Abstract:Riparian soils are an important source of dissolved organic matter (DOM) to their connected rivers. The transport of DOM from riparian soils to rivers exerts a significant impact on both terrestrial and aquatic organic matter cycles. However, few studies focused on the underlying changes in DOM composition and potential biogeochemical processes involved. By combining optical techniques and Fourier transform ion cyclotron mass spectrometry (FT-ICR MS), here we show the variation in molecular composition between DOM in the riparian soil (S-DOM), submerged soil (SS-DOM), and river water (R-DOM) along a typical tributary of the Yangtze River. Variations in relative inputs of humic- and aromatic-like sourced DOM (R-DOM > S-DOM > SS-DOM) and protein-like sourced DOM (SS-DOM > S-DOM > R-DOM) were observed at both optical and molecular levels, indicating significant alterations in DOM composition during its transport. In particular, we have identified two preliminary mechanisms of DOM transport from soils to rivers at the molecular level by FT-ICR MS: (i) lower molecular weight (MW) (344 +/- 9 Da in average) and moderate aromatic (modified aromaticity index; AI(mod): 0.28 +/- 0.01 in average) DOM compounds are released to the river without modifications; (ii) moderate MW (370 +/- 1 Da in average) and less aromatic (AI(mod) : 0.25 +/- 0.01 in average) DOM compounds are produced (likely due to transformation or degradation of higher MW and aromatic DOM) and released to the river. Further incubation experiments suggested that DOM compounds associated with the first mechanism were more refractory (both bio- and photoresistant) than those of the second one. Therefore, we speculate that the first mechanism likely relates to the DOM transportation to the downstream, but the second mechanism mainly contributes to the in situ CO2 emissions in rivers. Our results (i) highlight the variation in DOM composition across the soil-river interface; (ii) confirm the preferential mobilization of specific DOM compounds from soils to rivers; and (iii) provide an avenue for further investigation of the mechanisms responsible for the observed changes. In particular, further studies are encouraged to investigate the spatial and temporal dynamics of DOM transport along the terrestrial-aquatic-continuum with different sources or composition of organic matter and under different hydrological scenarios.

Abstract:Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR MS) has been widely applied to characterize the molecular composition of dissolved organic matter (DOM) in different ecosystems. Most previous studies have explored the molecular composition of DOM focused on one or a few ecosystems, which prevents us from tracing the molecular composition of DOM from different sources and further exploring its biogeochemical cycling across ecosystems. In this study, a total of 67 DOM samples, including soil, lake, river, ocean, and groundwater, were analyzed by negative-ion electrospray ionization FT-ICR MS. Results show that molecular composition of DOM varies dramatically among diverse ecosystems. Specifically, the forest soil DOM exhibited the strongest terrestrial signature of molecules, while the seawater DOM showed the most abundant of biologically recalcitrant components, for example, the carboxyl-rich alicyclic molecules were abundant in the deep-sea waters. Terrigenous organic matter is gradually degraded during its transport along the river-estuary-ocean continuum. The saline lake DOM showed similar DOM characteristics with marine DOM, and sequestrated abundant recalcitrant DOM. By comparing these DOM extracts, we found that human activities likely lead to an increase in the content of S and N-containing heteroatoms in DOM, this phenomenon was commonly found in the paddy soil, polluted river, eutrophic lake, and acid mine drainage DOM samples. Overall, this study compared molecular composition of DOM extracted from various ecosystems, providing a preliminary comparison on the DOM fingerprint and an angle of view into biogeochemical cycling across different ecosystems. We thus advocate for the development of a comprehensive molecular fingerprint database of DOM using FT-ICR MS across a wider range of ecosystems. This will enable us to better understand the generalizability of the distinct features among ecosystems.

