Abstract:Objective To explore the effects and mechanisms of Kang Fuyan capsule for the treatment of inflammation through observing the activity of polymorphonuclear neutrophils (PMN) infected by Chlamydia trachomatis (CT). Methods Rat peripheral blood PMN was separated and cultured. After infecting CT for 6 h, PMN was treated with Kang Fuyan capsule [10 ng/mL (K-10 ng group), 10 μg/mL (K-10 μg group), 1 mg/mL (K-1 mg group)] for 6 h, 12 h and 24 h. Cells and cell supernatant were collected to analyze reactive oxygen species (ROS), myeloperoxidase (MPO) and CT loads. Molecular mechanisms of Kang Fuyan capsule affecting the activity of PMN in inflammatory response caused by CT were researched by TLR2/4 agonist or inhibitors. Results PMN intracellular ROS level was increased significantly (P<0.001) after CT infecting PMN. Kang Fuyan capsule significantly inhibited the production of ROS (K-1 mg: 6 h P=0.000 03, 12 h P=0.000 001, 24 h P=0.000 003; K-10 μg: 6 h P=0.014, 12 h P=0.009, 24 h P=0.009; K-10 ng: 24 h P=0.000 5) and MPO (K-1 mg: 6 h P=0.046, 12 h P=0.004, 24 h P=0.038; K-10 μg: 12 h P=0.048, 24 h P=0.011; K-10 ng: 12 h P=0.005, 24 h P=0.017) and inhibited the CT load of cell culture medium. Compared with Kang Fuyan capsule group, TLR4 inhibitors and Kang Fuyan capsule acting on cells at the same time significantly inhibit the secretion of MPO (P=0.011); TLR2/4 agonist and Kang Fuyan capsule acting on cells at the same time significantly decreased the CT load in cell supernatant (PGN: P=0.002; LPS: P=0.000 08). Conclusion Kang Fuyan capsule regulates the sterilization ways of PMN by suppressing the production and secretion of ROS and MPO, and promoting PMN phagocytosis. But the molecular mechanisms of Kang Fuyan capsule to PMN had nothing to do with TLR2/4 and still need to be studied. Bioactivity of pyocyanin of Pseudomonas aeruginosa clinical isolates against a variety of human pathogenic bacteria and fungi species

Abstract:Pyocyanin is blue pigment redox active, secondary metabolites produced by P. aeruginosa. The present study investigated the bioactivity of pyocyanin against certain types of bacteria and fungi causing human infections Objectives: Pyocyanin is blue pigment redox active, secondary metabolites produced by P. aeruginosa. The current study deals with biosynthesis, purification and bioactivity of pyocyanin produced by P. aeruginosa. Design: Pyocyanin extraction was done by chloroform method and concentration was determined by multiplying the optical density at 520 nm by 17.072 expressed as μg/ml. Biological activity of pyocyanin was determined by well diffusion procedure. Results: According to the source of infection, results showed that P. aeruginosa were most common in ear infection (30%) followed by wounds (22%), burns (17%), urine (13%) and each in stool and diabetes (9%). In this study the high resistance of P. aeruginosa isolates to antibiotics were 19 (82.6 %) to piperacillin followed by 10(43.5%) to aztreonem, 8(34.8%) to meropenem, 6(26.1%) to amikacin, 5(21.7%) to ciprofloxacin then 2(8.7%) to cefotaxime. the urine isolate produced the largest amount of pyocyanin (15.894 μg/ml). pyaocyanin have antimicrobial activity against Pathogenic bacteria: Shigella, Staphyllococcus aureus and Staphyllococcus epidermedis. and pathogenic fungi and yeast: Aspergillus niger, Penicillium spp., Rhizopus spp, Trichophyton mentagrophyte, Rhodotorula spp., Alternaria alternate , Trichophyton rubrum and Candida spp Conclusions: cefotaxime is the best antibiotic for P. aeruginosa. Antimicrobial activity of pyocyanin against gram positive more than gram negative bacteria but less than that observed against fungi (molds and yeast).

