Abstract:Fluoride toxicity through potable water, particularly ground water, is not uncommon in countries such as India, China, Iran, Iraq, Turkey, parts of Africa and Afghanistan. Kidney being the main organ involved in fluoride removal, it accumulates considerable amount of fluoride. Here, we report toxic effects of oral exposure of Swiss albino mice to fluoride (sub-acute: 190 mg/kg body wt. for 7 days; and sub-chronic: 94 mg/kg body wt. for 90 days) and recovery of sub-chronic fluoride exposed mice after 90 days of sodium fluoride (NaF) withdrawal. The role of diet supplements (Spirulina and tamarind fruit pulp @ 230 mg/kg body wt. independently as well as in combination) in amelioration of fluoride toxicity has also been screened. Compared with controls, feed intake decreased from 3-43%, body wt. 4-18%, and kidney wt. 5-12% in treated mice (except diet supplement groups of sub-chronic exposure) while their water intake increased from 4-43%. Histopathological changes in the cortical region of kidney in fluoride treated mice were as follows: dilation of bowman's capsule and thickening of its parietal and visceral layer; alterations in glomeruli size and their sclerotization; increase in bowman's space; proliferation of mesangial cells; reduction in podocyte counts; and dilation of proximal and distal tubules. Fluoride exposure altered tissue biochemistry (protein, acid phosphatase and alkaline phosphatase content) and increased urea (23-58%) and creatinine content (14-127%) in the serum. Sub-acute exposure was found more toxic. The diet modulation not only reduced fluoride toxicity but also led to better recovery of treated mice after withdrawal, especially in combination.

Abstract:Recent studies have shown a strong correlation between ambient fine particulate matter (PM2.5) exposure and diabetes risk, including abnormal lipid accumulation and systemic insulin resistance (IR). Hawthorn total flavonoids (HF) are the main groups of active substances in Hawthorn, which showed anti-hyperlipidemic and anti-hyperglycemic effects. Therefore, we hypothesized that HF may attenuate PM2.5-induced IR and abnormal lipid accumulation. Female C57BL/6 N mice were randomly assigned to the filtered air exposure (FA) group, concentrated PM2.5 exposure (PM) group, PM2.5 exposure maintained on a low-dose HF diet (LHF) group, and PM2.5 exposure maintained on a high-dose HF diet (HHF) group for an 8-week PM2.5 exposure using a whole-body exposure device. Body glucose homeostasis, lipid profiles in the liver and serum, and enzymes responsible for hepatic lipid metabolism were measured. We found that exposure to PM2.5 impaired glucose tolerance and insulin sensitivity. In addition, triacylglycerol (TAG) in serum elevated, whereas hepatic TAG levels were decreased after PM2.5 exposure, accompanied by inhibited fatty acid uptake, lipogenesis, and lipolysis in the liver. HF administration, on the other hand, balanced the hepatic TAG levels by increasing fatty acid uptake and decreasing lipid export, leading to alleviated systemic IR and hyperlipidemia in PM2.5-exposed mice. Therefore, HF administration may be an effective strategy to protect against PM2.5-induced IR and metabolic abnormalities of lipids.
Reduction of blood lead levels in lead-exposed mice by dietary supplements and natural antioxidants.

Abstract:BACKGROUND: Lead exposure is a global environmental problem that induces lifelong adverse health effects. Our aim was to investigate the effect of dietary supplements and natural antioxidants on blood lead levels (BLL) in lead-exposed mice, and observe their impact on the absorption of calcium, iron and zinc in vivo. RESULTS: All of selected dietary supplements (calcium lactate, zinc sulfate, ferrous sulfate, ascorbic acid and calcium-rich milk) and natural! antioxidants (extract of Chinese wolfberry, extract of Hangzhou white chrysanthemum and antioxidant extract of bamboo leaves) have promising capacity of reducing BLL in lead-exposed mice with an reduction range from 56.2% to 65.1%. The metal ion-mediated chelating and competitively inhibitory mechanisms may elucidate their reduction effect. Besides, blood calcium, iron and zinc levels were not significantly changed in all of the experimental groups, indicating that the intake of all additives does not disturb the absorption of essential mineral elements in mice. CONCLUSION: All the studied additives not only effectively reduce BLL, but also maintain normal calcium, iron and zinc absorption in mice. The formulation of calcium, iron and zinc supplements and/or polyphenol and vitamin-rich antioxidants may constitute an important secondary prevention effort to reduce BLL and lead exposure. Vitamin E supplementation protects oxidative stress during arsenic and fluoride antagonism in male mice

