Abstract:Background-Preeclampsia is a prevalent life-threatening hypertensive disorder of pregnancy. The circulating antiangiogenic factor, soluble endoglin (sEng), is elevated in the blood circulation of women with preeclampsia and contributes to disease pathology; however, the underlying mechanisms responsible for its induction in preeclampsia are unknown.Methods and Results-Here, we discovered that a circulating autoantibody, the angiotensin receptor agonistic autoantibody (AT(1)-AA), stimulates sEng production via AT(1) angiotensin receptor activation in pregnant mice but not in nonpregnant mice. We subsequently demonstrated that the placenta is a major source contributing to sEng induction in vivo and that AT(1)-AA-injected pregnant mice display impaired placental angiogenesis. Using drug screening, we identified tumor necrosis factor-alpha as a circulating factor increased in the serum of autoantibody-injected pregnant mice contributing to AT(1)-AA-mediated sEng induction in human umbilical vascular endothelial cells. Subsequently, among all the drugs screened, we found that hemin, an inducer of heme oxygenase, functions as a break to control AT1-AA-mediated sEng induction by suppressing tumor necrosis factor-alpha signaling in human umbilical vascular endothelial cells. Finally, we demonstrated that the AT1-AA-mediated decreased angiogenesis seen in human placenta villous explants was attenuated by tumor necrosis factor-alpha-neutralizing antibodies, soluble tumor necrosis factor-alpha receptors, and hemin by abolishing both sEng and soluble fms-like tyrosine kinase-1 induction.Conclusions-Our findings demonstrate that AT1-AA-mediated tumor necrosis factor-alpha induction, by overcoming its negative regulator, heme oxygenase-1, is a key underlying mechanism responsible for impaired placental angiogenesis by inducing both sEng and soluble fms-like tyrosine kinase-1 secretion from human villous explants. Our results provide important new targets for diagnosis and therapeutic intervention in the management of preeclampsia. (Circulation. 2010;121:436-444.)
Effects of Autoantibodies Against the Angiotensin II Receptor on Rat Ventricular Myocardial Ionic Currents

Abstract:Objectives: The importance of agonistic antibodies against the angiotensin II receptor (AT1-AA) in the pathogenesis of preeclampsia has not been well researched. The aims of our study ware to obtain direct evidence that the concentration of AT1-AA is increased in preeclampsia; determine the effects of AT1-AA on ventricular ionic currents. Methds: Sera obtained from preeclamptic women (n = 18) were applied to detect AT1-AA and purify IgG and those collected from normal pregnant women (n = 17) were used as negative control. The effects of AT1-AA on rat ventricular ionic currents were studied by whole cell patch clamp techniques. Results: the concentration of AT1-AA is time-dependently increased in preeclampsia compared with normal pregnant women. AT1-AA could dose-dependently increase both I Ca -L and I Na /Ca current which blocked by losartan, a specific antagonist of AT1-receptor suggesting that the agonist-like activity of AT1-AA are mediated by stimulation of AT1-receptor. AT1-AA decreased dose-dependently both I to and I k1current which were also blocked by losartan, However these effects were not observed when IgG from normal pregnant women were administered. Conclusion: Our study provides direct evidence that the concentration of AT1-AA is markedly increased in preeclampsia; it has a remarkable positive inotropic effect on the heart, indicating that it may be play an important role in the pathogenesis of preeclampsia.
Angiotensin 1-7 and Its Analogue Decrease Blood Pressure but Aggravate Renal Damage in Preeclamptic Mice.

