Abstract:Recent studies indicate that the hypertensive action of angiotensin II (Ang II) mediated by its type 1 receptors in central cardiovascular control centers includes induction of the resident neuroimmune system, in particular microglia, in the paraventricular nucleus of the hypothalamus (PVN). Our objective here was to investigate the potential counteractive effects of the protective arms of the renin angiotensin system, angiotensin‐(1–7)/Mas and angiotensin type 2 receptors (AT2R), on activation of hypothalamic microglia. Real time PCR analyses revealed the presence of similar levels of Mas and AT2R in the PVN, as well as other cardiovascular control centers, of normotensive (SD; Sprague Dawley) and spontaneously hypertensive rats (SHR). Both Mas and AT2R were maintained in microglia prepared from the hypothalamus these rat strains, according to respective PCR and receptor binding analyses. Treatment of SD or SHR cultures with either Ang‐(1–7) (100–1000nM) or the AT2R agonist Compound 21 (C21; 100–1000nM) for 24 h elicited significant (~30%) decreases in basal levels of mRNAs for CD11b (marker of microglial activation) and the pro‐inflammatory cytokine interleukin‐1β. These data support the idea that both Mas and AT2R have an anti‐inflammatory role, and potentially an antihypertensive role, in the hypothalamus. Supported by HL‐076803

Abstract:The chronic hypertensive action of angiotensin II (Ang II) mediated within central cardiovascular control centers includes a pro-inflammatory effect and induction of microglial activation in the paraventricular nucleus (PVN) of the hypothalamus. The objective here was to investigate whether angiotensin-(1-7), a protective arm of the renin-angiotensin system (RAS), can exert anti-inflammatory effects in hypothalamic microglia. The presence of Mas, the receptor for Ang-(1-7), within the PVN and other cardiovascular control centers of normotensive (SD; Sprague Dawley) rats was demonstrated by quantitative real time PCR analyses. Mas was maintained in microglial cultures prepared from the hypothalamus of SD rat pups based on immunostaining and PCR. Treatment of microglial cultures with Ang-(1-7) (100nM, 3 -12h) elicited time-dependent significant (P<0.05) decreases in the basal levels of mRNAs for the pro-inflammatory cytokines interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumor-necrosis factor α (TNFα), and in the microglial-macrophage marker CD11b. For example, the levels of IL-1β mRNA (normalized to GAPDH) within control and Ang-(1-7) treated cells were 1.0±0.09, 0.70±0.06, 0.40±0.04, and 0.48±0.07 (n=6). There was no significant effect of Ang-(1-7) on basal IL-1β mRNA at 10 nM (6h), and 1000 nM Ang-(1-7) produced no greater effect than 100 nM of this peptide. Treatment of cultures with the RAS component prorenin (PRO; 10, 20 or 50 nM; 6h) elicited highly significant (P<0.001) increases in IL-1β mRNA (4.2±0.10, 7.4±0.35, 16.7±0.46, respectively, n=6) when compared with controls (0.78±0.06), effects that far exceeded the ~2.0 fold increase in IL-1β mRNA elicited by Ang II (100 nM). Similar stimulatory effects of PRO on TNFα, IL-6 and CD11b mRNAs were also observed. The stimulatory action of PRO (10 nM) on IL-1β and TNFα mRNAs, but not CD11b mRNA, was attenuated by co-treatment with Ang-(1-7) (100 nM). For example, levels of IL-1β mRNA in PRO and Ang-(1-7)/PRO treated cells were 34.1±1.3 and 24.4±1.1 respectively (n=6), a reduction of ~29% (P<0.001). Collectively, these data suggest that Ang-(1-7) exerts a direct anti-inflammatory action at hypothalamic microglia, and inhibits PRO-induced pro-inflammatory effects.