Abstract:With the on-going boom in the construction of dam reservoirs all over the world, the sources and composition of dissolved organic matter (DOM) in fluvial networks are expected to be altered. Considering the importance of DOM as a key biogeochemical component in inland waters, this might bring important ecological and environmental influences. However, limited information is available on the molecular composition of DOM in dam reservoirs. In this study, the spatial characteristics of DOM composition were investigated in Xiangxi tributary and mainstream of the Three Gorges Reservoir (TGR), the largest freshwater reservoir in the world. The concentration alteration of conservative cations revealed the water intrusion from mainstream into Xiangxi tributary, which mainly controlled the hydrological gradient. One tyrosine-like (C4), one tryptophan-like (C2), and two humic-like (C1 and C3) fluorescent components were identified in fluorescent DOM (FDOM) by parallel factor analysis (PAFACAC), potentially indicating algal, anthropogenic, and terrestrial inputs, respectively. Decreasing trends of C1, C3 and C4 components and an increasing trend of C2 component were observed from Xiangxi tributary to mainstream, indicating higher terrestrial and algal inputs but lower anthropogenic inputs in Xiangxi tributary compared to mainstream. The Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) characterization further revealed substantial heterogeneity of DOM at the molecular level. Interestingly, S-containing compounds related to synthetic surfactants were consistently detected, and their relative abundances showed an increasing trend from Xiangxi tributary to mainstream, in agreement with the distribution of the anthropogenic derived C2 component. Meanwhile, numerous lignin-like S-containing compounds were identified, likely the result of the incorporation of sulfide ions to lignin-like CHO compounds. This study represents the first molecular level characterization of DOM in the TGR system, which should aid the design and implementation of more detailed future studies.
Organic matter in geothermal springs and its association with the microbial community

Abstract:Organic matter (OM) plays an important role in the biogeochemical cycles of carbon, nitrogen, and other elements, shaping the structure of the microbiome and vice versa. However, the molecular composition of OM and its impact on the microbial community in terrestrial geothermal environments remain unclear. In this study, we characterized the OM in water and sediment from a typical geothermal field using ultra-high-resolution Fourier transform ion cyclotron resonance mass spectrometry. By combining high-throughput amplicon sequencing and multivariate analyses, we deciphered the association between OM components and microbial community. A surprisingly high chemodiversity of OM was observed in the waters (11,088 compounds) and sediments (7772 compounds) in geothermal springs. Sulfur-containing organic compounds, a characteristic molecular signature of geothermal springs, accounted for 21 % ± 5 % in waters and 33 % ± 4 % in sediments. Multivariate analyses revealed that both labile and recalcitrant fractions of OM (e.g., carbohydrates intensity and tannins chemodiversity) influenced the structure and function of the microbial community. Co-occurrence networks showed that Proteobacteria and Crenarchaeota accounted for most of the connections with OM in waters (33 % and 15 %, respectively) and sediments (15 % and 12 %, respectively), highlighting their key roles in carbon cycling. This study expands our understanding of the molecular compositions of OM in geothermal springs and highlights its potentially important role in global climate change through microbial carbon cycling.

Abstract:Understanding the degradation of dissolved organic matter (DOM) is vital for optimizing DOM control. However, the microbe-mediated DOM transformation during wastewater treatment remains poorly characterized. Here, microbes and DOM along full-scale biotreatment processes were simultaneously characterized using comparative genomics and high-resolution mass spectrometry-based reactomics. Biotreatments significantly increased DOM's aromaticity and unsaturation due to the overproduced lignin and polyphenol analogs. DOM was diversified by over five times in richness, with thousands of nitrogenous and sulfur-containing compounds generated through microbe-mediated oxidoreduction, functional group transfer, and C-N and C-S bond formation. Network analysis demonstrated microbial division of labor in DOM transformation. However, their roles were determined by their functional traits rather than taxa. Specifically, network and module hubs exhibited rapid growth potentials and broad substrate affinities but were deficient in xenobiotics- metabolism-associated genes. They were mainly correlated to liable DOM consumption and its transformation to recalcitrant compounds. In contrast, connectors and peripherals were potential degraders of recalcitrant DOM but slow in growth. They showed specialized associations with fewer DOM molecules and probably fed on metabolites of hub microbes. Thus, developing technologies (e.g., carriers) to selectively enrich peripheral degraders and consequently decouple the liable and recalcitrant DOM transformation processes may advance DOM removal.

