Abstract:Myocardial infarction (MI) remains a major challenge to cardiovascular health worldwide, with poor healing leaving a direct impact on patients' quality of life and survival. Metabolic abnormalities after MI are receiving increasing attention. Our previous studies showed that enhancing proline catabolism ameliorates hypoxic damage to myocardial cells; therefore, we sought to determine whether reducing the synthesis of endogenous proline also affects MI. We analysed GEO datasets associated with MI and western blot of mouse heart tissue in an MI model to demonstrate pyrroline-5-carboxylate reductase 1 (Pycr1) expression level after MI. We constructed Pycr1 KO mice by CRISPR/Cas9 technology to explore the effect of Pycr1 gene KO after MI using transcriptomic and metabolomic techniques. In this study, we found reduced mRNA and protein expression levels of Pycr1 in the hearts of mice after MI. We observed that Pycr1 gene KO has a protective effect against MI, reducing the area of MI and improving heart function. Using transcriptomics approaches, we found 215 upregulated genes and 247 downregulated genes after KO of the Pycr1 gene, indicating that unsaturated fatty acid metabolism was affected at the transcriptional level. Metabolomics results revealed elevated content for 141 metabolites and decreased content for 90 metabolites, among which the levels of fatty acids, glycerol phospholipids, bile acids, and other metabolites increased significantly. The changes in these metabolites may be related to the protective effect of Pycr1 KO on the heart after MI. Pycr1 gene KO has a protective effect against MI and our research will lay a solid foundation for the development of future Pycr1-related drug targets.

Abstract:MicroRNA-34a (miR-34a) promotes apoptosis via down-regulating many anti-apoptotic proteins. Aldehyde dehydrogenase 2 (ALDH2) is an anti-apoptotic enzyme whose activity decline associates with myocardial injury. We tested hypothesis that miR-34a might play a pro-apoptotic role in myocardial infarction (MI) by down-regulating ALDH2. MiR-34a was highly increased while ALDH2 expression was decreased after experimental MI. Overexpression of miR-34a in neonatal rat cardiomyocyte could significantly enhance apoptosis and down-regulate ALDH2 expression. In 293 cells, luciferase reporter assay results demonstrated that ALDH2 was a direct target of miR-34a. Serum miR-34a levels in acute myocardial infarction (AMI) patients and rats were significantly higher than healthy subjects and sham rats. Our results proved that miR-34a could promote cardiomyocyte apoptosis via negatively regulating ALDH2 and circulating miR-34a was increased in the condition of MI. Thus, miR-34a may constitute a new therapeutic target and diagnostic marker for patients with MI.
Irisin protects cardiomyocytes against hypoxia/reoxygenation injury via attenuating AMPK mediated endoplasmic reticulum stress

Abstract:Endoplasmic reticulum (ER) stress plays a central role in myocardial ischemia/reperfusion (I/R) injury. Irisin has been reported to have protective properties in ischemia disease. In this study, we aimed at investigating whether irisin could alleviate myocardial I/R injury by ER stress attenuation. The in vitro model of hypoxia/reoxygenation (H/R) was established, which resembles I/R in vivo. Cell viability and apoptosis were estimated. Expressions of cleaved caspase-3, cytochrome c, GRP78, pAMPK, CHOP, and eIF2α were assessed by western blot. Our results revealed that pre-treatment with irisin significantly decreased cytochrome c release from mitochondria and caspase-3 activation caused by H/R. Irsin also reduced apoptosis and increased cell viability. These effects were abolished by AMPK inhibitor compound C pre-treatment. Also, GRP78 and CHOP expressions were up-regulated in the H/R group compared to the control group; however, irisin attenuated their expression. The pAMPK level was significantly decreased compared to the control, and this effect could be partly reversed by metformin pre-treatment. These results suggest that ER stress is associated with cell viability decreasing and cardiomyocytes apoptosis induced by H/R. Irisin could efficiently protect cardiomyocytes from H/R-injury via attenuating ER stress and ER stress-induced apoptosis.
Cryptotanshinone Alleviates Myocardial Ischemia and Reperfusion Injury in Rats to Mitigate ER Stress-Dependent Apoptosis by Modulating the JAK1/STAT3 Axis

