Abstract:A novel perphenyl carbamate of beta-cyclodextrin immobilized onto aminized silica gel via multiple ureido chemical bonds was synthesized and its enantioseparation ability was evaluated as chiral stationary phase (CSP) through separation study of fluoxetine (FLU) on high performance liquid chromatography under reversed phase condition. The optimal separation condition of FLU was found and a good separation was obtained. The influences of mobile phase composition, ionic strength, pH, flow rates of mobile phase, and column temperature on retention behavior, resolution of (R)-FLU and (S)-FLU were studied. The results showed that the new CSP had good separation ability and built a strong basis for the large-scale separation of FLU.

Abstract:Fluoxetine hydrochloride enantiomers were separated by chromatography on Tris-(3,5-dimethylphenyl carbamate) cellulose as a chiral stationary phase (CSP). The effects of mobile phase composition, flow rate, and column temperature on capacity factor, separation factor, and resolution were studied systematically. Best separation was obtained with a mobile phase composition of hexane/isopropyl alcohol/diethylamine of 98/2/0.2 (v/v/v) at 15 degrees C. The optimal flow rate was 0.24 mL/min from the Van Deemter equation. However, baseline resolution (R S >1.5) was achieved under a flow rate of 0.8 mL/min. Thermodynamic parameters Delta H 0 , Delta S,R Delta H 0 , and Delta S,R Delta S 0 were calculated. In the temperature range examined, the enthalpic contribution to the enantiomer transfer energy was found to be more significant than the entropic one.

Abstract:Objective:Chromatographic method using amylose chiral stationary phase for separation of fluoxetine hydrochloride enantiomers has been developed.Tris (3,5-dimethylphenyl carbamate)amylose chiral stationary phase was used in the normal-phase mode.The effects of mobile phase composition,flow rate and column tempera- ture on the ehiral separation were discussed systematically.Method:The column used was Chiralpak ADH (250mm ×4.6mm,5μm).The mobile phase studied was hexane alcohol(include isopropanol,ethanol and 1-propanol). UV detector was used at 226 nm.Result:The optimal composition of mobile phase was hexane isopropanol diethyl amine(98:2:0.2,v/v/v).The optimal flow rate was about 0.4mL min~(-1)from Van Deemter equation,however, baseline separation was achieved (R_S1.5)at flow rate lower than 0.8mL·min~(-1).Within all the temperature range examined,the fluoxetine enantiomers were baseline separated.Conclusion:Using this method,the enantiomers of fluoxetine could be separated quickly and easily.

Abstract:To directly separate fluoxetine hydrochloride enantiomers, high performance liquid chromatography (HPLC) separation method on dimethylated β-cyclodextrin chiral stationary phase was investigated. By adopting the chromatography column of Φ4.6 mm × 250 mm, triethylamine acetate (TEAA) buffer with mass fraction of 0.1% was taken as the mobile phase, and acetonitrile or methanol was taken as modifier using ultraviolet detection wavelength of 226 nm. The effects of mobile phase composition, buffer pH, volume flow rate and column temperature on fluoxetine separation were investigated. The results show that when mass ratio of acetonitrile and pH 4.5 buffer in mobile phase is 12.1:87.9, fluoxetine resolution is 1.81 at column temperature of 35°C and volume flow rate of 1.0 mL/min. When mass ratio of methanol and pH 3.8 buffer in mobile phase is 25.3:74.7, fluoxetine resolution is 2.22 at column temperature of 20°C and volume flow rate of 0.8 mL/min. Dimethylated β-cyclodextrin chiral stationary phase can effectively separate fluoxetine hydrochloride enantiomers.

Abstract:Conventional methods to determine competitive adsorption isotherm are tedious, and require large quantities of pure isomers. In some cases pure enantiomers are not available, thereby making the full experiments to determine the competitive adsorption isotherm of enantiomers impossible. A new method, adjustable factor h-root method (AF-HRM), had been presented for the measurement of langmuir equilibrium coefficients. In the new approach, the retention factors and the frontal retention factors are required to determine the parameters of isotherms. As the ultimate goal is to separate enantiomers of fluoxetine on a preparative scale, a convenient method to determine the adsorption isotherm is essential. In this paper, we have demonstrated that AF-HRM can be applied to determine the parameters in the extended Langmuir model. Experimental results agree well with the prediction.

