Abstract:In this study, inulin was extracted from Jerusalem artichoke tuber (JAT) powder and then concentrated before spray drying. The aims of this study were to (1) determine the drying condition that provided high powder yield together with superior qualities of JAT inulin powder and (2) investigate the chemical and physical properties of inulin powder. The inulin extracts at different concentrations of 10, 20, and 30 °Brix were spray-dried and then compared. The spray drying experiments were conducted at the inlet/outlet air temperatures of 150/90, 170/90, and 190/90 °C for the chosen concentration of inulin extract. It appeared that spray drying of the 30 °Brix extract at the inlet/outlet drying air temperatures of 190/90 °C resulted in the highest value of powder recovery, bulk density, water solubility and the lowest moisture content and hygroscopicity in comparison with its counterparts. SEM micrographs showed that the powder produced by this condition was more stable and less sticky than others. The sugars, total fructo-oligosaccharides (1-kestose, nystose, and 1F-β-fructofuranosyl nystose) and inulin-type fructans contents of powder were 12.88, 11.12, and 64.36 g/100 g of powder, respectively. The moisture sorption data and models developed in this work could be used for determining the suitable condition of surrounding air for inulin powder storage.

Abstract:Inulin is used as an important food ingredient, widely used for its fiber content. In this study the operational extraction variables to obtain higher yields of inulin from Jerusalem artichoke tubers, as well as the optimal conditions, were studied. Response surface methodology and Box-Behnken design were used for optimization of extraction steps. The optimal extraction conditions were as follows: extraction temperature 74 °C, extraction time 65 min, and ratio of liquid to solid 4 mL/g. Furthermore, series connection of ion-exchange resins were used to purify the extraction solution where the optimal resin combinations were D202 strongly alkaline anion resin, HD-8 strongly acidic cation resin, and D315 weakly alkaline resin while the decolorization rate and decreased salinity reached 99.76 and 93.68, respectively. Under these conditions, the yield of inulin was 85.4 ± 0.5%.

Abstract:Ultrasonic extraction is an inexpensive, simple and efficient alternative to conventional extraction techniques, as compared with other novel extraction techniques such as microwave-assisted extraction & supercritical fluid extraction techniques, the ultrasound apparatus is cheaper and its operation is easier. Ultrasound assisted extraction has risen rapidly in the latest decade, and for most applications it has proven to be effective compared to traditional extraction techniques. In this paper, a method of ultrasonic-assisted extraction was used to extract Inulin from tubers of Jerusalem artichoke, which have been reported to have several medicinal properties and uses. Inulin is a storage carbohydrate found in many plants especially in chicory root, Jerusalem artichoke and dahlia tuber. In this study, the effect of time, temperature, pH and solid to liquid ratio on Inulin extraction from Jerusalem artichoke tubers by using ultrasonic water bath. The highest yield of Inulin were investigated from Jerusalem artichoke tuber was (99.47%) at temperature of 70°C, pH=7, 60 min and ratio of solid to solvent was (10gm/100ml). Then, The UV detector by colorimetric method with vanillin–sulfuric acid was used for the quantification of Inulin.

Abstract:The expression of intercellular adhesion molecule-1 (ICAM-1) on pulmonary endothelial cells after stimulus and subsequent binding of neutrophils is a first step leading to lung injury. A similar process may dictate the binding of tumor cells to the pulmonary endothelium during metastasis. We report the development of a new technique that allowed us to monitor the location and relative expression of ICAM-1 levels on the luminal surface of the pulmonary microvasculature in vivo. This technique uses intravital microscopy together with a two-step labeling procedure involving fluorescent microspheres. Constitutive expression of ICAM-1 was not detectable to a significant level by our model, but expression was observed after upregulation by the systemic administration of TNF alpha. Sprague-Dawley rats were injected with 0-5.0 micrograms/kg TNF alpha and ICAM-1 expression was monitored through 24 hr. ICAM-1 expression was related to both the dose of TNF alpha administered and the time elapsed between injection of TNF alpha and observation. Injection of 5 micrograms/kg TNF alpha caused upregulation of ICAM-1 protein expression from 0.30 +/- 2.76 binding events/175,000 microns2 to 62.6 +/- 5.48 through 4 hr observation, after which levels returned to near baseline within 24 hr. The delay required for maximal expression is likely related to the time required for the cell to respond to the stimulus and generate ICAM-1 protein. Reductions in the relative numbers of ICAM-1 protein expressed between 4 and 24 hr in vivo are likely a result of protein turnover after the initial stimulus.