Abstract:We recently proposed an eight-parameter model of the respiratory system to account for its mechanical behavior when flow is interrupted during passive expiration. The model consists of two four-parameter submodels representing the lungs and the chest wall, respectively. The lung submodel consists of an airways resistance together with elements embodying the viscoelastic properties of the lung tissues. The chest wall submodel has similar structure. We estimated the parameters of the model from data obtained in four normal, anesthetized, paralyzed, tracheostomized mongrel dogs. This model explains why lung tissue and chest wall resistances should be markedly frequency dependent at low frequencies and also permits a physiological interpretation of resistance measurements provided by the flow interruption method.
Insight into spatial variations of DOM fractions and its interactions with microbial communities of shallow groundwater in a mesoscale lowland river watershed

Abstract:Dissolved organic matter (DOM) plays a crucial role in driving biogeochemical processes and determining water quality in shallow groundwater systems, where DOM could be susceptible to dynamic influences of surface water influx. This study employed fluorescence excitation-emission matrix (EEM) spectroscopy combined with principal component coefficients, parallel factor analysis (PARAFAC), co-occurrence network analysis and structural equation modeling (SEM) to examine changes of DOM fractions from surface water to shallow groundwater in a mesoscale lowland river basin. Combining stable isotope and hydrochemical parameters, except for surface water (SW), two groups of groundwater samples were defined, namely, deeply influenced by surface water (IGW) and groundwater nearly non-influenced by surface water (UGW), which were 50.34 % and 19.39 % recharged by surface water, respectively. According to principal component coefficients, reassembled EEM data of these categories highlighted variations of the tyrosine-like peak in DOM. EEMs coupled with PARAFAC extracted five components (C1-C5), i.e. C1, protein-like substances, C2 and C4, humic-like substances, and C3 and C5, microbial-related substances. The abundance of the protein-like was SW > IGW > UGW, while the order of the humic-like was opposite. The bacterial communities exhibited an obvious cluster across three regions, which hinted their sensitivity to variations in environmental conditions. Based on co-occurrence, SW represented the highest connectivity between bacterial OTUs and DOM fractions, followed by IGW and UGW. SEM revealed that microbial activities increased bioavailability of the humic-like in the SW and IGW, whereas microbial compositions promoted the evolution of humic-like substances in the UGW. Generally, these results could be conducive to discern dissimilarity in DOM fractions across surface water and shallow groundwater, and further trace their interactions in the river watershed.

Abstract:<h3 class="a-plus-plus">Abstract</h3> Dynamics of dissolved organic matter (DOM) in ecosystems are controlled by a suite of interacting physical, chemical, and biological factors. Growing recognition of the associations between microbial communities and metabolism and intrinsic DOM characteristics, highlight the potential importance of microbe-DOM relationships to modulate the role and fate of DOM, yet these relationships are difficult to isolate because they often operate across confounding environmental gradients. In a controlled laboratory incubation (44 days), we integrated DOM bulk and molecular characterization, bacterial abundances, microbial assemblage composition, nutrient concentrations, and cellular respiration to discern the structural dynamics of biological processing among DOM sources from different allochthonous litters (grass, deciduous leaves, and evergreen needles). We identified two periods, consistent among DOM sources, where processing dynamics differed. Further, bulk fluorescent analyses showed shifts from low to high excitation and emission wavelengths, indicating the biological production of more complex/degraded materials over time. Molecular level analyses revealed similar temporal patterns among DOM sources in the production and consumption of individual chemical components varying in reactivity and heteroatomic content. Despite these similarities, total carbon (C) removed and carbon dioxide (CO2) accumulation differed by ~ 20% and 25% among DOM sources. This range in C processing was apparently tied to key chemical properties of the DOM (e.g., initial DOM composition, N content, and labile nature) as well as differential reorganization of the microbial populations that decomposed the DOM. We conclude that the production, transformation, and consumption of C in aquatic ecosystems is strongly dependent on the source and character of DOM as well as the structure of the microbial communities present, both of which change as DOM is processed over time. It is crucial that stream C processing models represent this complexity accurately.

