Abstract:Raw materials supplied as Aloe vera L. (sometimes referred to as Aloe barbadensis) samples often contain different composition of low and high molecular weight components when analyzed by size exclusion chromatography. One major reason for variable compositions of commercial A. vera L. materials is that they are produced by different manufacturing techniques. Consistent composition of matter based upon a given standard has been difficult to define. In addition, the method of quantifying and characterization of these commercially available materials has not been agreed upon within the industry. The end user, whether a researcher, a manufacturer, a marketing arm of industry or the consumer, should know that they are receiving a consistent product. A blind study of 32 various A. vera L. samples from different manufacturers, and a prepared sample of fresh A. vera L. gel with the commercial, biologic drug Acemannan Immunostimulanttrade mark, were analyzed for content of high molecular weight (polysaccharides) material by size exclusion chromatography with refractive index detection (SEC/RI) and SEC/RI coupled with multi-angle laser light scattering (MALLS) detection. Results from the SEC/RI analysis showed significant variation in the high molecular weight content, and the MALLS analysis also showed significant variation versus SEC/RI. In addition, HPLC analysis of the anthraquinone content showed that all samples contained significantly less than that of the raw, unwashed aloe gel. The variation of results from all analysis is attributed to differing methods in which the samples were processed by the different manufacturers.
Experimental investigation on the reliability of routine SEC–MALLS for the determination of absolute molecular weights in the oligomeric range

Abstract:Absolute number-average molecular weights were carefully measured for very-low to low molecular-weight poly(diisopropyl trimethylene-1,1-dicarboxylate) polymers that had been obtained from diisopropyl cyclopropane-1,1-dicarboxylate using a living anionic ring-opening polymerization technique (degree of polymerization in the range of 11–45 and polydispersity indices <1.13). Results obtained from four different analytical techniques, including end-group analysis (1H NMR), vapor pressure osmometry (VPO), size-exclusion chromatography coupled to a multi-angle laser light-scattering detector (SEC–MALLS), and matrix-assisted laser desorption ionization–time-of-flight mass spectrometry (MALDI–ToF), were compared and discussed. Although only crude estimates could be obtained by end-group analysis using 1H NMR (experimental errors of up to 20%), M̄n values estimated by SEC–MALLS were in perfect agreement with results obtained by VPO and MALDI–ToF. As the overall experimental protocol had been designed to prevent bias arising from some initial knowledge upon the exact molecular weights by the operator during the SEC–MALLS experiments, these results confirm a previous claim that SEC–MALLS is effective in measuring molecular weights in the oligomeric range.
Single-laboratory validation of an NMR method for the determination of aloe vera polysaccharide in pharmaceutical formulations

Abstract:This report presents a single-laboratory-validated NMR method for determining the quantity of aloe vera polysaccharide in product formulations. The ratio of signal intensities of the acetyl methyl protons to methyl protons of an internal reference varied linearly with concentration (r2 > 0.99) with a lower LOQ of 0.2 g/100 mL for two commercial aloe polysaccharide standards, Acemannan Hydrogel (AH) and Immuno-10 (I-10). The assay was used to quantify these standards in two nonacetylated polysaccharide matrices, dextrin and arabinogalactan, and in a pharmaceutical product. The concentrations of AH in samples containing the polysaccharide matrices agreed within 7% of values determined on the basis of weight and showed within- and between-run RSD values of < 3.5%. The assay of I-10 in the pharmaceutical product was within 7% of the expected values over a range from 50 to 125% of the targeted I-10 concentration, with a between-run RSD of < 7%. The assay showed no interference from other added polysaccharides or from other components of the pharmaceutical formulation and was independent of the molecular size distribution of the aloe polysaccharide present. The NMR assay can be used to validate aloe polysaccharide contained in a product and to follow any chemical degradation that may occur over time.
Quantitative colorimetric analysis of aloe polysaccharides as a measure of Aloe vera quality in commercial products

