Abstract:We developed a facile strategy to obtain a new kind of mesoporous core-shell structured up-conversion nanoparticles (mUCNPs), composed of a NaYbF4:2%Er core and a mesoporous NaGdF4 shell. This mesoporous shell not only enhanced the up-conversion luminescence but also endowed many other functionalities of the nanoparticles such as drug delivery and bio-imaging capabilities. Moreover, after being conjugated with polyethylenimine (PEI) and folic acid (FA), core-shell mUCNPs exhibited good water dispersibility, enhanced drug delivery efficiency, and remarkable targeting ability to cancer cells. To certify the folate receptors (FR)-mediated targeted drug delivery, cell viability assay, cell up-conversion luminescence imaging and flow cytometry analysis were carried out. Furthermore, apart from the application for targeted drug delivery, the as-prepared core-shell mUCNPs could also be employed as the contrast agents for X-ray computed tomography (CT) and magnetic resonance (MR) imaging, because of the strong X-ray attenuation ability of Yb and high longitudinal molar relaxivity (r1) of Gd in the nanoparticles, providing the potential for simultaneously bio-imaging and cancer-targeting therapy.
Direct imaging the upconversion nanocrystal core/shell structure at the subnanometer level: shell thickness dependence in upconverting optical properties

Abstract:Lanthanide-doped upconversion nanoparticles have shown considerable promise in solid-state lasers, three-dimensional flat-panel displays, and solar cells and especially biological labeling and imaging. It has been demonstrated extensively that the epitaxial coating of upconversion (UC) core crystals with a lattice-matched shell can passivate the core and enhance the overall upconversion emission intensity of the materials. However, there are few papers that report a precise link between the shell thickness of core/shell nanoparticles and their optical properties. This is mainly because rare earth fluoride upconversion core/shell structures have only been inferred from indirect measurements to date. Herein, a reproducible method to grow a hexagonal NaGdF(4) shell on NaYF(4):Yb,Er nanocrystals with monolayer control thickness is demonstrated for the first time. On the basis of the cryo-transmission electron microscopy, rigorous electron energy loss spectroscopy, and high-angle annular dark-field investigations on the core/shell structure under a low operation temperature (96 K), direct imaging the NaYF(4):Yb,Er@NaGdF(4) nanocrystal core/shell structure at the subnanometer level was realized for the first time. Furthermore, a strong linear link between the NaGdF(4) shell thickness and the optical response of the hexagonal NaYF(4):Yb,Er@NaGdF(4) core/shell nanocrystals has been established. During the epitaxial growth of the NaGdF(4) shell layer by layer, surface defects of the nanocrystals can be gradually passivated by the homogeneous shell deposition process, which results in the obvious enhancement in overall UC emission intensity and lifetime and is more resistant to quenching by water molecules.

Abstract:Lanthanide‐doped upconversion nanoparticles (UCNPs) have attracted a lot of interest due to their benefits in biological applications: They are not suffering from intermittence and provide nearly background‐free luminescence. The progress in synthesis nowadays enables access to complex core‐shell particles of controlled size and composition. Nevertheless, the frequently used doping ratio dates back to where mostly core‐only particles of relatively large size have been studied. Especially at low power excitation as needed in biology, a decrease in particle size leads to a drastic decrease in the upconversion efficiency. An enhancement strategy based on an increased absorption rate of near‐infrared light provided by an increase of the sensitizer content, together with the simultaneous blocking of the energy migration pathways to the particle surface, is presented. NaYbF4(20%Er) particles of 8.5 nm diameter equipped with an about 2 nm thick NaYF4 shell show significantly enhanced upconversion luminescence in the red (660 nm) compared to the most commonly used particles with only 20% Yb3+ and 2% Er3+. The impact of size, composition, and core‐shell architecture on photophysical properties are studied. The findings demonstrate that an increase in doping rates enables the design of small, bright UCNPs useful for biological applications. A higher lanthanide concentration of 80% sensitizer (Yb3+) and 20% activator (Er3+) increases the absorption rate of upconversion nanoparticles. A NaYF4 shell blocks the energy migration pathways to the particle surface, which enhances the upconversion luminescence upon near‐infrared excitation significantly. An investigation of particle diameter and shell thickness reveals significantly enhanced luminescence in the red (660 nm) for bioanalytical applications.

