Abstract:Rosa rugosa is a famous Chinese traditional flower with high ornamental value and well environmental adapt ability. The cultivation of new colorful germplasms to improve monotonous flower color could promote its landscape application. However, the mechanism of flower color formation in R. rugosa remains unclear. In this study, combined analyses of the chemical and transcriptome were performed in the R. rugosa germplasms with representative flower colors. Among the identified anthocyanins, cyanidin 3,5-O-diglucoside (Cy3G5G) and peonidin 3,5-O-diglucoside (Pn3G5G) were the two dominant anthocyanins in the petals of R. rugosa. The sum content of Cy3G5G and Pn3G5G was responsible for the petal color intensity, such as pink or purple, light- or dark- red. The ratio of Cy3G5G to Pn3G5G was contributed to the petal color hue, that is, red or pink/purple. Maintaining both high relative and high absolute content of Cy3G5G may be the precondition for forming red-colored petals in R. rugosa. Cyanidin biosynthesis shunt was the dominant pathway for anthocyanin accumulation in R. rugosa, which may be the key reason for the presence of monotonous petal color in R. rugosa, mainly pink/purple. In the upstream pathway of cyanidin biosynthesis, 35 differentially expressed structural genes encoding 12 enzymes co-expressed to regulate the sum contents of Cy3G5G and Pn3G5G, and then determined the color intensity of petals. RrAOMT, involved in the downstream pathway of cyanidin biosynthesis, regulated the ratio of Cy3G5G to Pn3G5G via methylation and then determined the color hue of petals. It was worth mentioning that significantly higher delphinidin-3,5-O-diglucoside content and RrF3'5'H expression were detected from deep purple-red-flowered 8-16 germplasm with somewhat unique and visible blue hue. Three candidate key transcription factors identified by correlation analysis, RrMYB108, RrC1, and RrMYB114, might play critical roles in the control of petal color by regulating the expression of both RrAOMT and other multiple structural genes. These results provided novel insights into anthocyanin accumulation and flower coloration mechanism in R. rugosa, and the candidate key genes involved in anthocyanin biosynthesis could be valuable resources for the breeding of ornamental plants in future.
Analysis of flower color diversity revealed the co-regulation of cyanidin and peonidin in the red petals coloration of Rosa rugosa

Abstract:Rosa rugosa is limited in landscaping applications due to its monotonous color, especially the lack of red-flowered varieties. Comprehensive assessment of petal color diversity in R. rugosa could promote to explore the mechanism of flower color formation. In this study, the variation and diversity of petal coloring of 193 R. rugosa germplasms were assessed by chromatic values (L∗, a∗, and b∗), and then divided into seven clusters belonging to three groups with pinkish-purple (185 individuals), white (6), and red (2) petals, respectively. Total anthocyanin content was the most important factor affecting flower color diversity and red hue formation of R. rugosa petals. There were significant correlations between petal color chromatic indexes and the sum content and the ratio of two major anthocyanin, namely cyanidin 3,5-O-diglucoside (Cy3G5G), peonidin 3,5-O-diglucoside (Pn3G5G). Both high levels of Cy3G5G + Pn3G5G and Cy3G5G/Pn3G5G were necessary conditions for red phenotype formation. Five cyanidin up-stream structural genes (RrF3'H1, RrDFR1, RrANS1, RrUF3GT1, RrUF35GT1) and one cyanidin down-stream structural gene (RrCCoAOMT1) were the key indicators which contributed to Cy3G5G + Pn3G5G and Cy3G5G/Pn3G5G, respectively. Functional verification showed that overexpression of RrDFR1, combined with silent expression of RrCCoAOMT1, could make R. rugosa petals redder by increasing the levels of Cy3G5G + Pn3G5G and Cy3G5G/Pn3G5G. These results provided a robust theoretical basis for further revealing the molecular mechanism of red petals coloration in R. rugosa.
Novel Insights into Pigment Composition and Molecular Mechanisms Governing Flower Coloration in Rose Cultivars Exhibiting Diverse Petal Hues

