Abstract:Chemical investigation of a co-culture of two endophytic fungi Aspergillus fumigatus D and Fusarium oxysporum R1 from two traditional medicinal plants, Edgeworthia chrysantha Lindl. and Rumex madaio Makino, led to isolation of a new amide 1 and six known compounds, including neovasinin (2), neovasifuranone B (3), N-(2-phenylethyl)acetamide (4), α-linolenic acid (5), α-elaeostearic acid (6), palmitoleic acid (7). On the basis of extensive spectroscopic analysis including 1D and 2D NMR, HR-ESI-MS and optical rotation measurement as well as comparison of literature data, chemical structure of 1 was unambiguous elucidated as (S, E)-methyl-2-(2,4-dimethylhex-2-enamido)acetate. Bioassay results indicated that none of these compounds exhibited strong inhibitory effect on three human pathogens Escherichia coli, Staphyloccocus aureus and Candida albicans with MIC values of ≥25 μM.

Abstract:A new cytotoxic and antiviral cyclic tetrapeptide, asperterrestide A(1), a new alkaloid, terremide C (2), and a new aromatic butenolide, aspernolide E (A), together with 10 known compounds were isolated from the fermentation broth of the marine-derived fungus Aspergillus terreus SCSGAF0162:. Their structures were elucidated by spectroscopic analysis, and the absolute configuration of 1 was determined by the Mosher ester technique, and analysis of the acid hydrolysates using a chiral, phase HPLC column. Compound 1 contains a rare 3-OH-N-CH3-Phe residue and showed cytotoxicity against U937 and MOLT4 human carcinoma cell lines and inhibitory effects on influenza virus strains H1N1 and H3N2.

Abstract:Aspersteroid A (1), a highly rearranged 1(10 → 6)-abeo-18,22-cyclosterol, together with two new 18,22-cyclosterols (2 and 3), was isolated from the culture extract of Aspergillus ustus NRRL 275. Their structures were determined by spectroscopic analysis, X-ray diffraction, modified Mosher's method, and Rh2(OCOCF3)4-induced electronic circular dichroism experiments. Compound 1 represents a new carbon skeleton with an uncommon 6/6/6/5/5 ring system, which is presumably biosynthesized from A-ring scission, double 1,2-shifts, and C-18/C-22 cyclization. Compound 1 exhibited potent immunosuppressive and antimicrobial activities.

Abstract:Asperginine (1), an alkaloid possessing a rare skeleton, was isolated from the cultural broth of the marine fungus Aspergillus sp. (Z-4) isolated from the gut of the marine isopod Ligia oceanica. The planar structure and relative configuration of 1 was determined by analysis of NMR and mass spectral data. Its absolute configuration was elucidated by Marfey's method, together with NOESY correlations of key hydrogen atoms. The cytotoxicity against prostate cancer PC3 and human HCT116 was assayed by the MTT method. Unfortunately, asperginine did not show any activity.

Abstract:A new sesquiterpenoid derivative, named aspergiketone (1), along with seven known compounds (2-8) were isolated from the coastal saline soil fungus Aspergillus fumigatus. Their structures were elucidated by spectroscopic analysis, and by comparison of experimental and reported data. The absolute configuration of compound 1 was defined by X-ray diffraction analysis. Compound 1 was cytotoxic towards HL-60 and A549 cell lines with IC50 values of 12.4 and 22.1 mu M, respectively.

Abstract:Fungi are important resource for the discovery of novel bioactive natural products. This study investigated the metabolites produced by Mariana-Trench-associated fungus Aspergillus sp. SY2601 in EY liquid and rice solid media, resulting in the isolation and structure determination of 28 metabolites, including five new compounds, asperindopiperazines A–C (1–3), 5-methoxy-8,9-dihydroxy-8,9-deoxyaspyrone (21), and 12S-aspertetranone D (26). Structures of the new compounds were elucidated based on extensive NMR spectral analyses, HRESIMS data, optical rotation, ECD, and 13C NMR calculations. The new compound 12S-aspertetranone D (26) exhibited antibacterial activity against both methicillin-resistant Staphylococcus aureus and Escherichia coli with MIC values of 3.75 and 5 μg/mL, respectively.