Abstract:NaYF4:Yb,Er@NaYF4 core–shell nanostructures were prepared to investigate their influence on upconversion (UC) luminescence. Tests revealed green radiation (4S3/2→4I15/2) and red radiation (4F9/2→4I15/2) first increased and then gradually decreased as Yb concentration increased in the NaYF4 shell. The strongest fluorescent radiation occurred at an Yb concentration of 5%. To investigate the complicated variation of luminescence, we designed a set of experiments to study the impact of Yb ion concentration on luminescence intensity, and we analyzed the corresponding enhancement mechanism. It is probable that the energy transfers between both Yb and Er ions and Yb and Yb ions are involved in the UC processes. The enhancement of hybrid nanostructures has huge potential in biological detection and solar cells.

Abstract:High sensitizer and activator concentrations have been increasingly examined to improve the performance of multi-color emissive upconversion (UC) nanocrystals (UCNC) like NaYF 4 :Yb,Er and first strategies were reported to reduce concentration quenching in highly doped UCNC. UC luminescence (UCL) is, however, controlled not only by dopant concentration, yet by an interplay of different parameters including size, crystal and shell quality, and excitation power density ( P ). Thus, identifying optimum dopant concentrations requires systematic studies of UCNC designed to minimize additional quenching pathways and quantitative spectroscopy. Here, we quantify the dopant concentration dependence of the UCL quantum yield ( Φ UC ) of solid NaYF 4 :Yb,Er/NaYF 4 :Lu upconversion core/shell nanocrystals of varying Yb 3+ and Er 3+ concentrations (Yb 3+ series: 20%–98% Yb 3+ ; 2% Er 3+ ; Er 3+ series: 60% Yb 3+ ; 2%–40% Er 3+ ). To circumvent other luminescence quenching processes, an elaborate synthesis yielding OH-free UCNC with record Φ UC of ∼9% and ∼25 nm core particles with a thick surface shell were used. High Yb 3+ concentrations barely reduce Φ UC from ∼9% (20% Yb 3+ ) to ∼7% (98% Yb 3+ ) for an Er 3+ concentration of 2%, thereby allowing to strongly increase the particle absorption cross section and UCNC brightness. Although an increased Er 3+ concentration reduces Φ UC from ∼7% (2% Er 3+ ) to 1% (40%) for 60% Yb 3+ . Nevertheless, at very high P (> 1 MW/cm 2 ) used for microscopic studies, highly Er 3+ -doped UCNC display a high brightness because of reduced saturation. These findings underline the importance of synthesis control and will pave the road to many fundamental studies of UC materials.

Abstract:Surface coating is a commonly used strategy to enhance upconversion emissions by shielding the luminescent core from surface quenching. In this work, we provide insights into the effect of surface coating on upconversion by investigating NaYF4 :Yb/Er nanoparticles and the corresponding NaYF4 :Yb/Er@NaYF4 core-shell nanoparticles, as a function of dopant concentration of Yb(3+) and excitation power. We observe declining emission enhancement factors with decreasing Yb(3+) concentration and increasing excitation power. Our mechanistic investigations suggest that the phenomenon originates from stepwise excitation in the upconversion process, as well as energy hopping among the Yb(3+) dopants. This increased understanding of the effect of surface coating on upconversion should be important towards the rational design of lanthanide-doped core-shell nanoparticles for various applications.

Abstract:NaYF4:Yb,Er nanocrystals with different concentrations of Gd3+ ions are prepared via a hydrothermal method. With increasing Gd3+ dopant, up-conversion (UC) emission is gradually enhanced and the strongest UC emission is improved by 50-fold for a sample with 15% Gd3+ doping compared with the one without Gd3+ incorporation. However, further increasing the Gd3+ dopant causes the reduction of UC emission which can be ascribed to the increased surface state recombination. Moreover, the self-assembly technique is used to fabricate Au nanosphere arrays with diameters of 300 nm to enhance the UC luminescence. It is found that the luminescence intensity is increased by 2-fold due to the surface plasmon resonance effect which is confirmed by the absorption spectra.

