Abstract:Micro and nano technologies are of increasing importance in microfluidics devices used for electroporation (electroporation – the permeabilization of the cell membrane with brief high electric field pulses). Electrochemical reactions of electrolysis occur whenever an electric current flows between an electrode and an ionic solution. It can have substantial detrimental effects, both on the cells and solutions during the electroporation. As electrolysis is a surface phenomenon, between electrodes and solution, the extent of electrolysis is increased in micro and nano electroporation over macro-electroporation, because the surface area of the electrodes in micro and nano electroporation is much larger. A possible way to eliminate the electrolytic effect is to develop non-electrolytic microelectroporation by coating the microelectroporation devices with a dielectric insulating layer. In this study, we examine the effect of a dielectric insulating layer on the performance of a singularity microelectroporation device that we have recently designed. Using numerical analysis, we study the effects of various design parameters including, input sinusoidal voltage amplitude and frequency, geometrical configuration and material electrical properties on the electroporation performance of the non-electrolytic microelectroporation device. In the simulation, we used properties of four real dielectric materials and four solutions of interest for microelectroporation. We characterized the effect of various design parameters of relevance to singularity based microelectroporation, on non-electrolytic microelectroporation. Interestingly, we found that the system behaves in some aspects as a filter and in many circumstances saturation of performance is reached. After saturation is reached, changes in parameters will not affect the performance of the device.

Abstract:We present a novel nanostructured micro cell chip with impedance spectroscopy for monitoring cell morphology change non-invasively, in real time and independent of any fluorescent or radioactive probes. The key strategy is to integrate the nanoporous alumina membrane with silicon based microfluidic devices for the impedance monitoring. In this configuration, the impedance of even single cell can be measured at the low frequency range but not be affected by the electrode polarization. The KYSE30 human oesophageal cancer cells have been successfully cultured on nanoporous alumina membrane. Initial electrochemical experiments with lipid layers have been done to testify the functionality of this device. Further experiments for cancer cells will be explored in near future.
Soft Electroporation Through 3D Hollow Nanoelectrodes.

Abstract:Generally, electroporation of in vitro cells is performed under very high electric fields to overcome the physical barrier of plasma membrane. Since traditional electroporation techniques make use of very high voltages, which is critical to cell viability, this study presents a microfluidic platform able to perform cell membrane electroporation with the application of low voltages (1.5-2 V). The platform is manufactured based on the milling by mean of focused ionic beam, which offers an established approach to fabricate ordered arrays of 3D gold hollow nanoelectrodes protruding from an insulating substrate. The novelty of this fabrication relies on the fact that the nanoelectrodes used for electroporation are simultaneously metallic, hollow and communicate through its nanochannels with an isolated microfluidic chamber beneath the device. Adherent cultured cells on the nanoelectrodes can be electroporated in this platform, and molecules can be selectively delivered only inside the porated cells.

Abstract:Lab-on-a-chip micro-devices utilizing electric field-mediated particle movement provide advantages over current cell rotation techniques due to the flexibility in configuring micro-electrodes. Recent technological advances in micro-milling, three-dimensional (3D) printing and photolithography have facilitated fabrication of complex micro-electrode shapes. Using the finite-element method to simulate and optimize electric field induced particle movement systems can save time and cost by simplifying the analysis of electric fields within complex 3D structures. Here we investigated different 3D electrode structures to obtain and analyse rotational electric field vectors. Finite-element analysis was conducted by an electric current stationary solver based on charge relaxation theory. High-resolution data were obtained for three-, four-, six- and eight-cylindrical electrode arrangements to characterize the rotational fields. The results show that increasing the number of electrodes within a fixed circular boundary provides larger regions of constant amplitude rotational electric field. This is a very important finding in practice, as larger rotational regions with constant electric field amplitude make placement of cells into these regions, where cell rotation occurs, a simple task - enhancing flexibility in cell manipulation. Rotation of biological particles over the extended region would be useful for biotechnology applications which require guiding cells to a desired location, such as automation of nuclear transfer cloning.