Abstract:Arsenic and fluoride are common environmental contaminants. Coexposure to these elements can occur through groundwater. We investigated the effects of sodium meta arsenite (50 mg/L in drinking water) and sodium fluoride (50 mg/L in drinking water) individually and in combination. Biochemical parameters suggestive of alterations in heme synthesis pathway, oxidative stress in liver and kidneys, and concentration of essential metals in blood and soft tissues were studied in Swiss albino male mice given the chemicals for 3 weeks. The possible beneficial effect of vitamin E administration (25 mg/kg, oral, alternate days after arsenic/fluoride exposure) on the above variables was investigated. Exposure to arsenic or fluoride caused a significant depletion in blood delta-aminolevulinic acid dehydratase (ALAD) activity, platelet counts (PLT), and glutathione (GSH) level. Blood white blood cell (WBC) counts also decreased. These changes were accompanied by increased reactive oxygen species (ROS) levels. Arsenic and fluoride exposure led to a significant depletion of super oxide dismutase (SOD) activity with no effect on catalase and glutathione peroxidase (GPx) activities. Combined exposure to these toxicants had no synergistic effect on blood ALAD activity and WBC counts, and the effects seen appeared to result predominantly from arsenic. Hepatic catalase activity, on the other hand, increased significantly on exposure to arsenic and fluoride. There was only moderate antagonistic effect on arsenic and fluoride concentration in blood and liver, and kidney arsenic concentration was less pronounced during coexposure compared with arsenic alone. Interestingly, fluoride concentration showed less pronounced uptake during concomitant exposure compared with fluoride exposure alone. Vitamin E supplementation during coexposure to arsenic and fluoride provided only moderate recovery in the altered antioxidant enzymes and in depleting ROS level, but the altered essential metal concentration, particularly calcium level, responded more favorably to vitamin E administration. It can be concluded from the current study that (i) coadministration of arsenic and fluoride was less toxic to the animals compared with individual toxic effects of these toxicants, and (ii) vitamin E supplementation during coexposure had only limited additional beneficial effects in restoring altered biochemical variables, maintaining pro-oxidant/antioxidant balance, and reducing body arsenic store but plays a significant role in maintaining essential metal balance.
The Pathophysiological Changes and Clinical Effects of Tetramethylpyrazine in ICR Mice with Fluoride-Induced Hepatopathy

Abstract:The excessive intake of fluoride, one of the trace elements required to maintain health, leads to liver injury. Tetramethylpyrazine (TMP) is a kind of traditional Chinese medicine monomer with a good antioxidant and hepatoprotective function. The aim of this study was to investigate the effect of TMP on liver injury induced by acute fluorosis. A total of 60 1-month-old male ICR mice were selected. All mice were randomly divided into five groups: a control (K) group, a model (F) group, a low-dose (LT) group, a medium-dose (MT) group, and a high-dose (HT) group. The control and model groups were given distilled water, while 40 mg/kg (LT), 80 mg/kg (MT), or 160 mg/kg (HT) of TMP was fed by gavage for two weeks, with a maximum gavage volume for the mice of 0.2 mL/10 g/d. Except for the control group, all groups were given fluoride (35 mg/kg) by an intraperitoneal injection on the last day of the experiment. The results of this study showed that, compared with the model group, TMP alleviated the pathological changes in the liver induced by the fluoride and improved the ultrastructure of liver cells; TMP significantly decreased the levels of ALT, AST, and MDA (p < 0.05) and increased the levels of T-AOC, T-SOD, and GSH (p < 0.05). The results of mRNA detection showed that TMP significantly increased the mRNA expression levels of Nrf2, HO-1, CAT, GSH-Px, and SOD in the liver compared with the model group (p < 0.05). In conclusion, TMP can inhibit oxidative stress by activating the Nrf2 pathway and alleviate the liver injury induced by fluoride.

