Abstract:To optimally penetrate biological hydrogels such as mucus and the tumor interstitial matrix, nanoparticles (NPs) require physicochemical properties that would typically preclude cellular uptake, resulting in inefficient drug delivery. Here, we demonstrate that (poly(lactic-co-glycolic acid) (PLGA) core)-(lipid shell) NPs with moderate rigidity display enhanced diffusivity through mucus compared with some synthetic mucus penetration particles (MPPs), achieving a mucosal and tumor penetrating capability superior to that of both their soft and hard counterparts. Orally administered semi-elastic NPs efficiently overcome multiple intestinal barriers, and result in increased bioavailability of doxorubicin (Dox) (up to 8 fold) compared to Dox solution. Molecular dynamics simulations and super-resolution microscopy reveal that the semi-elastic NPs deform into ellipsoids, which enables rotation-facilitated penetration. In contrast, rigid NPs cannot deform, and overly soft NPs are impeded by interactions with the hydrogel network. Modifying particle rigidity may improve the efficacy of NP-based drugs, and can be applicable to other barriers.

Abstract:The enrichment of M2-phenotype tumor-associated macrophages (TAMs) is the main factor to create an immunosuppressive microenvironment, which seriously attenuates antitumor treatment. Herein, we explored the capability of rigid nanoparticles to reprogram TAMs to the M1-like phenotype via mechanical force. We designed and synthesized amphiphilic copolymer PLA- b -PItEG initiated by the HO-Pd(II) complex. In contrast to polyethylene glycol, the hydrophilic PItEG segments have a helical conformation, which endows the self-assembled PLA- b -PItEG nanoparticles with stiff surface characteristics of higher Young's modulus and hydrophobicity. The rigid nanoparticles displayed strong interaction with TAMs to produce greater deformation of cell membrane and activated mechanosensitive potassium ion channel. Consequently, rigid nanoparticles exerted a positive effect on the proinflammatory polarization of TAMs. The expression level of CD86, a typical M1 macrophage marker, was increased by 60% compared with that in the soft PEG-NP group while CD206, a typical M2 macrophage marker, was decreased. Furthermore, several M1-related cytokines such as TNF-α, IL12p40, IL-6 and IL-1β were upregulated. In vitro and in vivo immune responses triggered by rigid nanoparticles could significantly inhibit B16F10 tumor growth with 71.44% tumor inhibitory rate. This study offers new insight into the use of mechanical forces for tumor immunotherapy.

Abstract:Cells can sense the mechanical stimulation of nanoparticles (NPs) and then regulate the cellular uptake process. The enhanced endocytosis efficiency can improve the concentration of NPs in tumor cells significantly, which is the key prerequisite for achieving efficient biological performance. However, the preparation methods of NPs with flexible and tunable stiffness are relatively limited, and the impact of stiffness property on their interaction with tumor cells remains unclear. In this study, soft liquid metal (LM) core was coated with hard silica layer, the obtained core-shell NPs with a wide range of Young's modulus (130.5 +/- 25.6 MPa - 1729.2 +/- 146.7 MPa) were prepared by adjusting the amount of silica. It was found that the NPs with higher stiffness exhibited superior cellular uptake efficiency and lysosomal escape ability compared to the NPs with lower stiffness. The silica layer not only affected the stiffness, but also improved the photothermal stability of the LM NPs. Both in vitro and in vivo results demonstrated that the NPs with higher stiffness displayed significantly enhanced tumor hyperthermia capability. This work may provide a paradigm for the preparation of NPs with varying stiffness and offer insights into the role of the mechanical property of NPs in their delivery.
Non-specific binding and steric hindrance thresholds for penetration of particulate drug carriers within tumor tissue

