Abstract:SUMMARY Nutraceuticals, including amino acids, carbohydrates, minerals and vitamins, have gained more attention, owing to multiple nutritional and health benefits, including maintaining the normal physiological status and disease prevention, thereby improving production performance of poultry birds. In commercial poultry production systems, chicks do not have access to feed and water for about 24–48 h during transportation between hatchery and production farm. This delay time poses negative effects on early feed intake, organ development, natural exposure to microbiota and immune sensitisation of chicks. Early feeding via in ovo inoculation of nutrients and feed additives to the developing embryo during 12th or 17th to 18th days of incubation significantly mitigates the adverse effects of starvation and enhances post-hatch performance and health of poultry birds during pre-andpost-hatch periods. The direct administration of various supplements, nutrients, drugs, vaccines, immunostimulants and biological substances through an in ovo route has diverse advantages including boosted health status, immunity and production performance of post-hatch poultry birds. Early feeding of nutrients during embryonic life has numerous benefits, such as improvements in weight gain, feed efficiency, growth rate and digestive organ development; decrease in morbidity and mortality of embryo; reduction in skeletal disorder; and boosted immunity, muscular development and meat yield of poultry birds. Furthermore, early in ovo feeding of nutraceuticals has provided better protection against a number of infectious pathogens than post-hatch supplementation. The current review aimed to provide a promising insight for the promotion of in ovo technology in order to supply different nutraceuticals (amino acid, vitamins, minerals and carbohydrates) directly into the developing embryo to achieve an improvement in post-hatch growth, immune responses and antioxidant defence along with the development of gastrointestinal tract. Additionally, this review highlights the future of in ovo technology as a strategy to improve the productive performance of poultry birds.

Abstract:The poultry industry has improved genetics, nutrition, and management practices, resulting in fast-growing chickens; however, disturbances during embryonic development may affect the entire production cycle and cause irreversible losses to broiler chicken producers. The most crucial time in the chicks' development appears to be the perinatal period, which encompasses the last few days of pre-hatch and the first few days of post-hatch. During this critical period, intestinal development occurs rapidly, and the chicks undergo a metabolic and physiological shift from the utilization of egg nutrients to exogenous feed. However, the nutrient reserve of the egg yolk may not be enough to sustain the late stage of embryonic development and provide energy for the hatching process. In addition, modern hatchery practices cause a delay in access to feed immediately post-hatch, and this can potentially affect the intestinal microbiome, health, development, and growth of the chickens. Development of the in ovo technology allowing for the delivery of bioactive substances into chicken embryos during their development represents a way to accommodate the perinatal period, late embryo development, and post-hatch growth. Many bioactive substances have been delivered through the in ovo technology, including carbohydrates, amino acids, hormones, prebiotics, probiotics and synbiotics, antibodies, immunostimulants, minerals, and microorganisms with a variety of physiological effects. In this review, we focused on the physiological effects of the in ovo delivery of these substances, including their effects on embryo development, gastrointestinal tract function and health, nutrient digestion, immune system development and function, bone development, overall growth performance, muscle development and meat quality, gastrointestinal tract microbiota development, heat stress response, pathogens exclusion, and birds metabolism, as well as transcriptome and proteome. We believe that this method is widely underestimated and underused by the poultry industry.

Abstract:SUMMARYThe key objective of poultry husbandry is to provide safe and nutritious food for the growing population. The technique of delivering various nutrients, nutraceuticals, vaccines, immunostimulants, phytochemicals and medicinal herbs via the in ovo route is gaining wide attention among scientists worldwide for improving poultry productivity, boosting immune response and safeguarding the health. Embryonic growth in poultry has been manipulated through direct administration of biological compounds into embryonated eggs during the mid and late phase of incubation. This shows potential to enhance hatchability, superior nutritional status, higher chick quality, improve post-hatch performance and ultimately reduce the challenges associated with infections and oxidative stress, as well as minimize the use of synthetic antibiotic growth promoters in poultry production. In the last two decades, several studies have demonstrated that the in ovo supplementation of herbs and their derivatives, such as chicoric acid, rosmarinic acid, astragalus polysaccharides boost immunity, upregulates antioxidant defense, and improves the health status and production output of poultry birds, making herbal compounds an alternative to antibiotic growth promoters. In ovo injection of herbs and their phytochemicals has shown diverse advantages, including improvements in weight gain, feed efficiency, growth rate, decreased morbidity and mortality of embryos, boosted immunity, and improved health status of poultry birds. Furthermore, in ovo injection of herbal compounds has shown better protection against a number of infectious pathogens than post-hatch supplementation. The current review aimed to provide a promising insight for the promotion of in ovo technology in order to supply natural biological compounds/phytochemicals directly into the developing embryo to achieve an improvement in post-hatch growth, immune responses, antioxidant defense and highlights the future of in ovo technology as a strategy to minimize the use of AGPs and improve the health status and production performance of poultry.

