Abstract:The neurogenomic mechanisms mediating male–male reproductive cooperative behaviours remain unknown. We leveraged extensive transcriptomic and behavioural data on a neotropical bird species (Pipra filicauda) that performs cooperative courtship displays to understand these mechanisms. In this species, the cooperative display is modulated by testosterone, which promotes cooperation in non‐territorial birds, but suppresses cooperation in territory holders. We sought to understand the neurogenomic underpinnings of three related traits: social status, cooperative display behaviour and testosterone phenotype. To do this, we profiled gene expression in 10 brain nuclei spanning the social decision‐making network (SDMN), and two key endocrine tissues that regulate social behaviour. We associated gene expression with each bird's behavioural and endocrine profile derived from 3 years of repeated measures taken from free‐living birds in the Ecuadorian Amazon. We found distinct landscapes of constitutive gene expression were associated with social status, testosterone phenotype and cooperation, reflecting the modular organization and engagement of neuroendocrine tissues. Sex‐steroid and neuropeptide signalling appeared to be important in mediating status‐specific relationships between testosterone and cooperation, suggesting shared regulatory mechanisms with male aggressive and sexual behaviours. We also identified differentially regulated genes involved in cellular activity and synaptic potentiation, suggesting multiple mechanisms underpin these genomic states. Finally, we identified SDMN‐wide gene expression differences between territorial and floater males that could form the basis of 'status‐specific' neurophysiological phenotypes, potentially mediated by testosterone and growth hormone. Overall, our findings provide new, systems‐level insights into the mechanisms of cooperative behaviour and suggest that differences in neurogenomic state are the basis for individual differences in social behaviour.

A Model of Evaluating Integrating Emerging Technology into Formal Technology Curriculum

Abstract:The purpose of this study was to recognize a formative evaluation model for integrating emerging technology into formal technology curriculum. For coping with new contents brought by fast advancing technology, education system should provide ways to integrating that new information of emerging technology into our curriculum for preparing students with up-to-date knowledge. There is a need to find out how to establish an evaluation system for effects of integrating emerging technology into formal curriculum. Several evaluation models were reviewed and compared. Upon the evaluation characteristics, a systematic approach was applied for designing evaluating standards for the context, input, process, and output. A model with fourteen stages was proposed and conducted for research purpose. The experience of creating evaluation system for high scope curriculum innovation was described for supporting the proposed model. The evaluation target was first identified based on the evaluation project, high scope curriculum development. Evaluating items of each category were also identified. Ecological modulation of reproductive behaviour in the peacock blenny: a mini-review

Abstract:The inter-populational variation in the reproductive behaviour of the peacock blenny Salaria pavo (Risso 1810), particularly the influence of the ecologic environment, was reviewed in the present work. Two populations of this species inhabiting contrasting environments were studied: the Ria Formosa population, a coastal lagoon with sandy/muddy substrate located in the south of Portugal, and the Gulf of Trieste, an area presenting rocky substrate located in the northern Adriatic sea. The remarkable differences in the mating system and reproductive behaviour between the two populations (namely sex-role reversal and high frequency of alternative reproductive tactics (ARTs) at the Ria Formosa vs typical sex roles and low frequency of ARTs at the Gulf of Trieste) are interpreted as a plastic behavioural and physiological response to contrasting ecological conditions. Androgens, namely 11-ketotestosterone, seem to act as mediators of this response. The expression and activity of the enzyme aromatase in the brain also seems to play a key role in fine-tuning the behavioural output in all male morphs as well as females.

Abstract:In this mini-review, we summarize the brain distribution of aromatase, the enzyme catalyzing the synthesis of estrogens from androgens, and the mechanisms responsible for regulating estrogen production within the brain. Understanding this local synthesis of estrogens by neurons is pivotal as it profoundly influences various facets of social behavior. Neuroestrogen action spans from the initial processing of socially pertinent sensory cues to integrating this information with an individual's internal state, ultimately resulting in the manifestation of either pro-affiliative or - aggressive behaviors. We focus here in particular on aggressive and sexual behavior as the result of correct individual recognition of intruders and potential mates. The data summarized in this review clearly point out the crucial role of locally synthesized estrogens in facilitating rapid adaptation to the social environment in rodents and birds of both sexes. These observations not only shed light on the evolutionary significance but also indicate the potential implications of these findings in the realm of human health, suggesting a compelling avenue for further investigation.