Abstract:AT1R expression is increased in the nucleus of solitary tract (NTS) of many animal models of hypertension. However, the impact of its chronic regulation on cardiovascular control is not well understood. In this study, we investigated the hypothesis that NTS AT1R is involved in peripheral inflammatory status and hypertension-linked cardiovascular pathophysiology. AAV2- mediated expression of a shRNA targeted the AT1R in the NTS has been used to test the hypothesis. First,the efficacy of the AAV2-AR1R-shRNA was established with the use of neuronal cells in primary culture. Transduction of these cultures by AAV2-AT1R-shRNA resulted in a 72% decrease in AT1R mRNA compared with the AAV2- Sc-shRNA transduced control. This was accompanied by complete attenuation of AngII-induced increase in ERK1/2 phosphorylation and neuronal action potential frequency (control: 2.2±0.4 vs. AT1R-shRNA: 0.5±0.3 hz). We then microinjected AAV2-AT1R-shRNA or control vector, AAV2-Scrambled (Sc)-shRNA into the NTS of 8-week-old SHR and WKY rats. Nine weeks following microinjection, AAV2-AT1R-shRNA group of SHRs showed a 29 mmHg increase in mean arterial pressure (MAP) (control: 154.2±4.7; shRNA: 183.0±10.2 mmHg), insignificant effects on the MAP of WKY rats was observed. The increase in MAP in the SHR by AAV2-AT1R-shRNA was associated with a 42% decrease in baroreflex bradycardia (control: 0.57±0.02; shRNA: 0.33±0.06 ms/mmHg), profound cardiac hypertrophy, and significant increase in the plasma norepinephrine (control: 6.36±0.39, shRNA: 13.0±4.13 ng/mL). In addition, AAV2-AT1R-shRNA rats showed significant increases in water intake (control: 32.5±1.5; shRNA: 12.5±2.5 ml/16h) and urine output (control: 1.6 ±0.5; shRNA: 9.3±2.5 ml/16h). Furthermore, AAV2-AT1R-shRNA treated SHR exhibited 74% decrease in circulating endothelial progenitor cells (EPC, CD90+, CD4/5/8-), and ∼300% increases in inflammatory cells (IC) including CD4/8+, T lymphocytes (CD45/3+) and macrophages (CD68+) . As a result, EPC/IC ratio was decreased by 8∼15 fold in AT1R-shRNA treated SHR. These observations suggest that increased AT1R in the NTS of SHR may present a counter-hypertensive mechanism involving inflammatory/angiogenic cells.

Abstract:Heatstroke is characterized by excessive hyperthermia associated with systemic inflammatory responses, which leads to multiple organ failure, in which brain disorders predominate. This definition can be almost fulfilled by a mouse model of heatstroke used in the present study. Unanesthetized mice were exposed to whole body heating (41.2°C for 1 hour) and then returned to room temperature (26°C) for recovery. Immediately after termination of whole body heating, heated mice displayed excessive hyperthermia (body core temperature ~42.5°C). Four hours after termination of heat stress, heated mice displayed (i) systemic inflammation; (ii) ischemic, hypoxic, and oxidative damage to the hypothalamus; (iii) hypothalamo-pituitary-adrenocortical axis impairment (reflected by plasma levels of both adrenocorticotrophic-hormone and corticosterone); (iv) decreased fractional survival; and (v) thermoregulatory deficits (e.g., they became hypothermia when they were exposed to room temperature). These heatstroke reactions can be significantly attenuated by human umbilical cord blood-derived CD34 + cells therapy. Our data suggest that human umbilical cord blood-derived stem cells therapy may improve outcomes of heatstroke in mice by reducing systemic inflammation as well as hypothalamo-pituitary-adrenocortical axis impairment.

