Abstract:Microplastics (MPs), the emerging environmental pollutants, have attracted global attention due to the potential public health challenge and ecological security risk. Recent studies suggested liver as a vulnerable organ to MPs exposure, evidenced by abnormal hepatic lipid metabolism upon MPs intake in multiple animal species. How-ever, the specific changes of lipid metabolism in mammalian livers, as well as the underlying mechanisms, remain to be elucidated. In the present study, C57BL/6 mice were randomly assigned to normal drinking water or drinking water containing 100 mu g L-1 or 1000 mu g L-1 polystyrene (PS) MPs for 8 weeks. MPs exposure exerted no significant effect on body weight, serum triglyceride or total cholesteryl esters. However, mice showed impaired glucose tolerance and hepatic lipid deposition in response to high-dose MPs administration. Further lipidomic analysis showed significant alteration in hepatic lipid species particularly with free fatty acids (FFAs) and tri-acylglycerols (TAGs) in mice exposed to MPs. Meanwhile, the liver transcriptional profile indicated MPs exposure-induced differentially expressed genes (DEGs) were enriched in pathways of lipid metabolism and unfolded protein response. Furthermore, most altered lipid species were significantly correlated with DEGs enriched in lipid metabolic signaling. These findings provide lipidomic and transcriptional signatures of liver in response to MPs exposure, which will shed light on further understanding of the metabolic toxicity of MPs.
Effects of exposure to waterborne polystyrene microspheres on lipid metabolism in the hepatopancreas of juvenile redclaw crayfish, Cherax quadricarinatus

Abstract:Previous research has identified microplastics as new environmental pollutants that are widely distributed in a variety of environments, including aquaculture environments. However, the potential hazard of microplastics to aquaculture animals, especially toward lipid metabolism involved with the survival and growth of aquatic animal, has not yet been investigated. In the present study, redclaw crayfish (Cherax quadricarinatus) were exposed to different concentrations of 200 nm-sized polystyrene microspheres (0, 0.5, and 5 mg/L) for 21 days, to investigate the effects of microplastics on lipid metabolism.After ingestion, the microplastics were distributed in the intestines and hepatopancreas, and appeared to inhibit the growth of Cherax quadricarinatus. Subsequently, the lipid levels in the hepatopancreas and hemolymph was detected, and found that after 21 days of exposure, the lipid content and free fatty acids in the hepatopancreas and hemolymph decreased significantly, and total cholesterol and triglycerides levels increased significantly in the hemolymph. This might have been caused by insufficient intake of exogenous fat. A significant decrease in lipase activity also supported this view. The activity of lipoprotein lipase related to lipolysis in the hepatopancreas increased significantly, while the activity of fatty acid synthase related to fat synthesis increased, and the activity of acetyl-CoA carboxylase decreased. These results indicated disturbed lipid metabolism in the hepatopancreas. The significant increase of lipid transport-related low-density lipoprotein indicated that the lipolytic capacity was higher than the lipid synthesis capacity. The expression levels of fatty acid metabolism-related genes FAD6 and FABP decreased significantly, indicating that the fatty acid utilization ability of hepatopancreas cells was inhibited, which was consistent with the results of enzyme activities.Thus, microplastics represent a potential hazard to redclaw crayfish, at least on lipid metabolism. This study provided basic data on the ecotoxicological effects of microplastics on crustaceans.

Toxic effect of chronic exposure to polyethylene nano/microplastics on oxidative stress, neurotoxicity and gut microbiota of adult zebrafish (Danio rerio)

Abstract:The rapid development of aquaculture industry has provided a large amount of high-quality animal protein, while the food safety caused by microplastics and nanoplastics (MP/NPs) has become a major concern. In addition, recent evidence has shown the potential toxic effect of PE-MP/NPs, highlighting the need for further research into their environmental and health impacts. Chronic exposure of polyethylene microplastics (PE-MPs) and nanoplastics (PE-NPs) on adult zebrafish were conducted in the present study for 21 d. Organ-dependent oxidative damage induced by MP/NPs was observed. Insignificant differences in neurotoxicity and dysbiosis of gut microbiota were found between MPs and NPs. Changes in glutathione S-transferase (GST), glutathione (GSH), catalase (CAT), lipid peroxidation (LPO), and superoxide dismutase (SOD) showed that MP/NPs induced oxidative damage in gill and intestinal cells of zebrafish. The inhibited AChE activity suggested the potential neurotoxicity of microplastics and nanoplastics (MP/NPs). In addition, chronic exposure increased the alpha-diversity of intestinal microbiota. At the phylum level, the average relative abundance of Proteobacteria increased from 29.73% (control group) to 66.10% (microplastics), 54.84% (nanoplastics) and 60.03% (combined exposure), respectively. Tenericutes decreased from 55.43% (control group) to 20.02% (microplastics), 22.44% (nanoplastics) and 31.77% (combined exposure), respectively. Overall, this study provides new insights and objective evidence for the toxicity assessment of PE-MPs.