Abstract:The growing applications of upconversion nanocrystals in bioimaging, therapeutics, and photonics have given rise to a demand of high quality nanocrystals with desirable luminescence intensity. Although the design of optimal nanocrystals such as core–shell nanostructures has improved the intensity, the internal links between dopant concentration balance, epitaxial growth protection, and shell thickness effect encounter a compromised situation that lacks of integrated consideration and comprehensive assessment. Here we propose an integrated strategy based on a core–shell design for the enhancement of upconversion luminescence intensity. Epitaxial protection can enable higher activator accommodation capacity in limited spatial scale, which leads to an Er3+ concentration threshold improvement in β-NaYF4 core–shell nanocrystals from 2 to 6 mol %. We further perform a comprehensive assessment of the nanocrystals with convincing performance improvement in ensemble spectroscopic intensity, upconversion quantum yield, and single nanocrystal intensity. Our findings provide improved understanding of electronic behaviors in multiphoton upconversion and opportunities for diverse applications requiring high quality upconversion nanocrystals.

Abstract:In this work, the energy transfer process and upconversion luminescence properties of NaYF4: Ln3+@NaYF4: Ln3+ (Ln = Yb3+, Er3+, Tm3+) core-shell microcrystals were studied through introducing different Yb3+ ions in core and shell. Compared with Yb3+/Tm3+/Er3+ co-doped NaYF4 microcrystals, the NaYF4:Yb3+/Tm3+@NaYF4: Yb3+/Er3+ core-shell microcrystals displays greater stability spectral characteristics under NIR 980 nm excitation. This is because the core-shell structures can control the excitation energy acquisition channel and avoid interactions between Er3+ and Tm3+ ions. Meanwhile, the luminescence properties of NaYF4@NaYF4 core-shell microcrystals can also be precisely controlled by changing concentrations of Yb3+ ions in the core and shell, respectively. The experimental results further indicate that upconversion emission of microcrystals can also be precisely controlled by constructing core-shell structures. The multicolor emission core-shell microcrystals may have great potential in display, anti-counterfeiting and micro-photoelectric technology applications.

Abstract:A new type of core-shell upconversion nanoparticles which can be effectively excited at 795 nm has been designed and synthesized through spatially confined doping of neodymium (Nd(3+)) ions. The use of Nd(3+) ions as sensitizers facilitates the energy transfer and photon upconversion of a series of lanthanide activators (Er(3+), Tm(3+), and Ho(3+)) at a biocompatible excitation wavelength (795 nm) and also significantly minimizes the overheating problem associated with conventional 980 nm excitation. Importantly, the core-shell design enabled high-concentration doping of Nd(3+) (~0 mol %) in the shell layer and thus markedly enhanced the upconversion emission from the activators, providing highly attractive luminescent biomarkers for bioimaging without autofluorescence and concern of overheating.
Revisiting the Optimized Doping Ratio in Core/shell Nanostructured Upconversion Particles.

Abstract:The development of rare-earth doped upconversion nanoparticles (RE-UCNPs) in various applications is fuelling the demand for nanoparticles with highly enhanced upconversion luminescence (UCL). Although the core/shell structure is proved to enhance the UCL effectively, there is still plenty of room to further improve the UCL by optimizing the doping ratio of the materials. In this article, a general strategy is demonstrated to achieve highly-enhanced visible UCL in core/shell nanostructured NaREF4 by increasing the doping ratio of Yb3+ in the core region. The energy transfer from RE-UCNPs to surface quenching sites through Yb3+-Yb3+ energy migration is demonstrated to be the main reason for restricting the doping ratio of Yb3+. Notable UCL enhancement (ca. 15 times) of core/shell structured α-NaYF4:Yb,Er@CaF2 nanoparticles is observed by increasing the concentration of Yb3+ to 98 mol%. The highly-enhanced visible UCL signal is used to guide the lymphatic vessel resection with the naked eye.
Changing J774A.1 Cells to New Medium Perturbs Multiple Signaling Pathways, Including the Modulation of Protein Kinase C by Endogenous Sphingoid Bases*

