast response to HNO in the presence of various biological relevant reductants, and is successfully employed to trap nitroxyl in different living cells and zebrafish models. These results suggest that DCM-P is a suitable near-infrared metal-free fluorescent probe for motoring nitroxyl in living samples.

Abstract:We report a novel colorimetric and red-emitting fluorescent probe for hydrazine detection based on resorufin platform. This OFF-ON fluorescent probe shows a large (117nm) red-shifted absorption spectrum and the color changes from colorless to red upon addition of hydrazine in the aqueous solution, which can serve as a "naked-eye" probe for hydrazine. Moreover, this probe also shows a significant fluorescence increase (~16 folds) and excellent linear relationship at physiological pH. Utilizing this sensitive and selective probe, we have successfully detected hydrazine in living cells.

Abstract:Nitroreductase (NTR), one of the flavin-dependent enzymes and an upregulated enzyme under tumor hypoxia, has been studied for decades. Many fluorescent probes were developed to detect NTR activity; however, these probes tend to diffuse away from their reaction site (NTR) inevitably, leading to inappropriate sample fixation, lower accuracy of NTR localization, and weaker signal-to-noise ratio. Herein, we present the design, synthesis, in vitro evaluation, and biological applications of an NTR-activatable fluorogenic and labeling probe FY. By integrating with quinone methide (QM) proximity-based protein labeling, the additional fluoromethyl group on FY serves as a potential origin of QM. Compared with conventional fluorescent probes, this new NTR probe not only offers mitochondrial localizable and fluorogenic response but also achieves permanent retention on the site of activation with an enhanced spatial resolution to improve the detection sensitivity even after cell fixation. We believe our work could offer an expandable synthetic approach to develop these permanent labeling and imaging fluorescence probes for deciphering complex biological events.

Abstract:Nitroreductase (NTR) is to be pivotal in the biodegradation of nitroaromatics. NTR is produced in tumor tissues under hypoxic conditions, which is one of the markers for early tumor diagnosis. In this study, a novel probe FD-NTR was developed for NTR detection. Probe FD-NTR can exhibit a specific reaction with NTR in the presence of NADH. The probe displayed satisfactory selectivity and sensitivity towards NTR with a calculated detection limit of 12 ng/mL. Under the conditions of low cytotoxic hypoxia, the FD-NTR probe has shown successful application in imaging both MCF-7 cells and tumor tissues, which indicated that the FD-NTR probe holds promising application prospects for detecting NTR in tumors.

Abstract:A novel near-infrared (NIR) fluorescent probe based on cyanine dyes was designed and synthesized to detect nitroreductase (NTR) activity. Upon reaction with NTR, the nitro-group of p-nitrobenzyl moiety was reduced to amino-group and therefore renews the π-electron conjugation of the dye, resulting in the intense NIR fluorescent emission (708 nm). This redox-induced NIR-fluorescence probe has been successfully applied to the detection of NTR activity under physiological conditions with high selectivity.

Abstract:Nitroxyl (HNO) has been discovered to play a vital role in diverse physiological and pathological processes. In this work, we report a new reductant-resistant fluorescent probe (NHMC-P) for highly effective tracking of HNO in vitro and in vivo in the near-infrared regions. NHMC-P contains a nitrogen-hydroxylmerocyanine (NHMC) as the fluorophore, linked with a 2-(diphenylphosphino)benzoic acid through an ester bond as the recognition moiety. Upon reaction with HNO, the recognition moiety will be triggered to an aza-ylide, which then attacks the intramolecular ester carbonyl group to release the fluorophore NHMC and restore the fluorescence. NHMC-P exhibited ultra-sensitivity and high selectivity for HNO in the near-infrared region. Furthermore, ascribed to the excellent water-solubility, the probe was successfully employed to track HNO in living cancer cells and zebrafish models. Our work provides a potential tool for in vivo HNO tracking and is helpful for HNO-related pathological studies in living organisms.

