Abstract:The central regulation of the food intake is organized by a long-loop mechanism involving humoral signals and afferent neuronal pathways to the hypothalamus, obligatory processing in hypothalamic neuronal circuits, and descending commands through vagal and spinal neurons to the body. Receptors sensitive to glucose metabolism, body fat reserves, distension of the stomach, as well as neuropeptide and cannabinoid receptors have been identified and localized in the hypothalamus. Five groups of cells in the hypothalamus--arcuate, paraventricular, ventromedial and dorsomedial nuclei, and the dorsolateral hypothalamic area--contain neurons with either anorexic actions (alpha-MSH, CART peptide, corticotropin-releasing hormone, urocortin III, cholecystokinin, glucagon-like peptides) or that stimulate food intake (neuropeptide Y, agouti-related peptide, orexins, melanin concentrating hormone, galanin). Intrahypothalamic neuronal circuits exist between these peptidergic neurons including the arcuate-paraventricular and arcuate-dorsolateral hypothalamic projections. Circulating substances carrying signals connected to changes in body food homeostasis and energy balance (leptin, ghrelin, insulin, glucose) enter the hypothalamus mainly through the arcuate nucleus. Neurons in the medulla oblongata that express leptin and insulin receptors, as well as neuropeptide mediators project to the hypothalamus. Vica versa, hypothalamic neurons give rise to projections to autonomic centers in the brainstem and the spinal cord with potential for stimulation or inhibition of food intake, energy balance and ingestion behavior.
N-Nitrosodimethylamine formation from anthropogenic nitrogenous compounds during preozonation and post-chloramination with characteristic low treatment dose

Abstract:One effective option to minimize N-nitrosodimethylamine (NDMA) in finished drinking water is to identify and control its precursors. However, previous works to identify significant precursors use formation potential (FP) tests using high doses to assure the maximum NDMA formation rather than the NDMA formation in finished waters. In this study, we applied characteristic low treatment doses of ozone (O3)-to-dissolved organic carbon (DOC) of target compounds of 0.8 mg/mg and NH2Cl of 2.5 ± 0.2 mg Cl2/L to evaluate the NDMAFP yields of organic compounds bearing N,N-dimethylamine (DMA) and N,N-dimethylhydrazine (DMH) during preozonation and post-chloramination. The results in pH-buffered Milli-Q water showed a significant decrease from ≤ 52% to non-detectable levels in the O3-NDMAFP yields of O3-reactive precursors (i.e., DMH-like compounds) after preozonation and post-chloramination. Similarly, a significant decrease from 0.5 to 12% to nonquantifiable levels was observed for the NH2Cl-NDMAFP yields of NH2Cl-reactive precursors; however, the NH2Cl-NDMAFP yields of N,N-dimethylbenzylamine (DMBzA)-like compounds only decreased from ~ 110 to ≤ 43%, suggesting that these compounds could contribute to NH2Cl-NDMAFPs even after preozonation. The effect of the matrix in sewage-effluent and lake water samples varied and was specific for precursors; for example, the O3-NDMAFP yield of 1,1,1',1'-tetramethyl-4,4'-(methylene-di-p-phenylene) disemicarbazide (TMDS), an important O3-reactive NDMA precursor, did not significantly decrease when tested in sewage-effluent samples. Based on the previous occurrence concentration of TMDS in sewage samples, we estimated an NDMAFP of ~ 315 ng/L. This estimate exceeds the guidance concentrations of NDMA (3-100 ng/L), highlighting the importance of TMDS and its related compounds for NDMA formation.
Oxidation of amine-based pharmaceuticals with unactivated peroxymonosulfate: Kinetics, mechanisms, and elimination efficiency of NDMA formation