Abstract:The organism's normal physiological function is greatly impacted in a febrile environment, leading to the manifestation of pathological conditions including elevated body temperature, dehydration, gastric bleeding, and spermatogenic dysfunction. Numerous lines of evidence indicate that heat stress significantly impacts the brain's structure and function. Previous studies have demonstrated that both animals and humans experience cognitive impairment as a result of exposure to high temperatures. However, there is a lack of research on the effects of prolonged exposure to high-temperature environments on learning and memory function, as well as the underlying molecular regulatory mechanisms. In this study, we examined the impact of long-term heat stress exposure on spatial memory function in rats and conducted transcriptome sequencing analysis of rat hippocampal tissues to identify the crucial molecular targets affected by prolonged heat stress exposure. It was found that the long-term heat stress impaired rats' spatial memory function due to the pathological damages and apoptosis of hippocampal neurons at the CA3 region, which is accompanied with the decrease of growth hormone level in peripheral blood. RNA sequencing analysis revealed the signaling pathways related to positive regulation of external stimulation response and innate immune response were dramatically affected by heat stress. Among the verified differentially expressed genes, the knockdown of Arhgap36 in neuronal cell line HT22 significantly enhances the cell apoptosis, suggesting the impaired spatial memory induced by long-term heat stress may at least partially be mediated by the dysregulation of Arhgap36 in hippocampal neurons. The uncovered relationship between molecular changes in the hippocampus and behavioral alterations induced by long-term heat stress may offer valuable insights for the development of therapeutic targets and protective drugs to enhance memory function in heat-exposed individuals.

Abstract:We have sequenced one complete rDNA tandem repeat from the nematode C. elegans. By comparative analysis we derive secondary structures for the 18s, 5.8s, and 26s rRNA molecules, and comment on other important features of the sequence. We also present the sequence of a junction between the rDNA and non-ribosomal DNA. Finally, we use our data to quantify the evolutionary relationships among several organisms currently studied in developmental biology.
Activation of angiotensin converting enzyme 2 promotes hippocampal neurogenesis via activation of Wnt/β-catenin signaling in hypertension

Abstract:Hypertension-induced brain renin-angiotensin system (RAS) activation and neuroinflammation are hallmark neuropathological features of neurodegenerative diseases. Previous studies from our lab have shown that inhibition of ACE/Ang II/AT1R axis (by AT1R blockers or ACE inhibitors) reduced neuroinflammation and accompanied neurodegeneration via up-regulating adult hippocampal neurogenesis. Apart from this conventional axis, another axis of RAS also exists i.e., ACE2/Ang (1-7)/MasR axis, reported as an anti-hypertensive and anti-inflammatory. However, the role of this axis has not been explored in hypertension-induced glial activation and hippocampal neurogenesis in rat models of hypertension. Hence, in the present study, we examined the effect of ACE2 activator, Diminazene aceturate (DIZE) at 2 different doses of 10 mg/kg (non-antihypertensive) and 15 mg/kg (antihypertensive dose) in renovascular hypertensive rats to explore whether their effect on glial activation, neuroinflammation, and neurogenesis is either influenced by blood-pressure. The results of our study revealed that hypertension induced significant glial activation (astrocyte and microglial), neuroinflammation, and impaired hippocampal neurogenesis. However, ACE2 activation by DIZE, even at the low dose prevented these hypertension-induced changes in the brain. Mechanistically, ACE2 activation inhibited Ang II levels, TRAF6-NFκB mediated inflammatory signaling, NOX4-mediated ROS generation, and mitochondrial dysfunction by upregulating ACE2/Ang (1-7)/MasR signaling. Moreover, DIZE-induced activation of the ACE2/Ang (1-7)/MasR axis upregulated Wnt/β-catenin signaling, promoting hippocampal neurogenesis during the hypertensive state. Therefore, our study demonstrates that ACE2 activation can effectively prevent glial activation and enhance hippocampal neurogenesis in hypertensive conditions, regardless of its blood pressure-lowering effects.

