Abstract:Improving maternal nutrition during pregnancy/lactation is a promising strategy to maximise the intestinal health of piglets undergoing abrupt weaning under commercial production conditions. This experiment investigated the effects of maternal supplementation of a casein hydrolysate and yeast β-glucan (CH-YBG) from day 83 of gestation until weaning (day 28) on sow faecal microbial populations and measures of piglet gastrointestinal health parameters at weaning. Sows (n = 10 sows/group) were assigned to: (1) control diet, and (2) control diet + CH-YBG. Maternal supplementation increased the abundance of the phylum Firmicutes, including members Lactobacillus in the sows faeces, with a concomitant increase in the caecal abundance of Lactobacillus in the weaned piglets compared to the controls Piglets weaned from the supplemented sows had increased villus height in the duodenum (P < 0.05) and increased villus height to crypt depth ratio in the jejunum, as well as a decreased expression of the proinflammatory cytokine genes ( IL6 / TNF / TGFB) , the tight junction gene CLDN3 and the mucin gene MUC2 in the duodenum/jejunum compared to the controls (P < 0.05). In conclusion, maternal CH-YBG supplementation during pregnancy/lactation improved microbial, structural, and inflammatory measures of gastrointestinal health of piglets at weaning. This is a promising strategy to alleviate the challenges that occur with early abrupt weaning in commercial pig production.

Abstract:Abstract Feeding probiotics like live yeast Saccharomyces cerevisiae var. boulardii (SB) in pig diets has been suggested to preserve health and reduce antibiotic use during critical periods like weaning. This study was conducted to determine whether SB added to the diet of sows during the last 2 mo of gestation and the 4 wk of lactation may contribute to support the health and performance of piglets before and after weaning through changes in sow physiology, milk composition, and fecal microbiota. Crossbred sows (n = 45) from parity 1 to 9 were allocated to two dietary treatments: Control (n = 23) and SB (n = 22). Sows in the SB group were fed the same standard gestation and then lactation diet as the Control sows but with the addition of SB at 1 × 109 colony-forming units/kg of feed. Piglets were weaned under challenging conditions consisting of mixing of litters, no pen cleaning, and a 2-h period of nonoptimal temperature exposure. Blood and feces were collected from sows on days 28 and 113 of gestation and days 6 (feces only) and 28 of lactation, and from piglets on days 6 (feces) and 28 of lactation and day 5 after weaning. Colostrum was collected during parturition and milk on day 6 of lactation. Supplementation of sow diets with SB influenced the fecal microbiota of the sows and their piglets. Five days after weaning, the alpha-diversity was lower (P < 0.05) in piglets from SB sows than in piglets from Control sows. Analysis of microbiota with partial least square discriminant analysis discriminated feces from SB sows from that of Control sows at 110 d of gestation (29.4% error rate). Piglet feces could also be discriminated according to the diet of their mother, with a better discrimination early after birth (day 6 of lactation) than after weaning (day 5 postweaning, 3.4% vs. 12.7% error rate). Five days after weaning, piglets had greater white blood cell count, plasma haptoglobin concentration, and oxidative stress than before weaning (P < 0.001). Nevertheless, SB supplementation in sow diets had no effect (P > 0.05) on most of health criteria measured in blood and growth performance of piglets during lactation and the postweaning period. Moreover, dietary supplementation of SB to sows did not elicit any changes (P > 0.05) in their reproductive performance, metabolic and health status, nor in the concentration of immunoglobulins and nutrients in colostrum and milk. In the present experimental conditions, feeding SB to sows influenced sow and piglet microbiota with no consequences on their health and performance.

