Abstract:The effects of moderate hypothermia (28 °C) on the response of human varicose spermatic vein to α1-adrenoceptor agonist phenylephrine and the role of endothelial nitric oxide (NO) in these effects were studied. Concentration-response curves for phenylephrine (10-9 to 3 × 10-4 M) were recorded in rings with and without endothelium at 37 and 28 °C. To further analyze the role of NO, in the response to phenylephrine during hypothermia, the effects of this agonist in the presence of NG-nitro-L-arginine methyl ester (10-4 M) were also determined. Under every condition tested, phenylephrine produced a marked, concentration-dependent contraction. Sensitivity of intact veins to the agonist was consistently lower at 28 °C than at 37 °C. There was no significant difference in phenylephrine response at 28 and 37 °C in vessels without endothelium but at 28 °C veins without endothelium showed a higher sensitivity than intact veins to phenylephrine. The sensitivity of veins with and without endothelium to nitroprusside (10-9 to 3 × 10-3 M) was significantly decreased during hypothermia, and endothelium removal did not affect the relaxation to this nitrovasodilator. These results suggest that moderate hypothermia decreases the sensitivity of human varicose spermatic vein to phenylephrine probably by increasing the availability of endothelial NO.
Vasomotor Effect of Cyclovirobuxine D and Its Underlying Mechanisms

Abstract:To investigate the vasomotor effects of Cyclovirobuxine D (CVB) on the rat aortic rings in vitro and underlying mechanisms, rat aorta was obtained, and aortic rings with or without endothelium were pre-treated with potassium chloride (KCl) or phenylephrine (PE). Then, CVB was added and its vasomotor effects were observed. In addition, the influence of other drugs on the CVB induced vasomotor effects was detected. The effects of CVB and verapamil (VER) on the dilation of the aortic rings were observed. After incubation in Ca2+ free solution containing CVB, the KCl pre-treated aortic rings were treated with CaCl2 aiming to investigate the effects of CVB on the influx of extracellular calcium, or with PE and caffeine to observe the effects of CVB on the release of intracellular calcium. In aortic rings precontracted with PE or KCl, CVB produced concentration-dependent relaxation in both endothelium-intact and denuded rings. But the dilation of rings with intact endothelium was more potent than that of rings without endothelium. In intact rings, L-NAME (100 mu mol/L) or indomethacin (10 mu mol/L) reduced the degree of CVB-induced relaxation. CVB (3.2x10(-4) mol/L) and VER (1 mu mol/L) could induce the dilation of KCl precontracted rings and these effects could be partially reversed by CaCl2 (1.25 mmol/L). In Ca2+ free solution, calcium-dependent KCl contractions were inhibited by CVB (3.2x10(-4) mol/L). Furthermore, CVB (3.2x10(-4) mol/L) inhibited the PE (1 mu mol/L) induced contraction of aortic rings, but had no effect on the caffeine (10 mmol/L) induced contraction. CVB vasodilates by endothelium-dependent and endothelium-independent mechanism, Endothelium-dependent involving nitric oxide or prostaglandin released from the endothelium. Its endothelium-independent vasodilation probably occurs via the suppression of voltage-sensitive Ca2+ channel and inhibition of IP3-sensitive Ca2+ released in vascular smooth muscles, but has no effect on ryanodine receptor.
Effect of Emodin on NO-cGMP Signal Pathway in Rat Vascular Endothelium in Vitro