Abstract:Hypertension is associated with blood-brain barrier alteration and brain function decline. Previously, we established the 2-kidney,1-clip (2K1C) hypertensive mice model by renin-angiotensin system (RAS) stimulating. We found that 2K1C-induced hypertension would impair hippocampus-related memory function and decrease adult hippocampal neurogenesis. Even though large studies have investigated the mechanism of hypertension affecting brain function, there remains a lack of efficient ways to halt this vicious effect. The previous study indicated that running exercise ameliorates neurogenesis and spatial memory function in aging mice. Moreover, studies showed that exercise could normalize RAS activity, which might be associated with neurogenesis impairment. Thus, we hypothesize that running exercise could ameliorate neurogenesis and spatial memory function impairment in the 2K1C-hypertension mice. In this study, we performed 2K1C surgery on eight-weeks-old C57BL/6 mice and put them on treadmill exercise one month after the surgery. The results indicate that running exercise improves the spatial memory and neurogenesis impairment of the 2K1C-mice. Moreover, running exercise normalized the activated RAS and blood-brain barrier leakage of the hippocampus, although the blood pressure was not decreased. In conclusion, running exercise could halt hypertension-induced brain impairment through RAS normalization.

Abstract:d Exercise training is believed to have a positive effect on cardiac hypertrophy after hypertension. However, its mechanism is still not fully understood. Herein, our findings suggest that heat shock transcription factor 1 (HSF1) improves exercise-initiated myocardial angiogenesis after pressure overload. A sustained narrowing of the diagonal aorta (TAC) and moderately- intense exercise training protocol were imposed on HSF1 heterozygote (KO) and their littermate wild-type (WT) male mice. After two months, the cardiac function was assessed using the adaptive responses to exercise training, or TAC, or both of them such as catheterization and echocardiography. The HE stains assessed the area of myocyte cross-sectional. The Western blot and real-time PCR measured the levels of expression for heat shock factor 1 (HSF1), vascular endothelial growth factor (VEGF) and hypoxia inducible factor-1 alpha (HIF-1 alpha) in cardiac tissues. The anti-CD31 antibody immunohistochemical staining was done to examine how exercise training influenced cardiac ontogeny. The outcome illustrated that exercise training significantly improved the cardiac ontogeny in TAC mice, which was convoyed by elevated levels of expression for VEGF and HIF-1 alpha and preserved the heart microvascular density. More importantly, HSF1 deficiency impaired these effects induced by exercise training in TAC mice. In conclusion, exercise training encourages cardiac ontogeny by means of HSF1 activation and successive HIF-1 alpha/VEGF up-regulation in endothelial cells during continue pressure overload.

Abstract:We explored the mechanism underlying the improvement of tolerance to exercise-heat stress through physical training at a moderate temperature. Rats were randomly divided into an aerobic exercise (AE) group and a control (C) group. Rats in the former group were subjected during 5 weeks for AE in a cool environment (20°C). The oxidative damage level in the brain to AE rats after both 1-h-long heat exposure and exhaustive exercise in heat and the rate of core temperature rise were lower than those in the control (P < 0.05). The expression level of HSP70 in the motor cortex and the exhaustion time in group AE were found to be significantly greater compared to those in group C (P < 0.05). Our results indicate that the enhanced expression of HSP70 may be an important factor for physical training in a cool environment, and this factor improves tolerance to exercise-heat stress.

Abstract:To explore the influence of short-term (0, 30 and 60 min) exposure to high temperatures on the integrity of hippocampal-dependent learning and memory in neonatal rats. Confocal laser scanning microscopy (CLSM) was used to observe neuronal apoptosis in the hippocampi of neonatal rats. In situ hybridization was conducted for the detection of hippocampal c-fos messenger RNA (mRNA) expression, and reverse transcription-polymerase chain reaction was performed to test the expression of NR1 (an N-methyl-D-aspartate receptor [NR] subunit) gene in the hippocampus. Finally, the Morris water maze (MWM) was used to evaluate hippocampal-dependent learning and memory. The number of c-fos mRNA-positive cells in the hippocampus was positively correlated with increasing heat exposure times. NR1 mRNA expression in the hippocampal CA1 area was positively correlated with the proportion of dead and apoptotic cells. In addition, neonatal rats exposed to high temperatures had significantly reduced platform crossings in the MWM compared to controls. The duration of heat exposure during early pregnancy was correlated with severe deficits in hippocampal-dependent learning and memory in neonatal rats.