An Event-related Potential Investigation on Associative Encoding and the Effects of Intra-list Semantic Similarity

Abstract:Event-related potentials were recorded as subjects were presented with pairs of words, one word at a time, to examine the electrocortical manifestations of association formation and the effect of intra-list semantic similarity. Two types of lists were presented: Same – all pairs belonged to the same semantic category; Different – all pairs belonged to a different semantic category. Subjects were told to memorize the pairs for a following paired associate recall test. Recall was better for the Different than Same lists. Subsequent recall was predicted by the amplitudes of a potential lasting throughout the epoch and the P555 to each word of a pair (likely reflecting stateand item-related encoding activity, respectively), as well as a late positive wave that occurred after the offset of the second word, which is thought to reflect association formation. A larger N425 was elicited by pairs in the Different than Same lists, likely reflecting semantic integration.

Abstract:Since Darwin proposed that human musicality evolved through sexual selection, empirical evidence has supported intersexual selection as one of the adaptive functions of artistic propensities. However, intrasexual competition has been overlooked. We tested their relative importance by investigating the relationship between the self-perceived talent/expertise in 16 artistic and 2 sports modalities and proxies of intersexual selection (i.e., mate value, mating and parenting efforts, sociosexuality, and number of sexual partners) and intrasexual competition (i.e., aggressiveness, intrasexual competitiveness) in heterosexuals. Participants were 82 Brazilian men, 166 Brazilian women, 146 Czech men, and 458 Czech women (Mage = 26.48, SD = 7.12). Factor analysis revealed five factors: Literary-arts (creative writing, humor, acting/theater/film, poetry, storytelling), Visual-arts (painting/drawing, sculpting, handcrafting, culinary arts, architecture design), Musical-arts (playing/instruments, singing, dance, whistling), Circus-arts (juggling, acrobatics), and Sports (individual, collective). Multivariate General Linear Model (GLM) showed more associations of the arts to intersexual selection in women and to intrasexual selection in men, and overall more relationships in women than in men. In women, literary and musical-arts were related to elevated inter- and intrasexual selections proxies, visual and circus-arts were related to elevated intersexual selection proxies, and sports were related to intrasexual selection proxies. In men, literary-arts and sports were related to elevated inter- and intrasexual selection proxies, musical-arts were related to intrasexual proxies, and circus-arts were related to intersexual proxies; visual-arts did not have predictors. Although present in both sexes, each sexual selection component has different relative importance in each sex. Artisticality functions to attract and maintain long/short-term partners, and to compete with mating rivals.

Abstract:Predators exert a powerful selective force, however, predator avoidance can conflict with other important activities such as attracting mates. Decisions over whether to court mates versus avoiding predators are vital to fitness, yet the mechanistic underpinnings of how animals manage such tradeoffs are poorly understood. Here, we investigate the flexibility of behaviour and gene regulation in response to a tradeoff between avoiding predators (survival) and courting potential mates (reproduction) in three-spined stickleback ( Gasterosteus aculeatus ). We compared behavioural and transcriptomic responses of male sticklebacks faced with a courtship opportunity and cues of a predator simultaneously with the responses of males faced with a courtship opportunity or cues of a predator alone, and found that males behaviourally compromised courtship in favour of predator avoidance when faced with a tradeoff between them. The need to manage this tradeoff elicited dynamic changes in brain gene expression, and sets of functionally connected genes were organized into discrete modules based on co-expression. Additionally, we found that behavioural flexibility in response to tradeoffs corresponded to flexibility in gene regulatory network structure. Combined, these results uncover the coordinated response by the brain to a fundamental ecological tradeoff, providing insight into the structure and function of genetic networks underpinning how animals make fitness-influencing decisions.