Abstract:OBJECTIVE To investigate the vasorelaxation effect of emodin and its relationship with NO-cGMP signal pathway. METHODS Changes of tension of rat thoracic aortic rings were measured by MedLab biologic signal collection system, and the activity of total nitric oxide synthase (tNOS), constitutive NOS (cNOS) and inducible NOS (iNOS) in endothelium after being treated with emodin was determined with nitric acid reductase method. RESULTS Emodin relaxed the phenylephrine and potasium chlorate induced contraction of aortic rings, either with or without intact endothelium, in a concentration-dependent manner. Pretreatment of no-specific potassium channel blocker strontium chloride (CsCL) could attenuate the vasorelaxation effect of emodin on aortic rings without intact endothelium, but it could not inhibit vasorelaxation of emodin on aortic rings with intact endothelium. This vasorelaxation action of emodin (40 micromol/L) could be partial blocked by NOS inhibitor L-NAME and guanylate cyclase inhibitor ODQ, with the vasorelaxation range dropped to 64.76 +/- 13.73% and 6.28 +/- 4.79% respectively. Moreover, emodin (40 micromol/L) increased iNOS activity significantly. CONCLUSION The concentration-dependent vasorelaxation effect of emodin might act by activating the NO-cGMP pathway in vascular endothelium.
Growth Rate Determination through Automated TEM Image Analysis: Crystallization Studies of Doped SbTe Phase-Change Thin Films

Abstract:Abstract A computer-controlled procedure is outlined here that first determines the position of the amorphous-crystalline interface in an image. Subsequently, from a time series of these images, the velocity of the crystal growth front is quantified. The procedure presented here can be useful for a wide range of applications, and we apply the new approach to determine growth rates in a so-called fast-growth-type phase-change material. The growth rate (without nucleation) of this material is of interest for comparison with identical material used in phase-change random access memory cells. Crystal growth rates in the amorphous phase-change layers have been measured at various temperatures using in situ heating in a transmission electron microscope. Doped SbTe films (20 nm thick) were deposited on silicon nitride membranes, and samples with and without silicon oxide capping layer were studied. The activation energy for growth was found to be 3.0 eV. The samples without capping layer exhibit a nucleation rate that is an order of magnitude higher than the samples with a silicon oxide capping layer. This difference can be attributed to the partial oxidation of the phase-change layer in air. However, the growth rates of the samples with and without capping are quite comparable.
EXPERIMENTAL STUDY OF MECHANISM AND MEASURES OF PREVENTION AND TREATMENT OF HYPOTHERMAL VASOCONSTRICTION

Abstract:OBJECTIVE:To investigate the mechanism and to explore the measures of prevention and treatment of hypothermal vasoconstriction.METHODS:By the techniques of endothelial cell culture and scanning electron microscopy, and vasomotor functional test of isolated vascular vessels, the relation of hypothermal vasoconstriction and the release of endothelium-derived contractile and vasodilative factors were observed.RESULTS:Hypothermia obviously induced vasoconstriction of isolated vascular vessels, whether endothelium was intact or removed, the lower the temperature, the higher the vascular tension. Removal of endothelium could decrease the effect of vasoconstriction by hypothermia. The conditioned medium of bovine aortic endothelial cell could induce significantly vasoconstriction of isolated rat common neck arterial ring in hypothermia. It indicated that the bovine aortic endothelial cells secreted contractile factors into the medium. Reheating to 37 degrees C or vasodilator or reheating plus vasodilator did not obviously influence the hypothermia-induced vasoconstriction within 2 hours. When reheating to 50 degrees C, vascular tension was decreased, but only changed in range of 28% to 42%.CONCLUSION:Hypothermia vasoconstriction is relative to vasoconstrictor factors secreted by endothelium. Reheating to 37 degrees C or vasodilator does not antagonize the constriction of vascular vessels. Reheating to 50 degrees C only partially eliminates the constrict effect of blood vessels, so the prevention of hypothermia vasoconstriction should be emphasized.
Guanylate cyclase and not ATP-dependent K(+) channels seems temperature-dependent in smooth muscle relaxation of human umbilical arteries