Abstract:Therapeutic nanoparticles (NPs) approved for clinical use in solid tumor therapy provide only modest improvements in patient survival, in part due to physiological barriers that limit delivery of the particles throughout the entire tumor. Here, we explore the thresholds for NP size and surface poly(ethylene glycol) (PEG) density for penetration within tumor tissue extracellular matrix (ECM). We found that NPs as large as 62nm, but less than 110nm in diameter, diffused rapidly within a tumor ECM preparation (Matrigel) and breast tumor xenograft slices ex vivo. Studies of PEG-density revealed that increasing PEG density enhanced NP diffusion and that PEG density below a critical value led to adhesion of NP to ECM. Non-specific binding of NPs to tumor ECM components was assessed by surface plasmon resonance (SPR), which revealed excellent correlation with the particle diffusion results. Intravital microscopy of NP spread in breast tumor tissue confirmed a significant difference in tumor tissue penetration between the 62 and 110nm PEG-coated NPs, as well as between PEG-coated and uncoated NPs. SPR assays also revealed that Abraxane, an FDA-approved non-PEGylated NP formulation used for cancer therapy, binds to tumor ECM. Our results establish limitations on the size and surface PEG density parameters required to achieve uniform and broad dispersion within tumor tissue and highlight the utility of SPR as a high throughput method to screen NPs for tumor penetration.
Shape switching of CaCO3-templated nanorods into stiffness-adjustable nanocapsules to promote efficient drug delivery

Abstract:Geometry and mechanical property have emerged as important parameters in designing nanocarriers, in addition to their size, surface charge, and hydrophilicity. However, inconsistent and even contradictory demands regarding the shape and stiffness of nanoparticles have been noted in blood circulation, tumor accumulation, and tumor cell internalization. Herein, CaCO3 nanorods (NRs) with an aspect ratio of around 2.4 are assembled with hyaluronic acid (HA) hydrogel layers to prepare CaCO3@HA NRs. The rod geometry enables lower recognition by macrophages and higher extravasation into tumor tissues than the spherical counterpart. In response to the slightly acidic tumor matrix, the acid-labile removal of CaCO3 templates achieves shape switching into spherical HA nanocapsules (NCs). The shape switchable CaCO3@HA NRs show significantly higher uptake and cytotoxicities to 4T1 cells than CaCO3-Si@HA NRs with silica layers on CaCO3 cores to inhibit shape switching. In addition, HA NCs with 2 - 8 layers of HA hydrogels exhibit stiffness from 1.85 to 12.3 N/m, and the assembly of 4 layers shows 2- to 3-fold higher cellular uptake than those of other NCs. The shape shift satisfies long-term blood circulation of NRs, and the resulting stiffness-adjustable NCs promote tissue infiltration and intracellular accommodation, resulting in a 4-fold higher drug accumulation in tumors. The CaCO3@HA NR treatment significantly suppresses tumor growth; prolongs animal survival; inhibits lung metastasis; and eliminates systemic toxicities to blood, liver, kidney, and heart tissues. This study achieves a comprehensive understanding of the shape and stiffness effects and demonstrates a hierarchical targeting strategy to address the multiple delivery barriers for chemotherapeutic agents. STATEMENT OF SIGNIFICANCE: The different barriers involved in the drug delivery pathway have inconsistent and even contradictory demands on the shape and stiffness of nanoparticles. In the current study, in situ switching of nanorods (NRs) into spherical nanocapsules (NCs) in tumor tissues is proposed to address these dilemmas. The NR shape ensures long-term blood circulation and high tumor tissue accumulation, while the in situ switching into NCs promotes tissue infiltration and cellular internalization. NCs with different numbers of hydrogel layers also provide a robust system wherein NC stiffness is controlled as a single variable to study stiffness-dependent cellular behaviors. Thus, this straightforward design offers a comprehensive understanding of how the shape and stiffness of nanocarriers affect their biological pathways.
Penetration of Nanoparticles Across a Lipid Bilayer: Effects of Particle Stiffness and Surface Hydrophobicity

Abstract:The cellular uptake of nanoparticles (NPs) has drawn significant attention due to their great importance and potential in drug delivery, bioimaging, and specific targeting. Here, we conduct a computational study on the translocation process of soft nanoparticles with different elasticities and surface hydrophobicities through a lipid bilayer membrane. It is shown that the translocation abilities of hydrophilic NPs can be enhanced by increasing their stiffness, while the penetrability of hydrophobic NPs is weakened by increasing the particle stiffness. The free energy analysis indicates that rigid hydrophilic NPs and soft hydrophobic NPs encounter lower energy barriers during penetration. In direct translocation, different deformation modes are observed for NPs with different surface hydrophobicities during cellular internalization. Further, deformation analysis demonstrates that hydrophilic NPs are flattened in the membrane plane, while hydrophobic NPs are elongated along the membrane norm during penetration. We conclude that the elasticity of NPs has an obvious impact on their ability to penetrate across the lipid bilayer membrane through different morphological responses of hydrophilic and hydrophobic NPs. These results shed light on the coupled effects of particle elasticity and surface hydrophobicity on the cellular uptake of elastic NPs, which may provide useful guidelines for designing effective nanocarrier systems for drug delivery.