Abstract:Abstract A 2 × 2 factorial experiment was conducted to investigate the effect of maternal supplementation from day 83 of gestation and/or direct supplementation from weaning of a bovine casein hydrolysate plus a yeast β-glucan (CH-YBG) on pig performance and intestinal health on day ten post-weaning. Twenty cross bred gilts (Large White × Landrace) were randomly assigned to one of two dietary groups ( n = 10 gilts/group): basal diet (basal sows) and basal diet supplemented with CH-YBG (supplemented sows) from day 83 of gestation until weaning (2g/sow/day). At weaning, 120 pigs (6 pigs/sow) were selected. The two dam groups were further divided, resulting in four experimental groups (10 replicates/group; 3 pigs/pen) as follows: 1) BB (basal sows + basal pigs); 2) BS (basal sows + supplemented pigs); 3) SB (supplemented sows + basal pigs); 4) SS (supplemented sows + supplemented pigs). Supplemented pigs were offered 0.5g CH-YBG/kg of feed for 10 days post-weaning. On day 10 post-weaning, 1 pig/pen was humanely sacrificed and samples were taken from the gastrointestinal tract for analysis. Pigs weaned from supplemented sows (SS, SB) had reduced faecal scores and incidence of diarrhoea (P<0.05) compared to pigs weaned from basal sows (BB, BS), with SS pigs not displaying the transient rise in faecal scores seen in the other three groups from day 3 to day 10 post-weaning (P<0.05). Pigs weaned from supplemented sows had reduced feed intake (P<0.05), improved feed efficiency (P<0.05), increased butyrate concentrations (P<0.05), increased abundance of Lactobacillus (P<0.05) and decreased abundance of Enterobacteriaceae and Campylobacteraceae (P<0.05) compared to pigs weaned from basal sows. In conclusion, a combination of both maternal and direct supplementation with CH-YBG is the desirable feeding strategy to ameliorate the post-weaning challenge in pig production systems.
Factors affecting sow colostrum yield and composition, and their impact on piglet growth and health

Abstract:The present study investigated sow colostrum yield (CY), colostrum composition and factors affecting them, and their relation to piglet survivability, growth and mortality. The study included 230 sows with 3,210 live-born piglets from five Finnish and one Dutch sow herd. Sow farrowing was supervised, and piglets were individually weighed at birth (BWB) and 24 h after birth of first piglet in order to calculate piglet CI and sow CY. Colostrum nutritional composition, immunoglobulin (Ig), serum amyloid A (SAA) and haptoglobin (Hp) contents were assessed. Sow plasma SAA, Hp and progesterone around farrowing were also assessed. Selected ear-tagged piglets were weighed at 3 to 4 weeks of age to calculate individual average daily gain. Sow CY was positively correlated with plasma Hp (P = 0.029) and number of live-born piglets (P < 0.01). An additional minute of farrowing duration lowered the CY by 2.2 g (P = 0.01). Piglet CI was positively associated with piglet weight at birth (P < 0.001) and negatively associated with the number of live-born piglets in the litter and percentage of protein in the colostrum (P < 0.001). Both piglet CI and birth weight were positively associated with piglet average daily gain (ADG) (P < 0.001). Piglet survival from birth to weaning depends on CI. We established that the risk of piglet death or of a piglet being treated with antibiotic before weaning increases with a decrease in sow back fat thickness at farrowing (P = 0.04). Similarly, we found that piglets from litters with low BWB and low CI had a higher risk of death before weaning (P < 0.001). Piglets born from sows with lower levels of colostrum IgA and SAA and high plasma progesterone at the end of farrowing had a higher risk of neonatal diarrhea (P = 0.04; P = 0.05; P = 0.04). Piglets born from sows having higher back fat thickness at weaning had a higher risk of developing weaning diarrhea (P = 0.02). In conclusion, longer farrowing duration can be detrimental and can negatively influence sow CY. Sow body condition and physiological status around farrowing can also affect CY, and thereby increase piglet mortality and use of antibiotics in neonatal piglets. Neonatal piglets can benefit from higher colostrum immunoglobulins, SAA, and decreased level of plasma progesterone in sows at the end of farrowing.