Abstract:The pigmentation of various components leads to different colors of roses. However, the intricate molecular machinery and metabolic pathways underlying rose pigmentation remain largely unexplored. In this study, we determined that pink and black-red petals contain abundant anthocyanins, reaching concentrations of 800 μg/g and 1400 μg/g, respectively, significantly surpassing those in white and yellow petals. We identified 22 key anthocyanin components, predominantly cyanidin, pelargonidin, delphinidin, peonidin, and petunidin, which were preferentially enriched in pink and black-red petals. Additionally, we confirmed the presence of five carotenoid species—lutein, zeaxanthin, ζ-carotene, α-carotene, and β-carotene—with zeaxanthin and carotenoids notably accumulating in yellow petals at significantly higher levels compared with other colors. Furthermore, RNA-seq and qRT-PCR analyses revealed the association between pigment accumulation and the expression patterns of genes involved in anthocyanin and carotenoid biosynthesis pathways. Through promoter core element prediction and transcriptional metabolic co-expression analyses, we found that the MYB transcription factor likely positively modulates the expressions of key biosynthetic genes such as CHS, F3′H, and DFR, while the NAC transcription factor enhances the transcriptional activities of PSY, ZISO, and LYCB. Overall, this study explores the components of flower color, unravels the synthesis of anthocyanins and carotenoids, identifies regulatory factors, and highlights the prospects of rose breeding.

Abstract:: Petal blotch is a specific flower color pattern commonly found in angiosperm families. In particular, Rosa persica is characterized by dark red blotches at the base of yellow petals. Modern rose cultivars with blotches inherited the blotch trait from R. persica. Therefore, understanding the mechanism for blotch formation is crucial for breeding rose cultivars with various color patterns. In this study, the metabolites and genes responsible for the blotch formation in R. persica were identified for the first time through metabolomic and transcriptomic analyses using LC-MS/MS and RNA-seq. A total of 157 flavonoids were identified, with 7 anthocyanins as the major flavonoids, namely, cyanidin 3-O-(6′′-O-malonyl) glucoside 5-O-glucoside, cyanidin-3-O-glucoside, cyanidin 3-O-galactoside, cyanidin O-rutinoside-O-malonylglucoside, pelargonidin 3-O-glucoside, pelargonidin 3,5-O-diglucoside, and peonidin O-rutinoside-O-malonylglucoside, contributing to pigmentation and color darkening in the blotch parts of R. persica, whereas carotenoids predominantly influenced the color formation of non-blotch parts. Zeaxanthin and antheraxanthin mainly contributed to the yellow color formation of petals at the semi-open and full bloom stages. The expression levels of two 4-coumarate: CoA ligase genes (Rbe014123 and Rbe028518), the dihydroflavonol 4-reductase gene (Rbe013916), the anthocyanidin synthase gene (Rbe016466), and UDP-flavonoid glucosyltransferase gene (Rbe026328) indicated that they might be the key structural genes affecting the formation and color of petal blotch. Correlation analysis combined with weighted gene co-expression network analysis (WGCNA) further characterized 10 transcription factors (TFs). These TFs might participate in the regulation of anthocyanin accumulation in the blotch parts of petals by modulating one or more structural genes. Our results elucidate the compounds and molecular mechanisms underlying petal blotch formation in R. persica and provide valuable candidate genes for the future genetic improvement of rose cultivars with novel flower color patterns.

Abstract:Abstract Background Roses are famous ornamental plants worldwide. Floral coloration is one of the most prominent traits in roses and is mainly regulated through the anthocyanin biosynthetic pathway. In this study, we investigated the key genes and metabolites of the anthocyanin biosynthetic pathway involved in color mutation in miniature roses. A comparative metabolome and transcriptome analysis was carried out on the Neptune King rose and its color mutant, Queen rose, at the blooming stage. Neptune King rose has light pink colored petals while Queen rose has deep pink colored petals. Result A total of 190 flavonoid-related metabolites and 38,551 unique genes were identified. The contents of 45 flavonoid-related metabolites, and the expression of 15 genes participating in the flavonoid pathway, varied significantly between the two cultivars. Seven anthocyanins (cyanidin 3- O -glucosyl-malonylglucoside, cyanidin O -syringic acid, cyanidin 3- O -rutinoside, cyanidin 3- O -galactoside, cyanidin 3- O -glucoside, peonidin 3- O -glucoside chloride, and pelargonidin 3- O -glucoside) were found to be the major metabolites, with higher abundance in the Queen rose. Thirteen anthocyanin biosynthetic related genes showed an upregulation trend in the mutant flower, which may favor the higher levels of anthocyanins in the mutant. Besides, eight TRANSPARENT TESTA 12 genes were found upregulated in Queen rose, probably contributing to a high vacuolar sequestration of anthocyanins. Thirty transcription factors, including two MYB and one bHLH, were differentially expressed between the two cultivars. Conclusions This study provides important insights into major genes and metabolites of the anthocyanin biosynthetic pathway modulating flower coloration in miniature rose. The results will be conducive for manipulating the anthocyanin pathways in order to engineer novel miniature rose cultivars with specific colors.