Abstract:The effects of K(+) channel opener, nicorandil [N-(2-hydroxyethyl)-nicotinamide nitrate], on isolated human umbilical arteries were investigated at two temperatures: 37 degrees C and 25 degrees C. The purpose of this investigation was: (1) to confirm the relaxant effect of nicorandil, (2) to elucidate the influence of endothelium and temperature on nicorandil-induced relaxation, (3) to determine which of guanylate cyclase or ATP-sensitive K(+) channels was implicated in temperature-induced relaxation of smooth muscles. Rings, 3-mm-wide, were suspended in organ chambers for isometric force measurement. All solutions were aerated with 95% O(2)-5% CO(2) and maintained at 37 degrees C or 25 degrees C (cooling), pH 7.4. The presence of an intact endothelium was confirmed by immunohistochemistry. During the first set of experiments after contraction with 5-hydroxytryptamine (5-HT 10(-5) M), nicorandil (10(-9)-10(-4) M) was added to the organ chambers with controls and in with rings incubated with L-arginine, N-nitro-L-arginine (L-NNA) an inhibitor of nitric oxide (NO) synthase, [1-H-(1,2,4) oxadiazole (4,3-a) quinoxalin-1-one] (ODQ), a specific inhibitor of guanylate cyclase, or glibenclamide, an antagonist of nicorandil, all at 10(-5) M. In another set of experiments, rings were contracted with 5-HT (10(-5) M) and relaxed with 3-morpholinosydnonimine [SIN-1 (10(-9)-3x10(-5) M) or cromakalim (10(-9)-3x10(-5) M)]. Our results showed that nicorandil induced concentration-dependent relaxation. At 37 degrees C, in the control, the maximum relaxation was 90+/-5%, and 60+/-8% at 25 degrees C (P<0.01). However, the relaxation at 37 degrees C or 25 degrees C remained unchanged after pretreatment with L-arginine, L-NNA, this suggests that the same concentration of drugs used in this type of vessel does not appear to modulate the relaxant effect of nicorandil. On the other hand, we observed that the relaxant effect of SIN-1 was 72+/-5% at 37 degrees C and only 28+/-7% at 25 degrees C (P<0.01). However, relaxations with cromakalim were partly influenced by cooling. In the presence of ODQ, the nicorandil-induced relaxation observed at 37 degrees C or 25 degrees C was less than that in the control and in the rings incubated with glibenclamide. These results for human umbilical arteries indicate that cooling decreases the relaxation response of smooth muscles and that this is partly due to a decreased response to guanylate cyclase.
Rate dependency and role of nitric oxide in the vascular response to direct cooling in human skin

Abstract:Local cooling of nonglabrous skin without functional sympathetic nerves causes an initial vasodilation followed by vasoconstriction. To further characterize these responses to local cooling, we examined the importance of the rate of local cooling and the effect of nitric oxide synthase (NOS) inhibition in intact skin and in skin with vasoconstrictor function inhibited. Release of norepinephrine was blocked locally (iontophoresis) with bretylium tosylate (BT). Skin blood flow was monitored from the forearm by laser-Doppler flowmetry (LDF). Cutaneous vascular conductance (CVC) was calculated as the ratio of LDF to blood pressure. Local temperature was controlled over 6.3 cm2 around the sites of LDF measurement. Local cooling was applied at -0.33 or -4 degrees C/min. At -4 degrees C/min, CVC increased (P < 0.05) at BT sites in the early phase. At -0.33 degrees C/min, there was no early vasodilator response, but there was a delay in the onset of vasoconstriction relative to intact skin. The NOS inhibitor N(G)-nitro-L-arginine methyl ester (L-NAME) (intradermal microdialysis) decreased (P < 0.05) CVC by 28.3 +/- 3.8% at untreated sites and by 46.9 +/- 6.3% at BT-treated sites from the value before infusion. Rapid local cooling (-4 degrees C/min) to 24 degrees C decreased (P < 0.05) CVC at both untreated (saline) sites and L-NAME only sites from the precooling levels, but it transiently increased (P < 0.05) CVC at both BT + saline sites and BT + L-NAME sites in the early phase. After 35-45 min of local cooling, CVC decreased at BT + saline sites relative to the precooling levels (P < 0.05), but at BT + L-NAME sites CVC was not reduced below the precooling level (P = 0.29). These findings suggest that the rate of local cooling, but not functional NOS, is an important determinant of the early non-adrenergic vasodilator response to local cooling and that functional NOS, adrenergic nerves, as well as other mechanisms play roles in vasoconstriction during prolonged local cooling of skin.
Homogeneous nucleation rates of higher n-alcohols measured in a laminar flow diffusion chamber.

