Redox-responsive self-assembled peptide hydrogel for mitochondrial-targeted anticancer drug delivery

Abstract:Achieving controlled release and organelle-targeted drug delivery to improve therapeutic efficacy while minimizing side effects on surrounding healthy cells remains challenging. Peptide-based delivery approaches provide a new platform for cancer therapy. To achieve selective drug release in cancer cells, we developed a redox-responsive Pep-CS-LND hydrogel with mitochondria-targeting potential by incorporating disulfide linker and KLAK sequences. We characterized the self-assembly mechanism, redox-responsive drug release and mitochondrial targeting ability of Pep-CS-LND hydrogel. Furthermore, we assessed its antitumor activity and biocompatibility through in vitro cell and in vivo animal experiments. The self-assembled Pep-CS-LND hydrogel formed a "drug repository" around the tumor, efficiently releasing LND-KLAK in response to high glutathione (GSH) levels. It also effectively targeted mitochondria demonstrated efficacy in targeting mitochondria, reducing GSH, adenosine triphosphate, and mitochondrial membrane potential, while concurrently increasing levels of reactive oxygen species. Furthermore, both the in vitro and in vivo results revealed that Pep-CS-LND hydrogel had the effective tumor accumulation capacity, excellent antitumor activity, and good biocompatibility. Pep-CS-LND hydrogel shows great potential for effective mitochondrial drug delivery and enhanced anticancer efficacy.