Abstract:This study investigates the transition of Rosa canina L. petals from pink to white, driven by genetic and biochemical factors. It characterizes the expression of ten key genes involved in anthocyanin and flavonoid biosynthesis across five developmental stages, correlating gene expression with flavonoid and anthocyanin concentrations and colorimetric changes. Initially, the petals exhibit a rich flavonoid profile, dominated by Rutin and Kaempferol derivatives. The peak anthocyanin concentration, corresponding to the deepest color saturation, occurs in the subsequent stage. Advanced chromatographic analyses identify key flavonoids persisting into the final white petal stage. Notably, the ANS gene shows a dramatic 137.82-fold increase in expression at the final stage, indicating its crucial role in petal color maturation despite the absence of visible pigmentation. The study provides a comprehensive characterization of the genetic and biochemical mechanisms underlying petal pigmentation, suggesting that reduced anthocyanin synthesis and increased flavonol concentration led to white petals. It also highlights the roles of other genes such as PAL, CCD1, FLS, CHI, CHS, UFGT, F3H, DFR, and RhMYB1, indicating that post-translational modifications and other regulatory mechanisms may influence anthocyanin stability and degradation.

Abstract:Lycoris longituba is one of the species belonging to the Amaryllidaceae family. Despite its limited distribution, endemic to central eastern China, this species displays an exceptionally wide diversity of flower colors from purple, red, orange, to yellow, in nature. We study the natural variation of floral color in L. longituba by testing the components of water-soluble vacuolar pigments--anthocyanins--in its petals using high-performance liquid chromatography coupled with photodiode array detection and electrospray ionization mass spectrometry. Four anthocyanins were identified, cyanidin-3-sophoroside (Cy3So), cyanidin-3-xylosylglucoside (Cy3XyGlc), cyanidin-3-sambubioside (Cy3Sa), and pelargonidin-3-xylosylglucoside (Pg3XyGlc), which occur at various amounts in L. longituba petals of different colors. A multivariate analysis was used to explore the relationship between pigments and flower color. Anthocyanins have been thought to play a major role in acting as a UV screen that protects the plant's DNA from sunlight damage and attracting insects for the purpose of pollination. Thus, knowledge about the content and type of anthocyanins determining the petal coloration of Lycoris longituba will help to study the adaptive evolution of flowers and provide useful information for the ornamental breeding of this species.

Abstract:By use of the specific substrates, 7alpha-hydroxy-5beta-cholanic acid and 3alpha, 7alpha, 12alpha-trihydroxy-3,12-diacetyl-5beta-cholanic acid methyl ester, crude extracts of P. testosteroni grown on a steroid-containing medium have been shown to exhibit 7alpha-hydroxysteroid: NADP-oxidoreductase activity. The enzyme is highly specific for NADP. Both free and conjugated 7alpha-hydroxy bile acids can act as substrates, but those of low polarity (few hydroxyl groups) seem to be preferred, judging from initial reaction velocity studies. Optimal conditions appears to be at pH 8.5-9.5 and at 25 degress C. Free SH-groups are essential for maximum catalytic activity, since the enzyme is inhibited by SH-reacting substances such as p-chloromercuribenzoate and monoiodoacetic acid. Also the ketone-trapping agents hydrazine hydrate and semicarbazide act as inhibitors. Upon dilution, the storage stability is severely reduced, but this effect may be counteracted by the addition of glycerol at concentration of 20% or more. By gel filtration experiments on Sephadex G-100, the molecular weight was estimated to about 80,000.
Analysis of petal anthocyanins to investigate coloration mechanism in herbaceous peony cultivars