Abstract:This study was conducted to evaluate the effect of in ovo feeding of cationic amino acids on hatchability, hatch weights, and organ developments in pigeon squabs. Two experiments were conducted in this study. Eggs in Exp. 1 were subjected to modification of in ovo feeding in pigeons. Optimal time was determined by checking amniotic fluid volume, and suitable length was confirmed through ink injection. Results showed that the optimum time of in ovo feeding was on d 13 of embryonic development, and the suitable injected length was 20 mm to reach the amniotic cavity of the embryo. Eggs in Exp. 2 were transferred to access in ovo feeding of cationic amino acids. A total of 75 fertile pigeon eggs was randomly distributed into 5 treatments of 15 replicate eggs. Treatments in Exp. 2 consisted of non-injected controls (Control), a sterile buffered solution (0.75% saline), or a cationic amino acid mixture (> 98.5% purity crystalline L-arginine, > 98% purity crystalline L-lysine, and > 98.5% purity L-histidine) containing 0.1, 1, or 10% concentration (Conc.), which were relative to their total content in the eggs, respectively. The crystalline amino acids were dissolved in 200 μL buffered solution prior to in ovo feeding. After hatching, hatch weight (HW) and organ weight (OW) of the squabs were measured immediately. In ovo feeding of cationic amino acids increased the proportions of yolk-free hatch weight to hatch weight (YFHW/HW) (quadratic P = 0.01), and those of OW to YFHW including the heart (quadratic P = 0.01), kidney (quadratic P < 0.01), and liver (quadratic P = 0.02) compared to the control group, and the levels of those ratios were maximized in the 1% Conc group. Also, a proportion of small intestine weight to YFHW improved (linear P = 0.02, quadratic P = 0.05) after in ovo feeding. The organ weight of the head, leg, heart, lung, kidney, proventriculus, pancreas, liver, and small intestine correlated with YFHW positively (0.4 < correlation coefficient < 0.8, P < 0.05). In conclusion, cationic amino acids injection into amnion can improve the embryonic development, which may be mediated by the increment of relative organ weight.

Abstract:Pathogens, such as Escherichia coli (E. coli), have been identified as significant causes of poultry mortality. Poultry can serve as potential sources of E. coli transmission, even when asymptomatic, posing a substantial threat to food safety and human health. The in ovo administration of antimicrobials is crucial for preventing and/or effectively combating acute and chronic infections caused by poultry pathogens. To achieve this goal, it is critical that antimicrobials are properly injected into embryonic fluids, such as the amnion, to reach target tissues and trigger robust antimicrobial responses. Several protocols based on antimicrobials were evaluated to meet these requirements. This review analyzed the impacts of antimicrobial substances injected in ovo on the control of E. coli in poultry. The reduction in infection rates, resulting from the implementation of in ovo antimicrobials, combined with efforts aimed at hygienic-sanitary action plans in poultry sheds, reinforces confidence that E. coli can be contained before causing large scale damage. For example, antimicrobial peptides and probiotics have shown potential to provide protection to poultry against infections caused by E. coli. Issues related to the toxicity and bacterial resistance of many synthetic chemical compounds represent challenges that need to be overcome before the commercial application of in ovo injection protocols focused on microbiological control.