Abstract:Preeclampsia (PE) is a multisystem disease that affects the health of both the pregnant women and the fetus during pregnancy. Agonistic autoantibodies to the angiotensin II type I receptor (AT1-AA) play a significant role in the pathogenesis of PE. This study aimed to determine the effects of Angiotensin 1-7 (Ang 1-7) and its analogue AVE0991 on AT1-AA-induced PE model. Pregnant mice were divided into five groups: the normal pregnant group, AT1-AA-induced preeclampsia group, and AT1-AA-induced preeclampsia group treated with Losartan, Ang 1-7, and AVE0991, respectively. AT1-AA-induced PE model was established on gestational day 13 by tail intravenous injection of purified AT1-AA polyclonal antibody from serum of guinea pigs. Blood urea nitrogen (BUN), urine albumin and urinary creatinine were measured on day 18 of pregnancy. The systolic blood pressure (SBP) was measured from gestational day 13 to day 18. Renal structure changes were observed via light and electron microscopy. Compared with the normal pregnant group (NP group), AT1-AA-induced preeclampsia group (PE group) exhibited elevated blood pressure and proteinuria, consistent with the characteristics of PE. Ang 1-7 or AVE0991 treatment decreased blood pressure without showing renoprotective effects. The findings indicated that Ang 1-7 and its analogue reduced blood pressure but aggravated renal damage in AT1-AA-induced PE mice.
Angiotensin II type 1 receptor autoantibody blockade improves cerebral blood flow autoregulation and hypertension in a preclinical model of preeclampsia

Abstract:Introduction:Women with preeclampsia (PE) and reduced uterine perfusion pressure (RUPP) pre-clinical rat model of PE have elevated angiotensin II type 1 receptor agonistic autoantibodies (AT1-AA) and cerebrovascular dysfunction. Methods:Sprague Dawley rats had RUPP surgery with/without AT1-AA inhibitor ('n7AAc'144 μg/day) osmotic minipumps. Mean arterial pressure (MAP), CBF autoregulation, blood brain barrier (BBB) permeability, cerebral edema, oxidative stress, and eNOS were assessed. Results:'n7AAc' improved MAP, restored CBF autoregulation, prevented cerebral edema, elevated oxidative stress, and increased phosphorylated eNOS protein in RUPP rats. Conclusion:Inhibiting the AT1-AA in placental ischemic rats prevents hypertension, cerebrovascular dysfunction, and improves cerebral metabolic function.

Abstract:AT1R expression is increased in the nucleus of solitary tract (NTS) of many animal models of hypertension. However, the impact of its chronic regulation on cardiovascular control is not well understood. In this study, we investigated the hypothesis that NTS AT1R is involved in peripheral inflammatory status and hypertension-linked cardiovascular pathophysiology. AAV2- mediated expression of a shRNA targeted the AT1R in the NTS has been used to test the hypothesis. First,the efficacy of the AAV2-AR1R-shRNA was established with the use of neuronal cells in primary culture. Transduction of these cultures by AAV2-AT1R-shRNA resulted in a 72% decrease in AT1R mRNA compared with the AAV2- Sc-shRNA transduced control. This was accompanied by complete attenuation of AngII-induced increase in ERK1/2 phosphorylation and neuronal action potential frequency (control: 2.2±0.4 vs. AT1R-shRNA: 0.5±0.3 hz). We then microinjected AAV2-AT1R-shRNA or control vector, AAV2-Scrambled (Sc)-shRNA into the NTS of 8-week-old SHR and WKY rats. Nine weeks following microinjection, AAV2-AT1R-shRNA group of SHRs showed a 29 mmHg increase in mean arterial pressure (MAP) (control: 154.2±4.7; shRNA: 183.0±10.2 mmHg), insignificant effects on the MAP of WKY rats was observed. The increase in MAP in the SHR by AAV2-AT1R-shRNA was associated with a 42% decrease in baroreflex bradycardia (control: 0.57±0.02; shRNA: 0.33±0.06 ms/mmHg), profound cardiac hypertrophy, and significant increase in the plasma norepinephrine (control: 6.36±0.39, shRNA: 13.0±4.13 ng/mL). In addition, AAV2-AT1R-shRNA rats showed significant increases in water intake (control: 32.5±1.5; shRNA: 12.5±2.5 ml/16h) and urine output (control: 1.6 ±0.5; shRNA: 9.3±2.5 ml/16h). Furthermore, AAV2-AT1R-shRNA treated SHR exhibited 74% decrease in circulating endothelial progenitor cells (EPC, CD90+, CD4/5/8-), and ∼300% increases in inflammatory cells (IC) including CD4/8+, T lymphocytes (CD45/3+) and macrophages (CD68+) . As a result, EPC/IC ratio was decreased by 8∼15 fold in AT1R-shRNA treated SHR. These observations suggest that increased AT1R in the NTS of SHR may present a counter-hypertensive mechanism involving inflammatory/angiogenic cells.