Effects of Fluorosis on Lipid Peroxidation and Antioxidant Systems in Young Pigs

Abstract:This study was conducted to investigate effects of fluoride on lipid peroxidation and antioxidant systems in young pigs. Three groups of crossbred barrows about 50 days old were exposed to 100, 250, and 400 mg F-/kg (from NaF) in their diets for 50 days. Serum malondialdehyde (MDA) and nitric oxide (NO) levels were significantly increased, and the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) were significantly decreased. In thyroid, liver, and kidney tissues, the MDA level was significantly increased, and SOD and GSH-Px activities were significantly decreased. These results suggest that fluoride induces excessive production of NO and reactive oxygen species (ROS), enhances lipid peroxidation, and disturbs the antioxidant system of pigs. Oxidative damage from oxidative stress could therefore be an important pathway for fluoride toxicity in soft tissues.
Possible Protective Role of Aqueous Tomato Extract on Hemato-Biochemical Parameters against Sodium Arsenite Toxicity in Albino Rats

Abstract:Due to rapid industrialization and technological development, humans are constantly exposed to heavy metals at minimum to maximum tolerable levels. Arsenic is ubiquitously distributed and most concerned element now-a-days, inorganic arsenicals mainly enter into the body via the route of lungs and gastrointestinal tract and 55-95% of it gets absorbed. Sodium arsenite poisoning leads to disturbance in hematological and biochemical processes. In the present study, toxic effects of sodium arsenite on the hematological parameters, serum transaminases and possible modulatory role of aqueous tomato extract were studied. Female rats were allocated into four different groups: 1. Control (given normal diet), 2. Sodium arsenite treated (10mg/kg B.W.), 3. Combination of sodium arsenite (10 mg/kg B.W.) and aqueous tomato extract (50 mg/kg B.W.) treated rats 4. Only aqueous tomato extract treated (50mg/kg B.W.) rats. It was observed that arsenic significantly decreased the total erythrocytes count (RBC), hemoglobin concentration (Hb), hematocrit value (HCT) and platelets (PLT), whereas significantly elevated the total leucocytes count (TLC) and activities of serum transaminases such as ALT and AST. Supplementation of aqueous tomato extract to arsenic treated rats showed decrease in values of WBC, ALT and AST which almost matched to values of control rats. So, from this study it can be concluded that antioxidants and vitamins present in the aqueous tomato extract showed protective role against arsenic toxicity in albino rats.
Effect of excess dietary fluoride on laying performance and antioxidant capacity of laying hens.

Abstract:The objective of this study was to investigate the effects of excess dietary fluoride (F) on laying performance and antioxidant capacity of laying hens. A total of 576 laying hens, 51 wk old, was randomly divided into 6 groups, each of which included 6 replicates of 16 hens. Graded amounts of sodium fluoride (NaF) were added to the basal diet to achieve concentrations of 16 (control), 200, 400, 600, 800, and 1,000 mg/kg F, respectively. Dietary F at 1,000 mg/kg significantly decreased ADFI, laying rate, and average egg weight, and increased feed conversion ratio (FCR) (P < 0.05). No significant differences were observed in serum total antioxidant capacity (T-AOC) level or catalase (CAT) concentration among all the treatments, while hens fed F at 800 and 1,000 mg/kg had higher activity of serum glutathione peroxidase (GSH-PX) and concentration of malondialdehyde (MDA) (P < 0.05) as compared to the control group. Compared with the control group, dietary F at 400 mg/kg increased liver MDA concentration (P < 0.001), and decreased CAT concentration of liver (P < 0.001); 600 mg/kg F decreased liver T-AOC levels (P < 0.001); and 800 mg/kg of F decreased liver total superoxide dismutases (T-SOD) activity (P < 0.001). Compared with the control group, feeding F at 600 mg/kg decreased kidney T-AOC levels and T-SOD activity (P < 0.001), and increased MDA concentration of kidney (P < 0.001), while dietary 1,000 mg/kg of F decreased kidney GSH-PX activity (P < 0.05) and CAT concentration (P < 0.001). In conclusion, these results indicated that excessive F ingestion had an adverse effect on laying performance by inducing oxidative stress and impairing the antioxidant system of laying hens.