Abstract:Healthy gastrointestinal tract (GIT) is crucial for optimum performance, better feed efficiency, and overall health of poultry. In the past, antibiotic growth promoters (AGP) were commonly used to modulate the gut health of animals. However, considering the public health concern, the use of AGP in animal feeding is banned or regulated in several jurisdictions around the world. This necessitates the need for alternative nutritional strategies to produce healthy poultry. For that, several alternatives to AGP have been attempted with some success. However, effective modulation of the gut health parameters depends on the methods and timing of the compound being available to host animals. Routinely, the alternatives to AGP and other nutrients are provided in feed or water to poultry. However, the GIT of the newly hatched poultry is functionally immature, despite going through significant morphological, cellular, and molecular changes toward the end of incubation. Thus, early growth and development of GIT are of critical importance to enhance nutrients utilization and optimize the growth of poultry. Early nutrition programming using both in ovo and post-hatch feeding has been used as a means to modulate the early growth and development of GIT and found to be an effective strategy but with inconsistent results. This review summarizes the information on in ovo and post-hatch-feeding of different nutrients and feeds additives and their effects on gut development, histomorphology, microbiology, and immunology. Furthermore, this review will provide insight on the future of early nutrition programming as a strategy to enhance gut health, thereby improving overall health and production so that the poultry industry can benefit from this technique.

Abstract:Glucose is the major energy source of living organisms and its homeostatic maintenance during the days preceding hatching and post-hatch is a great challenge in chick's life. Establishment of a stable and sufficient glucose status is critical for the late-term embryonic developmental hatching process and post-hatch development of poultry until feed consumption is initiated. In ovo feeding has been used to enhance post-hatch development in broilers. However, there is paucity of information on the effect of in ovo feeding on performance of turkey poults. Therefore, effect of in ovo feeding on performance of turkey poults were investigated from different research works. The hatching quality in turkey has been of great concern and interest to researchers over the years. Turkey hatchlings usually do not survive critical post-hatch adjustment period and most of the survivor exhibit stunted growth, and inefficient feed utilization. This has led to loss of a lot of money yearly to the poultry industry. Several researches have been done but were seen to have no lasting effect on the post-hatch quality of the poults. Although, early feeding was recorded to be of benefit to the animals but had little orno effect on struggles experienced by embryos during pipping and hatching. Improvement on poults' quality aside the body weight and growth becomes difficult at this level. This work reviewed the technology of using in ovo feeding which is the administration of food into the amnion of late embryo in the efficiency of turkey hatchlings. Le glucose est la principale source d'énergie des organismes vivants et son maintien homéostatique pendant les jours précédant l'éclosion et après l'éclosion est un grand défi dans la vie du poussin. L'établissement d'un état de glucose stable et suffisant est essentiel pour le processus d'éclosion du développement embryonnaire tardif et le développement post-éclosion de la volaille jusqu'au début de la consommation d'aliments. L'alimentation Inovo a été utilisée pour améliorer le développement post-éclosion des poulets à griller. Cependant, il y a peu d'informations sur l'effet de l'alimentation in ovo sur les performances des dindonneaux. Par conséquent, l'effet de l'alimentation in ovo sur les performances des dindonneaux a été étudié à partir de différents travaux de recherche. Les nouveau-nés de dinde ne survivent généralement pas à la période critique d'ajustement post-éclosion et la plupart des survivants présentent un retard de croissance et une utilisation inefficace des aliments. Cela a conduit à la perte de beaucoup d'argent chaque année pour l'industrie de la volaille. Plusieurs recherches ont été effectuées, mais elles n'ont eu aucun effet durable sur la qualité post-éclosion des dindonneaux. Bien que l'alimentation précoce ait été enregistrée comme étant bénéfique pour les animaux, mais n'a eu que peu ou pas d'effet sur les difficultés rencontrées par les embryons lors de la naissance et de l'éclosion. L'amélioration de la qualité des dindonneaux en dehors du poids vif et de la croissance devient difficile à ce niveau. Ce travail a passé en revue la technologie d'utilisation de l'alimentation in ovo qui est l'administration de nourriture dans l'amnios de l'embryon tardif dans l'efficacité des nouveau-nés de dinde.