Abstract:Microbes play a critical role in regulating the size, composition, and turnover of dissolved organic matter (DOM), which is one of the largest pools of carbon in aquatic ecosystems. Global change may alter DOM-microbe associations with implications for biogeochemical cycles, although disentangling these complex interactions remains a major challenge. Here we develop a framework called Energy-Diversity-Trait integrative Analysis (EDTiA) to examine the associations between DOM and bacteria along temperature and nutrient gradients in a manipulative field experiment on mountainsides in contrasting subarctic and subtropical climates. In both study regions, the chemical composition of DOM correlated with bacterial communities, and was primarily controlled by nutrients and to a lesser degree by temperature. At a molecular-level, DOM-bacteria associations depended strongly on the molecular traits of DOM, with negative associations indicative of decomposition as molecules are more biolabile. Using bipartite networks, we further demonstrated that negative associations were more specialized than positive associations indicative of DOM production. Nutrient enrichment promoted specialization of positive associations, but decreased specialization of negative associations particularly at warmer temperatures in subtropical climate. These global change drivers influenced specialization of negative associations most strongly via molecular traits, while both molecular traits and bacterial diversity similarly affected positive associations. Together, our framework provides a quantitative approach to understand DOM-microbe associations and wider carbon cycling across scales under global change. ### Competing Interest Statement The authors have declared no competing interest.
Influence of Ce Content on the Mechanical Properties of Sintered (Ce, Nd)–Fe–B Magnets

Abstract:The (R,Ce)–Fe–B magnets have been successfully industrialized in recent years. The mechanical property of sintered permanent magnets is one important aspect of their comprehensive performances, which directly influences the service reliability and the production cost. In this paper, the bending strength, fracture toughness, Vickers hardness, and brittleness index of commercial (R1–<italic>x</italic>Ce<italic>x</italic>)30.5–31.5Febal.B1M1 magnets with different cerium contents have been investigated. The micro-fractures of the magnets were observed by a scanning electron microscopy equipped with energy dispersive X-ray analysis system. It shows that the bending strength and the fracture toughness of (R,Ce)–Fe–B magnets have a downward tendency with increasing Ce content x, while the Vickers hardness of the magnets varies irregularly with Ce contents in this paper. The optimum mechanical properties have been obtained in the (R1–<italic>x</italic>Ce<italic>x</italic>)30.5–31.5Febal.B1M1 magnet with <inline-formula> <tex-math notation="LaTeX">$x = 0.15$ </tex-math></inline-formula>; the bending strength, fracture toughness and brittleness index of the magnet with <inline-formula> <tex-math notation="LaTeX">$x =0.15$ </tex-math></inline-formula> are obviously superior to those of the ordinary sintered Nd–Fe–B magnets. Some flocculent oxide phases have been discovered in the (R,Ce)–Fe–B magnet with <inline-formula> <tex-math notation="LaTeX">$x = 0.15$ </tex-math></inline-formula>. The flocculent phases may absorb the crack propagating energy, and reduce the stress concentration at a crack tip, which is beneficial to strengthening and toughening of (R,Ce)–Fe–B magnets. However, the mechanical properties are obviously worse for the magnet with <inline-formula> <tex-math notation="LaTeX">$x = 0.45$ </tex-math></inline-formula>(Ce/<inline-formula> <tex-math notation="LaTeX">$\sum $ </tex-math></inline-formula>RE = 45%). That is probably because the microstructures of the magnet with <inline-formula> <tex-math notation="LaTeX">$x = 0.45$ </tex-math></inline-formula> become deteriorated, in which abnormally large grains have been observed.
Microbial degradation of various types of dissolved organic matter in aquatic ecosystems and its influencing factors

