Abstract:With the increasing demands for high-throughput multiplexed bioassays, quantum dot (QD)-encoded microbeads as biocarriers for various bioreactions have attracted considerable attention. However, three key requirements for these biocarriers are still longstanding issues: a stable fluorescence intensity, a large encoding capacity and abundant surface functional groups. Here, a novel one-pot strategy is developed, generating functionalized QD-encoded microspheres with a strong fluorescence intensity and optical stability. With poly(styrene-co-maleic anhydride) (PSMA) molecules as mediators, the encapsulation of QDs and carboxylation of the bead surface are integrated together, greatly improving the preparation efficiency and guaranteeing their potential application in biodetection. Moreover, the mechanism for preparing QD-doped beads is further proposed, which helps to precisely manipulate the preparation process and accurately encode the beads. Through this approach, a single- and dual-color barcode library of QD-encoded microspheres has been successfully established, which demonstrates their great potential in suspension arrays.

Abstract:Fluorescence-encoded microspheres are largely demanded in high-throughput assays. A strategy for preparing encoded microspheres based on fluorescence resonance energy transfer (FRET) was advanced. Two fluorophores, poly(phenylenevinylene) nanoparticles (PPV NPs) as the donor and rhodamine 6G (R6G) as the acceptor were employed to construct the energy-transfer pair, and introduced onto the substrate polymeric microspheres. The fluorescence intensity ratio between PPV NPs and R6G molecules were tuned based on energy transfer mechanism. Nine codes (wavelength-intensity combinations) were obtained at the same excitation by keeping the concentration of PPV NPs constant and varying the concentration of R6G in the dipping solutions. Then, a melamine–formaldehyde (MF) resin layer was coated onto the encoded spheres to prevent the leakage of fluorophores. The encoded spheres were found to have good fluorescence stability against various solvents and even "dissociation agent", and no significant variation of the codes were observed. Further modification of this MF layer with silanization reaction provided the amino reactive sites. In all, this study provides a general strategy for the preparation of fluorescence encoded microspheres based on energy transfer as well as a stable protective layer with reactive sites.

Abstract:We report a facile strategy for fabricating fluorescent quantum dot (QD)-loaded microbeads by means of microfluidic technology. First, a functional fluorine-containing microemulsion was synthesized with poly[(2-(N-ethylperfluorobutanesulfonamido)ethyl acrylate)-co-(methyl methacrylate)-co-(butyl acrylate)] (poly(FBMA-co-MMA-co-BA)) as the core and glycidyl methacrylate (GMA) as the shell via differential microemulsion polymerization. Then, CdTe QDs capped with N-acetyl-l-cysteine (NAC) were assembled into the poly(FBMA-co-MMA-co-BA-co-GMA) microemulsion particles through the reaction of the epoxy group on the shell of the microemulsion and the carboxyl group of the NAC ligand capped on the QDs. Finally, fluorescent microbeads were fabricated using the CdTe QD-loaded fluorine-containing microemulsion as the discontinuous phase and methylsilicone oil as the continuous phase by means of a simple microfluidic device. By changing flow rate of methylsilicone oil and hybrid microemulsion system, fluorescent microbeads with adjustable sizes ranging from 290 to 420 mu m were achieved. The morphology and fluorescent properties of the microbeads were thoroughly investigated using optical microscopy and fluorescence microscopy. Abstract:Quantum dots encoded microbeads have various advantages such as large surface area, superb optical properties and the ability of multiplexing. Molecularly imprinted polymer that can mimic the natural recognition entities has high affinity and selectivity for the specific analyte. Here, the concept of utilizing the quantum dots encoded microbeads as the supporting material and the polydopamine as the functional monomer to form the core-shell molecular imprinted polymer was proposed for the first time. The resulted imprinted polymer can provide various merits: polymerization can complete in aqueous environment; fabrication procedure is facile and universal; the obvious economic advantage; the thickness of the imprinting layer is highly controllable; polydopamine coating can improve the biocompatibility of the quantum dot encoded microbeads. The rabbit IgG binding and flow cytometer experiment result showed the distinct advantages of this strategy: cost-saving, facile and fast preparation procedure. Most importantly, the ability for the multichannel detection, which makes the imprinted polydopamine modified encoded-beads very attractive in protein pre-concentration, recognition, separation and biosensing.