Abstract:In ovo feeding refers to a method of delivering nutrients or supplements directly to the developing embryo inside the egg before it hatches. This method is commonly used in the poultry industry to improve the health and performance of the chicks after they hatch. One specific application of in ovo feeding is the use of probiotics. Probiotics are beneficial bacteria that can help improve gut health and overall immune function. By delivering probiotics directly to the developing embryo in ovo feeding can help establish a healthy gut microbiota early on leading to improved health and performance in the hatchlings. Probiotics can compete with harmful bacteria in the gut preventing them from colonizing and causing infections. This can reduce the need for antibiotics as the chicks are better equipped to fight off infections naturally. This can lead to better growth rates and feed conversion efficiency in the chicks. In ovo feeding probiotics is a relatively new and evolving technique in the poultry industry. Research is ongoing to optimize the delivery methods doses and types of probiotics used. Overall, in ovo feeding probiotics is a promising approach to improve the health and performance of poultry. By supporting the gut microbiota early on in the development of the chicks the benefits can be seen throughout their lifespan. Continued research and advancements in in ovo feeding techniques will likely lead to further improvements in the future.
The Effects of In Ovo Injection of Synbiotics on the Early Growth Performance and Intestinal Health of Chicks

Abstract:In this study, the effects of synbiotic inclusion at the intra-amniotic stage in layer chicks were evaluated with different parameters, such as performance, immunological function, intestinal development, and cecal microflora content. A total of 1,200 eggs with fertile embryos were allocated into four treatment groups. For every treatment, five replicates were used, and 60 eggs were included in each replicate. The following four treatment groups were established: the non-injected group, 0.9% physiological saline injection (saline) group, 1 × 10 6 CFU/egg Lactobacillus plantarum injection (probiotic) group, and 1 × 10 6 CFU/egg L. plantarum + 2 mg/egg Astragalus polysaccharide injection (synbiotic) group. In ovo injection was carried out at 18.5 days of incubation. The results showed that in ovo injection of probiotics or synbiotics did not affect the hatching or growth performance of the chicks but significantly increased their feed intake (FI), body weight (BW), and the feed conversion ratio (FCR). Additionally, in ovo injection of synbiotics enhanced the levels of serum interleukin-2 (IL-2), interferon-γ (IFN-γ), and secretory immunoglobulin A (SIgA) in intestinal lavage fluid and the histomorphological development of the small intestine. Our results also indicated that intra-amniotic synbiotic injection significantly increased Lactobacillus and Bifidobacterium colonization while decreasing the relative abundance of Escherichia coli in the chicken cecum ( P < 0.05). In summary, in ovo injection of synbiotics had positive impacts on the performance, immunological function, gut development, and microbiota of growing chicks.

In ovo feeding of probiotic lactobacilli differentially alters expression of genes involved in the development and immunological maturation of bursa of Fabricius in pre-hatched chicks

Abstract:Compelling evidence indicates that immunological maturation of the gut-associated lymphoid tissues, including the bursa of Fabricius, is dependent upon antigenic stimulation post-hatch. In view of these data, the present study investigated the impact of exposing the immune system of chick embryos to antigenic stimuli, via in ovo delivery of poultry-specific lactobacilli, on the expression of genes associated with early bursal development and maturation. Broiler line embryonated eggs were inoculated with 106 and 107 colony-forming units (CFUs) of an individual or a mixture of Lactobacillus species, including L. crispatus (C25), L. animalis (P38), L. acidophilus (P42), and L. reuteri (P43), at embryonic day 18 (ED18). The bursa of Fabricius was collected from pre-hatched chicks (ED20) to measure the expression levels of various immune system genes. The results revealed that L. acidophilus and the mixture of Lactobacillus species at the dose of 106 CFU consistently elicited higher expression of genes responsible for B cell development, differentiation, and survival (B cell activating factor (BAFF), BAFF-receptor (BAFF-R)), and antibody production (interleukin (IL)-10) and diversification (TGF-β). Similar expression patterns were also noted in T helper (Th) cell-associated cytokine genes, including Th1-type cytokines (interferon (IFN)-γ and IL-12p40), Th2-type cytokines (IL-4 and IL-13) and Th17 cytokine (IL-17). Overall, these results suggest that the supplementation of poultry-specific lactobacilli to chick embryos might be beneficial for accelerating the development and immunological maturation of the bursa of Fabricius. However, further studies are required to determine if the changes in gene expression are associated with the developmental trajectory and phenotypes of bursal cells.
In-ovo injection of Bacillus subtilis, raffinose, and their combinations enhances hatchability, gut health, nutrient transport- and intestinal function-related genes, and early development of broiler chicks