Abstract:Petal coloration and anthocyanin compositions of 41 herbaceous peony cultivars were analyzed. Anthocyanins were identified by high-performance liquid chromatography–electrospray ionization-mass spectrometry (HPLC–ESI-MSn) coupled with photodiode array detection (DAD). Peonidin-3,5-di-O-glucoside (Pn3G5G), pelargonidin-3,5-di-O-glucoside (Pg3G5G), cyanidin-3,5-di-O-glucoside (Cy3G5G), peonidin-3-O-glucoside (Pn3G), and cyanidin-3-O-glucoside (Cy3G) were the five major anthocyanins in herbaceous peony cultivars. Deep purple or reddish purple cultivars contained 4–5 anthocyanins, whereas pink cultivars only contained Cy3G5G and Pn3G5G, and their contents were much lower than those of purple cultivars. According to the chemical structures of three anthocyanidins in association with petal coloration, flowers were classified into three phenotypes: 1. “Pn, Cy, and Pg” (all purple flowers including two pink flowers); 2. “Pn, Cy” (pink flowers); 3 “Pn” (light pink and white flowers). The coloration mechanisms of cultivars with the pink and purple flowers were quite different. Correlations between lightness (L*) and chroma (C*), chromatic component a* and total anthocyanins (TA) value, a* and co-pigmentation index (CI) showed opposite tendencies, whereas L* and TA showed the same tendency in each group. High contents of Pn3G5G and Pg3G5G may responsible for the purple coloration of herbaceous peony cultivars.

Characterisation of Flower Colouration in 30 Rhododendron Species Via Anthocyanin and Flavonol Identification and Quantitative Traits

Abstract:Floral colour is a key reproductive character, often associated with environmental adaptation, and subject to human intervention. A large number of Rhododendron species differ widely in flower colour, providing a good model for flower colouration. The chromatic features and anthocyanin compositions of 30 species from seven subgenera were systematically analysed. The Royal Horticultural Society Colour Chartand CIE L*a*b* system were employed to describe and investigate flower colours. The UPLC-PDA/ESI-MSn system was used to identify and quantify anthocyanins in petal extracts. The flower colours of 30 Rhododendron species were categorised into four groups - red, purplish pink, purple and white. Seven anthocyanins were identified and quantified in petals: delphinidin, cyanidin and malvidin 3-O-arabinoside-5-O-glucosides, cyanidin 3,5-di-O-glucoside, 3-O-galactoside and 3-O-arabinoside, and delphinidin 3-O-glucoside. The red-flowered species mainly contained cyanidin monoglycosides and had much higher total anthocyanin content than purplish pink- and purple-flowered species. Purplish pink- and purple-flowered species had similar anthocyanin types and content. The chromatic differences were significant among groups, except the purplish pink and purple groups. Statistical analysis showed that Cy3Gal and Cy3Arb are characteristic for red-flowered species, and Mv3Arb5G and Dp3Arb5G play important roles in purple colouration; their contents were major components that greatly affected the chromatic parameters. In total, 21 flavonol derivates were identified. However, total flavonol content and co-pigmentation index showed no significant difference or correlation among/with colour groups, suggesting that flavonols might not play a major role in colouration. These results enhance our knowledge of the biochemical basis of flower colouration in Rhododendron species, and provide a foundation for genetic variation studies and aid in breeding cultivars with novel flower colours.
Traits and anthocyanin genetic characteristics of the progeny of crosses between white 2 azalea 'Rhododendron hybridum Hot' and pink Rhododendron pulchurum 'Wan Xia'

