Abstract:A new analytical method was developed to simultaneously determine residues of 109 pesticides (including isomers) in the foods of animal origin. Acetonitrile was selected for accelerated solvent extraction (ASE) for effectively extracting the pesticides from the fatty samples. The cleanup was performed with an automated gel permeation chromatography (GPC) cleanup system. The prepared samples were analysed with GC–MS in the selected ion monitoring mode (SIM) using one target and two qualitative ions for each analyte. Chlorpyrifos-d10 was used as an internal standard. The lowest limit of detection was 0.3μgkg−1 for some pesticides. The recoveries and relative standard deviations (RSDs) were checked by spiking untreated samples with pesticides at 0.05, 0.1 and 0.2mgkg−1. The average recoveries of most pesticides were from 62.6% to 107.8%. The precision values expressed as RSD were all ⩽20.5% (n=6). Good linearity (r⩾0.99) was observed between 0.05 and 1.5μgmL−1.

Abstract:This article reports a comprehensive analytical method for the identification and quantification of a broad range of pesticides in green plant crops. The sample preparation method for pesticides involved an optimization of the QuEChERS-based extraction protocol, with sample mass, volume of added water, and the type of cleanup sorbent as variables. A sorbent combination based on ENVI-Carb and ChloroFiltr was examined. A highly efficient method was developed for the purification of plant extracts with 900 mg MgSO 4 , 150 mg PSA, and 15 mg ENVI-Carb at the d-SPE stage, combined with gas chromatography and liquid tandem mass spectrometry for the determination of 197 pesticides in crop plants containing chlorophyll. The method was validated in accordance with the requirements of international guidelines SANTE/11312/2021. The method was applied to quantify pesticide residues in 29 pairs of green crop plants and plants from the corresponding crop protection zone to verify whether the zones are effective barriers to prevent pesticides from penetrating outside agricultural areas. The number and types of agrochemical preparations were chosen by farmers. In total, more than 60 one- and several-component pesticide formulations were applied to the crops included in the study. The pesticide residues were detected in 21 crop samples and 3 samples from protection zones. Epoxiconazole, an active substance that was banned for use in 2021, was found in a spring barley sample. Based on the conducted research, the effectiveness of the protection zones has been clearly demonstrated, and it has been proven that environmental migration of pesticides and unauthorized agricultural practices pose a risk to ecosystems.

Abstract:An overview of the actual state-of-the-art sample treatment procedures for trace and ultra-trace determination of neonicotinoids (NQs) in environmental, food and biological samples is presented. Analytical procedures for NQs, sometimes together with their metabolites/by-products, and NQ-related new generation pesticides have been considered, focusing on sample preparation, which is essential in most cases due to both the complexity of the matrices and low concentrations of residual analytes. Different strategies have been practised for analytes extraction/preconcentration/cleanup, entailing various techniques as solid-liquid extraction (SLE), liquid-liquid extraction (LLE), offline and online solid-phase extraction (SPE), dispersive SPE (d-SPE), magnetic SPE (MSPE), solid-phase microextraction (SPME) and the quick, easy, cheap, effective, rugged and safe (QuEChERS) approach. New solid phases, including inorganic and carbon-based materials, have been prepared, also starting from “green” precursors, and successfully employed in the sample treatment step before hyphenated liquid chromatography or direct electrochemical detection. As well, new environmentally friendly solvents have been evaluated for LLE. The most recent studies (2022-present), around 50 papers, have been reviewed by matrix type, briefly described and commented concerning extraction efficiency, matrix effect, sensitivity, ease of application and sample throughput. Advantages and drawbacks were highlighted, as well as new achievements paving the way for further improvements.