Abstract:An experiment was conducted to assess the response of chicks to in-ovo injection of Bacillus subtilis (probiotic), raffinose (prebiotic), and their combinations. The study used 1,500 embryonated eggs allotted to 10 groups/ 6 replicates (150 eggs/group). The experimental treatments were: 1) un-injected control (NC); 2) sham (sterile distilled water) (PC); 3) probiotic 4 × 105CFU/egg (LBS); 4) probiotic 4 × 106CFU/egg (HBS); 5) prebiotic 2 mg/egg (LR); (6 prebiotic 3 mg/egg (HR); 7) probiotic 4 × 105CFU + prebiotic 2 mg/egg (LBS+LR); 8) probiotic 4 × 105CFU + prebiotic 3 mg/egg (LBS+HR); 9) probiotic 4 × 106CFU + prebiotic 2 mg/egg (HBS+LR); and 10) probiotic 4 × 106CFU + prebiotic 3 mg/egg (HBS+HR). Results showed that in-ovo inclusion of Bacillus subtilis, prebiotic, and their combinations improved hatchability, yolk-free chick weight, and chick weight compared to the control group. Moreover, the in-ovo treatment reduced residual yolk weight on the day of hatch compared to the control group. Different levels of in-ovo B. subtilis alone or combined with raffinose significantly (P ≤ 0.001) reduced total bacterial count and total yeast and mold count compared to the negative control group. Total coliform and E. coli decreased significantly (P ≤ 0.001) in groups treated with probiotics, prebiotics, and synbiotics with different doses during incubation compared to those in the control. Clostridium spp. was not detected in the groups injected with B. subtilis alone or combined with raffinose. In-ovo probiotics and synbiotics (LBS+LR & LBS+HR) significantly (P ≤ 0.001) increased ileal villus length compared to other groups. In-ovo treatment increased mRNA expression of JAM-2 compared to the control group. The fold change significantly increased in group LBS+HR for genes MUC-2, OCLN, VEGF, SGLT-1, and EAAT-3 compared to the negative control. In conclusion, in-ovo injection of a low dose of B. subtilis plus a high or low dose of raffinose can positively affect hatching traits, cecal microbial populations, intestinal histomorphometry, nutrient transport- and intestinal function-related genes, and chick quality of newly hatched broiler chicks.
Effects of stage of broiler embryo development on coccidiosis vaccine injection accuracy, and subsequent oocyst localization and hatchling quality

Abstract:Control of coccidiosis in broiler chickens continues to pose challenges to commercial poultry producers, especially in an era of increased consumer demand for antibiotic-free broiler production. As a result, coccidiosis vaccines are now commonly used in rotation programs to achieve effective coccidiosis control. Inovocox EM1 vaccine (EM1) is a coccidiosis vaccine that allows for earlier immune acquisition through oocyst cycling, which reduces the effects of wild-type coccidia. The EM1 vaccine is administered to embryonated broiler hatching eggs between 18 and 19 D of incubation (doi). In the U.S., commercial broiler hatcheries vaccinate embryonated eggs at either 18.5 or 19 doi. However, it is unclear whether a difference in embryo age at the time of in ovo injection can impact the actual site of vaccine delivery. In addition, it is unclear where oocysts eventually become localized within the embryo following the in ovo injection of EM1. Therefore, the objective of this study was to determine the effects of stage of embryonic development on the actual deposition site of the EM1 vaccine oocysts when they are in ovo injected and to subsequently investigate the movement and eventual location of EM1 oocysts after in ovo injection. Because all eggs were injected at the same time, a 12-h difference in set time was a means to derive 18.5 and 19.0 incubation age of injection (IAN) treatments. The experimental design was a 3 injection treatment (noninjected, diluent-injected, and vaccine-injected) × 2 IAN factorial. There was a significant main effect of IAN on site of vaccine oocysts delivery, and subsequent hatching chick quality. Qualitative histological evaluation revealed the oral uptake of vaccine oocysts through the amnion, with their subsequent presence in the gizzard and intestinal lumen by 24 to 36 h postinjection. In conclusion, physiological development influenced the site of injection, and oocysts imbibed along with the amniotic fluid in late stage broiler embryos are subsequently transported to the gastrointestinal tract.
Effects of in Ovo Injection of Carbohydrate Solution on Small Intestine Development in Domestic Pigeons (columba Livia)