Abstract:An increase in inulin and plant-protein based nutraceutical demand ultimately puts pressure on available resources. Therefore, there is a need to prospect for supplementary feedstocks and sustainable ways to exploit them. The aim of this study was to explore the technical feasibility of sequential extraction of inulin and protein from Jerusalem artichoke tubers and understand the interrelationships between processes and product functional properties. The response surface methodology was used to determine the optimal parameters for sequential extraction. Protein functional properties analysis was done to identify the effects of the extraction process. The extraction approach adopted in this study was preceded by mechanical pressing of the tuber to yield a protein-rich juice. However, only 40.8% of the protein was recovered from the juice, therefore a subsequent solvent extraction step followed to extract the residual protein and inulin retained in the solids. Selective extraction was achieved when protein was solubilised in the first step of solvent extraction. The overall protein and inulin yields from pressing and both sequential extraction steps were 71.88 and 67.6%, respectively. The inulin yields were substantially higher than the maximum overall yields when inulin extraction, from the pressed tuber, was performed first thus improving yields from 57.3 to 67.6%. Consequently, mechanical pressing improved the overall protein yield. Sequential extraction resulted in an inulin extract with minimal protein contamination compared to the conventional method. Therefore, sequential extraction was efficient in yielding extracts with reduced impurities and good functional properties.

Abstract:Inulin has become an attractive ingredient in the food industry because of its nutritional and functional characteristics. Extraction of inulin from Jerusalem artichoke and the effects of its enzymatic hydrolyzate on the textural and sensorial characteristics of yogurt were investigated. The extraction yield (98%) of inulin was obtained with dry Jerusalem artichoke powder. The thin-layer chromatography and the ion chromatogram analysis of the enzymatic hydrolyzate revealed that inulinase hydrolyzed inulin into fructooligosaccharides with degrees of polymerization from 2 to 10. Utilization of inulin and hydrolyzate in yogurts improved their textural properties. Sensorial evaluation of yogurts showed that yogurts with inulin or its hydrolyzate had a better body and smoother texture than yogurts with sucrose only. The findings of this study suggest that inulin or its hydrolyzate can partially replace sucrose in formulations in the food industry.

Abstract:According to global analytical agencies the most promising area of development of the food industry is the enrichment of food products with physiologically functional ingredients – dietary fiber, vitamins, minerals, etc. Analysis of the market for functional ingredients indicates the prospects of organizing the production of domestic inulin and other biologically active components from various anatomical parts herbaceous tuberous plant of the Sunflower genus – Jerusalem artichoke, the distinctive features of which are high yield of tubers, unpretentiousness to cultivation conditions, resistance to diseases and pests. The purpose of the research was to develop a technology for the production of inulin and chlorophyll from Jerusalem artichoke tubers and leaves. The work used generally accepted physical and chemical methods for studying plant objects. The results of the research made it possible to establish optimal technological parameters for the process of extracting inulin from Jerusalem artichoke tubers with a 60% aqueous-alcohol solution. The technology has been proposed for the production of inulin from Jerusalem artichoke tubers, based on the fractionation of a mixture of components with different molecular weights, by passing them through polyamide composite membranes of reverse osmosis roll type. It has been established that the molecular weight of inulin isolated from Jerusalem artichoke tubers of the «Interes» variety is 5.84 kDa. It has been revealed that Jerusalem artichoke leaves contain a and b chlorophyll in amounts of 6.98 and 0.74 mg/g of wet matter. Copper chlorophyll derivatives isolated from Jerusalem artichoke leaves are promising not only as food colorings, but also as biologically active substances with a wide spectrum of action. The target products obtained using the proposed technology – inulin and copper derivatives of chlorophylls – can find wide practical application in the food industry and in public catering.