Abstract:Amine-based pharmaceuticals are a significant class of N-nitrosodimethylamine (NDMA) precursors. This study investigated the use of unactivated peroxymonosulfate (PMS) to control amine-based pharmaceuticals and their NDMA formation potential. Kinetic analysis and product identification revealed that sumatriptan and doxylamine primarily underwent reactions at their tertiary amine group, while ranitidine and nizatidine had both tertiary amine and thioether group as reaction sites. The NDMA formation from sumatriptan and doxylamine during post-chloramination was significantly reduced with the abatement of the parent contaminants, while the formation of NDMA remained high even if full abatement of ranitidine and nizatidine was achieved. Product formation kinetics and reference standard tests revealed the great contribution of transformation products to NDMA formation. Ranitidine could be oxidized to sulfoxide-type product ranitidine-SO and N-oxide type product ranitidine-NO. Ranitidine-SO exhibited a high NDMA yield comparable to that of ranitidine (>90%), while ranitidine-NO showed a low NDMA yield (2%). With further oxidation of ranitidine-SO at the tertiary amine group, NDMA formation was reduced by more than 90%. The underlying mechanism for the importance of the tertiary amine group in NDMA formation was demonstrated by quantum chemical calculation. These findings underscore the potential of PMS pre-oxidation on NDMA control.

Abstract:This review summarizes major findings over the last decade related to N-nitrosodimethylamine (NDMA) formed upon ozonation, which was regarded as highly toxic and carcinogenic disinfection by-products. The reaction kinetics, chemical yields and mechanisms were assessed for the ozonation of potential precursors including dimethylamine (DMA), N,N-dimethylsulfamide, hydrazines, N-containing water and wastewater polymers, dyes containing a dimethylamino function, N-functionalized carbon nanotubes, guanidine, and phenylurea. The effects of bromide on the NDMA formation during ozonation of different types of precursors were also discussed. The mechanism for NDMA formation during ozonation of DMA was re-summarized and new perspectives were proposed to assess on this mechanism. Effect of hydroxyl radicals (•OH) on NDMA formation during ozonation was also discussed due to the noticeable oxidation of NDMA by •OH. Surrogate parameters including nitrate formation and UV254 after ozonation may be useful parameters to estimate NDMA formation for practical application. The strategies for NDMA formation control were proposed through improving the ozonation process such as ozone/hydrogen peroxide, ozone/peroxymonosulfate and catalytic ozonation process based on membrane pores aeration (MEMBRO3X).

Fitzpatrick skin types and clindamycin phosphate 1.2%/benzoyl peroxide gel: efficacy and tolerability of treatment in moderate to severe acne.

Abstract:BACKGROUND Acne in skin of color is an increasing problem, presenting unique challenges. Although combination therapy is now standard of care in acne, concerns exist with the increased potential irritation and dryness in skin of color. Although individual medications can be titrated or applied at different times of the day to minimize irritation, this is not always practical or desirable. There is a paucity of clinical studies that evaluate the safety and efficacy of acne medications in skin of color. METHODS A post-hoc analysis of efficacy and cutaneous tolerability in 797 subjects receiving clindamycin phosphate 1.2% benzoyl peroxide (BPO) 2.5% gel from two 12-week, multi-center, double-blind studies that enrolled 2,813 subjects with moderate to severe acne. Efficacy, tolerability, and subject satisfaction in Fitzpatrick skin types I-III subjects were compared to subjects with Fitzpatrick skin types IV-VI. RESULTS Median reductions in inflammatory lesions were comparable between the two groups. There was a small difference in noninflammatory and total lesions in favor of those patients with Fitzpatrick skin types I-III (P=0.013 and P=0.024, respectively). Median reductions in inflammatory, noninflammatory, and total lesions at week 12 were 63%, 50%, and 52.4%, respectively for Fitzpatrick skin types I-III and 65%, 47%, and 51.4%, respectively for Fitzpatrick skin types IV-VI. Treatment success was comparable between the two groups and both groups had a high level of subject satisfaction at week 12. Cutaneous tolerability was excellent, with all mean scores less than or equal to 0.2 at week 12 (where 1=mild). Results in the two groups were comparable, although there was slightly more erythema reported in the Fitzpatrick skin types I-III subjects (0.2 versus 0.1). This could be due to the difficulty in visualizing erythema in subjects with darker skin. CONCLUSIONS Acne subjects with Fitzpatrick skin types IV-VI were not found to be more susceptible to cutaneous irritation from treatment with clindamycin phosphate 1.2%/BPO 2.5% gel and both efficacy and tolerability was comparable across the two treatment groups.
NDMA formation during ozonation of metformin: Roles of ozone and hydroxyl radicals