Abstract:The effect of combined administration of calcium (Ca), iron (Fe), zinc (Zn), chrysanthemum flavonoids, and meso‐2,3‐dimercaptosuccinic acid (DMSA) on the treatment of lead (Pb) intoxication in mice was studied. One hundred ninety female mice (SPF level, aged 18‐22 days) were randomly divided into two groups as experimental animals. Mice in group I (10 mice) served as normal control animals, and were administered deionized water containing 12.5 μL/L acetate acid for 6 weeks, whereas mice in group II (180 mice) were exposed to 0.1% (wt/vol) of lead acetate in deionized water for 6 weeks and served as experimental animals. After 6 weeks of successful modeling, 180 mice from group II (lead‐exposed) were divided into 18 groups of 10 mice each, 16 of which were treated by the combined administration of Ca, Fe, Zn, chrysanthemum flavonoids, and DMSA by L16 (215) orthogonal design. The remaining two groups were given treatment with low and high doses of DMSA, respectively. After three weeks of intervention (ig), the optimal treatment group was identified according to its blood lead level, as well as some antioxidant indices in the blood, liver, and hippocampus. The results indicated that the combined administration of Fe, Zn, chrysanthemum flavonoids, and DMSA with low dosage had the most significant effect on increasing the activities of blood delta‐aminolevulinic acid dehydratase and superoxide dismutase (SOD), hepatic SOD and hippocampus nitric oxide synthase while decreasing the blood lead level, the content of hepatic malondialdehyde and hippocampus nitric oxide; this was considered the optimal treatment group. There was no difference in the level of blood hemoglobin between the optimal treatment group and the model control group (the first group of the orthogonal experiment). The activities of blood glutathione (GSH), hepatic GSH and glutathione peroxidase of the optimal treatment group were the same as other groups', and the recovery of the related indexes in the optimal effect group closely resembled the high dosage DMSA group. It can be concluded that the coadministration of Fe, Zn, and chrysanthemum flavonoids along with a low‐dose DMSA effectively reduces Pb poisoning and lead‐induced oxidative damage in lead‐exposed mice; the result may provide a theoretical reference for the treatment of Pb poisoning.

Alleviation of fluoride-induced colitis by tea polysaccharides: Insights into the role of Limosilactobacillus vaginalis and butyric acid

Abstract:Endemic fluorosis has gained increasing attention as a public health concern, and the escalating risk of colitis resulting from excessive fluoride intake calls for effective mitigation strategies. This study aimed to investigate the potential mechanisms underlying the alleviation of fluoride-induced colitis by Tea polysaccharides (TPS). Under conditions of excessive fluoride intake, significant changes were observed in the gut microbiota of rats, leading to aggravated colitis. However, the intervention of TPS exerted a notable alleviating effect on colitis symptoms. Antibiotic intervention and fecal microbiota transplantation (FMT) experiments provided evidence that TPS-mediated relief of fluoride-induced colitis is mediated through its effects on the gut microbiota. Furthermore, TPS supplementation was found to modulate the structure of gut microbiota, enhance the relative abundance of Limosilactobacillus vaginalis in the gut microbiota, and promote the expression of short-chain fatty acid (SCFAs) receptors in colonic tissue. Notably, L. vaginalis played a significant role in alleviating fluoride-induced colitis and facilitating the absorption of butyric acid in the rat colon. Subsequent butyric acid intervention experiments confirmed its remarkable alleviating effect on fluoride-induced colitis. Overall, these findings provide a potential preventive strategy for fluoride-induced colitis by TPS intervention, which is mediated by L. vaginalis and butyric acid.

Turmeric counteracts manganese-associated deteriorations in liver and kidney tissues: histomorphometric and biochemical insights