Abstract:Interactions between chemodiverse dissolved organic matter (DOM) and biodiverse microbes are governed by a myriad of intrinsic and extrinsic factors which are not well understood. Here, we update and bridge the gap of this interdisciplinary theme comprehensively. At an ecosystem level, aquatic ecosystems dominated by algae-sourced DOM (e.g., eutrophic lake or coastal upwelling areas) harbor more biolabile DOM, such as directly assimilable monomers and readily hydrolysable biopolymers. However, other ecosystems prevailed by DOM supply from soil and vascular plants (e.g., river or wetland) have more biorefractory DOM, such as low molecular weight (LMW) residue of aliphatic C skeletons and geopolymers. A variety of heterotrophic bacteria, archaea, fungi, phagotrophic protists, and even photoautotrophic phytoplankton shows genomic and/or culturing experimental evidence of being able to process a diverse type of organics. The various biodegradable organics have different chemical structures and chemical bonds such as carbohydrates, amino acids, proteins, lignins, lipids, carboxylic acids, humic acids, hydrocarbons, and nanoplastics. Meanwhile, bio-production of metabolism intermediates and/or biorefractory organics (e.g., carboxyl-rich alicyclic molecules, CRAM) is observed despite general decay of bulk dissolved organic carbon (DOC) during bioassay experiments. In particular, emerging evidence shows that archaea contribute significantly to biomass in the marine mesopelagic zone and subsurface environments and their abundance often increases with depth in sediments. Furthermore, not only intrinsic factors (e.g., DOM composition and structure), but also extrinsic ones (e.g., sunlight and dissolved oxygen) play important roles in interplays between DOM and microbes.

Abstract:The mineralization of dissolved organic matter (DOM) in sediments is an important factor leading to the eutrophication of macrophyte-dominated lakes. However, the changes in the molecular characteristics of sediment-derived DOM during microbial degradation in macrophyte-dominated lakes are not well understood. In this study, the microbial degradation process of sediment-derived DOM in Lake Caohai under aerobic and hypoxic conditions was investigated using Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) and metagenomics . The results revealed that the microbial degradation of sediment-derived DOM in macrophyte-dominated lakes was more intense under aerobic conditions. The microorganisms mainly metabolized the protein-like substances in the macrophyte-dominated lakes, and the carbohydrate-active enzyme genes and protein/lipid-like degradation genes played key roles in sediment-derived DOM degradation. Organic compounds with high H/C ratios such as lipids, carbohydrates, and protein/lipid-like compounds were preferentially removed by microorganisms during microbial degradation. Meanwhile, there was an increase in the abundance of organic molecular formula with a high aromaticity such as tannins and unsaturated hydrocarbons with low molecular weight and low double bond equivalent. In addition, aerobic/hypoxic environments can alter microbial metabolic pathways of sediment-derived DOM by affecting the relative abundance of microbial communities (e.g., Gemmatimonadetes and Acidobacteria) and functional genes (e.g., ABC.PE.P1 and ABC.PE.P) in macrophyte-dominated lakes. The abundances of lipids, unsaturated hydrocarbons, and protein compounds in aerobic environments decreased by 58 %, 50 %, and 44 %, respectively, compared to in hypoxic environments under microbial degradation. The results of this study deepen our understanding of DOM biodegradation in macrophyte-dominated lakes under different redox environments and provide new insights into nutrients releases from sediment and continuing eutrophication in macrophyte-dominated lakes.

Soil DOM molecules were characterized by Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR MS) and the results showed that the soil DOM consisted mainly of lignin/carboxylic rich alicyclic molecule (CRAM)-like structures, while the ratios of lipids and aliphatic/protein decreased in sequence with recovery time. Lipids and aliphatic/proteins with high H/C DOM (labile DOM) degrade preferentially, while lignin/CRAM-like structures and tannins with low H/C DOM (recalcitrant DOM) are recalcitrant during vegetation restoration. With the restoration of vegetation, DOM molecules tend to be diversified and complicated, and DOM compounds with low double bond equivalent (DBE), low aromatic, and low alkyl structures will be converted into persistent organic matter with high carbon numbers and high DBE. The diversity of soil microorganisms was determined by high-throughput sequencing. The results showed that the abundance and diversity of soil bacteria increased significantly after revegetation, while the abundance and diversity of soil fungi began to increase when the ecosystem became a more mature coniferous forest. The soil microbial community exhibited complex connectivity and strong interaction with DOM molecules during vegetation restoration. As most of the DOM molecules are recalcitrant, vegetation restoration facilitates C sequestration in the soil, thereby contributing to climate change mitigation.