Abstract:Abstract Rhododendron is a world-famous ornamental flower with an abundance of genetic resources for producing different flower color varieties through breeding. However, the lack of genetic knowledge involved mechanisms behind the flower color formation have hindered the breeding process. In this study, a cross was made between the female pink Rhododendron pulchurum 'WanXia' and the male white 'Rhododendron hybridum Hot'. A total of 61 hybrid progenies with four different flower colors including white, pink, rose and red were produced and analyzed to identify the inheritance pattern of flower color as well as their genetic diversity in the whole group. The results of the combined qualitative and quantitative trait analysis disclosed that the germplasm resources in the hybrid progeny plants were genetically enriched, which can provide an excellent germplasm for screening and breeding new Rhododendron varieties. The anthocyanin-3-O-glucoside (Cy3G) contents of hybrid progeny petals were measured using high performance liquid chromatography-mass spectrometry (HPLC-MS). The total anthocyanin (TA) contents were calculated by first developing a linear equation (y=36.52x-59.18, R2=0.998) with anthocyanin-3-O-glucoside (Cy3G) specimens. The results revealed that TA was positively and significantly correlated with the color parameter a* values as the flower color of the hybrid progeny was gradually deepened from white to red. The value of parameter L* was gradually decreased, while a*, C* and TA contents were gradually increased. L* displayed to be both significantly and negatively correlated with a*, C*and TA contents. From the trait characteristics of each progeny plants, it was observed that No.52 Pink azalea has the highest pedicel length and petal number, while No.37 Red 2 azalea has the highest corolla lobe length, corolla lobe width and TA content. Our findings indicate that it may be feasible to breed Rhododendron varieties with new flower colors based on width and TA content. In conclusion, our outcomes propose that there is probability to produce Rhododendron varieties with new flower colors by crossing and screening hybrid progenies.
Light-mediated anthocyanin biosynthesis in rose petals involves a balanced regulatory module comprising transcription factors RhHY5, RhMYB114a, and RhMYB3b

Abstract:Roses are significant botanical species with both ornamental and economic value, displaying diverse floral traits, particularly an extensive array of petal colors. The red pigmentation of rose petals is predominantly attributed to anthocyanin accumulation. However, the underlying regulatory mechanism of anthocyanin biosynthesis in roses remains elusive. This study presents a novel light-responsive regulatory module governing anthocyanin biosynthesis in rose petals, which involves the transcription factors RhHY5, RhMYB114a, and RhMYB3b. Under light conditions (1000-1500 μmol m-2 s-1), RhHY5 represses RhMYB3b expression and induces RhMYB114a expression, positively regulating anthocyanin biosynthesis in rose petals. Notably, activation of anthocyanin structural genes probably involves an interaction and synergy between RhHY5 and the MYB114a-bHLH3-WD40 complex. Additionally, RhMYB3b is activated by RhMYB114a to prevent excessive accumulation of anthocyanin. Conversely, under low light conditions (<10 μmol m-2 s-1), the degradation of RhHY5 leads to down-regulation of RhMYB114a and up-regulation of RhMYB3b, which in turn inhibits the expression of both RhMYB114a and anthocyanin structural genes. Additionally, RhMYB3b competes with RhMYB114a for binding to RhbHLH3 and the promoters of anthocyanin-related structural genes. Overall, our study uncovers a complex light-mediated regulatory network that governs anthocyanin biosynthesis in rose petals, providing new insights into the molecular mechanisms underlying petal color formation in rose.

Abstract:Roses are consistently ranked at the forefront in cut flower production. Increasing demands of market and changing climate conditions have resulted in the need to further improve the diversity and quality of traits. However, frequent hybridization leads to highly heterozygous nature, including the allelic variants. Therefore, the absence of comprehensive genomic information leads to them making it challenging to molecular breeding. Here, two haplotype-resolved chromosome genomes for Rosa chinensis 'Chilong Hanzhu' (2n = 14) which is high heterozygous diploid old Chinese rose are generated. An amount of genetic variation (1,605,616 SNPs, 209,575 indels) is identified. 13,971 allelic genes show differential expression patterns between two haplotypes. Importantly, these differences hold valuable insights into regulatory mechanisms of traits. RcMYB114b can influence cyanidin-3-glucoside accumulation and the allelic variation in its promoter leads to differences in promoter activity, which as a factor control petal color. Moreover, gene family expansion may contribute to the abundance of terpenes in floral scents. Additionally, RcANT1, RcDA1, RcAG1 and RcSVP1 genes are involved in regulation of petal number and size under heat stress treatment. This study provides a foundation for molecular breeding to improve important characteristics of roses.
Rosa1, a Transposable Element-Like Insertion, Produces Red Petal Coloration in Rose Through Altering RcMYB114 Transcription