Abstract:Nowadays, Manganese (Mn) become an unavoidable ingredient in agriculture, medical and manufacturing industries. Manganese deficiency is rare, and even though recurrent exposure to manganese is inevitable for humans, concerns have been voiced regarding public health hazards. This research was designed to evaluate the manganese toxicity and potential protective effects of turmeric in Swiss albino male mice. Eight-week albino mice (33 ± 01 g) were randomized equally into seven groups ( n = 10), as control (untreated), VC-I (received 0.1 ml of saline water), Mn-I, Mn-II & Mn-III administrated manganese (5 mg/kg, 10 mg/kg & 20 mg/kg body-weight (B.W.) respectively), Mn + Tu (co-administrated with 20 mg/kg Mn &100 mg/kg B. W turmeric), VC-II (received 0.1 ml of olive oil), orally for four weeks consecutively. Before administrations, ferric reducing antioxidant power (FRAP), total phenolic content (TPC) and gass chromatography-mass spectroscopy (GC- MS) assays were performed to assess the antioxidant capacity of turmeric. After 28 days of dosing, mice were acclimatized for 10 days, then sacrificed and recovered samples (tissues & blood). Morphological observations showed changes in color, shape disruption, dilation and flattening of organs as compared to controls. Morphometric studies showed that the average body weight, organs weight and size of Mn intoxicated mice reduced remarkably ( P ≤ 0.05) as compared to control groups. Sideways Mn + Tu group showed non-significant change in both parameters. Hematological (CBC) and biochemical markers, liver function test & renal function test (LFT & RFT) showed significant differences in manganese groups against controls. Histopathological findings of both tissues (liver & kidney) showed multiple anatomic alterations in Mn exposed as compared to other groups. While the aforementioned deviations were less obvious in Mn + Tu administrated group. Manganese also decreased glutathione-s-transferase, superoxide dismutase, reduced glutathione contents and increased lipid peroxidation as compared to control groups. It is concluded by the abovementioned findings that turmeric showed remedial efficacy against Mn-instigated histomorphometric, biochemical and enzymatic alterations in mice.

Abstract:Lead (Pb) intoxication is a serious food safety issue, and the development of relevant dietary strategies is an area of ongoing research. In this study, two different dietary supplements were designed and evaluated for their effects against Pb toxicity in mice. Dietary supplement A contained grape seed extract, tea polyphenols and Lactobacillus plantarum CCFM8661, and dietary supplement B contained vitamin C, calcium carbonate, zinc acetate, and L. plantarum CCFM8661. The results showed that both dietary supplements could effectively decrease Pb levels, protect aminolevulinic acid dehydratase, superoxide dismutase and catalase activities and recover glutathione, zinc protoporphyrin and malondialdehyde levels in tissues and blood of mice. A step-through passive avoidance task confirmed that the dietary supplements could recover the learning and memory capacities of Pb-exposed mice. The protective effects of both dietary supplements to alleviate oxidative stress and cognitive impairments were superior to the chelator treatment. Administration of the dietary supplements during Pb exposure offered more significant protection than administration after Pb exposure. Animal safety evaluation also indicated that these dietary supplements barely induced side effects in the mice. This study provides evidence that dietary supplements containing probiotics, micronutrients, and plant extracts can be considered a new dietary strategy against Pb toxicity.

Abstract:Background: Fluoride is an inorganic element present everywhere in the environment and has cumulative toxic effects from prolonged ingestion. Green tea used since ancient times as a medical drink due to its multiple health benefits owing to its anti-oxidative and anti-apoptotic activities. Aim: To the best of our knowledge there were no studies until now concerning the possible protective role of green tea extract on the intestinal mucosal changes induced by fluoride on adult albino rats, so this work tried to investigate this role. Materials and Methods: Forty male adult albino rats randomly and equally divided into four groups, Group I: received distilled water only orally via a gastric tube. Group II: received green tea extract (GTE) 150 mg/kg dissolved in distilled water and freshly prepared for 35 days. Group III: received sodium fluoride (NaF) once daily oral dose of 10 mg/kg dissolved in distilled water for 35 days. Group IV: received NaF in the same dose as group III concomitant with GTE as the previously adjusted dose for 35 days. Body weight of all the studied groups was recorded at the start and after 35 days from the beginning of this study. After that the study tested the antioxidant and oxidant parameters in intestinal tissue as MDA, reduced GSH, and CAT, histopathological and immunohistochemical examination of intestinal tissue using PCNA marker were also included. Results: the rate of weight gain was little in NaF group compared to other groups. Concomitant administration of GTE with NaF resulted in highly significant decrease in MDA level, highly significant increase in reduced GSH level and highly significant elevation in CAT level when compared with NaF group. Many of abnormalities as moderate increase in goblet cell mass and highly expression of PCNA stain in a form of brown nuclear coloration were detected in intestinal tissues in NaF group by histopathological and immunohistochemical study that improved with administration of GTE. Conclusion: concomitant administration of GTE was improved the biochemical, histopathological and immunohistochemical alterations in intestinal mucosa that treated with NaF.
Attenuation of chromium (VI) and arsenic (III)-induced oxidative stress and hepatic apoptosis by phloretin, biochanin-A, and coenzyme Q10 via activation of SIRT1/Nrf2/HO-1/NQO1 signaling