Abstract:Aloe vera inner leaf gel has been used as a medicinal remedy for many years. Yet some aloe products do not demonstrate beneficial effects, indicating that poor quality products are reaching the market. Therefore, an efficient and accurate method is needed to evaluate the quality of aloe products. This paper describes a quick, quantitative colorimetric assay that has been developed for the determination of glucomannan in aloe gel and products. With this method, interference by non-aloe polysaccharides or other extraneous components was absent or negligible. Data indicate that the glucomannan can be determined at parts per million (mg/L) in aqueous solutions with an accuracy of 100 +/- 5% at a 10 mg/L concentration. The correlation coefficient is 0.999, and linearity is from 0.9 to 72.7 mg/L in the test solution. The method is inexpensive, simple, sensitive, and reproducible. This method was applied to determine the polysaccharide content of commercial aloe products. Both qualitative and quantitative information can be obtained in about 5 min.
Comparative Studies on Three Dendrobium Medicinal Plants

Abstract:To investigate the diversities of polysaccharides content, distribution, composition and molecular mass between three medicinal aendrobium plants Dendrobium officinale Kimura et Migo, Dendrobium moniliforme (L.) Sw., Dendrobium hancockii Rolfe. Methods measureo polysaccharides content was used to analyze total polysaccharides(TPs)and water soluble polysaccharides(WPs)in different parts of the plants. Measured uronic acid and protein content, ion exchange column chromatography, and gel column chromatography were applied to determine WPs. The content of TPs and WPs extracted from Dendrobium officinale Kimura et Migo were highest.While the content of WPs from Dendrobium hancockii Rolfe was lower than that of the others. And polysaccharides content in different parts of the three medicinal plants is various. Moreover, the composition and molecular mass of WPs are distinct. In conclusion, differences of polysaccharides content, distribution, composition and molecular mass are evident between the three medicinal plants.
Chemical Investigation of Major Constituents in Aloe vera Leaves and Several Commercial Aloe Juice Powders

Abstract:Background: There are a substantial number of papers in the scientific literature reporting on the chemical composition of the Aloe vera plant. None of these investigations are truly comprehensive nor address the differences in composition that occur through processing variations in fresh leaves and commercially available product forms. Objectives: This work was to analytically examine a range of these forms and compile the findings. Methods: Fresh A. vera leaves and a number of commercial aloe juice powders were investigated for their major chemical constituents. Samples included fresh leaves from China and Mexico, plus commercial powders from different manufacturers made from different plant parts and/or manufacturing processes. The test results include moisture, ash, fiber, protein, lipids, minerals, organic acids, free sugars, and polysaccharides. The analytical methods employed comprise inductively coupled plasma-optical emission spectroscopy for minerals, high-performance anion-exchange chromatography equipped with pulsed amperometric detection for free sugars, HPLC for organic acids, and size exclusion chromatography (SEC)-multi-angle laser light scattering (MALS)-differential refractive index (dRI) for polysaccharide analyses. The absolute MW and MW distribution were determined using MALS measurement. Results: The major constituents of A. vera fresh leaf are fibers, proteins, organic acids, minerals, monosaccharides, and polysaccharides, which accounted for 85-95% of the total composition determined. In the commercial powdered aloe juice samples, four major components-organic acids, minerals, monosaccharides, and polysaccharides-accounted for 78-84% of the total composition. Apart from the four major components, products manufactured by ethanol precipitation contained high amounts of fiber and protein, while the free sugars were removed. In ethanol-precipitated products, the polysaccharide MW was less affected by manufacturing conditions and the concentration of aloe polysaccharides was higher than in products made in the nonethanol manufacturing processes. The overall chemical profiles were found to be consistent, except for the MW and content of polysaccharides in the commercial aloe samples analyzed, which were largely dependent on the types of manufacturing processes employed. Conclusions: This present study provides a comprehensive investigation of the major chemical composition of A. vera leaf and commercially derived products. The use of the SEC combined with MALS and differential RI detectors has proved to be an improved tool for the accurate determination of polysaccharide MW and contents of the various commercially available A. vera products in this study.

