Abstract:Brown rot, caused by Monilinia fructicola, is considered one of the devasting diseases of pre-harvest and post-harvest peach fruits, restricting the yield and quality of peach fruits and causing great economic losses to the peach industry every year. Presently, the management of the disease relies heavily on chemical control. In the study, we demonstrated that the volatile organic compounds (VOCs) of endophyte bacterial Pseudomonas protegens QNF1 inhibited the mycelial growth of M. fructicola by 95.35% compared to the control, thereby reducing the brown rot on postharvest fruits by 98.76%. Additionally, QNF1 VOCs severely damaged the mycelia of M. fructicola. RNA-seq analysis revealed that QNF1 VOCs significantly repressed the expressions of most of the genes related to pathogenesis (GO:0009405) and integral component of plasma membrane (GO:0005887), and further analysis revealed that QNF1 VOCs significantly altered the expressions of the genes involved in various metabolism pathways including Amino acid metabolism, Carbohydrate metabolism, and Lipid metabolism. The findings of the study indicated that QNF1 VOCs displayed substantial control efficacy by disrupting the mycelial morphology of M. fructicola, weakening its pathogenesis, and causing its metabolic disorders. The study provided a potential way and theoretical support for the management of the brown rot of peach fruits.
Streptomyces virginiae XDS1-5, an antagonistic actinomycete, as a biocontrol to peach brown rot caused by Monilinia fructicola

Abstract:Background: Peach brown rot, caused by the pathogen Monilinia fructicola, represents a significant postharvest infectious disease affecting peach fruit. This disease is responsible for a substantial increase in fruit decay rates, leading to significant economic losses, often exceeding 50%. Currently, there is a growing interest in identifying biocontrol agents to mitigate peach brown rot, with a predominant interest in Bacillus species. Results: In this investigation, we isolated 410 isolates of actinomycetes from non-farmland ecosystem soil samples. Subsequently, 27 isolates exhibiting superior inhibitory capabilities were selected. Among these, strain XDS1-5 demonstrated the most robust fungistatic effect against brown rot disease, achieving an 80% inhibition rate in vitro and a 66% inhibition rate in vivo. XDS1-5 was identified as belonging to the Streptomyces virginiae species. Furthermore, a fermentation filtrate of XDS1-5 exhibited the ability to metabolize 34.21% of the tested carbon sources and 7.37% of the tested nitrogen sources. Particularly noteworthy was its capacity to disrupt the cell membrane structure directly, leading to increased cell membrane permeability and cytoplasmic leakage. Additionally, our investigation indicated that indoline, a metabolite produced by XDS1-5, played a pivotal role in inhibiting the growth of M. fructicola. Conclusion: In summary, our study has identified a biocontrol actinomycete, XDS1-5, with the potential to effectively inhibit postharvest brown rot disease in peaches. This finding holds great significance for the biological control of peach brown rot, offering promising prospects for mitigating the economic losses associated with this devastating disease. © 2024 Society of Chemical Industry.

Abstract:Microorganism-produced volatile organic compounds (VOCs) are considered promising environmental-safety fumigants in food preservation. In this study, the VOCs from fungal Ceratocystis fimbriata strains (WSJK-1, Mby) were tested against postharvest fungi Monilinia laxa , Fusarium oxysporum , Monilinia fructicola , Botrytis cinerea , Alternaria solani , and Aspergillus flavus in vitro. The mycelial growth was significantly inhibited, in particular M. fructicola and B. cinerea (76.95, 76.00%), respectively. VOCs were identified by headspace solid-phase microextraction coupled with Gas Chromatography–Mass Spectrometry (HS-SPME-GC–MS); 40 compounds were identified. The antifungal activity of 21 compounds was tested by the minimum inhibitory concentrations (MIC) value. Benzaldehyde, 2-Phenylethanol, and 1-Octen-3-ol showed strong antifungal activity with the MIC in vitro ranging from 0.094 to 0.284 ml L −1 depending on the pathogen tested. The optical microscope showed serious morphological damage, including cell deformation, curling, collapse, and deficiency in mycelial or conidia cell structures treated with C. fimbriata VOCs and pure compounds. In vivo tests, C. fimbriata VOCs decreased brown rot severity in peaches, and compounds Benzaldehyde and 2-Phenylethanol could reduce peach brown rot in peaches at 60 μl L −1 . The VOCs produced by C. fimbriata strain have good antifungal effects; low concentration fumigation could control peach brown rot. Its fragrance is fresh, safe, and harmless, and it is possible to replace chemical fumigants. It could be used as a potential biofumigant to control fruit postharvest transportation, storage, and food preservation. To the best of our knowledge, this is the first report on the antifungal activity and biocontrol mechanism of VOCs produced by C. fimbriata .