Abstract:The influence of macroalgae cultivation on aquaculture carbon sinks is significant, with microbial carbon (C) pumps contributing to a stable inert dissolved carbon pool in this context. Concurrently, dissolved organic matter (DOM) exchange at the marine sediment-water interface profoundly affects global ecosystem element cycling. However, the interactions between DOM and bacterial communities at the sediment-water interface in kelp cultivation areas, especially regarding microbial function prediction, have not been fully explored. This study analyzed the DOM characteristics, environmental factors, and bacterial community structure in the Tahewan kelp--Saccharina japonica cultivated area and compared them with those in non-cultivated areas. The results indicated significantly higher dissolved organic carbon (DOC) concentrations in the kelp culture area, particularly in surface seawater and overlying water. The dominant bacterial phyla in both regions included Pseudomonadota, Actinomycetota, and Bacteroidota in both regions, while Desulfobacterota was more prevalent in the sediment environment of the cultivated region. Parallel factor analysis (EEM-PARAFAC) was used to identify DOM components, among which component C2 (a microbial humic-like substance DOM) was highly resistant to microbial degradation. We infer that C2 has similar properties to recalcitrant dissolved organic matter (RDOM). Analysis of the predicted functional genes based on 16S rRNA gene data showed that methanol oxidation, methylotrophy, and methanotrophy were significant in the bottom seawater of the cultivation area. The carbon (C), nitrogen (N), and sulfur (S) cycle functional genes in the sediment environment of the kelp cultivation area were more active than those in other areas, especially in which sulfate reduction and denitrification were the two main processes. Furthermore, a DOM priming effect was identified in the cultivated sediment environment, where kelp-released labile dissolved organic matter (LDOM) stimulates rapid degradation of the original RDOM, potentially enhancing C sequestration.

Abstract:The production of refractory dissolved organic matter (RDOM) is complex and closely related to microbial consortia in aquatic ecosystems; however, it is still unclear how microorganisms regulate the production of RDOM and its molecular composition in inland waters. Therefore, we conducted a large-scale survey of inland waters and analyzed the optical and mass spectrometric characteristics of DOM, the microbial community and functional genes, as well as related environmental parameters, to understand the abovementioned issues. Here, the RDOM production was found mainly regulated by microbial (e.g., phylogeny and community assembly) rather than other environmental factors in inland waters. Biostatistical analyses and carbon isotopic evidence indicated that the successive microbial processing from labile DOM to RDOM (i.e., carboxyl-rich alicyclic molecules, CRAMs) was widely present in inland waters, involving the microbially mediated carbon skeleton turnover and heteroatom conversion. There was a significant empirical relationship between CRAMs and the ratio of Proteobacteria to Actinobacteria, highlighting the intraspecific interaction of bacteria more important than other microbial groups (i.e., archaea, eukaryotes, and fungi) for the RDOM production. This study demonstrated a fundamental role of microbial regulation in RDOM production within the inland waters, thereby facilitating future estimation of carbon sequestration potential in inland aquatic ecosystems.

Abstract:Controlled surface water systems, including those with dams lead to dynamic stage changes that alter the fluctuation directions of flow exchange in the hyporheic zones (HZ). However, the nitrogen transformation, dissolved organic matter (DOM) composition, and microbial community responding to variable scenarios of water source and hyporheic exchange are poorly studied. The present work investigated nitrogen transformation in HZ sediments, focusing on how microbial community structure and biological functions related to nitrogen transformation and sediment-attached DOM compositions. Upwelling of synthesized groundwater, downwelling of synthesized river water and exchangeable elution of both feed water created distinct microbial zonation and N-transformation processes. Mixing of river water and groundwater enhanced microbial diversity, microbial co-occurrence network complexity and N-transformation functions. In terms of the sediment-attached DOM properties after hyporheic exchanges, humic fractions occupied the predominant chromophoric DOM. Correlation analysis implied that there were more DOM properties, e.g., tryptophan-like proteins, humic-like fractions, and the source of humic fractions, involved in affecting the microbial community under downwelling flow. Co-occurrence network analysis verified that fluorescent components, protein-like and lignin-like fractions in sediment-detached DOM were clustered with microbial communities in one module in downwelling column, implying closer interactions among microbial communities and DOM fractions. The strains of Nitrospinae, Dinghuibacter, and Lentimicrobium etc. were key species collaborating to metabolize both nitrogen and DOM in HZ sediments. The work provides insights into how the nitrogen transformation, DOM compositional changes, as well as the linkages between community structure and DOM factions, response to the changes in water chemistry, leading to valuable insights into hyporheic zone functions.