Abstract:Objective: Myocardial ischemia is the stoppage or insufficiency of blood flow to the myocardium, depriving cells of oxygen supply which leads to their apoptosis or death. Currently, the management of patients has improved, making it possible to reduce myocardial infarction injury with new strategies of reperfusion and pharmacologic treatment. Methods: A rat model of myocardial ischemia and reperfusion injury (MIRI) was created and subjected to cryptotanshinone (CRY) with or without JAK1 inhibitor filgotinib (FILGO) treatment. H&E staining was used for histopathologic evaluation of heart injury, and TTC staining was employed for evaluation of the infarct size. Western blotting and immunofluorescence were used to measure the protein expression and qRT-PCR for determining mRNA expression. Results: CRY significantly reduced the area of the infarct, the number of apoptotic cells, and the concentrations of lactate dehydrogenase (LDH) and creatine kinase-MB (CK-MB) induced by ischemia/reperfusion (I/R). Subsequent analysis showed that CRY repressed the expression of caspase-12, CHOP, and GRP78, but enhanced the phosphorylation of JAK and STAT3. However, FILGO treatment markedly abolished the beneficial effect of CRY pretreatment on cardiomyocyte damage, apoptosis, cardiac function, and inhibition of endoplasmic reticulum stress (ERS)-dependent apoptosis marker proteins. Conclusion: CRY may alleviate MIRI by inhibiting ERS-dependent apoptosis by activating the JAK1/STAT3 signaling pathway.
Crocin protects against endoplasmic reticulum stress-related tubular injury in diabetic nephropathy via the activation of the PI3K/AKT/Nrf2 pathway

Abstract:Objectives: Diabetic nephropathy (DN) is the main cause of end-stage renal disease, but the current treatment is not satisfactory. Crocin is a major bioactive compound of saffron with antioxidant and anti-endoplasmic reticulum stress (ERS) abilities used to treat diabetes. This study specifically investigated whether crocin has a regulatory role in renal injury in DN. Materials and methods: The experiment was divided into control, (db/m mice), model (db/db mice), and experimental groups (db/db mice were intraperitoneally injected with 40 mg/kg crocin). Renal function-related indicators (Scr, BUN, FBG, UP, TG, TC, ALT, and AST) and oxidative stress-related indicators (ROS, MDA, GSH, SOD, and CAT) were assessed. The pathological changes of renal tissues were confirmed by HE, Masson, PAS, and TUNEL staining. The levels of ERS-related proteins (GRP78 and CHOP), apoptosis-related proteins, and PI3K/AKT and Nrf2 pathways-related proteins in renal tissue were detected. Results: In db/db mice, renal function-related indicators, apoptotic cells of renal tissues, the contents of ROS and MDA as well as the expressions of CHOP, GRP78, and Bax were increased, the degree of renal tissue damage was aggravated, while the contents of GSH, SOD, and CAT, as well as the protein levels of Nrf2, PARP, anti-apoptotic proteins (Mcl-1, Bcl-2, Bcl-xl) were decreased compared to the db/m mice. However, crocin treatment reversed the above-mentioned situation. The expressions of the PI3K/AKT and Nrf2 pathways-related proteins were also activated by crocin. Conclusion: Crocin inhibited oxidative stress and ERS-induced kidney injury in db/db mice by activating the PI3K/AKT and Nrf2 pathways.

Abstract:The Green's-function Monte Carlo method is used to calculate the ground-state wave function of the muonic-molecular ion composed of a negative muon bound to a deuteron and a triton. Using the sudden approximation, the probability that the muon will remain bound to the escaping alpha particle after fusion occurs is found to be 0.90%, about 25% smaller than previous estimates based on the Born-Oppenheimer approximation. The numerical method for determining the wave function is discussed in detail.

Abstract:Diabetes and myocardial ischemia reperfusion (MIR) injury are characterized by oxidative stress, inflammation, autophagy disorders, and cardiac contractile dysfunction. Klotho and SIRT1 regulate the level of oxidative stress to participate in the regulation of many physiological functions such as cell survival, aging, apoptosis, autophagy, mitochondrial biogenesis, and inflammation. We hypothesized that the activation of Klotho/SIRT1 signaling pathway could attenuate MIR in diabetic rats. Type 1 diabetes and MIR injury model were established to examine this hypothesis in vivo . Primary rat cardiomyocytes and H9c2 cells were exposed to high glucose conditions and hypoxia/reoxygenation (H/R) insult in vitro . Hemodynamic parameters of heart function, myocardial infarct size, oxidative stress, markers of MIR injury or cell viability, and the mRNA and protein expression of Klotho and SIRT1 were measured. There was lower expression of Klotho and SIRT1 in diabetic MIR hearts than in nondiabetic rats, as well as significantly increased oxidative stress levels and decreased autophagy levels. Recombinant Klotho (rKlotho) protein and the SIRT1 agonist SRT1720 could significantly attenuate MIR injury in diabetes by activating Klotho/SIRT1 signaling pathway to reduce oxidative stress and restore autophagy levels. These findings suggest that the Klotho/SIRT1 pathway plays an important role in MIR injury in diabetic rats, and rKlotho protein and agonist SRT1720 have therapeutic potential for alleviating diabetic myocardial IR injury by activating Klotho/SIRT1 to reduce oxidative stress and restore autophagy levels.