Abstract:Pyocyanin is an extracellular diffusible pigment conferring virulence to Pseudomonas strains for their survival. Surge in antimicrobial resistance demands alternate potential candidates of biological origin. Combining the virulence and antagonistic potential, pyocyanin pigment shall be developed as a biocontrol agent with experimental studies. Media optimization and cross streak method were employed to screen the competent strain and identified using 16 S rRNA sequencing. Agar diffusion, broth dilution and MTT assay were adopted to determine the antimicrobial potential and biocompatibility of pyocyanin. Further, UV-vis spectrophotometry, GC-MS and 1 H NMR analyses were used to validate the structural identity of pyocyanin. Results revealed that Potato glycerol broth supported maximum pyocyanin production from competent Pseudomonas aeruginosa WS1. Significant antimicrobial activity in a dose dependent fashion was observed with MIC 64 – 32 μg / mL. Cytotoxicity with IC 50 @ 108.5 mg L -1 was found to be associated with clumping and cell rounding with consistent lactate dehydrogenase (LDH) assay result. The mass spectrum ( m/z ) at 210 and N–CH 3 proton resonating at 1.4229 ppm and 7.4 – 7.5 ppm confirmed the condensed nitrogen aromatic ring of pyocyanin. These findings suggest that the combined virulence and antagonistic potential of pyocyanin interfering with the ETC may serve as a promising bio-control agent.

Abstract:Current understanding of reverberation chambers is firmly funded upon their being operated as overmoded cavities. A proper definition of this condition has been lacking for a long time, while being regarded as automatically satisfied as soon as a large (and unspecified) number of normal modes are set to resonate at the frequency of operation of the chamber. In this paper we summarize the most important steps needed to introduce in a formal way a proper definition of the overmoded condition, showing how, from a strict mathematical viewpoint, it is not related to any threshold frequency, but rather requires a statistical framework.
The MraY Inhibitor Muraymycin D2 and Its Derivatives Induce Enlarged Cells in Obligate Intracellular Chlamydia and Wolbachia and Break the Persistence Phenotype in Chlamydia

Abstract:Chlamydial infections and diseases caused by filarial nematodes are global health concerns. However, treatment presents challenges due to treatment failures potentially caused by persisting Chlamydia and long regimens against filarial infections accompanied by low compliance. A new treatment strategy could be the targeting of the reduced peptidoglycan structures involved in cell division in the obligate intracellular bacteria Chlamydia and Wolbachia, the latter being obligate endosymbionts supporting filarial development, growth, and survival. Here, cell culture experiments with C. trachomatis and Wolbachia showed that the nucleoside antibiotics muraymycin and carbacaprazamycin interfere with bacterial cell division and induce enlarged, aberrant cells resembling the penicillin-induced persistence phenotype in Chlamydia. Enzymatic inhibition experiments with purified C. pneumoniae MraY revealed that muraymycin derivatives abolish the synthesis of the peptidoglycan precursor lipid I. Comparative in silico analyses of chlamydial and wolbachial MraY with the corresponding well-characterized enzyme in Aquifex aeolicus revealed a high degree of conservation, providing evidence for a similar mode of inhibition. Muraymycin D2 treatment eradicated persisting non-dividing C. trachomatis cells from an established penicillin-induced persistent infection. This finding indicates that nucleoside antibiotics may have additional properties that can break bacterial persistence.

Abstract:Introduction: Pseudomonas aeruginosa is notorious for its multidrug resistance and its involvement in hospital-acquired infections. In this study, 20 bacterial strains isolated from soil samples near the Hindan River in Ghaziabad, India, were investigated for their biochemical and morphological characteristics, with a focus on identifying strains with exceptional drug resistance and pyocyanin production. Methods: The isolated bacterial strains were subjected to biochemical and morphological analyses to characterize their properties, with a particular emphasis on exopolysaccharide production. Strain GZB16/CEES1, exhibiting remarkable drug resistance and pyocyanin production. Biochemical and molecular analyses, including sequencing of its 16S rRNA gene (accession number LN735036.1), plasmid-curing assays, and estimation of plasmid size, were conducted to elucidate its drug resistance mechanisms and further pyocynin based target the Candida albicans Strain GZB16/CEES1 demonstrated 100% resistance to various antibiotics used in the investigation, with plasmid-curing assays, suggesting plasmid-based resistance gene transmission. The plasmid in GZB16/CEES1 was estimated to be approximately 24 kb in size. The study focused on P. aeruginosa 's pyocyanin production, revealing its association with anticandidal activity. The minimum inhibitory concentration (MIC) of the bacterial extract against Candida albicans was 50 μg/ml, with a slightly lower pyocyanin-based MIC of 38.5 μg/ml. Scanning electron microscopy illustrated direct interactions between P. aeruginosa strains and Candida albicans cells, leading to the destruction of the latter. Discussion: These findings underscore the potential of P. aeruginosa in understanding microbial interactions and developing strategies to combat fungal infections. The study highlights the importance of investigating bacterial-fungal interactions and the role of pyocyanin in antimicrobial activity. Further research in this area could lead to the development of novel therapeutic approaches for combating multidrug-resistant infections.