Abstract:The goal of this study was to evaluate the effects of a mixture of yeast culture, cell wall hydrolysates, and yeast extracts (collectively “yeast products,” YP) on the performance, intestinal physiology, and health of weaned piglets. A total of 90 piglets weaned at 21 d of age were blocked by body weight, sex, and litter and randomly assigned to one of three treatments for a 14-d feeding experiment, including (1) a basal diet (control), (2) 1.2 g/kg of YP, and (3) 20 mg/kg of colistin sulfate (CSE). No statistically significant differences were observed in average daily feed intake, average daily weight gain, or gain-to-feed ratio among CSE, YP, and control piglets. Increased prevalence of diarrhea was observed among piglets fed the YP diet, whereas diarrhea was less prevalent among those fed CSE. Duodenal and jejunal villus height and duodenal crypt depth were greater in the control group than they were in the YP or CSE groups. Intraepithelial lymphocytes (IEL) in the duodenal and jejunal villi were enhanced by YP, whereas IEL in the ileal villi were reduced in weaned piglets fed YP. Secretion of jejunal and ileal interleukin-10 (IL-10) was higher and intestinal and serum antioxidant indexes were affected by YP and CSE. In YP- and CSE-supplemented animals, serum D-lactate concentration and diamine oxidase (DAO) activity were both increased, and intestinal mRNA expressions of occludin and ZO-1 were reduced as compared to the control animals. In conclusion, YP supplementation in the diets of weaned piglets appears to increase the incidence of diarrhea and has adverse effects on intestinal morphology and barrier function.

Abstract:The present study was conducted to determine effects of different forms of yeast (Saccharomyces cerevisiae, strain Y200007) on the growth performance, intestinal development, and systemic immunity in early-weaned piglets. A total of 96 piglets (14-d old, initial average body weight of 4.5 kg) were assigned to 4 dietary treatments: (1) basal diet without yeast (Control); (2) basal diet supplemented with 3.00 g/kg live yeast (LY); (3) basal diet supplemented with 2.66 g/kg heat-killed whole yeast (HKY); and (4) basal diet supplemented with 3.00 g/kg superfine yeast powders (SFY). Diets and water were provided ad libitum to the piglets during 3-week experiment. Growth performance of piglets was measured weekly. Samples of blood and small intestine were collected at days 7 and 21 of experiment. Dietary supplementation with LY and SFY improved G:F of piglets at days 1-21 of the experiment (P < 0.05) compared to Control group. Serum concentrations of growth hormone (GH), triiodothyronine (T3), tetraiodothyronine (T4), and insulin growth factor 1 (IGF-1) in piglets at day 21 of the experiment were higher when fed diets supplemented with LY and SFY than those in Control group (P < 0.05). Compared to Control group, contents of serum urea nitrogen of piglets were reduced by the 3 yeast-supplemented diets (P < 0.05). Diets supplemented with LY increased villus height and villus-to-crypt ratio in duodenum and jejunum of piglets (P < 0.05) compared to other two groups at day 7 of the experiment. Feeding diets supplemented with LY and SFY increased (P < 0.05) serum concentrations of IgA, IL-2, and IL-6 levels in piglets compared to Control. The CD4+/CD8+ ratio and proliferation of T-lymphocytes in piglets fed diets supplemented with LY were increased compared to that of Control group at day 7 of the experiment (P < 0.05). In conclusion, dietary supplementation with both LY and SFY enhanced feed conversion, small intestinal development, and systemic immunity in early-weaned piglets, with better improvement in feed conversion by dietary supplementation with LY, while dietary supplementation with SFY was more effective in increasing systemic immune functions in early-weaned piglets.

Abstract:Hydrolyzed yeast (HY)-derived protein from Saccharomyces cerevisiae has a high digestible protein content and nucleotides and is a sweetener immunostimulatory substance. This could be used in nursery diets to minimize diarrhea and improve the growth rate and gut health of early-weaned piglets. This research was conducted with the objective of examining the effect of the inclusion level of HY as a potential protein ingredient for early-weaned piglets. A total of 72 crossbred weaned piglets [(Landrace × Large White) × Duroc] were assigned to three dietary treatments in six replicates with four pigs per pen. Dietary treatments were: (i) control (CON), piglets weaned at 18 days; (ii) CON diet with 5% HY inclusion (HY5); and (iii) CON diet with 10% HY inclusion (HY10) in a corn–soybean meal-based basal diet. Increasing HY levels positively improved body weight, average daily gain, and average daily feed intake (linear effect, p < 0.05). Furthermore, there was a linear increase in N-retention, albumin, jejunal villus height, villus height-to-crypt depth ratio, immunoglobulin A, acetate and propionate production, and Lactobacillus spp. count proportional to the dose of the HY-supplemented diet (p < 0.05). It also observed a decrease in diarrheal rate, jejunal crypt depth, blood urea nitrogen, pro-inflammatory cytokines, branched amino acids, and E. coli corresponding to the HY-supplemented levels (p < 0.05). However, the changes in the apparent total tract digestibility (dry matter, crude ash, and crude fat), blood glucose, butyrate, and Salmonella spp. were unaffected by the dietary HY level. Therefore, the supplementation of HY in the diet for early-weaned pigs not only supported the growth rate and immune function but also activated the beneficial bacterial growth of the early-weaned piglets.