Abstract:Introduction This study aimed to investigate the preventive and therapeutic effects of crocetin (Cro) on ischaemic stroke in cell and animal models. Material and methods A cell model of oxygen and glucose deprivation (OGD) and a rat model of middle cerebral artery occlusion were established to simulate ischaemic stroke. The infarct volume was measured by TTC assay, and the apoptotic cell number was counted by TUNEL. Relative protein and gene expression levels in the rats were measured by immunohistochemical and RT-qPCR assays. The apoptosis rate and relative protein and gene expression levels were determined by flow cytometry, WB and RT-qPCR assay, respectively. Results Compared with those in the normal control (NC) group, the brain tissue injury and apoptotic cell number significantly increased (p < 0.001) and the miR-145-5p gene expression significantly decreased in the cell and animal experiments. In the animal experiment, the infarct volume, apoptotic cell number and pathological status improved in the Cro-treated groups. In the cell experiment, the apoptosis rates significantly depressed in the Cro-treated groups (p < 0.05). However, the cell apoptosis rate significantly increased after miR-145-5p inhibitor transfection (p < 0.001). The protein and gene expression levels of Toll-like receptor 4, myeloid differentiation factor 88 and nuclear factor (NF)-κB (p65) significantly decreased (p < 0.05). In addition, p-NF-κB(p65) nuclear volume significantly decreased (p < 0.05). Conclusions Crocetin improved ischaemic stroke by regulating the miR145-5p/TLR4 axis in cell and animal experiments.

The silent information regulatory factor 1 (SIRT1), peroxisome proliferators‐activated receptor‐γ coactivator‐1α (PGC‐1α), and Bcl‐2 proteins expression of quercetin treated were significantly upregulation compared with MC group ( P < 0.05, respectively), and Bax protein expression of quercetin treated group was significantly downregulation compared with MC group ( P < 0.05, respectively). In the vivo study, the myocardial pathological morphology of quercetin treated groups was improved. The cell apoptosis number of quercetin treated group were significantly suppressed compared with MC group by terminal deoxynucleotidyl transferase dUTP nick end labeling assay ( P < 0.05, respectively). SIRT1, PGC‐1a, Bcl‐2, and Bax proteins expressions of quercetin treated groups were significant differences compared with MC group in myocardial tissue ( P < 0.05, respectively). Conclusion Quercetin had improved the myocardial ischemia/reperfusion‐induced cardiomyocyte apoptosis via SIRT1/PGC‐1α signaling.
Inhibition of Endoplasm Reticulum Stress by Ghrelin Protects Against Ischemia/reperfusion Injury in Rat Heart.

Abstract:Ghrelin is a multi-functional polypeptide with cardiovascular protective effects. We aimed to explore whether the cardioprotective effect of ghrelin is mediated by inhibiting myocardial endoplasmic reticulum stress (ERS). A Langendorff model of isolated rat heart was used with ischemia/reperfusion (I/R; 40/120min). Cardiac function was monitored, and histomorphologic features, degree of myocardial injury, level of ERS markers, and number of apoptotic cardiomyocytes were determined. Compared with control group, the I/R group showed significantly decreased cardiac function, seriously damaged myocardial tissue, increased number of apoptotic cells, and overexpression of mRNA and protein of ERS markers. However, preadministration of ghrelin in vivo (10−8mol/kg, intraperitoneal injection, every 12h, twice in all) greatly ameliorated the damaged heart function, attenuated myocardial injury and apoptosis, and decreased the expression of ERS markers: it decreased the mRNA and protein levels of glucose-regulated protein78 (GRP78) and C/EBP homologous protein (CHOP), with reduced caspase-12 protein expression. Furthermore, in vitro, ghrelin directly inhibited the myocardial ERS response induced by tunicamycin or dithiothreitol in rat cardiac tissue. Ghrelin could protect the heart against I/R injury, at least in part, through inhibiting myocardial ERS.