Abstract:Pyocyanin is a bioactive pigment produced by Pseudomonas aeruginosa . It is an important virulence factor that plays a critical role in P. aeruginosa infections as a redox-active secondary metabolite and a quorum sensing (QS) signaling molecule. Pyocyanin production from chorismic acid requires the involvement of two homologous operons, phz1 and phz2 , which are activated by QS regulatory proteins. Pyocyanin inhibits the proliferation of bacterial, fungal, and mammalian cells by inducing oxidative stress due to which it acts as a potent antibacterial, antifungal, and anticancer agent. Its potential role as a neuroprotectant needs further exploration. However, pyocyanin exacerbates the damaging effects of nosocomial infections caused by P. aeruginosa in immunocompromised individuals. Further, cystic fibrosis (CF) patients are highly susceptible to persistent P. aeruginosa infections in the respiratory system. The bacterial cells form colonies and three interconnected QS networks— pqs, las , and rhl —get activated, thus stimulating the cells to produce pyocyanin which exacerbates pulmonary complications. As an opportunistic pathogen, P. aeruginosa produces pyocyanin to impede the recovery of injuries like burn wounds through its anti-proliferative activity. Moreover, pyocyanin plays a vital role in compounding P. aeruginosa infections by promoting biofilm formation. This review begins with a brief description of the characteristics of pyocyanin, its activity, and the different aspects of its production including its biosynthesis, the role of QS, and the effect of environmental factors. It then goes on to explore the potential applications of pyocyanin as a biotherapeutic molecule while also highlighting the biomedical challenges and limitations that it presents.

Abstract:Chlamydiae are obligate intracellular Gram-negative bacteria and widespread pathogens in humans and animals. Broad-spectrum antibiotics are currently used to treat chlamydial infections. However, broad-spectrum drugs also kill beneficial bacteria. Recently, two generations of benzal acylhydrazones have been shown to selectively inhibit chlamydiae without toxicity to human cells and lactobacilli, which are dominating, beneficial bacteria in the vagina of reproductive-age women. Here, we report the identification of two acylpyrazoline-based third-generation selective antichlamydials (SACs). With minimal inhibitory concentrations (MIC) and minimal bactericidal concentrations (MBC) of 10-25 μM against Chlamydia trachomatis and Chlamydia muridarum, these new antichlamydials are 2- to 5-fold more potent over the benzal acylhydrazone-based second-generation selective antichlamydial lead SF3. Both acylpyrazoline-based SACs are well tolerated by Lactobacillus, Escherichia coli, Klebsiella, and Salmonella as well as host cells. These third-generation selective antichlamydials merit further evaluation for therapeutic application.

Abstract:Inclusion membrane proteins (Incs) play an important role in the structure and stability of chlamydial inclusion and the interaction between Chlamydia spp. and their hosts. Following Chlamydia infection through the respiratory tract, human polymorphonuclear neutrophils (hPMN) not only act as the primary immune cells reaching the lungs, but also serve as reservoir for Chlamydia. We have previously identified a Chlamydia psittaci hypothetical protein, CPSIT_0556, as a medium expressed inclusion membrane protein. However, the role of inclusion membrane protein, CPSIT_0556 in regulating hPMN functions remains unknown. In the present study, we found that CPSIT_0556 could not only inhibit hPMN apoptosis through the PI3K/Akt and NF-κB signaling pathways by releasing IL-8, but also delays procaspase-3 processing and inhibits caspase-3 activity in hPMN. Up-regulating the expression of anti-apoptotic protein Mcl-1 and down-regulating the expression of pro-apoptotic protein Bax could also inhibit the translocalization of Bax in the cytoplasm into the mitochondria, as well as induce the transfer of p65 NF-κB from the cytoplasm to the nucleus. Overall, our findings demonstrate that CPSIT_0556 could inhibit hPMN apoptosis through PI3K/Akt and NF-κB pathways and provide new insights towards understanding a better understanding of the molecular pathogenesis and immune escape mechanisms of C. psittaci .