Abstract:This paper reports a method for label‐free single‐cell biophysical analysis of multiple cells trapped in suspension by electrokinetic forces. Tri‐dimensional pillar electrodes arranged along the width of a microfluidic chamber define actuators for single cell trapping and selective release by electrokinetic force. Moreover, a rotation can be induced on the cell in combination with a negative DEP force to retain the cell against the flow. The measurement of the rotation speed of the cell as a function of the electric field frequency define an electrorotation spectrum that allows to study the dielectric properties of the cell. The system presented here shows for the first time the simultaneous electrorotation analysis of multiple single cells in separate micro cages that can be selectively addressed to trap and/or release the cells. Chips with 39 micro‐actuators of different interelectrode distance were fabricated to study cells with different sizes. The extracted dielectric properties of HeLa, HEK 293, and human immortalized T lymphocytes cells were found in agreements with previous findings. Moreover, the membrane capacitance of M17 neuroblastoma cells was investigated and found to fall in in the range of 7.49 ± 0.39 mF/m2.This article is protected by copyright. All rights reserved

Abstract:Multielectrode arrays for interfacing with neurons are of great interest for a wide range of medical applications. However, current electrodes cause damage over time. Ultra small carbon fibers help to address issues but controlling the electrode site geometry is difficult. Here we propose a methodology to create small, pointed fiber electrodes (SPFe). We compare the SPFe to previously made blowtorched fibers in characterization. The SPFe result in small site sizes with consistently sharp points (20.8 ± 7.64°). Additionally, these electrodes were able to record and/or stimulate neurons multiple animal models including rat cortex, mouse retina, Aplysia ganglia and octopus axial cord. In rat cortex, these electrodes recorded significantly higher peak amplitudes than the traditional blowtorched fibers. These SPFe may be applicable to a wide range of applications requiring a highly specific interface with individual neurons.

Abstract:There is an enduring quest for technologies that provide - temporally and spatially - highly resolved information on electric neuronal or cardiac activity in functional tissues or cell cultures. Here, we present a planar high-density, low-noise microelectrode system realized in microelectronics technology that features 11,011 microelectrodes (3,150 electrodes per mm(2)), 126 of which can be arbitrarily selected and can, via a reconfigurable routing scheme, be connected to on-chip recording and stimulation circuits. This device enables long-term extracellular electrical-activity recordings at subcellular spatial resolution and microsecond temporal resolution to capture the entire dynamics of the cellular electrical signals. To illustrate the device performance, extracellular potentials of Purkinje cells (PCs) in acute slices of the cerebellum have been analyzed. A detailed and comprehensive picture of the distribution and dynamics of action potentials (APs) in the somatic and dendritic regions of a single cell was obtained from the recordings by applying spike sorting and spike-triggered averaging methods to the collected data. An analysis of the measured local current densities revealed a reproducible sink/source pattern within a single cell during an AP. The experimental data substantiated compartmental models and can be used to extend those models to better understand extracellular single-cell potential patterns and their contributions to the population activity. The presented devices can be conveniently applied to a broad variety of biological preparations, i.e., neural or cardiac tissues, slices, or cell cultures can be grown or placed directly atop of the chips for fundamental mechanistic or pharmacological studies.