Abstract:AT(1) receptor subtype a (AT(1)Ra) expression is increased in the nucleus of the solitary tract (NTS) in spontaneously hypertensive rat (SHR) compared with Wistar Kyoto controls. However, the chronic role of AT(1)Ra in the NTS for cardiovascular control is not well understood. In this study, we investigated the hypothesis that the NTS AT(1)Ra is involved in the neural regulation of the peripheral inflammatory status and linked with hypertension. Transduction of brain neuronal cultures with recombinant adeno-associated virus type 2 (AAV2)-AT(1)R-small hairpin RNA (shRNA) resulted in a 72% decrease in AT(1)Ra mRNA and attenuated angiotensin II-induced increase in extracellular signal-regulated kinase 1/2 phosphorylation and neuronal firing. Specific NTS microinjection of AAV2-AT(1)R-shRNA vector in the SHR resulted in a approximate to 30 mm Hg increase in the mean arterial pressure compared with control vector-injected animals (Sc-shRNA: 154 +/- 4 mm Hg; AT(1)R-shRNA: 183 +/- 10 mm Hg) and induced a resetting of the baroreflex control of heart rate to higher mean arterial pressure. In addition, AAV2-AT(1)R-shRNA-treated SHRs exhibited a 74% decrease in circulating endothelial progenitor cells (CD90(+), CD4(-)/CD5(-)/CD8(-)) and a 300% increase in the circulating inflammatory cells, including CD4(+)+CD8(+), CD45(+)/3(+) T lymphocytes, and macrophages (CD68(+)). As a result, the endothelial progenitor cell/inflammatory cells ratio was decreased by 8- to 15-fold in the AT(1)R-shRNA-treated SHR. However, identical injection of AAV2-AT(1)R-shRNA into the NTS of Wistar Kyoto rats had no effect on mean arterial pressure and inflammatory cells. These observations suggest that increased expression of the AT(1)Ra in SHR NTS may present a counterhypertensive mechanism involving inflammatory/angiogenic cells.

Abstract:Inflammation is critical in progression of hypertension. However, how an elevated blood pressure (BP) initiates inflammation is unknown, as are the precise effects of high BP on the immune responses. To study this, we first challenged mice with antigen ovalbumin (OVA). Two-fold more OVA-specific CD8 + T cells were present in the blood (2.7 ± 0.23% vs. 1.2 ± 0.47%) and spleen (2.62 ± 0.31% vs. 1.31 ± 0.17%) of angiotensin (Ang) II induced hypertensive mice as compared to those of normotensive controls. To address whether the over-activation of T cell-mediated immune responses is pathogenic, two models of autoimmune disease were studied. RIP-mOVA is a transgenic mouse line that expresses OVA in pancreatic islet β cells. When OVA-specific OT-I T cells were infused into Rip-mOVA mice, the animals with hypertension induced by either Ang II or L-NAME developed more severe diabetes (blood glucose (mg/dl): Ang II 331, L-NAME 315 vs. Control 168). Concanavalin A-induced hepatitis was also significantly worsened by hypertension as compared to the pathology observed in normotensive mice. After examining the characteristics of T cells and antigen presenting cells (APCs), we found that the most distinguishable difference in hypertensive mice was the upregulation of CD86 on APCs. Blocking CD86 by antibody completely abrogated the elevated immune responses in hypertensive mice. To understand what causes CD86 upregulation in APCs of hypertensive mice, we studied damage-associated molecular patterns and found that plasma ATP levels rose as early as 3 days after the induction of hypertension and reached from about 1μM of baseline to a peak level of 3 μM after 2 weeks of hypertension, which exactly parallels the kinetics of CD86 elevation on APCs. Hydrolyzing ATP or blocking its P2X 7 receptor normalized APC CD86 expression and eliminated hypertension-induced T cell over-activation. Further, untreated human hypertensive patients have substantially elevated plasma ATP levels (2.21 ± 0.99 μM, n = 27) compared to treated hypertensive patients (0.94 ± 0.34 μM, n = 17) or normotensive controls (0.63 ± 0.38 μM, n =30). A linear trend was observed between ATP levels and BP among all subjects. These studies indicate ATP is critical in initiating hypertension-associated inflammation.

