Abstract:This study aimed to evaluate the effect of chemical modifications of the structure of active compounds on the skin permeation and accumulation of ibuprofen [IBU] from the acrylic pressure-sensitive adhesive used as a drug-in-adhesives matrix type transdermal patch. The active substances tested were ibuprofen salts obtained by pairing the ibuprofen anion with organic cations, such as amino acid isopropyl esters. The structural modification of ibuprofen tested were Ibuprofen sodium salt, [GlyOiPr][IBU], [AlaOiPr][IBU], [ValOiPr][IBU], [SerOiPr][IBU], [ThrOiPr][IBU], [(AspOiPr)2][IBU], [LysOiPr][IBU], [LysOiPr][IBU]2, [PheOiPr][IBU], and [ProOiPr][IBU]. For comparison, the penetration of unmodified ibuprofen and commercially available patches was also investigated. Thus, twelve transdermal patches with new drug modifications have been developed whose adhesive carrier is an acrylate copolymer. The obtained patches were characterized for their adhesive properties and tested for permeability of the active substance. Our results show that the obtained ibuprofen patches demonstrate similar permeability to commercial patches compared to those with structural modifications of ibuprofen. However, these modified patches show an increased drug permeability of 2.3 to even 6.4 times greater than unmodified ibuprofen. Increasing the permeability of the active substance and properties such as adhesion, cohesion, and tack make the obtained patches an excellent alternative to commercial patches containing ibuprofen.

Abstract:OBJECTIVE To investigate the release behavior of drugs with different physico-ehemical properties in styrene-isoprene-styrene(SIS)hot-melt pressure sensitive adhesive(HMPSA),and to evaluate the applicability of HMPSA in dermal patches. METHODS Adhesive dispersion-type patches which containing cryptotanshinone,α-asarone,caffeine,quercetin, baicalin,sinomenine hydrochloride,lappaconitine hydrobromide were prepared using SIS HMPSA,respectively.The release experiments was carried on single chamber diffusion cell using 60% ethanol solution as release media.The effects of the type of adhesive and the content of drugs on release rate were evaluated.The compatibility of HMPSA with cryptotanshinone and baicalin was also studied by differential scanning calorimetry(DSC).RESULTS The drug release behavior of HMPSA patches followed Higuchi equation.The release rate of patches was related with the drug properties.High lipophilic drugs were showed higher release rates.The HMPSA had good compatibility with high lipophilie cryptotanshinone,while low lipophilic baicalin was incompatibility.CONCLUSION The SIS HMPSA is a suitable and compatible pressure sensitive adhesive for the development of transdermal drug delivery systerns for high lipophilic drugs.
A semi-interpenetrating network acrylic pressure-sensitive adhesive for efficient transdermal application with high cohesion and adhesion

Abstract:The application of pressure-sensitive adhesives (PSAs) in transdermal drug delivery systems (TDDSs) is limited. Insufficient cohesion in TDDS can lead to cold flow, patch displacement, dose inaccuracies, and the 'dark ring' phenomenon. To address these issues, we developed a semi-interpenetrating network (semi-IPN) PSA using a 2-hydroxyethylacrylate-modified acrylic copolymer as the linear polymer and poly[tri(propylene glycol) diacrylate] as the cross-linked network. Semi-IPN PSAs have better mechanical properties compared to traditional PSAs. The cohesion of the semi-IPN PSA was 31 times and 1.6 times that of commercially available DURO-TAK®87-2287 and DURO-TAK® 87–4098, respectively, and the cold flow of the semi-IPN PSA was significantly reduced. Furthermore, the semi-IPN PSA adhered to the human forearm for longer time and did not exhibit the 'dark ring' phenomenon after removal. When loaded with flurbiprofen, the semi-IPN PSA was found to maintain high levels of drug release and skin permeation amount, and flurbiprofen loading did not affect its superior cohesive performance. In conclusion, the innovative application of the semi-IPN PSA for TDDS showed excellent mechanical properties, effective adhesion to the skin without irritation, and highly effective drug release. These findings indicate that the use of the semi-IPN is both simple and highly effective for TDDS.