Abstract:Caste differentiation happens early in development to produce gynes as future colony germlines and workers as present colony soma. However, gynes need insemination to become functional queens, a transition that initiates reproductive role differentiation relative to unmated gynes. Here, we analyze the anatomy and transcriptomes of brains during this differentiation process within the reproductive caste of Monomorium pharaonis. Insemination terminated brain growth, whereas unmated control gynes continued to increase brain volume. Transcriptomes revealed a specific gene regulatory network (GRN) mediating both brain anatomy changes and behavioral modifications. This reproductive role differentiation GRN hardly overlapped with the gyne-worker caste differentiation GRN, but appears to be also used by distantly related ants where workers became germline individuals after the queen caste was entirely or partially lost. The genes corazonin and neuroparsin A in the anterior neurosecretory cells were overexpressed in individuals with reduced or nonreproductive roles across all four ant species investigated.
Dialogues in colour and behaviour - Integration of complex signalling traits and physiology

Abstract:Animal communication can be complex, often involving multiple static and dynamic traits. The extent to which these traits are correlated can elucidate their function as either redundant or multiple messages. Using the agamid lizard, Psammophilus dorsalis, as a model system we examined patterns of trait expression and the role of steroid hormones in mediating these traits during social interactions. We staged male-male interactions in the lab and measured the repertoire of display behaviour and colours, which change dynamically in the visible and ultraviolet ranges in different body regions. Additionally, we measured testosterone and corticosterone levels before and after the social trials. Our results show that within behaviour and colour trait categories, components were strongly correlated within individuals, suggesting either a shared physiological pathway or redundant information content. However, across trait categories, correlation patterns varied. The chromatic contrast of the (yellow) dorsal region of lizards was correlated with both body size and level of aggression, whereas the size of UV patches was correlated with body size only. We also found a negative association between baseline corticosterone levels, body size and dorsal yellow chromatic contrast, suggesting a mechanistic link between these traits. However, social interaction induced testosterone and corticosterone levels were uncorrelated with the expression of the dynamic behavioural and colour displays during the social interactions itself. Notably, the intensity of colour and behavioural displays of males were matched by their opponents. Overall, our results suggest that multiple signalling traits can ensure both redundancy as well as provide multiple messages to receivers, thus improving the robustness of information transfer, particularly during competitions which have high fitness consequences.

Abstract:Abstract Almost all life on earth is facing environmental change, and understanding how populations will respond to these changes is of urgent importance. One factor that is known to affect the speed by which a population can evolve when faced with changes in the environment is strong sexual selection. This increases the adaptive capacity of a population by increasing reproductive skew toward well-adapted (usually) males who will, on average, be best able to compete for matings. This effect could potentially be disrupted when males pursue alternative reproductive tactics (ARTs), whereby males within a species exhibit qualitatively different behaviors in their pursuit of matings. ARTs are diverse, but one common class is those expressed through condition-dependent polyphenism such that high-quality, well-adapted males compete aggressively for mates and low-quality, poorly adapted males attempt to acquire matings via other, nonaggressive behaviors. Here, using an individual-based modeling approach, we consider the possible impacts of ARTs on adaptation and evolutionary rescue. When the ART is simultaneous, meaning that low-quality males not only engage in contests but also pursue other tactics, adaptive capacity is reduced and evolutionary rescue, where a population avoids extinction by adapting to a changing environment, becomes less likely. This is because the use of the ART allows low-quality males to contribute more maladaptive genes to the population than would happen otherwise. When the ART is fixed, however, such that low-quality males will only use the alternative tactic and do not engage in contests, we find the opposite: adaptation happens more quickly and evolutionary rescue when the environment changes is more likely. This surprising effect is caused by an increase in the mating success of the highest quality males who face many fewer competitors in this scenario—counterintuitively, the presence of males pursuing the ART increases reproductive skew toward those males in the best condition.