Abstract:Nucleation rate isotherms of n-butanol, n-pentanol, n-hexanol, n-heptanol, and n-octanol were measured in a laminar flow diffusion chamber using helium as carrier gas. The measurements were made at 250-310 K, corresponding to reduced temperatures of 0.43-0.50, and at atmospheric pressure. Experimental nucleation rate range was from 10(3) to 10(7) cm(-3) s(-1). The expression and accuracy of thermodynamic parameters, in particular equilibrium vapor pressure, were found to have a significant effect on calculated nucleation rates. The results were compared to the classical nucleation theory (CNT), the self-consistency corrected classical theory (SCC) and the Hale's scaled model of the CNT. The average ratio between the experimental and theoretical nucleation rates for all alcohols used was 1.5x10(3) when the CNT was used, and 0.2x10(-1) when the SCC was used and 0.7x10(-1) when the Hale's scaled theory was used. The average values represent all the alcohols used at the same reduced temperatures. The average ratio was about the same throughout the temperature range, although J(exp)/J(the) calculated with the Hale's scaled theory increased slightly with increasing temperature. The saturation ratio dependency was predicted closest to experiment with the classical nucleation theory. The nucleation rates were compared to those found in the literature. The measurements were in reasonable agreement with each other. The molecular content of critical alcohol clusters was between 35 and 80 molecules. At a fixed reduced temperature, the number of molecules in a critical cluster decreased as a function of alcohol carbon chain length. The number of molecules in critical clusters was compared to those predicted by the Kelvin equation. The theory predicted the critical cluster sizes well.
Hypothermia Inhibits Dexmedetomidine-Induced Contractions in Isolated Rat Aortae

Abstract:Dexmedetomidine is widely used to induce sedation in the perioperative period. This study examined the effect of hypothermia (33 and 25 °C) on dexmedetomidine-induced contraction in an endothelium-intact aorta with or without the nitric oxide synthase inhibitor NW-nitro-L-arginine methyl ester (L-NAME). In addition, the effect of hypothermia on the contraction induced by dexmedetomidine in an endothelium-denuded aorta with or without a calcium-free Krebs solution was examined. The effects of hypothermia on the protein kinase C (PKC), myosin light chain (MLC20) phosphorylation, and Rho-kinase membrane translocation induced by dexmedetomidine were examined. Hypothermia inhibited dexmedetomidine-induced contraction in the endothelium-intact aorta with L-NAME or endothelium-denuded aorta. Hypothermia had almost no effect on the dexmedetomidine-induced contraction in the endothelium-denuded aorta with the calcium-free Krebs solution; however, the subsequent contraction induced by the addition of calcium was inhibited by hypothermia. Conversely, the transition from profound hypothermia back to normothermia reversed the hypothermia-induced inhibition of subsequent calcium-induced contractions. Hypothermia inhibited any contraction induced by KCl, PDBu, and NaF, as well as PKC and MLC20 phosphorylation and Rho-kinase membrane translocation induced by dexmedetomidine. These results suggest that hypothermia inhibits dexmedetomidine-induced contraction, which is mediated mainly by the impediment of calcium influx and partially by the attenuation of pathways involving PKC and Rho-kinase activation.

Dependence of Myocardial Hypothermia Tolerance on Sources of Activator Calcium.