Abstract:Aqueous mode size exclusion chromatography (SEC) was employed for the analysis and construction of molecular weight (MW) calibration curves of three water-soluble polymers, namely, polyethylene glycol, polyethylene oxide, and polyacrylic acid sodium salt. Several calibration curves were obtained, varying chromatographic conditions such as columns arrangement, ionic strength, temperature and pH, in addition trends in polymeric chromatographic behavior were examined. The variation in SEC distribution coefficients at different temperatures was found to be below 10%, indicating that the studied polymers follow an ideal SEC mechanism under the tested conditions. Thus, differences in chromatographic behavior were ascribed to changes in polymer configuration induced by media and/or temperature. These variations in morphology were consistent with the observed SEC behavior. Regarding MW calibration, polynomial regression models ranging from first to fifth order were applied, and the most adequate ones were selected based on their fit and prediction capabilities. Third order polynomials were the preferred models for polyethylene glycol and polyacrylic acid sodium salt, independently of chromatographic conditions. Meanwhile for polyethylene oxide, either third or fifth-order polynomial models were optimal depending on the chromatographic conditions. All the selected regression models presented coefficients of multiple determination ( R 2 ) above 0.990, while achieving relative errors of prediction (REP%) in MW ranging from 0.3 to 4% for cross-validation.

Abstract:Aloe vera gel is a globally popular natural product used for the treatment of skin conditions. Its useful properties are attributed to the presence of bioactive polysaccharides. Nearly 25% of the 600 species in the genus Aloe are used locally in traditional medicine, indicating that the bioactive components in Aloe vera may be common across the genus Aloe. The complexity of the polysaccharides has hindered development of relevant assays for authentication of Aloe products. Carbohydrate detecting microarrays have recently been suggested as a method for profiling Aloe polysaccharide composition. The aim of this study was to use carbohydrate detecting microarrays to investigate the seasonal variation in the polysaccharide composition of two medicinal and two non-medicinal Aloe species over the course of a year. Microscopy was used to explore where in the cells the bioactive polysaccharides are present and predict their functional role in the cell wall structure. The carbohydrate detecting microarrays analyses showed distinctive differences in the polysaccharide composition between the different species and carbohydrate detecting microarrays therefore has potential as a complementary screening method directly targeting the presence and composition of relevant polysaccharides. The results also show changes in the polysaccharide composition over the year within the investigated species, which may be of importance for commercial growing in optimizing harvest times to obtain higher yield of relevant polysaccharides.

Abstract:ABSTRACT In traditional Chinese medicine research, seaweeds used for drug and health food development mainly refer to Sargassum pallidum (Turn.) C. Ag. (SP) and Sargassum fusiforme (Harv.) Setch. (SF). In the Chinese Pharmacopoeia, polysaccharides are the main quality control indicators. The total sugar content is determined by the anthrone–sulfuric acid method to evaluate the quality of seaweeds. However, this method cannot reflect the structural characteristics of seaweed polysaccharides and cannot distinguish among different seaweed varieties. Given this, to comprehensively evaluate the quality of different seaweeds, this study conducted a thorough analysis of the primary structure of polysaccharides in 11 batches of SF and 7 batches of SP. The neutral sugar content was determined by the anthrone–sulfuric acid colorimetric method, weight average molecular weight ( M w) by high‐performance size exclusion chromatography coupled with multi‐angle laser light scattering and refractive index detector method, sulfate content by ion chromatography and monosaccharide composition by ultrahigh‐performance liquid chromatography method, while also using professional software for statistical and similarity analysis. The results showed significant differences ( p < 0.05) in neutral sugar content and M w between S. fusiforme polysaccharides and S. pallidum polysaccharides, which can be used to distinguish SF and SP. The monosaccharide composition fingerprint was analyzed using chemometric methods, and it was found that fucose and glucose could serve as differential markers to distinguish SF and SP. This study further deepens the understanding of polysaccharides in seaweeds and more comprehensively evaluates the quality of different seaweeds.

Abstract:<p>Characterizing polysaccharides with large molecular weights and isomeric heterogeneity with mass spectrometry (MS) is generally difficult. In this work, we demonstrate how coupling size exclusion chromatography (SEC) and high-resolution MS with source-induced dissociation (SID) can be used for the separation and direct structural evaluation of intact polysaccharides. The analytical method was successfully developed using dextran standards up to 3755 kDa. This method was used to separate naturally occurring plant polysaccharides based on size, after which numerous polysaccharide fragments were identified from the resulting MS spectra. The results provided strong evidence for structural diversity, complexity, and heterogeneity among polysaccharides. MS showed superior sensitivity and reliability for the polysaccharides in eluted fractions when compared to a refractive index detector. Putative compositions for the fragments were proposed based on exact mass values. The work demonstrated that SEC-SID-MS is a feasible alternative for obtaining valuable structural information from the analysis of intact polysaccharides.</p>