Does Sepsis Case Mix Heterogeneity Prevent Outcome Comparisons?

For starters, it is a syndrome with etiologic agents not only across three taxonomic kingdoms of life (bacteria, protozoa, and fungi) but also outside of the taxonomy of life (viruses). Multiplying the possible flavors of sepsis, many of these pathogens can infect a variety of human tissues, and each infective episode can in turn induce different combinations of systemic inflammatory responses and distant organ failures. Just how many flavors of sepsis can there be? As a simple thought experiment, let us ignore the rainbow of etiologic agents, assume one source of infection per sepsis episode and narrow the field of organ dysfunctions to five. To get the total number of sepsis combinations under these stipulations, we multiply the five infection sources by the 11 different combinations of systemic inflammatory response syndrome (SIRS) criteria that meet a definition of sepsis by the 31 combinations of organ dysfunctions (Fig. 1). This gives us 1,705 shades of sepsis, admittedly only the tip of the iceberg given that we have ignored the myriad of etiologic agents. Yet, like choosing which variation of beige to paint your dining room, the important question is whether there is any meaningful difference between these myriad tones of sepsis? The article published in this issue of Critical Care Medicine by Shankar-Hari et al (2) adds to a body of evidence suggesting that there “is” a meaningful difference. To summarize, the article is a cohort study of the 241,603 adults admitted with a diagnosis of infection to a general critical care unit in England from 2000 to 2012. Systemic inflammatory response and organ dysfunction criteria were extracted from physiologic data within the first 24 hours of critical care admission and hospital mortality was the primary outcome. The study uses data from the Case Mix Programme, a high quality, professionally and independently maintained database of nearly every patient of every general critical care unit in the United Kingdom (3). The analyses examine the unadjusted hospital mortality of sepsis from many different angles: source of infection, quantity and combination of SIRS criteria, and quantity and combination of organ dysfunctions. The results display a wide range of hospital mortality along every facet. For source of infection, hospital mortality was lowest for genitourinary infections (19.4%) and highest for cardiovascular infections (44.2%). For SIRS criteria, even within groupings of two and three total criteria met within the first 24 hours of critical care admission, different combinations were associated with wide within-group variation in unadjusted hospital mortality (from approximately 20–40%, estimating from the graphs). Similarly, among different combinations of organ dysfunctions, there is a considerable range of unadjusted hospital mortality rates. From this data, the investigators create four risk categories of organ dysfunction combinations, grouped by the associated hospital mortality rates rather than the sum or physiologic relationships of organ dysfunctions. They demonstrate that within each risk category, hospital mortality has followed a similar downward trend from 2000 to 2012, an observation that increases confidence in the notion that overall sepsis mortality is improving due to changes in care rather than increases in diagnosis of less severe cases. Furthermore, the investigators use these risk categories to hypothesize an explanation as to why the hospital mortality from sepsis in England was on average 1.5 times higher than that reported in a well-known study over the same time period in Australia and New Zealand (4). Their analyses suggest that the sepsis case mix in England represents a higher risk case mix than that found in Australia and New Zealand. However, while they illustrate that overall mortality reported in Australia and New Zealand is similar to the mortality from risk category 2 sepsis in England, we do not see the risk categories of the Australia and New Zealand data, limiting any direct comparisons. The study by Shankar-Hari et al (2) graphically depicts the wide variations in unadjusted hospital mortality of a wide variety sepsis syndromes defined by various combinations of infection source, SIRS criteria, and organ dysfunctions. Although this is an important and well-presented reminder of the significant variety under the umbrella of sepsis, many of the concepts are not new. The notion that increasing numbers of abnormal vital signs and organ dysfunctions was associated with increased mortality in sepsis was demonstrated in the early clinical studies of sepsis from the mid-1990s (5–7). Abstract:Intracerebral hemorrhage (ICH) is the deadliest type of stroke. Oxidative stress was considered to play an important role in ICH-induced secondary injury. Crocin, the main compound isolated from Crocus sativus L., possesses a potential anti-oxidative function in many types of diseases including ICH. In the current study, the protective role of crocin in ICH-induced brain injury was investigated in the ICH model. The ICH-induced brain edema and neurological deficits were analyzed by brain edema measurement and neurological testing. The superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) activity and the content of malondialdehyde (MDA) were assessed by a total superoxide dismutase assay kit. The expressions of ferroptosis-related genes were verified by quantitative real-time PCR (qPCR) and western blotting. The ICH-induced brain edema and neurological deficits were significantly decreased after treatment with crocin. Moreover, the SOD and GSH-px activities were obviously increased in the ICH with crocin-treated group compared with the ICH group, while the content of MDA was markedly decreased after treatment with crocin. Crocin inhibited ferroptosis of neuron cells, as evidenced by increased Fe2+ concentration and the expression of GPX4, FTH1, and SLC7A11. Mechanistically, crocin treatment increased the expression and nuclear translocation of Nrf2. Our data suggest that crocin alleviates intracerebral hemorrhage–induced neuronal ferroptosis by facilitating Nrf2 nuclear translocation.

