Abstract:ABSTRACT Anaplasma marginale is an obligate, intracellular, tick-borne bacterial pathogen that causes bovine anaplasmosis, an often severe, production-limiting disease of cattle found worldwide. Methods to control this disease are lacking, in large part due to major knowledge gaps in our understanding of the molecular underpinnings of basic host–pathogen interactions. For example, the surface proteins that serve as adhesins and, thus, likely play a role in pathogen entry into tick cells are largely unknown. To address this knowledge gap, we developed a phage display library and screened 66 A . marginale proteins for their ability to adhere to Dermacentor andersoni tick cells. From this screen, 17 candidate adhesins were identified, including OmpA and multiple members of the Msp1 family, including Msp1b, Mlp3, and Mlp4. We then measured the transcript of ompA and all members of the msp1 gene family through time, and determined that msp1b , mlp2 , and mlp4 have increased transcript during tick cell infection, suggesting a possible role in host cell binding or entry. Finally, Msp1a, Msp1b, Mlp3, and OmpA were expressed as recombinant protein. When added to cultured tick cells prior to A. marginale infection, all proteins except the C-terminus of Msp1a reduced A. marginale entry by 2.2- to 4.7-fold. Except OmpA, these adhesins lack orthologs in related pathogens of humans and animals, including Anaplasma phagocytophilum and the Ehrlichia spp., thus limiting their utility in a universal tick transmission-blocking vaccine. However, this work greatly advances efforts toward developing methods to control bovine anaplasmosis and, thus, may help improve global food security.

Abstract:Tick saliva contains a number of effector molecules that inhibit host immunity and facilitate pathogen transmission. How tick proteins regulate immune signaling, however, is incompletely understood. Here, we describe that loop 2 of sialostatin L2, an anti-inflammatory tick protein, binds to annexin A2 and impairs the formation of the NLRC4 inflammasome during infection with the rickettsial agent Anaplasma phagocytophilum. Macrophages deficient in annexin A2 secreted significantly smaller amounts of interleukin-1 beta (IL-1 beta) and IL-18 and had a defect in NLRC4 inflammasome oligomerization and caspase-1 activation. Accordingly, Annexin a2-deficient mice were more susceptible to A. phagocytophilum infection and showed splenomegaly, thrombocytopenia, and monocytopenia. Providing translational support to our findings, better binding of annexin A2 to sialostatin L2 in sera from 21 out of 23 infected patients than in sera from control individuals was also demonstrated. Overall, we establish a unique mode of inflammasome evasion by a pathogen, centered on a blood-feeding arthropod.

Abstract:Anaplasmosis is increasingly common in the United States, with cases being reported over an expanding geographic area. To monitor for changes in risk of human infection, the U.S. Centers for Disease Control and Prevention monitors the distribution and abundance of host-seeking vector ticks ( Ixodes scapularis and Ixodes pacificus ) and their infection with Anaplasma phagocytophilum . While several variants of A. phagocytophilum circulate in I. scapularis , only the human-active variant (Ap-ha) appears to be pathogenic in humans. Failure to differentiate between human and non-human variants may artificially inflate estimates of the risk of human infection. Efforts to differentiate the Ap-ha variant from the deer variant (Ap-V1) in ticks typically rely on traditional PCR assays coupled with sequencing of PCR products. However, laboratories are increasingly turning to Next Generation Sequencing (NGS) to increase testing efficiency, retain high sensitivity, and increase specificity compared with traditional PCR assays. We describe a new NGS assay with novel targets that accurately segregate the Ap-ha variant from other non-human variants and further identify unique clades within the human and non-human variants. Recognizing that not all investigators have access to NGS technology, we also developed a PCR assay based on one of the novel targets so that variants can be visualized using agarose gel electrophoresis without the need for subsequent sequencing. Such an assay may be used to improve estimates of human risk of developing anaplasmosis in North America.