Abstract:Poly(ethylene glycol) (PEG) is usually used to protect nanoparticles from rapid clearance in blood. The effects are highly dependent on the surface PEG density of nanoparticles. However, there lacks a detailed and informative study in PEG density and in vivo drug delivery due to the critical techniques to precisely control the surface PEG density when maintaining other nano-properties. Here, we regulated the polymeric nanoparticles' size and surface PEG density by incorporating poly(ε-caprolactone) (PCL) homopolymer into poly(ethylene glycol)-block-poly(ε-caprolactone) (PEG–PCL) and adjusting the mass ratio of PCL to PEG–PCL during the nanoparticles preparation. We further developed a library of polymeric nanoparticles with different but controllable sizes and surface PEG densities by changing the molecular weight of the PCL block in PEG–PCL and tuning the molar ratio of repeating units of PCL (CL) to that of PEG (EG). We thus obtained a group of nanoparticles with variable surface PEG densities but with other nano-properties identical, and investigated the effects of surface PEG densities on the biological behaviors of nanoparticles in mice. We found that, high surface PEG density made the nanoparticles resistant to absorption of serum protein and uptake by macrophages, leading to a greater accumulation of nanoparticles in tumor tissue, which recuperated the defects of decreased internalization by tumor cells, resulting in superior antitumor efficacy when carrying docetaxel.
Morphological and Mechanical Determinants of Cellular Uptake of Deformable Nanoparticles

Abstract:Understanding the interactions of nanoparticles (NPs) with cell membranes and regulating their cellular uptake processes are of fundamental importance to the design of drug delivery systems with minimum toxicity, high efficiency and long circulation time. Employing the procedure of coarse-graining, we built an elastically deformable NP model with tunable morphological and mechanical properties. We found that the cellular uptake of deformable NPs depends on their shape: an increase in the particle elasticity significantly slows the uptake rate of spherical NPs, slightly retards that of prolate NPs, and promotes the uptake of oblate NPs. The intrinsic mechanisms have been carefully investigated through analysis of the endocytic mechanisms and free energy calculations. These findings provide unique insights into how deformable NPs penetrate across cell membranes and offer novel possibilities for designing effective NP-based carriers for drug delivery.

Abstract:Tumor penetration is a critical determinant of the therapy efficacy of nanomedicines. However, the dense extracellular matrix (ECM) in tumors significantly hampers the deep penetration of nanomedicines, resulting in large drug-untouchable areas and unsatisfactory therapy efficacy. Herein, we synthesized a third-generation PAMAM-cored multiarm copolymer and modified the polymer with collagenase to enhance its tumor penetration. Each arm of the copolymer was a diblock copolymer of poly(glutamic acid)-b-poly(carboxybetaine), in which the polyglutamic acid block with abundant side groups was used to link the anticancer agent doxorubicin through the pH-sensitive acylhydrazone linkage, and the zwitterionic poly(carboxybetaine) block provided desired water solubility and anti-biofouling capability. The collagenase was conjugated to the ends of the arms via the thiol-maleimide reaction. We demonstrated that the polymer-bound collagenase could effectively catalyze the degradation of the collagen in the tumor ECM, and consequently augmented the tumor penetration and antitumor efficacy of the drug-loaded polymers.

Solid Tumor Penetration by Integrin-Mediated Pegylated Poly(trimethylene Carbonate) Nanoparticles Loaded with Paclitaxel.