Abstract:Residues of various highly polar pesticides and their metabolites are commonly found in numerous food products. Some of these compounds, such as glyphosate, are not only used in large amounts in agriculture, but are also controversially discussed in public. Here, we present a method, employing ion chromatography (IC) coupled to tandem mass spectrometry (IC-MS/MS), for the analyses of glyphosate, aminomethyl phosphonic acid (AMPA), N -acetyl-glyphosate (NAGly), fosetyl, and 10 further highly polar pesticides and metabolites in various plant and animal matrices following a minimal sample preparation by means of the QuPPe method. Thorough investigations showed that an AS19 column enabled the analysis of all 14 compounds within 30 min. The best sensitivity could be obtained with the make-up solvent acetonitrile being admixed to the mobile phase at a 1:2 flow rate ratio. Matrix effects were thoroughly studied in terms of ion suppression and retention time shifts. Conductivity detection was used to monitor elution profiles of matrix co-extractives in comparison with matrix effect profiles obtained by continuous post-column infusion of a mix with 13 highly polar pesticides and metabolites. These tests indicated that a fivefold dilution of QuPPe extracts was suitable for the routine analysis of samples for MRL-conformity, as it considerably reduced matrix effects maintaining sufficient sensitivity and high recovery rates in eight different commodities. The suitability of the final method for its application in routine analysis was verified by the analysis of >130 samples containing incurred residues where the results were compared with two existing LC-MS/MS methods.

Abstract:Bioactive compounds obtained from plants, microorganisms and minerals show some specific properties like insecticidal, herbicidal, repellent, antifeedant and toxicant activities called bio pesticide. They have specific modes of action against different pests. Due to their environmental eco-friendly nature, low cost, economic effectiveness, less pollution, and target specific quality they are in high demand in agriculture compared to chemical or synthetic pesticides. Extraction, purification, identification and characterization of these compounds from the plants materials are found always challenging. There are various types of traditional and non-traditional methods of extraction have been proposed such as maceration, distillation, ultrasonic-assisted extraction, soxhlet extraction, enzyme assisted extraction, microwave assisted extraction, accelerated solvent extraction, etc. have been reported for extraction of bioactive ingredients from plants complex matrix samples. The chromatographic separation techniques like thin layer chromatography(TLC), high performance thin layer chromatography (HPTLC), high performance liquid chromatography (HPLC) and gas chromatographic (GC) are used for their separation followed by the identification in order to determine their structure with the help of UV-Vis, fluorescence, NMR spectrometry, Fourier transforms infra-red spectrometry (FTIR) and mass spectrometry (MS). This review summarized the extraction procedure, formulation of biopesticide, structural identification and their application in agriculture.
Automated micro-solid-phase extraction clean-up and gas chromatography-tandem mass spectrometry analysis of pesticides in foods extracted with ethyl acetate

Abstract:Generic extraction methods for the multi-compound pesticide analysis of food have found their solid place in laboratories. Ethyl acetate and acetonitrile extraction methods have been developed as fast and easy to handle standard multi-compound methods, both feature benefits and limitations. The direct injection to gas chromatography can be impaired by a high burden of coextracted matrix, resulting in deterioration of the chromatographic system and matrix effects, requiring frequent maintenance. Therefore, common clean-up methods, such as dispersive solid-phase extraction, freeze-out of fats, or gel permeation chromatography, have been applied in clean-up. Automated clean-up using micro-solid-phase extraction (µSPE) is a recent development with several demonstrated advantages when employed in the analysis of pesticides and other contaminants in foods extracted with acetonitrile, but it has not yet been evaluated in this application using ethyl acetate for extraction. In this study, an automated procedure using µSPE cartridges was developed and established on an x,y,z robotic sampler for the raw extract clean-up and preparation of diluted samples for injection on a GC-MS/MS system. Validation experiments for 212 pesticides, polychlorinated biphenyls, and polycyclic aromatic hydrocarbons in lettuce, avocado, raspberry, paprika, egg, and liver extracts were performed using µSPE with MgSO4, PSA, C18, and CarbonX. The performance in routine operation is briefly discussed.