Abstract:Plant polysaccharides have numerous medicinal functions. Due to the differences in their origins, regions of production, and cultivation conditions, the quality and the functions of polysaccharides can vary significantly. They are macromolecules with large molecular weight (MW) and complex structure, and pose great challenge for the analytical technology used. Taking Dendrobium officinale (DO) from various origins and locations as model samples. In this investigation, mechanochemical extraction method was used to successfully extract polysaccharides from DO using water as solvent, the process is simple, fast (40 s) and with high yield. The MWs of the intact saccharides from calibration curve and light scattering measurement were determined and compared after separation with size exclusion chromatography (SEC). The large polysaccharide was acid hydrolyzed to oligosaccharides and the products were efficiently separated and identified using liquid chromatography coupled to a high resolution tandem mass spectrometry (LC-MS2). Obvious differences were observed among LC-MS2 chromatograms of digested products, and the chemical structures for the products were proposed based on accurate mass values. More importantly, isomeric digested carbohydrate compounds were explored and characterized. All the chromatographic and mass spectrometric results in this study provided a multi-dimensional characterization, fingerprint analysis, and molecular structure level assessment of plant polysaccharides.
[Separation and characterization of Gastrodia elata polysaccharides based on asymmetrical flow field-flow fractionation: steric transition phenomenon]

Abstract:Asymmetrical flow field-flow fractionation (AF4), a gentle tool for the separation and characterization of particles and macromolecules, has attracted increased interest in recent years owing to its broad dynamic size range and utilization of "open channel" voids in the packing or stationary phase. A steric transition phenomenon in which the sample elution mode change from the normal mode to the steric/hyperlayer mode occurs. Accurate characterization by AF4 requires the absence of steric transition, particularly when the sample has a broad size distribution, because the effect of the combination of different modes is difficult to interpret. In this study, the relative molecular mass (M), radius of gyration (Rg), and conformation of Gastrodia elata polysaccharides (GEPs) were characterized using AF4 coupled with online multi-angle light scattering (MALS) and differential refractive index (dRI) detection (AF4-MALS-dRI). Steric transition was observed during GEP separation by AF4 owing to the broad size distribution of the molecules. This phenomenon would result in the inaccurate characterization of the GEPs in terms of M and Rg because two GEP groups of different sizes may elute together. In this study, the effects of constant and exponentially decaying cross-flow rates, sample mass concentration, and spacer thickness on steric transition were systematically investigated. The results indicated that a high GEP mass concentration (i. e., 0.75 mg/mL) can lead to steric transition. The spacer thickness affected the resolution and retention time of the GEPs and changed the steric transition point (di). An exponentially decaying cross-flow rate not only adjusted the di of the polydisperse GEP samples but also improved the GEP resolution and shortened the analysis time. The influence of steric transition was solved under the following operating conditions: injected GEP mass concentration=0.5 mg/mL; injection volume=50 μL; spacer thickness=350 μm; detector flow rate=1.0 mL/min; and cross-flow rate exponentially decayed from 0.2 to 0.05 mL/min with a half-life of 2 min. Moreover, the influence of GEP origins and ultrasound treatment time on the M and Rg distributions and conformation of GEPs were investigated under the optimized operating conditions. The results showed that the M and Rg distributions of Yunnan and Sichuan GEPs decreased with increasing ultrasound time. When the ultrasound treatment time was 15 min, the Yunnan GEPs had a loosely hyperbranched chain conformation, whereas the Sichuan GEPs had a spherical conformation. When the ultrasound treatment time was increased to 30 or 60 min, the GEPs from both Yunnan and Sichuan had a hyperbranched chain conformation, indicating that ultrasound treatment resulted in GEP degradation. Under the same extraction conditions, GEPs from Yunnan had larger M and Rg values than those from Sichuan. AF4-MALS-dRI showed good repeatability for the characterization of GEPs under the optimized operating conditions. The relative standard deviations of Rg and M were 0.5% and 1.7%, respectively. The data presented in this study can be used as a starting point for in-depth studies on the structural bioactivity of GEPs.
Determination of Polysaccharides in Undaria pinnatifida by Ionic Liquid-Modified Silica Gel Size Exclusion Chromatography