Abstract:Several non-viral techniques involving the use of liposomes, particle bombardment and electroporation have been used for efficient transfection of plasmids and other molecules into cells. Current approaches target whole or bulk regions of tissue, lacking the desired spatial control over the transfection process. In this study, we present a novel approach using microsystems to achieve spatial and temporal control over the transfection process in adherent cells. A 6x6 MEA (microelectrode array) with 100 microm microelectrode dimension was developed on a silicon substrate using standard microfabrication procedures and passivated with a biocompatible layer. Using finite element models, electric field intensities were simulated and locations of optimal electroporation zones in the cell culture on the microelectrode surface were predicted. The MEA was subsequently tested using 3T3 fibroblasts cultured on the MEA surface for 96 h and stimulation voltages in the range of 2-5 V in the presence of propidium iodide (PI), a cell impermeant dye. Maximum electric field intensities in the z-direction were estimated to be in the range of 320-820 V/cm for applied differential voltages in the range of 2-5 V. Cells directly on the top and on the edges of the stimulating microelectrodes in the MEA were preferentially transfected with PI as predicted by the simulations. The results of these experiments demonstrate that spatial and temporal control of desired regions of transfection in vitro can be achieved using MEAs and electroporation.
Transparent and Conformal Microcoil Arrays for Spatially Selective Neuronal Activation

Abstract:Micromagnetic stimulation (μMS) using small, implantable microcoils is a promising method for achieving neuronal activation with high spatial resolution and low toxicity. Herein, we report a microcoil array for localized activation of cortical neurons and retinal ganglion cells. We developed a computational model to relate the electric field gradient (activating function) to the geometry and arrangement of microcoils, and selected a design that produced an anisotropic region of activation <50 μm wide. The device was comprised of an SU-8/Cu/SU-8 tri-layer structure, which was flexible, transparent and conformal and featured four individually-addressable microcoils. Interfaced with cortex or retina explants from GCaMP6-expressing mice, we observed that individual neurons localized within 40 μm of a microcoil tip could be activated repeatedly and in a dose- (power-) dependent fashion. These results demonstrate the potential of μMS devices for brain-machine interfaces and could enable routes toward bioelectronic therapies including prosthetic vision devices.

Abstract:Microelectrode arrays (MEAs) are state-of-the-art devices for extracellular recording and stimulation on biological tissue. Furthermore, they are a relevant tool for the development of biomedical applications like retina, cochlear and motor prostheses, cardiac pacemakers and drug screening. Hence, research on functional cell-sensor interfaces, as well as the development of new surface structures and modifications for improved electrode characteristics, is a vivid and well established field. However, combining single-cell resolution with sufficient signal coupling remains challenging due to poor cell-electrode sealing. Furthermore, electrodes with diameters below 20 µm often suffer from a high electrical impedance affecting the noise during voltage recordings. In this study, we report on a nanocavity sensor array for voltage-controlled stimulation and extracellular action potential recordings on cellular networks. Nanocavity devices combine the advantages of low-impedance electrodes with small cell-chip interfaces, preserving a high spatial resolution for recording and stimulation. A reservoir between opening aperture and electrode is provided, allowing the cell to access the structure for a tight cell-sensor sealing. We present the well-controlled fabrication process and the effect of cavity formation and electrode patterning on the sensor's impedance. Further, we demonstrate reliable voltage-controlled stimulation using nanostructured cavity devices by capturing the pacemaker of an HL-1 cell network.
Microtrap electrode devices for single cell trapping and impedance measurement

Abstract:This paper reports the design and fabrication of electrode microtraps for single cell trapping and impedance measurement. In this work, the microtrap electrodes of parallel and elliptical geometry have been fabricated by electroplating of gold electrodes of optimum thickness. This has enabled the formation of electrode traps without requiring any precision alignment between separate insulating traps like PDMS and the bottom gold electrodes. Further the improved uniformity of the electric field between the trapping electrodes as observed from COVENTORWARE simulation significantly reduces the effect of cell position inside the microwell on the electrical measurement unlike previous reports. This makes it possible to directly extract the equivalent cell parameters from the electrical measurement without introducing any correction factor corresponding to cell position. We have performed impedance spectroscopy with both the microwell electrode structures with single HeLa cell at two different positions of trapping. It has been observed that there is almost no change in the extracted values of cell resistance and capacitance for different positions within parallel electrodes and there is only 0.7 % and 0.85 % change in cell resistance and capacitance for the two positions within elliptical electrodes. Thus these microwell electrode structures can be used as an improved and a more convenient platform for single cell electrical characterization.