Abstract:Limited penetration of antineoplastic agents is one of the contributing factors for chemotherapy failure of many solid tumors. In order to enhance drug penetration into solid cancer, especially, into the avascular regions inside tumors, we proposed cyclic RGD peptide functionalized PEGylated poly(trimethylene carbonate) nanoparticles (c(RGDyK)-NP). By integrin-mediated transcytosis and enhanced drug permeation, c(RGDyK)-NP could access the neoplastic cells distant from blood vessels, and consequently, avoiding the capability of cancer regeneration from these tumor cells. In the present study, the solid tumor penetration, homing specificity and anticancer efficacy were evaluated both on the ex vivo 3D tumor spheroids and on the subcutaneous xenograft mice model. In comparison with conventional nanoparticles (NP/PTX) and Taxol, c(RGDyK)-NP/PTX showed the strongest penetration and accumulation into 3D tumor spheroids, a marked tumor-homing specificity in vivo and the greatest tumor growth inhibitory effect in vitro and in vivo. Histochemistry analysis revealed that no obvious histopathological abnormalities or lesions were observed in major organs after intravenous administration with the treatment doses. In conclusion, cyclic RGD peptide-conjugated PEG-PTMC nanoparticle could facilitate drug penetration and accumulation in tumor tissues and may be a promising vehicle for enhancing the chemotherapy of solid cancers.
The Effect of Hydrophilic Chain Length and Irgd on Drug Delivery from Poly(ε-Caprolactone)-poly(n-vinylpyrrolidone) Nanoparticles.

Abstract:Poly(ε-caprolactone)-b-Poly(N-vinylpyrrolidone) (PCL-b-PVP) copolymers with different PVP block length were synthesized by xanthate-mediated reverse addition fragment transfer polymerization (RAFT) and the xanthate chain transfer agent on chain end was readily translated to hydroxy or aldehyde for conjugating various functional moieties, such as fluorescent dye, biotin hydrazine and tumor homing peptide iRGD. Thus, PCL-PVP nanoparticles were prepared by these functionalized PCL-b-PVP copolymers. Furthermore, paclitaxel-loaded PCL-PVP nanoparticles with satisfactory drug loading content (15%) and encapsulation efficiency (>90%) were obtained and used in vitro and in vivo antitumor examination. It was demonstrated that the length of PVP block had a significant influence on cytotoxicity, anti-BSA adsorption, circulation time, stealth behavior, biodistribution and antitumor activity for the nanoparticles. iRGD on PCL-PVP nanoparticle surface facilitated the nanoparticles to accumulate in tumor site and enhanced their penetration in tumor tissues, both of which improved the efficacy of paclitaxel-loaded nanoparticles in impeding tumor growth and prolonging the life time of H22 tumor-bearing mice.

Abstract:The electrospraying process has been exploited in recent times to prepare micro- and nano-scaled polymer based particulate structures for a range of biomedical and pharmaceutical applications. Conventionally, three main parameters (applied voltage, flow rate and base solvent in processing polymeric media) are used to manipulate their morphologies during preparation (size, surface and porosity). In this work, an unconventional approach is demonstrated to yield biocompatible core-shell encapsulated (poly-epsilon-caprolactone (PCL) and silicon oil) particles with a range of structural morphologies (Le, spherical, hemi-spherical and oblate shaped). Retaining its advantageous single-step fabrication aspect and by using two co-axial needles, a range of collection media was used to demonstrate shape and size variation (from similar to 28-48 mu m) using optical and scanning electron microscopy. The morphological variation is directly linked to surface tension of the collection liquid. In addition, the non-volatile and flexible nature of the oil core plays a role in geometry formation. In vitro release from these structures demonstrates significant and non-significant variations; providing pathways to control release as well mimic release profile but alter other biomedical process (e.g. ultrasound). These findings demonstrate an alternative approach to manipulate the shape of bi-phasic structures which is an important functional property in several biomedical remits (e.g. drug delivery and ultrasound).[GRAPHICS]. Abstract:Novel methods used in studying the proliferation activity and cell kinetics of malignant neoplasms such as: the in vivo and in vitro bromodeoxyuridine labeling, the flow cytometric DNS analysis, the immunohistochemical demonstration of the so called proliferation associated antigens and the counting of nucleolar organized regions are briefly introduced. The data obtainable by these methods are reviewed and critically discussed. The prognostic value of the obtained data and their utility in planning and monitoring of cancer patients' therapy are also discussed.