Abstract:Abstract In the present study, we investigated the effects of heat shock protein 70 (HSP70) on novel object recognition, cell proliferation, and neuroblast differentiation in the hippocampus. To facilitate penetration into the blood–brain barrier and neuronal plasma membrane, we created a Tat-HSP70 fusion protein. Eight-week-old mice received intraperitoneal injections of vehicle (10% glycerol), control-HSP70, or Tat-HSP70 protein once a day for 21 days. To elucidate the delivery efficiency of HSP70 into the hippocampus, western blot analysis for polyhistidine was conducted. Polyhistidine protein levels were significantly increased in control-HSP70- and Tat-HSP70-treated groups compared to the control or vehicle-treated group. However, polyhistidine protein levels were significantly higher in the Tat-HSP70-treated group compared to that in the control-HSP70-treated group. In addition, immunohistochemical study for HSP70 showed direct evidences for induction of HSP70 immunoreactivity in the control-HSP70- and Tat-HSP70-treated groups. Administration of Tat-HSP70 increased the novel object recognition memory compared to untreated mice or mice treated with the vehicle. In addition, the administration of Tat-HSP70 significantly increased the populations of proliferating cells and differentiated neuroblasts in the dentate gyrus compared to those in the control or vehicle-treated group based on the Ki67 and doublecortin (DCX) immunostaining. Furthermore, the phosphorylation of cAMP response element-binding protein (pCREB) was significantly enhanced in the dentate gyrus of the Tat-HSP70-treated group compared to that in the control or vehicle-treated group. Western blot study also demonstrated the increases of DCX and pCREB protein levels in the Tat-HSP70-treated group compared to that in the control or vehicle-treated group. In contrast, administration of control-HSP70 moderately increased the novel object recognition memory, cell proliferation, and neuroblast differentiation in the dentate gyrus compared to that in the control or vehicle-treated group. These results suggest that Tat-HSP70 promoted hippocampal functions by increasing the pCREB in the hippocampus.
Heat shock factor HSF1 regulates BDNF gene promoters upon acute stress in the hippocampus, together with pCREB

Abstract:Heat shock factor (HSF1) is a master stress‐responsive transcriptional factor, protecting cells from death. However, its gene regulation in vivo in the brain in response to neuronal stimuli remains elusive. Here, we investigated its direct regulation of the brain‐derived neurotrophic factor (BDNF) gene (Bdnf) in response to acute neuronal stress stimuli in the brain. The results of immunohistochemistry and chromatin immunoprecipitation quantitative PCR (ChIP‐qPCR) showed that administration of kainic acid (a glutamate receptor agonist inducing excitotoxity) to young adult mice induced HSF1 nuclear translocation and its binding to multiple Bdnf promoters in the hippocampus. Footshock, a physical stressor used for learning, also induced HSF1 binding to selected Bdnf promoters I and IV. This is, to our knowledge, the first demonstration of HSF1 gene regulation in response to neuronal stimuli in the hippocampus in vivo. HSF1 binding sites (HSEs) in Bdnf promoters I and IV were also detected when immunoprecipitated by an antibody of phosphorylated (p)CREB (cAMP‐responsive element‐binding protein), suggesting their possible interplay in acute stress‐induced Bdnf transcription. Interestingly, their promoter binding patterns differed by KA and footshock, suggesting that HSF1 and pCREB orchestrate to render fine‐tuned promoter control depending on the types of stress. Further, HSF1 overexpression increased Bdnf promoter activity in a luciferase assay, while virus infection of constitutively active‐form HSF1 increased levels of BDNF mRNA and protein in vitro in primary cultured neurons. These results indicated that HSF1 activation of Bdnf promoter was sufficient to induce BDNF expression. Taken together, these results suggest that HSF1 promoter‐specific control of Bdnf gene regulation plays an important role in neuronal protection and plasticity in the hippocampus in response to acute stress, possibly interplaying with pCREB.