Abstract:Rose ( Rosa sp.) flowers have a rich diversity of colors resulting from the differential accumulation of anthocyanins, flavonols, and carotenoids. However, the genetic and molecular determinants of the red-petal trait in roses remains poorly understood. Here we report that a transposable element-like insertion (Rosa1) into RcMYB114 , a R2R3-MYB transcription factor’s promoter region causes its transcription, resulting in red petals. In red-petal varieties, RcMYB114 is expressed specifically in flower organs, but is absent from non-red varieties. Sequencing, yeast two-hybrid, transient transformation, and promoter activity assays of RcMYB114 independently confirmed the role of Rosa1 in altering RcMYB114 ’s transcription and downstream effects on flower color. Genetic and molecular evidence confirmed that the Rosa1 transposable element-like insertion, which is a previously unknown DNA transposable element, is different from those in other plants and is a reliable molecular marker to screen red-petal roses.

Abstract:Rhododendron dauricum is a cold-hardy shrub integrating ornamental, medicinal, and aromatic functions. Flower color is an important feature related to ornamental value for breeders and consumers. Nevertheless, the coloration mechanism of flower color in R. dauricum is still unclear. R. dauricum var. albiflorum is a white flower variety of R. dauricum. In this study, an integrative analysis of the metabolome and transcriptome was conducted between R. dauricum var. albiflorum and R. dauricum. A total of nine anthocyanins and two proanthocyanidins were differentially accumulated between R. dauricum var. albiflorum and R. dauricum. A decrease in malvidin-, delphinidin-, cyanidin-, peonidin-, and petunidin-based anthocyanins and an increase in procyanidin A2 and procyanidin B2 were responsible for the white flowers of R. dauricum var. albiflorum. Furthermore, a total of 4376 differentially expressed genes (DEGs) were identified using transcriptome sequencing. Integrated analysis of the metabolome and transcriptome showed that 21 DEGs encoding 9 enzymes (PAL, C4H, CHS, CHI, F3H, F3′5′H, DFR, ANR, and UFGT) were identified as structural genes involved in anthocyanin and proanthocyanidin biosynthesis, and 15 MYBs and 10 bHLHs were the transcriptional regulators of the anthocyanin biosynthesis pathways in R. dauricum var. albiflorum. Our results deepen the understanding of variations in azalea flower color, which is helpful for identifying important genes in the genetic engineering of azalea shrubs.

Abstract:Camellia japonica petals are colorful, rich in anthocyanins, and possess important ornamental, edible, and medicinal value. However, the regulatory mechanism of anthocyanin accumulation in C. japonica is still unclear. In this study, an integrative analysis of the metabolome and transcriptome was conducted in five C. japonica cultivars with different petal colors. Overall, a total of 187 flavonoids were identified (including 25 anthocyanins), and 11 anthocyanins were markedly differentially accumulated among these petals, contributing to the different petal colors in C. japonica . Moreover, cyanidin-3- O- (6 ′′ - O- malonyl) glucoside was confirmed as the main contributor to the red petal phenotype, while cyanidin-3- O- rutinoside, peonidin-3- O- glucoside, cyanidin-3- O- glucoside, and pelargonidin-3- O- glucoside were responsible for the deep coloration of the C. japonica petals. Furthermore, a total of 12,531 differentially expressed genes (DEGs) and overlapping DEGs (634 DEGs) were identified by RNA sequencing, and the correlation between the expression level of the DEGs and the anthocyanin content was explored. The candidate genes regulating anthocyanin accumulation in the C. japonica petals were identified and included 37 structural genes (especially CjANS and Cj4CL ), 18 keys differentially expressed transcription factors (such as GATA , MYB , bHLH , WRKY , and NAC ), and 16 other regulators (mainly including transporter proteins, zinc-finger proteins, and others). Our results provide new insights for elucidating the function of anthocyanins in C. japonica petal color expression.