Abstract:Heavy metal contamination is an alarming concern on a global scale, as drinking tainted water significantly increases human susceptibility to heavy metals. In a realistic scenario, humans are often exposed to a combination of harmful chemicals rather than a single toxicant. Phloretin (PHL), biochanin-A (BCA), and coenzyme Q10 (CoQ10) are bioactive compounds owning plentiful pharmacological properties. Henceforth, the current research explored the putative energizing effects of selected nutraceuticals in combined chromium (Cr) and arsenic (As) intoxicated Swiss albino mice. Potassium dichromate (75 ppm) and sodium meta-arsenite (100 ppm) were given in the drinking water to induce hepatotoxicity, conjugated with PHL and BCA (50 mg/kg each), and CoQ10 (10 mg/kg) intraperitoneally for 2 weeks. After the statistical evaluation, it was observed that the hepato-somatic index, metal load, and antioxidant activity (lipid peroxidation and protein carbonyl content) increased along with the concomitant decrease in the antioxidants (catalase, glutathione-S-transferase, superoxide dismutase, reduced glutathione, and total thiol) in the Cr and As intoxicated mice. Additionally, light microscopy observations, DNA breakages, decreased silent information regulator 1 (SIRT1), nuclear factor (erythroid-derived 2)-like 2 (Nrf2), heme oxygenase (HO-1), and NAD(P)H quinone dehydrogenase 1 (NQO1) gene expressions, together with stimulated apoptotic cell death manifested by the increased expressions of caspase 8 and caspase 3, thus, proved consistency with the aforementioned outcomes. Importantly, the treatment with nutraceuticals not only restored the antioxidant activity but also favorably altered the expressions of SIRT1, Nrf2, HO-1, and NQO1 signaling and apoptosis markers. These findings highlight the crucial role of the PHL, BCA, and CoQ10 combination in reducing Cr and As-induced hepatotoxicity in mice. By averting the triggered apoptosis in conjunction with oxidative stress, this combination increases the SIRT1, Nrf2, HO-1, and NQO1 signaling, thereby reassuringly maintaining the cellular equilibrium.
Selenomethionine supplementation mitigates fluoride-induced liver apoptosis and inflammatory reactions by blocking Parkin-mediated mitophagy in mice

Abstract:As an environmental pollutant, fluoride-induced liver damage is directly linked to mitochondrial alteration and oxidative stress. Selenium's antioxidant capacity has been shown to alleviate liver damage. Emerging research proves that E3 ubiquitin ligase Park2 (Parkin)-mediated mitophagy may be a therapeutic target for fluorosis. The current study explored the effect of diverse selenium sources on fluoride-caused liver injury and the role of Parkin-mediated mitophagy in this intervention process. Therefore, this study established a fluoride-different selenium sources co-intervention wild-type (WT) mouse model and a fluoride-optimum selenium sources co-intervention Parkin gene knockout (Parkin-/-) mouse model. Our results show that selenomethionine (SeMet) is the optimum selenium supplementation form for mice suffering from fluorosis when compared to sodium selenite and chitosan nano‑selenium because mice from the F-SeMet group showed more closely normal growth and development levels of liver function, antioxidant capacity, and anti-inflammatory ability. Explicitly, SeMet ameliorated liver inflammation and cell apoptosis in fluoride-toxic mice, accomplished through downregulating the mRNA and protein expression levels associated with mitochondrial fusion and fission, mitophagy, apoptosis, inflammatory signalling pathway of nuclear factor-kappa B (NF-κB), reducing the protein expression levels of PARKIN, PTEN-induced putative kinase1 (PINK1), SQSTM1/p62 (P62), microtubule-associated protein light chain 3 (LC3), cysteinyl aspartate specific proteinase 3 (CASPAS3), as well as restraining the content of interleukin-1β (IL-1β), interleukin-6 (IL-6), tumor necrosis factor α (TNF-α), and interferon-γ (IFN-γ). The Parkin-/- showed comparable positive effects to the SeMet in the liver of fluorosis mice. The structure of the mitochondria, mRNA, protein expression levels, and the content of proinflammatory factors in mice from the FParkin-/- and F + SeMetParkin-/- groups closely resembled those in the F + SeMetWT group. Overall, the above results indicated that SeMet could alleviate fluoride-triggered inflammation and apoptosis in mice liver via blocking Parkin-mediated mitophagy.
Ginkgo biloba ameliorates fluoride toxicity in rats by altering histopathology, serum enzymes of heme metabolism and oxidative stress without affecting brain mGluR5 gene