Abstract:Microplastics have become emerging contaminants, causing widespread concern about their potential toxic effects. In this study, the uptake and tissue accumulation of polystyrene microplastics (PS-MPs) in zebrafish were detected, and the toxic effects in liver were investigated. The results showed that after 7 days of exposure, 5 μm diameter MPs accumulated in fish gills, liver, and gut, while 20 μm diameter MPs accumulated only in fish gills and gut. Histopathological analysis showed that both 5 μm and 70 nm PS-MPs caused inflammation and lipid accumulation in fish liver. PS-MPs also induced significantly increased activities of superoxide dismutase and catalase, indicating that oxidative stress was induced after treatment with MPs. In addition, metabolomic analysis suggested that exposure to MPs induced alterations of metabolic profiles in fish liver and disturbed the lipid and energy metabolism. These findings provide new insights into the toxic effects of MPs on fish.

Abstract:Microplastics (MPs) are a serious threat to the living environment of aquatic organisms. However, there are fewer studies on the toxicity of microplastics to freshwater organisms. This study aimed to establish a polystyrene microplastics (PS-MPs) model by feeding carp (Cyprinus carpio) PS-MP (1000 ng/L) particles 8 μm in size. HE staining revealed a mass of inflammatory cells infiltrated in the carp hepatopancreas. The activities of alkaline phosphatase (AKP), aspartate transaminase (AST), lactate dehydrogenase (LDH), and alanine transaminase (ALT) were strengthened considerably, suggesting that PS-MPs cause injury to the hepatopancreas of carp. Real-Time polymerase chain reaction and western blotting results indicated increased levels of glucose-regulated protein 78 (GRP78), (PKR)-like ER kinase (PERK), eukaryotic translation initiation Factor 2α (EIF2α) and activating transcription Factor 4 (ATF4) genes and increased levels of inflammatory factors downstream of endoplasmic reticulum stress (ERs) thioredoxin-interacting protein (TXNIP), NOD-like receptor protein 3 (NLRP3), interleukin-18 (IL-18), interleukin-1β (IL-1β), and caspase 1. Increased expression of microtubule-associated protein-2 (LC3II), autophagy-related 5 (ATG5) and autophagy-related 12 (ATG12) genes revealed that PS-MPs promoted autophagy in carp hepatocytes. The enhanced expression of the Caspase 12, Caspase 3, and Bax genes suggested that PS-MPs led to the apoptosis of carp hepatocytes. These results suggest that PS-MPs result in serious injury to the hepatopancreas of carp. The present study of PS-MPs in freshwater fish from the aspect of endoplasmic reticulum stress was conducted to provide references and suggestions for toxicological studies of PS-MPs in freshwater environments.

Abstract:Microplastics (5 mm - 1 μm) have become one of the major pollutants in the environment. Numerous studies have shown that microplastics can have negative impacts on aquatic organisms, affecting their liver function levels. However, the extent of these effects and their potential toxicological mechanisms are largely unknown. In this study, a meta-analysis and systematic review were conducted to assess the effects of microplastics on fish liver function and summarize the potential toxicological mechanisms of microplastic-induced liver toxicity. The meta-analysis results indicate that compared to the control group, exposure to microplastics significantly affects fish liver indicators: aspartate aminotransferase (AST) ( p < 0.001), alanine aminotransferase (ALT) ( p < 0.001), alkaline phosphatase (ALP) ( p < 0.001), total protein (TP) ( p < 0.001), and lactate dehydrogenase (LDH) ( p < 0.001), including oxidative stress indicators: superoxide dismutase (SOD) ( p < 0.001), glutathione S-transferase (GST) ( p < 0.001), glutathione (GSH) ( p < 0.001), and malondialdehyde (MDA) ( p < 0.001) in fish liver. For fish living in different environments, the potential toxicological mechanisms of microplastics exposure on fish liver may exhibit some differences. For freshwater fish, the mechanism may be that microplastics exposure causes overproduction of reactive oxygen species (ROS) in fish hepatocyte mitochondria. ROS promotes the expression of toll-like receptor 2 (TLR2) and activates downstream molecules myeloid differentiation factor 88 (MyD88) and tumor necrosis factor receptor-associated factor 6 (TRAF6) of the TLR2 signaling pathway, leading to phosphorylation of NF-κB p65. This leads to the release of inflammatory factors and oxidative stress and inflammation in fish liver. In addition, for seawater fish, the mechanism may be that microplastics exposure can cause damage or death of fish hepatocytes, leading to continuous pathological changes, inflammation, lipid and energy metabolism disorders, thereby causing significant changes in liver function indexes.