Abstract:Bead-based optical encoding or magnetic encoding techniques are promising in high-throughput multiplexed detection and separation of numerous species under complicated conditions. Therefore, a self-assembly strategy implemented in an organic solvent is put forward to fabricate fluorescent-magnetic dual-encoded nanospheres. Briefly, hydrophobic trioctylphosphine oxide-capped CdSe/ZnS quantum dots (QDs) and oleic acid-capped nano-γ-Fe2O3 magnetic particles are directly, selectively and controllably assembled on branched poly(ethylene imine)-coated nanospheres without any pretreatment, which is crucial to keep the high quantum yield of QDs and good dispersibility of γ-Fe2O3. Owing to the tunability of coating amounts of QDs and γ-Fe2O3 as well as controllable fluorescent emissions of deposited-QDs, dual-encoded nanospheres with different photoluminescent emissions and gradient magnetic susceptibility are constructed. Using this improved layer-by-layer self-assembly approach, deposition of hydrophobic nanoparticles onto hydrophilic carriers in organic media can be easily realized; meanwhile, fluorescent-magnetic dual-functional nanospheres can be further equipped with readable optical and magnetic addresses. The resultant fluorescent-magnetic dual-encoded nanospheres possess both the unique optical properties of QDs and the superparamagnetic properties of γ-Fe2O3, exhibiting good monodispersibility, huge encoding capacity and nanoscale particle size. Compared with the encoded microbeads reported by others, the nanometre scale of the dual-encoded nanospheres gives them minimum steric hindrance and higher flexibility.

Abstract:Fluorescent probes have long been regarded as tools for imaging living organisms with advantages such as high sensitivity, good designability and multifunctional potential. Many fluorescent probes, especially the probes based on aggregation-induced emission (AIE) dyes, have received increasing attention since the AIE phenomenon was discovered. These AIE dye-based fluorescent probes could elegantly overcome the notorious quenching effect caused by aggregation of conventional organic dyes. However, it is still difficult to directly apply these AIE-active dyes for biomedical applications owing to their hydrophobic nature. Therefore, the development of novel and facile strategies to endow them with water dispersibility is of critical importance. In this work, we exploit an efficient and simple strategy to fabricate an AIE dye-based fluorescent copolymer through the combination of reversible addition-fragmentation chain transfer and the Biginelli reaction. Moreover, the copolymer can self-assemble to fluorescent polymeric nanoparticles (FPNs) in water solution. Hydrophilic poly(PEGMA-co-AEMA) was reacted with the AIE-active dye 4',4‴-(1,2-diphenylethene-1,2-diyl)bis([1,1'-biphenyl]-4-carbaldehyde (CHO-TPE-CHO) to form amphiphilic luminescent polymers using urea as the connection bridge. The successful synthesis of the final products (poly(PEGMA-co-AEMA-TPE) FPNs) was confirmed by various instruments. Furthermore, Transmission electron microscopy (TEM) images manifest that poly(PEGMA-co-AEMA-TPE) copolymers will self-assemble into spherical nanoparticles in aqueous environments with sizes between 100 nm and 200 nm. The cell uptake and bioimaging experiment confirm that poly(PEGMA-co-AEMA-TPE) FPNs have excellent biocompatibility and emit strong green fluorescence in a cellular environment. Thus, poly(PEGMA-co-AEMA-TPE) FPNs are excellent candidates for biomedical applications.

Abstract:Encoded microparticles (EMPs) have shown demonstrative value for multiplexed high-throughput bioassays such as drug discovery and diagnostics. Herein, we propose for the first time the incorporation of thermally activated delayed fluorescence (TADF) dyes with low-cost, heavy metal-free, and long-lived luminescence properties into polymer matrices via a microfluidic droplet-facilitated assembly technique. Benefiting from the uniform droplet template sizes and polymer-encapsulated structures, the resulting composite EMPs are highly monodispersed, efficiently shield TADF dyes from singlet oxygen, well preserve TADF emission, and greatly increase the delayed fluorescence lifetime. Furthermore, by combining with phase separation of polymer blends in the drying droplets, TADF dyes with distinct luminescent colors can be spatially separated within each EMP. It eliminates optical signal interference and generates multiple fluorescence colors in a compact system. Additionally, in vitro studies reveal that the resulting EMPs show good biocompatibility and allow cells to adhere and grow on the surface, thereby making them promising optically EMPs for biolabeling.