Abstract:Abstract Background Placental ischemia and hypertension, characteristic features of preeclampsia, are associated with impaired cerebral blood flow (CBF) autoregulation and cerebral edema. However, the factors that contribute to these cerebral abnormalities are not clear. Several lines of evidence suggest that angiotensin II can impact cerebrovascular function; however, the role of the renin angiotensin system in cerebrovascular function during placental ischemia has not been examined. We tested whether the angiotensin type 1 (AT1) receptor contributes to impaired CBF autoregulation in pregnant rats with placental ischemia caused by surgically reducing uterine perfusion pressure. Methods Placental ischemic or sham operated rats were treated with vehicle or losartan from gestational day (GD) 14 to 19 in the drinking water. On GD 19, we assessed CBF autoregulation in anesthetized rats using laser Doppler flowmetry. Results Placental ischemic rats had impaired CBF autoregulation that was attenuated by treatment with losartan. In addition, we examined whether an agonistic autoantibody to the AT1 receptor (AT1-AA), reported to be present in preeclamptic women, contributes to impaired CBF autoregulation. Purified rat AT1-AA or vehicle was infused into pregnant rats from GD 12 to 19 via mini-osmotic pumps after which CBF autoregulation was assessed. AT1-AA infusion impaired CBF autoregulation but did not affect brain water content. Conclusions These results suggest that the impaired CBF autoregulation associated with placental ischemia is due, at least in part, to activation of the AT1 receptor and that the RAS may interact with other placental factors to promote cerebrovascular changes common to preeclampsia.

Vascular Endothelial Growth Factor Attenuates N Omega-Nitro-L-Arginine Methyl Ester-Induced Preeclampsia-Like Manifestations In Rats

Abstract:Objectives. To verify the hypothesis that vascular endothelial growth factor (VEGF) attenuates N omega-Nitro-L-arginine Methyl Ester (L-NAME)-induced preeclampsia-like manifestations in rats. Study design. Forty pregnant Wistar rats were randomly divided into four groups: control, preeclampsia model, VEGF treatment, and VEGF prophylactic. On day 5 of gestation, L-NAME was injected subcutaneously in rats of the preeclampsia model, VEGF treatment, and VEGF prophylactic groups. VEGF was given after the occurrence of hypertension and proteinuria in the VEGF treatment group and from day 5 in the VEGF prophylactic group. Blood pressure was monitored and urine protein was assayed. Blood platelet was counted, and serum nitric oxide metabolites, endothelin-1, 6-keto-PGF-1 alpha, and TXB(2) were determined. Results. Blood pressure increased significantly on day 8 of gestation in the preeclampsia model and VEGF treatment groups compared with control (p < 0.05 for both) and remained elevated through the pregnancy in the preeclampsia model group. Blood pressure was significantly decreased after the administration of VEGF in the VEGF treatment group (p < 0.05). There was no significant difference in blood pressure between the VEGF prophylactic group and control (p < 0.05). Urine protein, platelet count, serum nitric oxide metabolites, endothelin-1, 6-keto-PGF-1, and TXB2 were significantly different between control and the preeclampsia model group (p < 0.05), but not between control and the VEGF treatment or VEGF prophylactic groups (p > 0.05 for all). Conclusion. VEGF attenuates L-NAME-induced preeclampsia-like manifestations in rats, suggesting the important role of VEGF in preeclampsia and providing a potential strategy for the prevention and treatment of preeclampsia.