Abstract:Three endophytic Muscodor isolates isolated from needle tissues of the critically threatened Abies beshanzuensis can produce intense volatiles in vitro, which in general cause obvious inhibition or even death to a wide range of plant pathogens, indicating their strong antibiotic activity. In this paper, the fungal agents derived from solid-state fermentation were applied to efficiently control the fruit postharvest diseases, including Botrytis cinerea, Physalospora pyricola and Monilinia fructicola. The SPME-GC-MS technique was used to detect the volatile profiling of Mucodor, and the result showed that there were obvious difference in the chemical components and relative content among the three Mucodor isolates. Muscodor sp. M25 had relatively simple chemical composition of volatiles, while Muscodor fengyangensis M112-2 and M153 had complicated composition. The difference in their chemical composition of volatiles was, to some degree, correlated with their bioactivity. Propanoic acid, 2-methyl-, propanoic acid, 2-methyl-, methyl ester and β-phellandrene were the main volatile small molecules. Thus, those Muscodor isolates would be ideal candidates to be exploited as novel ‘myco-fumigation’ agents.

Abstract:We examine the statistical power of fundamental and behavioral factors with regards to stock returns of the Dow Jones Industrials Index. With a novel sentiment dataset from over 3.6 million Reuters news articles, we nd signi cant correlations between Reuters sentiment and stock returns. We show with vector autoregression and error correction models that sentiment can explain and predict changes in stock returns better than macroeconomic factors. Considering positive and negative sections of Reuters sentiment, we nd that negative sentiment performs better in simple trading strategies to predict stock returns than positive sentiment, while the sentiment e ect remains over months.
Screening of core microorganisms in healthy and diseased peaches and effect evaluation of biocontrol bacteria (Burkholderia sp.)

Abstract:Biological antagonists serve as the most important green alternatives to chemical fungicides, a class of microorganism that inhibits the growth of pathogenic fungi to reduce fruit incidence. In this paper, healthy and diseased peach fruit was selected for amplicon sequencing of the epiphytic microbiota on their surface to obtain a comprehensive understanding. Community structure, diversity and LefSe analysis were performed to screen Acetobacter, Muribaculaceae and Burkholderia as the core bacteria, Mycosphaerella, Penicillium and Alternaria as the core fungi, they showed significant differences and were highly enriched. Two strains fungi (Penicillium K3 and N1) and one strain antagonistic bacteria (Burkholderia J2) were isolated. The in intro test results indicated the bacterial suspension, fermentation broth and volatile organic compounds of antagonistic bacteria J2 were able to significantly inhibit pathogen growth. In vivo experiments, peach was stored at 28 °C for 6 days after different treatments, and samples were taken every day. It was found that Burkholderia J2 enhanced peach resistance by increasing the activities of antioxidant-related enzymes such as SOD, POD, PAL, PPO, GR, MDHAR, and DHAR. The results improved that Burkholderia J2 has great biocontrol potential and could be used as a candidate strain for green control of blue mold.