Effects of nuclear factor kappa B inhibitor pyrrolidine dithiocarbamate on the expression of pro- and anti-inflammatory cytokines in mice with Chlamydia pneumoniae pneumonia

Abstract:Objective To investigate the effects of nuclear factor-kappa B(NF-κB) inhibitor pyrrolidine dithiocarbamate (PDTC) on the expression of pro- and anti-inflammatory cytokines in the tissues of mice infected with C.pneumoniae and the associated regulating mechanism.Methods Ninety male C3H/HeJ mice were randomly divided into 3 groups:normal control; C.pneumoniae infection;and C.pneumoniae infection plus NF-κB inhibitor intervention.The infection was induced by intranasal inoculation of 4.0×10~6 IFU /mL of C.pneumoniae per mouse in the C.pneumoniae-infected animals.The animals of intervention group were injected with NF-kB inhibitor PDTC(50 mg/kg) intraperitoneally.Mice in the three groups were killed on Day 1,4,7,14,and 21,respectively.Lung tissue of the animals was sampled for analysis of the expression of NF-kB protein kinase by Western blot.The concentrations of TNF-αand IL-10 in the lung tissue were measured by ELISA.Results The expression of NF-kB in C.pneumoniae infection group increased significantly compared to the normal control group.The level peaked on day 4[(12.44±1.42) pg/mg] and decreased on day 14[(5.61±0.81 pg/mg).The expression levels of TNF-αand IL-10 in the lung tissue were higher than that of normal control group at different time points.PDTC intervention significantly decreased the expression of NF-kB in C.pneumoniae infected animals.The concentrations of TNFαand IL-10 in the lung tissue were lower than that of C.pneumoniae infection group at different time points(P<0.01).Conclusions NF-kB can activate pulmonary inflammatory reaction.The specific NF-kB inhibitor PDTC can effectively down-regulate the expression of pro-inflammatory cytokines.It may inhibit the release of inflammatory mediators and so attenuate lung inflammation.

Abstract:The Chlamydiaceae comprise a group of highly adapted bacterial pathogens sharing a unique intracellular lifestyle. Three Chlamydia species are pathogenic to humans: Chlamydia trachomatis, Chlamydia pneumoniae, and Chlamydia psittaci. C. trachomatis is the leading bacterial cause of sexually-transmitted infections and infectious blindness worldwide. Chlamydia pneumoniae is a major cause of community-acquired atypical pneumonia. C. psittaci primarily affects psittacine birds and can be transmitted to humans causing psittacosis, a potentially fatal form of pneumonia. As opposed to other bacterial pathogens, the spread of clinically relevant antimicrobial resistance genes does not seem to be a major problem for the treatment of Chlamydia infections. However, when exposed to stressing conditions, like those arising from exposure to antimicrobial stimuli, these bacteria undergo a temporary interruption in their replication cycle and enter a viable but non-cultivable state known as persistence. When the stressing conditions are removed, Chlamydia resumes replication and generation of infectious particles. This review gives an overview of the different survival strategies used by Chlamydia to evade the deleterious effects of penicillin and IFNγ, with a focus on the different models used to study Chlamydia persistence, their contribution to elucidating the molecular basis of this complex phenomenon and their potential implications for studies in animal models of infection.

Critical Role Of The Interleukin-17/Interleukin-17 Receptor Axis In Regulating Host Susceptibility To Respiratory Infection With Chlamydia Species

Abstract:The specific contribution of interleukin-17/interleukin-17 receptor (IL-17/IL-17R)-mediated responses in regulating host susceptibility against obligatory intracellular Chlamydia infection was investigated in C57BL/6 and C3H/HeN mice during Chlamydia muridarum respiratory infection. We demonstrated that Chlamydia stimulated IL-17/IL-17R-associated responses in both Chlamydia-resistant C57BL/6 and Chlamydia-susceptible C3H/HeN mice. However, C3H/HeN mice developed a significantly greater IL-17/IL-17R-associated response than C57BL/6 mice did. This was reflected by an increase in IL-17 mRNA expression, a higher recall IL-17 production from splenocytes upon antigen restimulation, and higher production of Th17-related cytokines (IL-23 and IL-6) and chemokines (chemokine [C-X-C motif] ligand 2 [CXCL1]/keratinocyte-derived chemokine [KC] and CXCL2/macrophage inflammatory protein 1 [MIP2]) in C3H/HeN mice than in C57BL/6 mice. Furthermore, C3H/HeN mice displayed a massive accumulation of activated and preactivated neutrophils in the airway and lung parenchyma compared to their C57BL/6 counterparts. We further demonstrated that the skewed IL-17/Th17 profile in C3H/HeN mice was predisposed by a higher basal level of IL-17 receptor C (IL-17RC) expression and then further amplified by a higher inducible IL-17RA expression in lungs. Most importantly, in vivo delivery of IL-17RA antagonist that resulted in a 50% reduction in the neutrophilic infiltration in lungs was able to reverse the susceptible phenotype of C3H/HeN mice to respiratory Chlamydia infection. Thus, our data for the first time have demonstrated a critical role for the IL-17/IL-17R axis in regulating host susceptibility to Chlamydia infection in mice.
Nafamostat mesylate inhibits chlamydial intracellular growth in cell culture and reduces chlamydial infection in the mouse genital tract