Abstract:Sphingosine, sphinganine, and other long-chain (sphingoid) bases are highly bioactive intermediates of sphingolipid metabolism that have diverse effects when added to cells, including the inhibition of protein kinase C (PKC) as evaluated by both enzymatic activity and [3H]phorbol dibutyrate ([3H]PDBu) binding. Nonetheless, changes in endogenous sphingoid bases have not been proven to affect PKC or other signal transduction pathways. We have discovered recently that changing J774A.1 cells to new medium results in up to 10-fold increases in sphingoid bases (Smith, E. R., and Merrill, A. H., Jr. (1995) J. Biol. Chem. 270, 18749-18758); therefore, this system was used to elevate sphingosine and sphinganine and determine if PKC was affected. Incubation of J774A.1 cells in new medium for 30 min increased the levels of these endogenous sphingoid bases to approximately 0.5 nmol/mg of protein and decreased [3H]PDBu binding by 40-60%. Addition of NH4Cl, which suppresses the change in sphingosine, restored [3H]PDBu binding. Elevation of endogenous sphinganine by a second method (addition of fumonisin B1, an inhibitor of ceramide synthase) also reduced [3H]PDBu binding; therefore, elevations in sphingosine and sphinganine can both affect PKC. The elevation in sphingoid bases was also associated with an increase in the amount of PKC-δ (the major PKC isozyme in J774A.1 cells) in the cytosol, as determined by activity assays and immunoblot analyses. Changing the culture medium affected other PKC isozymes, increased cellular levels of diacylglycerol, dihydroceramide, and ceramide, and altered the expression of two genes (the expression of JE was increased, and the induction of MnSOD by TNF-α was potentiated). Thus, changing the culture medium has numerous effects on J774A.1 cells, including the modulation of PKC by endogenous sphingoid bases.

Abstract:For the purpose of finding out a strain which has stronger cytolytic activity on Ehrlich ascites carcinoma cells, the authors isolated 113 strains of Bacillus natto from straws, which were collected at various areas in Japan, and measured the cytolytic activities by cylinder plate method. As the results, the authors found out a strain of Bacillus natto (tentatively called KMD 2311) which has the strongest cytolytic activity in the 113 strains. There were at least two kind of cytolytic substances on Ehrlich ascites carcinoma cells in the culture medium of Bacillus natto KMD 2311. One was extracted with AcOEt from the culture medium, which constitute approximately 20% of the cytolytic activity in the culture medium and it was stable. This cytolytic substance was purified and colorless crystalline compound (mp 247-249°) was obtained by recrystallization from acetone-petr. ether. Chemical structure of this compound was examined by elementary analysis, infrared, nuclear magnetic resonance, and mass spectroscopy and study of decomposed products. As the results, this compound was proved to be identical with surfactin (mp 140°), which was obtained from culture medium of Bacillus subtilis by Kakinuma, et al. and from Bacillus natto KMD 1126 by the authors. Melting point of this compound (mp 249°) was higher about 100° than that of surfactin (mp 140°), but high mp compound was obtained from surfactin by recrystallization from acetone-petr. ether. From these results, it was indicated that the cytolytic substance was identified with surfactin and dimorphic.
Upconverting Core-Shell Nanocrystals with High Quantum Yield under Low Irradiance: On the Role of Isotropic and Thick Shells