Abstract:OBJECTIVE Compare maximum concentration (Cmax), time to maximum concentration (Tmax), mean serum concentration of vasopressin, return of spontaneous circulation (ROSC), time to ROSC, and odds of survival relative to vasopressin administration by tibial intraosseous (TIO), humerus intraosseous (HIO), and intravenous (IV) routes in a hypovolemic cardiac arrest model. DESIGN Prospective, between subjects, randomized experimental design. SETTING TriService Research Facility. SUBJECTS Yorkshire-cross swine (n = 40). INTERVENTION Swine were anesthetized, exsanguinated to a Class III hemorrhage, and placed into cardiac arrest. After 2 minutes, cardiopulmonary resuscitation was initiated. After an additional 2 minutes, a dose of 40 units of vasopressin was administered by TIO, HIO, or the IV routes. Blood samples were collected over 4 minutes and analyzed by high-performance liquid chromatography tandem mass spectrometry. MAIN OUTCOME MEASUREMENTS ROSC, time to ROSC, Cmax, Tmax, mean concentrations over time, and odds ratio. RESULTS There was no significant difference in rate of ROSC or time to ROSC between the TIO, HIO, and IV groups (p > 0.05). The Cmax was significantly higher in the IV group compared to the TIO group (p = 0.015), but no significant difference between the TIO versus HIO or HIO versus IV groups (p > 0.05). The Tmax was significantly shorter for the HIO compared to the TIO group (p = 0.034), but no significant differences between the IV group compared to the TIO or HIO groups (p > 0.05). The odds of survival were higher in the HIO group compared to all other groups. CONCLUSION The TIO and HIO provide rapid and reliable access to administer life-saving medications during cardiac arrest.

Abstract:Limited nutrient intake in combination with the increased demand for milk production required to support a large litter often leaves sows in a severe catabolic condition and reduces sow longevity for reproduction. Dietary supplementation with yeast culture (YC) in the gestation and lactation diets of sows has been shown to increase the performance of progeny. In the present study, a novel type of YC, Saccharomyces cerevisiae refermented SDDGS (SSDDGS), was developed, and the effects of supplementing sow diets with SSDDGS from late gestation to weaning on the performance of the sows and their progeny were studied. In Exp. 1, 24 sows at 85 d of gestation were allocated to the following 3 dietary treatments: 1) sows fed a basal diet from late gestation to weaning (n = 8), 2) sows fed a diet with 2% SSDDGS (n = 8), and 3) sows fed a diet with 4% SSDDGS (n = 8). The 4% SSDDGS treatment significantly improved the sows' ADFI, the litter weaning alive rate, and the individual piglet weaning weights and significantly reduced the litter stillbirth rate and the levels of urea N and somatic cell counts (SCC) in the milk. However, the 2% SSDDGS treatment did not alter the performance of the sows or progeny. Therefore, we considered the volume of 4% SSDDGS to be more efficient than 2% SSDDGS. To verify the results of Exp. 1, we performed Exp. 2, in which 60 sows at 85 d of gestation were allocated into the following 2 dietary treatments: 1) sows fed a basal gestation diet from 85 d of gestation to weaning (n = 30) and 2) sows fed a basal diet with 4% SSDDGS from 85 d of gestation to weaning (n = 30). The 4% SSDDGS supplementation tended to increase the sows' ADFI, litter weaning size, litter weight gain during lactation, and individual piglet weaning weight and weight gain during lactation, and it also increased the milk yield and the fat and DM contents of the milk. This treatment also decreased the levels of urea N and SCC in the milk. Therefore, the present study indicates that supplementing sow diets with 4% SSDDGS from late gestation to weaning has the potential to 1) increase sow ADFI, 2) promote progeny growth performance, 3) increase sow milk production and quality, and 4) improve the maternal health status as indicated by improved protein utilization and reduced potential inflammatory response.
Supplementing Synbiotic in Sows' Diets Modifies Beneficially Blood Parameters and Colonic Microbiota Composition and Metabolic Activity in Suckling Piglets