Abstract:Three sample preparation procedures were compared for a multi-residue analysis of pesticides in hops: (a) modified Hengel's method based on extraction with acetonitrile in combination with clean-up on a C18 SPE column, (b) miniaturized Biendl's method based on acetone extraction and PSA SPE sample clean-up, and (c) modified Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) method which utilizes a specific mixture of three sorbents (PSA, C18, and Z-Sep) for dispersive SPE sample clean-up. The performance of the methods was evaluated and validated for a mixture of 56 pesticides analyzed by the liquid chromatography–high resolution mass spectrometry in compliance with the analytical quality control criteria of the SANTE/11813/<span class="CitationRef"><a href="#CR14">2017</a></span> guidelines. Strong matrix-dependent signal suppression caused by the co-eluting hop matrix was observed for all sample preparations involved in this method comparison study. The matrix effects in percentages (%ME) were used for the evaluation of the effectiveness of the individual sample clean-up procedures. The recovery experiments were performed by spiking pesticides at the concentration level 0.50 mg/kg into the blank matrix to evaluate the extraction efficiency of the compared methods. Recoveries obtained for the modified Hengel's method were in the range of 70–120% with RSDs of less than 20% for all studied pesticides. The performances of the methods were tested on the set of 24 samples of hops harvested in the Czech Republic. The method comparison on the determined concentration levels of the pesticide residues clearly showed that the extraction efficiency of the QuEChERS method is significantly less effective for the extraction residues presented in a sample above the level 20 mg/kg. In terms of time consumption, labor, materials, and solvents consumptions, the methods were thoroughly compared, and these demands increase in the following order QuEChERS < modified Hengel's < miniaturized Biendl's method.

Abstract:A new method was evaluated and developed for the analysis of pesticides in sandy-loam soil by direct-immersion solid phase microextraction (DI-SPME) followed by gas chromatography tandem-mass spectrometry (GC-MS/MS) determination. Ten pesticides were selected based on a literature survey of the compounds reported to be present in EU soils. The extraction was performed using SPME LC-Tips, a new SPME configuration with the coated fibers attached to a disposable and easy-to-handle micropipette tip, which was immersed into a soil slurry made by the addition of an aqueous solution to the soil sample. Ten experimental parameters were evaluated with a Plackett-Burman design, after which the extraction time and percentage of organic solvent in the aqueous extraction were optimized separately. The two fiber chemistries available (PDMS/DVB and C 18 ) were evaluated in parallel for the entire work. In the final method, slurry samples were made by adding an aqueous solution (6% methanol v/v) to 2 g of soil. The fiber was conditioned and then inserted, for extraction, into the samples, stirred by a magnetic bar. Afterwards, the analytes were desorbed onto 100 μ L of methanol. After the addition of analyte protectants (ethylglycerol, gulonolactone, and sorbitol) the extract was injected into the GC-MS/MS system. Isotopically labelled penconazole was used as internal standard. A calibration was performed by extracting spiked soil with analyte concentrations of 0.1-50 μg/kg. Coefficients of determination of the linear calibration were between 0.94-0.98 for the PDMS/DVB and 0.92-0.99 for the C 18 . Limits of detection range between 0.01-10 μg/kg for the PDMS/DVB and 0.1-10 μg/kg for the C 18 . Overall, the C 18 analytically outperformed the PDMS/DVB but required a longer extraction time (120 min vs 75 min for the PDMS/DVB). This method allows automation and generates low residual toxic waste, having the potential to be introduced as a greener and simpler alternative to currently used sample preparation methodologies.