Abstract: Background: To evaluate the therapeutic potential of Ginkgo biloba extract (GBE) in experimental model of fluorosis. Objectives: To study the protective effect of GBE in fluoride toxicity by assessment of oxidative stress, serum biochemical parameters, acetylcholinesterase (AChE) activity, histopathology and brain mGluR5 gene expression. Materials and Methods: Fifteen adult male Wistar rats were randomly assigned to 5 groups (n = 3 rats in each group). Group 1 (control) received water, Groups 2–5 were treated with 100 ppm of sodium fluoride for 30 days, while the Groups 3, 4 and 5 were GBE treated with 50 mg/kg, 100 mg/kg and 200 mg/kg body weight for 15 days, after sodium fluoride treatment for 30 days. Results: Elevated serum delta aminolevulinic acid dehydratase and delta aminolevulinic acid synthatase levels in fluoride intoxicated rats were ameliorated by various doses of GBE treatment. Elevated serum glutathione and decreased oxidized gluatathione levels observed in fluoride intoxicated rats were also ameliorated by GBE treatment but effectively at 100 mg/kg dose. Reduced AChE activity of hippocampus in fluoride-induced toxicity was reverted by 50 mg/kg of GBE whereas other doses (100 and 200 mg/kg) caused significant inhibition of AChE activity in comparison with fluoride group. Fluoride group rats showed significant reduction of mGluR5 gene expression levels whereas in all GBE treatment groups those changes Were not significantly reverted. GBE treatment to fluoride intoxicated rats almost reverted the degenerative changes in liver and kidney caused by fluorosis. Conclusion: The present study concluded beneficial effects of GBE in experimental model of fluorosis.

Abstract:In recent years, the influence of chronic fluorosis on the brain has been widely reported. Our study aimed to demonstrate the potential mechanism underlying the impairment of memory function by excessive fluorine intake. We also evaluated whether improvement of intestinal microflora could be a potential therapy to prevent the negative influences from the perspective of gut-brain axis. Male ICR mice were randomly divided into three groups and administered with either phosphate buffered saline (PBS) (Control and F groups) or Lactobacillus johnsonii BS15 (FP group; daily amounts of 1 × 109 CFU/mL), a probiotic strain, by oral gavage throughout a 98-day experimental period. Sodium fluoride (100 mg/L) was added to the drinking water of the F and FP groups. Animals were sacrificed for sampling with or without water avoidance stress (WAS) at two phases of the experiment and behavioral tests including T-maze test and passive avoidance test were also performed. Based on the results of behavioral tests, probiotic reversed the fluorine-induced memory dysfunction. In addition, L. johnsonii BS15 also increased the antioxidant capacities (serum and hippocampal tissue) and hippocampal synaptic plasticity-related mRNA expression after excessive fluoride ingestion. Moreover, the increased colonization of L. johnsonii BS15 also protected the small intestines from the damages of growth performance, visceral indexes, intestinal development, digestive, and secretory functions by changing the structure of the microflora and then improving intestinal permeability and integrity. L. johnsonii BS15 also improved the ability of flourosis mice against psychological stress indicated by the changes in behavioral tasks, hippocampal antioxidant levels, and synaptic plasticity-related mRNA expressions. Lactobacillus johnsonii BS15 intake appears as a promising way to ameliorate fluorine-induced memory dysfunction, especially under psychological stress.