Abstract:Background: Preeclampsia, a pregnancy-specific complication occurring in the second half of human pregnancy and one of the leading causes for perinatal mortality and morbidity, is characterized by the onset of hypertension and proteinuria. We measured circulating adipocyte fatty acid binding protein (AFABP) and retinol binding protein-4 (RBP-4) in patients with preeclampsia.Methods: Twenty-seven healthy non-pregnant women, 27 healthy pregnant women at third trimester and 26 women with preeclampsia were recruited and blood samples were taken. Concentrations of serum AFABP and RBP-4 were measured with ELISA.Results: There were significant differences in serum AFABP (median: 0.99, 0.93 and 1.81 ng/ml, respectively, P < 0.001) and RBP-4 (mean: 3.16, 2.65 and 4.27 ng/ml, respectively, P = 0.022) among non-pregnancy, normal pregnancy and preeclampsia. Serum AFABP was significantly higher in preeclampsia than non-pregnancy (P < 0.001) and normal pregnancy (P = 0.001). Serum RBP-4 was significantly higher in preeclampsia than normal pregnancy (P = 0.007) but not non-pregnancy (P = 0.061). There were no significant differences in serum RBP-4 and AFABP between non-pregnancy and normal pregnancy. Serum RBP-4 was significantly higher in severe than mild preeclampsia (mean: 5.15 vs 2.84 ng/ml, P = 0.046). There was no significant difference in serum AFABP between mild and severe preeclampsia.Conclusion: Increased circulating AFABP and RBP-4 concentrations were demonstrated, suggesting it be an important pathophysiology of preeclampsia. Stimulation of Angiotensin II Receptor Subtype 2 Reduces Preeclampsia-like Symptoms in a Mouse Model of Preeclampsia

Abstract:Angiotensin II (AngII) receptor subtype 1 (AT1R) is involved in the pathogenesis of preeclampsia (PE). Angiotensin II receptor subtype 2 (AT2R) can antagonize the effects of AT1R, but its effects during pregnancy are not known. We investigated the effect of AT2R on the pathogenesis of PE using a mouse model and recently developed AT2R agonist (compound 21 [C21]). Blastocysts collected from pregnant imprinting control region (ICR) mice were incubated with adenovirus containing the CD40L gene and transferred into the uterine horns of pseudo-pregnant ICR mice to express PE-like features. Osmotic pumps were placed subcutaneously on the dorsal side with C21 or saline. C21 reduced the plasma soluble fms-like tyrosine kinase 1 (sFlt-1) concentration, ameliorating hypertension. The splenic T and B cell profiles in model mice were analyzed by flow cytometry. The gated percentage of IFN-γ-positive Th cells was significantly increased and the percentage of plasma cells in B cells was significantly decreased; however, the percentages were not altered by C21. sFlt-1 and soluble endoglin concentrations in plasma were measured with an enzyme-linked immunosorbent assay, and sFlt-1 was reduced. C21 could become a candidate PE drug as it ameliorated the pathophysiology of PE as a result of decreased production of sFlt-1.
Effect of Vascular Endothelial Growth Factor on Pre-Eclampsia in Pregnant Rats

Abstract:OBJECTIVE:To investigate relationship between the vascular endothelial growth factor (VEGF) and the pathogenesis of pre-eclampsia in pregnant rats.METHODS:Pregnant rats were divided into two groups randomly. Saline solution or L-nitro arginine methyl ester (L-NAME) 125 mg/d was given subcutaneously from day 7 of gestation till establishing pre-eclampsia. Systolic blood pressure, urine protein, platelet count, and weight of pups and placenta were determined. The levels of VEGF in pregnant rats venous serum, placenta and decidual tissue from normal pregnancy and pre-eclampsia rats were detected by ELISA and immunohistochemistry, respectively.RESULT:Pregnant rats which were given L-NAME produced physical signs similar to those of pre-eclampsia, such as increase in systolic blood pressure [(145.3 +/-4.6)mmHg] and urine protein [(814.3 +/-57.5)mg/L], and decrease in platelet count [(467.1 +/-76.3) x 10(9)/L] and weight of pups and placenta. Compared with controls, the intensity of VEGF immunostaining in trophoblast or decidual cells were significantly reduced. The serum levels of VEGF were significantly lower in pre-eclampsia group than in normal pregnancy.CONCLUSION:Decreased serum levels of VEGF and reduced expression of VEGF in placental tissues might in part explain the pathogenesis of pre-eclampsia in pregnant rats.
Effects of Autoantibodies Against AT1 Receptor from Patients with Refractory Hypertension on Intracellular Ca2+ Mobilization of Rat Vascular Smooth Muscle Cells