Abstract:Colloidal upconverter nanocrystals (UCNCs) that convert near-infrared photons to higher energies are promising for applications ranging from life sciences to solar energy harvesting. However, practical applications of UCNCs are hindered by their low upconversion quantum yield (UCQY) and the high irradiances necessary to produce relevant upconversion luminescence. Achieving high UCQY under practically relevant irradiance remains a major challenge. The UCQY is severely limited due to non-radiative surface quenching processes. We present a rate equation model for migration of the excitation energy to show that surface quenching does not only affect the lanthanide ions directly at the surface but also many other lanthanide ions quite far away from the surface. The average migration path length is in the order of several nanometers and depends on the doping as well as the irradiance of the excitation. Using Er3+-doped beta-NaYF4 UCNCs, we show that very isotropic and thick (~10 nm) beta-NaLuF4 inert shells dramatically reduce the surface-related quenching processes, resulting in much brighter upconversion luminescence at simultaneously considerably lower irradiances. For these UCNCs embedded in PMMA, we determined an internal UCQY of 2.0+-0.2 % using an irradiance of only 0.43+-0.03 W/cm2 at 1523 nm. Normalized to the irradiance, this UCQY is 120x higher than the highest values of comparable nanomaterials in the literature. Our findings demonstrate the important role of isotropic and thick shells in achieving high UCQY at low irradiances from UCNCs. Additionally, we measured the additional short-circuit current due to upconversion in silicon solar cell devices as a proof concept and to support our findings determined using optical measurements.

Significantly Boosted Upconversion Emission in Cryogenic Er@Yb@Y Core–Shell–Shell Nanostructures

Abstract:The advantages of active shell sensitization and the cryogenic field‐induced cross relaxation suppression are combined in this study, which significantly promotes the upconversion emission brightness of Er3+‐rich core–shell nanostructures 1–2 orders of magnitude. Based on the unique property of the material, a temperature‐induced high‐level information encryption application with QR code is successfully developed. Recent advances reveal that due to the cross‐relaxation restriction, impressive upconversion (UC) enhancement (≈100‐folds) can be achieved in cryogenic Er3+‐rich core‐inert shell nanostructures (e.g., NaErF4@NaYF4), which opens up exciting opportunities in diverse frontier applications. However, further promotion of UC intensity is still highly desired, in which the rational design of nanostructures can play a key role. Herein, it is demonstrated that adopting an active shell design will constantly benefit the UC within a wide temperature range (40–300 K). Specifically, through constructing the luminescent core@active shell@inert shell sandwich nanostructure (e.g., NaErF4@NaYbF4@NaYF4), 8.3–73‐folds UC enhancement will be achieved (taking the corresponding core@inert shell structures as competitors). Moreover, from spectral‐domain and time‐domain spectroscopic experiments, the relevant UC enhancement is convincingly attributed to a temperature‐dependent energy injection process (from the active shell to the luminescent core). More interestingly, the unique property of the material makes a temperature‐induced high‐level encryption application possible, which is obtained by employing the nanomaterials on a quick response (QR) code. These results not only deepen the UC mechanism in multi‐layer nanostructures, but also introduce an expanded dimension (via low temperatures) in information security.

Abstract:The ability to synthesize upconversion nanocrystals (UCNCs) with tailored upconversion luminescence and controlled size is of great importance for biophotonic applications. However, until now, limited success has been met to prepare bright, ultrasmall, and monodispersed β-NaYF₄:Yb3+/Er3+ UCNCs. In this work, we report on a synthetic method to produce monodisperse hexagonal NaYF₄:Yb3+/Er3+ nanocrystals of ultrasmall size (5.4 nm) through a precise control of the reaction temperature and the ratio of Na⁺/Ln3+/F-. We determined the optimum activator concentration of Er3+ to be 6.5 mol % for these UCNCs, yielding about a 5-fold higher upconversion luminescence (UCL) intensity than the commonly used formula of NaYF₄:30% Yb3+/2% Er3+. Moreover, a thin epitaxial shell (thickness, 1.9 nm) of NaLnF₄ (Ln = Y, Gd, Lu) was grown onto these ultrasmall NaYF₄:Yb3+/Er3+ NCs, enhancing its UCL by about 85-, 70- and 50-fold, respectively. The achieved sub-10-nm core and core-shell hexagonal NaYF₄:Yb3+/Er3+ UCNCs with enhanced UCL have strong potential applications in bioapplications such as bioimaging and biosensing.