SIRT1 Suppresses Doxorubicin-Induced Cardiotoxicity by Regulating the Oxidative Stress and P38mapk Pathways

Abstract:Background: SIRT1, which belongs to the Sirtuin family of NAD-dependent enzymes, plays diverse roles in aging, metabolism, and disease biology. It could regulate cell survival and has been shown to be a protective factor in heart function. Hence, we verified the mechanism by which SIRT1 regulates doxorubicin induced cardiomyocyte injury in vivo and in vitro. Methods: We analyzed SIRT1 expression in doxorubicin-induced neonatal rat cardiomyocyte injury model and adult mouse heart failure model. SIRT1 was over-expressed in cultured neonatal rat cardiomyocyte by adenovirus mediated gene transfer. SIRT1 agonist resveratrol was used to treat the doxorubicin-induced heart failure mouse model. Echocardiography, reactive oxygen species (ROS) production, TUNEL, qRT-PCR, and Western blotting were performed to analyze cell survival, oxidative stress, and inflammatory signal pathways in cardiomyocytes. Results: SIRT1 expression was down-regulated in doxorubicin induced cardiomocyte injury, accompanied by elevated oxidative stress and cell apoptosis. SIRT1 over-expression reduced doxorubicin induced cardiomyocyte apoptosis with the attenuated ROS production. SIRT1 also reduced cell apoptosis by inhibition of p38MAPK phosphorylation and caspase-3 activation. The SIRT1 agonist resveratrol was able to prevent doxorubicin-induced heart function loss. Moreover, the SIRT1 inhibitor niacinamide could reverse SIRT1's protective effect in cultured neonatal rat cardiomyocytes. Conclusions: These results support the role of SIRT1 as an important regulator of cardiomyocyte apoptosis during doxorubicin-induced heart injury, which may represent a potential therapeutic target for doxorubicin-induced cardiomyopathy.

Abstract:MicroRNAs have been shown to play critical regulatory roles in various ischemia/reperfusion (I/R) injury. However, its role in myocardial I/R injury remains largely unknown. The aim of this study was to investigate the role of microRNA-34a (miR-34a) in the development of myocardial I/R injury. Rat myocardial I/R model was established, followed by PCR array analyses to investigate the expressional alterations of miRNAs in the hearts of rats. H9c2 cardiomyocytes were subjected to hypoxia/reoxygenation (H/R), and lactate dehydrogenase (LDH) release, cell viability and rat cardiomyocyte apoptosis were measured as well as miR-34a expression. Knockdown experiments were performed to determine the effects of manipulating miR-34a on apoptotic responses. Our results revealed that miR-34a was significantly up-regulated in I/R group compared with sham group. Inhibition of miR-34a promoted cell proliferation, reduced LDH release and apoptosis which were accompanied by a significant decrease in caspase-3 activity. Furthermore, Bcl-2, a well know anti-apoptotic gene was identified to be a functional target of miR-34a. Taken together, knockdown of miR-34a mediates H/R-induced apoptosis in cardiomyocytes by targeting Bcl-2 and represents a potential target for prevention of myocardial I/R injury.