Abstract:This study aimed to investigate the potential of photoreactive acrylate patches as systems for transdermal drug delivery, in particular, using more renewable alternatives and more environmentally friendly synthesis routes of transdermal patches. Therefore, the aim of this study was to develop a transdermal patch containing ibuprofen and investigate its performance in vitro through the pigskin. Transparent patches were prepared using four acrylate copolymers with an incorporated photoinitiator. Two types of transdermal patches based on the photocrosslinking acrylic prepolymers with isobornyl methacrylate as biocomponent and monomer increasing Tg (“hard”) were manufactured. The obtained patches were characterized for their adhesive properties and tested for permeability of the active substance. It turns out that patches whose adhesive matrix is photoreactive polyacrylate copolymers have a higher cohesion than patches from commercial adhesives, while the modification of the copolymers with isobornyl methacrylate resulted in an improvement in adhesion and tack. This study demonstrates the feasibility of developing photoreactive acrylic-based transdermal patches that contain biocomponents that can deliver a therapeutically relevant dose of ibuprofen.

Abstract:BACKGROUND: The purpose of this work was to develop novel pressure-sensitive adhesives (PSAs) for transdermal drug-delivery systems (TDDS) with proper adhesive properties, hydrophilicity, biocompatibility and high drug loading.METHOD: Polyethyleneglycol-modified polyurethane PSAs (PEG-PU-PSAs) were synthesized by prepolymerization method with PEG-modified co-polyether and hexamethylene diisocyanate. The effects of reaction temperature, catalyst, ratios of NCO/OH, co-polyether composition, and chain extender were investigated. Drug loading was studied by using thiamazole (hydrophilic drug), diclofenac sodium (slightly hydrophilic drug), and ibuprofen (lipophilic drug) as model drugs. In vitro drug-release kinetics obtained with Franz diffusion cell and dialysis membrane.RESULTS: The results showed that when reaction temperature at 80 degrees C, weight percentage of stannous octoate as catalyst at 0.05%, ratio of NCO/OH at 2.0-2.2, ratio of PEG/polypropylene glycol (PPG)/polytetramethylene ether glycol (PTMG) at 30/25-30/50-55, and weight percentage of glycol as chain extender at 4.5%, PEGPU-PSAs synthesized performed well on adhesive properties. Actually, PEG on the main chain of the PU could improve the hydrophilicity of PSAs, whereas PPG and PTMG could offer proper adhesive properties. Skin compatibility test on volunteers indicated that PEG-PU-PSAs would not cause any skin irritations. All the model drugs had excellent stabilizations in PEG-PU-PSAs. In vitro drug-release kinetics demonstrated that the drug release depended on drug-loading level and solubility of the drug.CONCLUSION: These experimental results indicated that PEG-PU-PSAs have good potential for applications in TDDS.

Abstract:The aim of this study was to design a tulobuterol (TUL) patch with good penetration behavior and mechanical properties. Particular attention was paid to the effect of transdermal permeation enhancers on the release process of metal ligand-based acrylic pressure-sensitive adhesive (AA-NAT/Fe3+). The type and dosage of the enhancers were screened by in vitro transdermal penetration in rat skin. The optimized formulation was evaluated in a pharmacokinetic study in rats. Furthermore, the molecular mechanism by which Azone (AZ) improves the release rate of TUL from AA-NAT/Fe3+ was investigated by FT-IR, shear strength test, rheological study, and molecular simulation. As a result, the optimized formula using AA-NAT/Fe3+ showed better mechanical properties compared to commercial products. Meanwhile, the AUC0-t and Cmax of the optimized patch were 1045 ± 89 ng/mL·h and 106.8 ± 28.5 ng/mL, respectively, which were not significantly different from those of the commercial product. In addition, AZ increased the mobility of the pressure-sensitive adhesive (PSA) rather than decreasing the drug-PSA interaction, which was the main factor in enhancing TUL release from the patch. In conclusion, a TUL transdermal drug delivery patch was successfully developed using metal-coordinated PSA, and a reference was provided for the design of metal-coordinated acrylic PSA for transdermal patch delivery applications.

Formulation Development and Molecular Mechanism Characterization of Long-Acting Patches of Asenapine for Efficient Delivery by Combining API-ILs Strategy and Controlled-Release Polymers