Abstract:Nutrients in the maternal diet favor the growth and development of suckling piglets and alter their gut microbiota composition and metabolic activity, thus affecting the hosts. The present study analyzed, in suckling piglets from sows receiving antibiotic or synbiotic supplements from pregnancy to lactation, several biochemical parameters, oxidative/anti-oxidative indices, inflammatory cytokines, and ingestion-related factor levels in plasma, as well as colonic microbiota composition and metabolic activity, and mucosal expression of genes related to the intestinal barrier function. Compared with the control group, maternal synbiotic supplementation decreased ( P < 0.05) the plasma levels of glucose, AMM, TC, low-density lipoprotein-cholesterol (LDL-C), MDA, H 2 O 2 , ghrelin, CCK, PP, IL-1β, IL-2, IL-6, TNF-α, Ala, Cys, Tau, and β-AiBA, the levels of propionate and total short-chain fatty acids (SCFAs) in the colonic luminal content, and colonic abundances of RFN20, Anaerostipes , and Butyricimonas ; while increased ( P < 0.05) the plasma levels of urea nitrogen (UN), Ile, Leu, α-AAA, α-ABA, and 1-Mehis, as well as colonic abundances of Sphingomonas, Anaerovorax, Sharpea , and Butyricicoccus . Compared with the antibiotic group, maternal synbiotic supplementation decreased ( P < 0.05) the plasma levels of glucose, gastrin, and Ala, as well as abundances of Pasteurella and RFN20 and propionate level in the colonic content. Expression of genes coding for E-cadherin, Occludin, ZO-1, ZO-2, IL-10, and interferon-α were down-regulated in the colonic mucosa. The synbiotic supplementation increased ( P < 0.05) the plasma levels of UN, Leu, α-ABA, and 1-Mehis, the abundances of Anaerovorax, Sharpea , and Butyricicoccus and expression of genes coding for E-cadherin, Occludin, ZO-1, ZO-2, IL-10, and interferon-α. Spearman correlation analysis showed that there was a positive correlation between colonic Anaerostipes abundance and acetate and SCFAs levels; whereas a negative correlation between Fusobacteria and Fusobacterium abundances and acetate level. These findings suggest that synbiotic supplementation in the maternal diet improved nutrient metabolism and intestinal barrier permeability, reduced oxidative stress, and modified colonic microbiota composition and metabolic activity in suckling piglets.

Research on fault diagnosis of turbine generator unit based on improved CPN neural network

Abstract:Steam turbine generator unit is the core of thermal power plant, whose structure is complex and the operating environment is special. The fault diagnosis research of turbine generator unit has a practical significance in many aspects because of the inevitable failure of the turbine, which can improve the operational safety, reliability and the economic efficiency for the unit. In this paper, it takes the advantages of the combination of supervised and unsupervised types learning process of the Counter-Propagation Network, uses the fault spectrum feature vectors of turbine generator unit as the learning samples to train the CPN, and then improves the algorithm of CPN training process, intervenes the neurons artificially so that the information of the failure modes can be recorded within different neurons. In this way, the network can reflect the mapping relationship between the fault spectrum feature vectors and the fault types directly. Compared with the BP neural network and the improved CPN neural network, the simulation results show that the improved CPN neural network can overcome the shortcomings and deficiencies of BP neural network, can be better applied to the fault diagnosis of turbine generator unit.
Effects of Β-Carotene Microcapsules on Reproducibility, Immunity and Antioxidant Capability in Late-Pregnancy Sows.