Abstract:Self-assembly of amphiphilic luminescent copolymers is a general route to fabricate fluorescent polymeric microparticles (FPMs). In this work, the FPMs with aggregation-induced emission (AIE) feature were fabricated via the combination of the chain transfer free radical polymerization and "one-pot" multicomponent reaction, which conjugated the aldehyde-containing AIE active dye AIE (CHO-An-CHO) and amino-terminated hydrophilic polymer (ATPPEGMA) using mercaptoacetic acid (MTA) as the "lock" molecule. The structure, chemical compositions, optical properties as well as biological properties of the PPEGMA-An-PPEGMA FPMs were characterized and investigated by means of a series of techniques and experiments in detail. We demonstrated the final copolymers showed amphiphilic properties, strong yellow fluorescence and high water dispersibility. Biological evaluation suggested that PPEGMA-An-PPEGMA FPMs possess low cytotoxicity and can be used for cell imaging. More importantly, many other AIE active FPMs are expected to be fabricated using the similar strategy because of the good substrate and monomer applicability of the multicomponent reaction and chain transfer living radical polymerization. Therefore, we could conclude that the strategy described in this work should be of great interest for fabrication of multifunctional AIE active nanoprobes for biomedical applications.

Abstract:Droplet-based single cell sequencing technologies, such as inDrop, Drop-seq, and 10X Genomics, are catalyzing a revolution in the understanding of biology. Barcoding beads are key components for these technologies. What is limiting today are barcoding beads that are easy to fabricate, can efficiently deliver primers into drops, and thus achieve high detection efficiency. Here, this work reports an approach to fabricate dissolvable polyacrylamide beads, by crosslinking acrylamide with disulfide bridges that can be cleaved with dithiothreitol. The beads can be rapidly dissolved in drops and release DNA barcode primers. The dissolvable beads are easy to synthesize, and the primer cost for the beads is significantly lower than that for the previous barcoding beads. Furthermore, the dissolvable beads can be loaded into drops with >95% loading efficiency of a single bead per drop and the dissolution of beads does not influence reverse transcription or the polymerase chain reaction (PCR) in drops. Based on this approach, the dissolvable beads are used for single cell RNA and protein analysis.

Abstract:Water dispersion aggregation-induced emission (AIE) dyes based nanomaterials have recently attracted increasing attention in the biomedical fields because of their unique optical properties, outstanding performance as imaging and therapeutic agents. The methods to conjugate hydrophilic polymers with AIE dyes to solve the hydrophobic nature of AIE dyes and makeS them widely used in biomedicine, which have been extensively explored and paid great effort previously. Although great advance has been made in the fabrication and biomedical applications of AIE-active polymeric nanoprobes, facile and efficient strategies for fabrication of biodegradable AIE-active nanoprobes are still high desirable. In this work, amphiphilic biodegradable fluorescent organic nanoparticles (PLL-TPE-O-E FONs) have been fabricated for the first time by conjugation of AIE dye tetraphenylethene acrylate (TPE-O-E) with Poly-l-Lysine (PLL) through a facile one-step Michael addition reaction, which was carried out under rather mild conditions, included air atmosphere, near room temperature and absent of metal catalysts or hazardous reagents. Due to the unique AIE properties, these amphiphilic copolymers tend to self-assemble into high luminescent water dispersible nanoparticles with size range from 400 to 600nm. Laser scanning microscope and cytotoxicity results revealed that PLL-TPE-O-E FONs can be internalized into cytoplasm with negative cytotoxicity, which implied that PLL-TPE-O-E FONs are promising for biological applications.
An Efficient Method for Preparing High-Performance Multifunctional Polymer Beads Simultaneously Incorporated with Magnetic Nanoparticles and Quantum Dots

Abstract:An efficient modified method combining conventional swelling method with high-temperature swelling method was applied to prepare high-stable multifunctional MNPs-QD-encoded polystyrene beads simultaneously with large encoding capacity and fast separation. In this paper, the results demonstrate that encapsulating QDs into pre-prepared MNPs-beads is the best sequence, which keeps high brightness and fast separation. The MNPs-QD-encoded microcarriers designed by our proposed method exhibit excellent performance for magnetic manipulation and optical encoding. What is more, they also have better physical and chemical stabilities. Compared with beads prepared via conventional swelling method, the leakage of nanoparticles from the designed MNPs-QD-encoded poly (styrene-co-ethylene glycol dimethacrylate-co-methacrylic acid) beads (PSEMBs) induced by organic solvents (e. g. cyclohexane) is less than 6%; meanwhile, fluorescence intensity of MNPs-QDs-encoded PSEMBs fluctuates slightly more in a wide range of pH 2-12 buffers. In addition, immunoassay performance for human IgG detections indicates that carboxyl groups on fluorescent microsphere surface facilitate efficient attachment of biomolecule and therefore they can be further applied to fast separation and multiplexed biomolecular assays.