Effect of polyethylene microplastics on oxidative stress and histopathology damages in Litopenaeus vannamei

Abstract:There has been a significant increase in the microplastic (MP) polluting the ocean in recent time which is regarded as toxic for living organisms. In this study, red fluorescent polyethylene microspheres (FRPE) were administered intramuscularly to Litopenaeus vannamei juveniles at the concentration of 0.1, 0.2, 0.5 and 1.0 μg (g shrimp)−1, and the survival rate was recorded. Analysis of the hepatopancreas for antioxidant enzyme activity and gene expression were done after seven days. Further tissue morphology and accumulation of FRPE was analysed. The results showed that FRPE at 0.5 and 1.0 μg (g shrimp)−1 reduce the survival rate of L. vannamei. FRPE at 0.5 and 1.0 μg (g shrimp)−1 reduced superoxide dismutase (SOD) activity; FRPE at different concentrations reduced catalase (CAT) activity; FRPE at 0.2, 0.5 and 1.0 μg (g shrimp)−1 increased the lipid peroxide thiobarbituric acid (TBARS) content. FRPE at 0.1, 0.2, and 0.5 μg (g shrimp)−1 significantly affect the performance of SOD and CAT genes; FRPE at 0.2 and 0.5 μg (g shrimp)−1 significantly improves GPx gene performance; FRPE at 1.0 μg (g shrimp)−1 significantly reduced the expression of SOD and GPx genes. Analysis of tissue morphology shows that FRPE cause muscle, midgut gland, and hepatopancreas, and gill damage at different concentrations. In the results of accumulation of microplastic, FRPE accumulated in gill tissue at 0.2 and 0.5 μg (g shrimp)−1; FRPE accumulated in gill, muscle and hepatopancreas tissue at 1.0 μg (g shrimp)−1. Based on the above results, FRPE at 0.5 and 1.0 μg (g shrimp)−1 can regulate the antioxidant enzymes of L. vannamei, increase lipid peroxide content, cause tissue damage by accumulating in the tissues. The rate of survival decreased in L. vannamei, and the impact of FRPE at 1.0 μg (g shrimp)−1 was significant.

Abstract:Microplastics (MPs) are widely found in coastal areas and oceans worldwide. The MPs are environmentally concerning due to their bioavailability and potential impacts on a wide range of marine biota, so assessing their impact on the biota has become an urgent research priority. In the present study, we exposed Crassostrea gigas oysters to irregular MPs of two polymer types (polyethylene (PE) and polyethylene terephthalate (PET)) at concentrations of 10 and 1000 μg L−1 for 21 days. Accumulation of MPs, changes in metabolic enzyme activity, and histological damage were evaluated, and metabolomics analysis was conducted. Results demonstrated that PE and PET MPs were detected in the gills and digestive gland following exposure to both tested concentrations, confirming ingestion of MPs by the organisms. Moreover, both PE and PET MPs inhibited lipid metabolism, while energy metabolism enzyme activities were activated in the oysters. Histopathological damage of exposed oysters was also observed in this study. Integrated biomarker response (IBR) results showed that MPs toxicity increased with increasing MPs concentration, and the toxic effects of PET MPs on oysters was greater than PE MPs. In addition, metabolomics analysis suggested that MPs exposure induced alterations in metabolic profiles in oysters, with changes in energy metabolism and inflammatory responses. This study reports new insights into the consequences of MPs exposure in marine bivalves at environmentally relevant concentrations, providing valuable information for ecological risk assessment of MPs in a realistic conditions.