Abstract:A total of 48 sows(Landrace × Yorkshire) with the similar reproductive conditions were used to investigate the effects of β-carotene on reproductive performance,immune function and antioxidant capability in late-pregnancy sows.These sows were allocated to four treatments(12 sows per treatment) randomly at the 80th day of pregnancy,representing supplementation with 0(control),40,100 and 160 mg·kg-1 β-carotene microcapsules to the same basal diet till to parturition.At the 95th and 110th day of pregnancy,five sows were selected randomly from each treatment to obtain blood samples for serum immunoglobulin mensuration.The colostrums were collected after parturition for IgG mensuration.Dietary supplementation with 40,100 and 160 mg·kg-1 β-carotene increased the birth mass of piglets,the concentration of colostrums IgG and total antioxidant capability of serum in the sows on the 110th day of pregnancy(P0.05).Compared with the control group,supplementation with 160 mg·kg-1 β-carotene increased the litter mass and the concentrations of serum IgG,IgM and IgA(P0.05),but decreased the activities of serum GSH-PX and SOD in the sows on the 95th and 110th day of pregnancy(P0.05).These findings indicated that dietary β-carotene supplementation for late-pregnancy sows may improve birth mass of piglets,the immune function and the antioxidant capacity of sows,and the dose of 160 mg·kg-1 is in order.

Abstract:Introduction: This study was conducted to evaluate the effects of dietary galacto-oligosaccharides (GOS) and hyocholic acids (HCA) during late gestation and lactation on reproductive performance, colostrum quality, antioxidant capacity and gut microbiota in multiparous sows. Methods: A total of 60 healthy multiparous cross-bred sows (Landrace × Yorkshire) were randomly fed 4 groups diets as follows: the basal diets (CTRL group), or the basal diets containing only 600 mg/kg GOS (GOS group), 600 mg/kg GOS + 100 mg/kg HCA (GOS + Low HCA group), and 600 mg/kg + 200 mg/kg HCA (GOS + High HCA group) from d 85 of gestation to weaning. Multiple parameters of sows were determined. Results: There was a trend of shortening the labor process of sows ( p = 0.07) in the GOS group and GOS + Low/High HCA group. Compared with the CTRL group, the GOS + Low/High HCA group increased the average piglets weight at birth ( p < 0.05), and increased the IgA concentration of colostrum ( p < 0.05). In addition, serum triglyceride (TG) concentration was lower ( p < 0.05), and serum total antioxidant capacity (T-AOC) was higher ( p < 0.05) in the GOS and GOS + Low/High HCA groups than in the CTRL group at farrowing. Serum catalase (CAT) activities was higher in the GOS and GOS + High HCA groups than in the CTRL group at farrowing. The 16S rRNA analysis showed that GOS combination with high-dose HCA shaped the composition of gut microbiota in different reproductive stages (d 107 of gestation, G107; d 0 of lactation, L0; d 7 of lactation, L7). At the phylum level, the relative abundance of Bacteroidota and Desulfobacterota in G107, Bacteroidota , and Proteobacteria in L0, and Planctomycetota in L7 was increased in GOS + High HCA group ( p < 0.05). Spearman correlation analysis showed that Streptococcus was positively correlated with the serum TG but negatively correlated with the average piglets weight at birth ( p < 0.05). Conclusion: This investigation demonstrated that the administration of galacto-oligosaccharides (GOS) in conjunction with hyocholic acids (HCA), to sows with nutrient restrictions during late gestation and lactation, further improved their antioxidant capacity and milk quality. The observed beneficial effects of GOS + HCA supplementation could potentially be linked to an improvement in gut microbiota disorders of the sows.