Abstract:To determine the relationship between cardiac hypothermia tolerance and the sources of activator calcium, we selectively modified either the sarcolemmal calcium permeability by nifedipine or the sarcoplasmic reticulum function by caffeine in papillary muscles from both the rat, as a cold sensitive model, and the ground squirrel,Citellus dauricus,a deep hibernator. Both force–interval relationship and cooling performance were investigated. At 25°C, the slope of the force–interval curve of the ground squirrel was nearly double that of the rat. At shorter test intervals 0.5 μMnifedipine moved the curve down with little effect at longer intervals, and the curve slope increased. Caffeine (1 mM) decreased the peak force and eliminated its dependence upon test interval. When the temperature was lowered, rat preparations showed a marked increase of resting tension and aftercontraction between 7 and 12°C and became inexcitable. In contrast, they maintained contractility down to a few degrees above 0°C without aftercontraction and increased resting tension in the ground squirrel. In the rat nifedipine shortened the contractions, prevented the increase of resting tension, and minimized aftercontractions, with little improvement of contractility. Caffeine prolonged the contractions, caused a striking increase of resting tension and aftercontractions, and finally disabled the contractility at about 5–10°C, even in the ground squirrel. We conclude that depressed calcium influx helps to prevent hypothermic calcium overload of the cardiac cells. Good function of the sarcoplasmic reticulum is essential for tolerance of hypothermia by cardiac cells. A suggestion that may improve the hypothermic tolerance of the myocardium from nonhibernators is postulated.
Exploiting the Cellular Actions of Skca and Ikca Channels to Manipulate Endothelial Function and Vascular Tone

Abstract:Growing evidence indicates that small‐ and intermediate‐conductance, Ca2+‐activated K+ channels (SKCa and IKCa channels, respectively) play pivotal roles in the cellular mechanisms contributing to stimulated vasorelaxation (i.e. nitric oxide (NO) synthesis, actions of EDHF). Using single cell patch clamp and fluorimetry, along with arteriolar pressure myography, we have obtained novel results demonstrating that activators of SKCa and IKCa channels (i.e. NS309 and DCEBIO, 0.01 ‐ 1 μM) enhance a number of cellular processes associated with stimulated vasorelaxation (i.e. cytosolic Ca2+ elevations, membrane hyperpolarization and NO synthesis). These observed effects of NS309 and DCEBIO were largely prevented by the SKCa and IKCa channel blockers apamin and TRAM‐34 in combination, but not when applied separately. In rat cremaster arterioles pressurized to 70 mmHg, NS309 and DCEBIO reduced myogenic tone and further augmented ACh‐induced vasodilation. The eNOS inhibitor L‐NAME (0.1 mM) decreased ACh‐evoked inhibition of myogenic tone, but did not affect the dilatory responses to NS309 and DCEBIO. The observed inhibition of NS309/DCEBIO action following endothelial denudation confirmed that these agents acted predominantly via the vascular endothelium. Finally, we will discuss initial efforts to discern the contribution of STIM1 to SKCa and IKCa activity and endothelial vasodilatory processes.
Amlodipine, but Not Verapamil or Nifedipine, Dilates Rabbit Femoral Artery Largely Through A Nitric Oxide- and Kinin-Dependent Mechanism