Abstract:Microplastics (MPs) are currently one of the major environmental problems due to the large amount released in aquatic and terrestrial ecosystems as well as their potential impacts on organisms and human health. However, the molecular mechanisms underlying MP toxicity remain elusive and insufficiently understood. In this study, oysters were exposed to irregular MPs composed of polyethylene and polyethylene terephthalate at concentrations of 10 and 1000 μg L−1 for 21 days under static conditions, and physiological responses and changes in antioxidant enzymatic activity were investigated. Moreover, the protein expression profiles in the digestive glands were analyzed by using the isobaric tags for relative and absolute quantitation (iTRAQ) method. And the expression of ten potentially regulated genes was analyzed by qRT-PCR in different tissues under MP exposure. It was demonstrated that PE and PET MP exposure had no distinct adverse effects on physiological rates, while inducing oxidative stresses in the oyster. Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis of DEPs revealed that MP exposure mainly disturbed the "arachidonic acid metabolism", "linoleic acid metabolism" and "glycerophospholipid metabolism" processes in oysters. Gene Ontology enrichment analysis revealed that MPs affected the oxidation-reduction process, lipid metabolism process and pentose phosphate pathway. Moreover, a significant increase in the mRNA expression of genes related to lipid and aerobic metabolism and apoptosis pathways was observed after MP exposure. In summary, our results indicated that MPs could induce oxidative stress and alter lipid and glucose metabolism processes in oysters. Additionally, the weight of evidence (WOE) model analysis suggested that high concentrations of MPs were more hazardous than low concentrations and typically higher for PET than PE. This study provided the effects of PE and PET MP toxicity on oysters at the molecular level, which will contribute to better understanding the defense mechanisms of oysters against MP exposure.

Abstract:Microplastics (MPs) pose one of the major environmental threats to marine organisms and ecosystems on a global scale. Although many marine crustaceans are highly susceptible to MPs pollution, the toxicological effects and mechanisms of MPs on crustaceans are poorly understood. The current study focused on the impacts of MPs accumulation in shrimp Litopenaeus vannamei at the behavioral, histological and biochemical levels. The results demonstrated the accumulation of polystyrene MPs in various organs of L. vannamei, with highest MPs abundance in the hepatopancreas. The MPs accumulated in shrimp caused growth inhibition, abnormal swimming behavior and reduced swimming performance of L. vannamei. Following MPs exposure, oxidative stress and lipid peroxidation were also observed, which were strongly linked to attenuated swimming activity of L. vannamei. The above MPs-induced disruption in balance of antioxidant system triggered the hepatopancreatic damage in L. vannamei, which was exacerbated with increasing MPs concentrations (from 0.02 to 1 mg L-1). Furthermore, metabolomics revealed that MPs exposure resulted in alterations of metabolic profiles and disturbed glycolysis, lipolysis and amino acid metabolism pathways in hepatopancreas of L. vannamei. This work confirms and expands the knowledge on the sublethal impacts and toxic modes of action of MPs in L. vannamei.

Long-term PS micro/nano-plastic exposure: Particle size effects on hepatopancreas injury in Parasesarma pictum

Abstract:With the widespread use of plastic products, microplastics and nanoplastics have emerged as prevalent pollutants in coastal aquatic ecosystems. Parasesarma pictum, a common estuarine crab species, was selected as a model organism. P. pictum was exposed to polystyrene (PS) particles of sizes 80 nm (80PS), 500 nm (500PS), and 1000 nm (1000PS), as well as to clean seawater (CK) for 21 days. Histological and fluorescent staining results showed that PS particles of all three sizes induced hepatopancreatic nuclear pyknosis, cell junction damage, and necrosis. The degree of damage was observed as 1000PS > 80PS > 500PS. Transcriptomic analysis revealed that major differentially expressed genes (DEGs) were associated with cellular processes, membrane components, and catalytic activity. The respiratory chain disruptions and immune exhaustion induced by 1000PS were notably stronger than those by 80PS and 500PS. Additionally, necrosis caused hepatopancreas injury in P. pictum rather than apoptosis or autophagy after long-term PS particle exposure. Furthermore, PS particles of all three sizes inhibited innate immunity, while the complement pathway was not significantly affected in the 80PS group. This study elucidated potential distinctions in how plastic particles of varying sizes (nanoplastics, microplastics, and micro/nanoplastics) impact P. pictum, providing a reference for toxicological mechanism research on microplastics and nanoplastics in aquatic organisms. Future research should focus on exploring long-term effects and potential mitigation strategies for microplastics and nanoplastics of more types and a wider range of particle size pollution in aquatic environments.
Polystyrene microplastics induce endoplasmic reticulum stress, apoptosis and inflammation by disrupting the gut microbiota in carp intestines