Abstract:In this study, a series of NaYF4:Yb3+, Tm3+@NaYF4:Yb3+, Er3+ core@shell nanocrystals with different sensitizer contents in the shell were synthesized by a facile solvothermal method to investigate the impact of sensitizer Yb3+ on the upconversion luminescence. Transmission electron microscopy (TEM) results show that all prepared nanocrystals are well-crystallized with a hexagonal phase and have a desirable size distribution. The particle size increases after shell coating as shown in TEM images; however, it changes a little for samples with different sensitizer contents in the shell. Therefore, the effect of different particle sizes on luminescence properties, which is caused by different Yb3+ contents, could be minimized. Subsequently, it was assumed that the luminescent center content (Er3+) is same in all samples and does not influence the emission spectrum differences. Therefore, we investigated the underlying upconversion luminescence mechanism by determining the sample’s fluorescence spectrum and assuming that the energy transfer mechanism occurred in all the samples. The results show that the green upconversion luminescence decay was faster than its red counterpart when the Yb3+ content was increased and samples were excited using a 980 nm pulse laser. The transient spectra measured at different times after the excitation pulse reveal the intensity ratio associated with the green to red upconversion luminescence. It was concluded that the back energy transfer (BET) process from Er3+ to Yb3+ led to a conversion of the green emissions into the red emissions, which was beneficial to the red upconversion and the electron relaxation from 4I11/2 to 4I13/2. Therefore, the red to green upconversion luminescence ratio increases significantly in samples highly doped with Yb3+.

Abstract:Upconversion luminescence materials have gained extensive attentions owing to their unique luminescence characteristics of converting near-infrared excitation to visible light emission. However, how to improve the doping concentration of activator (usually <3 mol%) in traditional sensitizer-activator system is still a great challenge due to the luminescence quenching effect. In this work, NaErF4 crystals with diverse crystal phases and morphologies were prepared by simple hydrothermal method, constructing a 100% doping concentration system of activator (Er3+). With the increase of F− content, the phase of NaErF4 transfers from cubic to hexagonal, and the morphology changes from nanospheres to hexagonal microflakes, meanwhile, the upconversion luminescence intensity also increases gradually. In addition, in order to further inhibit the luminescence quenching effect of NaErF4 bare core, a series of NaErF4@SiO2 core-shell samples with different SiO2 shell thickness were successfully prepared. Compared with NaErF4 bare core, the morphology and structure of the NaErF4@SiO2 core-shell samples have no obvious change, but the UC luminescence intensity is significantly enhanced. Interestingly, the change of NaErF4 crystal phase and SiO2 shell thickness can both bring about multicolor luminescence. In addition, different from the traditional NaYF4:Yb, Er co-doping system, the as-prepared hydrophilic NaErF4 and NaErF4@SiO2 samples showed a greater red to green ratio (R/G > 1), which may expand their potential applications in biological imaging and sensing fields.

Studies in vitro on the effects of rhein on the chemotaxis of human leukocytes.

Abstract:Rhein (R: 1,8-dihydroxy-3-carboxyanthraquinone) is the active metabolite of the drug diacetylrhein (DAR), an anthraquinone molecule which has recently been proposed for the long-term treatment of osteoarthrosis. Its action mechanism in rheumatic pathology has not been fully explained. It is known that DAR, while not inhibiting the formation of prostaglandins, inhibits certain proteolytic enzymes, and acts on phlogistic cells by lysosomal enzymic and superoxide-anion modifications. Moreover DAR modifies phagocytic functions and the motility of cells. This paper is a contribution to the clarification of the last point, namely the effect of rhein on cell motility. It reports that in vitro no effect of R on random migration was found, but instead a double inhibiting effect on chemotaxis (i.e. a low-dosage and a high-dosage effect). Furthermore, R did not modify the inhibition or induce modification of chemotaxis by vinblastine. Finally R cancelled the stimulating effect of ionic potassium. The results thus indicate that R acts on the chemotaxis of the leukocytes with a complex action at different doses. The action mechanism is probably due to a membrane effect, since rhein (R) did not modify the chemotaxis-inhibiting activity of vinblastine but did interfere with the stimulating effect of K+.