Abstract:Metformin, a high-consumed pharmaceutical for diabetes, has been reported to generate carcinogenic nitroso-dimethylamine (NDMA) during treatment of its containing wastewater. However, whether it would produce NDMA during ozonation or not is unclear, let alone discriminate roles of ozone (O3) and hydroxyl radicals (OH). In this paper, effects of ozonation on NDMA formation from metformin were investigated, roles of O3 and OH were also distinguished by adding tert-butyl alcohol (tBA) as OH scavenger. Moreover, various influencing factors and reaction mechanisms were demonstrated. The results indicated that NDMA could be directly formed from metformin during ozonation, the addition of OH scavenger significantly enhanced its formation (0–46.2 ng/L vs 0–139.1 ng/L). The formation of NDMA by O3 and OH was more affected by bromide and HCO3− than those with only O3; while the impacts of pH and sulphate on the latter were more notable. No matter without/with tBA in the solution, the formed NDMA during ozonation of metformin increased with raising pH (from 5 to 9) and achieved the maximum 69.6 ng/L and 235.9 ng/L at pH 9, respectively; small amount of bromide (0.1 μM) promoted NDMA production, high levels of bromide (10 μM) inhibited its formation; the existence of HCO3− enhanced the amounts of NDMA from 44.5 to 73.5 ng/L (raised by 65.2%) by O3 and OH and from 102.9 to 130 ng/L with only O3 (raised by 26.3%); with the addition of sulphate, NDMA concentration raised by 43.8% by O3 and OH, while the value was high up to 134.6% with only O3. Based on the result of UPLC-Q-TOF and density functional theory, the oxidation intermediates were identified and possible transformation pathways of metformin during ozonation were proposed. The findings in this paper would provide reference when treating metformin-containing water in future.

Abstract:Aliphatic amines, common constituents that contribute to dissolved organic nitrogen (DON), can quickly react with ozone due to the lone electron pair on the nitrogen atom and this may produce carcinogen N-Nitrosodimethylamine (NDMA). 3-(Dimethylamino)-1-propylamine (DMAPA) was chosen as a representative to elucidate the NDMA formation characteristics, kinetic rates, reaction pathways, and influencing factors during ozonation in this study. The results demonstrated that NDMA generated directly from DMAPA during ozonation. Moreover, the NDMA yields increased with ozone dosages. The NDMA molar yield increased and then decreased when the pH raised from 5 to 9, achieving the maximum value at pH 8. Hydroxyl radical (∙OH) played a promotional role in NDMA formation, and its scavenger dramatically cut down its yields. Low levels of Br- facilitated NDMA formation, while the value significantly reduced when Br- was up to 1 mM. The NDMA amount was slightly raised by NO2-, but it was inhibited by NH4+ and NO3-. Moreover, it was also depressed by co-existing components in actual lake water. Based on the result of the Gaussian calculation, the LC-MS/MS and GC-MS analysis, four possible transformation pathways were proposed. The radical recombination was verified to be the primary pathway for ozone promoting NDMA formation from DMAPA.