Abstract:Urogenital Chlamydia trachomatis (C. trachomatis) infection is one of the most common bacterial sexually transmitted diseases worldwide. Untreated C. trachomatis infections that ascend to the upper genital tract lead to a series of severe complications. To search for novel antichlamydial drugs, we evaluated the effect of nafamostat mesylate (NM), a synthetic serine protease inhibitor, on chlamydial infection. NM inhibited chlamydial intracellular growth and reduced both the inclusion size and number in cell culture. NM may mainly target the intracellular reticulate bodies for inhibition. NM was also effective in enhancing chlamydial clearance from mouse genital tract when NM was applied to mice via intravaginal inoculation. The vaginal NM did not significantly alter inflammatory cytokine responses in the mouse genital tract. Thus, we have demonstrated a novel role of NM in inhibiting the obligate intracellular bacterium Chlamydia.

Efficacy of a Novel Affitoxin Targeting Major Outer Membrane Protein Against Chlamydia Trachomatis in Vitro and in Vivo

Abstract:Targeted therapy is an attractive approach for treating infectious diseases. Affibody molecules have similar capability to antibodies that facilitate molecular recognition in both diagnostic and therapeutic applications. Targeting major outer membrane protein (MOMP) for treating infection of Chlamydia trachomatis, one of the most common sexually transmitted pathogens, is a promising therapeutic approach. Previously, we have reported a MOMP-specific affibody (ZMOMP:461) from phage display library. Here, we first fused it with modified Pseudomonas exotoxin (PE38KDEL) and a cell-penetrating peptide (CPP) to develop an affitoxin, Z461X-CPP. We then verified the addition of both toxin and CPPs that did not affect the affinitive capability of ZMOMP:461 to MOMP. Upon uptake by C trachomatis-infected cells, Z461X-CPP induced cell apoptosis in vitro. In an animal model, Z461X significantly shortened the duration of C trachomatis infection and prevented pathological damage in the mouse reproductive system. These findings provide compelling evidence that the MOMP-specific affitoxin has great potential for targeting therapy of C trachomatis infection. Affitoxin constructed from MOMP-specific affibody carrying PE38KDEL toxin and a membrane-penetrating peptide could effectively target Chlamydia trachomatis-infected cells at the cell level in vitro and in mouse models. Abstract:The antimicrobial peptides Mytichitin-CB and Hispidalin were isolated from Mytilus coruscus and Benincasa hispida , respectively. Our previous studies showed that Mytichitin-CB, when expressed in Chlamydomonas reinhardtii , inhibits the bacterial growth with moderate efficiency. Naturally isolated or Pichia pastoris -expressed Hispidalin also exerts moderate efficiency for inhibiting the growth of bacteria. Here, we expressed a chimeric peptide composed of Mytichitin-CB and Hispidalin that we named as MCH in C. reinhardtii . MCH was stably expressed and its yield accounted for 0.18% of total soluble proteins of the host cells. C. reinhardtii -derived MCH inhibited the growth of both Gram-positive and Gram-negative bacteria by disrupting their cell membrane integrity. Its minimal inhibitory concentration (MIC) were ranged between 20 and 40 μg/ml, demonstrating that MCH is more potent for inhibiting the bacterial growth than individual microalgae-expressed Mytichitin-CB and yeast-expressed Hispidalin. Of more importance, MCH was temperature- and pH-tolerant, protease-resistant and exhibited low hemolytic activity against rat erythrocytes and low cytotoxicity to human embryonic kidney 293 (HEK 293) and Madin-Darby bovine kidney (MDBK) cells. Therefore, we conclude that C. reinhardtii -derived chimeric peptide MCH has a great potential for being developed to replace traditional antibiotics in the future.