Abstract:Novel cross-linked copolymers of PEG-IM-PhNH2 are successfully synthesized through PEGylation via radical polymerization of 2-isocyanatoethyl methacrylate and poly(ethylene glycol) monomethyl ether methacylate and subsequent cross-linking with an amino-terminated aggregation-induced emission fluorogen. Such obtained amphiphilic copolymers can self-assemble to form uniform fluorescent polymeric nanoparticles (FPNs) and be utilized for cell imaging. These cross-linked FPNs are demonstrated good water dispersibility with ultralow critical micelle concentration (∼ 0.002 mg mL(-1)), uniform morphology (98 ± 2 nm), high red fluorescence quantum yield, and excellent biocompatibility. More importantly, this novel strategy of fabricating cross-linked FPNs paves the way to the future development of more robust and biocompatible fluorescent bioprobes.

Abstract:In recent years, aggregation-induced emission (AIE) dyes based fluorescent organic nanoparticles (FONs) have achieved significant progress in various biomedical applications. In this work, we developed a covalent strategy to prepare biocompatible AIE-active dyes based cross-linked copolymers (MPC-POSS-PhE) via controllable reversible addition fragmentation chain transfer (RAFT) polymerization using zwitterionic 2-methacryloyloxyethyl phosphorylcholine (MPC), polymerizable AIE dye (named as PhE) and 8-vinyl polyoctahedral silsesquioxanes (POSS) as monomers. Due to the existence of hydrophilic MPC and hydrophobic PhE, the resultant copolymers will self-assemble into core-shell nanoparticles in aqueous solution with ultra-low critical micelle concentration (CMC). This could effectively overcome the drawbacks of non-crosslinked micelles and show more attractive properties and better performance for biomedical applications. Furthermore, the characterization results and biological assays demonstrated that the final MPC-POSS-PhE FONs show stable aqueous stability, uniform size and morphology, high water dispersity, desirable optical properties and low cytotoxicity. These remarkable properties make the resultant AIE-active nanoprobes great potential for biomedical applications.
Quantum Dot-Encoded Microbeads: Synthesis And Application To Microchip Flow Cytometer For Diagnosis

Abstract:A series of quantum dot(QD)-encoded polystyrene microbeads is prepared. Compared with microbeads loaded with traditional organic dyes, as-synthesized fluorescence microbeads have better photostability. These microbeads could conjugate with protein by further surface modification, so they could be applied to such fields as biochemistry, molecular biology, cell biology, immunology. A home-made microfluidic system is designed and fabricated, and coupled with flow cytometry for QD-encoded microbeads analysis. The microbeads modified with biomolecules can be used for pathogen detection. A method for detecting epidemic hemorrhagic fever and new-castle disease based on such microbeads was developed. It was shown the set-up could be a good platform for detecting multiple disease pathogens, simultaneously.

Abstract:Magnetic-fluorescent microbeads had been extensively used in various applications, particularly in the multiple detection of biological molecules. Currently, most commercially available magnetic-fluorescent microbeads had simple structures and the fluorescence intensity was inversely proportional to the concentration of magnetic particles, which affected performances of microbeads. In this work, a three-dimensional capillary microfluidic system was constructed to produce magnetic-fluorescent core-shell microbeads. Magnetic nanoparticles were dispersed in the core of the microbeads, while the shell consisted of quantum dots (QDs). Core-shell structures restricted fluorescence quenching caused by magnetic nanoparticles in the core area. Moreover, the number of cores could be adjusted precisely, which provided an additional encoding method besides the traditional fluorescence encoding. Three types of dual-encoded core-shell microbeads which exhibited excellent uniformity and fluorescence characteristics with additional magnetic response properties were produced. The self-assembly of silica nanoparticles on the surface of microbeads made the introduction of carboxyl groups easy. Furthermore, the capability of these microbeads for multiplex immunoassays was demonstrated using three types of IgG, and a calibration curve for the detection of human IgG was established using a sandwich immunoassay. These results indicated the potential application of this scheme in the field of biomedical detection.