Abstract:Bromide ion (Br - ) in water sources has been found to affect the formation of carcinogenic non-brominated Nnitrosodimethylamine (NDMA) during ozonation of typical amines. The increasing bromide levels bring more concern due to climate change and industrial discharge, but its effects on actual water matrixes were still unclear. In this study, NDMA formation from three actual water matrixes during ozonation as the Br - concentration increased from 0 to 100 mu M were investigated (promotion (100.4 - 231.6 ng/L), promotion then inhibition (399.5 - 461.2 - 428.2 ng/L), and subtle influence (51.9 +/- 6.9 ng/L)); from which the NDMA precursors were screened and 8 precursors were selected for exploring the influencing mechanisms. The results indicated that the roles of Br - on NDMA formation significantly depended on the water characteristics of actual matrixes. A total of 16 typical NDMA precursors, which were mainly derived from industrial, agricultural and pharmaceutical sources, were identified. Moreover, 6 brominated substance (Br -DMA, Br-UDMH, Br-DMSC, Br-DMS, Br-TMDS, Br 2 -TMDS), which were thought to play a crucial role in promoting NDMA formation, were also identified in actual water by precursor ion scan (PIS) method. Hypobromous acid (HOBr), which has been proved to be the culprit to the generation of brominated substances, tend to attack the electron -donating groups of amines, such as acylamino (-NHCO-R) or amidogen (-NH 2 ) groups. However, the reactive bromine species ( center dot Br and center dot Br 2 - ) was inclined to attack the aromatic and anti -oxidant moieties then result in Br - release, as no bromine substitution but free radical transfer reaction occurs. Therefore, decreasing HOBr formation is the crucial step to reduce NDMA generation. This work would provide technical guide for NDMA risks control during ozonation of Br - -amines cocontaining water matrixes.

Formation of N-Nitrosodiphenylamine and Two New N-Containing Disinfection Byproducts from Chloramination of Water Containing Diphenylamine

Abstract:N-nitrosodiphenylamine (NDPhA) is a disinfection byproduct (DBP) in drinking water, However, it is not known what governs the formation of NDPhA and which precursor(s) in the raw water is responsible for its formation. We report here diphenylamine (DPhA) as a key precursor of NDPhA, and we describe the effect of water pH and chloramination conditions on the formation of NDPhA. To identify precursors of NDPhA, raw water samples were collected from the same drinking water system in which NDPhA was previously detected. Analysis of the raw water samples showed the presence of 1.3 ng/L of DPhA and no detectable NDPhA. Seven hours after the treatment of the raw water with chloramines, the concentration of DPhA decreased to 0.4 ng/L with corresponding formation of NDPhA (0.4 ng/L). Controlled experiments involving chloramination of DPhA in water showed that chloramines were essential to the formation of NDPhA, and that increasing the pH from 4 to 10 resulted in 64-fold enhancement in NDPhA formation. Removal of DPhA and formation of NDPhA was found by mass imbalance, which led to the identification of two new DBPs, phenazine (MW 180 Da) and a chlorinated phenazine derivative (MW 216 Da), using liquid chromatography tandem mass spectrometry and gas chromatography mass spectrometry. Both new DBPs were detected only in the treated water and not in the raw water. Phenazine and N-chlorophenazine have never been reported as DBPs and neither their occurrence in drinking water nor their health effects are known.

Impact of ozonation on the formation of particulate nitrosodi-methylamine (NDMA) in atmosphere

Abstract:The contribution of ozonation to the formation of particulate nitrosodi-methylamine (NDMA) in the aqueous aerosol phase was investigated using measurement data from 2018 in Seoul, Republic of Korea and a box model. The correlation between the NDMA concentration and aerosol liquid water content and box model results showed that aqueous aerosol phase reactions, including nitrosation and ozonation, might contribute to the formation of NDMA. The concentration of NDMA and the ratio of O3/dimethylamine exhibited a negative correlation, suggesting that the contribution of ozonation to NDMA formation may not be significant. Furthermore, when the daily concentration of NDMA exceeded 10 ng/m3, the pH was 3.96 ± 0.48, indicating that the impact of ozonation on NDMA concentration might not be significant. To quantitatively investigate the contribution of ozonation, the ozonation mechanism that forms NDMA was included in the box model developed in our previous study. The model results showed that the ozonation contributed to the ambient concentration of NDMA (7.9 ± 3.8% (winter); 1.9 ± 3.0% (spring); 10.0 ± 0.77% (summer); 3.6 ± 7.3% (autumn)). It is estimated that the relatively higher O3/NOx ratio in summer (1.63 ± 0.69; 0.64 ± 0.52 (winter); 1.14 ± 0.92 (spring); 0.52 ± 0.54 (autumn)) could enhance ozonation and that relatively lower pH in summer (2.2 ± 0.4; 5.3 ± 1.2 (winter); 3.9 ± 1.2 (spring); 3.9 ± 0.7 (autumn)) could hinder nitrosation compared to that in other seasons.