Abstract:Pesticide extraction in rapeseed samples remains a great analytical challenge due to the complexity of the matrix, which contains proteins, fatty acids, high amounts of triglycerides and cellulosic fibers. An HPLC-MS/MS method was developed for the quantification of 179 pesticides in rapeseeds. The performances of the quick, easy, cheap, effective, rugged, and safe (QuEChERS) method were evaluated using different dispersive solid-phase extraction (d-SPE) sorbents containing common octadecylsilane silica/primary-secondary amine adsorbent (PSA/C18) and new commercialized d-SPE materials dedicated to fatty matrices (Z-Sep, Z-Sep+, and EMR-Lipid). The analytical performances of these different sorbents were compared according to the SANTE/12682/2019 document. The best results were obtained using EMR-Lipid in terms of pesticide average recoveries (103 and 70 of the 179 targeted pesticides exhibited recoveries within 70-120% and 30-70%, respectively, with low RSD values). Moreover, the limits of quantification (LOQ) range from 1.72 µg/kg to 6.39 µg/kg for 173 of the pesticides. Only the recovery for tralkoxydim at 10 μg/kg level was not satisfactory (29%). The matrix effect was evaluated and proved to be limited between -50% and 50% for 169 pesticides with this EMR-Lipid and freezing. GC-Orbitrap analyses confirmed the best efficiency of the EMR-Lipid sorbent for the purification of rapeseeds.
Influence of QuEChERS modifications on recovery and matrix effect during the multi-residue pesticide analysis in soil by GC/MS/MS and GC/ECD/NPD

Abstract:A QuEChERS extraction followed by GC/MS/MS and GC-μECD/NPD for 216 pesticide and metabolites determination in soil simultaneously were developed and compared. Volume of water, volume and polarity of solvent, and cleanup sorbents (C18, GCB, PSA) were optimized. The QuEChERS with and without purification step were applied to estimate effectiveness of the method. The recovery and matrix effect (ME) were critical parameters within each tested procedure. The optimal method without cleanup was validated. Accuracy (expressed as recovery), precision (expressed as RSD), linearity, LOQ, and uncertainty were determined. The recoveries at the three spiking levels using matrix-matched standards ranged between 65 and 116% with RSD ≤17 and 60–112% with RSD ≤18% for MS/MS and μEC/NP, respectively. The LOQ ranged from 0.005–0.01 mg/kg for MS/MS to 0.05 mg/kg for μEC/NP. The ME for most of pesticides resulted in enhancement of the signal and depended on the analyte and detection system: MS/MS showed ME from −25 to 74%, while μEC/NP from −45 to 96%. A principal component analysis was performed to explain the relationships between physicochemical parameters and ME of 216 pesticides. The QuEChERS protocol without the cleanup step is a promising option to make the method less expensive and faster. This methodology was applied in routine analysis of 263 soil samples in which p,p’ DDT was the most frequently detected (23.5% of samples) and pendimethalin with the highest concentration (1.63 mg/kg).

Abstract:Despite organophosphorus pesticides (OPPs) benefits in controlling vector-borne diseases and noxious insects, the bioaccumulation and persistence in the soil system may metamorphose into new substances which could pose a serious threat to the ecosystems and human health. The generally low levels of OPPs residues and often the complexity of the soil matrix are the issues that researcher must deal with. Thus, it is essential to isolate and preconcentrate the OPPs from the matrix to reduce interference effects to obtain a reliable detection. Researchers have reported sample preparation techniques as a promising approach to improve analytical measurement of merits including recovery, precision, linearity, limit of detection, and limit of quantification. Under the selected conditions, limits of detection range between 0.001 and 143 ng/mL, and extraction recovery range between 5 and 154% were obtained. This review evaluates the challenges and opportunities, emphasizes the prospects of sampling techniques and various (micro)extraction coupled with chromatographic methods in different soil samples. Based on the finding, the extraction efficiency depended largely on the interaction between OPPs and extraction media. The fate, migration, toxicity impact, sampling procedure, and storage which influenced the sample preparation were comprehensively discussed.

Abstract:Nutraceutical products are plant-based materials primarily valued for their possible medicinal or therapeutic properties. According to their “natural” origin they are often perceived as being safe; however, for the prevention of economic losses, pesticides may be used against pest, mold and insects causing plant damage. Bearing in mind that a nutraceutical is a concentrated form of the plant, pesticides could be found in the final product in possibly worrying concentration. Nutraceutical products and dried herb materials typically represent very complex matrices for pesticide residue analysis, therefore a proper sample preparation protocol with appropriate clean-up steps is necessary prior to the analysis. Hence, this article gives an overview of pesticide residue analyses in nutraceuticals and provides the state of the art for sample preparation techniques applied for different types of nutraceuticals.