Abstract:In this study, we focused on the fluorous affinity acting among fluorine compounds, and then developed a new separation medium and evaluated their performance. We prepared the stationary phases for a column using silica gel‐modified alkyl fluoride and investigated the characteristics of fluorous affinity by comparing them with a typical stationary phase, which does not contain fluorine, using high‐performance liquid chromatography (HPLC). In HPLC measurements, we confirmed that while all non‐fluorine compounds were not retained, retention of fluorine compounds increased as the number of fluorine increased with the stationary phase. It also revealed that the strength of fluorous affinity changes depending on the types of the organic solvent; the more polar the solvent, the stronger the effect. Additionally, the stationary phase was employed to compare the efficiency of our column with that of a commercially available column, Fluofix‐II. The retention selectivity was almost the same, but the absolute retention strength was slightly higher on our column, indicating that the column is available for practical use.

Abstract:Fluorine‐18 is the predominant radionuclide used to label Positron Emission Tomography (PET) tracers. One outstanding challenge in nucleophilic aromatic radiofluorination reactions is the sensitivity of precursors and catalysts for basic reaction conditions, which are necessary for the work‐up of [18F]fluoride, resulting in limited reproducibility. Triflyl [18F]fluoride is a new [18F]fluoride source that allows freedom in choice of type and amounts of base and cryptand. The aim of the current work is to explore the scope and limitations of triflyl [18F]fluoride in the late‐stage nucleophilic aromatic 18F‐fluorination of various functionalized precursors, exploring reduced amounts of base and cryptand. The assessment allowed for the application of this new nucleophilic [18F]fluoride reagent to the successful radiosynthesis of boron, stannane, hypervalent iodonium ylide and phenol substrates bearing electron‐deficient, ‐neutral and ‐rich functional groups as well as the clinically relevant PET tracers [18F]FPEB, [18F]mFBG and [18F]SynVesT‐1.

In conclusion, the retention behavior of glucuronides can be predicted by two ways: (1) prediction of capacity factor (K') for hydroxy compounds and their glucuronides is based on chemi-physical constants; (2) prediction of glucuronides capacity factor is based on the capacity factor of parent compound, The paper solved a knotty problem, the lack of glucuronide reference by using biological synthesis method, It is very useful for the study on drug phase II metabolism, Predicting retention behavior of metabolites based on the parent compounds is also a new idea for studying QSRR for drug metabolites.

Abstract:A new method was proposed for the determination of fluoride in soil water extract by ion chromatography with column-switching technique. A reversed-phase column was used as a pretreatment column on-line to separate fluoride and organic acids as well as to eliminate organics. Fluoride was collected by a collection loop and delivered to the analytical system. Under the optimum conditions, the linear range of the method for fluoride was 0.05-10. 0 mg/L (r=0. 999 9), and the limit of detection (S/N =3) was 5. 50 g/L. The recovery was in the range of 103. 4%-105. 3% with the relative standard deviation of 2. 0%-2. 1%. The method could be applied to determine fluoride in complicated samples.

Abstract:2-18F-PD153035 was synthesized by nucleophilic displacement of 4-(3-bromoanilino)-6,7-dimethoxyquinazoline-2-trimethylammonium chloride with 18F−. Biodistribution and micro-PET of the tracer in normal mice were studied. The 2-18F-PD153035 was obtained in 20–30 % yields with more than 99 % radiochemical purity. Biodistribution and micro-PET showed that the liver and the intestines were the primary sites of excretion of this drug.