Abstract:The objective of this study was to test the hypothesis that in ovo injection of carbohydrates into pigeon (Columba livia) amnion may improve the small intestine development. At d 14.5 of incubation, 80 fertile eggs were injected with 200 μL of carbohydrate solution, and 80 control eggs were not injected. The carbohydrate solution (wt/vol) contained 2.5% maltose + 2.5% sucrose, all dissolved in 0.75% saline. Twelve eggs from each treatment were randomly sampled at d 16 of incubation and the day of hatch, embryos or young pigeons were euthanized, and the jejunum samples were collected. Jejunal villus surface area, activity of the brush border enzymes, sucrase, maltase, aminopeptidase-N, and alkaline phosphatase, and mRNA expression of the digestion-absorption related genes oligopeptide transporter 1, sodium glucose transporter 1, glucose transporter 2, aminopeptidase-N, and sucrase-isomaltase were examined. Results showed that in ovo injection of carbohydrate solution caused a villus surface area increase of 38% on d 16 of incubation and 23% on day of hatch relative to controls (P < 0.05). The in ovo injected pigeons exhibited greater (P < 0.05) activities of jejunal sucrase, maltase, and alkaline phosphatase from d 16 of incubation to day of hatch compared with the controls. At day of hatch, aminopeptidase-N activity in embryos injected in ovo was approximately 27% greater (P < 0.05) than control embryos. Enhanced expressions of the jejunal sodium glucose transporter 1, glucose transporter 2, and aminopeptidase-N mRNA were found at d 16 of incubation in embryos that received carbohydrate solution into the amniotic fluid in comparison with the control group (P < 0.05). These results indicate that the in ovo injected pigeon may hatch with more mature enterocytes and greater intestinal digestive and absorptive capacity than the conventional hatchling. Therefore, the in ovo injected pigeons may become more precocial at hatch and easier to hand-rear during the immediate posthatch period.

Abstract:Egg yolk constitutes a relevant alternative source of antibodies. It presents some advantages over mammalian serum immunoglobulins regarding productivity, animal welfare and specificity. The main immunoglobulin present in avian blood (IgY) is transmitted to their offspring and accumulates in egg yolks, which enables the non-invasive harvesting of high amounts of antibodies. Moreover, due to structural differences and phylogenetic distance, IgY is more suitable for diagnostic purposes than mammalian antibodies, since it does not react with certain components of the human immune system and displays greater avidity for mammalian conserved proteins. IgY has been extensively used in health researches, as both therapeutic and diagnostic tool. This article aims to review its applications in both human and veterinary health.

Abstract:As a classical model system of embryo biology, the chicken embryo has been used to investigate embryonic development and differentiation. Delivering exogenous materials into chicken embryos has a great advantage for studying gene function, transgenic breeding, and chimera preparation during embryonic development. Here we show the method of in ovo intravascular injection whereby exogenous materials such as plasmid vectors or modified primordial germ cells (PGCs) can be transferred into donor chicken embryos at early developmental stages. The results show that the intravascular injection through the dorsal aorta and head allows injected materials to diffuse into the whole embryo through the blood circulatory system. In the presented protocol, the efficacy of exogenous plasmid and lentiviral vector introduction, and the colonization of injected exogenous PGCs in the recipient gonad, were determined by observing fluorescence in the embryos. This article describes detailed procedures of this method, thereby providing an excellent approach to studying gene function, embryo and developmental biology, and gonad-chimeric chicken production. In conclusion, this article will allow researchers to perform in ovo intravascular injection of exogenous materials into chicken embryos with great success and reproducibility.