Abstract:The objective of our study, which combined API-ILs strategy and controlled-release polymers, was to prepare a 72 h long-acting drug-in-adhesive patch for optimum delivery of asenapine (ASE). Special attention was paid to the permeation promotion mechanism and the controlled release behavior of ASE-ILs in pressure sensitive adhesives (PSA). Formulation factors were investigated by ex vivo transdermal experiments. The optimized patch was evaluated by pharmacokinetics study and skin irritation test. The obtained formulation was as follows, 15% w/w ASE-MA (about 1136 μg/cm2 ASE, 413 μg/cm2 MA), AACONH2 (Amide adhesive) as the matrix, 80 μm thickness, backing film of CoTran™ 9733. The optimized patch displayed satisfactory ex vivo and in vivo performance with Q 72 h of 620 ± 44 µg/cm2 and Fabs of 62.4%, which utilization rate (54.6%) was significantly higher than the control group (38.3%). By using the classical shake flask method, 13C NMR, DSC, and FTIR, the physicochemical properties and structure of ILs were characterized. log Do/w, ATR-FTIR, Raman, and molecular dynamics simulation results confirmed that ASE-MA (MA: 3-Methoxypropionic acid) had appropriate lipophilicity, and affected lipid fluidity as well as the conformation of keratin to improve the skin permeation. The FTIR, MDSC, rheology, and molecular docking results revealed that hydrogen bond (H-bond), were formed between ASE-MA and PSA, and the drug increased the molecular mobility of polymer chains. In summary, the 72 h long-acting patch of ASE was successfully prepared and it supplied a reference for the design of long-acting patches with ASE.

Abstract:In this study, the influence of the ratio of silicone (Si) to acrylic pressure-sensitive adhesive (PSA), polyvinyl pyrrolidone (PVP), and lauryl alcohol (LA) % (wt/wt) on the properties of a drug in adhesive patch containing 4% (wt/wt) fentanyl as model drug was evaluated. The dependent variables selected were drug solubility, in vitro drug release in the platforms as well as adhesion properties including peel strength and tack value. By using the central composite design of Design Expert software, it was found that the effect of each factor was different, yet all had influenced dependent variables significantly (p < .05). Quadratic model generated for various response variables using backward regression analysis was found to be statistically significant (p < .05). It was deduced that the presence of PVP and Si displayed similar trends on drug solubility and release. Each role played by Si with LA and PVP in release rate was separately investigated, and it was found that the presence of PVP and LA in lowering the amount of drug released was more dominant compared with that of Si. The release patterns at the early and later stages follow the Higuchi and semiempirical models, respectively. Effect of PVP as well as Si and LA were similar on tack value. The influence of LA compared to peeling characteristics of Si system was more pronounced.

Abstract:Poor skin adhesion and mechanical properties are common problems of pressure-sensitive adhesive (PSA) in transdermal drug delivery system (TDDS). Its poor water compatibility also causes the patch to fall off after sweating or soaking in the application site. To solve this problem, poly (2-Ethylhexyl acrylate- co - N -Vinyl-2-pyrrolidone- co - N -(2-Hydroxyethyl)acrylamide) (PENH), a cross-linked pyrrolidone polyacrylate PSA, was designed to improve the adhesion and water resistance of PSA through electrostatic force and hydrogen bonding system. The structure of PENH was characterized by 1 H NMR, FTIR, DSC, and other methods. The mechanism was studied by FTIR, rheological test, and molecular simulation. The results showed that the PENH patch could adhere to human skin for more than 10 days without cold flow, and it could still adhere after sweating or water contact. In contrast, the commercial PSA Duro-Tak® 87-4098 and Duro-Tak® 87-2852 fell off completely on the 3rd and 6th day, respectively, and Duro-Tak® 87-2510 showed a significant dark ring on the second day. Mechanism studies have shown that the hydrogen bond formed by 2-ethylhexyl acrylate (2-EHA), N-vinyl-2-pyrrolidinone (NVP), and N -(2-Hydroxyethyl)acrylamide (HEAA) enhances cohesion, the interaction with skin improves skin adhesion, and the electrostatic interaction with water or drug molecules enhances the ability of water absorption and drug loading. Due to the synergistic effect of hydrogen bonds and electrostatic force, PENH can maintain high cohesion after drug loading or water absorption. PENH provides a choice for the development of water-compatible patches with long-lasting adhesion. Statement of Significance Based on the synergistic effect of hydrogen bonding and electrostatic force, a hydrogen-bonded, cross-linked pyrrolidone acrylate pressure-sensitive adhesive for transdermal drug delivery was designed and synthesized, which has high adhesion and cohesive strength and is non-irritating to the skin. The patch can be applied on the skin surface continuously for more than 10 days without the phenomenon of "dark ring", and the patch can remain adherent after the patient sweats or bathes. This provides a good strategy for choosing a matrix for patches that require prolonged administration.