Intrinsic factors involved in the depression of neuronal activity induced by temperature increase in rat hippocampal neurons

Abstract:The intrinsic factors involved in the temperature-dependent impairment of neuronal activity in hippocampal CA2-CA1 regions were investigated using optical recording techniques. At 32 degrees C, stimulation of the Schaffer collaterals in the hippocampal CA2 region evoked depolarizing optical responses that spread toward the CA1 region. The optical response was characterized by fast and slow components that were mainly related to the presynaptic action potentials and excitatory postsynaptic response, respectively. The increase of the temperature to 38 degrees C was associated with a reversible depression of the neuronal activity in the hippocampal brain preparations. The depression of neuronal activity was irreversible when the temperature was increased to 40 degrees C. In the presence of 22 mM glucose, the depression of the neuronal activity at 38 degrees C was significantly attenuated. Pyruvate (22 mM), but not lactate (22 mM), also improved the depression of neuronal activity induced by the temperature increase. Adenosine (200 microM) strongly depressed the excitatory postsynaptic response, but not presynaptic action potentials. 8-Cyclopentyl-1,3-dimethylxanthine (8-CPT) (10 microM), an adenosine A1 receptor blocker, attenuated the adenosine-induced depression of the excitatory postsynaptic response. 8-CPT (10 microM) prevented the impairment of the excitatory postsynaptic response induced by the increase of the temperature to 38 degrees C. In contrast, the depression of presynaptic action potential at 38 degrees C was not prevented by 8-CPT (10 microM). N omega-nitro-L-arginine methyl ester (L-NAME), a nitric oxide synthase (NOS) inhibitor, and methylcobalamin (10 microM), a vitamin B12 analogue, attenuated the inhibition of pre- and postsynaptic activities induced by the increase of the temperature to 38 degrees C. Glibenclamide, a KATP channel blocker, did not protect neuronal activity from the effects of the increase of the temperature. These results suggest that the heat-induced depression of neuronal activity is mediated by multiple factors, such as impairment of energy metabolism and increase in extracellular adenosine and nitric oxide (NO) levels in hippocampal neurons.

Abstract:Heat acclimation (HA) is a key adaptive response in mammals to repeated heat exposure, essential for fitness and survival . HA improves cardiovascular function, thermal comfort, and exercise capacity . However, the lack of a genetically tractable model has hindered understanding of the molecular and neural mechanisms underlying HA. Here, we show that 10 days of daily 38°C exposure lowers core body temperature (T ) and reduces anxiety during subsequent heat exposures in mice. HA increases brain-derived neurotrophic factor (BDNF) expression in the medial preoptic area (MPO). BDNF-expressing MPO neurons (MPO ) show increased intrinsic heat sensitivity after HA. These neurons orchestrate downstream targets in the dorsomedial hypothalamus (DMH) and rostral raphe pallidus (rRPa) to mediate HA effects. BDNF, acting through its receptor tropomyosin-related kinase B (TrkB) in the DMH, facilitates the anxiolytic effect of HA by enhancing excitatory synaptic connections between MPO and DMH neurons. This study provides new insights into HA mechanisms, setting the stage for future research on heat stress reduction and exercise optimization.

Upregulation of AT1 Receptor Mediates a Pressor Effect Through ROS-SAPK/JNK Signaling in Glutamatergic Neurons of Rostral Ventrolateral Medulla in Rats with Stress-Induced Hypertension.