Abstract:Dissolved organic matter (DOM) play critical roles in arsenic (As) biotransformation in groundwater, but its compositional characteristics and interactions with indigenous microbial communities remain unclear. In this study, DOM signatures coupled with taxonomy and functions of microbial community were characterized in As-enriched groundwater by excitation-emission matrix, Fourier transform ion cyclotron resonance mass spectrometry and metagenomic sequencing. Results showed that As concentrations were significantly positively correlated with DOM humification (r = 0.707, p < 0.01) and the most dominant humic acid-like DOM components (r = 0.789, p < 0.01). Molecular characterization further demonstrated high DOM oxidation degree, with the prevalence of unsaturated oxygen-low aromatics, nitrogen (N 1 /N 2 )-containing compounds and unique CHO molecules in high As groundwater. These DOM properties were consistent with microbial composition and functional potentials. Both taxonomy and binning analyses demonstrated the dominance of Pseudomonas stutzeri , Microbacterium and Sphingobium xenophagum in As-enriched groundwater which possessed abundant As-reducing gene, with organic carbon degrading genes capable of labile to recalcitrant compounds degradation and high potentials of organic nitrogen mineralization to generate ammonium. Besides, most assembled bins in high As groundwater presented strong fermentation potentials which could facilitate carbon utilization by heterotrophic microbes. This study provides better insight into the potential role of DOM mineralization for As release in groundwater system.

Abstract:Microbially-mediated transformations of dissolved organic matter (DOM) in a marsh-dominated estuarine system were investigated at the molecular level using ultrahigh resolution mass spectrometry. In addition to observing spatial and temporal variability in DOM sources in the estuary, multiple incubations with endogenous microorganisms identified the influence of DOM composition on biodegradation. A clear microbial preference for degradation of compounds associated with marine DOM relative to those of terrestrial origin was observed, resulting in an overall shift of the remaining DOM toward a stronger terrigenous signature. During short, 1-day long incubations of samples rich in marine DOM, the molecular formulae that were enriched had slightly smaller mass (20–30 Da) and number of carbon atoms compared to the molecular formulae that were depleted. Over longer time scales (70 days), the mean differences in molecular mass between formulae that were depleted and enriched were substantially larger (~270 Da). The differences in elemental composition over daily time scales were consistent with transformations in functional groups; over longer time scales, the differences in elemental composition may be related to progressive transformations of functional groups of intermediate products and/or other reactions. Our results infused new data toward the understanding of DOM processing by bacterioplankton in estuarine systems.

Abstract:Abstract Soils are losing increasing amounts of carbon annually to freshwaters as dissolved organic matter (DOM), which, if degraded, can offset their carbon sink capacity. However, the processes underlying DOM degradation across environments are poorly understood. Here we show DOM changes similarly along soil-aquatic gradients irrespective of environmental differences. Using ultrahigh-resolution mass spectrometry, we track DOM along soil depths and hillslope positions in forest catchments and relate its composition to soil microbiomes and physico-chemical conditions. Along depths and hillslopes, we find carbohydrate-like and unsaturated hydrocarbon-like compounds increase in abundance-weighted mass, and the expression of genes essential for degrading plant-derived carbohydrates explains >50% of the variation in abundance of these compounds. These results suggest that microbes transform plant-derived compounds, leaving DOM to become increasingly dominated by the same (i.e., universal), difficult-to-degrade compounds as degradation proceeds. By synthesising data from the land-to-ocean continuum, we suggest these processes generalise across ecosystems and spatiotemporal scales. Such general degradation patterns can help predict DOM composition and reactivity along environmental gradients to inform management of soil-to-stream carbon losses.