Abstract:The Gram-negative, opportunistic pathogen Pseudomonas aeruginosa has become a serious threat to global health, with increasing resistance towards even the most potent antibiotics. Like other bacteria, the pathogen produces a number of virulence factors that promote its pathogenicity. Of these, metallophores constitute an important group, able to scavenge metal ions from the surrounding environment to secure the metabolic functions of the bacterium. Pseudomonads produce the iron-chelating metallophore (siderophore) pyochelin (PCH), which, in addition to its iron scavenging ability, is an effector for the transcriptional regulator PchR in its Feᴵᴵᴵ-bound form (ferripyochelin). In the present study, docking studies predicted a major ferripyochelin binding site in PchR, which prompted the exploration of nor-pyochelin analogs to produce tight binding to PchR, and thereby establish a lasting effector response associated with upregulation of the genes involved in the PCH metabolism. In addition, we investigated the effects of using the analogs to bind the antimicrobial cations Gaᴵᴵᴵ and Inᴵᴵᴵ, leading to enhanced uptake and further disruption of the cell machinery. Selected analogs of nor-pyochelin (nor-PCH) were synthesized and their Gaᴵᴵᴵ and Inᴵᴵᴵ-based complexes were assessed for antimicrobial activity. The results indicate that the Gaᴵᴵᴵ-complexes inhibit the pathogens under iron-limited conditions, while the Inᴵᴵᴵ-based systems are more effective in iron-rich media. Several of the Gaᴵᴵᴵ-complexes were shown to be highly effective against an MDR P. aeruginosa clinical isolate, with minimum inhibitory concentrations (MICs) of ≤1 µg/mL. Similarly, two of the Inᴵᴵᴵ-based systems were particularly effective against the isolate, with an MIC of 8 µg/mL. These results show high promise in comparison with other, traditionally potent antibiotics. Preliminary mechanistic investigations of the antimicrobial activity using pseudomonal transposon mutants suggested that the inhibitory effects of the Inᴵᴵᴵ-based systems could be due to acute iron deficiency as a result of Inᴵᴵᴵ-bound bacterioferritin (BfrB). Cytotoxiticy analysis indicated that the compounds generally show low toxicity toward mammalian cells under the experimental conditions.

Abstract:Bacterial quorum sensing (QS) is anticipated as a new potential target for the development of antimicrobial drugs. An anti-QS substance against Chromobacterium violaceum CV026 and Pseudomonas aeruginosa PA01 has been isolated and purified from the crude extracts of the marine fungus Penicillium chrysogenum DXY-1, and the accurate structure was identified as cyclo(l-Tyr-l-Pro). This cyclic dipeptide at sub-minimum inhibitory concentration can decrease the QS-regulated violacein production of C. violaceum CV026 by 79% and QS-mediated pyocyanin production, proteases, and elastase activity of P. aeruginosa PA01 by 41%, 20%, and 32%, respectively. In addition, it can also destroy the biofilm formation and decrease QS gene expression of P. aeruginosa PA01. Molecular docking was further performed, and the obtained data indicated that this dipeptide blocks the effect of QS autoinducers through competitive binding to the same pocket of the receptor proteins. We expect this anti-QS cyclic dipeptide to be a potential pro-drug treating drug-resistant P. aeruginosa infections, and these findings could relieve the alarming problem of microbial resistance to antimicrobial drugs.This article has not yet been cited by other publications.

Abstract:Targeted therapy is an attractive approach for treating infectious diseases. Affibody molecules have similar capability to antibodies that facilitate molecular recognition in both diagnostic and therapeutic applications. Targeting major outer membrane protein (MOMP) for treating infection of Chlamydia trachomatis , one of the most common sexually transmitted pathogens, is a promising therapeutic approach. Previously, we have reported a MOMP-specific affibody (Z MOMP :461) from phage display library. Here, we first fused it with modified Pseudomonas Exotoxin (PE38KDEL) and a cell-penetrating peptide (CPP) to develop an affitoxin, Z461X-CPP. We then verified the addition of both toxin and CPPs that did not affect the affinitive capability of Z MOMP :461 to MOMP. Upon uptake by C.trachomatis -infected cells, Z461X-CPP induced cell apoptosis in vitro . In animal model, Z461X significantly shortened the duration of C. trachomatis infection and prevented pathological damage in mouse reproductive system. These findings provide compelling evidence that the MOMP-specific affitoxin has great potential for targeting therapy of C. trachomatis infection.