Abstract:Nitroreductase (NTR) acts as a detective enzyme that can be used to judge the hypoxic level of solid tumors. To date, only a few optical probes can effectively and selectively monitor NTR in vivo. Therefore, it is important to develop an ideal probe with near-infrared (NIR) excitation and emission for NTR detection and imaging. In this study, we report a NIR frequency upconversion luminescence (FUCL) probe (Cy7-NO2) based on a cyanine structure decorated with an aromatic nitro group to detect NTR in vivo. Cy7-NO2 not only was excited at 850 nm and emitted at 790 nm, but also showed a fast and selective response to NTR. Docking calculations revealed that NTR was able to form hydrogen bonds with Cy7-NO2 and underwent an enzyme-catalyzed reduction reaction. Compared with absorption and the Stokes emission methods, FUCL method exhibited a lower detection limit (3.4 ng mL(-1)). In addition, Cy7-NO2 was used for FUCL bioimaging of NTR in living cells and mice. All of these results proved that Cy7-NO2 as a FUCL probe supplies a new method for biodetection and bioimaging of NTR in vitro and in vivo.

Abstract:We designed and synthesized a novel near-infrared (NIR) mitochondria-targeted fluorescent probe RQNA for the specific detection of mitochondrial Cu2+ because mitochondria are important reservoirs of intracellular copper. For the preparation of this probe, a novel π-extended fluorescent xanthene dye RQN was firstly synthesized via an intramolecular nucleophilic substitution of aromatic hydrogen (SNArH) strategy. Then, probe RQNA was prepared by the reaction of RQN and hydrazine hydrate, followed by treatment with acetone. RQNA exhibited selectivity, sensitivity (22 nM), and fast response time (20 s) for the detection of Cu2+ via a specific Cu2+-triggered ring-opening and hydrolysis cascade reaction. RQNA is cell-membrane permeable and mitochondria-targetable, and can be used for monitoring mitochondrial Cu2+ in living cells.

Abstract:Nitroreductase (NTR) plays a vital part in the activation of drugs and the biodegradation of nitroaromatic compounds. With the enhanced expression level of NTR in hypoxic solid tumors, it has been widely used as one of the biomarkers to evaluate the degree of tumor hypoxia. In this work, we reported a benzoindole-based fluorescent probe NFP-NTR for nitroreductase detection. NFP-NTR can react with NTR specifically to cleave the p-nitrobenzyl ether bond when nicotinamide adenine dinucleotide (NADH) is present, showing significant fluorescence augment characteristics. Encouragingly, NFP-NTR exhibited excellent selectivity and satisfactory sensitivity, with a detection limit of 17 ng/mL. In addition, this probe has been successfully used to image live Hela cells under hypoxic conditions with low cytotoxicity, indicating that probe NFP-NTR has great potential to further elucidate more biological applications of NTR.

Abstract:Rhodamine dyes bearing aromatic nitro group has been synthesized for nitroreductase enzyme chemosensing applications. The probe is showing very selective turn-on fluorescent response towards nitroreductase enzymes and in hypoxic conditions. The sensor displays a remarkable fluorescent enhancement at 557 nm (λex = 500 nm) without the interference of other biologically relevant species under hypoxic conditions in a physiological medium. The nitro group in the sensor is reduced by the nitroreductase enzyme to the amino group, resulting in the hydrolysis of the probe and subsequent release of highly fluorescent rhodamine 6G dye is observed. This rhodamine based fluorescent probe has been utilized for the imaging of nitroreductase enzymes as well as hypoxia in live cells.

Abstract:Reduced nicotinamide adenine dinucleotide (NADH) is a kind of coenzyme and widely works as a biomarker in cancer cells. It plays a crucial role in many cellular metabolic processes, especially NADH in mitochondria is indispensable for the mitochondrial respiration chain that produces ATP. Herein, we designed a fluorescent probe Mito-FCC based on an ethylene-bridging dual-salt structure, in which benzo[e]indolium fluorophore was used as the mitochondria-targeting group and 1-methylquinolinium moiety as the NADH recognition unit. Mito-FCC exhibited high sensitivity and selectivity for NADH with a rapid "turn-on" fluorescence signal. The dual-salt structure endowed the probe with a reliable mitochondria-targeted ability even after the recognition unit was reduced by NADH. With the help of the probe, the fluctuations of endogenous NADH induced by glucose or pyruvate were imaged. Besides, Mito-FCC had a capability to make a distinction between cancer cells and normal cells due that the content of NADH in cancer cells was distinctly higher than that in normal ones. Notably, the visualization of tumor in vivo through monitoring NADH using Mito-FCC was realized successfully. These experimental results showed that Mito-FCC hold a great perspective in study of mitochondrial function and potential diagnosis of cancer diseases.