Abstract:The present study examined whether angiotensin II (Ang II) mediates the pressor effect through nicotinamide adenine dinucleotide phosphate (NADPH) oxidase-derived reactive oxygen species (ROS)-mitogen-activated protein kinase (MAPK) signaling in the glutamatergic neurons of the rostral ventrolateral medulla (RVLM) in stress-induced hypertensive rats (SIHR). The SIHR model was established using electric foot-shocks combined with noises for 15 days. We observed that Ang II type 1 receptor (AT1R) and the glutamatergic neurons co-localized in the RVLM of SIHR. Furthermore, glutamate levels in the intermediolateral column of the spinal cord were higher in SIHR than in controls. Microinjection of Ang II into the RVLM of SIHR activated stress-activated protein kinase/Jun N-terminal kinase (SAPK/JNK), extracellular signal-regulated protein kinase (ERK) 1/2, and p38MAPK. Compared with controls, the activation of SAPK/JNK, ERK1/2, p38MAPK, and ROS in the RVLM were higher in SIHR, an effect that was blocked by an NADPH oxidase inhibitor (apocynin) and an AT1R antagonist (candesartan). RVLM microinjection of apocynin or a SAPK/JNK inhibitor (SP600125), but not an ERK1/2 inhibitor (U0126) or a p38MAPK inhibitor (SB203580), decreased AT1R mRNA and mean arterial blood pressure (MABP) in SIHR. The increase of AT1R protein expression and MABP was inhibited by intracerebroventricular infusion (ICV), for 14 days, of SP600125, but not U0126 or SB203580 in SIHR. We conclude that Ang II modulates the pressor effect through AT1R-dependent ROS-SAPK/JNK signaling in glutamatergic neurons in the RVLM of SIHR.
Pharmacological Activation Of Aldehyde Dehydrogenase 2 Protects Against Heatstroke-Induced Acute Lung Injury by Modulating Oxidative Stress and Endothelial Dysfunction

Abstract:Heatstroke (HS) can cause acute lung injury (ALI). Heat stress induces inflammation and apoptosis via reactive oxygen species (ROS) and endogenous reactive aldehydes. Endothelial dysfunction also plays a crucial role in HS-induced ALI. Aldehyde dehydrogenase 2 (ALDH2) is a mitochondrial enzyme that detoxifies aldehydes such as 4-hydroxy-2-nonenal (4-HNE) protein adducts. A single point mutation in ALDH2 at E487K (ALDH2*2) intrinsically lowers the activity of ALDH2. Alda-1, an ALDH2 activator, attenuates the formation of 4-HNE protein adducts and ROS in several disease models. We hypothesized that ALDH2 can protect against heat stress-induced vascular inflammation and the accumulation of ROS and toxic aldehydes. Homozygous ALDH2*2 knock-in (KI) mice on a C57BL/6J background and C57BL/6J mice were used for the animal experiments. Human umbilical vein endothelial cells (HUVECs) were used for the in vitro experiment. The mice were directly subjected to whole-body heating (WBH, 42°C) for 1 h at 80% relative humidity. Alda-1 (16 mg/kg) was administered intraperitoneally prior to WBH. The severity of ALI was assessed by analyzing the protein levels and cell counts in the bronchoalveolar lavage fluid, the wet/dry ratio and histology. ALDH2*2 KI mice were susceptible to HS-induced ALI in vivo. Silencing ALDH2 induced 4-HNE and ROS accumulation in HUVECs subjected to heat stress. Alda-1 attenuated the heat stress-induced activation of inflammatory pathways, senescence and apoptosis in HUVECs. The lung homogenates of mice pretreated with Alda-1 exhibited significantly elevated ALDH2 activity and decreased ROS accumulation after WBH. Alda-1 significantly decreased the WBH-induced accumulation of 4-HNE and p65 and p38 activation. Here, we demonstrated the crucial roles of ALDH2 in protecting against heat stress-induced ROS production and vascular inflammation and preserving the viability of ECs. The activation of ALDH2 by Alda-1 attenuates WBH-induced ALI in vivo.