Abstract:Sows undergo physiological stress during gestation and lactation, potentially leading to enteric dysbiosis and reduced reproductive potential. Phytogenic additives (PFs) may improve performance via their antioxidant, anti-inflammatory and antimicrobial properties. This study determined whether the provision of a gestation/lactation diet containing PAs would alter the gastrointestinal microbiota of sows and their piglets, and improve performance. Sows received a commercial diet throughout gestation and lactation (CTR; n = 64), a commercial diet throughout gestation and a diet containing PAs in lactation (CTR-PA; n = 63) or a commercial diet containing PAs in gestation and lactation (PA; n = 90). Sows were weighed and backfat recorded after mating and at entry and exit from the farrowing house and piglets were weighed on days 1 and 21 of life. Faecal samples collected from sows at farrowing house entry and piglets at 21 and 35 d were subjected to 16 S rRNA gene amplicon analysis. The addition of PAs to sow diets resulted in more piglets born (P = 0.03), however, it did not improve the number of liveborn piglets (P = 0.14). There were no differences in sow weight, P2 backfat depth or lactation feed intake observed. PAs had no effect on piglet weight or survival to weaning but did alter the faecal microbiota of sows, and this change was observed in piglets at 21 and 35 d. PA supplementation to sows has the potential to increase litter size, while also potentially influencing gastrointestinal tract health of the sow and piglets reared.
Enhanced Antioxidative Capacity Transfer between Sow and Fetus via the Gut–Placenta Axis with Dietary Selenium Yeast and Glycerol Monolaurate Supplementation during Pregnancy

Abstract:This study aims to investigate the impact of dietary supplementation with selenium yeast (SeY) and glycerol monolaurate (GML) on the transfer of antioxidative capacity between the mother and fetus during pregnancy and its underlying mechanisms. A total of 160 sows with similar body weight and parity of 3–6 parity sows were randomly and uniformly allocated to four groups (n = 40) as follows: CON group, SeY group, GML group, and SG (SeY + GML) group. Animal feeding started from the 85th day of gestation and continued to the day of delivery. The supplementation of SeY and GML resulted in increased placental weight and reduced lipopolysaccharide (LPS) levels in sow plasma, placental tissues, and piglet plasma. Furthermore, the redox balance and inflammatory markers exhibited significant improvements in the plasma of sows fed with either SeY or GML, as well as in their offspring. Moreover, the addition of SeY and GML activated the Nrf2 signaling pathway, while downregulating the expression of pro-inflammatory genes and proteins associated with inflammatory pathways (MAPK and NF-κB). Vascular angiogenesis and nutrient transportation (amino acids, fatty acids, and glucose) were upregulated, whereas apoptosis signaling pathways within the placenta were downregulated with the supplementation of SeY and GML. The integrity of the intestinal and placental barriers significantly improved, as indicated by the increased expression of ZO-1, occludin, and claudin-1, along with reduced levels of DLA and DAO with dietary treatment. Moreover, supplementation of SeY and GML increased the abundance of Christensenellaceae_R-7_group, Clostridium_sensus_stricto_1, and Bacteroidota, while decreasing levels of gut microbiota metabolites LPS and trimethylamine N-oxide. Correlation analysis demonstrated a significant negative relationship between plasma LPS levels and placental weight, oxidative stress, and inflammation. In summary, dietary supplementation of SeY and GML enhanced the transfer of antioxidative capacity between maternal-fetal during pregnancy via gut–placenta axis through modulating sow microbiota composition.

Abstract:Section 504 of the Rehabilitation Act of 1973 prohibits discrimination against the handicapped in any program receiving federal funds. With its implementing regulations, Section 504 may well apply to the selection of patients for medical treatments. This article examines patient selection for heart transplantation, in light of the Rehabilitation Act and its previous application in similar (but non-medical) contexts. With the growing need to allocate scarce medical resources comes a need to examine carefully the legal bounds for patient-selection procedures.