Abstract:Ionic liquid-modified silica particles with a large pore size were synthesized and used as the stationary phase for size exclusion chromatography coupled with a refractive index detector for the determination of polysaccharides, such as fucoidan, alginic acid, and laminarin from Undaria pinnatifida (seaweed). The molecular weight of polysaccharide was determined by a dextran standard curve (5–1100 kDa). The ionic liquid-modified silica column exhibited excellent size exclusion properties in separating laminarin from other components. The 1-butyl-3-methyl-imidazolium bis-(trifluoromethylsulfonyl)-amino silica column has superior resolution in laminarin separation than the other columns because the amino-group in ionic liquid provide π–π interactions due to aromaticity of the ring structure, which enhances the effect of the hydroxyl group in the target compound separation. The concentrations of polysaccharides were calculated using a standard linear equation to be 0.332–0.484 mg/g of fucoidan, 0.207–0.301 mg/g of alginic acid, and 0.154–0.297 mg/g of laminarin.

Abstract:Red seaweed carrageenans are frequently used in industry for its texturizing properties and have demonstrated antiviral activities that can be used in human medicine. However, their high viscosity, high molecular weight, and low skin penetration limit their use. Low-weight carrageenans have a reduced viscosity and molecular weight, enhancing their biological properties. In this study, ι-carrageenan from Solieria chordalis, extracted using hot water and dialyzed, was depolymerized using hydrogen peroxide and ultrasound. Ultrasonic depolymerization yielded fractions of average molecular weight (50 kDa) that were rich in sulfate groups (16% and 33%) compared to those from the hydrogen peroxide treatment (7 kDa, 6% and 9%). The potential bioactivity of the polysaccharides and low-molecular-weight (LMW) fractions were assessed using WST-1 and LDH assays for human fibroblast viability, proliferation, and cytotoxicity. The depolymerized fractions did not affect cell proliferation and were not cytotoxic. This research highlights the diversity in the biochemical composition and lack of cytotoxicity of Solieria chordalis polysaccharides and LMW fractions produced by a green (ultrasound) depolymerization method.

Abstract:Polyamic acid (PAA) is the precursor of polyimide (PI) prepared by a two-step method, its molecular weight and molecular weight distribution not only play an important role during the preparation and processing, but also influence the final performance of PI. However, precise characterization on molecular weight of PAA is still a challenge because of the existence of very complicated interactions in the solution system, including the electrostatic interaction, hydrogen bond interaction, dipole-dipole interaction, etc. Thus, it is necessary to establish a suitable strategy which can completely suppress these complex effects and get reasonable data on molecular weight. Herein, the gel permeation chromatography (GPC) coupled with differential refractive index (RI) and multi-angle laser light scattering (MALLS) detectors were applied to measure the molecular weight of (6FDA-DMB) PAA using different mobile phases, LiBr/DMF, LiBr/H3PO4/THF/DMF, LiBr/HAc/THF/DMF, and LiBr/HAc/DMF, respectively. It was found that combination of LiBr with HAc can shield the above-mentioned complex interactions and is more conducive to the separation of PAA than only addition of LiBr in DMF. LiBr/HAc/DMF was employed for the first time as a mild mobile phase to effectively separate PAA and determine its molecular weight. After a series of conditional experiments, 0.02 M LiBr/0.2 M HAc/DMF was fixed as an optimized mobile phase to measure the relative and absolute molecular weights of (6FDA-DMB) PAA prepared, and the obtained Mw from GPC-MALLS and GPC-RI were 35,300 g/mol and 125,000 g/mol, respectively. Abstract:A simple size-exclusion high-performance liquid chromatographic method was developed for rapid molecular mass screening to determine the average molecular mass, polydispersity, and quantity of Taizishen polysaccharides extract. The screening used a TOSOH TSK-GEL G3000SW high-performance liquid chromatographic column with 0.1 mol/L NaNO3 buffer as the mobile phase and refractive index detector. The molecular mass calibration curve was linear for polysaccharide standards from 10 to 100 kDa with a correlation coefficient of 0.9991. The method can be used to analyze the quantity, average molecular mass, and polydispersity of polysaccharides. In addition, the screening method is suitable for quality control of polysaccharide preparations in Chinese medicines and pharmaceutical products.