Abstract:Microplastics have been recognized as a widespread new pollutant in nature and have induced an increase in the occurrence of a variety of diseases in carp. An animal model of microplastic ingestion was successfully established in an aqueous environment. The gut microbiota was analysed using a metagenomic approach. The results showed a significant reduction in the relative abundances of Lactococcus garvieae, Bacteroides_paurosaccharolyticus, and Romboutsia_ilealis after PS-MPs treatment. The 16S Silva database was used to predict and analyse the known genes. Intestinal flora disorders related to infectious diseases, cancers, neurodegenerative diseases, endocrine and metabolic diseases, cardiovascular diseases, and other diseases were found. The intake of PS-MPs resulted in damage to carp intestinal tissue and apoptosis of intestinal epithelial cells. The levels of the inflammatory cytokines IL-1β, IL-6, and TNF-α were significantly increased with the intake of PS-MPs. The gene and protein levels of GRP78, Caspase-3, Caspase-7, Caspase-9, Caspase-12, PERK, IRE1, and ATF6 were further examined in PS group. The occurrence of ERS and apoptosis in carp intestines was confirmed. These results suggest that the accumulation of PS-MPs in the aquatic environment can disturb the carp gut microbiota and induce ERS, apoptosis, and inflammation in the intestinal tissue.

Abstract:As ubiquitous emerging pollutants, microplastics (MPs) in aquatic environments have aroused critical global concerns. Despite the occurrence and characteristics of MPs in freshwater agroecosystems well-described by our previous study, their ecotoxicological implications in Monopterus albus remains unfathomed. Herein, we dissected toxic effects and mechanisms of PS-NPs exposure against M. albus hepatic tissues at concentrations of 0.5 (L), 5 (M), 10 (H) mg/L for 28 days using physiochemical measurements, histopathological analysis and transcriptomic sequencing. Results showed that upon PS-NPs treatments, levels of ROS, MDA, 8-OHdG and MFO activity were significantly enhanced relative to the control (C) group, while SP content and T-AOC activity were dramatically suppressed, suggesting ROS burst, lipid peroxidation and DNA damage may occur in liver tissues. This oxidative damage further triggered impaired hepatic function and histopathology, disordered lipid metabolism and hepatocyte apoptosis, as reflected by significantly diminished activities of GPT, GOT, ACP, AKP and LDH, paralleled with augmented levels of TG, TC and HSI as well as Cytc and Caspase-3,8,9 activities. Noticeably, concentration-dependent rises of apoptotic rate, vacuolar degeneration and lipid deposition were manifest in TUNEL, H&E and ORO staining. In addition, a total of 375/475/981 up-regulated as well as 260/611/1422 down-regulated DEGs in C vs L, C vs M and C vs H categories were identified based on RNA-seq, respectively. These DEGs were significantly annotated and enriched into GO terms (membrane, cytoplasm, response to stimuli, oxidation-reduction process) as well as KEGG pathways (ether lipid metabolism, apoptosis, chemical carcinogenesis-reactive oxygen species, non-alcoholic fatty liver disease). Moreover, signaling cascades Keap1-Nrf2, p53 and PPAR were either substantially initiated or dysregulated to orchestrate PS-NPs hepatotoxicity featuring oxidative damage, hepatocyte apoptosis and lipid steatosis. Collectively, this study not only expounded on toxicological mechanisms whereby PS-MPs exerted deleterious effects on M. albus , but also pointed to ecological risks of PS-MPs-induced hepatoxicity and lipid steatosis in this commercially-important species.