Obvious different from traditional fluorescence quenching materials, AIE-active materials could emit stronger fluorescence in concentrated and solid state. In this work, an AIE-active molecule (named as TPE-DB) was synthesized and applied to prepare amphiphilic fluorescent copolymers through light-induced RAFT polymerization in the present of air. The poly(ethylene oxide) monomethacrylate (PEGMA), itaconic acid (IA) and TPE-DB were selected as monomers and Ir3+ complex was used as photoredox catalyst. The samples were carefully characterized and analyzed by different techniques. More interesting, we demonstrated that TPE-PEGMA-IA could form nanoparticles with small size and high water dispersibility. The results from cell viability evaluation and confocal laser scanning microscopy imaging suggested that these fluorescent nanoparticles possess low cytotoxicity and are promising for biological imaging. Owing to the introduction of IA, these fluorescent polymeric nanoparticles (FPNs) could also be utilized for drug delivery. Taken together, we believe that the light-induced polymerization should be a simple and useful route for synthesizing AIE-active fluorescent copolymers, which could greatly advance the biomedical applications of AIE-active functional materials.

Abstract:Luminophore stained micro- and nanobeads made from organic polymers like polystyrene (PS) are broadly used in the life and material sciences as luminescent reporters, for bead-based assays, sensor arrays, printable barcodes, security inks, and the calibration of fluorescence microscopes and flow cytometers. Initially mostly prepared with organic dyes, meanwhile luminescent core/shell nanoparticles (NPs) like spherical semiconductor quantum dots (QDs) are increasingly employed for bead encoding. This is related to their narrower emission spectra, tuneability of emission color, broad wavelength excitability, and better photostability. However, correlations between particle architecture, morphology, and photoluminescence (PL) of the luminescent nanocrystals used for encoding and the optical properties of the NP-stained beads have been rarely explored. This encouraged us to perform a screening study on the incorporation of different types of luminescent core/shell semiconductor nanocrystals into polymer microparticles (PMPs) by a radical-induced polymerization reaction. Nanocrystals explored include CdSe/CdS QDs of varying CdS shell thickness, a CdSe/ZnS core/shell QD, CdSe/CdS quantum rods (QRs), and CdSe/CdS nanoplatelets (NPLs). Thereby, we focused on the applicability of these NPs for the polymerization synthesis approach used and quantified the preservation of the initial NP luminescence. The spectroscopic characterization of the resulting PMPs revealed the successful staining of the PMPs with luminescent CdSe/CdS QDs and CdSe/CdS NPLs. In contrast, usage of CdSe/CdS QRs and CdSe QDs with a ZnS shell did not yield luminescent PMPs. The results of this study provide new insights into structure–property relationships between NP stained PMPs and the initial luminescent NPs applied for staining and underline the importance of such studies for the performance optimization of NP-stained beads.

Abstract:To achieve the dual-channel (analog and digital) encoding, microbeads assembled with quantum dots (QDs) and element coding nanoparticles (ECNPs) have been prepared. Dual-spectra, including fluorescence generated from quantum dots (QDs) and laser induced breakdown spectrum obtained from the plasma of ECNPs, including Ago, MgO and ZnO nanoparticles, has been adopted to provide more encoding amounts and more accurate dual recognition for encoded microbeads in multiplexed utilization. The experimental results demonstrate that the single microbead can be decoded in two optical channels. Multiplexed analysis and contrast adsorption experiment of anti-IgG verified the availability and specificity of dual-channel-coded microbeads in bioanalysis. In gradient detection of anti-IgG, we obtained the linear concentration response to target biomolecules from 3.125 x 10(-10) M to 1 x 10(-8) M, and the limit of detection was calculated to be 2.91 x 10(-11) M. Abstract:With the rapid development of suspension array technology, microbeads‐based barcodes as the core element with sufficient encoding capacity are urgently required for high‐throughput multiplexed detection. Here, a novel structure‐fluorescence combinational encoding strategy is proposed for the first time to establish a barcode library with ultrahigh encoding capacities. Based on the never revealed transformability of the structural parameters (e.g., porosity and matrix component) of mesoporous microbeads into scattering signals in flow cytometry, the enlargement of codes number has been successfully realized in combination with two other fluorescent elements of fluorescein isothiocyanate isomer I (FITC) and quantum dots (QDs). The barcodes are constructed with precise architectures including FITC encapsulated within mesopores and magnetic nanoparticles as well as QDs immobilized on the outer surface to achieve the ultrahigh encoding level of 300 accompanied with superparamagnetism. To the best of knowledge, it is the highest record of single excitation laser‐based encoding capacity up to now. Moreover, a ten‐plexed tumor markers bioassay based on the tailored‐designed barcodes has been evaluated to confirm their feasibility and effectiveness, and the results indicate that the barcodes platform is a promising and robust tool for practical multiplexed biodetection.