Abstract:The artichoke tuber is full of nutrients, inulin, and phytochemicals. It has been used to treat illnesses including diabetes and colon cancer, as well as in food product formulation, but limited information on the Jerusalem artichoke tuber (JAT) powder characterization exists in the literature, hence in this paper, JAT was freeze and oven-dried. It was powdered into JAT-freeze-dried-(FD)-powder and oven-dried (OD)-powder. This enabled the JAT powder's functional and physical properties to be studied. As a result, JAT powder's morphology, microstructure, and functional groups, as well as the powder foaming, swelling, solubility, antioxidant, color pasting, bulk, packed, and particle distribution properties were studied. Results indicated that the average particle distribution size at Dx90 and Dx80 displayed a distinct difference at p ≤ 0.05, while the bulk (0.39 g/cm-3) and packed (0.48 g/cm-3) densities recorded a lower value for FD powder. The FD powder's foaming capacity (24.0%) was significantly distinct (p ≤ 0.05) from the OD powder. Also, the solubility of FD powder was 6.2 g/g at 50 °C, and that of OD powder was recorded as 2.3 g/g. Again, the FD powder had a higher ABTS+ (34.3 mM (TE)/g dw) and CUPRAC (94.61 mM (TE)/g dw) capacity. Besides, a significant (p ≤ 0.05) dissimilarity among the powder color parameters (L∗, a∗, b∗, C∗, and whiteness) was observed. More so, the XRD and FT-IR characterization established a semi-crystalline or amorphous nature of the powder containing polysaccharides, and a broad halo pattern 2 θ at an angle 19.3° and 20 ° for FD powder and OD powder respectively. The FD powder particles were more agglomerated than those of OD powder. This was seen as a microscopic image, again FD powder revealed a higher pasting temperature and a drop in peak viscosity. Based on the results obtained, JAT (FD and OD) powder has all the quality attributes required of a powder for culinary product formulation.

Abstract:Jerusalem artichoke is promoted as a plant with great potential for use due to its specific chemical composition, having inulin as a reserve carbohydrate, unlike other plants that contain starch. Because the tubers deteriorate very easily during storage, it is recommended that the harvest be carried out according to use. In this work the soluble solids, inulin and reducing sugars content of three Jerusalem artichoke cultivars (Dacic, Rares and Dabuleni) grown at Agricultural Research Development Station Caracal was evaluated at two harvest times (autumn and spring). The obtained results show that the studied chemical indices vary depending on the analyzed cultivar and the stage of plant development. The tubers of the experimented Jerusalem artichoke cultivars present appreciable content of soluble solids and inulin at both harvest times. In all investigated cultivars, tubers harvested in spring have lower content of inulin and soluble solids and higher content of reducing sugars than those harvested in autumn.
Investigating the effect of different inulin-rich substrate preparations from Jerusalem artichoke (Helianthus tuberosus L.) tubers on efficient inulooligosaccharides production

Abstract:Commercial production of inulooligosaccharides (IOS) relies largely on chicory roots. However, Jerusalem artichoke (JA) tubers provide a suitable alternative due to their high inulin content and low cultivation requirements. In this study, three inulin-rich substrate preparations from JA were investigated to maximize IOS production, namely powder from dried JA tuber slices (Substrate 1), solid residues after extracting protein from the JA powder (Substrate 2) and an inulin-rich fraction extracted from protein extraction residues (Substrate 3). The preferred temperature, pH and inulin substrate concentration were determined after which enzyme dosage and extraction time were optimized to maximize IOS extraction from the three substrates, using pure chicory inulin as benchmark. Under the optimal conditions, Substrate 3 resulted in the highest IOS yield of 82.3% (w/winulin). However, IOS production from the Substrate 1 proved more efficient since it renders the highest overall IOS yield (mass of IOS per mass of the starting biomass). In the case of co-production of protein and IOS from the JA tuber in a biorefinery concept, IOS production from the Substrate 2 is preferred since it reduces the inulin losses incurred during substrate preparation. For all the inulin-rich substrates studied, an enzyme dosage of 14.8 U/ginulin was found to be optimal at reaction time less than 6 h. JA tuber exhibited excellent potential for commercial production of IOS with improved yield and the possible advantage of a reduced biomass cost.