Abstract:Microplastics are ubiquitous pollutants in the global marine environment. However, few studies have adequately explored the different toxic mechanisms of microplastics (MPs) and nanoplastics (NPs) in aquatic organisms. The sea cucumber, Apostichopus japonicus, is a key organism in the marine benthic ecosystem due to its crucial roles in biogeochemical cycles and food web. This study investigated the bioaccumulation and adverse effects of polystyrene micro- and nanoplastics (PS-M/NPs) of different sizes (20 μm, 1 μm and 80 nm) in the regenerated intestine of A. japonicus using multi-omics analysis. The results showed that after 30-day exposure at the concentration of 0.1 mg L-1, PS-MPs and PS-NPs accumulated to 155.41-175.04 μg g-1 and 337.95 μg g-1, respectively. This excessive accumulation led to increased levels of antioxidases (SOD, CAT, GPx and T-AOC) and reduced activities of immune enzymes (AKP, ACP and T-NOS), indicating oxidative damage and compromised immunity in the regenerated intestine. PS-NPs had more profound negative impacts on cell proliferation and differentiation compared to PS-MPs. Transcriptomic analysis revealed that PS-NPs primarily affected pathways related to cellular components, e.g., ribosome, and oxidative phosphorylation. In comparison, PS-MPs had greater influences on actin-related organization and organic compound metabolism. In the PS-M/NPs-treated groups, differentially expressed metabolites were mainly amino acids, fatty acids, glycerol phospholipid, and purine nucleosides. Additionally, microbial community reconstruction in the regenerated intestine was severely disrupted by the presence of PS-M/NPs. In the PS-NPs group, Burkholderiaceae abundance significantly increased while Rhodobacteraceae abundance decreased. Correlation analyses demonstrated that intestinal regeneration of A. japonicus was closely linked to its enteric microorganisms. These microbiota-host interactions were notably affected by different PS-M/NPs, with PS-NPs exposure causing the most remarkable disruption of mutual symbiosis. The multi-omic approaches used here provide novel insights into the size-dependent toxicity of PS-M/NPs and highlight their detrimental effects on invertebrates in M/NPs-polluted marine benthic ecosystems.

Abstract:Microplastics and heavy metal pollution frequently co-occur in the marine environment, raising concerns about their potentially harmful impacts on marine fish. This study undertook a comprehensive evaluation of the individual and combined stress effects of polylactide microplastics (PLA-MPs) and chromium (Cr) on marine medaka larvae. Following a 14-day exposure to PLA-MPs (100 μg/L) and Cr (50 μg/L), both individually and in combination, significant increases in heart rate and body length were observed. Notably, the combined exposure to PLA-MPs and Cr caused marked histopathological alterations, including shedding, atrophy, and lysis of the intestinal tissues. Furthermore, both individual and combined exposure induced oxidative stress in fish larvae, leading to changes in various enzyme activity indices. Individual exposure to either PLA-MPs or Cr led to anxious behavior in the larvae, whereas combined exposure not only caused anxious behavior but also altered swimming patterns. These findings suggest that combined exposure to PLA-MPs and Cr can exacerbate the toxic effects on marine medaka larvae.

Abstract:Microplastics are plastic fragments of particle sizes less than 5 mm, which are widely distributed in marine and terrestrial environments. In this study, earthworms Eisenia fetida were exposed to 100 and 1000 mu g of 100 nm and 1300 nm fluorescent polystyrene microplastics (PS-MPs) per kg of artificial soil for 14 days. Uptake or accumulation of PS-MPs in earthworm intestines, histopathological changes, oxidative stress, and DNA damage were assessed to determine the toxicological effects of PS-MPs on E. fetida. The results showed that the average accumulated concentrations in the earthworm intestines were higher for 1300 nm PS-MPs (0.084 +/- 0.005 and 0.094 +/- 0.003 mu g/mg for 100 and 1000 mu g/kg, respectively) than for 100 nm PS-MPs (0.015 +/- 0.001 and 0.033 +/- 0.002 mu g/mg for 100 and 1000 mu g/kg, respectively). In addition, histopathological analysis indicated that the intestinal cells were damaged after exposure to PS-MPs. Furthermore, PS-MPs significantly changed glutathione (GSH) level and superoxide dismutase (SOD) activity. The GSH levels were 86.991 +/- 7.723, 165.436 +/- 4.256 - 167.767 +/- 18.642, and 93.590 +/- 4.279-173.980 +/- 15.523 mu mol/L in the control, 100 nm, and 1300 nm PS-MPs treatment groups. In addition, the SOD activities were 10.566 +/- 0.621, 9.039 +/- 0.787-9.408 +/- 0.493, and 7.959 +/- 0.422-9.195 +/- 0.327 U/mg protein for the control, 100 nm, and 1300 nm PS-MPs treatment groups, respectively, indicating that oxidative stress was induced after PS-MPs exposure. Furthermore, the comet assay suggested that exposure to PS-MPs induced DNA damage in earthworms. Overall, 1300 nm PS-MPs showed more toxic effect than 100 nm PS-MPs on earthworms. These findings provide new insights regarding the toxicological effects of low concentrations of microplastics on earthworms, and on the ecological risks of microplastics to soil animals. Polyethylene microplastics trigger cell apoptosis and inflammation via inducing oxidative stress and activation of the NLRP3 inflammasome in carp gills