Abstract:Monilinia fructicola is the causal agent of brown rot disease in several species of fruit crops. Volatile organic compounds (VOCs) produced by some microorganisms have been shown to inhibit postharvest disease development in fruit crops and are being explored for use in disease management. Our previous studies have shown that Bacillus subtilis, strain CF-3 VOCs can inhibit the growth of M. fructicola [ACCC 36262]. In the present study, the effect of B. subtilis CF-3 VOCs on M. fructicola was subjected to transcriptomic and proteomic analyses to provide a theoretical basis for the mechanism of action of CF-3 VOCs. A total of 1,602 differentially expressed genes (DEGs) and 1,008 differentially expressed proteins (DEPs) were identified by quantitative transcriptome (RNA-sequencing) and quantitative proteomics (Tandem Mass Tag, TMT). The DEGs and DEPs were mainly associated with the GO and KEGG categories cell wall, cell membrane, extracellular enzyme, energy metabolism, and ribosome biosynthesis. Our results indicate that CF-3 VOCs influence both the growth and pathogenicity of M. fructicola and provide a theoretical basis for the potential mechanism of action of B. subtilis CF-3 in the protection of fruits against M. fructicola and perhaps other pathogens.

The Potential of Novel Bacterial Isolates from Natural Soil for the Control of Brown Rot Disease (Monilinia fructigena) on Apple Fruits

Abstract:Monilinia fructigena is one of the most important fungal pathogens causing brown rot on apple and is heavily affecting fruit production. The main objective of this study was to screen for potential bacterial isolates with higher antagonistic activity against M. fructigena. Our study focused on the identification of potential bacterial isolates capable of reducing both the mycelial growth of M. fructigena and the disease severity using in vitro and in planta trials, respectively. To achieve this goal, thirteen bacteria, isolated from natural soil, were evaluated for their abilities to produce lytic enzymes (amylase, cellulase and protease), hydrocyanic acid (HCN) and lipopeptides (bacillomycin, fengycin, iturin and surfactin). Further, results from the dual culture method, volatile and bacterial free-cell filtrate bioassays indicated that tested isolates showed a fungicidal activity against the mycelial growth of M. fructigena. Thus, out of the 13 isolates tested, 12 exhibited significant mycelial inhibition (more than 70%) against M. fructigena, while remaining the last isolate displayed only a partial inhibition (up to 43%). Further, 12 of the bacteria isolates displayed an amylase production, 10 showed cellulase production, 11 revealed protease production, while only 2 displayed HCN production. In addition, most bacterial isolates were found to have genes encoding for different lipopeptides: bacillomycin (10), fengycin (3), iturin (11) and surfactin (1). Interestingly, two bacterial isolates, Bacillus amyloliquefaciens B10W10 and Pseudomonas sp. B11W11 were found to be the most effective and displayed the lowest disease severity in planta trial. These two bacteria reduced the brown rot incidence compared to the synthetic fungicide in a semi-commercial large-scale trial. Therefore, our findings suggest that these two later bacterial isolates provide apple protection against M. fructigena via direct and indirect mechanisms. These isolates may be used, therefore, as potential biological control agents (BCAs) in preventive treatment to control brown rot disease on apple fruits.
Inhibition of Monilinia fructicola sporulation and pathogenicity through eucalyptol-mediated targeting of MfCat2 by Streptomyces lincolnensis strain JCP1-7

Abstract:Peach brown rot, attributed to Monilinia fructicola, presents a significant threat to postharvest peach cultivation, causing losses of up to 80%. With an increasing number of countries, spearheaded by the European Union, imposing bans on chemical agents in fruit production, there is a growing interest in mining highly active antibacterial compounds from biological control strains for postharvest disease management. In this study, we highlight the unique ability of Streptomyces lincolnensis strain JCP1-7 to inhibit M. fructicola sporulation, despite its limited antimicrobial efficacy. Through GC-MS analysis, eucalyptol was identified as the key compound. Fumigation of diseased fruits with eucalyptol at a concentration of 0.0335 μg cm-3 demonstrated an in vivo inhibition rate against M. fructicola of 93.13%, completely suppressing spore formation. Transcriptome analysis revealed the impact of eucalyptol on multiple pathogenesis-related pathways, particularly through the inhibition of catalase 2 (Cat2) expression. Experiments with a MfCat2 knockout strain (ΔMfCat2) showed reduced pathogenicity and sensitivity to JCP1-7 and eucalyptol, suggesting MfCat2 as a potential target of JCP1-7 and eucalyptol against M. fructicola. Our findings elucidate that eucalyptol produced by S. lincolnensis JCP1-7 inhibits M. fructicola sporulation by regulating MfCat2, thereby effectively reducing postharvest peach brown rot occurrence. The use of fumigation of eucalyptol offers insights into peach brown rot management on a large scale, thus making a significant contribution to agricultural research.
Control Effects of Bacillus Siamensis G-3 Volatile Compounds on Raspberry Postharvest Diseases Caused by Botrytis Cinerea and Rhizopus Stolonifer