Abstract:Abstract Jerusalem artichoke tubers (Helianthus tuberosus) are distinguished by their protein, minerals (potassium, calcium, magnesium, iron, etc.) and inulin content. Inulin can be used in the diet of diabetics as a substitute of sugar, without having an impact on blood glucose. At the same time, an international study had shown that due to their inulin content, regular consumption of Jerusalem artichoke tubers can help to prevent type 2 diabetes. In this paper are presented the results of the researches performed to achieve a functional ingredient (powder) with high nutritional value by processing of Jerusalem artichoke tubers. Thus, the Jerusalem artichoke tubers (Red Jerusalem artichoke and White Jerusalem artichoke varieties) were subjected to a convective drying process at 50°C, to protect bioactive components (vitamins, phenolic compounds, etc.) to a moisture content that allow their milling and turning them into powder and, at the same time, their stability in terms of quality. The achieved functional ingredient was evaluated sensory, physicchemically and microbiologically. The powder obtained from Jerusalem artichoke tubers is characterized by their inulin-type fructans (51.60... 57.45%), crude fiber (6.85...8.27%), total polyphenols (18.51... 44.03 mg GAE/g), proteins (8.75...9.26%), iron (12.45...13.88 mg/100g), potassium (1905.44...2100.35 mg/100g), calcium (50.21...57.45mg/100g), magnesium (84.55...89.95mg/100g) and phosphorus content (300.12...345.35 mg/100g). At the same time, powder achieved from Jerusalem artichoke tubers has antioxidant potential. Due to its complex biochemical composition, the functional ingredient achieved from Jerusalem artichoke tubers can be used to fortify food and also as a sweetening agent for products destined to diabetics.

Abstract:The purpose of this work is development of the universal technology of processing of a girasol and chicory for an inulin according to requirements to quality of a feed stock. An inulin — the reserve carbohydrate widespread in flora. There is the most of inulin quantity in chicory, a girasol, much less is in onions, garlic, inula, yakena, etc. The inulin is widely applied as a low-calorie product in a dietary and diabetic food, just also as a jelly former in food. In the All-Russian Research Institute of Starch Products there are conducted researches on development of the universal technology for an inulin from inulin-containing raw materials. The technological assessment of a girasol and chicory from various regions of the Russian Federation is carried out, their physical and chemical indexes are defined. Requirements to processing behavior of raw materials are developed for inulin processing. By results of researches initial requirements to inulin-containing raw materials are defined: a mass fraction of dry solids is not less than 25 %, an inulin is not less than 16 %; mono — and disaccharides are 2 %. Diffusion from freshly harvested and dry chicory shavings and a girasol is studied, optimal established conditions are temperature 80–85oC; the hydromodule for dry shaving — 1:7; for crude one — 1:2. It is developed the purification mode for extract and girasol and chicory syrups with use of various adsorbents such as the fissile coal, ion-exchange resins. The established high extent of extract and syrup purification is resulted in considerable decrease in a chromacity, mass fraction of a protein, ashes, etc. For researches as control specimens there was used an inulin of the foreign companies. Significance of the work is the creation of domestic technology of an inulin as prebiotic component for a treatmentand-prophylactic nutrition of the population and ensuring its import substitution.

Abstract:Globe artichoke is an intriguing source of indigestible sugar polymers such as inulin-type fructans. In this study, the effect of ultrasound in combination with ethanol precipitation to enhance the extraction of long chain fructans from artichoke wastes has been evaluated. The inulin-type fructans content both from bracts and stems was measured using an enzymatic fructanase-based assay, while its average degree of polymerization (DP) was determined by HPLC-RID analysis. Results show that this method provides artichoke extracts with an inulin-type fructans content of 70% with an average DP between 32 and 42 both in bracts and in stems. The prebiotic effect of long chain inulins from artichoke extract wastes was demonstrated by its ability to support the growth of five Lactobacillus and four Bifidobacterium species, previously characterized as probiotics. Besides, we considered the possibility to industrialize the process developing a simpler method for the production of inulin-type fructans from the artichoke wastes so that the artichoke inulin preparation could be suitable for its use in synbiotic formulations in combination with different probiotics for further studies including in vivo trials.