Abstract:We use a theoretical approach to examine the effect of a radial fluid flow or electric current on the growth and homeostasis of a cell spheroid. Such conditions may be generated by a drain of micrometric diameter. To perform this analysis, we describe the tissue as a continuum. We include active mechanical, electric, and hydraulic components in the tissue material properties. We consider a spherical geometry and study the effect of the drain on the dynamics of the cell aggregate. We show that a steady fluid flow or electric current imposed by the drain could be able to significantly change the spheroid long-time state. In particular, our work suggests that a growing spheroid can systematically be driven to a shrinking state if an appropriate external field is applied. Order-of-magnitude estimates suggest that such fields are of the order of the indigenous ones. Similarities and differences with the case of tumors and embryo development are briefly discussed.

Abstract:This paper reports a novel microarray chip for in-situ , real-time and selective electroporation on individual cells integrated with cell positioning and impedance monitoring. An array of quadrupole-electrode units (termed positioning electrodes) and pairs of planar center electrodes located at the centers of each quadrupole-electrode unit were fabricated on the chip. The positioning electrodes are used to trap and position living cells onto the center electrodes based on negative dielectrophoresis (nDEP). The center electrodes are used for in-situ cell electroporation and also used to measure cell impedance for monitoring cellular dynamics in real time. Controllably selective electroporation and electrical measurement on the cells in array are realized. We present an evidence of selective electroporation through use of fluorescent dyes. Subsequently we use in-situ and real-time impedance measurement to monitor the process, which demonstrates the dynamic behavior of the cell electroporation. Finally, we show the use of this device to perform successful transfection onto individual HeLa cells with vector DNA encoding a green fluorescent.

Abstract:Multi-electrode arrays serve to record electrical signals of many neurons in the brain simultaneously. For most of the past century, electrodes that penetrate brain tissue have had exactly one shape: a straight needle. Certainly this was a good starting choice at the time, but there is no reason to think that a straight line would be the optimal shape in all Neuroscience applications. Here I argue that, in fact, a wide variety of curved shapes is equally practical: all possible helices. I discuss the manufacture and manipulation of such devices, and illustrate a few use cases where they will likely outperform conventional needles. With some collective action from the research community, curved arrays could be manufactured and distributed at low cost.

Abstract:Cells are the basic unit of life. Electrochemical analysis of single cells/organelles is essential for uncovering the molecular mechanisms of physiological and pathological processes that are difficult to elucidate on a larger scale. This paper provides an overview of the commonly used fabrication methods for micro/nanoelectrodes applied in the investigations of single cells/organelles as well as the corresponding electrochemical measurements over the last four years including extracellular measurement, combination of extra and intracellular measurement, intracellular reactive oxygen species and reactive nitrogen species (ROS/RNS) measurement, and isolated organelles measurement.

A micro cell lysis device

Abstract:A new micromachined cell lysis device is developed. It is designed for miniature bio-analysis systems where cell lysing is needed to obtain intracellular materials for further analysis such as DNA identification. It consists of multi-electrode pairs to apply electric fields to cells. We adopt the means of using electric field lysing because it can greatly simplify purification steps for preparation of biological samples compared to conventional chemical methods. Yeast, Chinese cabbage, radish cells and Escherichia coli are tested with the device. The lysis of yeast, Chinese cabbage and radish cells is observed by a microscope. The experimental observation suggests that E. coli are also lysed by the pulsed electric field. The range of electric field for the lysis is on the order of 1 kV/cm to 10 kV/cm. The Teflon coated on the electrodes can protect the electrodes during the pulsing period. In addition, for practical reasons, we reduce the voltage required for lysing to less than 10 V by making the electrode gap on the order of microns.