Abstract:Objective:To explore the effect of autoantibodies against AT1 receptor of patients with refractory hypertension on the intracellular Ca2+ mobilization in vascular smooth muscle cells (VSMC).Method:Forty-six patients with hypertension were recruited and divided into two groups based on whether their blood pressure was above or below 140/90 mmHg after combination drug therapy: a refractory hypertension group (n=22) and a non-refractory hypertension group (n=24). The epitope of the second extracellular loop of AT1 receptor was synthesized and used as antigen to screen the antibodies by ELISA. IgG from patients with the refractory hypertension was obtained by affinity purification, and was used to prove the affinity with AT1-receptor in the cultured vascular smooth muscles (VSMC) by immunocytochemistry. Then, Fluo-3/AM, a Ca2+ fluorescent indicator was used to load VSMC. IgG from patients was used to stimulate VSMC and the change of fluorescence was monitored by confocal microscopy.Result:Frequency of the autoantibodies against AT1 receptor in refractory hypertension group was higher than that in non-refractory hypertension group (P0.05). And VSMC showed increased intracelluar Ca2+ mobilization induced by the patients’ antibodies against AT1 receptor. Furthermore, we found the autoantibody-induced Ca2+ increase could be blocked by losartan, an AT1 receptor antagonist.Conclusion: Antibodies against AT1 receptor with the ability to activate AT1 receptor may account for Ca2+ increase in VSMC. And the autoantibodies may involve in the pathogenesis of hypertension, especially in patients refractory to drug treatment.
The Inhibition of Protein Kinase C Β Contributes to the Pathogenesis of Preeclampsia by Activating Autophagy.

Abstract:BACKGROUND:Preeclampsia is a devastating hypertensive disorder of pregnancy with unknown mechanism. Recent studies have considered abnormal autophagy as a new cellular mechanism for this disorder, while little is known about how autophagy is specifically involved and what factors are implicated. Here, we report a previously unrecognized preeclampsia-associated autophagic regulator, PKCβ, that is involved in placental angiogenesis.METHODS:PKCβ levels were evaluated by quantitative real-time PCR, western blotting, immunofluorescence and by the analysis of public data. The autophagy-regulating role of PKCβ inhibition in preeclampsia pathogenesis was studied in a mouse model, and in human umbilical vein endothelial cells (HUVECs) and human choriocarcinoma cells (JEG-3).FINDINGS:PKCβ was significantly downregulated in human preeclamptic placentas. In a mouse model, the selective inhibition of PKCβ by Ruboxistaurin was sufficient to induce preeclampsia-like symptoms, accompanied by excessive autophagic flux and a disruption in the balance of pro- and anti-angiogenic factors in mouse placentas. In contrast, autophagic inhibition by 3-methyladenine partially normalized hypertension, proteinuria and placental angiogenic imbalance in PKCβ-inhibited mice. Our in vitro experiments demonstrated that PKCβ inhibition activated autophagy, thus blocking VEGFA-induced HUVEC tube formation and resulting in the significant upregulation of sFLT1 and downregulation of VEGFA in JEG-3 cells.INTERPRETATION:These data support a novel model in which autophagic activation due to PKCβ inhibition leads to the impairment of angiogenesis and eventually results in preeclampsia.FUNDING:Shanghai Key Program of Clinical Science and Technology Innovation, National Natural Science Foundation of China and Shanghai Medical Center of Key Programs for Female Reproductive Diseases.
The role of T cell stimulated agonistic autoantibodies to the angiotensin II type I receptor (AT1‐AA) in mediating multiorgan dysfunction in IL‐17 induced hypertension during pregnancy