Abstract:Newly designed micro-solid phase extraction cartridges are now available, reflecting the increasing shift towards laboratory automation, especially in the clean-up step for the analysis of pesticide residues in food and feed. In the present study, the introduction of different sorbents on the newly designed PAL μSPE CTC cartridges was investigated for the removal of matrix interferents and the recovery of pesticides. Eight cartridges containing different sorbent combinations and different amounts were used including EMR-lipid (not activated), Z-sep, chitin, C18, PSA, and GCB. The evaluation of co-extractive removal for each cartridge showed that the optimal choice for removing fatty acids was the cartridges containing PSA and Z-sep as clean-up sorbents. However, the presence of C18 and EMR-lipid was still required for the removal of sterols and tocopherols. Two grams of sample, fish feed (FF) and rapeseed cake (RSC) were extracted using QuEChERS citrate buffer, followed by a freeze-out step. The recoveries and repeatability of QuEChERS using μ-SPE clean-up were evaluated for 216 pesticide residues (112 compounds analyzed by GC-MS/MS and 143 compounds by LC-MS/MS, from which 39 compounds were analyzed using both techniques). The best results, with recovery between 70 and 120% and RSD <20%, were achieved when FF samples were cleaned-up with 15 mg EMR-lipid and 20 mg MgSO 4 . This was achieved for 94% of GC-amenable compounds and 86% of LC-amenable compounds. In the case of RSC, the best results were seen when samples were cleaned-up with the cartridge containing only 20 mg Z-sep and 20 mg MgSO 4 . This was achieved for 88% of GC-amenable compounds and 90% of LC-amenable compounds. Although these cartridges yielded optimal results in terms of recovery, their use could require more instrument maintenance, especially for GC-MS/MS, due to the lower removal of co-extractives.

Abstract:Many plants have a high polyphenol content, which causes the matrix effect and makes the analysis of trace pesticide residues highly challenging. A common approach to improving matrix effects is to purify pesticides through the use of sorbents, but this requires a combination of multiple sorbents and extensive use. Zeolitic imidazolate framework-8 is widely used for pesticide analysis due to its high porosity, large specific surface area, and versatility. Here, we established and validated a modified quick, easy, cheap, effective, rugged, and safe method based on a zeolitic imidazolate framework-8 that was used to test the removal ability for polyphenols. To this end, 145 pesticide residues in peppermint, perilla, fennel, and mulberry leaves were analyzed by the modified method coupled with liquid chromatography with tandem mass spectrometry. The mean recoveries of all pesticide residues were in the range of 74.3-103.7%, with mean relative standard deviations ≤9.1% at spiked concentrations of 1, 10, 50, and 100 μg/kg for mulberry leaves. The limits of quantitation of the method ranged from 1 to 50 μg/kg. This study offers a reliable approach for the accurate quantitative analysis of various trace substances in the polyphenol-rich plants.

Abstract:Pesticides, widely applied in agriculture, can produce a variety of transformation products and their continuous use causes deleterious effects to ecosystem. Efficient and sensitive analytical techniques for enrichment and analysis of pesticides samples are highly required. Compared with other extraction methods, solid-phase micro extraction is a solvent free, cost effective, robust, versatile, and high throughput sample preparation technique, especially for the analysis of pesticides from complicated matrices. Coupling of solid-phase micro extraction with gas chromatography and mass spectrometry and liquid chromatography-mass spectrometry has been extensively applied in pesticide analysis. [Coronary angioplasty with the Monorail system via 6 French diagnostic catheters].