Abstract:Microband electrodes display a very strong edge effect. The diffusion layer at a microband electrode may be considered as a semicylinder. When a microband-array electrode is electrolysed for a long time, these diffusion layers partially overlap each other and the total current is smaller than the sum of the currents at all single-microband electrodes. This is the so-called shielding effect. The degree of shielding can be expressed by a shielding factor Sf which depends on the electrolysis time, the distance between electrodes and the electrode width. A formula to calculate the shielding factor Sf is presented here. The results of theoretical calculations are found to be in good agreement with experimental results reported previously.
Nanofocused Electric Field for Localized Single Cell Nanoelectroporation with Membrane Reversibility.

Abstract:Despite the significant research in electroporation, high electric field was applied to the whole cells resulted in permeabilizing the membrane of millions of cells without reversibility [1]. To deliver biomolecules through the specific region of the cell membrane with high cell viability and high transfection rate is important for various biological and therapeutic applications.This report presents a new type localized single cell membrane electroporation (LSCMEP), at specific region of the single cell with the application of 800 μs electric pulse. The ITO nano-electrodes with 100nm thickness and 500 nm gap between two electrodes can generate an intense electric field to track biomolecules inside HeLa cell in our studies. This small gap between two nano-electrodes can neglect thermal effect on cell membrane and permit reversible electroporation with high cell viability (90%) and minimum effected electroporation region (0.48 μm). Our approach successfully delivers biomolecules through a specific region of single cell with high transfection rate (82%) and high cell viability. This process, not only generates well-controlled nano-pores allowing rapid recovery of cell membrane, but also it provides a clear optical path potentially tracking of drugs to deliver inside single cell.
[Comparison of migration characteristics of MSCs in different assay systems].

Abstract:The aim of this study was to explore the difference of MSC migration mediated by SDF-1/CXCR4 axis through Boyden chamber in vitro migration assay. The SDF-1 density-dependence of MSC migration was observed. Subsequently, the effects of different blocking agents on hSDF-MSC migration were observed after MSC were treated with 50 nmol/L wortmannin, 10 micromol/L LY294002, 50 micromol/L PD98059, 10 micromol/L U73122, 126 micromol/L AMD3100 and 50 nmol/L verapamil respectively. The results showed the efficiency of MSC migration increased gradually with the increasing of hSDF-1 density. And after MSCs treatment with 50 nmol/L wortmannin, 10 micromol/L LY294002, 50 micromol/L PD98059, 10 micromol/L U73122 and 126 micromol/L AMD3100 respectively, the ability of MSC migration decreased. The ability of MSCs migration obviously decreased when MSCs were treated with U73122, AMD3100. It is concluded that the SDF-1/CXCR4-mediated MSC migration may be related to mitogen-activated protein kinase (MAPK), phosphatidylinositol phospholipase C (PI-PLC) and protein kinase (PKC) signal pathways.
Circular shaped microelectrodes for single cell electrical measurements for lab-on-a-chip applications

Abstract:The impedimetric sensing techniques for single cell characterization have witnessed growing interest due to their high sensitivity and widespread applications. However, adapting the method to different biological measurements in microfluidic environments under various input conditions can result in feeble signal detection leading to a drastic decrease in the sensor sensitivity. The reduced signal-to-noise ratio (SNR) hinders the signal differentiation, sensor accuracy and prohibits fully integrated point-of-care applications. Here, we address the sensitivity enhancement for microfluidic impedimetric sensing of micron and submicron-sized microparticles by exploring novel circular shape electrodes in a simulation study. The influence of radial electrode parameters on differential electrical signal is systematically analyzed in COMSOL Multiphysics using spherical particles ranging from 0.75 µm to 5 µm in diameter. Detailed analysis revealed the strong impact of the circular shape microelectrode geometry and the electrode gap on the signal strength, resulting SNR, and device sensitivity for multiple bioparticles detection. Specifically, ˃ 50 dB improvement in SNR was enabled by optimizing the circular electrode geometrical parameters. Our proposed sensing modality can be adapted for nanoparticles detection by further optimizing the microfluidic device parameters.