Abstract:We investigated the nitric oxide (NO) dependence of vasorelaxation in response to different calcium channel blockers (CCB), in rabbit femoral artery in vivo. Anaesthetized rabbits underwent femoral artery ligation, and blood from the proximal artery was returned distal to the ligature through a constant infusion pump. The effects of local injection of CCB on perfusion pressure and plasma nitrite+nitrate (NOx, which reflects local NO biosynthesis) concentration in this system were determined. Intra‐arterial verapamil, nifedipine or amlodipine 10 μmol kg−1 each reduced perfusion pressure. Pre‐treatment with intra‐arterial NG‐nitro‐L‐arginine methyl ester (L‐NAME, a NO synthase inhibitor) 1 μmol kg−1 did not affect responses to verapamil or nifedipine, but attenuated the reduction in perfusion pressure to amlodipine, from 33.2±2.1% to 22.5±1.6% (P=0.002). Intra‐arterial amlodipine – unlike verapamil or nifedipine – increased femoral venous NOx, from 9.1±0.4 μM to 14.1±0.5 μM (P=0.005). The bradykinin B2 receptor antagonist HOE 140, 30 mg kg−1, attenuated the reduction in perfusion pressure and abolished the rise in venous NOx concentration, following intra‐arterial amlodipine. Amlodipine potently inhibited serum angiotensin converting‐enzyme (ACE) activity in vitro, as effectively as enalapril at similar concentrations. These results suggest that the vasorelaxant effects of nifedipine and verapamil are NO‐independent, whereas those of amlodipine are partly NO‐dependent, in rabbit femoral artery in vivo. This effect of amlodipine occurs through B2 receptor activation, and may be related to an increase in local bradykinin through inhibition of ACE. [Hemodynamic effects in high-dose infusion of nimodipine, a new calcium antagonist]

Abstract:Nimodipine, a new calcium channel blocker, seems to be effective in the treatment of vasospasm in cerebral vasculature. Typical cardiovascular side effects have limited the dose in neurology and neurosurgery to 0.03 mg/kg X h. This study was designed to examine the influence of an infusion of high dose nimodipine (0.09 mg/kg X h) on haemodynamics. 52 patients undergoing aorto-coronary bypass surgery and prospectively randomised in a nimodipine group and a control group having received 0.9% saline solution as placebo were investigated at 3 different times: 1. before induction of anaesthesia (n = 6) 2. during anaesthesia (n = 10) 3. during extracorporeal circulation (ECC n = 10). Predominant effect of high-dose nimodipine was a decrease in total systemic resistance (TSR), followed by a decrease in mean arterial pressure (MAP) and a significant increase in cardiac output. Haemodynamic effects were much more pronounced during anaesthesia in comparison to patients before induction of anaesthesia, thus demonstrating an interaction between anaesthetics and calcium channel blocker. Heart rate (HR -9.3%) and dp/dtmax (-17%) showed a decrease, too. The decrease in MAP and HR in connection with a decrease in left ventricular pressure (-21.9%) and left ventricular end diastolic pressure (-42.8%) indicate a reduction in myocardial oxygen demand. An increasing dosage of nimodipine is accompanied by increasing cardiovascular effects. From the haemodynamic point of view high dosage of nimodipine seems to be of advantage in patients with hypertension and/or coronary heart disease suffering simultaneously from cerebral vasospasm.

Abstract:The effect of cold on the reactivity of the rat superior mesenteric artery (SMA), estimated by the magnitude of contractile responses of its segments to noradrenaline (NA, 0.01–10.0 μM) and electrical field stimulation (EFS) of the perivascular nerves at a frequency of 3, 10 and 40 Hz in both the absence and presence of NA in the medium at a temperature of 37 or 25°C, was studied in isometric experiments. Cooling did not alter SMA tone evoked by NA at all its concentrations used. In both the absence and presence of low NA concentrations (0.01–0.10 μM), cooling led to a significant decrease in the neurogenic SMA response at all EFS frequencies, while in the presence of NA at a high concentration (1–10 μM), cooling had no significant effect on this response. In the presence of NA at a low concentration, cooling insignificantly (at low EFS frequencies) and significantly (at high EFS frequencies) reduced the potentiating effect of NA on the neurogenic SMA constriction. At a high concentration under normal temperature conditions (37°C), NA reduced the neurogenic SMA constriction at all EFS frequencies, while under cooling conditions (25°C), it had a potentiating effect at low and a depressing effect at high EFS frequencies. These results demonstrate that the depressing effect of cold on the neurogenic constriction of the rat SMA, occurring at low and disappearing at high NA concentrations, as well as on the NA-evoked potentiation of the neurogenic vasoconstriction, is irrelevant to a decrease in the contractile effect of NA, which is the main neurotransmitter in this artery. These effects may contribute to blood rerouting from the surface to the deeper regions of the body, thus reducing heat loss and improving thermoregulation.