Abstract:Post-harvest diseases of raspberries affect their storage and subsequently the ability to provide consumers with fresh products. The antifungal activity of microbial volatile organic compounds (MVOCs) against both pre and postharvest diseases has previously been demonstrated, but the control of raspberry postharvest diseases caused by Botrytis cinerea and Rhizopus stolonifer using MVOCs has not been extensively studied. We have thus investigated the effects of Bacillus siamensis G-3 volatile organic compounds (VOCs) on controlling the diseases caused by B. cinerea and R. stolonifer. Bioassays of the VOCs of B. siamensis G-3 exhibited significant control effects (89.4%) on the two pathogens. The VOCs composition was studied with solid phase micro extraction and GC/mass spectrometry (SPME-GC-MS), and 22 VOCs, different from the controls, were identified. Among these compounds, only 2, 6-di-tert-butyl-4-methylphenol (BHT), and 2,4-di-tert-butylphenol (2,4-DTBP) had significant control effects on the pathogens (*P < 0.05), 52.38 and 93.33%, respectively, compared with the controls in vivo. With the 2,4-DTBP treatments, the raspberries could be stored for 20 d at 0 degrees C, with the disease rate maintained at similar to 10%. In addition, we have reported for the first time, that 2,4-DTBP may be a better preservative than BHT and could be used as a bio-fumigant for controlling raspberry postharvest diseases caused by B. cinerea and R. stolonifer in vivo.

Abstract:urine of adrenalectomized rats decreased significantly, whereas there was slight increase in that of dioxogulonate. Also, there was slight decrease in the total ascorbate content of blood. Decreases of about 20 and 30% were observed in the activities of D-glucurono-8-lactone hydrolase and L-gulono-y-lactone oxidase respectively, whereas that of L-gulono-y-lactone hydrolase remained practically unchanged. An appreciable increase in the activity of dehydroascorbatase and a moderate increase in that of dioxoaldonate decarboxylase were observed in liver and kidney of adrenalectomized rats. All corresponding alterations were more conspicuous in rats 12 days after adrenalectomy.

Abstract:Fruit discoloration and fruit rot disease in longan is a serious disease caused by the pathogenic fungi Pestalotiopsis oxyanthi and Lasiodiplodia pseudotheobromae. This study investigated the potential of antagonistic bacteria as an eco-friendly alternative to traditional fungicides for managing longan diseases caused by these fungal pathogens. Two bacterial isolates, UP-JLS050 and UP-JLS067, demonstrated significant inhibitory activity against the pathogenic fungi P. oxyanthi and L. pseudotheobromae, with high inhibitory activity. The bacterial strains were isolated from leaves and soil in a longan garden, and dual culture methods showed that the isolates UP-JLS050 and UP-JLS067 exhibited the highest inhibitory activity on the fungal pathogens, with percentage inhibitions of 72.67 ± 2.31 and 70.37 ± 1.28 %, respectively. Biochemical methods and molecular techniques, including sequencing of the 16S rDNA gene, were then used to identify the isolates. This confirmed their similarity to Bacillus subtilis and Bacillus amyloliquefaciens with 100 and 99 % identity, respectively. In vitro tests on longan fruit indicated that these antagonistic bacteria, administered at a concentration of 108 CFU/mL, significantly reduced disease incidence by up to 45 % after 9 days of incubation. The incubation of B. subtilis (UP-JLS050) and B. amyloliquefaciens (UP-JLS050) reduced disease incidence by 45 ± 5.77 and 32.5 ± 5.00 %, respectively. Identification of the Fungal Pathogens of Postharvest Disease on Peach Fruits and the Control Mechanisms of Bacillus subtilis JK-14