Abstract:Rationale N ‐Nitroso dimethylamine (NDMA) is a mutagenic impurity detected in several ranitidine products. The amino functional group of ranitidine is a risk factor for classical nitrosation‐induced NDMA formation in ranitidine drug products during storage conditions. The United States Food and Drug Administration (US FDA) recommended the use of antioxidants to control NDMA in drug products. Considering the need for sensitive analytics, a liquid chromatography/high‐resolution mass spectrometry (LC–HRMS) method was developed and validated to detect NDMA in this pilot study to demonstrate the antioxidants as inhibitors of nitrosation reactions. Methods The method, utilizing an EC‐C18 column and tuned to atmospheric pressure chemical ionization/selected ion monitoring (APCI/SIM) mode, separated NDMA ( m/z : 75.0553; tR: 3.71 min) and ranitidine ( m/z : 315.1485; tR: 8.61 min). APCI mode exhibited four times higher sensitivity to NDMA than electrospray ionization (ESI) mode. Classical nitrosation of the dimethyl amino group of ranitidine was studied with sodium nitrite in solid pellets. Antioxidants (alpha‐tocopherol, ascorbic acid, and trolox) were evaluated as NDMA attenuators in ranitidine pellets under vulnerable storage conditions. The developed method quantified NDMA levels in samples, extracted with methanol through vortex shaking for 45 min. Results The method achieved a limit of detection (LOD) and limit of quantitation (LOQ) of 0.01 and 0.05 ng/mL, respectively, with linearity within 1–5000 ng/mL ( R 1 : 0.9995). It demonstrated good intra‐day and inter‐day precision (% RSD [relative standard deviation]: <2) and accuracy (96.83%–101.72%). Nitrosation of ranitidine induced by nitrite was significant ( p < 0.001; R 2 = 0.9579) at various sodium nitrite levels. All antioxidants efficiently attenuated NDMA formation during ranitidine nitrosation. Ascorbic acid exhibited the highest NDMA attenuation (96.98%), followed by trolox (90.58%). This study recommends 1% ascorbic acid and trolox as potent NDMA attenuators in ranitidine drug products. Conclusions This study compared the effectiveness of antioxidants as NDMA attenuators in ranitidine under storage conditions susceptible to NDMA generation. The study concluded that ascorbic acid and trolox are potent inhibitors of NDMA formation and nitrosation attenuators in ranitidine drug products.

This study demonstrates that the existence of environmentally relevant concentrations of nitrite (NO 2 − ) can lead to the formation of N -nitrosodimethylamine (NDMA), a probable human carcinogen, when heat activated peroxydisulfate (heat/PDS) is applied to address contaminants with dimethylamine moieties, such as tetracyclines. NO 2 − effectively competes with tetracyclines for SO 4 •− at a high second-order reaction rate constant of 8.8×10 8 M −1 s −1 , thus suppressing the degradation. Simultaneously, SO 4 •− reacts with NO 2 − to form the nitrogen dioxide radical (NO 2 • ) which rapidly dimerizes to a potent nitrosating agent named dinitrogen tetroxide (N 2 O 4 ). N 2 O 4 , in turn, attacks the dimethylamine moiety in tetracyclines via nucleophilic substitution, contributing to the degradation of parent compounds and generating an active intermediate that quickly decomposes to NDMA, along with the release of R + and NO 3 − . The released R + can be further oxidized by SO 4 •− to form phenolic intermediates which combine with NO 2 • to generate nitrated products. When 5 μM oxytetracycline (OTC) was treated with 0.5 mM PDS in the presence of 20 μM NO 2 − at 60 °C, the highest formation of NDMA was 0.045 μM in 1 h. NDMA formation was also observed for other compounds with dimethylamine moieties, such as methylene blue, phenylurea herbicides, etc., with the molar yield ranging from 0.07 to 3.53%, negatively related to the R-N bond dissociation energy of the precursors. These findings suggest that NDMA can be commonly generated when SO 4 •− is applied to the degradation of dimethylamine pollutants in wastewater treatment or groundwater remediation where NO 2 − ubiquitously exists, which may pose a serious threat to the aquatic environment.