Abstract:To evaluate the protective role of lycopene (LYC) against aflatoxin B-1 (AFB(1))-induced erythrocyte dysfunction and oxidative stress, male kunming mice were treated with LYC (5 mg/kg) and/or AFB(1) (0.75 mg/kg) by intragastric administration for 30 d. Hematological indices were detected to assess erythrocyte function. The erythrocytes C3b receptor rate (E-C3bRR) and erythrocytes C3b immune complex rosette rate (E-ICRR) were detected to assess erythrocyte immune function. Hydrogen peroxide (H2O2) and malondialdehyde (MDA) contents and superoxide dismutase (SOD) and catalase (CAT) activities were determined to evaluate erythrocyte oxidative stress. The results showed that LYC administration significantly relieved AFB(1)-induced erythrocyte dysfunction by increasing the levels of red blood cell count (RBC), hemoglobin (HGB) and hematocrit (HCT), as well as reducing red blood cell volume distribution width (RDW) level, while the levels of mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC) and mean platelet volume (MPV) had no significant differences among the four groups. Besides, LYC ameliorated AFB(1)-induced erythrocyte immune dysfunction by increasing E-C3bRR and decreasing E-ICRR. Furthermore, LYC also alleviated AFB(1)-induced erythrocyte oxidative stress by decreasing H2O2 and MDA contents and increasing SOD and CAT activities. These results indicated that LYC protected against AFB(1)-induced erythrocyte dysfunction and oxidative stress in mice. The findings could lead a possible therapeutics for the management of AFB(1)-induced erythrocyte toxicity.
Fucoidan Protects against Acute Sulfoxaflor-Induced Hematological/Biochemical Alterations and Oxidative Stress in Male Mice

Abstract:Fucoidan is a sulfated polysaccharide which can be found among a number of macroalgea species. It has a broad spectrum of biological activities including anti-oxidant, anti-tumor, immunoregulation, anti-viral and anti-coagulant. The current study was performed to investigate possible protective effects of fucoidan for sulfoxaflor-induced hematological/biochemical alterations and oxidative stress in the blood of male Swiss albino mice. For this purpose, sulfoxaflor was administered at a dose of 15 mg/kg/day (1/50 oral LD50), and fucoidan was administered at a dose of 50 mg/kg/day by oral gavage alone and combined for 24 h and 7 days. Hematological parameters (RBC, HGB, HCT, MCV, MCH, MCHC, Plt, WBC, Neu, Lym and Mon), serum biochemical parameters (AST, ALT, GGT, LDH, BUN, Cre and TBil), and serum oxidative stress/antioxidant markers (8-OHdG, MDA, POC and GSH) were analyzed. The results indicated that sulfoxaflor altered hematological and biochemical parameters and caused oxidative stress in mice; fucoidan ameliorated some hematological and biochemical parameters and exhibited a protective role as an antioxidant against sulfoxaflor-induced oxidative stress.

Abstract:This study aims to investigate the mechanisms involved in the action of lutein (LU) alleviating arsenic-induced reproductive toxicity using mice model. Forty male Kunming mice were received following treatments by gavage: normal saline solution (control), arsenic trioxide (ATO; 5 mg/kg/day), LU (40 mg/kg/day), and ATO + LU (5 mg/kg/day + 40 mg/kg/day). At the end, the mice were killed by cervical dislocation and weighed. Pathological examination was done on the testis. The biomedical parameters including superoxide dismutase (SOD), glutathione (GSH), total antioxidative capability, malondialdehyde (MDA), 8-hydroxydeoxyguanosine (8-OHdG), and reproductive indexes were analyzed. The messenger RNA (mRNA) and protein expression of Nrf2, heme oxygenase 1 (HO-1), glutathione S-transferase (GST), nicotinamide adenine dinucleotide phosphate dehydrogenase, quinone 1 (NQO1) in testis were detected by real-time polymerase chain reaction and Western blot. We found that there was a decrease in sperm count; testis somatic index; the activities of SOD, GSH, total antioxidative capacity ( p < 0.01, respectively) in ATO-treated mice, while there was an increase in the levels of sperm abnormalities, MDA, and 8-OHdG than control ( p < 0.01, respectively). The groups treated with ATO + LU showed recovery of the measured parameters between those of ATO or saline-treated group. The antagonized interaction between ATO and LU was statistically significant ( p < 0.01). Mice treated with ATO + LU also showed greater mRNA expression of Nrf2, HO-1, NQO1, and GST than ATO or saline-treated groups. These findings suggest that LU alleviates reproductive toxicity induced by arsenic in male mice via Nrf2 signaling, which implicates a possible mechanism of LU in preventing the reproductive injury, and elucidates that consuming the rich plant sources of LU will alleviate the reproductive toxicity induced by chemicals.