Abstract:Postharvest fungal disease is one of the significant factors that limits the storage period and marketing life of peaches, and even result in serious economic losses worldwide. Biological control using microbial antagonists has been explored as an alternative approach for the management of postharvest disease of fruits. However, there is little information available regarding to the identification the fungal pathogen species that cause the postharvest peach diseases and the potential and mechanisms of using the Bacillus subtilis JK-14 to control postharvest peach diseases. In the present study, a total of six fungal isolates were isolated from peach fruits, and the isolates of Alternaria tenuis and Botrytis cinerea exhibited the highest pathogenicity and virulence on the host of mature peaches. In the culture plates, the strain of B. subtilis JK-14 showed the significant antagonistic activity against the growth of A. tenuis and B. cinerea with the inhibitory rates of 81.32% and 83.45% at 5 days after incubation, respectively. Peach fruits treated with different formulations of B. subtilis JK-14 significantly reduced the mean disease incidences and lesion diameters of A. tenuis and B. cinerea. The greatest mean percent reduction of the disease incidences (81.99% and 71.34%) and lesion diameters (82.80% and 73.57%) of A. tenuis and B. cinerea were obtained at the concentration of 1 × 107 CFU mL−1 (colony forming unit, CFU). Treatment with the strain of B. subtilis JK-14 effectively enhanced the activity of the antioxidant enzymes-superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) in A. tenuis and B. cinerea inoculated peach fruits. As such, the average activities of SOD, POD and CAT were increased by 36.56%, 17.63% and 20.35%, respectively, compared to the sterile water treatment. Our results indicate that the isolates of A. tenuis and B. cinerea are the main pathogens that cause the postharvest peach diseases, and the strain of B. subtilis JK-14 can be considered as an environmentally-safe biological control agent for the management of postharvest fruits diseases. We propose the possible mechanisms of the strain of B. subtilis JK-14 in controlling of postharvest peach diseases.

Abstract:Gray mold and brown rot, caused respectively by Botrytis cinerea and Monilinia spp., are fungal diseases responsible for significant losses during the storage of fruit and vegetables. Nowadays, the control of postharvest diseases is shifting towards more sustainable strategies, including the use of plant secondary metabolites. In this study, the antifungal activity of Origanum vulgare, Thymus vulgaris, Thymus serpyllum, Melaleuca alternifolia, Lavandula officinalis, Lavandula hybrida, Citrus bergamia, Rosmarinus officinalis, Cinnamomum zeylanicum essential oils (EOs) in vapor phase was tested in vitro against B. cinerea, Monilinia fructicola, Monilinia fructigena, and Monilinia laxa. For the experiments, a protocol using a volatile organic compounds (VOC) chamber was designed. Results indicate a dose-dependent inhibitory activity of all the tested EOs, with O. vulgare, T. vulgaris, and T. serpyllum being the most active ones, with minimum inhibitory concentrations (MIC) of 22.73, 45.45, and 22.73 µl/L, respectively, against B. cinerea and a range between 5.64 and 22.73 µl/L against the three Monilinia spp. Overall, B. cinerea presented lower sensitivity to vapor-phase EOs than any of the Monilinia strains, except for the C. zeylanicum EO, which consistently showed higher inhibition against B. cinerea. Among the three Monilinia spp., M. fructicola was the least sensitive, while M. fructigena was the most sensitive. The use of VOC chambers proved to be a reliable protocol for the assessment of antimicrobial activities of EOs. These results suggest that the VOC emitted by the tested EOs are effective towards important decay-causing fungi, and that they could be used for the control of gray mold and brown rot in in vivo trials.
Biocontrol potential of volatile organic compounds from Pseudomonas chlororaphis ZL3 against postharvest gray mold caused by Botrytis cinerea on Chinese cherry