Abstract:Microplastics cause varying degrees of damage to aquatic organisms. Exposure to microplastics contaminated water, the gills are among the first tissues, after the skin, to be affected by microplastics. As an essential immune organ, prolonged stimulation by microplastics disrupts immune function not only in the gills but throughout the body, yet the underlying mechanisms remain elusive. A model of gill injury from exposure to polyethylene (PE) microplastics was developed in this study. H&E staining revealed that polyethylene microplastics caused gill inflammation, vascular remodeling, and mucous cell proliferation. An increase in collagen indicates severe tissue damage. Additional analysis showed that polyethylene microplastics profoundly exacerbated oxidative stress in the gills. TUNEL assay demonstrated cell apoptosis induced by polyethylene microplastic. The mRNA levels were subsequently quantified using RT-PCR. The results showed that polyethylene microplastics increased the expression of the nuclear factor-κB (NF-κB) pathway (NF-κB p65, IKKα, IKKβ) and apoptosis biomarkers (p53, caspase-3, caspase-9, and Bax). Nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasomes, which is an influential component of innate immunity, were overactive. What's more, the pro-inflammatory factors (TNF-α, IFN-γ, IL-2, IL-6, IL-8, IL-1β) that induce immune disorder also increased significantly, while the anti-inflammatory factors (IL-4, IL-10) decreased significantly. These results suggested that oxidative stress acted as an activation signal of apoptosis triggered by the NF-κB pathway and activating the NLRP3 inflammasome to promote inflammatory immune responses. The present study provided a different target for the prevention of toxin-induced gill injury under polyethylene microplastics.
Adverse Physiological and Molecular Level Effects of Polystyrene Microplastics on Freshwater Microalgae.

Abstract:Microplastics have aroused widespread concern because of their adverse effects on aquatic organisms. However, the underlying toxicity mechanisms have not been examined in detail. This study investigated the interactions between polystyrene microplastics (PS-MPs) and the model freshwater microalgae Euglena gracilis. The results of transmission electron microscopy showed that the vacuoles of microalgae were induced after 24 h exposure to 1 mg/L PS-MPs (5 μm and 0.1 μm). Furthermore, PS-MPs significantly (p < 0.05) reduced pigment contents. Moreover, superoxide dismutase activities were significantly (p < 0.05) induced in all PS-MPs treated groups. Peroxidase activities were also significantly (p < 0.05) affected by two sizes of PS-MPs (5 μm and 0.1 μm), indicating that oxidative stress was induced after exposure to PS-MPs. At the molecular level, PS-MPs dysregulated the expression of genes involved in cellular processes, genetic information processing, organismal systems, and metabolisms. The KCS gene and the CTR1 gene may be key pathways to induce adverse effects on the E. gracilis after exposure to 5 μm PS-MPs. These findings will help to elucidate the underlying molecular mechanism of microplastics toxicity on freshwater organisms.

Abstract:A nickel complex has been shown to promote conformation-specific oxidation of guanosine in polynucleotide RNA. In all cases, reaction was strictly dependent on the solvent exposure and surface properties of guanine N7. Modification of native tRNA(Phe) (yeast) was detected at G18, G19, G20, and Gm34 and concurred with predictions based on its crystal structure. Additional guanine derivatives became exposed to oxidation only after the tRNA unfolded in the absence of Mg2+. Reaction of the Tetrahymena group I intron RNA (L-21 ScaI) also compared favorably to its three-dimensional model by appropriately identifying guanosine residues in hairpin loops, duplex termini, and the essential cofactor binding site. These results complemented prior data generated by hydroxyl radical, and in combination they served to distinguish the solvent accessibility of sugar backbone and base positions in guanosine residues. Most importantly, this nickel complex exhibited greater selectivity than either dimethyl sulfate or RNase T1 for characterizing tRNA(Phe) and intron RNA.