Abstract:Jerusalem artichoke (JA) is a tuber considered a functional food. The tubers accumulate a high amount of fructans (inulin and fructo-oligosaccharides) although tubers also have a high content of minerals and proteins of high biological value. In this study Jerusalem artichoke flour (JAF) was obtained by means of two drying methodologies: convective air oven drying and freeze-drying. Physico-chemical characteristics were analyzed in both samples, as well as a colorimetric analysis and microstructure. As a by-product of the production of JAF, a residual liquid is obtained from which fructans can be extracted, this process included separation steps in aqueous medium without the addition of organic solvents. The optimal extraction conditions were: solid/solvent ratio of 1:2 (Kg JA / L distilled water) during 40 min at 90 ± 5 °C. The flours obtained presented a high protein and fiber content, 9.73–9.94% and 1.21–1.47% respectively, while the morphology of the matrix of protein and inulin could be observed by microscopy. These results allow them to be considered as an alternative raw material for the production of gluten-free products in order to overcome the nutritional deficiency that gluten-free products generally have. The fructans extraction yield was affected by the temperature and storage time of the tubers. The concentration of fructans was performed by ultrafiltration, the process was stopped when the desired volume concentration ratio was achieved (VCR = 3.24). The concentrate stream was lyophilized. The results showed the following centesimal compositions: 6.65% proteins; 0.22% fat; 7.50% ash; 6.60% moisture and 4% inulin. The use of JA tubers allowed to obtain a gluten-free alternative flour with good nutritional characteristics. Therefore, JA tubers present a high potential for the production of inulin through chemical-free methods, from the by-product of the JAF process.

Abstract:Inulin is best found in the tubers of Jerusalem artichokes (JAT) (Helianthus tuberosus). Inulin has useful, nutritive, and functional traits. JAT was identified based on its chemical analysis, mineral content, and inulin concentration. In addition, inulin extracted from JAT was determined to have certain physiochemical properties. The sensory attributes, qualitative characteristics, mineral content, and quality attributes of chips substituted with various levels of JAT (3, 6 and 9%) were evaluated. The JAT comprised 85.25% moisture, 15.96% protein, and 4.23% ash. The JAT-produced powders were a rich source of K, Ca, Mg, Fe, and P minerals. The optimum sample to the solvent ratio for recovering the greatest amount of inulin (73.82%) was 1 g of sample to 9 mL of water at 540 W for 7 min. Inulin from JAT may hold 1.57 g/g water and 2.93 g/g oil. In addition, the sensory evaluation of the 3 and 6% JAT-enriched chips revealed excellent features. The amount of moisture and salt in the chips produced by adding JAT was lower than that of the control chips. Finally, JAT-enriched chips might be a beneficial ingredient that could be added to food products, particularly chips, to boost their nutritional value and act as a sweetener for diabetic products, particularly those marketed to children.

Abstract:-- Jerusalem artichoke (Helianthus tuberosus L.) is primarily used as a biomass for ethanol production. It is also utilized for human consumption as a healthy snack by having good nutritional value and inulin content. In this study, ultrasound pre-treatment (USPT) was selected to minimize drying defects and 5, 10 and 15 min. treatments were applied before hot air drying at 60 Â°C and 80 Â°C. The effect of pre-treatment on drying performances and properties as color, total phenolic content and total antioxidant were investigated. Effective diffusion coefficient values â€‹â€‹ranged from 1.72 Ã— 10-8 m2/min to 2.94 Ã— 10-8 to m2/min, and it was observed that the effective moisture diffusion coefficient was higher in the samples where ultrasound was applied compared to the control sample. L * value 72.72 Â± 2.71 was found highest at samples that were 5 minutes ultrasound treated and dried at 60 Â° C for 60 minutes. The highest antioxidant and phenolic content in the drying process at 60 Â°C was determined in the samples where ultrasound was applied for 15 minutes. Consequently, more conserved quality in shorter drying time with desired end product moisture content can be achieved by the application of USPT.