Abstract:Abstract Background Emerging tick-transmitted illnesses are increasingly recognized in the United States. To identify multiple potential tick-borne pathogens in patients from the Upper Midwest and Northeast US with suspected anaplasmosis, we used state-of-the-art methods (PCR and paired serology) to test samples from patients in whom anaplasmosis had been excluded. Methods 568 patients without anaplasmosis had optimal samples available for confirmation of alternative tick-borne pathogens, including PCR and/or paired serology (acute-convalescent interval ≤ 42 days). Results Among 266 paired serology evaluations, for which the median acute-convalescent sampling interval was 28 days (IQR 21-33), we identified 35 acute/recent infections [24 (9%) Borrelia burgdorferi; 6 (2%) Ehrlichia chaffeensis/ Ehrlichia muris subsp. eauclairensis (EC/EME); 3 (1%) spotted fever group rickettsioses (SFGR)], and 2 (<1%) Babesia microti] in 33 (12%) patients. Two had concurrent or closely sequential infections (1 B. burgdorferi and EC/EME, and 1 B. burgdorferi and SFGR). Using multiplex PCR and reverse transcriptase-PCR we identified 7 acute infections [5/334 (1%) Borrelia miyamotoi and 2/334 (1%) B. microti infections] in 5 (1%) patients, including 2 with B. microti-B. miyamotoi co-infection, but no B. mayonii, SFGR, Candidatus Anaplasma capra, Heartland virus, or Powassan virus infections. Thus, amongst 568 patients with ruled out anaplasmosis, 38 (6.7%) had ≥1 agents of tickborne illness identified, with 33 patients (35 infections) diagnosed by paired serology and 5 additional patients (7 infections) by PCR. Conclusions By identifying other tickborne agents in patients in whom anaplasmosis had been excluded, we demonstrate that emerging tickborne infections will be identified if specifically sought.

Abstract:To the Editor: Anaplasma phagocytophilum, an emerging human pathogen of public health importance, is transmitted to humans most commonly by tick bites (1). The agent has been detected in various species of Ixodes ticks around the world (2) and in Dermacentor silvarum ticks in northeastern People’s Republic of China (3), where 3 A. phagocytophilum strains were isolated from wild and domestic animals (4). In the Asiatic region of Russia adjacent to China, A. phagocytophilum was identified in Ixodes persulcatus ticks, and A. bovis in Haemaphysalis concinna ticks (5). Human granulocytic anaplasmosis was reported in the southern area of the Russian Far East that borders China (6). The objectives of this study were to investigate the prevalence of A. phagocytophilum in ticks collected from the China–Russia border and to characterize the agent by molecular biology techniques.

Abstract:The invasive Asian longhorned tick Haemaphysalis longicornis that vectors and transmits several animal pathogens is significantly expanding in the United States. Recent studies report that these ticks also harbor human pathogens including Borrelia burgdorferi sensu lato, Babesia microti , and Anaplasma phagocytophilum . Therefore, studies that address the interactions of these ticks with human pathogens are important. In this study, we report the characterization of H. longicornis organic anion-transporting polypeptides (OATPs) in interactions of these ticks with A. phagocytophilum . Using OATP-signature sequence, we identified six OATPs in the H. longicornis genome. Bioinformatic analysis revealed that H. longicornis OATPs are closer to other tick orthologs rather than to mammalian counterparts. Quantitative real-time PCR analysis revealed that OATPs are highly expressed in immature stages when compared to mature stages of these ticks. In addition, we noted that the presence of A. phagocytophilum upregulates a specific OATP in these ticks. We also noted that exogenous treatment of H. longicornis with xanthurenic acid, a tryptophan metabolite, influenced OATP expression in these ticks. Immunoblotting analysis revealed that antibody generated against Ixodes scapularis OATP cross-reacted with H. longicornis OATP. Furthermore, treatment of H. longicornis with OATP antibody impaired colonization of A. phagocytophilum in these ticks. These results not only provide evidence that the OATP-tryptophan pathway is important for A. phagocytophilum survival in H. longicornis ticks but also indicate OATP as a promising candidate for the development of a universal anti-tick vaccine to target this bacterium and perhaps other rickettsial pathogens of medical importance.