Abstract:How does complex behavior arise from the genome across biological levels? Specific cell types have evolutionarily conserved functional roles in regulating and maintaining various aspects of social behavior. There is a long history of research demonstrating how specific cells in the hypothalamus and POA are critical in regulating social status and reproductive behavior. This cell function is dependent not just on one gene, but the entire gene expression network working within each cell. The brain is a heterogenous tissue made up of a broad diversity of cells. Targeting the cellular-level transcriptomes in socially relevant brain regions enables the identification of social status mediators. Single-nuclei RNA-sequencing (snRNA-seq) provides the resolution to identify cell-specific transcriptomes, with more practical benefits compared to single-cell approaches. Single-nucleus sequencing of neuronal tissue has remained fairly limited to model mammalian organisms. Here we performed snRNA-seq in the hypothalamus and POA of Astatotilapia burtoni, a fish system ideal for social neuroscience. Male A. burtoni are characterized by plastic phenotypes associated with social status, dominant and subordinate, with well-established differences in neural gene expression patterns and specific neuron morphology. This provides a novel opportunity to apply snRNA-seq in a complex tissue of a non-model system using hypothesis driven approaches. We show how changes in social status are linked to distinct transcriptomic profiles at the cellular level.

Abstract:Precopulatory courtship is a high-cost, non-well understood animal world mystery. Drosophila's (=D.'s) precopulatory courtship not only shows marked structural similarities with mammalian courtship, but also with human spoken language. This suggests the study of purpose, modalities and in particular of the power of this language and to compare it to human language. Following a mathematical symbolic dynamics approach, we translate courtship videos of D.'s body language into a formal language. This approach made it possible to show that D. may use its body language to express individual information - information that may be important for evolutionary optimization, on top of the sexual group membership. Here, we use Chomsky's hierarchical language classification to characterize the power of D.'s body language, and then compare it with the power of languages spoken by humans. We find that from a formal language point of view, D.'s body language is at least as powerful as the languages spoken by humans. From this we conclude that human intellect cannot be the direct consequence of the formal grammar complexity of human language.

Abstract:Behavior is a complex phenotype that is plastic and evolutionarily labile. The advent of genomics has revolutionized the field of behavioral genetics by providing tools to quantify the dynamic nature of brain gene expression in relation to behavioral output. The honey bee Apis mellifera provides an excellent platform for investigating the relationship between brain gene expression and behavior given both the remarkable behavioral repertoire expressed by members of its intricate society and the degree to which behavior is influenced by heredity and the social environment. Here, we review a linked series of studies that assayed changes in honey bee brain transcriptomes associated with natural and experimentally induced changes in behavioral state. These experiments demonstrate that brain gene expression is closely linked with behavior, that changes in brain gene expression mediate changes in behavior, and that the association between specific genes and behavior exists over multiple timescales, from physiological to evolutionary.

Abstract:Revealing the mechanisms underlying experience-dependent plasticity is a hallmark of behavioral neuroscience. While the study of social behavior has focused primarily on the neuroendocrine and neural control of social behaviors, the plasticity of these innate behaviors has received relatively less attention. Here, we review studies on mating-dependent changes to social behavior and neural circuitry across mammals, birds, and reptiles. We provide an overview of species similarities and differences in the effects of mating experiences on motivational and performative aspects of sexual behaviors, on sensory processing and preferences, and on the experience-dependent consolidation of sexual behavior. We also discuss recent insights into the neural mechanisms of and developmental influences on mating-dependent changes and outline promising approaches to investigate evolutionary parallels and divergences in experience-dependent plasticity.

Abstract:Sexual dimorphism, the divergence in morphological traits between males and females of the same species, is often accompanied by sex-biased gene expression. However, the majority of research has focused on species with conventional sex roles, where females have the highest energy burden with both egg production and parental care, neglecting the diversity of reproductive roles found in nature. We investigated sex-biased gene expression in Syngnathus typhle , a sex-role reversed species with male pregnancy, allowing us to separate two female traits: egg production and parental care. Using RNA sequencing, we examined gene expression across organs (brain, head kidney and gonads) at various life stages, encompassing differences in age, sex and reproductive status. While some gene groups were more strongly associated with sex roles, such as stress resistance and immune defence, others were driven by biological sex, such as energy and lipid storage regulation in an organ- and age-specific manner. By investigating how genes regulate and are regulated by changing reproductive roles and resource allocation in a model system with an unconventional life-history strategy, we aim to better understand the importance of sex and sex role in regulating gene expression patterns, broadening the scope of this discussion to encompass a wide range of organisms.