Abstract:MicroRNA (miRNA)-mediated gene regulation is involved in various physiological processes in plants. Flower color is one of the vital ornamental traits of tree peony ( Paeonia suffruticosa Andr.). However, the yellow-flowered tree peony cultivars are particularly rare. To elucidate the miRNA-mediated gene regulatory mechanism underlying yellow pigmentation in tree peony, we combined pigment assessment, miRNA identification, expression analysis, and gene functional verification in two contrasting flower color cultivars “High Noon” and “Roufurong.” Flavones/flavonols and anthocyanins were found to be the main contributors to the coloration of “High Noon” and “Roufurong” petals, respectively. Subsequently, miRNA analysis based on available genome data identified 9 differentially expressed miRNAs and 12 relevant target genes implicated in flavonoid biosynthesis. Their dynamic expression patterns determined the key role of mdm-miR156b-PsSPL2 module in yellow pigmentation of tree peony flowers. The sequence analysis and subcellular localization validated that PsSPL2 might function as a nuclear-localized transcription factor. Overexpression of PsSPL2 in tobacco resulted in a decrease of anthocyanin content and down-regulation of NtF3′H and NtDFR transcripts. PsSPL2 -silenced petals exhibited lighter yellow color, and the contents of THC, Ap, and Ch decreased significantly. Meanwhile, expression levels of PsCHS , PsCHI , and PsF3H were significantly decreased in the petals with PsSPL2 silencing, while those of PsF3′H and PsDFR were remarkably increased. This study offers a novel insight into yellow pigmentation-related miRNA regulation network in tree peony, and further provides the valuable information on physiological changes during yellow coloring process of tree peony.

Abstract:Abstract Rhododendron( Rhododendron simsii Planch ) is one of the most diverse genera of woody plants in the world. It is rich in germplasm resources, but the molecular regulation of Rhododendron colour formation is poorly understood. In this study, two Belgian Rhododendron varieties with red and white flowers were subjected to RNA-sequencing and Protein sequencing analyses. Integrative analysis of transcriptome and proteome data was used to identify anthocyanin synthesis genes and proteins specifically expressed in flowers of different colours. The key differences between red and white flowers across flower development stages were analysed using the KEGG database, while the expression levels of differentially expressed genes in the anthocyanin/flavonoid biosynthesis pathways were compared using RNA-seq and qRT‒PCR data. The key pathways affecting flower colour in Rhododendron were identified by correlation analysis of the transcriptome data. We identified 6 anthocyanin biosynthesis-related genes. The differences in the gene sequences of RsCHS, RsCHI, RsF3H, RsFLS, RsDFR, and RsANS and cis-acting elements might be related to the accumulation of anthocyanins. RsDFR gene overexpression caused Arabidopsis leaves to turn red. These results provide valuable information on the molecular mechanism underlying Rhododendron flower colour formation.
The Identification of Flavonoids and the Expression of Genes of Anthocyanin Biosynthesis in the Chrysanthemum Flowers

Abstract:In order to provide additional information on the coloration of chrysanthemum flowers, the flavonoid composition and the expression of six structural genes involved in anthocyanin pathway in the ray florets of a pink flowering (cv. H5) and two white flowering (cvs. Keikai and Jinba) Chrysanthemum grandiflorum cultivars were examined. HPLCDAD/ESI-MSn analysis showed that cyanidin 3-O-(6″-O-malonylglucoside) and cyanidin 3-O-(3″,6″-O-dimalonylglucoside) were the two major flavonoids presented in H5, while white flowering cultivars contained flavones instead of anthocyanins. Nine flavone derivatives were detected in the three cultivars, the amount of each flavone varied upon cultivars, and seven of these were identified as luteolin 7-O-arabinosylglucuronide, apigenin 7-O-glucoside, luteolin 7-O-malonylglucoside, apigenin 7-O-malonylglucoside, chrysoeriol 7-O-malonylglucoside, acacetin 7-O-rutinoside and acacetin 7-O-malonylglucoside. The two white flowering cultivars showed similar total flavonoid content, which was about two fold higher than that in H5. A high expression of the genes encoding dihydroflavonol 4-reductase and 3-O-glucosyltransferase was detected only in H5 but not in Keikai or Jinba. Chalcone synthase, chalcone isomerase, flavanone 3-hydroxylase, and flavonoid 3′-hydroxylase were expressed in all flowers, suggesting that the lack of anthocyanin in white flowering cultivars cannot be due to any blockage of their expression.