Long-term quality assurance of [(18)F]-fluorodeoxyglucose (FDG) manufacturing

Abstract:Nine years of experience with 2286 commercial synthesis allowed us to deliver comprehensive information on the quality of (18)F-FDG production. Semi-automated FDG production line using Cyclone 18/9 machine (IBA Belgium), TRACERLab MXFDG synthesiser (GE Health, USA) using alkalic hydrolysis, grade "A" isolator with dispensing robotic unit (Tema Sinergie, Italy), and automatic control system under GAMP5 (minus2, Slovakia) was assessed by TQM tools as highly reliable aseptic production line, fully compliant with Good Manufacturing Practice and just-in-time delivery of FDG radiopharmaceutical. Fluoride-18 is received in steady yield and of very high radioactive purity. Synthesis yields exhibited high variance connected probably with quality of disposable cassettes and chemicals sets. Most performance non-conformities within the manufacturing cycle occur at mechanical nodes of dispensing unit. The long-term monitoring of 2286 commercial synthesis indicated high reliability of automatic synthesizers. Shewhart chart and ANOVA analysis showed that minor non-compliances occurred were mostly caused by the declinations of less experienced staff from standard operation procedures, and also by quality of automatic cassettes. Only 15 syntheses were found unfinished and in 4 cases the product was out-of-specification of European Pharmacopoeia. Most vulnerable step of manufacturing was dispensing and filling in grade "A" isolator. Its cleanliness and sterility was fully controlled under the investigated period by applying hydrogen peroxide vapours (VHP). Our experience with quality assurance in the production of [(18)F]-fluorodeoxyglucose (FDG) at production facility of BIONT based on TRACERlab MXFDG production module can be used for bench-marking of the emerging manufacturing and automated manufacturing systems.

Abstract:A fluorinated covalent organic framework composed of 2,3,5,6 -tetrafluoroterephthaldehyde (TFA) and 1,3,5 -tri(4-aminophenyl)benzene (TAPB) is proposed for electrochromatographic separation. TFA-TAPB is for the first time regarded as the stationary phase of capillary electrochromatography. The TFA-TAPB-coated capillary columns exhibited satisfactory separation (resolution values > 1.5) and good reproducibility towards fluoroquinolones. The intraday relative standard deviations (RSDs) of retention time and peak areas were 0.54–0.68% and 1.69–2.82%, respectively. The interday RSDs of retention time and peak areas were less than 1.79% and 2.30%, respectively. The column-to-column RSDs of retention time and peak areas were 0.22–0.73% and 0.74–1.86%, respectively. And, the inter-batch RSDs of retention time and peak areas were less than 0.39% and 1.67%, respectively. Moreover, the possible separation mechanism was discussed, and it was found that the π-π stacking effect, hydrophobic interaction, hydrogen bonding, and fluorous interactions were the main factors. Overall, these results demonstrated that TFA-TAPB has high prospect for CEC separation.
High yield synthesis of 6-[18F]fluoro-L-dopa

Abstract:The radiofluorination of L-dopa with [18F]F2 was investigated with the purpose of improving the yield of 6-[18F]fluoro-L-dopa. When boron trifluoride was added to the reaction mixture in hydrogen fluoride (HF), the yield was increased threefold. Nine millicuries of 6-[18F]fluoro-L-dopa were produced from 100 mCi [18F]F2 routinely and reliably after 2 hr of preparation. If acetonitrile or water were substituted for HF, little or no 6-fluoro-L-dopa was made.

Abstract:Fluorous solid-phase extraction (FSPE) has been employed to isolate target compounds from complex chemical or biological samples in many research areas. However, the lack of efficient and economical available perfluorinated materials impeded its development. In this study, we present a novel nanographite fluoride-based fluorous solid-phase extraction (GF-FSPE) as a replacement of commercially available cartridge-based FSPE, which showed remarkable selectivity, low LOD, high post enrichment recovery, and large enrichment capacity. To demonstrate the potency of GF-FSPE, it was designed and successfully applied to isolate cysteine-containing peptide subsets from complex protein samples to improve the proteome coverage. Additionally, since graphite fluoride was inexpensive and highly commercialized, this study was expected to promote the popularization of FSPE in both chemical and biological separations as well as encourage the synthesis and broaden the application of highly fluorinated carbon fluoride in material science.