Abstract:Maternal diet has a profound impact on growth and immune development of offspring. This study aimed to evaluate the effects of maternal supplementation with a combination of wheat bran (WB, a source of insoluble dietary fiber) and sugar beet pulp (SBP, a source of soluble dietary fiber) on growth and intestinal morphology, immunity, barrier function and microbiota in piglets. Thirty sows (Landrace × Yorkshire; 3-6 parity) were randomly allocated to 2 dietary treatments from d 85 of gestation to weaning (d 21 of lactation). The 2 dietary treatments were: a control diet (CON, a corn-soybean meal diet) and a dietary fiber diet (DF, 15% WB and 10% SBP during gestation and 7.5% WB and 5% SBP during lactation). Maternal DF supplementation improved growth, serum growth hormones and ileal morphology in piglets. Piglets fed DF showed enhanced intestinal barrier function as indicated by reduced serum concentrations of diamine oxidase and endotoxin, and increased ileal mRNA level of occludin. Maternal DF supplementation reduced pro-inflammatory cytokines in the colostrum, milk and serum of piglets. Furthermore, maternal DF supplementation decreased the colonic abundance of Subdoligranulum and Mogibacterium, and increased the colonic abundance of Lactobacillus and norank_f__Bacteroidales_S24-7_group and the colonic concentration of acetate and butyrate in piglets. In summary, maternal supplementation with a combination of SBP and WB during late gestation and lactation improved cytokines in colostrum and milk, growth, immune responses, intestinal morphology, barrier function and microbiota in piglets, which may be a potential strategy to improve offspring growth and intestinal functions.

Abstract:In piglets, it is observed that early weaning can lead to poor weight gain due to an underdeveloped gastrointestinal (GI) tract, which is unsuitable for an efficient absorption of nutrients. Short-chain fatty acids (SCFAs) such as butyrate have demonstrated their ability to improve intestinal development by increasing cell proliferation, which is vital during this transition period when the small and large intestinal tracts are rapidly growing. Previous reports on butyrate inclusion in feed demonstrated significantly increased feed intakes (FIs) and average daily gains (ADGs) during piglet weaning. Similar benefits in piglet performance have been observed with the inclusion of yeast cell wall in diets. A proprietary mix of yeast cell wall, SCFAs, and zinc proteinate (YSM) was assessed here in vitro to determine its impact on cellular growth, metabolism and appetite-associated hormones in ex vivo small intestinal pig cells and STC-1 mouse intestinal neuroendocrine cells. Intestinal cells demonstrated greater cell densities with the addition of YSM (150 ppm) compared to the control and butyrate (150 ppm) at 24 h. This coincided with the higher utilisation of both protein and glucose from the media of intestinal cells receiving YSM. Ghrelin (an appetite-inducing hormone) demonstrated elevated levels in the YSM-treated cells on a protein and gene expression level compared to the cells receiving butyrate and the control, while satiety hormone peptide YY protein levels were lower in the cells receiving YSM compared to the control and butyrate-treated cells across each time point. Higher levels of ghrelin and lower PYY secretion in cells receiving YSM may drive the uptake of protein and glucose, which is potentially facilitated by elevated gene transporters for protein and glucose. Greater ghrelin levels observed with the inclusion of YSM may contribute to higher cell densities that could support pig performance to a greater extent than butyrate alone.

Abstract:A total of 20 healthy white × landrace sows were evenly and randomly divided into two groups, and fed basal diets unsupplemented or supplemented with 500 g/t Meriden-Stim® from day 100 of gestation until day 21 of lactation. Serum and fecal samples were collected from the sows on the final day for subsequent analysis. Compared to the control group, there were no significant differences in the sows’ performances; however, an increase was observed in the piglets’ weight at weaning (p = 0.08). Moreover, oregano essential oil (OEO) significantly reduced the levels of urea (UREA) (p < 0.01), total cholesterol (TC) (p < 0.05), low-density lipoprotein (LDL-C) (p < 0.05) and alanine aminotransferase (ALT) (p < 0.05) in serum. In terms of antioxidant indexes in serum, the catalase (CAT) and glutathione (GSH) levels showed significant increases (p < 0.05) while the malondialdehyde (MDA) level exhibited a decrease tendency (p = 0.09). 16S rRNA analysis identified the specific bacteria taxa in feces. OEO significantly decreased the relative abundance of Proteobacteria and Actinobacteria at the phylum level (p < 0.05). At the genus level, OEO significantly increased the relative abundance of Lactobacillus and Prevotellaceae UCG 003 and UCG 005, while decreasing that of Escherichia-Shigella (p < 0.05). Taken together, OEO supplementation in maternal diets during late gestation and lactation improved serum metabolites, antioxidant capacity and regulated the intestinal-flora balance of sows, thereby tending to increase the piglets’ weight at weaning.