Abstract:The review informs on the results of the research of the application of inulin as a bioactive prebiotic in functional nutritional products, in cosmetic formulations and pharmaceuticals as the excipient of anticancer agents on the basis of the system analysis. The steady trend of the world growth of inulin production of 8-10 % a year has been noted. The aim of the research is to reveal and to analyze the effect of the system complex of inulin production in sequential chain from the inulin synthesis, its accumulation in plants and extraction. The Jerusalem artichoke is the most advanced kind of raw material which has the high crop yield of tubers up to 40 t/ha and inulin content up to 20 %. The sucrose and the synthesizing enzymes 1-SST and 1-FFT take an active part in the inulin biosynthesis as their genes can be edited with the purpose of changing the inulin content. At the Jerusalem artichoke breeding the most prospective trend is the cross-species hybridization drawing on the example of a topinsunflower with the crop yield of tubers of 40 t/ha. In the Jerusalem artichoke seed breeding the new methods of clonal micropropagation of tubers and its cultivation in the airhydroponic environment with the achievement of the inulin content no less than 20 % have been studied. At Jerusalem artichoke cultivation the breed is significative for the maximum crop yield of tubers with the row width of 90 cm and with the interval between tubers in a row – 30 cm. The least loss of the tubers weight and the inulin content in them under the long-term storage is achieved at the temperatures from -5 to 0 °C. The technology of the Jerusalem artichoke tubers processing into inulin is suggested as the system of partly optimized technological operations from peeling and washing the tubers to receiving the powdered inulin and the oligofructose syrup. The system complex is suggested in the form of the structural topological model combining 4 systems: «Selection and seed breeding», «Agritechnologies», «Harvesting and storage of tubers», «The tubers processing technologies» with the interrelations in the form of the technological requirements and the output data of every system and of the complex as a whole, which is the basis for the development of the agri-food technology of inulin from Jerusalem artichoke and other kinds of raw materials.

Abstract:The chemical composition and agrotechnical characteristics of Jerusalem artichoke (Helianthus tuberosus) determine the wide prospects for its use in various branches of the processing industry, including food biotechnology. The aim of the study was to determine the possibility to use Jerusalem artichoke processed products from various stages of alcohol production in native form without drying in the technology of extruded products and to assess the impact of the raw material selection stage on extrusion modes and quality indicators of the finished product. In the course of the experimental work, Jerusalem artichoke tubers were processed into alcohol, samples of the high-moisture dispersed fraction of Jerusalem artichoke were taken from different stages of the process. These fractions were added in an amount of 15 % without pre-drying into the mixture for extrusion based on rice groats. The obtained extrudates were dried and their structural-mechanical, color characteristics and nutritional value were determined. The control sample was without the addition of Jerusalem artichoke additives. It has been established that during the extrusion of test samples, compared with the control, the expansion index and the number of microfractures significantly decrease. These data show that with the addition of Jerusalem artichoke fractions, the samples become less porous, their crispness decreases. The expansion index of all experimental samples decreased by 42-49 %. The maximum hardness of 22.9 N, which is 56 % higher than the control value, was noted for a sample with a fraction taken after the fermentation stage. The use of food additives of Jerusalem artichoke from the stages of hydromechanical processing and after fermentation, added in the amount of 15 % to the mixture for extrusion, significantly, by 52-61 %, increased the content of dietary fiber. More promising is the use of Jerusalem artichoke as an ingredient after a full cycle of its processing into alcohol, which makes it possible to enrich the product with microbiological protein of alcohol yeast biomass and increase the total protein content.

Abstract:A three-stage homogenate extraction was used as a new method for inulin extraction from Jerusalem artichoke tubers. Compared with the results from conventional hot water extraction, the three-stage homogenate extraction gave higher yields and caused less degradation of the inulin. The inulin crude extract was then clarified by a carbonate-precipitation method, during which three variables — the quicklime mass, the reaction temperature and the reaction time were optimized for the main liming process to give the best clarification effect. A Plackett-Burman design, the path of steepest ascent method, a Box-Behnken design and response surface methodology (RSM) were employed in the experimental design. The optimal conditions for the main liming process were determined to be 12.0 g/L, 71.4°C and 8 min. The confirmatory tests proved that the best clarification efficiency (92.74%) was achieved at these conditions and this was approximately equal to the value predicted by the model.