Abstract:Four pharmaceuticals (ranitidine, nizatidine, doxylamine, and carbinoxamine) were selected as model compounds to assess the efficiency of four oxidants (ozone (O3), chlorine (Cl2), chlorine dioxide (ClO2) and potassium permanganate (KMnO4)) on the removal of amine-based pharmaceutical and personal care products (PPCPs), as well as the reduction of their N-Nitrosodimethylamine formation potentials (NDMAFPs). The changes in PPCPs and their NDMAFPs during oxidation were quantified using various oxidants and dosages. The relationship between oxidation product structures and NDMAFP changes was also analyzed. The results showed that oxidation with O3, Cl2 and ClO2 were effective in removing the selected PPCPs. However, only ozonation was effective in reducing their NDMAFPs. Ozonation at 6 mg/L removed approximately 90% PPCPs and 90% NDMAFPs for all PPCPs. In addition, the results indicated that ozonation products made little contribution to NDMAFPs. In contrast, some PPCP products had higher NDMAFPs than PPCPs after oxidation with Cl2, ClO2 and KMnO4. There were two possible reaction pathways that led to decrease in NDMAFPs after oxidation. One was oxygen transfer to nitrogen at the tertiary amine site and the other was N-dealkylation from the tertiary amine site.

Abstract:Microcystis aeruginosa (M. aeruginosa, blue-green algae) blooms frequently in drinking water reservoirs and subsequently causes the formation of disinfection by-products (DBPs) after disinfection, which may pose a potential health risk. In this study, the formation of N-nitrosodimethylamine (NDMA) was evaluated from algal organic matter (AOM) including extracellular organic matter (EOM) and intracellular organic matter (IOM) during the disinfection process of chlorination, chloramination, or ultraviolet (UV) irradiation. The effects of a variety of factors, including reaction times, disinfectant dosages and pH, on the NDMA formation by three different disinfection methods were investigated. Additionally, this study evaluated the nitrogen sources involved in NDMA formation during chloramination of EOM and IOM using 15N-labeled monochloramine. The results showed that the NDMA formation by three different disinfection methods were ranked in the order of chlorination > UV irradiation ≈ chloramination and the specific yield from EOM was greater than that from IOM regardless of disinfection method. The yields of NDMA firstly increased and then plateaued as time prolonged during the chlorination and chloramination of AOM. Similarly, the NDMA formation from EOM was firstly increased and then remained constant with the increase of the disinfectant dosage, while it was gradually increased for IOM. The solution pH highly influenced the NDMA formation during chlorination and chloramination, while exhibited a little impact under UV irradiation. Moreover, fluorescence excitation-emission (EEM) analysis confirmed that soluble microbial by-product-like (SMPs) in EOM and IOM were the major precursors in algal-derived organic matter that contributed to the NDMA formation. Chloramination of EOM and IOM using isotope 15N-labeled monochloramine indicated that the nitroso group of the formed NDMA originates mainly from EOM and IOM of algal cells.