Abstract:Natural polysaccharides are important active biomolecules. However, the analysis and structural characterization of polysaccharides are challenging tasks that often require multiple techniques and maps to reflect their structural features. This study aimed to propose a new heart-cutting two-dimensional liquid chromatography (2D–LC) method for separating and analyzing polysaccharides to explore the multidimensional information of polysaccharide structure in a single map. That is, the first-dimension liquid chromatography (1D–LC) presents molecular-weight information, and the second-dimension liquid chromatography (2D–LC) shows the fingerprints of polysaccharides. In this 2D–LC system, the size-exclusion chromatography–hydrophilic interaction chromatography (SEC–HILIC) model was established. Coupling with a charged aerosol detector (CAD) eliminated the need for the derivatization of the polysaccharide sample, allowing the whole process to be completed within 80 min. The methods were all validated in terms of precision, linearity, stability, and repeatability. The capability of the new 2D-LC method was demonstrated in determining various species of natural polysaccharides. Our experimental data demonstrated the feasibility of the whole systematic approach, opening the door for further applications in the field of natural polysaccharide analysis.

Abstract:Natural polysaccharides exhibit a wide range of biological activities, which are closely related to their structural characteristics, including their molecular weight distribution, size, monosaccharide composition, glycosidic bond types and spatial conformation, etc. Size exclusion chromatography (SEC) and asymmetrical flow field-flow fractionation (AF4), as two potent separation techniques, both harbor potential for continuous development and enhancement. This manuscript reviewed the fundamental principles and separation applications of SEC and AF4. The structural information and spatial conformation of polysaccharides can be obtained using SEC or AF4 coupled with multiple detectors. In addition, this manuscript elaborates in detail on the shear degradation of samples such as polysaccharides separated by SEC. In addition, the abnormal elution that occurs during the application of the two methods is also discussed. Both SEC and AF4 possess considerable potential for ongoing development and refinement, thereby offering increased possibilities and opportunities for polysaccharide separation and characterization.

Abstract:With the deepening of polysaccharide research, whether the real polysaccharide structure can be clearly observed directly affects researchers' understanding of polysaccharide structure and function. Therefore, the establishment of a suitable method to study polysaccharide structure is urgent. Konjac glucomannan (KGM), agar, and curdlan were selected in this paper to investigate the effects of drying temperature, plasticizer type and dosage on the molecular structure of polysaccharides in the preparation of polysaccharide samples for AFM testing. The experimental results showed that the molecular morphology changes of different polysaccharides varied at different drying temperatures. Adding a certain dose of plasticizer could relieve the shrinkage of the hydrophilic polysaccharide molecular chain during the drying process. Compared with Tween 20 and Tween 80, glycerol showed a superior effect on relieving molecular chain shrinkage with the same addition amount. Finally, a new polysaccharide sample preparation method for AFM was proposed, and the feasibility of the method was confirmed, which provided a new idea for better use of AFM to observe the morphology of polysaccharides.

Abstract:Glycoside linkage analyses of medicine and food homologous plant polysaccharides have always been a key point and a difficulty of structural characterization. The gas chromatography-mass spectrometry (GC-MS) method is one of the commonly used traditional techniques to determine glycoside linkages via partially methylated alditol acetates and aldononitrile acetates (PMAAs and PMANs). Due to the simplicity of derivatization and the highly structural asymmetry of PMANs, reverse thinking is proposed using liquid chromatography-electrospray ionization-multiple reaction monitoring mass spectrometry (LC-ESI-MRM-MS) for the first time to directly determine the neutral and acidic glycosyl linkages of polysaccharides. The complete characterization of glycoside linkages deduced from PMANs was achieved using a combination of tR values, characteristic MRM ion pairs, diagnostic ESI+-MS/MS fragmentation ions (DFIs), and optimal collision energy (OCE). The DFI and OCE parameters were confirmed to be effective for the auxiliary discrimination of some isomers of the PMANs. The practicality of LC-ESI+-MRM-MS was further verified by analyzing the glycoside linkages of polysaccharides in five medicine and food homologous plants. This method can serve as an alternative to GC-MS for the simultaneous determination of neutral and acidic glycosyl linkages in polysaccharides.