Abstract:Abstract In biological research, chicken embryos are a classic experimental model for the exploration of the embryonic development and cell differentiation. Transferring exogenous substances into chicken embryos for producing medical antibodies has been widely used in the production practice. However, there are few studies about the effect of the different injection site and dosage on chicken embryos. The aim of this study was to explore the effects of different injection sites and dosages on chicken embryo hatching rate and development, so as to provide a basis for further studies using the chicken embryo model. Freshly laid eggs (Rugao yellow chicken) were injected with different doses of saline at the tip, equatorial plane and the blunt end of the egg shell, respectively. Egg hatching rate was recorded and compared among injection sites and different doses. A trypan blue stain was also injected at the aforementioned sites and the growth of chicken embryos was observed. The SPSS (statistical package for the social science) software was used to analyze the relationship between the chicken eggs hatching rate and the different injection sites or the different dosages. Abstract:During the first week post-hatch, the neonatal chick is immunologically vulnerable and subject to infectious threats found in the environment. Probiotics are live, non-pathogenic microorganisms known to have a positive effect on the host by improving the natural balance of gut microbiota and promoting animal health. The objective of this study was to determine the effects of administering probiotics in ovo and in the diet on broiler chick hatchability, and post-hatch immune organ weights and ileal immune-related gene expression. At embryonic day 18, 1584 eggs were injected with nothing (Dry), 1 × 106, or 1 × 107 (P1 and P2 respectively) probiotic bacteria. The remaining 393 eggs were left non-injected to serve as a negative control. Immune organ weights and tissue samples were taken on DOH and d4, 6, 8, 14, and 20. No differences were observed for hatchability or relative bursa weights. Only on d6, the P2 birds receiving the probiotic-supplemented diet had larger spleens as a result of a 2-way interaction between in ovo treatment and post-hatch diet. Both in ovo and dietary administration of probiotics were able to modulate the expression of the immune-related genes in the ileum; however, expression patterns differed based on the gene, treatment, and time point evaluated. In conclusion, these results indicate that in ovo supplementation of this commercial probiotic product does not influence hatchability and is capable of differentially modulating expression of certain genes in the ileum. Furthermore, in ovo administration of probiotics has an effect similar to that of dietary supplementation endorsing its usage to potentially promote early colonization of beneficial bacteria to stimulate intestinal and immune system development.

Abstract:The effects of in ovo feeding of carbohydrates on hatchability, BW, yolk sac weights (YSW), pectoral muscle weights (PMW), liver and pectoral muscle glycogen concentration, serum glucose level, and hepatic glucose-6-phosphatase activity of domestic pigeons, hatched from eggs laid by a 40-wk-old breeder flock, were investigated. At 14.5 of incubation, fertile eggs were injected with 200 μL of 1.5% maltose (M) + 1.5% sucrose (S), 2.5% M + 2.5% S, 3.5% M + 3.5% S, or 4.5% M + 4.5% S in 0.75% saline, with controls not injected. Results showed that in ovo injection with 1.5% M + 1.5% S or 2.5% M + 2.5% S increased the hatchability compared with the control, whereas injection of 4.5% M + 4.5% S decreased the hatchability. The BW at hatch was quadratic, and BW was maximized by injecting 2.5% M + 2.5% S. The YSW at hatch decreased linearly by the injection with 3.5% M + 3.5% S compared with the control group. In ovo injection of 2.5% M + 2.5% S increased the PMW at hatch. There were no significant differences between any of the treatment groups for liver glycogen reserves. Serum glucose level at hatch was quadratic, and the glucose level was maximized between supplemental 2.5% M + 2.5% S and supplemental 3.5% M + 3.5% S. The pectoral muscle glycogen reserves increased quadratically as supplemental carbohydrates increased, and the response was maximized by injecting 2.5% M + 2.5% S. In conclusion, the present results demonstrate that the injected carbohydrates are available for use and storage. In ovo feeding of carbohydrates, especially at the level of 2.5% M + 2.5% S, on 14.5 d of incubation can improve the hatchability, BW, and PMW by elevating the pectoral muscle glycogen reserves in domestic pigeons at hatch. Results also suggested that in ovo injection of carbohydrates could increase the yolk sac nutrient utilization and hence might enhance the pigeon enteric development.

Abstract:Outbreaks of avian pathogens such as Newcastle disease virus, avian influenza virus, and salmonella have a major impact on economies and food security worldwide. Some pathogens also pose a significant zoonotic potential, especially avian influenza viruses. Vaccination plays a key role in controlling many poultry diseases, and there are many vaccines licenced in the United Kingdom for diseases of poultry caused by viruses, bacteria, and parasites. However, these vaccines often do not provide complete protection and can cause unwanted side effects. Several factors affect the potency of poultry vaccines, including the type of vaccination used, the mechanism of delivery, and the use of adjuvants. Advancements in technology have led to the study and development of novel vaccines and vaccine adjuvants for use in poultry. These induce stronger immune responses compared with current vaccine technology and have the potential to protect against multiple poultry diseases. This review aims to discuss the existing poultry vaccine technology; the effect of delivery mechanisms on vaccine efficacy; the use of current and novel adjuvants; the ability to target antigens to antigen-presenting cells; and the use of probiotics, multivalent vaccines, and nanotechnology to enhance the potency of poultry vaccines.