Histological grading of oral squamous cell carcinoma patients in Ibadan using Bryne's and Broders' grading systems--a comparative study.

Abstract:BACKGROUND Several histological grading systems for oral squamous cell carcinoma (OSCC) have been used previously to guide treatment and prognostication, amongst which are the Broders and the Bryne's methods. The Bryne's invasive tumour front (ITF) protocol has been adjudged to have good prognostic significance. We compared the use of Bryne's ITF system and Broders' grading system in grading OSCC at our centre in relation to clinico-demographic profile of patients. METHODS Thirty two formalin fixed paraffin embedded (FFPE) tissue samples of histologically diagnosed OSCC patients at the University College Hospital (UCH) Ibadan were selected, demographic data and site of lesions were retrieved from the medical records of the patients. The haematoxylin and eosin (H&E) slides of the 32 cases were prepared and evaluated using Bryne's ITF system and Broders classification. RESULTS Using Bryne's system, 28% of the cases had a high malignancy score while 72% had a low malignancy score. The high malignancy score lesions were most commonly seen in the palate (15.5%). Also Broders classification showed that 31.3% of cases were well differentiated tumours, 50% were moderately differentiated while 18.7% were poorly differentiated. CONCLUSION Either the Bryne's system or Broders classification can be used to grade OSCC with similar results being obtained.
Effects of Acute Transmural Pressure Elevation on Endothelium-Dependent Vasodilation in Isolated Rat Mesenteric Veins

Abstract:BACKGROUND/AIMS: The vascular regulatory function of the endothelium can be impaired by increases in transmural pressure (TMP). We tested the hypothesis that increasing TMP impairs the endothelial dilator function of rat mesenteric small veins (MSVs).METHODS: In PGF2α-preconstricted MSVs, bradykinin (BK), sodium nitroprusside (SNP) and S-Nitroso-N-acetylpenicillamine (SNAP) concentration-response curves were generated at intermediate (6 mm Hg) and high (12 mm Hg) pressures. BK-induced vasodilation was examined in the absence and presence of nitric oxide synthase inhibitor [N(ω)-nitro-L-arginine (L-NNA), 100 µM], cyclooxygenase inhibitor (indomethacin, 1 µM), and large (BKCa, paxilline, 500 nM) and small (SKCa, apamin, 300 nM) conductance Ca(2+)-activated K(+) channel blockers.RESULTS: BK, SNP and SNAP responses were not altered by TMP increases. BK-induced vasodilation was significantly reduced by L-NNA, indomethacin, apamin and paxilline at 6 mm Hg and L-NNA at 12 mm Hg, and was further reduced by coapplication of apamin and/or paxilline with L-NNA compared with responses obtained with either blocker. Endothelium removal completely abolished BK-induced vasodilation.CONCLUSION: Venous endothelial dilator function is not affected by TMP elevation. BK-induced vasodilation is completely dependent on the presence of functional endothelial cells and mediated in part by nitric oxide, BKCa and SKCa channels, while the participation of prostacyclin may be important at intermediate pressures.