Abstract:In transdermal patches, pressure-sensitive adhesives (PSAs) are considered the most critical excipients due to their ability to provide adhesion and act as a drug reservoir. However, the loaded drugs plasticize the patches, resulting in a cold flow phenomenon, which poses a product safety risk and reduces treatment efficacy. Herein, 3-methacryloxypropyltrimethoxysilane (KH-570)-modified hydroxyl acrylate PSA (AAOK) was proposed. Diclofenac, ketoprofen, tulobuterol, and oxybutynin were selected as model drugs. The results showed that when not loaded with drugs, the performance of AAOK was not significantly different from that of hydroxyl acrylate PSA (AAOH). Interestingly, when loaded with drugs, AAOK automatically forms a cross-linked system through drug catalysis, which exerts a strong antiplasticity without reducing the ability of AAOK to load and deliver the drug. Specifically, the shear strength of drug-loaded AAOK increased by more than 10-fold compared to AAOH. The cold flow area of drug-loaded AAOK was reduced by more than 15-fold compared to commercial DURO-TAK 87-2287. In addition, the “dark ring” phenomenon was not observed with AAOK compared to that with DURO-TAK 87-2287 on the human forearm after drug loading. After loading four model drugs into AAOK, a high drug loading and efficient drug delivery were endowed for AAOK in vitro and in vivo. The molecular mechanism for the improved antiplasticity of the drug-loaded AAOK was that the drug molecules had a catalyzing effect on the self-condensation reactions of KH-570. This study opens up ideas for the investigation of transdermal patches with high antiplasticity and efficient transdermal application.

Mechanistic insights of different release behaviors dominated by drug physicochemical properties in polyisobutylene pressure sensitive adhesive

Abstract:The purpose of this study was to investigate the effect of the physicochemical parameters of drugs on their own release behaviors in polyisobutylene pressure sensitive adhesive (PIB PSA), which provided a theoretical guidance for the application of PIB in transdermal drug delivery system (TDDS). Seven drugs with different physicochemical parameters including clonidine (CLO), flurbiprofen (FLU), diclofenac (DIC), ibuprofen (IBU), zolmitriptan (ZOL), lidocaine (LID), tulobuterol (TUL) and the mixed adhesive (7:3, w/w) of Oppanol® B 15 N (M.W. = 108,000 Da) and Oppanol® N 50 (M.W. = 565,000 Da) were selected for in vitro drug release and skin permeation studies. Regression analysis was used to study the relationship between physicochemical parameters and release behaviors. The release behaviors of drugs were a negative correlation with polarizability and dipole moment per molecular volume (μ/V), which represented van der Waals and dipole-dipole interaction, respectively. Fourier transform infrared spectroscopy (FT-IR), modulated temperature differential scanning calorimetry (MDSC) and molecular dynamics simulation were used to provide molecular details of the interaction between the drug and PIB. The free volume and molecular mobility of PIB were characterized using mechanical property tests, rheology study, MDSC and molecular dynamics simulation. Based on the above results, drugs with high polarizability and μ/V had stronger van der Waals and dipole-dipole interaction with PIB, reducing the free volume and molecular mobility of PIB, so that the drug struggled to release from PIB. In addition, the diffusion activation energy of the drug was calculated by using the variable temperature release study to characterize the ease of drug release from the kinetic aspect. And the trends of in vitro drug release and skin penetration profiles were basically similar. Thus, it was thought that the physicochemical parameters of the drug played a vital role in the drug release behavior of PIB PSAs and would affect the skin penetration process, which provided a reference for the design and application of patches based on PIB PSAs in TDDS.

Abstract:Eudragit® RL (EuRL) matrices have been proposed to release a drug to the skin. However, no information is available on both viscoelastic and adhesive properties of such compositions. This work focuses on the evaluation of both rheological and texture properties of EuRL differently plasticized with tributyl citrate (TBC) or triacetin (TRI) in order to design a pressure sensitive adhesive suitable for transdermal patch preparation. The patch adhesive properties (i.e. tack, peel adhesion and shear adhesion) as well as its in vitro biopharmaceutical performances were determined after loading ibuprofen, ketoprofen or flurbiprofen. The addition of 40-60% w/w TBC or 40-50% w/w TRI to EuRL permitted to obtain matrices with the desired adhesive properties. Moreover, the increase of plasticizer content and loading of the drug reduced the relaxation time (τR). Consequently, the shear adhesion values decreased and the in vitro drug release constants (k) increased. Indeed, the k values from patches containing TBC were lower than the corresponding with TRI because of the lower fluidity of such matrices. In conclusion, the 60/40 EuRL/TBC binary blend is suitable for the design of transdermal patches since the in vitro permeability of the three selected drugs appeared comparable to those described in literature for marketed products.