Abstract:A novel polycyclic aspochalasin, aspochalazine A (1), was isolated along with three related known compounds 2-4 from the culture broth of Aspergillus sp., which was found in the gut of a marine isopod Ligia oceanica. The structures were determined on the basis of ESI-MS, NMR spectrum data and single-crystal X-ray diffraction. This is the first report about aspochalasin derivative with an azabicyclo. Abstract:An unusual C18 norditerpenoid, aspergiloid I (1), was isolated from the culture broth of Aspergillus sp. YXf3, an endophytic fungus derived from Ginkgo biloba. Its structure was unambiguously established by analysis of HRMS–ESI and spectroscopic data, and the absolute configuration was determined by low-temperature (100 K) single crystal X-ray diffraction with Cu Kα radiation. This compound is structurally characterized by a new carbon skeleton with an unprecedented 6/5/6 tricyclic ring system bearing an α,β-unsaturated spirolactone moiety in ring B, and represents a new subclass of norditerpenoid, the skeleton of which is named aspergilane. The hypothetical biosynthetic pathway for 1 was also proposed. The cytotoxic, antimicrobial, anti-oxidant and enzyme inhibitory activities of 1 were evaluated.

Compound 1 contains a rare 5/6/6 tricyclic ring fused with the aspochalasin skeleton. The structures were determined on the basis of electrospray ionisation mass spectroscopy (ESIMS), nuclear magnetic resonance (NMR) spectral data, and the absolute configurations were further confirmed by modified Mosher's method. Cytotoxicity against the prostate cancer PC3 cell line were assayed by the MTT method. Compound 3 showed strong activity while the remaining compounds showed weak activity.

Abstract:Chemical investigation of the fungus Trichoderma asperellum SCNU-F0048 led to the discovery of two new steroids, ergosta-4,6,8 (14),22-tetraen-3-(3′-methyl-4′-hydroxyl- γ -butenolide) ( 1 ) and camphosterol B ( 2 ), as well as two known compounds, i.e. stigmasta-4,6,8(14),22-tetraen-3-one ( 3 ) and 4-hydroxy-17- methylincisterol ( 4 ). Their structures were elucidated by extensive nuclear mangnetic resonance, spectrum analysis and single crystal X-ray diffraction analysis. Bioassay disclosed that compound 1 showed strong cytotoxicity to a panel of tumor cell lines. Moreover, compounds 1 and 2 showed excellent antifungal activity against Penicillium italicum with IC 50 values of 0.016 and 0.022 μM, respectively.

Abstract:Two new sterol derivatives, namely ergosterimide B (1) and demethylincisterol A5 (2), along with eleven known metabolites (3–13), were isolated from the rice fermentation culture of Aspergillus tubingensis YP-2. The chemical structures were elucidated by detailed NMR and MS spectroscopic data and by comparison with reported data. Newly isolated compound 2 and known compound 3 were evaluated for their cytotoxicities against the A549 and HepG2 cell lines. Compound 2 showed weak activities with IC50 values of 11.05 and 19.15 μM, respectively, while 3 exhibited cytotoxicities with IC50 values of 5.34 and 12.03 μM, respectively, against the tested cell lines.

Abstract:Chemical examination of fermentation products of a deep-sea-derived fungus Aspergillus versicolor F77 led to the isolation of two new compounds acetyl-sydonol(1)and 2-hydroxydiorcinol D(2),a new natural product l-(2,4-dimethoxyphenyl)-l,2-dihydro-3H-pyrazol-3-one(3),together with four known compounds sydonol(4),(S)-sydonic acid(5),aspergillusene A(6)and diorcinol D(7).Their structures were determined through the extensive spectroscopic analysis and comparison with the reported data in literature.Compound 6 showed moderate NF-κB inhibitory activity.

Abstract:Marine natural products, characterized by fascinating drug-like functionalities and promising biological activities, are important base materials for innovative drugs and agrochemicals. Chemical investigations of the marine-algal-derived endophytic fungus Aspergillus sp. 1022LEF residing in the inner tissue of marine red alga yielded a novel polyketide-terpene hybrid metabolite, namely tennessenoid A ( 1 ), as well as six known biosynthetic congeners including two steroids, ergosta-4,6,8(14),22-tetraen-3-one ( 2 ) and (22 E ,24 R )-3α-ureido-ergosta-4,6,8(14),22-tetraene ( 3 ), and four sorbicillinoid-based compounds, saturnispol G ( 4 ), trichodimerol ( 5 ), and dihydrotrichodimer ethers A and B ( 6 and 7 ). Their structures were unambiguously determined based on extensive 1D/2D NMR and HRESIMS spectroscopic analyses. Tennessenoid A ( 1 ) was characterized as an unprecedented steroid−sorbicillinoid adduct via a C−C bond, which was rarely-observed in natural products. All of the isolated compounds were evaluated for their antifungal activities against eight plant pathogenetic fungi. 1 , in particular, demonstrated broad-spectrum activities against Sclerotium rolfsii Sacc., Fusarium oxysporum (Schl.) F.sp cucumerinum Owen, Coniella diplodiella Petrak et Sydow, Physalospora piricola Nose., Fusarium graminearum schw., Alternaria mali rob., Colletotrichum orbiculare Arx., and Alternaria porri (E11iott) Cifed., with the inhibition zone diameters ranging from 2 to 7 mm.