Transformations of neural representations in a social behaviour network

Abstract:Mating and aggression are innate social behaviours that are controlled by subcortical circuits in the extended amygdala and hypothalamus 1,2,3,4 . The bed nucleus of the stria terminalis (BNSTpr) is a node that receives input encoding sex-specific olfactory cues from the medial amygdala 5,6 , and which in turn projects to hypothalamic nuclei that control mating 7,8,9 (medial preoptic area (MPOA)) and aggression 9,10,11,12,13,14 (ventromedial hypothalamus, ventrolateral subdivision (VMHvl)), respectively 15 . Previous studies have demonstrated that male aromatase-positive BNSTpr neurons are required for mounting and attack, and may identify conspecific sex according to their overall level of activity 16 . However, neural representations in BNSTpr, their function and their transformations in the hypothalamus have not been characterized. Here we performed calcium imaging 17,18 of male BNSTpr Esr1 neurons during social behaviours. We identify distinct populations of female- versus male-tuned neurons in BNSTpr, with the former outnumbering the latter by around two to one, similar to the medial amygdala and MPOA but opposite to VMHvl, in which male-tuned neurons predominate 6,9,19 . Chemogenetic silencing of BNSTpr Esr1 neurons while imaging MPOA Esr1 or VMHvl Esr1 neurons in behaving animals showed, unexpectedly, that the male-dominant sex-tuning bias in VMHvl was inverted to female-dominant whereas a switch from sniff- to mount-selective neurons during mating was attenuated in MPOA. Our data also indicate that BNSTpr Esr1 neurons are not essential for conspecific sex identification. Rather, they control the transition from appetitive to consummatory phases of male social behaviours by shaping sex- and behaviour-specific neural representations in the hypothalamus.

Abstract:Many species, including humans exhibit a wide range of social behaviors that are crucial for the adaptation and survival of most species. Brain organization and function are shaped by genetic and environmental factors, although their precise contributions have been relatively understudied in the context of artificial selection. We used divergent lines of quail selected on their high versus low level of motivation to approach a group of conspecifics (S + and S-, respectively) to investigate the influence of genetic selection and early social environment on sociability. We observed distinct sex- and brain-region-specific expression patterns of three neuronal markers: mesotocin, and vasotocin, the avian homologues of mammalian oxytocin and vasopressin, as well as aromatase, the enzyme that converts androgens into estrogens. These markers displayed pronounced and neuroanatomically specific differences between S + and S- quail. Additionally, in a second experiment, we assessed the influence of early social environment on social skills in juvenile birds. Mixing S + and S- resulted in more S- males approaching the group without affecting the sociability of S + or other behaviors, suggesting that the early social environment may influence the results of genetic selection. In conclusion, the divergent quail lines offer a valuable model for unraveling the neuronal and behavioral mechanisms underlying social behaviors.

Cellular evolution of the hypothalamic preoptic area of behaviorally divergent deer mice

Abstract:Genetic variation is known to contribute to the variation of animal social behavior, but the molecular mechanisms that lead to behavioral differences are still not fully understood. Here, we investigate the cellular evolution of the hypothalamic medial preoptic area (MPOA), a brain region that plays a critical role in social behavior, across two sister species of deer mice (Peromyscus maniculatus and P. polionotus) with divergent social systems. These two species exhibit large differences in mating and parental care behavior across species and sex. Using single-nucleus RNA-sequencing, we build a cellular atlas of the MPOA for males and females of both Peromyscus species. We identify four cell types that are differentially abundant across species, two of which may account for species differences in parental care behavior. Our data further implicate two sex-biased cell types to be important for the evolution of sex-specific behavior. Finally, we show a remarkable reduction of sex-biased gene expression in P. polionotus, a monogamous species that also exhibits reduced sexual dimorphism in parental care behavior. Our MPOA atlas is a powerful resource to investigate how molecular neuronal traits may be evolving to give rise to innate differences in social behavior across animal species.