Abstract:Objective: This meta-analysis aimed to determine the efficacy of curcumin in preventing myocardial ischemia/reperfusion (I/R) injury in animal models. Methods: Studies published from inception to January 2023 were systematically searched in databases including PubMed, Web of Science, Embase, China's National Knowledge Infrastructure (CNKI), Wan-Fang database, and VIP database (VIP). The SYRCLE's RoB tool was used to determine methodological quality. Sensitivity analysis and subgroup analysis were performed when there was high heterogeneity. Publication bias was assessed using a funnel plot. Results: Thirty-seven studies involving 771 animals were included in this meta-analysis with methodology quality scores ranging from 4 to 7. The results indicated that curcumin treatment significantly improved myocardial infarction size standard mean difference (SMD) = −5.65; 95% confidence interval (CI): 6.94, −4.36; p < 0.01; I 2 = 90%). The sensitivity analysis for infarct size showed that the results were stable and reliable. However, the funnel plot was asymmetric. The subgroup analysis included species, animal model, dose, administration, and duration. The results showed that the subgroup dose was statistically significant between subgroups. In addition, curcumin treatment improved cardiac function, myocardial injury enzymes, and oxidative stress levels in animal models of myocardial I/R injury. The funnel plot revealed that there is publication bias for creatine kinase and lactate dehydrogenase. Finally, we performed a meta-analysis of inflammatory cytokines and apoptosis index. The results showed that curcumin treatment downregulated serum inflammatory cytokine levels and myocardial apoptosis index. Conclusion: This meta-analysis suggests that curcumin has excellent potential for the treatment of myocardial I/R injury in animal models. However, this conclusion needs to be further discussed and verified in large animal models and human clinical trials. Abstract:Background Ischemia reperfusion (I/R) injury is a frequent occurrence during liver transplantation surgery, resulting from the temporary cessation of blood flow and subsequent restoration of blood flow. Serious I/R injury is a significant factor causing transplant failure. Hepatic I/R process is characterized by excessive inflammation, oxidation, and apoptosis. Crocetin (Crt) is a natural compound exhibiting beneficial roles in various I/R-induced organ damages. However, Crt's potential role in hepatic I/R remains unexplored. Objective and Methods In order to reveal the impact of Crt on hepatic I/R and the associated signaling pathway, we utilized a syngeneic orthotopic liver transplantation rat model to induce hepatic I/R injury. Results Pretreatment with Crt significantly mitigated hepatic I/R injury. This was evident by decreased activities of serum ALT, AST and LDH, indicating improved liver function. Crt treatment also alleviated oxidative stress, as demonstrated by decreased serum MDA content and elevated serum SOD and GSH-Px activities. Furthermore, Crt suppressed inflammatory responses by downregulating both the serum and liver IL-1β, IL-6 and TNF-α while upregulating IL-10 expression. Additionally, Crt reduced apoptosis by decreasing pro-apoptotic Bax, cleaved caspase-3 and cleaved caspase-9, while increasing anti-apoptotic Bcl2 expression. Notably, these protective effects of Crt were dose-dependent. Moreover, our data indicates that Crt plays protective functions during hepatic I/R via disrupting Keap1/Nrf2 interaction and activating Nrf2/HO-1 signaling. This was further supported by observations of alleviated hepatic histopathological changes in I/R rats treated with Crt. Conclusions Crt shows potential as a therapeutic agent for preventing hepatic I/R injury during clinical liver transplantation.

Abstract:Background: Recent reports suggested the involvement of oxidative stress- and endoplasmic reticulum stress (ERS)-associated pathways in the progression of ischemia/reperfusion (I/R) injury. Notoginsenoside R1 (NGR1) is a novel saponin isolated from P. notoginseng, which has a history of prevention and treatment of cardiovascular diseases. Objective: We aimed to examine the cardioprotective effects of NGR1 on I/R-induced heart dysfunction ex vivo and in vitro. Methods: H9c2 cadiomyocytes were incubated with NGR1 for 24 h and exposed to hypoxia/reoxygenation. Isolated rat hearts were perfused by NGR1 for 15 min and then subjected to global ischemia/reperfusion. Hemodynamic parameters were monitored as left ventricular systolic pressure (LVSP), heart rate, and maximal rate of increase and decrease of left ventricular pressure (± dP/dt max/min). Results: NGR1 pretreatment prevents cell apoptosis and delays the onset of ERS by decreasing the protein expression levels of ERS-responsive proteins GRP78, P-PERK, ATF6, IRE, and inhibiting the expression of pro-apoptosis proteins CHOP, Caspase-12, and P-JNK. Besides, NGR1 scavenges free radical, and increases the activity of antioxidase. NGR1 inhibits Tunicamycin-induced cell death and cardiac dysfunction. Conclusion: We elucidated the significant cardioprotective effects of NGR1 against I/R injuries, and demonstrated the involvement of oxidative stress and ERS in the protective effects of NGR1.