Abstract:Aspernigerin (1), a novel cytotoxic alkaloid consisting of an unprecedented structural framework has been isolated from the extract of a culture of Aspergillus niger IFB-E003, an endophyte in Cyndon dactylon. Its structure was elucidated on the basis of comprehensive NMR spectral analysis and confirmed by single-crystal X-ray analysis. Aspernigerin (1) has been shown to be cytotoxic to the tumor cell lines nasopharynyeal epidermoid KB, cervical carcinoma Hela, and colorectal carcinoma SW1116 with corresponding IC50 values of 22, 46, and 35 mu m, respectively. A feasible total synthetic route for aspernigerin (1) has been established for further pharmacological research.

Abstract:From the marine sponge Xestospongia exigua, fungal isolates of Aspergillus versicolor (Vuill)Triab were obtained. Isolation and purification of ethyl acetate extracts from culture filtrates of the fungus led to yield six new chromone derivatives namely aspergione A, aspergione B, aspergione C, aspergione D. aspergione E, aspergione F. The structures of all the new compounds were established on the basis of extensive spectroscopy (UV, MS, H-1 and C-13 NMR, COSY, HMQC and HMBC) analysis.

Abstract:Two undescribed 30-norlanostane triterpenoids, named nidulanoids A and B, one ergostane-type steroid with an unusual double bond between C-17 and C-20 designated (17E,22E,24R)-3β,5α-dihydroxyergosta-7,17,22-trien-6,16-dione, and one pregnane, (7Z,9Z,17Z)-,2α,3β-dihydroxypregna-7,9,17 (20)-trien-18-al, along with six known steroids were isolated from the extract of the fungus Aspergillus nidulans. Among them, nidulanosides A and B represents the first example of naturally occurred 30-norlanostane triterpenoids featuring a C9 side-chain moiety at C-17 and a hemiacetal system formed between C-3 and C-19, as an intermediate between lanostane and the regular steriods; the structure of (17E,22E,24R)-3β,5α-dihydroxyergosta-7,17,22-trien-6,16-dione possesses an untypical Δ17,20 double bond; meanwhile, (7Z,9Z,17Z)-,2α,3β-dihydroxypregna-7,9,17 (20)-trien-18-al represents the first example of C-21 steroid with an aldehyde group at C-13. Their structures and absolute stereochemistry were elucidated based on spectroscopic data, electronic circular dichroism (ECD) calculations, and single-crystal X-ray diffraction analysis. (7Z,9Z,17Z)-,2α,3β-dihydroxypregna-7,9,17 (20)-trien-18-al showed moderate inhibitory activities against rat brain cancer (PC12) cell lines, with IC50 value of 7.34 μM. This study enriches the diversified structures of triterpenoids and steroids analogues from A. nidulans and indicated (7Z,9Z,17Z)-,2α,3β-dihydroxypregna-7,9,17(20)-trien-18-al to be a promising lead compound against PC12 cell lines.

Abstract:Chemical investigation of the extracts of Aspergillus sp. CSYZ-1 resulted in the identification of compound 1, aspergillactone, a new 3,5-dimethylorsellinic acid-based meroterpenoid, together with four known metabolites (2-5). The structure and relative configuration of 1 were unambiguously determined by nuclear magnetic resonance (NMR), mass spectrometry. The absolute configuration of 1 was defined by quantum chemical TDDFT calculated and the experimental ECD spectra. The possible biosynthetic pathway of compound 1 was also proposed. The new compound exhibited potent antimicrobial activity against Helicobacter pylori and Staphylococcus aureus with MIC values of around 1-4 and 2-16 mu g/mL, respectively.