Abstract:This study aimed to design a rizatriptan (RIZ) transdermal patch by combining of high-polarity hydroxyl pressure sensitive adhesive (PSA) AAOH-45 with an ion-pair strategy and investigate the molecular mechanism of high content hydroxyl PSA to enhance drug-PSA miscibility. RIZ free base, ion-pair complexes and PSAs containing hydroxyl group were prepared and characterized. Formulation factors including counter-ions, PSAs, drug-loading and others were optimized through single-factor studies and evaluated through pharmacokinetic studies and skin irritation tests. The properties of high polarity PSA and molecular mechanism of drug-PSA miscibility were investigated through molecular simulation, FTIR spectra, 13 C NMR spectra, DSC, and rheology study. The optimized formulation contained 20 % ( w/w ) RIZ-OA (Rizatriptan-Oleic acid), 80 % AAOH-45 ( w/w ) as the matrix, and had a thickness of 90 μm. Compared with the oral group ( MRT 0-t = 5.96 ± 0.97 h) and the control patch group ( MRT 0-t = 11.30 ± 1.78 h), the pharmacokinetic behavior of the optimization group demonstrated sustained drug delivery behavior ( MRT 0-t = 20.21 ± 0.61 h) with no irritation phenomenon. The miscibility of RIZ with PSAs was positively correlated with the mass percentage of 2-HEA. Higher polar similarity, lower flowability, and stronger intermolecular interaction were responsible for the higher compatibility of high hydroxyl PSA with the drug. This study provided a reference for increasing the drug-loading in PSA and developing RIZ patch.

Abstract:The aim was to assess the suitability of three nano-based transdermal drug delivery systems containing ibuprofen: a nano-emulsion, a nano-emulgel, and a colloidal suspension with ibuprofen-loaded nanoparticles. Understanding the transdermal delivery of ibuprofen using nano-based drug delivery systems can lead to more effective pain relief and improved patient compliance. Characterization tests assessed the suitability of the developed drug delivery systems. Membrane release and skin diffusion studies, along with tape stripping, were performed to determine drug release and skin permeation of ibuprofen. In vitro cytotoxicity studies on HaCaT cells were conducted using MTT and neutral red assays to evaluate the safety of the developed drug delivery systems. Characterization studies confirmed stable drug delivery systems with ideal properties for transdermal delivery. Membrane release studies demonstrated the successful release of ibuprofen. In vitro skin diffusion experiments and tape stripping, detecting ibuprofen in the receptor phase, stratum corneum-epidermis, and epidermis-dermis, indicating successful transdermal and topical delivery. The in vitro cytotoxicity studies observed only minor cytotoxic effects on HaCaT cells, indicating the safety of the developed drug delivery systems. The investigation demonstrated promising results for the transdermal delivery of ibuprofen using the developed drug delivery systems, which contributes to valuable insights that may lead to improved pain management strategies.

Abstract:Ionic liquid (IL)-based transdermal formulations have gained significant attention because of their high skin penetration efficiency and ease of design. However, many IL-based formulations exhibit low viscosity, which hinders direct administration and leads to less efficient drug delivery. Therefore, there is a pressing need for specialized formulations with improved viscosity and efficacy as a drug delivery platform. In this study, we report a ready-to-use transdermal patch system utilizing IL-in-oil (IL/O) microemulsions to deliver acyclovir, a sparingly soluble antiviral drug. To prepare the patch system, a mixture containing equal weights of IL/O microemulsions and adhesive (Durotak 87–4098) was homogenized, subsequently coated on a liner, and covered by a support film. These patches were produced with varying thicknesses (100 μm, 200 μm, 300 μm, and 500 μm) via a solvent evaporation technique. A 24-hour in vitro skin permeation study revealed that the thinnest patch (100 μm) delivered more than 45 % of the loaded drug. Drug delivery rate gradually reduced as the patch thickness increased, accounting for 22 % and 14 % drug permeation by 200 μm and 300 μm patches respectively. A 30-day stability study at room temperature demonstrated that neither the physical appearance nor the active ingredient content changed, validating the physical and chemical stability of the patches. Skin permeation mechanism by Fourier transform infrared spectroscopic analysis of the skin suggested that the IL/O patch possesses the capability to modify the lipid barrier in the stratum corneum thus facilitating drug diffusion. In vitro skin irritation studies on reconstructed human epidermal tissue and Yucatan micropig skin revealed that the patch exhibits no skin irritation. We believe that the adhesive nature of this newly developed IL-based patch holds significant promise as a transdermal drug delivery platform.