Abstract:Abstract Problem Preeclampsia (PE), new‐onset hypertension during pregnancy accompanied by organ dysfunction, is associated with chronic inflammation including elevated IL‐17, CD4+ T cells, B cells and natural killer (NK) cells. IL‐17 can serve as a signal for either the adaptive or innate immune activation. We have previously shown that IL‐17 contributes to increased blood pressure in association with elevated TH17 cells, NK cells and B cells secreting angiotensin II type 1 receptor agonistic autoantibodies (AT1‐AA) during pregnancy. Moreover, we have shown an important role for CD4+T cells and AT1‐AA in multiorgan dysfunction as measured by mitochondrial oxidative stress (mt ROS). However, we do not know the role of adaptive immune cells such as T cells or B cells secreting AT1‐AA in mediating the PE phenotype in response to elevated IL‐17. Method of study In order to answer this question, we infused IL‐17 (150 pg/day i.p.) into either Sprague Dawley (SD) or athymic nude rats via mini‐osmotic pump from gestational day (GD) 14–19 of pregnancy. On GD 19, blood pressure was determined and NK cells, mtROS and respiration and AT1‐AA production from B cells were measured. Results Infusion of IL‐17 increased blood pressure in the presence or absence of T cells. Mean arterial pressure (MAP) increased with IL‐17 from 98 ± 2 mm Hg ( n = 12) to 114 ± 2 ( n = 12) in SD rats and from 99 ± 4 mm Hg ( n = 7) versus 115 ± 2 mm Hg ( n = 7) in athymic nude rats. Similar trends were seen in NK cells and placental mt ROS. Knowing that IL‐17 stimulates AT1‐AA in SD pregnant rats, we included a group of SD and athymic nude pregnant rats infused with IL‐17 and the AT1‐AA inhibitor peptide (‘n7AAc’). The inhibitor attenuated blood pressure (104.9 ± 3.2, p = .0001) and normalized NK cells and mt function in SD pregnant rats. Importantly, the AT1‐AA was not produced in pregnant nude IL‐17 treated rats, nor did ‘n7AAc’ effect MAP, in nude athymic rats. Conclusion These findings suggest two conclusions; one is that IL‐17 causes hypertension and multiorgan dysfunction in the absence of T cells and AT1‐AA, possibly through its activation of innate cells and secondly, in the presence of T cells, blockade of the AT1‐AA attenuates the effect of IL‐17. This study indicates the critical effects of elevated IL‐17 during pregnancy and suggest treatment modalities to consider for PE women.

Abstract:We implement cross-correlated imaging in the frequency domain (fC(2)) in order to reconstruct different modes propagating in a multi-mode optical fiber, and measure their relative powers. Our system can complete measurements in under a second (950 ms), with a maximum signal to noise ratio of 25 dB. The system is capable of group-delay temporal resolution as high as 720 fs, and this number can be tailored for a variety of modal discrimination levels by choice of apodization functions and effective bandwidths of the tunable source we use. Measurements are made on a double-clad test fiber to demonstrate simultaneous reconstruction of six guided modes. Finally, the system is used to optimize alignment into the fiber under test and achieve mono-mode purity > 95%, underscoring the utility of fC(2) imaging for near-real-time modal content analysis.
Expression and influence of KATP in umbilical artery smooth muscle cells of patients with hypertensive disorders of pregnancy

Abstract:The objective of this study is to investigate the expression and influence of adenosine triphosphate-sensitive potassium channel (KATP) in human umbilical arterial smooth muscle cells (HUASMCs) of patients with hypertensive disorders of pregnancy (HDP). Western blotting was used to detect the protein expression levels of KATP inwardly rectifying potassium channel (Kir)6.1 and sulphonylurea receptor (SUR)2B subunits in HUASMCs from patients with normal parturients (NP), gestational hypertension (GH), chronic hypertension (CH), preeclampsia (PE) and chronic hypertension with superimposed preeclampsia (CHSP), respectively. There was no significant difference in the protein expression of Kir6.1 subunit in NP group, GH group, CH group, PE group and CHSP group ( P > 0.05). The protein expression of SUR2B subunit was gradually decreased in NP group, GH group, CH group, PE group and CHSP group, with statistically significant difference among the groups ( P < 0.05). The altered expression level of KATP SUR2B subunit may be involved in the pathogenesis of HDP. The severity of HDP may be related to the degree of decrease of SUR2B subunit.