Abstract:We studied the use of "Monorail" system with Express (Scimed) balloon catheters for coronary angioplasty through 6 French (F) "high-flow" diagnostic catheters (Novoste, USCI). Prospectively, from July 1992 to January 1993, angioplasty of 31 lesions in 24 patients was attempted (1.3 lesions/procedure). Twenty procedures were of a single lesion and four were multi-vessel angioplasty. Fourteen lesions were in the left anterior descending or in its branches, 10 in the left circumflex or in its branches, 6 in the right coronary artery, and one in the distal anastomosis of an internal mammary artery graft. Thirteen lesions (42%) were type A, 17 (55%) type B and one (3%) type C. Balloon sizes varied between 2.0 and 3.5 mm. Twenty-nine lesions could be successfully dilated (93.5%); two cases were unsuccessful due to an acute occlusion in one and residual stenosis of more than 50% in the other. For only one case, another balloon catheter different from the "Monorail" system was necessary to complete a multi-vessel angioplasty. Coronary visualization and manipulation of the balloon through the tip of the diagnostic catheter were satisfactory in all cases, except with the 3.5 mm balloon catheter. Coronary angioplasty with "Monorail" system balloon catheters through 6 F "high-flow" diagnostic catheters is feasible and provides a high success rate in simple and moderately complex selected lesions, including multivessel angioplasty with advantages of smaller artery punction and the feasibility of performing coronary angioplasty with the same catheter used for diagnostic angiography.

Abstract:Many conventional sample preparation methods are tedious, time-consuming, and use high volume of organic solvent. Minimized sample preparation methods, which use significantly smaller volume of organic solvent, represent an alternative to these methods. Three liquid phase microextraction (LPME) techniques: single drop microextraction, dispersive liquid-liquid microextraction, and hollow fiber liquid phase microextraction and a variety of its modifications are reviewed. Advantages and limitations are discussed. Extraction parameters influencing the extraction efficiency are evaluated. Applicability of LPME methods for pesticide residues analysis in various food matrices is overviewed. Determination of low-level pesticide residues was realized predominantly by chromatographic methods utilizing selective detectors. Powerful features of mass spectrometric detection for identification and determination of pesticide residues are pointed out.

Abstract:Phenolic resin based activated carbon fibers (ACFs) were applied for the first time as a reversed-dispersive solid-phase extraction (r-DSPE) sorbent. A modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) method was applied to determine 26 pesticides (organophosphates, organochlorines, synthetic pyrethroids, and herbicides) in different complex matrices, including cauliflower, cucumber, banana, apple, wheat, and black gram. Different physicochemical characterization techniques were used to investigate the engineering and structural properties of the r-DSPE sorbent. All the chromatographic analyses were performed with a gas chromatograph equipped with an electron capture detector. The recoveries of all 26 pesticides were acceptable (70–120%), with relative standard deviations of less than 15%. The limit of detection and the limit of quantification were 1.13–5.48 ng/g and 3.42–16.60 ng/g, respectively. In the original QuEChERS method, primary secondary amine is extensively used as the r-DSPE sorbent in the cleanup process, but it is eightfold more expensive than the ACFs used in this study. Therefore, the modified QuEChERS method using ACFs during the cleanup process is more efficient, cheaper, and more robust to determine pesticides from different types of matrices, including vegetables, grains, and fruits, and ACFs could be used as a cost-effective alternative to primary secondary amine.Graphical AbstractSample clean-up using PSA and ACF as r-DSPE sorbent in QuEChERS method

Abstract:Alcohol-use disorders (AUD’s) commonly occur in people with other severe mental illnesses, such as schizophrenia or bipolar disorder, and can exacerbate their psychiatric, medical, and family problems. Therefore, to improve detection of alcohol-related problems, establish correct AUD diagnoses, and develop appropriate treatment plans, it is important to thoroughly assess severely mentally ill patients for alcohol and other drug abuse. Several recent studies have indicated that integrated treatment approaches that combine AUD and mental health interventions in comprehensive, long-term, and stagewise programs may be most effective for these clients.