Abstract:Nitro-based compounds are the direct precursors of trichloronitromethane during chlorination disinfection. Two nitro-based pharmaceuticals ranitidine and nizatidine were selected as model compounds to assess the effect of oxidation on the removal of nitro-based pharmaceuticals, as well as the reduction of their trichloronitromethane formation potentials (TCNMFPs). The four oxidants were ozone (O3), chlorine (Cl2), chlorine dioxide (ClO2) and potassium permanganate (KMnO4). The changes in pharmaceuticals and their TCNMFPs during oxidation using various oxidants and dosages were quantified. The relationships between oxidation product structures and TCNMFP changes were also analyzed. The results showed that oxidation with Cl2 and KMnO4 were more effective than ClO2 and O3 in removing the nitro-based pharmaceuticals. Meanwhile, decreased TCNMFPs by KMnO4 oxidation but increased TCNMFPs by Cl2, ClO2 and O3 oxidation were observed. The results of product analysis indicated that chlorine transfer products had higher TCNMFPs, while oxygen transfer products made little contribution to TCNMFPs after oxidation. In addition, one possible reaction pathway leading TCNMFP increase was that chloro-nitromethane or nitromethane, which was a better TCNM precursor, formed when double bond was attacked by oxidants.
Removal of Dimethylamine by Ozone Combined with Hydrogen Peroxide (O3/H2O2)

Abstract:Ozonation of dimethylamine (DMA), the most direct and common precursor of high carcinogenic disinfection by-product N-nitrosodimethylamine (NDMA), in aqueous solutions and in the presence of H2O2 was studied at ozone doses of 1.8 mg/L. The removal ratio of DMA increases from 46.7% to 77.1% with the dosage of H2O2 increasing from 0 to 1 mM, and the removal rate can be improved by increasing pH. According to the results of H2O2 addition time experiments, hydroxyl radicals might play important roles in the destruction of DMA during combined oxidation process. With the treatment of O3/H2O2 for 24 h (containing 0.5 mM DMA), the yield of NDMA formation was below our detection limit (0.1Â¿g/L) and the yield of NDMA formation after chloramination (24 h) of DMA containing water could be reduced by 90%.

Abstract:The formation of cancinogenic nitrosamines, esp. N-nitrosodimethylamine (NDMA) in water and wastewater treatment plants has drawn much attention in recent years. Dissolved organic matter from the transported Luan River water as water source of Tianjin was fractionated with different XAD resins and a series of ultra-filtration membranes with molecular weight (MW) cut-offs of 5k Da, 3k Da, and 1k Da, respectively. The NDMA yields from the raw water and each fraction were measured to investigate their role in NDMAyield. Results indicated that the hydrophilic fraction had a higher NDMA yield than those of hydrophobic fraction and transphilic fraction. The fraction with MW below 1k Da had a higher NDMAyield than that with larger MW. NDMA formation increased as the dissolved organic carbon (DOC) to dissolved organic nitrogen (DON) ratio decreased, which indicated that DON might serve as the real important precursor for NDMA. The correlation between NDMA yield and specific ultraviolet absorbance at 254 nm (SUVA254) suggested that the latter might not represent the specific precursors for NDMA in the water. Besides the water quality, the influences of pH, disinfectant dosage, and disinfection contact time on the formation of NDMA were also examined. These results will help water treatment plants establish measures to control this harmful disinfection by-product.

Abstract:This review summarizes major findings over the last decade related to N-Nitrosodimethylamine (NDMA) in water and wastewater. In particular, the review is focused on the removal of NDMA and of its precursors by conventional and advanced water and wastewater treatment processes. New information regarding formation mechanisms and precursors are discussed as well. NDMA precursors are generally of anthropogenic origin and their main source in water have been recognized to be wastewater discharges. Chloramination is the most common process that results in formation of NDMA during water and wastewater treatment. However, ozonation of wastewater or highly contaminated surface water can also generate significant levels of NDMA. Thus, NDMA formation control and remediation has become of increasing interest, particularly during treatment of wastewater-impacted water and during potable reuse application. NDMA formation has also been associated with the use of quaternary amine-based coagulants and anion exchange resins. UV photolysis with UV fluence far higher than typical disinfection doses is generally considered the most efficient technology for NDMA mitigation. However, recent studies on the optimization of biological processes offer a potentially lower-energy solution. Options for NDMA control include attenuation of precursor materials through physical removal, biological treatment, and/or deactivation by application of oxidants. Nevertheless, NDMA precursor identification and removal can be challenging and additional research and optimization is needed. As municipal wastewater becomes increasingly used as a source water for drinking, NDMA formation and mitigation strategies will become increasingly more important. The following review provides a summary of the most recent information available.