Abstract:Postharvest gray mold is the most devastating fungal disease, and could lead to enormous losses on Chinese cherry. To formulate an effective and environment friendly management strategy for postharvest gray mold, an endophytic bacterial strain ZL3 with excellent antifungal activity was screened by using dual culture method. Strain ZL3 was identified as Pseudomonas chlororaphis based on 16S rRNA and gyrB gene sequence analysis. The maximum percent of mycelial growth inhibition (PGI) of VOCs to B. cinerea was up to 97.23% under the antifungal conditions (temperature of 31.96 °C, pH of 6.74 and glucose of 25.96 g/L) optimized by using Box-Behnken Design (BBD) of response surface method (RSM). The biological efficiency of VOCs was performed on cherry fruit at 0 °C for 21 d and 25 °C for 7 d. At 25 °C and 0 °C, the disease incidence reduced to 31.67% and 21.67%, whereas the lesion diameter decreased to 0.60 cm and 0.32 cm, respectively. Additionally, twenty-three VOCs, including 6 alkanes, 4 aldehydes and ketones, 4 alcohols, 4 alkenes, 3 acids & esters, 1 aromatic compound, and 1 sulfur compound, were detected by gas chromatograph-mass spectrometry (GC-MS). It is noticed that most VOCs, except for 1, 10-undecadiene and 1-dodecene, demonstrated significant differences among different collection time. The results of this work suggested that strain ZL3 could be used as a promising biocontrol agent to control postharvest gray mold on Chinese cherry.

Abstract:Phytopathogenic fungal growth in postharvest fruits and vegetables is responsible for 20–25% of production losses. Volatile organic compounds (VOCs) have been gaining importance in the food industry as a safe and ecofriendly alternative to pesticides for combating these phytopathogenic fungi. In this study, we analysed the ability of some VOCs produced by strains of the genera Bacillus , Peribacillus , Pseudomonas , Psychrobacillus and Staphylococcus to inhibit the growth of Alternaria alternata , Botrytis cinerea , Fusarium oxysporum , Fusarium solani , Monilinia fructicola , Monilinia laxa and Sclerotinia sclerotiorum , in vitro and in vivo . We analysed bacterial VOCs by using GC/MS and 87 volatile compounds were identified, in particular acetoin, acetic acid, 2,3-butanediol, isopentanol, dimethyl disulphide and isopentyl isobutanoate. In vitro growth inhibition assays and in vivo experiments using cherry fruits showed that the best producers of VOCs, Bacillus atrophaeus L193, Bacillus velezensis XT1 and Psychrobacillus vulpis Z8, exhibited the highest antifungal activity against B. cinerea , M. fructicola and M. laxa , which highlights the potential of these strains to control postharvest diseases. Transmission electron microscopy micrographs of bacterial VOC-treated fungi clearly showed antifungal activity which led to an intense degeneration of cellular components of mycelium and cell death.

Abstract:To select antagonistic yeasts for the control of fruit rot caused by Lasiodiplodia theobromae and anthracnose caused by Colletotrichum gloeosporioides in postharvest mango fruit, 307 yeast strains isolated from plant leaves were evaluated for their antagonistic activities against these two fungal pathogens in vitro. Torulaspora indica DMKU-RP31, T. indica DMKU-RP35 and Pseudozyma hubeiensis YE-21 were found to inhibit the growth of L. theobromae whereas only Papiliotrema aspenensis DMKU-SP67 inhibited the growth of C. gloeosporioides. Antagonistic mechanisms of these four antagonistic yeasts in vitro consisted of the production of antifungal volatile organic compounds (VOCs), biofilm formation and siderophore production. T. indica DMKU-RP35 was the most effective strain in controlling fruit rot on postharvest mango fruits. Its action was comparable to that of the fungicide, benomyl, reducing the disease severity by 82.4%, whereas benomyl revealed 87.5% reduction. P. aspenensis DMKU-SP67 reduced anthracnose severity by 94.1%, which was comparable to that of using benomyl (93.9%). The antifungal VOCs produced by these yeast strains also reduced the severity of these diseases on postharvest mango fruits but at lower rates than using yeast cells. Therefore, these antagonistic yeasts have the potential for use as biological control agents for the control of fruit rot and anthracnose diseases.