Abstract:18F-labelled radiopharmaceuticals are indispensable in positron emission tomography. The critical step in the preparation of 18F-labelled tracers is the anhydrous F-18 nucleophilic substitution reaction, which involves [18F]F- anions generated in aqueous media by the cyclotron. For this, azeotropic drying by distillation is widely used in standard synthesisers, but microfluidic systems are often not compatible with such a process. To avoid this step, several methods compatible with aqueous media have been developed. We summarised the existing approaches and two of them have been studied in detail. [18F]fluoride elution efficiencies have been investigated under different conditions showing high 18F-recovery. Finally, a large scope of precursors has been assessed for radiochemical conversion, and these hydrous labelling techniques have shown their potential for tracer production using a microfluidic approach, more particularly compatible with iMiDEV™ cassette volumes.

Abstract:Chromatographers often employ fully aqueous mobile phases to retain highly polar compounds in reversed-phase liquid chromatography (RPLC). However, when the flow rate is interrupted, either accidentally or intentionally, a substantial loss in retention occurs due to the spontaneous dewetting of water from the hydrophobic surface of conventional RPLC-C 18 particles. Previous studies have shown that maintaining a low C 18 surface coverage (approximately 1.5 μmol/m 2 ) can mitigate water dewetting by increasing chain disorder, facilitating the intercalation of water clusters between the C 18 -bonded chains, and keeping the mesopores wetted. In this research, we explore the potential and additional benefits of using two-component surface bonding materials (C 8 /C 18 and PhenylHexyl (PhHx)/C 18 ) at a constant and low total surface coverage of 1.51 ± 0.15 μmol/m 2 . We synthesized seven one- and two-component modified silica particles with a volume average particle size of 5.22 μm and an average mesopore size of 104 Å. The surface coverage was increased from 0 to 0.54, 1.00, and to 1.66 μmol 2 for C 8 chains and from 0 to 0.52, 0.70, and to 1.65 μmol 2 for PhHx ligands. To prevent interactions between water and any unreacted silanols, all seven derivatized particles were heavily endcapped with trimethylsilane (TMS) reagent. The fraction of the surface area remaining in contact with water was determined by measuring the retention times of weakly (thiourea) and strongly (thymine) retained compounds at intervals of 1, 2, 4, 8, 16, 32, and 64 minutes following the cessation of flow. Two distinct column temperatures, 24 °C and 60 °C, were employed in the experiments. Retention losses were found to be minimized in the presence of a small quantity of C 8 chains (less than 40% of the total surface coverage). Additionally, it is essential to consider substantial fractions of PhHx chains, as long as the presence of the PhHx ligand does not significantly impact retention and selectivity. Combining mixed RPLC bondings with a low total surface coverage of approximately 1.5 μmol/m 2 emerges as a viable solution for further minimizing retention loss in standard C18-bonded RPLC columns, particularly within the surface coverage range of 2.5-3.0 μmol/m 2 .

Abstract:We report on a free-standing microfluidic pipette made in poly(dimethylsiloxane) having a circulating liquid tip that generates a self-confining volume in front of the outlet channels. The method is flexible and scalable as the geometry and the size of the recirculation zone is defined by pressure, channel number, and geometry. The pipette is capable of carrying out a variety of complex fluid processing operations, such as mixing, multiplexing, or gradient generation at selected cells in cell and tissue cultures. Using an uptake assay, we show that it is possible to generate dose-response curves in situ from adherent Chinese hamster ovary cells expressing proton-activated human transient receptor potential vanilloid (hTRPV1) receptors. Using confined superfusion and cell stimulation, we could activate hTRPV1 receptors in single cells, measure the response by a patch-clamp pipette, and induce membrane bleb formation by exposing selected groups of cells to formaldehyde/dithiothreitol-containing solutions, respectively. In short, the microfluidic pipette allows for complex, contamination-free multiple-compound delivery for pharmacological screening of intact adherent cells.