Abstract:OBJECTIVE:To investigate the changes in serum levels of Th1- (IL-2 and TNF-alpha) and Th2-type cytokines (IL-10) and the ratios of Th1/Th2 (IL-2/IL-10 and TNF-alpha/IL-10) in preeclampsia and in gestational hypertension.METHODS:Levels of IL-2, IL-10 and TNF-alpha were determined with radioimmunoassay in serum samples from 22 women with preeclampsia, 15 women with gestational hypertension and 32 normal term pregnant women. The Th1/Th2 ratios were calculated accordingly.RESULT:There were no significant differences in serum levels of IL-2, IL-10 and TNF-alpha (P>0.05 for all) among normal pregnancy, gestational hypertension and preeclampsia. The ratio of serum IL-2/IL-10 was significantly higher in preeclampsia than that in controls (P < 0.05), and the ratio of TNF-alpha/IL-10 significantly higher in patients with preeclampsia than that in either controls or gestational hypertension (P<0.025 for both).CONCLUSION:Alterations of serum cytokine balance with predominance of Th1 immunity were observed in preeclampsia. These associations may offer insight into the pathogenesis of preeclampsia.

Abstract:The vasoactive peptide AngiotensinII (AngII) is an important factor in the cardiovascular system, exerting most of its effects through AngII receptor type 1 (AT(1)). Ib, a new nonpeptide AT(1) receptor antagonist, has been observed to play a positive role in the treatment of hypertension in preclinical tests. In this study, the inhibitory effects of Ib on AngII-induced vascular contraction in vitro were investigated, and its molecular mechanisms were further explored. In endothelium-denuded aortic rings from rabbits, Ib produced a rightward shift in the concentration-response curve for AngII with a decrease in the maximal contractile response and the pD'(2) was 7.29. In vascular smooth muscle cells (VSMCs), the specific binding of [I-125]AngII to AT(1) receptors was inhibited by Ib in a concentration-dependent manner with IC50 value of 0.96 nM. Ib could inhibit both AngII-induced Ca2+ mobilization from internal stores and Ca2+ influx. Moreover, the translocation of PKC-alpha stimulated by AngII was inhibited by Ib. Thus, the inhibitory effects of Ib might be related with the depression on AngII-induced increase in [Ca2+](i) and translocation of PKC-alpha through blocking AT(1) receptors. (C) 2007 Elsevier Inc. Abstract:An adverse relationship between suboptimal fetal environments and the development of adult diseases, such as hypertension, type II diabetes, and cardiovascular disease, has been reported in numerous studies. The purpose of this study was to investigate the strain difference of offspring’s response to maternal malnutrition during pregnancy and the involvement of the renin-angiotensin system (RAS) in the development of adult hypertension using C57BL/6J (C57) mice and angiotensin II (Ang II) type 1 receptor-associated protein-transgenic (ATRAP-Tg) mice. Pregnant dams were fed an isocaloric diet containing either 20% (normal protein; NP) or 8% (low protein; LP) protein. Birth weight was significantly reduced in C57-LP offspring, but not in ATRAP-Tg-LP offspring. Arterial blood pressure was higher in C57-LP offspring than in the other groups. In contrast, ATRAP-Tg-LP offspring did not show an increase in blood pressure compared with NP offspring. Renal angiotensin II type 1 (AT1) receptor expression was not altered by maternal malnutrition, whereas angiotensin II type 2 receptor expression was significantly decreased in C57-LP offspring. In conclusion, these findings suggest that a suboptimal intrauterine environment induces adult hypertension because of an alteration of expression of RAS components, which was partly suppressed by sustained ATRAP overexpression via attenuation of the AT1 receptor-mediated pathological response.