Abstract:Loquat fruit is one of the most perishable fruits in China, and has a very limited shelf life because of mechanical injury and microbial decay. Due to an increasing concern about human health and environmental security, antagonistic microorganisms have been a potential alternative for fungicides to control postharvest diseases. In this work, the antifungal effect of volatile organic compounds (VOCs) produced byBacillus methylotrophicusBCN2 andBacillus thuringiensisBCN10 against five postharvest pathogens isolated from loquat fruit,Fusarium oxysporum,Botryosphaeriasp.,Trichoderma atroviride,Colletotrichum gloeosporioides,andPenicillium expansumwere evaluated by in vitro and in vivo experiments. As a result, the VOCs released by BCN2 and BCN10 were able to suppress the mycelial growth of all targeted pathogens according to inhibition ratio in the double petri-dish assay as well as disease incidence and disease diameter on loquat fruits. The main volatile compounds were identified by solid-phase microextraction (SPME)-gas chromatography. These VOCs produced by the two strains played complementary roles in controlling these five molds and enabled loquat fruits to keep fresh for ten days, significantly. This research will provide a theoretic foundation and technical support for exploring the functional components of VOCs applicable in loquat fruit preservation.
Unleashing the Potential of Bacterial Isolates from Apple Tree Rhizosphere for Biocontrol of Monilinia laxa: A Promising Approach for Combatting Brown Rot Disease

Abstract:Monilinia laxa, a notorious fungal pathogen responsible for the devastating brown rot disease afflicting apples, wreaks havoc in both orchards and storage facilities, precipitating substantial economic losses. Currently, chemical methods represent the primary means of controlling this pathogen in warehouses. However, this study sought to explore an alternative approach by harnessing the biocontrol potential of bacterial isolates against brown rot in apple trees. A total of 72 bacterial isolates were successfully obtained from the apple tree rhizosphere and subjected to initial screening via co-cultivation with the pathogen. Notably, eight bacterial isolates demonstrated remarkable efficacy, reducing the mycelial growth of the pathogen from 68.75 to 9.25%. These isolates were subsequently characterized based on phenotypic traits, biochemical properties, and 16S rRNA gene amplification. Furthermore, we investigated these isolates’ production capacity with respect to two enzymes, namely, protease and chitinase, and evaluated their efficacy in disease control. Through phenotypic, biochemical, and 16S rRNA gene-sequencing analyses, the bacterial isolates were identified as Serratia marcescens, Bacillus cereus, Bacillus sp., Staphylococcus succinus, and Pseudomonas baetica. In dual culture assays incorporating M. laxa, S. marcescens and S. succinus exhibited the most potent degree of mycelial growth inhibition, achieving 68.75 and 9.25% reductions, respectively. All the bacterial isolates displayed significant chitinase and protease activities. Quantitative assessment of chitinase activity revealed the highest levels in strains AP5 and AP13, with values of 1.47 and 1.36 U/mL, respectively. Similarly, AP13 and AP6 exhibited the highest protease activity, with maximal enzyme production levels reaching 1.3 and 1.2 U/mL, respectively. In apple disease control assays, S. marcescens and S. succinus strains exhibited disease severity values of 12.34% and 61.66% (DS), respectively, highlighting their contrasting efficacy in mitigating disease infecting apple fruits. These findings underscore the immense potential of the selected bacterial strains with regard to serving as biocontrol agents for combatting brown rot disease in apple trees, thus paving the way for sustainable and eco-friendly alternatives to chemical interventions.