Abstract:The intrinsic pathological microenvironment of tumor tissue enforces barriers to the passive diffusion of nanomedicine, which results in inefficient tumor penetration of drugs and unsatisfactory therapeutic efficacy. Various strategies have been developed to improve the tumor penetration of nanomedicine, but they mostly overcome the barriers separately at different steps in a passive diffusion process. Here, the development of a large polymeric nanocarrier, DAT-PPED&F (≈180 nm), is reported, which comprises a flowable polyphosphoester core, ferrimagnetic nanocubes, and a tumor extracellular pH-sensitive transactivator of transcription (TAT). Compared with a polylactic acid-based nanocarrier with a rigid core, this deformable DAT-PPED&F with a similar diameter exhibits efficient penetration into the deep tumor tissue under magnetic actuation and an enhanced reactivation rate of the pHe-sensitive TAT. Therefore, DAT-PPED&F is able to efficiently deliver doxorubicin into most tumor cells in vivo, and the superior anticancer effect indicates the potential of DAT-PPED&F as a universal, responsive, and active nanocarrier to deliver various hydrophobic drugs into the deep tumor tissue.

Abstract:Core-shell nanoparticles were prepared from di-block copolymer of methoxy poly(ethylene glycol)-polycaprolactone (MePEG-PCL) and tri-block copolymer of polycaprolactone-poly(ethylene glycol)-polycaprolactone (PCL-PEG-PCL). The MePEG-PCL copolymers form nanoparticles with PEG "brush" on their surfaces and PCL-PEG-PCL copolymers form nanoparticles with a "mushroom-like" structure on their surfaces in aqueous solution. The morphology and size of nanoparticles were measured by field emission scanning electron microscopy (FESEM) and laser light scattering (LLS). All the nanoparticles are in spherical shape and the sizes are less than 200 nm. The sizes of the nanoparticles increases with increasing PCL segment length. The drug-loading content results showed that the optimal feeding ratio of paclitaxel to copolymer is dependent upon the copolymer composition and 5% is a suitable feeding ratio. The in vitro release behavior exhibits a sustained release manner and is affected by copolymer composition. Experimental results showed that cells would prefer to attach to more hydrophobic polymers. Comparing between MePEG-PCL and PCL-PEG-PCL of similar hydrophobicity, more HepG2 cells have attached to the MePEG-PCL copolymer films because a denser PEG layer was formed on the surfaces of PCL-PEG-PCL copolymers. In vitro cellular uptake experimental results indicated that HepG2 cells prefer smaller nanoparticles with the same PEG configuration on their surfaces. Nanoparticle Structure Transformation of Mpeg Grafted Chitosan with Rigid Backbone Induced by Α-Cyclodextrin

Abstract:This paper presented an interesting nanoparticle-based drug delivery system with morphology transition behavior depending on the content of exposed PEG chain on the particle surface, which is adjustable by addition of different amount of cyclodextrin (α-CD). The effect of α-CD inclusion to the self-assembly behavior of methoxy polyethylene glycol (mPEG) grafted chitosan (CS) was studied. The results showed that the mPEG grafted chitosan (mPEG-g-CS) forms self-assembled nanoparticles with either micelle or hollow sphere morphology depending on the ratio of α-CD to mPEG, as characterized by atomic force microscopy (AFM), transmission electron microscopy (TEM), and X-ray diffraction (XRD). Their sizes and zeta potential increased from 257.6 nmto 768.2nm and from +4.5mV to +11.6mV, respectively, with the increasing amount of α-CD. The correlation between zeta potential and particle size of α-CD/mPEG-g-CS nanoparticles indicated varied PEG density on surface of nanoparticles. Based on the above experimental observations, a likely mechanism for the morphological transition of the rod-coil graft copolymer mPEG-g-CS was proposed.

Abstract:To date, the role of elasticity in drug delivery remains elusive due to the inability to measure microscale mechanics and alter rheology without affecting chemistry. Herein, we describe the in vitro cellular uptake and in vivo tumor uptake of nanolipogels (NLGs). NLGs are composed of identical lipid bilayers encapsulating an alginate core, with tunable elasticity. The elasticity of NLGs was evaluated by atomic force microscopy, which demonstrated that they exhibit Young’s moduli ranging from 45 ± 9 to 19,000 ± 5 kPa. Neoplastic and non-neoplastic cells exhibited significantly greater uptake of soft NLGs (Young’s modulus <1.6 MPa) relative to their elastic counterparts (Young’s modulus >13.8 MPa). In an orthotopic breast tumor model, soft NLGs accumulated significantly more in tumors, whereas elastic NLGs preferentially accumulated in the liver. Our findings demonstrate that particle elasticity directs tumor accumulation, suggesting that it may be a design parameter to enhance tumor delivery efficiency.