Calcium antagonists and cardiac noradrenaline release in ischemia

Abstract:The effects of the calcium antagonists verapamil, gallopamil, nifedipine, felodipine and diltiazem on noradrenaline release during ischemia were studied in isolated perfused rat hearts. Endogenous levels of noradrenaline and its intraneuronal metabolite dihydroxyphenylethyleneglycol (DOPEG) were determined by high pressure liquid chromatography. Global isothermic ischemia of 20 min caused a release of endogenous noradrenaline amounting to 180 +/- 15 pmol/g. The calcium antagonists tested significantly suppressed ischemia-induced noradrenaline release (IC50 in mumols/l: verapamil 1, gallopamil 0.3, nifedipine 1, felodipine 3). Noradrenaline release during ischemia and the inhibitory effect of the calcium antagonists, were independent of extracellular calcium, indicating a nonexocytotic release mechanism. Interaction of the calcium antagonists with the major components of nonexocytotic release, intraneuronal storage and carrier-mediated transport, was tested in normoxic rat hearts. Vesicular storage was not stabilized by the calcium antagonists. In fact, verapamil, gallopamil, diltiazem and felodipine disturbed storage function, as indicated by an increased DOPEG release. Direct interaction with the noradrenaline carrier (uptake1) was demonstrated for verapamil, gallopamil, and diltiazem in a model of 3H-noradrenaline uptake. In conclusion, the calcium antagonists investigated inhibit noradrenaline release in ischemia by a mechanism which is different from blockade of neuronal calcium influx, and is rather due to an interaction with carrier-mediated transport of noradrenaline across the plasma membrane.

Abstract:The execution of positioning tasks by using image-based visual servoing can be easier if a trajectory planning mechanism exists. This paper deals with the problem of generating image plane trajectories (a trajectory is made of a path plus a time law) for tracked points in an eye-in-hand system which has to be positioned with respect to a fixed object. The generated image plane paths must be feasible i.e. they must be compliant with rigid body motion of the camera with respect to the object so as to avoid image jacobian singularities and local minima problems. In addition, the image plane trajectories must generate camera velocity screws which are smooth and within the allowed bounds of the robot. We show that a scaled 3D motion planning algorithm can be devised in order to generate feasible image plane trajectories.
Effects of antihypertensive therapy on factors mediating endothelium-dependent relaxation in rats treated chronically with L-NAME

Abstract:Objectives: To evaluate the relative participation of endothelium-derived factors mediating relaxation in response to acetylcholine in isolated mesenteric vascular beds from rats treated chronically with N(G)-nitro-L-arginine methylester (L-NAME); and to compare the consequences of prolonged treatment with either an angiotensin converting enzyme inhibitor or a calcium channel blocker on the components of acetylcholine-induced relaxation in this vascular preparation. Materials and methods: Male Sprague- Dawley rats were treated for 8 weeks with L-NAME (40 mg/kg per day), quinapril (10 mg/kg per day), diltiazem (100 mg/kg per day), L-NAME + quinapril and L-NAME + diltiazem. Systolic blood pressure was estimated by a tail-cuff plethysmograph. Relaxing responses to acetylcholine (10(-12) to 10(-8) mol) in mesenteric vascular beds precontracted with phenylephrine (10(-5) mol/l) were studied in the presence and absence of L-NAME (10(-5) mol/l), L-NAME + indomethacin (10(-5) mol/l) or L-NAME + indomethacin + potassium chloride (6 x 10(-5) mol/l). The area under the dose- response curve was used to calculate the approximate participation of nitric oxide, prostaglandins or endothelium-derived hyperpolarizing factor in the acetylcholine-induced relaxation. Results: Chronic administration of L-NAME increased blood pressure levels and vascular responsiveness to phenylephrine. Treatments with either quinapril or diltiazem reduced blood pressure levels and attenuated the increased response to phenylephrine. Relaxing responses to acetylcholine were similar in all groups, independently of the treatment received. The calculated participation of endothelium-derived hyperpolarizing factor in the acetylcholine-induced relaxation was higher than that of nitric oxide and prostaglandins in all groups, but was higher in L-NAME-treated than in untreated rats. In contrast, the participation of both nitric oxide and prostaglandins was higher in control than in L-NAME-treated rats. Quinapril increased the participation of prostaglandins in L-NAME-treated rats. Diltiazem increased the participation of nitric oxide in L-NAME-treated rats. Conclusions: The administration of L-NAME in Sprague-Dawley rats increased the production of endothelium-derived hyperpolarizing factor as a compensatory mechanism to maintain acetylcholine-induced relaxation. Antihypertensive therapy with either quinapril or diltiazem produced a selective redistribution of the endothelial factors mediating acetylcholine-induced relaxation.
